Abstract:The present study examined the effect of Lactobacillus plantarum ATCC14917 fermentation on the chemical composition and antioxidant activity of apple juice. Apple juice was fermented and examined of its antioxidant activity using chemical models and cellular antioxidant assay. Furthermore, the chemical composition of fermented apple juice was characterized by LC-MS/MS. Lactobacillus plantarum ATCC14917 fermentation showed an increase in DPPH and ABTS radical scavenging activity as well as cellular antioxidant activity of apple juice. However, fermentation decreased the total phenolic and flavonoid content. Subsequent LC-MS/MS analysis of the phenolic profile indicated that the content of 5-O-caffeoylquinic acid (5-CQA), quercetin, and phloretin with strong antioxidant activity was increased significantly after fermentation. The modified phenolic composition may contribute to the increased antioxidant activity of fermented apple juice. Our findings showed that Lactobacillus plantarum ATCC14917 fermentation may be an efficient way to enhance the bioavailability of phenolic compounds and to protect cells from oxidative stress.

Abstract:The ability of natural plants to treat chronic diseases is closely related to their antioxidant function. Lactic acid bacteria (LAB) fermentation is an effective way to improve the nutritional value, biological activity and flavor of food. This study investigated the pH, titratable acidity, total polysaccharide, total flavone, total saponin, total polyphenol, and antioxidant activity of the FH06 beverage before and after probiotic fermentation. Results: After fermentation, FH06 had lower contents of total polysaccharides, total flavonoids, total saponins and total polyphenols but higher titratable acidity. The antioxidant activity was tested by total antioxidant capacity (FRAP method) and DPPH· scavenging ability. The FRAP value significantly increased after fermentation ( P < 0.05), and the maximum increase was observed for Lactobacillus fermentum grx08 at 25.87%. For DPPH· scavenging ability, the value of all fermentations decreased, and L. fermentum grx08 had the smallest reduction at 2.21% ( P < 0.05). The results of GC–MS and sensory analysis showed that fermentation eliminated bad flavors, such as grass, cassia and bitterness, and highlighted the fruit aroma and soft sour taste. Conclusion: The FRAP value and sensory flavor of FH06 fermentation by L. fermentum grx08 were significantly improved, indicating its great potential as a functional food with both strong antioxidant activity and good flavor.

Abstract:The scattering of light within paper and the ink penetration in the substrate are the key factors which affect the color reproduction. A reflectance model for color halftone prints is introduced in this paper which considers these factors. The model is obtained by the extended Murray-Davies model and iterative algorithm. The model described in this paper contains two parameters, n and v . The n factor equals to the sum of w and v . The w factor relates to the optical spread function of paper relative to the spatial frequency of the halftone dots. The v factor relates to the distribution of colorant within the dots. The value of n and v are obtained by simulation according to the experimental data. For offset lithographic halftone data at 150lpi, the value of n and v are 1.578 and 0.02, thermal wax transfer halftone ( n =2.292, v =0.0209) and stochastic halftone ( n =1.1853, v =-0.006).
The Antioxidant and Anti-Fatigue Effects of Rare Ginsenosides and γ-Aminobutyric Acid in Fermented Ginseng and Germinated Brown Rice Puree

Abstract:γ-aminobutyric acid (GABA) and rare ginsenosides are good antioxidant and anti-fatigue active components that can be enriched via probiotic fermentation. In this study, ginseng and germinated brown rice were used as raw materials to produce six fermented purees using fermentation and non-fermentation technology. We tested the chemical composition of the purees and found that the content of GABA and rare ginsenoside (Rh4, Rg3, and CK) in the puree made of ginseng and germinated brown rice (FGB) increased significantly after fermentation. The antioxidant activity of the six purees was determined using cell-free experiments, and it was found that FGB had better ferric-ion-reducing antioxidant power (FRAP) and 1,1-diphenyl-2-picryl-hydrazyl (DPPH) free radical scavenging rates, exhibiting better antioxidant effects. We then evaluated the antioxidant effect of FGB in HepG2 cells induced by H2O2 and found that FGB can reduce the generation of reactive oxygen species (ROS) in HepG2 cells and increase the membrane potential level, thereby improving oxidative damage in these cells. In vivo experiments also showed that FGB has good antioxidant and anti-fatigue activities, which can prolong the exhaustive swimming time of mice and reduce the accumulation of metabolites, and is accompanied by a corresponding increase in liver glycogen and muscle glycogen levels as well as superoxide dismutase and lactate dehydrogenase activities. Finally, we believe that the substances with good antioxidant and anti-fatigue activity found in FGB are derived from co-fermented enriched GABA and rare ginsenosides.

Abstract:This study aimed to investigate the changes in physicochemical properties, bioactivities and metabolites of fermented goji juice (FGJ) by Lacticaseibacillus rhamnosus at different fermentation stages. The results showed that Lacticaseibacillus rhamnosus fermentation significantly decreased the content of soluble protein, total phenolic, total flavonoid and total sugar. Meanwhile, the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging ability and the inhibition rate of xanthine oxidase (XOD) activity were remarkably enhanced by Lacticaseibacillus rhamnosus fermentation. Flavor profiles analysis indicated that FGJ produced novel volatile compounds such as 4-methylpentanol and 2-butanol, which provide its distinct aroma. The non-targeted metabolomics analysis showed that the differential metabolites in the FGJ28 vs. FGJ0 group were mainly included 1,7-bis (3,4-dihydroxyphenyl) heptan-3-yl acetate, isoplumbagin, triacetylresveratrol, sulochrin, indole-3-acetaldehyde, etc., which might have an effect on the promotion of the bioactivity of goji juice. These findings will contribute to understanding the biotransformation effect of Lacticaseibacillus rhamnosus fermentation on goji juice.

Abstract:DaiDai fruit, a medicinal and edible plant fruit, is abundant in biologically active compounds and has a long history of use in traditional Chinese medicine. This research focuses on utilizing fermentation to develop a functional DaiDai fruit fermentation broth. Lactobacillus, Bacillus subtilis and Saccharomyces cerevisiae were employed in the fermentation process. By conducting screenings of bacterial strains, single factor experiments, and response surface methodology, the total flavonoids, polysaccharides, polyphenols, and 1,1-diphenyl-2-trinitrophenylhydrazine (DPPH) free radical scavenging rate were used as the index for selection, ultimately identifying Lactobacillus L-13 as the optimal fermentation strain. The optimal fermentation conditions were determined to be a time of 108 h, a temperature of 43.6°C, and a solid-liquid ratio of 1:15.157 (w/v). Under these conditions, the total flavonoid content reached 412.01 mg/g, representing a 36.71% increase compared to conventional extraction methods. The contents of polysaccharides and polyphenols and the DPPH scavenging rate were also increased. The fermentation broth of DaiDai fruit exhibited inhibitory effects on tyrosinase and melanin production in mouse melanoma cells B16-F10 induced by α-MSH and anti-inflammatory properties in a zebrafish inflammation model. These indicate that the DaiDai fruit fermentation broth possesses anti-melanoma, whitening, and anti-inflammatory properties, showcasing significant potential for applications in medicine, cosmetics, and other industries.

Abstract:Nonalcoholic fermentation could improve the organoleptic quality and functionality of fruit juice, and lactic acid bacteria (LAB)-fermented fruit juice has been proven to be beneficial to human health. However, LAB exhibit strain specificity with different food matrices. In the present study, the effects of L. plantarum and L. acidophilus on the physicochemical characteristics, antioxidant activity and metabolomics of loquat juices were investigated. Both LAB strains fermentation could significantly enhance the antioxidant activity of loquat juice. L. acidophilus grows faster than L. plantarum in loquat juice, but L. plantarum fermentation more effectively increased the total flavonoid content. Nontargeted metabolomics analysis based on UHPLC-Q Exactive HF-X/MS were performed to reveal the different metabolic profiles between LAB fermentation. A total of 776 metabolites were identified and annotated. The significant differentially regulated metabolites were concentrated mainly in lipids and lipid-like molecules; organic acids and derivatives; amino acids, peptides, and analogs; phenolic compounds and terpenoids. The regulation of metabolites plays a very important role in the flavor reconstruction and functional improvement of loquat juices. In addition, the metabolic profiles between L. plantarum- and L. acidophilus-fermented loquat juices were different, and the application of cofermentation of these two strains may be considered in future studies.

Abstract:This study used brewer’s yeast to ferment Dendrobium officinale and single-factor and orthogonal experiments were conducted to determine the optimal fermentation conditions. The antioxidant capacity of Dendrobium fermentation solution was also investigated by in vitro experiments, which showed that different concentrations of fermentation solution could effectively enhance the total antioxidant capacity of cells. The fermentation liquid was found to contain seven sugar compounds including glucose, galactose, rhamnose, arabinose, and xylose using gas chromatography–mass spectrometry (GC–MS) and high performance liquid chromatography-quadrupole-time of flight mass spectrometry (HPLC-Q-TOF-MS), with the highest concentrations of glucose and galactose at 194.628 and 103.899 μg/ml, respectively. The external fermentation liquid also contained six flavonoids with apigenin glycosides as the main structure and four phenolic acids including gallic acid, protocatechuic acid, catechol, and sessile pentosidine B.

Abstract:The main objective of this study was to optimize the fermentation conditions of goji and to analyze the sugar, organic acid, amino acid, and flavor compounds before and after the fermentation of goji. Based on the single-factor experiment, orthogonal array design and the range analysis, the optimized conditions for fermented goji were 30% raw material concentration, 28 h fermentation time and 0.05% inoculum size, respectively. Under optimal conditions, the biomass of Lactiplantibacillus plantarum NCU137 and sensory score of fermented goji reached to 8.84 +/- 0.05 log CFU/g and 81.3 +/- 1.5, respectively. After fermentation, the content of malate, succinic acid, and lactate increased, while the content of pyruvate, glucose, fructose, and the overall free amino acid decreased. After fermentation, volatile components except for the phenols and acids are declining. This study would provide theoretical basis and experimental data for the industrialization of Lycium barbarum fermentation. Novelty impact statement After Lactiplantibacillus plantarum NCU137 fermentation, the contents of organic acids and total acidity acid in fermented goji significantly increased, which could inhibit microbiological spoilage and growth. Moreover, we get a delicious fermented goji juice through the optimization of fermentation conditions. Therefore, the data obtained in this study provides a better understanding of the effects of raw material content, fermentation time, and inoculum size on the quality of fermented goji, which would be very beneficial for the production of high-quality fermented goji for subsequent consumption.
Nutritional and flavor properties of grape juice as affected by fermentation with lactic acid bacteria

Abstract:Two lactic acid bacteria (LAB) strains of Lactobacillus plantarum 21802 and Lactobacillus brevis 6239 were selected and combined for fermentation of grape juice. The changes of sugars, organic acids, volatiles, phenolic compounds as well as antioxidant capacity of the grape juice were characterized. Results showed that fermentation with LAB led to a slight decrease in sugar content and increased in organic acid content. The sugars were mainly consumed at the initial stage of fermentation, while the malic acid was consumed completely during the fermentation. The contents of total phenolics and flavonoids in grape juice decreased gradually during the fermentation process. As compared with the initial contents in unfermented grape juice, the contents of proanthocyanin B2, rutin, isoquercitrin, rhamnose-3-O-glucoside in the final fermented grape juice were increased by 26.83%, 249.24%, 138.48%, 139.60%, respectively, while the contents of gallic acid, proanthocyanin B1, catechin and epicatechin were decreased by 24.49%, 14.85%, 71.68%, 42.52%, respectively. The mice fed with the fermented grape juice showed higher activity of superoxide dismutase and lower level of malondialdehyde both in serum and liver tissue than those fed with the unfermented grape juice. Fermentation with LAB also enhanced the flavor profile by increasing the total number and content of volatile compounds, especially those of monoterpene alcohols, which are the main and characteristic aroma volatile compounds in Muscat aroma grapes. This research confirmed that fermentation with LAB might be a desirable option for improving the flavor and nutritional attributes of grape juice by altering the ratio of sugar to organic acid, improving the phenolic compounds, enhancing the antioxidant capacity, intensifying of the characteristic aroma and pleasant flavor of the grape juice.

Abstract:Abstract Consumption of functional juice is becoming increasingly popular. This study aimed to evaluate the effect of probiotic fermentation with Bacillus sp. DU-106; Lactobacillus planturum Lp-43 and Lactobacillus rhamnosus Lr-156 on the biochemical and functional characteristics, antioxidant activities and storage stability of Dendrobium officinale and banana (DOB) juice. The cell levels of the lactic acid bacteria reached near 9 log copies/mL after fermentation at 32 °C for 36 h and could keep in this level after storage at 4 °C for 28 days. After fermentation, total acidity, flavonoids and polysaccharide contents of DOB juice increased, while pH, dissolved oxygen, nitrite, nitrate, vitamin C, β-carotene, sugar contents and antioxidant capacity slightly decreased. Compared with nonfermented DOB juice, polyphenol oxidase (PPO) and peroxidase (POD) activities of fermented DOB decreased significantly, but the color value and the content of total phenols and vitamin C decreased slightly in fermented DOB juice during storage. Overall, the probiotic fermentation with selected strains could improve the quality the DOB juice, enhance antioxidant capacity and storage stability, and destroy accumulation of nitrite and nitrate during storage. Findings of this study would help in the development of beneficial beverages in industrial production.

Abstract:To enrich the flavor compounds and retain the content of polyphenolics in grape juice (GJ) under long-term storage, Lactiplantibacillus plantarum, Lactobacillus acidophilus, Lacticaseibacillus casei, and Lacticaseibacillus paracasei, were screened and the optimal fermentation conditions were determined as fermentation temperature of 41.2 °C for 24 h with an initial LAB density of 8.5 × 106 CFU/mL. Surprisingly, the retention rates of TPC still remained at 50% after storage for 45 days at 4 °C. Moreover, 251 different metabolites were identified, include 23 polyphenolics, 11 saccharides, and 9 organic acids. Most importantly, the total content of polyphenolics reserved was 92.65% at the end of fermentation. Among them, ephedrannin A content significantly decreased; however, 2',6'-Di-O-acetylononin gradually increased with the fermentation time, which resulted in FGJ maintaining excellent bioactivity. Meanwhile, organic acid content (palmitoylethanolamide, tetraacetylethylenediamine) increased with saccharides (linamarin) decreasing, which leads to FGJ having a unique taste. Furthermore, a total of 85 Volatile organic compounds (VOCs) were identified, mainly including esters, aldehydes, and alcohols. Interestingly, key VOCs could be formed by carboxylic acids and derivatives, and fatty acyls via complex metabolic pathways.
Effect of Lactobacillus acidophilus fermentation on bioaccessibility: The relationship between biotransformation and antioxidant activity of apple polyphenols based on metabolomics

Abstract:Apple ( Malus domestica ) polyphenols are strong antioxidants. However, the functional properties of apple polyphenols are limited due to their considerable low digestibility. Therefore, in this study, the effects of fermentation of apple pulp by Lactobacillus acidophilus on its total polyphenol content and antioxidant activity were assessed using metabolomics. The results showed that the total polyphenol content and antioxidant activity of apple pulp fermented with L. acidophilus zrx02 were significantly higher than those in the unfermented apple pulp. Six different significantly changed metabolites, including caffeic acid, hypericin, p -coumaric acid, p -hydroxybenzoic acid, ferulic acid, and eugenic acid, were selected using ultra-performance liquid chromatography-tandem mass spectrometry and a metabolomics analysis. A cluster analysis revealed that one group contained caffeic acid, while the other one contained ferulic acid, p -hydroxybenzoic acid, p -coumaric acid, hypericin, and syringic acid. Ten metabolic pathways with significant differences were screened using the hypergeometric method. Overall, the results suggest that after fermentation by L. acidophilus zrx02, apple polyphenols can be biotransformed into small molecules that are easily digested and absorbed, and they have improved antioxidant activity. It is a raw material source of antioxidant-rich active substances that merits full utilization.
Enhanced DPPH Radical Scavenging Activity and Enriched γ-Aminobutyric Acid in Mulberry Juice Fermented by the Probiotic Lactobacillus brevis S3

Abstract:Mulberries, known for their high sugar content and rich bioactive compounds, have attracted attention for their potential health benefits. γ-Aminobutyric acid (GABA) is an amino acid with multiple physiological functions. To increase the GABA content and enhance the antioxidant capacity in fermented mulberry beverages, we screened a high-yielding strain, Lactobacillus brevis S3, known for its probiotic properties. L. brevis S3 demonstrated an excellent tolerance to simulated gastric acid, gastric juice, intestinal fluid, bile salts, osmotic pressure, and phenol, making it a safe and valuable probiotic candidate for mulberry fermentation. We attempted the addition of different nutritional components to enhance the GABA content in mulberry juice, including 1% yeast extract; 0.5% peptone; 0.01% metal ion complex (magnesium sulfate, manganese sulfate, and ferrous sulfate); combinations of yeast extract and peptone, and all three components. Mulberry juice supplemented with all three components reached a viable cell count of 1.2 × 1010 CFU/mL after 72 h. The antioxidant capacity and GABA titer were enhanced. The DPPH free radical scavenging capacity increased by 1.62 times, and the GABA content reached 7.48 g/L. By utilizing L. brevis S3 with excellent probiotic properties and supplementation with nutritional components, it is possible to produce low-sugar mulberry functional beverages with a high DPPH free radical scavenging capacity that are rich in GABA.

Abstract:Probiotic fermented fruit juice could improve intestinal health with better sensory attributes. The effects of fermentation conditions on flavor quality of fermented bayberry juice were compared through microbial viability and sensory evaluation. Fermentation by combined Streptococcus thermophilus (ST) and Lactobacillus acidophilus (LA) resulted in higher microbial viability and sensory evaluation scores. The fermentation conditions were optimized by orthogonal experimental design and Technique for Order Preference by Similarity to an Ideal Solution analysis (ST and LA in a mass ratio of 3:1; 0.6% (mass concentration) inoculation; fermentation time, 48 h, at 37 degrees C; 10% (mass concentration) added sucrose). Fermentation under these conditions decreased the anthocyanin content of the juice, maintained the total phenolic content, and slightly decreased the antioxidant capacity. Fermentation reduced aroma volatiles from 42 in fresh juice to 33, as determined by gas chromatography-mass spectrometry-olfactometry (GC-MS-O), the aldehyde content decreased and the alcohol content increased. Lactic acid fermented bayberry juice by ST and LA produced a beverage with desirable consumer attributes, combining the health benefits of fresh juice and probiotics.

Abstract:Untargeted and targeted metabolomics based on LC-MS/MS was applied here to investigate the changes in metabolites following Lactiplantibacillus plantarum ( L. plantarum ) fermentation of goji juice. The changes in physicochemical characteristics, free amino acid and organic acid levels were quantified. Total phenols, total flavonoids, and antioxidant activity were increased after these L. plantarum fermentation. 31 primary metabolites were screened out by comparison between two groups using multivariate statistical analysis. L. plantarum consumed reducing sugar and most varieties of amino acids. Gamma amino butyric acid (GABA) and phenylacetaldehyde were significantly accumulated via precursor of glutamate and phenylalanine, respectively. Indole-3-lactic acid (ILA) was the characteristic secondary metabolite after fermentation. There were 6.25 and 4.19 mg/L ILA accumulated after LP-D and LP-F fermentation, respectively, whereas it was not detected in goji juice. The accumulation of tyrosol was strain-dependent. Part of the bitter-taste amino acids, including l -methinoine, l -phenylalanine and tryptophan, were converted into their corresponding acetyl-derivatives, which represented umami or kokumi taste. Overall, fermentation by L. plantarum strains altered the metabolite profile and increased the levels of several biologically active substances of goji juice. These results deepen the understanding of L. plantarum metabolism, and the bioconversion associated with flavor and bioactive compounds unraveled here can be used as references in other plant-based materials for LAB fermentation.

Abstract:The impact of fermentation on the structure and antioxidant potential of phenolic compounds in foods have received wide attention. In this study, five common phenolic compounds (ferulic acid, polydatin, cyanidin-3- O -glucoside, rutin, tannic acid) were fermented by Aspergillus oryzae, Lactobacillus plantarum and Saccharomyces cerevisiae respectively. The qualitative and quantitative analyses of the fermentation products were separately performed by HPLC-DAD-ESI-QTOF-MS/MS and HPLC-DAD, and the antioxidant properties (assessed by DPPH and ABTS assays) were also estimated. The results demonstrated that the glycosidic bonds in the phenolic compounds were hydrolyzed during fermentation, which led to the formation of additional phenolic metabolites such as gallic acid, protocatechuic acid and quercetin, as evidenced by the identification of 15 phenolic compounds in the fermented samples. The contents of these phenolics were considerably decreased ( p < 0.05) to 1.72%–97.99% during fermentation, while the phenolic metabolites were detected in certain amounts (0.44–15.53 mg/L). The antioxidant activities of these phenolics also varied among different fermenting microorganisms. This study provides direct evidence of the metabolites and antioxidant properties of these common food phenolic compounds after fermentation.

Abstract:Background: Douzhi, a traditional Chinese fermented beverage, features microbial communities primarily composed of lactic acid bacteria (LAB). As fermented foods continue to gain recognition and popularity, douzhi is attracting growing interest. However, investigation of the critical aspects of douzhi's fermentation processes, including fermentation characteristics and microbial community dynamics, remains vital for enhancing food safety and quality for douzhi, as well as for similar fermented food products. Method: In this study, we collected douzhi microbial communities from four chain stores, using them as fermentation starter cultures. The microbial dynamics of the fermentation were analyzed, focusing on the inoculation of LAB strains and the transition from a mung bean-based matrix to skimmed milk. The metabolomic profiles of the fermented mung bean matrices were also studied. Results: Douzhi samples obtained from representative chain stores were found to be overwhelmingly dominated by LAB. When inoculated along with the douzhi community, both LAB strains exhibited notable and substantial reductions in the pH value of the designated mung bean matrices compared to those inoculated indigenous microbiota. Specifically, Lactiplantibacillus plantarum CGMCC 1.1856 retained its population, whereas Pediococcus pentosaceus CGMCC 1.2695 exhibited a decrease in relative abundance. Using skimmed milk as a fermentation substrate instead of the mung bean matrix resulted in significant shifts in microbial communities, particularly leading to an increase in Escherichia sp. The metagenomic analyses and functional predictions illustrated that various metabolic functions were enhanced during the fermentation process due to LAB inoculation. The liquid chromatography–mass spectrometry based metabolomic analysis revealed that the inoculation of Lactiplantibacillus plantarum and Pediococcus pentosaceus in mung bean matrix did not introduce new metabolites but significantly altered the concentration and profile of existing metabolites, especially increased low molecular carbohydrates, which may enhance the nutritional potential of the fermented product. Discussion: This study examines the microbial dynamics of douzhi microbiota fermentation, emphasizing the role of lactic acid bacteria in enhancing fermentation activity and metabolite profiles. These insights contribute to improving manufacturing processes and ensuring the safety and quality of douzhi and similar fermented foods.

Abstract:Structural modification of polysaccharides using microbial fermentation is of great interest due to its specificity and environmental friendliness. The aim of this research was to compare structure, physicochemical properties and in vitro bioactivities of the Dendrobium officinale polysaccharides before and after fermentation using Lactobacillus paracasei and Lactobacillus acidophilus . The main fractions of crude polysaccharides from unfermented Dendrobium officinale (DOP-1A) and fermented Dendrobium officinale (FDOP-1A) were obtained by column chromatography purification. Compare to DOP-1A, the molecular weight of FDOP-1A was significantly reduced (from 412.566 to 25.161 kDa), smaller particle size (from 21.05 to 19.35 μm) and lower apparent viscosity. Although the microscopic surface morphologies of DOP-1A and FDOP-1A were significantly different, neither formed a triple-helix structure. Additionally, FDOP-1A contained more t-Man p and less 4-Man p, and interestingly the (1 → 4)- β -D-Glc p glycosidic bond site between the repeat units →4)-2- O -acetyl- β -D-Man p -(1 → 4)- β -D-Glc p -(1→ and →4)- β -D-Man p -(1 → 4)- β -D-Glc p -(1→ was cleaved. Moreover, FDOP-1A exhibited excellent scavenging capacity for ABTS + , DPPH, hydroxyl radicals and inhibited Nitric Oxide production by RAW 264.7 macrophages, thereby demonstrating effective antioxidant and anti-inflammatory effects. In conclusion, Lactobacillus fermentation could improve physicochemical properties and enhance biological activities of Dendrobium officinale polysaccharide, which has potential prospects for development and application in dietary supplements.

Abstract:To assess the effects of microbial fermentation on Gynostemma pentaphyllum leaves (GPL), four probiotics were used to ferment GPL (FGPL) for 7 days. At different stages of fermentation, changes in the active components and biological activities of FGPL were determined. The findings suggest that short-term fermentation with probiotics can enhance both the content and bioactivity of active components in GPL. However, prolonged fermentation may lead to a decline in these aspects. Among them, the best effect was observed with SWFU D16 fermentation for 2 days. This significantly improved the total phenolic and total flavonoid content, antioxidant capacity, and inhibitory ability against α-glucosidase activity with an increase of 28%, 114.82%, 7.42%, and 31.8%, respectively. The high-performance liquid chromatography (HPLC) analysis results also supported this trend. Untargeted metabolomics analysis revealed metabolite changes between GPL and FGPL and the key metabolites associated with these functional activities. These key metabolites are mainly organic acids, flavonoids, carbohydrates, terpenoids, and other substances. KEGG analysis demonstrated that microbial metabolism in diverse environments and carbon metabolism were the most significantly enriched pathways. Among them, 3-(3-hydroxyphenyl) propanoic acid, d-glucose, gallic acid, gluconic acid, l-lactic acid, and l-malic acid were mostly involved in the microbial metabolism of diverse environmental pathways. In contrast, D-glucose, gluconic acid, and l-malic acid were mainly related to the carbon metabolism pathway. This study revealed the positive effect of probiotic fermentation on GPL and its potential metabolism mechanism, which could provide supporting data for further research.
