Abstract:OBJECTIVE:To optimize the inclusion technology of cinnamon volatile oii-β-cyclodextrin inclusion complex and to investigate the effects of the type of penetration enhancer on the permeation rate of inclusion compounds.METHODS: The experiment was carried out in accordance with L_9(3~4)orthogonal design table;the 3 factors affecting saturated solucion were investigated,and the formula was optimized with steady transdermal permeation rate as evaluation index.With the skin of SD male rat as media,the penetration experiments in vitro were performed on modified Franz diffusion cells.The amount of drug penetration concentration of inclusion compound was determined by HPLC.RESULTS:The optimized inclusion condi- tion was as follows:cinnamon oil:β-cyciodextrin=1:5(mL:g),stirring for 2 hour at 50℃;after addition of the penetration enhancers-DMSO and 1,2-trimethylene glycol,the enhancement ratios were 1.38 and 1.65 respectively(in ,some cases,the penetration rate was decreased).CONCLUSION:The inclusion technology is simple,convenient and practical,and by which transformation of dosage form of cinnamon volatile oil was achieved.β-CD inclusion compound is expected to become a new transdermal cinnamon delivery system.

Abstract:Cannabidiol (CBD), which has a wide variety of biological activities, is one of the phytocannabinoids from Cannabis sativa L. However, due to the poor aqueous solubility, its applications in food, cosmetic, and pharmaceutical fields are limited. Moreover, the previous researches on the improvement in the solubility of CBD are rare. Cyclodextrins (CDs) are useful materials to improve the solubility of poorly water-soluble molecules. Therefore, this work was aimed to study the physicochemical properties, water solubility, and dissolution rate of the inclusion complexes of CBD with β-cyclodextrin (β-CD) and 2, 6-di-O-methyl-β-cyclodextrin (DM-β-CD), and to determine the influence of complexation on the antioxidant activity of CBD. Job's plot and phase solubility study revealed that CBD formed inclusion complexes with β-CD and DM-β-CD at a 1:1 stoichiometric ratio. The thermodynamics analysis demonstrated that the inclusion process was spontaneous and endothermic. Then, the inclusion complexes of CBD with β-CD or DM-β-CD were prepared and characterized by PXRD, FT-IR, DSC, SEM, and 1H NMR. The loading efficiency (LE) of the two inclusion complexes was 20.4% and 17.7% respectively, while the complexation efficiency (CE) was 92.4% and 90.8% respectively. Moreover, the results of molecular docking further showed that CBD was encapsulated successfully. The results also displayed that the water solubility of CBD in CBD/β-CD inclusion complex (IC) and CBD/DM-β-CD IC was significantly increased to 0.395 and 14.118 μg/mL, which was enhanced by 17-fold and 614-fold respectively at such a high LE, and the in vitro dissolution rate of CBD was also promoted after complexation. Besides, the antioxidant activity of CBD was improved after encapsulation with β-CD and DM-β-CD. Significantly, the ABTS free radical scavenging ability of the two inclusion complexes was better than that of Vitamin C (Vc).

Abstract:Improved solubility and anti-inflammatory (AI) properties are imperative for enhancing the effectiveness of poorly water-soluble drugs, particularly non-steroidal anti-inflammatory drugs (NSAIDs). To address these critical issues, our focus is on obtaining NSAID materials in the form of inclusion complexes (IC) with methyl-beta-cyclodextrin (MCD). Ketoprofen (KTP) is selected as the NSAID for this study due to its potency in treating various types of pain, inflammation, and arthritis. Our objective is to tackle the solubility challenge followed by enhancing the AI activity. Confirmation of complexation is achieved through observing changes in the absorbance and fluorescence intensities of KTP upon the addition of MCD, indicating a 1:1 stoichiometric ratio. Phase solubility studies demonstrated improved dissolution rates after the formation of ICs. Further analysis of the optimized IC is conducted using FT-IR, NMR, FE-SEM, and TG/DTA techniques. Notable shifts in chemical shift values and morphological alterations on the surface of the ICs are observed compared to their free form. Most significantly, the IC exhibited superior AI and anti-arthritic (AA) activity compared to KTP alone. These findings highlight the potential of ICs in expanding the application of KTP, particularly in pharmaceuticals, where enhanced stability and efficacy of natural AIs and AAs are paramount.

A Cohort Study to Assess the Impact of Depression on Patients with Kidney Disease

Abstract:Aims: This study assessed the impact of depression on mortality and quality of life of patients with kidney disease. Material: This study was conducted at Hospital São Lucas of Pontificia Universidade Católica of Rio Grande do Sul, Porto Alegre, Brazil, with a cohort of 125 patients seen from 1996 to 2004 (mean follow-up=77.6 months). Patients undergoing renal replacement therapy were asked to fill in the Beck Depression Inventory (BDI). At the completion of follow-up, 72 patients were alive, and 56 answered the BDI and the Medical Outcome Study Short Form General Health Survey (SF-36) to assess quality of life. Main outcome measures were mortality and quality of life. Results: Initial analysis showed that mortality was associated with age, treatment modality—continuous ambulatory peritoneal dialysis (CAPD) or hemodialysis (hd)—and depression. When controlling for age, gender, depression, and treatment modality in a multiple regression analysis, transplantation (tx), gender and age was associated with lower mortality. However, levels of depression at the end of follow-up were strongly associated with quality of life. Conclusions: Depression was not associated with mortality of patients with kidney disease when controlling for gender, age, and treatment modality. Transplantation was the main factor associated with lower mortality. Depression was, however, a strong predictor of quality of life. Number of depressive symptoms was directly associated with lower quality of life in most of the items of the SF-36.
Enhanced Aqueous Solubility and Bioavailability of Capsaicin by the Preparation of an Inclusion Complex

Abstract:To increase the aqueous solubility and poor bioavailability of capsaicin (CAS 404-86-4), in this paper, the effects of hydroxypropyl-beta-cyclodextrin (HP-beta-CD) on the aqueous solubility and the pharmacokinetic characteristics of capsaicin were investigated. The corresponding inclusion complex of capsaicin/HP-beta-CD at the molar ratio of 1:1 was obtained by the method of saturated aqueous solution and characterized by differential scanning calorimetry and X-ray diffractometry. The dissolution rate of capsaicin was significantly increased by the complexation with HP-beta-CD, due to its solubilizing activity. The absorption behavior and bioavailability of capsaicin and its complexation were evaluated after their subcutaneous (s. c.) injection in rats. The absorption rates of capsaicin across subcutaneous tissues and its bioavailability were significantly enhanced by the formation of an inclusion complex with HP-beta-CD. The present results indicate the potential of utilizing HP-beta-CD to improve the subcutaneous absorption and bioavailability of capsaicin.

Abstract:Methylthioadenosine phosphorylase (MTAP), a key enzyme in the methionine salvage pathway, is inactivated in a variety of human cancers. Since all human tissues express MTAP, it would be of potential interest to identify compounds that selectively inhibit the growth of MTAP-deficient cells. To determine if MTAP inactivation could be targeted, the authors have performed a differential chemical genetic screen in isogenic MTAP+ and MTAP− Saccharomyces cerevisiae. A low molecular weight compound library containing 30,080 unique compounds was screened for those that selectively inhibit growth of MTAP− yeast using a differential growth assay. One compound, containing a 1,3,4-thiadiazine ring, repeatedly showed a differential dose response, with MTAP− cells exhibiting a 4-fold shift in IC50 compared to MTAP+ cells. Several structurally related derivatives of this compound also showed enhanced growth inhibition in MTAP− yeast. These compounds were also examined for growth inhibition of isogenic MTAP+ and MTAP− HT1080 fibrosarcoma cells, and 4 of the 5 compounds exhibited evidence of modest but significant increased potency in MTAP− cells. In summary, these studies show the feasibility of differential growth screening technology and have identified a novel class of compounds that can preferentially inhibit growth of MTAP− cells.
Enhancing the solubility and potency of tetrahydrocurcumin as an anti-cancer agent using a β-cyclodextrin inclusion complex approach

Abstract:Curcuminoids originating from turmeric roots are renowned for their diverse pharmacological applications, particularly as a natural anticancer agent. Unfortunately, harnessing the full potential of curcumin derivatives in cancer therapy has been impeded by its inherent limitations, specifically instabilities owing to poor solubility, leading to low systemic bioavailability under normal physiological circumstances. To circumvent this, a novel organic-based drug delivery system employing physically adsorbed β-cyclodextrin (βCD) as an excipient was developed in this study. This resulted in improved aqueous dispersion coupled with anticancer enhancements of tetrahydrocurcumin (THC) at a molar ratio of 2:1. Encapsulation of this agent was confirmed by physicochemical characterisation using UV-vis spectroscopy, differential scanning calorimetry (DSC), Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), and both in vitro and in vivo approaches. Through the presence of an inclusion complex, a higher aqueous dispersion (65-fold) resulting in a higher drug content and an elevated release profile was achieved. Athymic nude (Nu/Nu) mice exposed to this treatment displayed improvements in tumour regression compared to stand-alone agents, consistent with in vitro cytotoxicity assays with an SI value > 10. The inclusion complex further enhanced apoptosis, as well as anti-migration and anti-invasion rates. Mechanistically, this formulation was consistent in terms of caspase 3 activation. Furthermore, the inclusion complex exhibited reduced systemic toxicity, including reduced inflammation in vital organs as examined by hematoxylin and eosin (H&E) staining. This study also revealed a notable sequential reduction in serum levels of tumour markers, including carcinoembryonic antigen (CEA) and mouse Cytochrome P450 1A2 (CYP1A2), correlating with a significant decrease in tumour bulk volume upon treatment commencement. These compelling findings highlight the potential of this formulation to empower insoluble or poorly soluble hydrophobic agents, thus offering promising prospects for their effective utilisation in colorectal cancer (CRC) chemotherapy.

Abstract:This study explores the formation of inclusion complexes between the solubility enhancement of Flufenamic acid (FNA) and various β-Cyclodextrins (β-CD, Hβ-CD, Mβ-CD, and Sβ-CD) using ultrasonic probe sonication method. The investigation encompasses a multi-faceted analysis, employing UV–Visible, FTIR, XRD, FE-SEM, TGA-DSC techniques and molecular docking studies. The UV–Visible spectra reveal absorption peak shifts and binding constants (K a ) indicate interaction strength, and thermodynamic parameters suggest spontaneous and favorable inclusion processes of β-CDs:FNA. Solid inclusion complexes of β-CDs:FNA were further characterized using FTIR, revealing alterations in functional groups indicative of complex formation. XRD analysis elucidates changes in crystallinity, while FE-SEM provides insights into morphological variations. Thermal stability is assessed through TGA-DSC, demonstrate the impact of inclusion on the complexes stability. Phase solubility diagrams illustrate the improved solubility of FNA in the presence of β-CDs. Mβ-CD:FNA exhibits the highest stability constant (1204 M −1 ), emphasizing its potential as an effective carrier. The binding energy of Mβ-CD:FNA (−46.72 kcal/mol) was lower than that of β-CD:FNA (−39.03 kcal/mol), Hβ-CD:FNA (−46.40 kcal/mol), and Sβ-CD:FNA (−46.29 kcal/mol), indicating that the hydroxypropyl group substitution enhanced the inclusion ability of β-CDs. Antibacterial activity studies against S. aureus and E. coli highlight enhanced efficacy of inclusion complexes with Mβ-CD:FNA (92.3 ± 0.18 % and 94.2 ± 0.17 %) demonstrating superior antibacterial activity. Cytotoxicity evaluations on HCT-116 cells reveal the safety profile of the β-CDs:FNA inclusion complexes, with Mβ-CD:FNA showing lower toxicity and high cell internalization compared to other formulations. The findings hold promise for applications in drug delivery, antimicrobial packaging, and the broader pharmaceutical and biotechnological fields.

In vitro and in silico studies of the inclusion complexation of 8-bromobaicalein with β-cyclodextrins

Abstract:Baicalein, a flavone derived from Scutellaria baicalensis Georgi, exhibits potent anti-inflammatory, antiviral, and anticancer properties. Its derivative, known as 8-bromobaicalein (BB), has been found to have strong cytotoxic effect on MCF-7 human breast cancer cells. However, its limited solubility in water has hindered its potential for wider applications. To address this issue, we investigated the use of cyclodextrins specifically βCD, 2,6-di-O-methyl-β-cyclodextrin (DMβCD), and hydroxypropyl-β-cyclodextrin (HPβCD) to improve the solubility of BB through inclusion complexation. During 250 ns molecular dynamics simulations, it was found that BB can form inclusion complexes with all βCDs. These complexes exhibit two distinct orientations: chromone group insertion (C-form) and phenyl group insertion (P-form). The formation of these complexes is primarily driven by van der Waals interactions. DMβCD has the highest number of atom contacts with BB and the lowest solvent accessibility in the hydrophobic cavity. These results coincide with the highest binding affinity from the MM/GBSA-based free energy calculation method. Experimental phase solubility diagrams revealed a 1:1 stoichiometric ratio (AL type) between BB and βCDs, in which BB/DMβCD showed the highest stability. The formation of inclusion complexes was confirmed by differential scanning calorimetry and scanning electron microscope methods. Additionally, the BB/DMβCD inclusion complex demonstrated significantly higher anticancer activity against MCF-7 human breast cancer cells compared to BB alone. These findings underscore the potential of DMβCD for formulating BB in pharmaceutical and medical applications.

Abstract:Cannabidiol (CBD) is one specific kind of the cannabinoid in Cannabis sativa L with a wide range of pharmacological activities. However, the poor water solubility and specificity of CBD limits its application in pharmaceutical field. For solving these problems, in this work, we successfully prepared a targeted carrier by grafting biotin (BIO) onto ethylenediamine-β-Cyclodextrin (EN-CD) in a single step to generate a functionalized supramolecule, named BIO-CD. Subsequently, an amantadine-conjugated cannabinoids (AD-CBD) was prepared and self-assembled with the BIO-CD. A series of methods were used to characterize the inclusion behavior and physicochemical properties of AD-CBD and BIO-CD. The results showed that AD-CBD entered the cavity of BIO-CD and formed a 1:1 host-guest inclusion complex. MTT assay and confocal laser scanning microscopy (CLSM) revealed that the targeting effect and anticancer activity of AD-CBD/BIO-CD inclusion complex against three human cancer cell lines were higher than BIO-CD, AD-CBD and free CBD. Moreover, the inclusion complex could release drugs under weakly acidic conditions. These results demonstrated that AD-CBD/BIO-CD inclusion complex possess excellent targeted and anticancer activity, which is hopeful to be applied in clinic as a new therapeutic approach.

Abstract:This study examines the use of cyclodextrins (CDs) to improve the stability, bioavailability, and water solubility of 7-O-methyl-aromadendrin (7-OMA), a natural compound with antioxidant, anti-inflammatory, antimicrobial, and antiviral properties. Natural CDs (Alpha-, Beta-, and Gamma-cyclodextrin (α-CD, β-CD and γ-CD)) and modified CDs (hydroxypropyl-, amino-, and random methyl-beta-cyclodextrin (HP-β-CD, AM-β-CD and RM-β-CD)) were studied for their potential to form stable inclusion complexes (ICs) with 7-OMA using computational methods e.g. molecular docking and molecular dynamics (MD) simulation. MD simulation revealed reduced hydrogen bonding and radial distribution function (RDF) plots in the encapsulated CDs compared to pure CDs, with HP-β-CD showing the highest number of hydrogen bonds due to its hydroxypropyl substituent. Moreover, the results of molecular docking (-8.0 kcal/mol) and van der Waals interaction energy (-28.0 kcal/mol (117.3 kJ/mol)) indicated that HP-β-CD could form a stable IC with 7-OMA. The interaction energy analysis confirmed the importance of van der Waals interactions and hydrogen bonding in the formation of CDs: 7-OMA ICs. These findings demonstrate the potential of both unmodified and modified CDs as promising excipients for enhancing the antioxidant and anti-inflammatory activity, bioavailability, solubility, and stability of various substances, including 7-OMA.

Abstract:The objective of this study was to improve the stability and water-solubility of patchouli alcohol by complexing with β-cyclodextrin (β-CD). The interactions between patchouli alcohol and β-CD were characterized by differential scanning calorimetry (DSC), Fourier transformation-infrared (FT-IR) spectroscopy, powder X-ray diffraction (PXRD), and Scanning electron microscope (SEM), respectively. According to molecular modeling method, the enthalpy formation of host-guest illustrated the predominant configuration and the lowest value ΔbGo was -10.8174±1.9235 kcal/mol, suggesting the complex could reduce the energy of the system. The characterization analysis confirmed the formation of PA-CD inclusion complex, and the results indicated the advantage of the inclusion complex in stability and dissolution rates. These results identified PA-CD inclusion complex an effective way for the storage of PA, and better inclusion method still needed to be studied.

Abstract:Liposomes- encapsulated indomethacin/ cyclodextrins ( IMC/ CD) inclusion complexes were prepared. The characteristics and pharmacokinetics of the combined system were investigated. The high drug entrapment values of 2.38 +/- 0.16 mu g/mg and 2.48 +/- 0.12 mu g/mg for liposomes-encapsulated IMC/beta-CD and IMC/HP-beta-CD inclusion complexes were achieved, as only 1.60 +/- 0.09 mu g/ mg for conventional liposomes. Encapsulating IMC/CD inclusion complexes into liposomes resulted in a slow release of drug. Following intravenous administration, both liposomes- encapsulated inclusion complexes showed significantly improved AUC(0-infinity) compared with that of conventional liposomes ( p < 0.05). After intramuscular administration, Cmax has been increased to 5.21 +/- 1.14 mu g . ml(-1) and 6.02 +/- 1.22 mu g . ml(-1) for liposomes- encapsulated IMC/beta-CD and IMC/HP-beta-CD inclusion complexes, respectively, whereas only 2.43 +/- 0.69 mu g . ml(-1) for liposomes- encapsulated free drug ( p < 0.01).

Abstract:Essential oils (EOs) are primarily isolated from medicinal plants and possess various biological properties. However, their low water solubility and volatility substantially limit their application potential. Therefore, the aim of the current study was to improve the solubility and stability of the Mosla Chinensis (M. Chinensis) EO by forming an inclusion complex (IC) with β-cyclodextrin (β-CD). Furthermore, the IC formation process was investigated using experimental techniques and molecular modeling. The major components of M. Chinensis 'Jiangxiangru' EOs were carvacrol, thymol, o-cymene, and terpinene, and its IC with β-CD were prepared using the ultrasonication method. Multivariable optimization was studied using a Plackett-Burman design (step 1, identifying key parameters) followed by a central composite design for optimization of the parameters (step 2, optimizing the key parameters). SEM, FT-IR, TGA, and dissolution experiments were performed to analyze the physicochemical properties of the ICs. In addition, the interaction between EO and β-CD was further investigated using phase solubility, molecular docking, and molecular simulation studies. The results showed that the optimal encapsulation efficiency and loading capacity of EO in the ICs were 86.17% and 8.92%, respectively. Results of physicochemical properties were different after being encapsulated, indicating that the ICs had been successfully fabricated. Additionally, molecular docking and dynamics simulation showed that β-CD could encapsulate the EO component (carvacrol) via noncovalent interactions. In conclusion, a comprehensive methodology was developed for determining key parameters under multivariate conditions by utilizing two-step optimization experiments to obtain ICs of EO with β-CD. Furthermore, molecular modeling was used to study the mechanisms involved in molecular inclusion complexation.

Abstract:Abstract In our work, the inclusion ability and binding pattern of flavonoids (catechin, iso‐liquiritigenin, luteolin, puerarin, rutin, and curcumin) with β‐cyclodextrin (β‐CD) and curcumin (CUR) with CDs (α‐CD, β‐CD, γ‐CD, dimethyl‐β‐CD, Hydroxypropyl‐β‐CD, and glucosyl‐β‐CD) were studied by molecular docking. The results showed that CUR among the six flavonoids was more likely to form stable inclusion complexes with β‐CD, and the HP‐β‐CD is the most suitable for CUR inclusion among the 6 types of CD. Phase solubility studies showed that CUR with β‐CD or HP‐β‐CD, respectively, could form stable inclusion complexes in a stoichiometric ratio of 1 : 1. The CUR/β‐CD and CUR/HP‐β‐CD inclusion complexes (ICs) were prepared by freeze‐drying method. The successful preparation of IC was confirmed by SEM images, FT‐IR spectra, and TG‐DSC. The results of in vitro release showed that the release of CUR from IC prepared with β‐CD or HP‐β‐CD could be improved significantly, and the release effect of HP‐β‐CD was better than that of β‐CD. The inclusion with CD could be used to improve the solubility and bioavailability of CUR.

Abstract:An inclusion complex between 2,6-di-O-methyl-β-cyclodextrin (DM-β-CD) and chlorzoxazone was prepared and investigated. Freeze-drying method was used to synthetize the complex; Fourier transform infrared spectroscopy, laboratory powder X-ray diffraction, 1H nuclear magnetic resonance spectroscopy, Rotating-frame overhauser effect spectroscopy, and Differential scanning calorimetry were utilized to confirm the formation of the complex. Furthermore, the solubilization and dissolution study results showed that the water solubility and dissolution rate of CZX were obviously improved through complexation with DM-β-CD. Finally, the cytotoxicity of the complex was determined by MTT assay. As expected, the complex expressed lower toxicities for normal liver cells than free CZX. This study provides a promising idea to develop novel CZX solid dosage form with good dissolution rate and low liver toxicity.

Abstract:Cannabinoids: cannabidiol (CBD), cannabidiolic acid (CBDA), and cannabichromene (CBC) are lipophilic compounds with limited water solubility, resulting in challenges related to their bioavailability and therapeutic efficacy upon oral administration. To overcome these limitations, we developed co-dispersion cannabinoid delivery systems with the biopolymer polyvinyl caprolactam-polyvinyl acetate-polyethylene glycol (Soluplus) and magnesium aluminometasilicate (Neusilin US2) to improve solubility and permeability. Recognizing the potential therapeutic benefits arising from the entourage effect, we decided to work with an extract instead of isolated cannabinoids. Cannabis sativa inflorescences (Henola variety) with a confirming neuroprotective activity were subjected to dynamic supercritical CO2 (scCO2) extraction and next they were combined with carriers (1:1 mass ratio) to prepare the co-dispersion cannabinoid delivery systems (HiE). In vitro dissolution studies were conducted to evaluate the solubility of CBD, CBDA, and CBC in various media (pH 1.2, 6.8, fasted, and fed state simulated intestinal fluid). The HiE-Soluplus delivery systems consistently demonstrated the highest dissolution rate of cannabinoids. Additionally, HiE-Soluplus exhibited the highest permeability coefficients for cannabinoids in gastrointestinal tract conditions than it was during the permeability studies using model PAMPA GIT. All three cannabinoids exhibited promising blood-brain barrier (BBB) permeability (Papp higher than 4.0 × 10−6 cm/s), suggesting their potential to effectively cross into the central nervous system. The improved solubility and permeability of cannabinoids from the HiE-Soluplus delivery system hold promise for enhancement in their bioavailability.

Abstract:Oxyresveratrol (OXY), a natural stilbenoid in mulberry fruits, is known for its diverse pharmacological properties. However, its clinical use is hindered by low water solubility and limited bioavailability. In the present study, the inclusion complexes of OXY with β-cyclodextrin (βCD) and its three analogs, dimethyl-β-cyclodextrin (DMβCD), hydroxypropyl-β-cyclodextrin (HPβCD) and sulfobutylether-β-cyclodextrin (SBEβCD), were investigated using in silico and in vitro studies. Molecular docking revealed two binding orientations of OXY, namely, 4',6'-dihydroxyphenyl (A-form) and 5,7-benzenediol ring (B-form). Molecular Dynamics simulations suggested the formation of inclusion complexes with βCDs through two distinct orientations, with OXY/SBEβCD exhibiting maximum atom contacts and the lowest solvent-exposed area in the hydrophobic cavity. These results corresponded well with the highest binding affinity observed in OXY/SBEβCD when assessed using the MM/GBSA method. Beyond traditional simulation methods, Ligand-binding Parallel Cascade Selection Molecular Dynamics method was employed to investigate how the drug enters and accommodates within the hydrophobic cavity. The in silico results aligned with stability constants: SBEβCD (2060 M -1 ), HPβCD (1860 M -1 ), DMβCD (1700 M -1 ), and βCD (1420 M -1 ). All complexes exhibited a 1:1 binding mode (A L type), with SBEβCD enhancing OXY solubility (25-fold). SEM micrographs, DSC thermograms, FT-IR and 1 H NMR spectra confirm the inclusion complex formation, revealing novel surface morphologies, distinctive thermal behaviors, and new peaks. Notably, the inhibitory impact on the proliferation of breast cancer cell lines, MCF-7, exhibited by inclusion complexes particularly OXY/DMβCD, OXY/HPβCD, and OXY/SBEβCD were markedly superior compared to that of OXY alone.

Abstract:β-cyclodextrin and hydroxypropyl-β-cyclodextrin were combined with baicalein to improve its solubility in water. The physicochemical properties and pharmacokinetic characteristics of the two complexes and their therapeutic effects on dextran sulfate sodium-induced ulcerative colitis were studied in mice. Differential scanning calorimetry, X-ray diffraction, infrared spectroscopy, and scanning electron microscopy showed that the two complexes were successfully prepared and significantly improved baicalein's solubility and dissolution. The t1/2, Cmax, and AUC0-∞ values of the inclusion complexes were substantially better than those of the original drug in rats. Analysis of the tissue distribution of acute ulcerative colitis in mice showed that the peak concentrations of baicalein and its metabolite, baicalin, in the small intestines, liver, and colon were higher than that of the free drug. The therapeutic effect of the inclusion compound was significantly better than that of the original drug. This study provides a new reference for applying cyclodextrin inclusion complex in ulcerative colitis.

Abstract:Cyclodextrin (CD) complexation has emerged as a promising approach for enhancing the solubility or/and stability of hydrophobic small molecule substances. Here the inclusion complexes of Vitamin A (VA), Vitamin A acetate (VAA), and Vitamin A palmitate (VAP) with β-cyclodextrin (β-CD) were prepared by co-precipitation with freeze-drying methods. Results from FT-IR, NMR, and SEM confirmed the formation of inclusion complexes. Phase solubility diagrams showed a molecular ratio of 2:1, 1:1, and 1:1 for β-CD complexes with VA, VAA, and VAP, respectively. Among these vitamins tested, free VA has the lowest thermal stability, but it was the highest after embedding with β-CD. β-CD:VA also showed a better water solubility. Although VAP achieved the highest increase rate in water solubility among the three vitamins after embedding with β-CD, water solubility of β-CD:VAP was still relatively small due to the poor water solubility of free VAP in nature. The obtained results revealed that β-CD complexation is more suitable for vitamin A than the other two vitamin A esters in promoting thermal stability and water solubility.On the basis of these experimental studies, we furthermore performed molecular docking simulations to predict possible binding mode and binding affinities of three vitamins with β-CD. Gibbs free energy changes (ΔG°) of β-CD:VA with 2:1 molecular ratio was lowest, which explained the enhanced thermal stability of VA by β-CD encapsulation. This work is useful for enlarged applications of β-CD inclusion complexes with vitamin A and its esters.
