Abstract:Decreased antigen presentation contributes to the ability of cancer cells to evade the immune system. We used the minimal gene regulatory network of type 1 conventional dendritic cells (cDC1) to reprogram cancer cells into professional antigen-presenting cells (tumor-APCs). Enforced expression of the transcription factors PU.1, IRF8, and BATF3 (PIB) was sufficient to induce the cDC1 phenotype in 36 cell lines derived from human and mouse hematological and solid tumors. Within 9 days of reprogramming, tumor-APCs acquired transcriptional and epigenetic programs associated with cDC1 cells. Reprogramming restored the expression of antigen presentation complexes and costimulatory molecules on the surfaces of tumor cells, allowing the presentation of endogenous tumor antigens on MHC-I and facilitating targeted killing by CD8+ T cells. Functionally, tumor-APCs engulfed and processed proteins and dead cells, secreted inflammatory cytokines, and cross-presented antigens to naïve CD8+ T cells. Human primary tumor cells could also be reprogrammed to increase their capability to present antigen and to activate patient-specific tumor-infiltrating lymphocytes. In addition to acquiring improved antigen presentation, tumor-APCs had impaired tumorigenicity in vitro and in vivo. Injection of in vitro generated melanoma-derived tumor-APCs into subcutaneous melanoma tumors delayed tumor growth and increased survival in mice. Antitumor immunity elicited by tumor-APCs was synergistic with immune checkpoint inhibitors. Our approach serves as a platform for the development of immunotherapies that endow cancer cells with the capability to process and present endogenous tumor antigens.

Abstract:Immunotherapy is revolutionizing cancer treatment, but success is limited to a fraction of patients. Tumor immunosurveillance and immunotherapy relies on presentation of tumor-associated antigens by conventional dendritic cells type 1 (cDC1). However, tumors develop mechanisms to avoid immune recognition such as downregulation of antigen presentation and exclusion of cDC1. We have previously demonstrated that enforced expression of the transcription factors PU.1, IRF8 and BATF3 (PIB) imposes the lineage conversion of fibroblasts to cDC1 by direct cell reprogramming. Here, we hypothesize that PIB reprograms cancer cells directly into functional tumor-antigen presenting cells (tumor-APCs) with enhanced immunogenicity. First, we show that enforced expression of PIB in a wide range of murine and human cancer cells from different origins is sufficient to induce surface expression of hematopoietic and DC-lineage specific markers (CD45 and Clec9a). Moreover, reprogramming restored the expression of antigen presentation complexes (MHC-I and MHC-II) and activated the expression of the co-stimulatory molecules CD40, CD80 and CD86, required for productive T cell activation. Transcriptomic analysis using mRNA-sequencing showed that PIB imposes a global cDC1 gene signature and an antigen presentation program in tumor cells as early as day 3 of reprogramming, overriding the original cancer cell program. Furthermore, Assay for Transposase-Accessible Chromatin (ATAC) sequencing analysis revealed that PIB-mediated cDC1 reprogramming elicited rapid epigenetic remodeling followed by gradual rewiring of transcriptional program and stabilization of cDC1 identity. Functionally, tumor-APCs present endogenous antigens on MHC-I, prime naïve CD8+ T and become prone to CD8+ T cell mediated killing. Tumor-APCs secrete pro-inflammatory cytokines (IL-12) and chemoattractants (CXCL10), uptake and process exogenous antigens, phagocyte dead cells, and cross-present exogenous antigens to activate naïve T-cells. In addition, reprogrammed tumor cells harboring TP53, KRAS and PTEN mutations downregulated proliferation and showed impaired tumorigenicity in vitro and in vivo. Importantly, we show that intra-tumoral injection of reprogrammed tumor-APCs elicited tumour growth control in vivo alongside increasing infiltration of CD8+ T and NK cells in B16-OVA tumors. Finally, we showed that our approach can be employed to convert primary cancer cells derived from melanoma, lung, breast, pancreatic, urothelial, and head and neck carcinomas as well as cancer associated fibroblasts. In summary, we provide evidence for the direct reprogramming of tumor cells into immunogenic cDC1-like cells, with restored antigen presentation capacity and the ability to reinstate anti-tumor immunity. Our approach elicits the immune system against cancer and counteract major tumor evasion mechanisms including tumor heterogeneity and impaired antigen presentation, laying the foundation for developing immunotherapeutic strategies based on the cellular reprogramming of human cancer cells. Abstract:Deregulation of antigen presentation is a common cancer feature that leads to immune escape. A new study reports an approach to reprogramme tumour cells in vivo, enabling them to present antigens, establish systemic immunity and induce tumour regression. Using adenoviral delivery of the transcription factors PU.1, IRF8 and BATF3, Ascic et al. reprogrammed mouse tumour cells into type 1 conventional dendritic (cDC1)-like cells. In mouse melanoma models, reprogrammed tumour cells presented antigens similar to cDC1s and induced tumour regression. The authors found that reprogrammed tumour cells caused the formation of dense lymphocyte clusters resembling tertiary lymphoid structures, enhanced the infiltration of memory and stem-like T cells, and induced polyclonal cytotoxic and memory T cell responses. Moreover, the researchers showed that using the same transcription factors could induce a cDC1-like phenotype with enhanced immunogenicity in human xenograft models. Finally, several experiments, including in spheroids, revealed that cDC1 reprogramming and cytotoxicity induction within the human tumour microenvironment was unaffected by immunosuppression.

Abstract:By overexpression of specific transcription factors, in vivo cell reprogramming has potential for treating 'cold' tumors that are often resistant to immune checkpoint blockade (ICB). A study in Science now shows that tumor cells in vivo can be reprogrammed to resemble type 1 conventional dendritic cells (cDC1s) and in so doing overcome these poorly immunogenic tumor conditions by boosting the presentation of tumor antigens. The researchers had previously found that the combination of the transcription factors PU.1, IRF8 and BATF3 (PIB) in vitro was sufficient to differentiate cancer cells in this manner, but they now demonstrate that this can be done in vivo using a gene-therapy approach in mouse melanoma models (and human spheroids). They injected replication-deficient viral vectors carrying PIB into the tumors and saw conversion of melanoma cells into cDC1-like cells with professional antigen-presenting capacity to activate anti-tumor T cell responses and limit tumor growth. In addition, the treatment resulted in immunological memory as there was durable protection against tumor rechallenge and against lung metastases. This method could avoid the costs and difficulties associated with ex vivo cell culture immunotherapies and boost or even replace ICB in resistant tumors. Abstract:Cancer immunotherapy induces a tumor antigen-specific T lymphocyte response that kills tumor cells. Dendritic cells, specifically conventional type 1 dendritic cells (cDC1s), have a crucial role in antigen presentation, making them an important cell type for immunotherapy. Tumor cell reprogramming into cDC1-like cells by the transcription factors PU.1, IRF8 and BATF2 (PIB) results in robust T cell activation in vitro. Ascic et al. demonstrate that PIB-mediated cDC1 reprogramming triggers anti-tumor immunity and hampers tumor growth in melanoma mouse models. Moreover, it remodels the tumor microenvironment by increasing memory and stem-like T cell infiltration while reducing the exhausted population. Single-cell transcriptomics reveals expansion of the CD4 + T cell population, supporting PIB-mediated immunity. Among the gene therapy delivery strategies tested, adenoviral vectors show robust tumor transduction and reprogramming in situ, triggering an efficient T cell response in lymph nodes and tumors. As a result, mice remain tumor free and resistant to metastatic spreading. Importantly, PIB overexpression in human xenografts and tumor spheroids also induces cDC1 reprogramming and immunogenicity independently of immunosuppression. This study introduces a cDC1 reprogramming-based gene therapy approach that promotes long-term, safe and efficient anti-tumor immunity. Abstract:Abundant cancer-associated fibroblasts (CAFs) in highly fibrotic breast cancer constitute an immunosuppressive barrier for T cell activity and are closely related to the failure of immune checkpoint blockade therapy (ICB). Inspired by the similar antigen-processing capacity of CAFs to professional antigen-presenting cells (APCs), a "turning foes to friends" strategy is proposed by in situ engineering immune-suppressed CAFs into immune-activated APCs for improving response rates of ICB. To achieve safe and specific CAFs engineering in vivo, a thermochromic spatiotemporal photo-controlled gene expression nanosystem was developed by self-assembly of molten eutectic mixture, chitosan and fusion plasmid. After photoactivatable gene expression, CAFs could be engineered as APCs via co-stimulatory molecule (CD86) expression, which effectively induced activation and proliferation of antigen-specific CD8 + T cells. Meanwhile, engineered CAFs could also secrete PD-L1 trap protein in situ for ICB, avoiding potential autoimmune-like disorders caused by "off-target" effects of clinically applied PD-L1 antibody. The study demonstrated that the designed nanosystem could efficiently engineer CAFs, signif-icantly enhance the percentages of CD8+ T cells (4-folds), result in about 85% tumor inhibition rate and 83.3% survival rate at 60 days in highly fibrotic breast cancer, further inducing long-term immune memory effects and effectively inhibiting lung metastasis.

A growing body of evidence indicates that intratumoral conventional type 1 dendritic cells (cDC1) are required to mount effective Tcell-mediated antitumor immunity and associate with better response to immune checkpoint blockade (ICB) therapy. However, approaches to generate cDC1s at clinical scale for immunotherapy have met limited success. We have previously demonstrated that overexpression of the transcription factors PU.1, IRF8 and BATF3 (PIB) drives cell fate reprogramming of cancer cells into functional antigen-presenting cDC1-like cells. Here, we investigated whether in vivo reprogramming of cancer cells into immunogenic antigen-presenting cDC1s can elicit systemic and durable antitumor immunity and explored the potential of in vivo delivery of PIB as a novel cancer immunotherapy modality. In vivo cDC1 reprogramming and antitumor efficacy were assessed upon subcutaneous implantation of in vitro transduced cancer cells in xenografts and ICB-resistant cDC1-dependent mouse syngeneic models. The impact of in vivo reprogramming in the TME was assessed using flow cytometry, single-cell transcriptomics, immunohistochemistry and microscopy. Tumor spheroids generated with human cancer cells in combination with cancer-associated fibroblasts or anti-inflammatory cytokines were used to investigate the impact of immunosuppressive microenvironment in cDC1 reprogramming. To select the optimal platform for in vivo delivery of PIB, we benchmarked PIB-encoding replication-deficient adenoviral (AdV), adeno-associated viral (AAV) and lentiviral (LV) vectors using mouse and human cancer cell lines and patient-derived cancer spheroids. Intratumoral delivery of PIB was performed in B16 syngeneic model to confirm in vivo efficacy induced by the gene therapy product.In vivo reprogramming to cDC1s drove complete responses (CR) in YUMM1.7 (100% CR), BRAFV600ECOX2KO (50% CR) and B2905 (80% CR) models and synergized with ICBs leading to 40% CR in B16 model, expansion of tumor-reactive T-cells and abscopal effect. Reprogrammed cancer cells recruited lymphocytes, promoted formation of tertiary lymphoid-like structures and expansion of polyclonal cytotoxic T cells. PIB-transduced human cancer cells acquired cDC1 and antigen-presentation markers in vivo with similar efficiency and kinetics as in vitro independently of immunosuppressive cells and cytokines. AdV was selected as the most efficient vector for in vivo delivery of PIB. Intra-tumoral injection of PIB-encoding AdV allowed in situ phenotypic reprogramming, enhanced infiltration of cytotoxic T cells and drove 50% CR, abscopal effect, long-term immune memory and protection against local and metastatic re-challenge. These findings provide preclinical proof-of-concept for an off-the-shelf, personalized cancer immunotherapy based on in situ cDC1 reprogramming, paving the way for first-in-human trials. Abstract:Advances in understanding how cancer cells interact with the immune system allowed the development of immunotherapeutic strategies, harnessing patients' immune system to fight cancer. Dendritic cell-based vaccines are being explored to reactivate anti-tumor adaptive immunity. Immune checkpoint inhibitors and chimeric antigen receptor T-cells (CAR T) were however the main approaches that catapulted the therapeutic success of immunotherapy. Despite their success across a broad range of human cancers, many challenges remain for basic understanding and clinical progress as only a minority of patients benefit from immunotherapy. In addition, cellular immunotherapies face important limitations imposed by the availability and quality of immune cells isolated from donors. Cell fate reprogramming is offering interesting alternatives to meet these challenges. Induced pluripotent stem cell (iPSC) technology not only enables studying immune cell specification but also serves as a platform for the differentiation of a myriad of clinically useful immune cells including T-cells, NK cells, or monocytes at scale. Moreover, the utilization of iPSCs allows introduction of genetic modifications and generation of T/NK cells with enhanced anti-tumor properties. Immune cells, such as macrophages and dendritic cells, can also be generated by direct cellular reprogramming employing lineage-specific master regulators bypassing the pluripotent stage. Thus, the cellular reprogramming toolbox is now providing the means to address the potential of patient-tailored immune cell types for cancer immunotherapy. In parallel, development of viral vectors for gene delivery has opened the door for in vivo reprogramming in regenerative medicine, an elegant strategy circumventing the current limitations of in vitro cell manipulation. An analogous paradigm has been recently developed in cancer immunotherapy by the generation of CAR T-cells in vivo. These new ideas on endogenous reprogramming, cross-fertilized from the fields of regenerative medicine and gene therapy, are opening exciting avenues for direct modulation of immune or tumor cells in situ, widening our strategies to remove cancer immunotherapy roadblocks. Here, we review current strategies for cancer immunotherapy, summarize technologies for generation of immune cells by cell fate reprogramming as well as highlight the future potential of inducing these unique cell identities in vivo, providing new and exciting tools for the fast-paced field of cancer immunotherapy.

This strategy might open avenues to enhance cancer cell recognition and elimination by the immune system. However, currently existing in-vitro and in-vivo testing platforms do not qualify to reproduce all complex cell interactions essential for the approbation of this hypothesis. Here, we report the development of the InSphero 3D InSightTM Oncology Platform for in-vitro assessment of efficacy and immune-stimulatory effects of this novel cancer immunotherapy approach. The feasibility of 3D spheroid formation for several GFP-expressing tumor cell lines was evaluated by varying seeding conditions in AKURA 96 well plate. We have measured the growth (ATP content) and GFP signal overtime and analyzed the morphology of the spheroids by IHC. With this, we have established spheroid models of T98G (glioblastoma), PK59 (pancreatic cancer), and A375 (melanoma) cell lines that are growing and viable for at least 10 days. In parallel, using 2D cultures, we have identified the optimal multiplicity of infection of a lentiviral vector encoding for PIB and mCherry to enable high transduction (mCherry+ cells), reprogramming efficiency (mCherry+CD45+HLA-DR+ cells), and cell viability, quantified by flow cytometry and IHC. Then, we have demonstrated that cDC1 reprogramming progresses in the context of 3D cancer spheroids and tumor cells acquire expression of CD45+ and HLA-DR+ cells using IHC and confocal microscopy analysis. We developed an algorithm enabling automated analysis of confocal images and quantification of cDC1 reprogramming efficiency from individual image stacks calculated as a ratio of mCherry+, CD45+ and HLA-DR+ cells versus the number of DAPI+ nuclei. Using the new algorithm we have evaluated the reprogramming efficacy of the different virus dosages in all three types of 3D tumor spheroids. Lastly, we have cocultured tumor spheroids transduced with PIB with naïve or activated HLA-matched PBMCs and evaluated cytokine secretion as a readout of immune cell activation. We observed that reprogramming induces activation of T cells and correlated it to the number of reprogrammed cells in the tumor spheroid, evaluated by the HC imaging. In summary, we developed the InSphero 3D InSightTM Oncology Platform that allowed us to demonstrate the effects of direct reprogramming of tumor cells into immunogenic dendritic cells. Abstract:Dendritic cells (DCs) are the main professional antigen-presenting cells for induction of T-cell adaptive responses. Cancer cells express tumor antigens, including neoantigens generated by nonsynonymous mutations, but are poor for antigen presentation and for providing costimulatory signals for T-cell priming. Mounting evidence suggests that antigen transfer to DCs and their surrogate presentation on major histocompatibility complex class I and II molecules together with costimulatory signals is paramount for induction of viral and cancer immunity. Of the great diversity of DCs, BATF3/IRF8-dependent conventional DCs type 1 (cDC1) excel at cross-presentation of tumor cell-associated antigens. Location of cDC1s in the tumor correlates with improved infiltration by CD8+ T cells and tumor-specific T-cell immunity. Indeed, cDC1s are crucial for antitumor efficacy using checkpoint inhibitors and anti-CD137 agonist monoclonal antibodies in mouse models. Enhancement and exploitation of T-cell cross-priming by cDC1s offer opportunities for improved cancer immunotherapy, including in vivo targeting of tumor antigens to internalizing receptors on cDC1s and strategies to increase their numbers, activation and priming capacity within tumors and tumor-draining lymph nodes.

Abstract:Inadequate antigen-specific T cells activation hampers immunotherapy due to complex antigen presentation. In addition, therapeutic in vivo T cell expansion is constrained by slow expansion rates and limited functionality. Herein, we introduce a model fusion protein termed antigen-presenting cell-mimic fusion protein (APC-mimic), designed to greatly mimicking the natural antigen presentation pattern of antigen-presenting cells and directly expand T cells both in vitro and in vivo. The APC-mimic comprises the cognate peptide-human leukocyte antigen (pHLA) complex and the co-stimulatory marker CD80, which are natural ligands on APCs. Following a single stimulation, APC-mimic leads to an approximately 400-fold increase in the polyclonal expansion of antigen-specific T cells compared with the untreated group in vitro without the requirement for specialized antigen-presenting cells. Through the combination of single-cell TCR sequencing (scTCR-seq) and single-cell RNA sequencing (scRNA-seq), we identify an approximately 600-fold monoclonal expansion clonotype among these polyclonal clonotypes. It also exhibits suitability for in vivo applications confirmed in the OT-1 mouse model. Furthermore, T cells expanded by APC-mimic effectively inhibits tumor growth in adoptive cell transfer (ACT) murine models. These findings pave the way for the versatile APC-mimic platform for personalized therapeutics, enabling direct expansion of polyfunctional antigen-specific T cell subsets in vitro and in vivo.

Abstract:Abstract Dendritic cells (DCs) are antigen-presenting myeloid cells that regulate T cell activation, trafficking and function. Monocyte-derived DCs pulsed with tumor antigens have been tested extensively for therapeutic vaccination in cancer, with mixed clinical results. Here, we present a cell-therapy platform based on mouse or human DC progenitors (DCPs) engineered to produce two immunostimulatory cytokines, IL-12 and FLT3L. Cytokine-armed DCPs differentiated into conventional type-I DCs (cDC1) and suppressed tumor growth, including melanoma and autochthonous liver models, without the need for antigen loading or myeloablative host conditioning. Tumor response involved synergy between IL-12 and FLT3L and was associated with natural killer and T cell infiltration and activation, M1-like macrophage programming and ischemic tumor necrosis. Antitumor immunity was dependent on endogenous cDC1 expansion and interferon-γ signaling but did not require CD8 + T cell cytotoxicity. Cytokine-armed DCPs synergized effectively with anti-GD2 chimeric-antigen receptor (CAR) T cells in eradicating intracranial gliomas in mice, illustrating their potential in combination therapies.

Abstract:Antigen-presenting cells expand antigen-specific T cells ex vivo and in vivo for tumor immunotherapy, but are time-consuming to generate and, as live cells, raise biosafety concerns. An alternative is found in cell-free artificial antigen-presenting cells (aAPC), but these only present two or three kinds of immune molecules. Here, we describe a multipotent artificial antigen-presenting cell (MaAPC) that delivered 11 kinds of immune moleclues. This MaAPC simulated natural APCs through the concurent coupling of target antigens (H-2Kb/TRP2180-188-Ig dimers and H-2Db/gp10025-33-Ig dimers), costimulatory molecules (anti-CD28, anti-4-1BB, and anti-CD2), and "self-marker" CD47-Fc onto surface-modified polylactic-co-glycolic acid microparticles (PLGA-MP). These PLGA-MPs also encapsulated cytokines (IL2 and IL15), a chemokine (CCL21), and checkpoint inhibitors (anti-CTLA-4 and anti-PD-1). Culture of MaAPCs with naïve T cells for 1 week elevated the frequencies of TRP2180-188-specific and gp10025-33-specific CTLs to 51.0% and 43.3%, respectively, with enhanced cytotoxicity. Three infusions of MaAPCs inhibited subcutaneous melanoma growth in a mouse model and expanded TRP2180-188 and gp10025-33-specific CTLs 59-86-fold in peripheral blood, 76-77-fold in spleen, and 205-212-fold in tumor tissue, in an antigen-specific manner. Compared with conventional aAPCs carrying two or three immune molecules, the 11-signal MaAPCs exerted greater impact on T cells, including activation, proliferation, cytotoxicity, differentiation to memory CTLs or regulatory T cells and cytokines profiles, without detected side effects. Such MaAPCs could be used to individualize tumor immunotherapy.

Abstract:The conventional type 1 dendritic cells (cDC1) play a pivotal role in protective immunity against pathogens and cancer. However, their low frequency in the blood and tissues limits their use in immune therapy. We have recently described a method to vaccinate against neoantigens that are induced in tumor cells by targeted delivery of a TAP siRNA to dendritic cells using a TLR9 binding CpG oligonucleotide. Since TLR9 is also expressed in immune suppressive myeloid populations TLR9 targeting could reduce the effectiveness of this approach. Here, we describe a modular multivalent antibody platform to target the TAP siRNA to resident Clec9a expressing cDC1 and show that it leads to selective and sustained TAP downregulation in cDC1 and inhibits tumor growth in mice more effectively than CpG targeted siRNA. To induce DC maturation an agonistic CD40 antibody was administered to the siRNA treated mice. To obviate the need for a second drug formulation and reduce the risk of toxicity, we exploited the multivalent nature of this targeting platform to co-deliver the TAP siRNA and a DC maturation agent, a CpG containing oligonucleotide, to cDC1 in vivo and show that it was more effective than Clec9a targeting of TAP siRNA in combination with CD40 antibody. This study describes a way to manipulate the function of cDC1 cells in vivo using a broadly applicable antibody-based targeting platform to deliver multiple biological agents to specific cells in vivo to potentiate (immune) therapy and to probe the biology of specific cell types in their natural settings.

Abstract:Therapeutic use of a peptide corresponding to the aminoacid sequence of the second extracellular loop of human M2 muscarinic acetylcholine receptor (M2 mAChR peptide) was studied. Expression and biological activity of M2 mAChR in association with circulating M2 mAChR-related antibodies in cardiac tissue from chagasic mice were evaluated. Mice infected or not with trypomastigotes Tulahuen strain either treated or not treated with M2 mAChR peptide were sacrificed at 8-9 weeks post-infection. Morphological, binding and contractility studies were performed on all animal groups. Hearts from infected mice showed a mAChR-related dysfunction, with a decrease in heart contractility, impaired response to exogenous mAChR agonist (carbachol) and a significant reduction of mAChR binding sites. Treating infected mice with M2 mAChR peptide reversed those effects. Moreover, autoantibodies from infected mice recognized the M2 mAChR peptide. In addition, serum from infected mice and the corresponding affinity purified IgG was capable of interacting with cardiac mAChR, reducing the number of binding sites and inhibiting the contractile response to exogenous agonist. In conclusion, (1) the development of alterations in mAChR related to cardiac dysfunction, may be associated with the presence of circulating antibodies against these receptors and (2) the chronic treatment with M2 mAChR peptide prevented infected mice heart dysfunction. The mechanism could be explained by the ability of the M2 mAChR peptide to inhibit the chronic interaction of autoantibodies specific to mAChR. The implication of M2 mAChR peptide treatment in the host's immune response is discussed.

Abstract:Abstract Dendritic cells (DCs) are professional antigen-presenting cells that are integral to the induction of primary, antigen-specific T cell responses. In the cancer setting, DCs mediate cross-priming of tumor-reactive T cells by presenting tumor antigens acquired from viable or dead cancer cells. Due to their unique functional properties, DCs have been utilized as both vectors and targets for immunological intervention in numerous diseases and are optimal candidates for vaccination protocols in cancer. In addition to their antigen presentation function(s), recent evidence suggests that DCs may also perform an innate immune effector function, with human DCs reported to mediate direct tumoricidal activity in vitro. However, the mechanism(s) by which DCs directly kill tumor cells remain unclear. The goal of this study is to further characterize the mechanism(s) associated with murine DC tumoricidal function and to determine whether and how this function may be enhanced to promote anti-tumor immune responses that translate into therapeutic effectiveness. One way we sought to enhance this DC effector function was through the genetic engineering of DCs themselves. After transduction with mIL-12 and/or mIL-18 cDNA using recombinant adenoviral vectors, DCs exhibited significantly elevated tumor killing activity. This was mediated, at least in part, by TNF ligand-receptor complexes, as demonstrated by antibody blocking assays. When injected in situ, these engineered DCs exhibited prolonged survival, in association with enhanced levels of tumor apoptosis proximal to imaged DCs and our capacity to image DC that had engulfed tumor apoptotic bodies. We also observed notable therapeutic benefits upon intratumoral delivery of these DCs in concert with an expanded in vivo repertoire of anti-tumor CD8+ T cells. In addition to DC modification, we also evaluated treatments applied to tumor cells that resulted in enhanced sensitivity to (control) DC-mediated killing. Specifically, we found that pretreatment of A20 lymphoma cells with a nitric oxide (NO) donor compound, PAPA-NO, markedly increased the sensitivity of tumor cells to consequent apoptosis mediated by DCs. This appeared to provide DCs with a preferred source of tumor antigens, with which, they were capable of activating specific T cells via a cross-presentation pathway. We have also discovered that multiple TNF family ligands participated in DC-mediated tumoricidal function and that tumor cell-expressed survivin may represent a critical downstream factor regulating the apoptotic sensitivity of tumor cells to DC-mediated apoptosis. When taken together, these studies provide novel details regarding mechanisms involved in DC anti-tumor effector function, and suggest two DC-based, combinational cancer therapies that target the effective cross-priming of therapeutic T cells. The results presented in this dissertation support an efficient model in which DCs may not only serve as the gatherers and presenters of antigens, but also the hunters as well, with tumoricidal activity mediated via TNF family ligands.
