Abstract:Our current study examines the significant impact of aniline on the fish feed freshwater benthic annelid, Tubifex tubifex , including mortality, changes in total protein concentration and antioxidant enzyme activity (catalase, superoxide dismutase, glutathione-s transferase and glutathione peroxidase), as well as the generation of oxidative stress (by assessing malondialdehyde concentration). The 96-hour LC 50 value of aniline is 276.940 mg/L. The General Unified Threshold Model of Survival (GUTS) was used to validate the survival rate pattern of the test organism in response to aniline at the acute level, with the GUTS-SD model better correctly predicting the expected course of population death. During long-term bioassay antioxidant enzymes activities (superoxide dismutase, glutathione-s transferase and glutathione peroxidase) and oxidative stress biomarkers concentration (malondialdehyde concentration) is substantially enhanced while protein concentration and Catalase activity are markedly reduced during the bioassay. The stress and detrimental effects of aniline on Tubifex tubifex were examined in the current study, taking into account all biomarkers, by utilising the Integrated Biomarker Response index. The results show that exposure to toxicants like aniline has a detrimental effect on Tubifex tubifex 's survival rate and general health.

A comparison of the effects of first premolar extractions on third molar angulation.

Abstract:The effect of third molars on the stability of orthodontic treatment has been studied extensively. Yet the effect of orthodontic treatment, particularly premolar extractions, on third molars has not been substantially studied. The purpose of this investigation was to compare the changes in third molar angulation in patients treated with and without extractions. Records of 45 Class I, nonextraction and 33 Class I, first premolar extraction patients were examined. The pretreatment and posttreatment pantographs were digitized, and the angles between the third molar long axes and the occlusal plane were measured. Changes in third molar angulation from pretreatment to posttreatment for the two groups were compared for statistical differences using a Student's t-test. Statistical analysis revealed there were no significant differences in the change in third molar angulation in either group. On average, the maxillary and mandibular third molars showed an improvement in angulation relative to the occlusal plane. The results suggest that factors other than first premolar extractions may influence third molar angulation.

Abstract:Benzo[a]pyrene (B[a]P) is a source of profound global concern due to its intrinsic toxicity and potential to cause deleterious impacts on aquatic organisms. This study aims to assess B[a]P-induced genotoxic perturbations in fish, Channa punctatus . The experiment was designed for a 96-hour acute exposure period, involving four groups i.e., I- control, II- solvent control, III- 1/20th of 96-h LC 50, and IV- 1/10th of 96-h LC 50 of B[a]P, respectively, with four fish in each group. Following the completion of the 96-hour exposure period, micronuclei (MN), nuclear abnormalities (NAs), and DNA tail% were evaluated in the erythrocytes of test fish. Significant ( p < 0.05) increments in MN, NAs, and DNA tail% were recorded in both the B[a]P exposed groups after a 96-h exposure period. The study conclusively confirms the DNA damage induced by B[a]P and underscores its profound significance in the conservation and effective mitigation of fish diversity in delicate aquatic ecosystems.

Abstract:Aim: This study aimed to investigate the oxidative stress, genotoxicity and histopathology after sub-lethal exposure of chromium (Cr6+) and copper (Cu+2) to common food fish Channa punctatus. Metal exposure to fishes of River Ganga is a very common phenomenon due to their ubiquitous nature and through discharge of domestic and industrial waste into the river water. Methodology: An in-vitro experiment was performed to study ill effects of metallic stressors on fish. Fishes were divided into seven groups. Group I served as control, Group II, III, IV as 1/20th ,1/10th and 1/5th of 96 hr-LC50 of Cr6+. Group V, VI and VII were similarly treated with Cu at 1/20th, 1/10th and 1/5th of 96 hr LC50 of Cu+2, respectively. All the treatments were done for 15, 30 and 45 days of exposure period. Genotoxicity was evaluated by micronuclei (MN) induction. Enzymatic and non-enzymatic oxidative stress markers, including reactive oxygen species (ROS), superoxide dismutase (SOD), catalase (CAT), glutathione (GSH), glutathione reductase (GR), and glutathione peroxidase (GPx) were evaluated. Histopathological studies of liver and kidney tissue were also performed. Results: Stress biomarkers were more profound in fishes of chromium treated groups in comparison to copper. Particularly, SOD and CAT showed a significant hike in the activity levels. ROS levels in blood cells increased significantly (p < 0.05) in all the treated groups (Group II, III, and IV) in a dose dependent manner as compared with control (Group I). There was significant induction in micronuclei (MN) frequency in all the treated groups for both the metals. The highest frequency of MN induction was recorded in Group IV after 96 hr of the exposure period.Significant histological alterations were observed in liver and kidney of treated fish, changes were more pronounced in chromium treated fish than copper. Interpretation: Acute exposure of chromium and copper induces ROS and generates oxidative stress mediated genotoxicity and histopathological perturbations in the prime organs of fish.Cr and Cu induces the changes in the production and accumulation of enzymatic antioxidant system for the regulation and scavenging of ROS induced by Cr and Cu. Abstract:background Dog-ears often lead to lengthening of an excision, and it is desirable to avoid them. objective Facial skin, including the subepidermal connective tissue, is flexible and can be used advantageously to minimize dog-ears using a novel buried suture technique. methods After removing a round lesion, the first horizontal square buried suture (HSBS) was deeply placed parallel to the longitudinal direction of the defect beneath superficial fascia. After the first HSBS was tied, the defect became fusiform but was still largely open. The second HSBS was also placed parallel to the longitudinal direction of the defect but in more superficial fascia and using smaller horizontal buried loops than those of the first deep suture. After the second HSBS in the middle of the dermis was tied, the wound was almost closed without dog-ears. Consequently, few skin sutures were required to finish the operation. conclusions Using this technique, a small circular or oval defect on the face up to 1 cm in diameter can be closed without any additional excision of the skin and without creating dog-ears.
An in vivo analysis of Cr6+ induced biochemical, genotoxicological and transcriptional profiling of genes related to oxidative stress, DNA damage and apoptosis in liver of fish, Channa punctatus (Bloch, 1793)

Abstract:Present study was designed to assess the hexavalent chromium (Cr6+) mediated oxidative stress that induces DNA damage and apoptosis in adult fish, Channa punctatus (35 ± 3.0 g; 14.5 ± 1.0 cm; Actinopterygii). Fishes were maintained in three groups for 15, 30 and 45 d of exposure periods. They were treated with 5% (Group T1) and 10% (Group T2) of 96 h-LC50 of chromium trioxide (Cr6+). Controls were run for the similar duration. A significant (p < 0.05) increment in the activities of antioxidant enzymes, SOD and CAT in liver tissues of the exposed fish evinces the persistence of oxidative stress. A significant (p < 0.05) increase in induction of micronuclei (MN) coupled with transcriptional responses of target genes related to antioxidant enzymes, DNA damage and apoptosis (sod, cat, gsr, nox-1, p53, bax, bcl-2, apaf-1 and casp3a) establishes the impact of oxidative stress due to in vivo, Cr6+ accumulation in liver as compared to control (0 mg/L), in a dose and exposure-dependent manner. Initially, the increased level of reactive oxygen species (ROS) in liver coincided with that of enhanced mRNA expression of antioxidant enzymes, sod, cat, gsr and nox-1 but, later, the overproduction of ROS, after 45 d of exposure of Cr6+, resulted in a significant (p < 0.05) up-regulation of p53. Our findings also unveil that the up-regulation of bax, apaf-1 and casp3a and down-regulation of bcl-2 are associated with Cr6+-induced oxidative stress mediated-apoptosis in liver of test fish. Aforesaid molecular markers can, thus, be efficiently utilized for bio-monitoring of aquatic regimes and conservation of fish biodiversity.
The Effects of Acute Exposure to Ammonia on Oxidative Stress, Hematological Parameters, Flesh Quality, and Gill Morphological Changes of the Large Yellow Croaker (Larimichthys crocea)

Abstract:Ammonia is considered to be the major chemical pollutant causing fish poisoning in aquaculture. This research aimed to evaluate the impact of acute ammonia exposure on the large yellow croaker's meat quality, gill morphology, liver oxidative stress, and hematological parameters. The fish were exposed to total ammonia nitrogen concentrations of 0, 2.96, 5.92, and 8.87 mg/L for 48 h, respectively. The findings demonstrated that all ammonia-exposed fish had higher liver lactate dehydrogenase and glutamic oxalate transaminase activities. The glucose, blood urea nitrogen, and creatinine levels in 8.87 mg/L total ammonia nitrogen (TAN) were higher than other samples. The total protein, albumin, and triglyceride levels in serum decreased significantly in ammonia-exposed samples. After 48 h of ammonia exposure, superoxide dismutase activities showed a 76.1%, 118.0%, and 156.8% increase when fish were exposed to 2.96, 5.92, and 8.87 mg/L TAN, respectively. Catalase activities and glutathione contents were considerably higher (p < 0.05) in all ammonia-treated samples compared to 0 mg/L TAN. The ammonia-treated gill lamellae become thicker, shorter, and curved. Additionally, the ammonia exposure resulted in the accumulation of free amino acids and the loss of nucleotides. The inosine monophosphate and adenosine monophosphate contents in the flesh were decreased after 12 h of exposure to 2.96, 5.92, and 8.87 mg/L ammonia compared to the control group. Overall, large yellow croakers exposed to ammonia for 6 h presented not only changes in serum composition but also oxidative stress, liver and gill tissue damage and flesh quality deterioration.

Assessment of aquatic experimental versus predicted and extrapolated chronic toxicity data of four structural analogues

Abstract:The present study was developed to assess the chronic toxicity predictions and extrapolations for a set of chlorinated anilines (aniline (AN), 4-chloroaniline (CA), 3,5-dichloroaniline (DCA) and 2,3,4-trichloroaniline (TCA)). Daphnia magna 21 d chronic experimental data was compared to the chronic toxicity predictions made by the US EPA ECOSAR QSAR tools and to acute-to-chronic extrapolations. Additionally, Species Sensitivity Distributions (SSDs) were constructed to assess the chronic toxicity variability among different species and to investigate the acute versus chronic toxicity in a multi-species context. Since chlorinated anilines are structural analogues with a designated polar narcotic mode of action, similar toxicity responses were assumed. However, rather large interchemical and interspecies differences in toxicity were observed. Compared to the other three test compounds, TCA exposure had a significantly larger impact on growth and reproduction of D. magna. Furthermore, this study illustrated that QSARs or a fixed ACR are not able to account for these interchemical and interspecies differences. Consequently, ECOSAR was found to be inadequate to predict the chronic toxicity of the anilines and the use of a fixed ACR (of 10) led to under of certain species. The experimental ACRs determined in D. magna were substantially different among the four aromatic amines (ACR of 32 for AN, 16.9 for CA, 5.7 for DCA and 60.8 for TCA). Furthermore, the SSDs illustrated that Danio rerio was rather insensitive to AN in comparison to another fish species, Phimphales promelas. It was therefore suggested that available toxicity data should be used in an integrative multi-species way, rather than using individual-based toxicity extrapolations. In this way, a relevant overview of the differences in species sensitivity is given, which in turn can serve as the basis for acute to chronic extrapolations.

Abstract:Nigrosine Black (Acid Black-2) is popularly used in tanning industries and a huge amount of this dye comes out with effluent of tanning industries, this dye reached near water bodies, and what impact produced on their aquatic life not reveal yet, Hence a study was plan to find out the impact of Nigrosine black dye on the renal cell of freshwater fish Channa punctata. Sub-acute and chronic exposure to Nigrocine Black produced significant renal cellular degeneration in fish C. punctata. The 1/20th LC50 (96h) of the Nigrocine Black produced degenerating changes in the kidney tissues of fish. Renal corpuscles were revealing swollen and contained some degenerated RBC and thmbrocytes, which appeared to be cellular debris in Bowman space, due to the rapid destruction of the nuclei of erythrocytes. The liberated hemoglobin and other cytoplasmic materials were also observed, while 1/20th of LC50 (96hr) of Nigrosine Black produced renal degeneration in Bowman’s capsule and swelling in the glomerulus in the intoxicated fish along with renal tubules atrophy, degeneration as compared to the control group of fish. The low dose of Nigrosine Black Toxicity in fish produced nephropathy, and nephrosis. While the high dose of Nigrosine black dye produces Glomerulonephritis in experimental fish. The kidney cells of experimental fish are damaged by both doses of Nigrosine Black. The physico-chemical property of aquarium water was kept constant during whole duration of experiment.
Distinctive fingerprints of genotoxicity induced by As, Cr, Cd, and Ni in a freshwater fish

Abstract:Genotoxicity of three toxic elements (chromium, cadmium, nickel) and a metalloid (arsenic) has been studied in a freshwater fish, Channa punctatus using micronuclei (MN) test, comet assay, and erythrocyte nuclear alterations (ENAs) as fingerprints of genotoxicity. These tests yielded different results suggesting involvement of different mechanisms for their genotoxicity. While highest frequency of blebbed nuclei was observed in chromium-treated fish (6.5 ± 0.76), lowest was observed in cadmium-treated fish (4.0 ± 1.0). Maximum number of notched nuclei was recorded in arsenic-treated fish (5.5 ± 1.15) whereas highest numbers of lobed nuclei were found in cadmium-treated fish (4.5 ± 0.13). These differences might be attributed to selective bioaccumulation and chemodynamics of each element. Other parameters used to determine genotoxicity viz.: lipid peroxidation and DNA damage also suggested different mechanisms of their genotoxicity. It is suggested that an integrative approach, using a battery of tests for determining genotoxicity, should be made while making environmental health risk assessment and ecotoxicological studies of these toxic elements.
Environmentally relevant concentration of copper elated hematological impairment, branchiotoxicity, myotoxicity, nephrotoxicity and antioxidants imbalance in fish Channa punctatus

Abstract:Copper is a mineral that organisms need to stay healthy on one hand and on the other hand prolonged overexposure may result into various dangerous implications. The present study was outlined to assess various deleterious effects being caused to the edible fish C. punctatus exposed to an environmentally relevant concentration of copper (ERCC). For this purpose, well-acclimatized fish were classified into four groups. Group I was maintained as control while groups II, III, and IV were exposed to the ERCC (0.85 mg/L), 10% increase in ERCC (0.935 mg/L) and 20% increase in ERCC (1.02 mg/L), respectively, for 15, 30, 45 and 60d. A significant (p <0.05) reduction in Hb% and RBC count while escalation in WBC counts was documented after all exposure periods and the maximum change was recorded after the longest exposure period of 60 days, respectively. Enzymatic antioxidants viz., superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR) and lipid peroxidation (LPO) levels were significantly (p <0.05) raised and non-enzymatic antioxidant, i.e., glutathione (GSH) activity was reduced in all exposed groups in a dose-dependent manner. Severe histopathological modifications were observed in the treated fish’s gills, muscles, and kidneys. The outcomes of the present investigation substantiate that ERCC induces toxicity in fish at various levels and warn about the possible repercussions of increased ERCC in the near future as the concentration of copper continuously rises with the ever-increasing pollution.
Genotoxic impact of agricultural insecticides as contaminants of river Teesta on the resident fish Pethia Conchonius

Abstract:Fish, being highly sensitive to changes in the physico-chemical parameters of water, are good indicators of contamination. Teesta, a prominent northern West Bengal River system, is increasingly contaminated due to anthropogenic activities. This study aims to determine agricultural pesticide contamination and its genotoxic impact on the resident fish, Pethia conchonius , as an experimental organism. Sample water analysis from three riverine sites I, II & III, showed the presence of the insecticides imidacloprid (IMI), chlorpyrifos (CPF), bifenethrin (BF), cypermethrin (CP), difenthiuron, acetamiprid (AC) in the sites II and III only with adjoining agricultural lands. Comet assay revealed a significantly lower % Head DNA (~ 1.2 times), higher %Tail DNA (~ 16 times), and %Tail length (~ 3.1 times) in the gills of Pethia conchonius from sites II and III. About 4 and 10 times increase of micronuclei and other nuclear abnormalities were also noted in the erythrocytes of the fish from sites II and III than I, which was not contaminated. The antioxidant enzymes SOD, CAT, and GST activity and MDA levels were significantly higher ( p < 0.05) in the liver samples from sites II and III, while AChE activity was significantly decreased ( p < 0.001) in the brain tissues. Moreover, the sod , cat , and gpx expression in the hepatic cells were significantly upregulated compared to the β actin mRNA indicating increased oxidative stress. Increased genomic damage, antioxidant enzyme activity, higher MDA levels, decreased AChE activity in the brain, and the upregulation of hepatic genes strongly suggested the genotoxic effects of the detected insecticides in combination with other contaminants.

Abstract:Coastal ecosystems are vital for maintaining the biodiversity and human livelihoods, but they are increasingly subjected to anthropogenic pressures, including pollution from various sources. Present work intends to assess the possible threats in coastal ecosystem as well as coastal fish species, in particular, through haematological parameters caused due to exposure of environmental contaminants like polycyclic aromatic hydrocarbons (PAHs), potentially toxic metals (PTMs), etc. This study analysed the haematological parameters and probable toxicity levels in two important coastal fish species, viz., Mystus sp. and Mugil sp. widely available in Digha coastal belt. Different haematological parameters, such as WBCs (White Blood Cells), Lym (Lymphocytes), Gran (Granulocytes), Mid (Monocytes), RBCs (Red Blood Cells), HCT (Haematocrit) value, MCV (Mean Corpuscular Volume), MCH (Mean Corpuscular Haemoglobin), MCHC (Mean Corpuscular Haemoglobin Concentration), RDW- CV (Red Cells Distribution Width-Co-efficient of Variation), RDW- SD (Red Cells Distribution Width-Standard Deviation), PLT (Total Platelet Count), MPV (Mean Platelet Volume), PDW- SD (Platelet Distribution Width-Standard Deviation), PDW- CV (Platelet Distribution Width-Co-efficient of Variation), PCT (Plateletcrit), PLCR (Platelet Large Cell Ratio), PLCC (Platelet Large Cell Count) and many others were measured directly through Erba H360 Haematology Analyser, simultaneously air dried blood smear was stained by Haematoxylin-Eosin(H-E) and Giemsa stain for assessing morphometric alterations of RBCs, WBCs, platelets as well as to determine the differential counts of WBCs by observing through Leica DM2000 microscope. Evidence of several abnormalities in the erythrocyte's nucleus (ENAs) and the abundance of abnormal celled erythrocytes (ECAs), carcinoma (lymphoproliferative disorder, polycythaemia vera, Hodgkin lymphoma and non-Hodgkin lymphoma), elevation of WBCs content, Lym %(Lymphocyte percentage), Eo(Eosinophils), monocytes, HCT and gross depletion of Ne(Neutrophils), basophils, and PLCR levels indicated a sign of major impact of contamination to two intoxicated fishes which may also affect the human being through food chain and may result into leukaemia in mammalian species, finally. However, comprehensive evaluation of the long-term impacts of the contaminants like PAHs and/or PTMs, etc., on fish populations, human health risk and coastal ecosystem is required to be addressed.

Abstract:Studies on heavy metal induced toxicity have been conducted in many water bodies across the globe and such effects have been evaluated in various fish species. The present study was designed to determine the load of some heavy metals in select sites in Southern Assam, India, along with estimating their concentration in tissues of Channa punctatus Bloch. inhabiting those niches. The effect of heavy metals in oxystress generation, genotoxicity and subsequent immune response in fish was also evaluated. In all of these sites, the concentration of Hg, Cd, Pb and Cr were above the permissible ranges while their concentrations were several folds higher in the piscine tissues due to bioaccumulation and possible biomagnification. Kidney showed the highest metal pollution index followed by liver and gills. Generation of ROS was significantly elevated and that in turn triggered oxystress, as is evident from enhanced lipid peroxidation, protein carbonylation and respiratory burst activity. These were in association with the compromised antioxidant enzyme levels with concomitant damage to DNA as evident from Comet parameters. The innate immune potential was significantly impaired as evident from the compromised cell adhesion, phagocytosis, intracellular killing activity in head kidney macrophages (HKM) along with decreased release of nitric oxide (NO) and myeloperoxidase (MPO). Immunosuppression was further validated at protein levels where compromised release of cytokines viz. TNF-α, IL-1β, IL-6, IL-10 and IL-12 and cell signaling molecules iNOS and NF-κβ were noted. Thus the present study indicates genotoxicity along with a compromise in immune status of Channa punctatus Bloch. living in a habitat laden with heavy metals.

Abstract:The fish ecology of Deepor Beel is being threatened by the continuous flow of effluents from the solid wastes of Municipal garbage dump. The present study has been aimed to investigate the genotoxic effect of the solid wastes on Channa punctata of Deepor Beel using micronucleus assay. Fishes were collected from garbage dump affected and unaffected area of Deepor Beel. The micronuclei assay demonstrated significantly higher frequency of micronuclei and other nuclear abnormalities in Channa punctata collected from affected site as compared to unaffected reference site. This study depicted the usefulness of micronucleus assay as an adequate approach for ecotoxicological monitoring of aquatic environment and its biodiversity.

Abstract:In the present study, pattern of changes in blood physiological parameters, structuralchanges in gills, liver and kidnney as well as gene expression patterns for ornithine-ureacycle (OUC) were studied, under hyper-ammonia stress (25 mM NH4Cl) for 1, 3, 6 and 9 hand 24 h in normal water condition after 9 h exposure (9h+N), in Clarias magur (Hamilton,1822). Analysis showed increase in blood ammonia upto 3 h, with decreasing trend (upto2 folds reduction) subsequently. It was accompanied by simultaneous increase in urealevels upto 6 folds at 9 h exposure. However, there were no significant changes in bloodparameters, except for serum protein, serum glutamic pyruvic transaminase (SGPT) andlactate dehydrogenase (LDH) levels after 1 h exposure. Histological analysis of gills revealedhighest (88%) percentage of damaged gills with congested blood capillaries, treated for 9 h,but declined by 65% at 9h+N. Histological alterations in liver and kidney were prominentonly at 9 h exposure, with nuclear hypertrophy and necrosis of hepatic cells in liver tissuesand distension as well as degeneration of Bowman’s capsule in kidneys. Significantup-regulation of cpsIII and otc genes at 9 h and 9h+N, points to activation of OUC in liver at6 h exposure and unique physiological characteristics of turning towards ureotelism fromammoniotelism, after short term exposure to high ammonia level. Evaluation of arsenic induced genotoxicity and its impact on life processes of Daphnia magna

Abstract:Heavy metals like arsenic is ubiquitously present in the environment. Geologic and anthropogenic activities are the root cause behind high concentration of arsenic in natural water bodies demanding strict monitoring of water quality prior to human consumption and utilization. In the present study, we have employed Daphnia magna for studying the biological effects of environmentally relevant high concentration of arsenic in water. In acute toxicity study, the LC50 value for 24hr exposure was found to be 2.504 mg/L, which gradually decreased with increase in time period (24hr- 96hr) to 2.011 mg/ L at 96hr. Sub-chronic toxicity was evaluated over 12 days using sub-lethal concentrations (5 %, 10 %, 15 %, and 20 % of the 24-hr LC50). Survivability in Daphnia showed a decreasing trend from 96 % to 91 % with increase in arsenic concentrations from 5 % of LC50 24 hr value to 20 % of LC 50 24hr value respectively. Alongside decreased survivability, there was a significant reduction in body size, with organisms exposed to the highest concentration of arsenic measuring 0.87±0.01 mm compared to 1.51±0.10 mm in the control group. Reproductive potential declined concentration dependently with exposure, with the highest reduction observed at 20 % of LC50 24hr value, where offspring numbers decreased to 7±1 from 23±5 in the control. Heart rate decreased in concentration and time-dependent manners, with the lowest rates observed at the highest arsenic concentration (279±16 bpm after 24hr and 277±27 bpm after 48hr). Comet assay and micronucleus assay conducted after 48 hrs of exposure revealed concentration-dependent genotoxic effects in Daphnia magna. Our results indicate negative impact on physiology and reproduction of Daphnia magna at environmentally existent concentration of arsenic. Also Daphnia magna could serve as a sensitive test system for investigating arsenic contamination in water bodies.

Abstract:λ-cyhalothrin, a synthetic type II pyrethroid, has become increasingly popular for control of aphids, butterfly larvae, and beetles, replacing other agricultural chemicals. As a result of which, residues of this synthetic pesticide are being reported across the globe in natural water, which poses a serious threat to aquatic life. Therefore, the present study was designed to understand the toxicity effects of λ-cyhalothrin on behaviour, oxidative stress and neurotoxicity in a vertebrate aquatic model, zebrafish ( Danio rerio ). The fish were exposed to 0.129, 0.194 and 0.388 μg/L corresponding to 5%, 10% and 20% of 96hLC 50 (1.94 μg/L) for 28 days. Upon exposure to the highest concentration (0.388 μg/L), the test animal exhibited significant alterations in behavioural patterns like number of entries to the top zone (n), decrease in average speed (m/s) and decrease in time spent in top zone (s). Moreover, the shoaling test demonstrated a significant decrease ( p < 0.05) in the relative time spent by the tested fish (%) near the stimulus fish. The change in behavioural alterations might be linked to a significant decrease ( p < 0.05) in the brain acetylcholine esterase activity. Furthermore, the present study also illustrates oxidative stress exerted by λ-cyhalothrin through an increase in the production of reactive oxygen species, which is again clearly depicted by a significant increase ( p < 0.05) in Superoxide dismutase, Catalase and Glutathione peroxidase activities. Overall, the present study systematically demonstrates the chronic effects of λ-cyhalothrin on adult fish behaviour and physiology, which will contribute to assessing the risks of λ-cyhalothrin to organismal health.

Abstract:Silver nanoparticles (AgNPs) have garnered significant global attention from researchers due to their unique physicochemical properties and wide-ranging applications in industry and medicine. However, their release into aquatic ecosystems has raised concerns regarding potential ecotoxicological consequences. The present study investigated the effects of polyvinyl pyrrolidone-coated silver nanoparticles on Labeo rohita fingerlings, focusing on behavioural reactions, genotoxic effects, histological changes and bioaccumulation. L. rohita fingerlings were exposed to polyvinyl pyrrolidone-coated silver nanoparticles with sizes ranging from 18 to 29 nm for 7 days at concentrations of 100, 200, 400 and 800 ug/l. The nanoparticle zeta potential was found to be extremely negative, measuring - 55.5 mV for 18 nm and - 31.4 mV for 29 nm. Behavioural abnormalities, including respiratory distress, reduced responsiveness and erratic swimming, were observed in exposed groups compared to controls, with severity increasing with higher nanoparticle concentrations. Genotoxicity assessment revealed significantly higher DNA damage in kidney cells compared to gill cells. Histological examination of gill tissues showed clogging in primary and secondary lamellae, along with distorted anatomy, necrosis and vacuolar atrophy in peripheral tubules of the kidneys. The kidneys exhibited greater nanoparticle accumulation than the gills with prolonged exposure. Moreover, 18 nm AgNPs induced more pronounced DNA damage and histological alterations in the kidney and gill tissues compared to 29 nm nanoparticles. This study elucidates the critical role of monitoring AgNPs in aquatic systems, providing essential data on their behaviour and environmental impacts. The findings highlight the need for improved detection techniques and effective management of AgNP contamination. Future research should focus on developing more sensitive analytical methods, understanding long-term ecological effects and exploring innovative remediation strategies.
