Abstract:This study aimed to investigate the effect of exogenous nanoparticles on the rheological properties of artificial saliva. There are four reasons for undertaking this type of research: Firstly, the number of solid particles of various origins present in the air is still high. Secondly, nanoparticles (including silver and gold nanoparticles) are increasingly used in food packaging and can migrate into food. Thirdly, saliva is the first biological fluid that comes into contact with exogenous particles. Finally, the function of saliva is also closely related to its rheological properties. Due to the remarkable properties of nano-objects, nanoparticles of various origins in the body may cause effects that have not been realised until now. Therefore, each type of nanoparticle must be tested in terms of its impact on the body/body fluid. We used silver and gold nanoparticles because they are used in the food industry, and diesel exhaust particles because they are standard components of air pollution. The effect of various nanoparticles (e.g., their size and shape) on the rheology of saliva at two temperatures was investigated. The constants of the power law constitutive equation were also estimated. Studies showing the impact of nanoparticles on the rheology of body fluids are rare because it is one of the less obvious ways of their affecting the human body. However, the results show that nanoparticles are not neutral to the biological fluid, which may translate into a change in its properties and thus disturb its functions.

Abstract:Every day, approximately 7 m3 of air flows through the lungs of an adult, which comes into contact with 80 m2 of the lung surface. This air contains both natural and anthropogenic particles, which can deposit on the surface of the mucus lining the respiratory tract. The presence of particles in the mucus leads to changes in its rheology and, consequently, in its functions. Therefore, this research aimed to determine how a non-Newtonian fluid suspension will behave during flow, illustrating the movement of mucus during coughing. The model mucus was an aqueous solution of carboxymethylcellulose (CMC). The tested particles suspended in a non-Newtonian fluid were Arizona Fine Dust, diesel exhaust particles, polyethylene microparticles, and pine pollen. It was noticed that as the fluid viscosity increases, the number of Kelvin–Helmholtz instabilities increases. The fluid's expansion angle at the output of the measuring cell decreased, and the values of parameters characterizing the aerosol generated at the outlet decrease for selected particles present in the fluid. The research shows that the deposition of particles from polluted air in the respiratory tract, although they do not enter the bloodstream, may affect the human body. Deposited particles can change the behavior of mucus, which may translate into its functions.

Understanding the interaction of micro(nano)plastics with simulated saliva, lysosomal and Gamble’s �uids and their impact on human lung epithelial A549 cells

Abstract:Micro(nano)plastics are considered an emerging threat to human health since they have potential to interact with biological systems and already been found in the human body, e.g., lungs. However, limited data is available for their behavior under biological conditions and impact on human cells, speci�cally alveolar epithelial cells. In this study, micro(nano)plastics weathered to various simulated biological �uids (saliva, arti�cial lysosomal �uids, Gamble’s solution) during 2 h to 80 h. Pristine and weathered plastic particles were characterized by their surface chemistry, zeta potentials, and elemental composition. After, various toxicological endpoints (mitochondrial membrane potential, lactate dehydrogenase, protein, and antioxidant levels) were examined through A549 lung carcinoma cells. The surface characteristics of micro(nano)plastics and the toxicological endpoints of A549 were in�uenced by the simulated biological weathering, speci�cally at high concentration of micro(nano)plastics and increasing exposure under biological conditions. The results also indicated that toxicological endpoints had strong linked with the chemistry of plastics and included the multiple processes to the response of the plastics, for instance, the cell membrane integrity and mitochondrial activity were declined with the micro(nano)plastics under saliva weathering, contrarily, their protein and antioxidant activities increased. However, different biological pathways were obtained under arti�cial lysosomal �uid and Gamble’s solution.

Abstract:Microplastics are a pervasive environmental contaminant and represent a newly emerging threat to human health. They enter the body through various pathways, including food consumption, drinking water, and inhalation. The present study is concerned with the issue of dietary and inhalational exposure to microplastics. The presence of microplastics has been identified in a range of food products and drinking water, with analyses of faecal samples also indicating their passage through the human digestive tract, thereby suggesting the potential for their absorption by the body. It is also important to consider the potential for inhalational exposure, given the presence of microplastics in the air. This suggests that inhalation by humans may also be a possibility. Furthermore, these particles have been identified in sputum samples from patients and lung tissues, indicating the potential for bioaccumulation and the induction of inflammatory responses. While there is documented evidence of microplastics in the human body, there is currently a lack of specific data on their impact on human health. The available evidence from animal studies and in vitro models indicates that microplastics have the potential to traverse epithelial barriers in the digestive and respiratory systems, resulting in inflammatory responses, oxidative stress, and, in extreme cases, tissue fibrosis. Furthermore, microplastics may act as carriers for heavy metals and toxic chemicals, thereby increasing the risk of neurotoxicity, nephrotoxicity, and potentially carcinogenic effects. Further research, including long-term clinical studies, is required to gain a full understanding of the impact of microplastics on human health.

Abstract:Fertilization results in the biphasic activation of polyphosphoinositide-specific phospholipase C (PLC) activity with an initial increase in activity coincident with the sperm-induced calcium transient, followed by a more sustained increase prior to mitosis. Immunoprecipitation studies demonstrated that the gamma isoform of PLC is present in both the unfertilized and the fertilized egg and contributes to the initial phase of PLC activation. Fertilization also resulted in translocation of a significant fraction of PLC-gamma from the cytosol to the membrane compartment of the egg.

In this study, micro(nano)plastics were exposed to various simulated biological fluids (artificial lysosomal fluids and Gamble's solution) for 2–80 h. Pristine and treated plastic particles were characterized based on their surface chemistry, zeta potentials, and elemental composition. Airborne Polystyrene Microplastics and Nanoplastics Induce Nasal and Lung Microbial Dysbiosis in Mice.

Abstract:Microplastics (MP) and nanoplastics (NP) have been found in multiple environments and creatures. However, their effects on the airway microbiota still remain poorly understood. In this study, a series of bioinformatic and statistical analyses were carried out to explore the influence of airborne MP and NP on the nasal and lung microbiota in mice. Both MP and NP were capable of inducing nasal microbial dysbiosis, and MP had a stronger influence on the lung microbiota than NP. Multiple nasal and lung bacteria were associated with MP and NP groups, among which nasal Staphylococcus and lung Roseburia were most associated with MP group, while nasal Prevotella and lung unclassified_Muribaculaceae were most associated with NP group. The nasal Staphylococcus, lung Roseburia, lung Eggerthella and lung Corynebacterium were associated with both MP and NP groups, which were potential biomarkers of micro/nanoplastics-induced airway dysbiosis. SAR11_Clade_Ia and SAR11_Clade_II were associated with both nasal and lung microbiota in MP group, while no such bacterium was determined in NP group. The relevant results suggest that both airborne MP and NP could induce nasal and lung microbial dysbiosis, and the relevant preventative and curable strategies deserve further investigations.
Pulmonary toxicity assessment of polypropylene, polystyrene, and polyethylene microplastic fragments in mice

Abstract:Polypropylene (PP), polystyrene (PS), and polyethylene (PE) plastics are commonly used in household items such as electronic housings, food packaging, bottles, bags, toys, and roofing membranes. The presence of inhalable microplastics in indoor air has become a topic of concern as many people spent extended periods of time indoors during the COVID-19 pandemic lockdown restrictions, however, the toxic effects on the respiratory system are not properly understood. We examined the toxicity of PP, PS, and PE microplastic fragments in the pulmonary system of C57BL/6 mice. For 14 days, mice were intratracheally instilled 5 mg/kg PP, PS, and PE daily. The number of inflammatory cells such as macrophages, neutrophils, and eosinophils in the bronchoalveolar lavage fluid (BALF) of PS-instilled mice was significantly higher than that in the vehicle control (VC). The levels of inflammatory cytokines and chemokines in BALF of PS-instilled mice increased compared to the VC. However, the inflammatory responses in PP- and PE-stimulated mice were not significantly different from those in the VC group. We observed elevated protein levels of toll-like receptor (TLR) 2 in the lung tissue of PP-instilled mice and TLR4 in the lung tissue of PS-instilled mice compared with those to the VC, while TLR1, TLR5, and TLR6 protein levels remained unchanged. Phosphorylation of nuclear factor kappa B (NF-κB) and IĸB-α increased significantly in PS-instilled mice compared with that in VC. Furthermore, Nucleotide‑binding oligomerization domain‑like receptor family pyrin domain‑containing 3 (NLRP3) inflammasome components including NLRP3, apoptosis-associated speck-like protein containing a caspase recruitment domain (ASC), and Caspase-1 in the lung tissue of PS-instilled mice increased compared with that in the VC, but not in PP- and PE-instilled mice. These results suggest that PS microplastic fragment stimulation induces pulmonary inflammation due to NF-ĸB and NLRP3 inflammasome activation by the TLR4 pathway. Abstract:Plastics are used all over the world. Unfortunately, due to limited biodegradation, plastics cause a significant level of environmental pollution. The smallest recognized to date are termed nanoplastics (1 nm [nm] up to 1 μm [μm]) and microplastics (1 μm-5 mm). These nano- and microplastics can enter the human body through the respiratory system via inhalation, the digestive tract via consumption of contaminated food and water, or penetration through the skin via cosmetics and clothes contact. Bioaccumulation of plastics in the human body can potentially lead to a range of health issues, including respiratory disorders like lung cancer, asthma and hypersensitivity pneumonitis, neurological symptoms such as fatigue and dizziness, inflammatory bowel disease and even disturbances in gut microbiota. Most studies to date have confirmed that nano- and microplastics can induce apoptosis in cells and have genotoxic and cytotoxic effects. Understanding the cellular and molecular mechanisms of plastics' actions may help extrapolate the risks to humans. The article provides a comprehensive review of articles in databases regarding the impact of nano- and microplastics on human health. The review included retrospective studies and case reports of people exposed to nanoplastics and microplastics. This research highlights the need for further research to fully understand the extent of the impact of plastics on human health.

Abstract:MR angiography of the aortic arch vessels and upper extremities is a noninvasive alternative to conventional catheter angiography without the risks of stroke or the potential toxicity of iodinated contrast. The use of optimized, breath hold gadolinium-enhanced three-dimensional MR angiography provides excellent image quality with the capability of display options not available with conventional angiography. This article demonstrates the utility of this new technique in the diagnosis of congenital and acquired conditions of the supraaortic vascular system.

Abstract:Globally, plastics are used in various products. Concerns regarding the human body's exposure to plastics and environmental pollution have increased with increased plastic use. Microplastics can be detected in the atmosphere, leading to potential human health risks through inhalation; however, the toxic effects of microplastic inhalation are poorly understood. In this study, we examined the pulmonary toxicity of polystyrene (PS), polypropylene (PP), and polyvinyl chloride (PVC) in C57BL/6, BALB/c, and ICR mice strains. Mice were intratracheally instilled with 5 mg/kg of PS, PP, or PVC daily for two weeks. PS stimulation increased inflammatory cells in the bronchoalveolar lavage fluid (BALF) of C57BL/6 and ICR mice. Histopathological analysis of PS-instilled C57BL/6 and PP-instilled ICR mice showed inflammatory cell infiltration. PS increased the NLR family pyrin domain containing 3 (NLRP3) inflammasome components in the lung tissue of C57BL/6 and ICR mice, while PS-instilled BALB/c mice remained unchanged. PS stimulation increased inflammatory cytokines, including IL-1β and IL-6, in BALF of C57BL/6 mice. PP-instilled ICR mice showed increased NLRP3, ASC, and Caspase-1 in the lung tissue compared to the control groups and increased IL-1β levels in BALF. These results could provide baseline data for understanding the pulmonary toxicity of microplastic inhalation.

Abstract:Micro- and nanoplastics (MNPs) can enter the atmosphere via sea spray aerosols (SSAs), but the effects of plastic characteristics on the aerosolization process are unclear. Furthermore, the importance of the transport of MNPs via these SSAs as a possible new exposure route for human health remains unknown. The aim of this study was two-fold: (1) to examine if a selection of factors affects aerosolization processes of MNPs, and (2) to estimate human exposure to MNPs via aerosols inhalation. A laboratory-based bubble bursting mechanism, simulating the aerosolization process at sea, was used to investigate the influence of MNP as well as seawater characteristics. To determine the potential human exposure to microplastics via inhalation of SSAs, the results of the laboratory experiments were extrapolated to the field based on sea surface microplastic concentrations and the volume of inhaled aerosols. Enrichment seemed to be influenced by MNP size, concentration and polymer type. With higher enrichment for smaller particles and denser polymers. Experiments with different concentrations showed a larger range of variability but nonetheless lower concentrations seemed to result in higher enrichment, presumably due to lower aggregation. In addition to the MNP characteristics, the type of seawater used seemed to influence the aerosolization process. Our human exposure estimate to microplastic via inhalation of sea spray aerosols shows that in comparison with reported inhaled concentrations in urban and indoor environments, this exposure route seems negligible for microplastics. Following the business-as-usual scenario on plastic production, the daily plastic inhalation in coastal areas in 2100 is estimated to increase but remain far below 1 particle per day. This study shows that aerosolization of MNPs is a new plastic transport pathway to be considered, but in terms of human exposure it seems negligible compared to other more important sources of MNPs, based on current reported environmental concentrations.

Abstract:Micro- and nanoplastic particles, including common forms like polyethylene and polystyrene, have been identified as relevant pollutants, potentially causing health problems in living organisms. The mechanisms at the cellular level largely remain to be elucidated. This study aims to visualize nanoplastics in bronchial smooth muscle (BSMC) and small airway epithelial cells (SAEC), and to assess the impact on mitochondrial metabolism. Healthy and asthmatic human BSMC and SAEC in vitro cultures were stimulated with polystyrene nanoplastics (PS-NPs) of 25 or 50 nm size, for 1 or 24 h. Live cell, label-free imaging by holotomography microscopy and mitochondrial respiration and glycolysis assessment were performed. Furthermore, 25 and 50 nm NPs were shown to penetrate SAEC, along with healthy and diseased BSMC, and they impaired bioenergetics and induce mitochondrial dysfunction compared to cells not treated with NPs, including changes in oxygen consumption rate and extracellular acidification rate. NPs pose a serious threat to human health by penetrating airway tissues and cells, and affecting both oxidative and glycolytic metabolism.

Abstract:Background Concerns on microplastics (MPs) in food are increasing because of our increased awareness of daily exposure and our knowledge gap on their potential adverse health effects. When particles are ingested, macrophages play an important role in scavenging them, potentially leading to an unwanted immune response. To elucidate the adverse effects of MPs on human health, insights in the immunotoxicity of MPs are essential. Objectives To assess the effect of environmentally collected ocean and land weathered MP particles on the immunological response of macrophages using a state-of-the art in vitro immunotoxicity assay specifically designed for measuring particle toxicity. Methods Environmentally-weathered macroplastic samples were collected from the North Pacific Subtropical Gyre and from the French coastal environment. Macroplastics were identified using (micro)Raman-spectrometry, FT-IR and Py-GC-MS and cryo-milled to obtain size-fractionated samples up to 300 μm. Physiochemical MP properties were characterized using phase contrast microscopy, gel-permeation chromatography, nuclear magnetic resonance, and differential scanning colorimetry. Macrophages (differentiated THP-1 cells) were exposed to particles (<300 μm) for 48 h before assessment of cell viability and cytokine release. Using both the physiochemical particle properties and biological data, we performed multi-dimensional data analysis to explore relationships between particle properties and immunotoxicological effects. Results We investigated land-derived polyethylene, polypropylene, polystyrene, and polyethylene terephthalate, water-derived polypropylene macroplastics, and virgin polyethylene fibers and nylon MPs. The different plastic polymeric compositions and MP size classes induced distinct cytokine responses. Macrophages had the largest response to polyethylene terephthalate-particle exposure, including a dose-related increase in IL-1β, IL-8, and TNF-α secretion. Smaller MPs induced cytokine production at lower concentrations. Additionally, a relationship between both physical and chemical particle properties and the inflammatory response of macrophages was found. Discussion This research shows that MP exposure could lead to an inflammatory response in vitro , depending on MP material and size. Whether this implies a risk to human health needs to be further explored.

Abstract:Throughout their lifecycle, from production to use and upon disposal, plastics release chemicals and particles known as micro- and nanoplastics (MNPs) that can accumulate in the environment. MNPs have been detected in different locations of the human body, including in our lungs. This is likely a consequence of MNP exposure through the air we breathe. Yet, we still lack a comprehensive understanding of the impact that MNP exposure may have on respiratory disease and health. In this review, we have collated the current body of evidence on the implications of MNP inhalation on human lung health from in vitro , in vivo and occupational exposure studies. We focused on interactions between MNP pollution and different specific lung-resident cells and respiratory diseases. We conclude that it is evident that MNPs possess the capacity to affect lung tissue in disease and health. Yet, it remains unclear to which extent this occurs upon exposure to ambient levels of MNPs, emphasising the need for a more comprehensive evaluation of environmental MNP exposure levels in everyday life.

Abstract:Microplastics pose a growing environmental threat with complex implications for human health. Despite the extensive research on the cytotoxicities of microplastics, gaps remain in understanding cellular responses to the interplay between environmental weathering and physiological processes. This study aims to fill this knowledge gap by evaluating and comparing the in vitro cellular responses to pristine polystyrene particles, particles weathered under UV light in DI water and seawater, and particles with subsequent incubation in simulated gastric fluid (SGF). In this study, Fourier transform infrared spectroscopy in attenuated total reflection mode (ATR-FTIR), Raman spectroscopy, X-ray photoelectron spectroscopy (XPS), and scanning electron microscopy-energy dispersive spectroscopy (SEM-EDS) were implemented to conduct surface chemistry and morphology characterizations of the particles. The combination of these techniques allowed a comprehensive understanding of surface morphology and chemistry alterations due to the weathering degradation and SGF incubation. Results showed nitrogen and carbonyl groups formed on weathered particles, and seawater-weathered particles showed a more pronounced weathering degree. After SGF incubation, stronger nitrogen and amide groups were detected on the surface of weathered particles, and more organic matter was attached. Two cell lines that are widely used for the evaluation of microplastic cytotoxicity were used, RAW264.7 macrophage and Caco-2 intestine epithelial cells. Results showed weathered and SGF-treated particles enhanced macrophage viability and pro-inflammatory effects compared to pristine particles. Elevated reactive oxygen species (ROS) generation was detected for all particle groups. Weathered particles caused higher cytotoxicity effects on Caco-2 cells and damaged tight junction integrity. The organic matter formation from the SGF incubation protected tight junction integrity and reduced cytotoxicity. These findings highlight the importance of taking both environmental and physiological factors into account for a more comprehensive assessment of microplastic toxicity.

Abstract:Abstract The continuously increasing use of plastics is supposed to result in a rising exposure of MNPs to humans. Available data on human health risks of microplastics after oral uptake increased immensely in the past years and indicates very likely only low risks after oral consumption. Concerning nanoplastics, uptake, transport and potential adverse effects after oral uptake are less well understood. This study aims to investigate differences between microplastic particles and particles in the submicron- and nanoscaled size derived from food-relevant polymers with a particle size range consistent with higher potential for cellular uptake, fate, and effects when applied to human intestinal and liver cells. This work includes the development of cellular and subcellular detection methods for synthetic polymeric particles in the micro- and nanometer-range, using Scanning Electron Microscopy, Small-Angle X-ray and Dynamic Light Scattering methods, Asymmetric Flow Field Flow Fractionation, octanol-water fractionation, fluorescence microscopy and flow cytometry. Polylactic acid (250 nm and 2 μm (polydisperse)), melamine formaldehyde (366 nm) and polymethylmethacrylate (25 nm) were thoroughly characterized. The submicro- and nanoplastic test particles showed an increased uptake and transport quantity through intestinal cells. Both types of particles resulted in observed differences of uptake behavior, most likely influenced by different lipophilicity, which varied between the polymeric test materials. Toxic effects were detected after 24 h only in overload situations for the particles in the submicrometer range. This study provides further evidence for gastrointestinal uptake of submicro- and nanoplastics and points towards differences regarding bioavailability between microplastics and smaller plastic particles that may result following the ingestion of contaminated food and beverages. Furthermore, the results reinforce the importance for studying nanoplastics of different materials of varying size, surface properties, polymer composition and hydrophobicity.
Detrimental effects of microplastic exposure on normal and asthmatic pulmonary physiology

Abstract:Concerns that airborne microplastics (MP) may be detrimental to human health are rising. However, research on the effects of MP on the respiratory system are limited. We tested the effect of MP exposure on both normal and asthmatic pulmonary physiology in mice. We show that MP exposure caused pulmonary inflammatory cell infiltration, bronchoalveolar macrophage aggregation, increased TNF-α level in bronchoalveolar lavage fluid (BALF), and increased plasma IgG1 production in normal mice. MP exposure also affected asthma symptoms by increasing mucus production and inflammatory cell infiltration with notable macrophage aggregation. Further, we found co-labeling of macrophage markers with MP incorporating fluorescence, which indicates phagocytosis of the MP by macrophages. A comparative transcriptomic analysis showed that MP exposure altered clusters of genes related to immune response, cellular stress response, and programmed cell death. A bioinformatics analysis further uncovered the molecular mechanism whereby MP stimulated production of tumor necrosis factor and immunoglobulins to activate a group of transmembrane B-cell antigens, leading to the modulation of cellular stress and programmed cell death in the asthma model. In summary, we show that MP exposure had detrimental effects on the respiratory system in both healthy and asthmatic mice, which calls for urgent discourse and action to mitigate environmental microplastic pollutants.
