Abstract:Nitrite (NO2-) widely exists in our daily diet, and its excessive consumption can lead to detrimental effects on the human central nervous system and an elevated risk of cancer. The fluorescence probe method for the determination of nitrite has developed rapidly due to its simplicity, rapidity and sensitivity. Despite establishing various nitrite sensing platforms to ensure the safety of foods and drinking water, the simultaneous achievement of rapid, specific, affordable, visualizing, and on-site nitrite detection remains challenging. Here, we designed a novel fluorescent probe by using Rhodamine 800 as the fluorescent skeleton and 5-aminoindole as the specific reaction group to solve this problem. The probe shows a maximal fluorescence emission at 602 nm, thereby avoiding background emission interference when applied to food samples. Moreover, this unique probe exhibited excellent sensing capabilities for detecting nitrite. These included: a rapid response time within 3 min, a noticeable color change that the naked eye can observe, a low detection limit of 13.8 nM, and a remarkable selectivity and specificity to nitrite. Besides that, the probe can detect nitrite quantitatively in barreled drinking water, ham sausage, and pickles samples, with good recoveries ranging from 89.0 % to 105.8 %. More importantly, based on the probe fixation and signal processing technology, a portable and smart sensing platform was fabricated and made convenient and rapid analysis the content of NO2- in real samples possible. The results obtained in this work provide a new strategy for the design of high-performance nitrite probes and feasible technology for portable, rapid and visual detection of nitrite, and this probe holds the potential as a practical tool for alleviating concern regarding nitrite levels.

Abstract:This study demonstrates a novel strategy for colorimetric/fluorescent/surface enhanced Raman scattering (SERS) triple-mode sensing of nitrite based on Griess reaction modulated gold nanorods (GNRs)-Azo-gold nanoparticles (GNPs) (GNRs-Azo-GNPs) assembly. The p-aminothiophenol (PATP)-capped GNRs (GNRs@PATP) and 1,8-diaminonaphthalene (DAP)-modified GNPs (GNPs@DAP) are first synthesized through Au–S and Au–N bonds, respectively. Upon being excited at 365nm, the dispersion of GNRs-GNPs (GNRs@PATP and GNPs@DAP) emits cyan fluorescence. Next, the addition of nitrite into the GNRs-GNPs induces the formation of GNRs-Azo-GNPs assembly, resulting in the enhancement of color and decrease of fluorescence. Therefore, the GNRs-Azo-GNPs assembly can not only be used as a naked-eye indicator of nitrite changed from orange-yellow to purple, but also as a highly selective ‘‘fluorescence quenching’’ probe due to the fluorescence resonance energy transfer (FRET) between azo-moiety and DAP. The limit of detection (LOD) for nitrite is 0.05μM by colorimetry and 0.01μM by fluorescence. Meanwhile, the GNRs-Azo-GNPs assembly possesses controllable core–satellites nanostructures and enables on-field SERS detection of nitrite with the LOD of 0.8nM. More importantly, the GNRs-GNPs sensing system can not only be used as a paper-based test strip for on-site fast screening of nitrite with a high sensitivity and selectivity, but also as a SERS substrate for reliable quantitative analysis of nitrite. This study offers a new method for on-site visual detection of nitrite in human urine and meat products, as well as provides a strategy for designing multi-mode sensing platform for various applications.
Rational design of carbon dot nanozymes for ratiometric dual-signal and smartphone-assisted visual detection of nitrite in food matrices

Abstract:Developing reliable nitrite (NO2-) sensors is essential for food safety and reducing health risks from NO2- exposure. In this study, we strategically designed nitrogen-doped carbon dot (N-CD) nanozymes to establish an accessible dual-signal ratiometric sensing system for detecting NO2- in food matrices. This system utilizes the photoluminescence and enzyme-like properties of N-CD nanozymes combined with NO2--triggered diazotization reactions of substrates such as o-phenylenediamine (OPD) or 3,3',5,5'-tetramethylbenzidine (TMB). The resulting N-CD/OPD and N-CD/TMB composites provide dual-mode detection-fluorescence and colorimetric-with high selectivity for NO2- and excellent resistance to interference. These sensors exhibit clear color changes under both ultraviolet and visible light, and can be combined with smartphones for visual, on-site detection of NO2-. By incorporating a ratiometric strategy, dual-signal output, and smartphone compatibility, our system achieved a low detection limit (≤ 1.92 μM) and satisfactory recovery rates (85.6-115 %) in environmental water and food samples. This highlights the potential of smartphone-assisted sensors for environmental monitoring and food safety applications. Our carbon dot-based platform offers a practical and effective solution for on-site NO2- detection, contributing valuable insights to the field.

Abstract:Nitrite (NO 2 − ) is a widely used food additive and long-term aging of cooked leftovers may also contribute to the formation of NO 2 − , excessive consumption of NO 2 − is harmful to human health. Developing an effective sensing strategy for on-site monitoring of NO 2 − has attracted considerable attention. Herein, a novel colorimetric and fluorometric probe ND-1 based on photoinduced electron transfer effect (PET) was designed for highly selective and sensitive detection of nitrite (NO 2 − ) in foods. The probe ND-1 was strategically constructed by employing naphthalimide as the fluorophore and o -phenylendiamine as the specific recognition site for NO 2 − . The triazole derivative ND-1-NO 2 - could be produced exclusively by reacting with NO 2 − , leading to a visible colorimetric change from yellow to colorless accompanied by a significantly enhanced fluorescence intensity at 440 nm. The probe ND-1 exhibited promising sensing performances towards NO 2 − including high selectivity, rapid response time (within 7 minutes), low detection limit (47.15 nM) and wide quantitative detection range (0–35 μM). In addition, probe ND-1 was capable of quantitative detecting of NO 2 − in real food samples (including pickled vegetables and cured meat products) with satisfactory recovery rates (97.61%–103.08%). Moreover, the paper device loaded by probe ND-1 could be utilized for visual monitoring of NO 2 − levels variation of stir-fried greens. This study provided a feasible method for the accurate, traceable and rapid on-site monitoring NO 2 − in foods.

Abstract:Accurate on-site detection of nitrite in complex matrices remains a significant challenge. Herin, we construct a self-ratio optical bimodal portable kit via co-assembling NaErF4:0.5%Tm@NaYF4@NaYbF4:0.5%Tm@NaYF4 (Er:Tm@Yb:Tm) and nitrogen-doped carbon platinum nanomaterials (Pt/CN) in sodium alginate (SA) hydrogel. Pt/CN nanomaterials are synthesized by high-temperature sintering using a zinc-based zeolite imidazolium framework as a sacrificial template. The Pt/CN nanozyme possesses excellent oxidase-like activity to produce the oxidation state 3,3',5,5'-tetramethylbenzidine (oxTMB). Nitrite mediates diazotization of oxTMB to trigger the change of absorption signals, accompanying the ratio fluorescence response of the Er:Tm@Yb:Tm. Crucially, Er:Tm@Yb:Tm and Pt/CN are embedded in SA hydrogel to fabricate a portable kit with efficient and sensitive performance. An image processing algorithm is used to analyze the nitrite-induced signal change of the portable hydrogel kit, resulting in detection limits of 0.63 μM. This method has great potential for point-of-care applications due to its reliability, long-term stability, accuracy, sensitivity, and portability.

Abstract:Nitrite (NO2-) is categorized as a carcinogenic substance and is subjected to severe limitations in water and food. To safeguard the public's health, developing fast and convenient methods for determination of NO2- is of significance. Point-of-care testing (POCT) affords demotic measurement of NO2- and shows huge potential in future technology beyond those possible with traditional methods. Here, a novel ratiometric fluorescent nanoprobe (Ru@MOF-NH2) is developed by integrating UiO-66-NH2 with tris(2,2'-bipyridyl)ruthenium(II) ([Ru(bpy)3]2+) through a one-pot approach. The special diazo-reaction between the amino group of UiO-66-NH2 and NO2- is responsible for the report signal (blue emission) with high selectivity and the red emission from [Ru(bpy)3]2+ offers the reference signal. The proposed probe shows obviously distinguishable color change from blue to red towards NO2- via naked-eye. Moreover, using a smartphone as the detection device to read color hue, ultra-sensitive quantitative detection of NO2- is achieved with a low limit of detection at 0.6 μΜ. The accuracy and repeatability determined in spiked samples through quantitative visualization is in the range of 105 to 117% with a coefficient of variation below 4.3%. This POCT sensing platform presents a promising strategy for detecting NO2- and expands the potential applications for on-site monitoring in food and environment safety assessment.

Abstract:A liquid-crystal (LC)-based sensor for detecting nitrite in aqueous solutions was developed using a diazotization reaction as the sensing mechanism. First, tetradecyl 4-aminobenzoate (14CBA) was synthesized and doped into a nematic LC, i.e., 4-cyano-4′-pentylbiphenyl (5CB). When the LC mixture was cast on a glass substrate and then immersed into an aqueous solution without nitrite, the orientation of LC was planar and the LC image was bright. In the presence of nitrite, it reacted with alkylanilines to give corresponding diazonium ions with a positive charge, which aligned at the LC/aqueous interface to cause homeotropic orientation of LC. As a result, a bright-to-dark transition of the LC image was observed. The limit of detection (LOD) of this system for nitrite is 25 μM with high selectivity. In addition, this system can work in environmental water samples such as tap water and pond water. Finally, we demonstrated that the optical signals of LC can be measured and recorded using a built-in digital camera of a smartphone, suggesting the portability of this system for on-site applications.This article has not yet been cited by other publications.

Abstract:A new method for the determination of nitrite (NO2–) by multicolor colorimetry and fluorescence dual-mode is presented. This sensing system involves the reaction of crystal violet modulated carbon dots (CV-CDs) with different concentrations of nitrite to produce multiple color signals, while the fluorescence intensity of CV-CDs increases with the increase of nitrite concentration. Therefore, the absorption wavelength shift and fluorescence intensity change can be used for the quantitative determination of nitrite. Furthermore, we combine the multicolor colorimetric output with a smartphone platform to perform on-site determination of nitrite without the need for a spectrometer. Intelligent recognition software was designed to take pictures of color signals and analyze R, G, and B channels. Good results of nitrite analysis were obtained.

Abstract:Nitrite has posed ubiquitous threats to the environment, human health, and public security because of its crucial role in food processing, construction, farming, and explosive manufacturing industries. To achieve accurate, sensitive, and on-site detection of trace nitrite, a colorimetric-fluorescent dual-mode sensing probe with unique photophysical process was designed for ultrasensitive determination of nitrite by precisely adjusting the amino site and significantly influencing the coupling reaction activity. This modulating strategy breaks through the dilemma of azo-product with only colorimetric phenomenon, facilitating the probe, 7-amino-4-methylcoumarin, to maintain a desirable sensitivity with detection limits of 98 nM and 629 nM in fluorescent and colorimetric modes, respectively, as well as an outstanding specificity even in the presence of 15 common anions and interfering substances. The probe was further integrated into a portable sensor by applying a polyvinyl alcohol hydrogel as a loading substrate, which showed superior sensing performances in analyzing nitrite in various real samples with visualized, anti-interferent, and dual-mode responses. Consequently, the present reaction-site modulation strategy would provide a general reference for ultrasensitive, on-site, and visual detection of nitrite, and it can be broadly applied in environmental monitoring and food safety assessment.

Abstract:Nitrite is widely used in production and daily life, and detecting this substance has always been crucial for human health or environment monitoring. Here, a dual-emitting dye@MOF composite, EY@UiO-67-CDC-(CH3)2, has been constructed and demonstrated that this material could detect nitrite ions with highly selective and anti-interference properties. By incorporating an organic anionic dye (Eosin Y) within a cationized EY@UiO-67-CDC-(CH3)2 framework through the post-synthetic method, the composite presents an effective combination due to electrostatic attraction. It features a dual emission effect, which belongs to the blue emission of MOF at 428 nm and yellow emission of EY at 559 nm, respectively. After adding NO2 −, the fluorescence emission peaks of EY and MOF showed opposite changes due to the inner filter effect of NO2 − and MOF, as well as effective excitation of the dye's fluorescence and slight dye leakage. Therefore, Idye/IMOF is used as the detection signal value instead of absolute fluorescence intensity, effectively reducing the detection limit to 0.224 μM and improving its detection accuracy. In addition, during the detection process, the fluorescence sensor showed noticeable color changes, so a smartphone-based analytical device without additional instruments was developed for the quantitative, visual analysis of nitrite by identifying the RGB values of the fluorescence sample solution, with a detection limit of 6.74 μM. The application of the sensor visualizes nitrite detection and reduces the cost of detection, providing a feasible method for on-site detection of nitrite in water samples.

Abstract:In this study, a dual-mode ratiometric fluorometric and colorimetric platform for the determination of nitrite in pickles was proposed by exquisitely employing the fact that non-fluorescent o -Phenylenediamine (OPD) was oxidized by nitrite under acidic conditions to form fluorescent 2,3-diaminophenazine (DAP) (EmвҖү=вҖү575), which meanwhile quench the fluorescent nitrogen-doped carbon dots (N-CDs) at 455В nm, the ratio of fluorescence intensity of DAP to N-CDs (F 575 /F 455 ) changed with the increase of nitrite accompanied by visible color changes. Thus, nitrite can be quantitatively detected within a wide linear range (10вҖ“500 ВөM) with a low detection limit of 0.45 ВөM due to the high quantum yield of 39.7% of N-CDs. In addition, the colour of the N-CDs/OPD system changed from transparent to yellow when the nitrite was introduced, enabling colorimetric and on-site visual detection. The detection limit of the colorimetric method was 3.03 ВөM with a linear range of 10вҖ“500 ВөM. The proposed ratiometric fluorometric method has pleasant selectivity and good immunity to interference.

Abstract:In this paper, a palm-size digital microfluidic (DMF) platform integrated with colorimetric analysis was developed for quantifying the concentration of nitrite. To realize the on-chip repeatable colorimetric analysis, a novel printed circuit board (PCB)-based DMF chip was designed with an embedded aperture on the actuator electrode, forming a vertical light path for online measurement of the droplets. The capabilities of the DMF platform enable automatic manipulation of microliter-level droplets to implement Griess assay without the use of external systems such as syringe, pump, or valve, which provides the benefits including high flexibility, portability, miniature size, and low cost. Results indicated the characteristics of good linearity (R2 = 0.9974), the ignorable crosstalk for reusability, and the limit of detection (LOD) of nitrite as low as 5 μg/L. Furthermore, the presented platform was successfully applied to determine nitrite levels in food products with reliable results and satisfactory recoveries. This integrated DMF platform can be a promising new tool for a wide range of applications involving step-by-step solution mixing and optical detection in environmental monitoring, food safety analysis, and point-of-care testing.This article has not yet been cited by other publications.

Abstract:Developing an effective and convenient nitrite detection method is indispensable in food safety, environmental monitoring, clinical diagnosis of diseases, and many other areas. Herein, a dicyanoisophorone derivative, TMN-NH2 with large Stokes shift and near-infrared (NIR) emission, was proposed as a ratiometric fluorescence and colorimetric dual-mode probe for the rapid determination of NO2- in acidic media, showing excellent selectivity and high sensitivity. The sensing mechanism is based on the diazotization of TMN-NH2 with NO2- and subsequent diazonium salt hydrolysis to form a hydroxyl-substituted product (TMN-OH). Under the optimized conditions of reaction and detection, a new quantitative analysis method based on TMN-NH2 was established for NO2- detection, exhibiting good linear relationships to NO2- in the range of 0.5 to 15 μM with practical detection limits of 26.6 nM and 17.6 nM for the colorimetric and fluorescent readout, respectively. The quantitative detection of NO2- in real samples demonstrated satisfactory recoveries and repeatability. Moreover, TMN-NH2 was successfully applied for monitoring NO2- in Escherichia coli by confocal fluorescence imaging.

Abstract:Nitrite is a typical food additive and preservative used in the food industry, which has attracted considerable attention due to its severe adverse effects on human health. Here, a sensitive and highly selective ratiometric colorimetric sensing platform for the detection of nitrite was created based on a polymetallic oxide nanozyme CoMnO 3 nanofibers (CMO). The nanozyme has superior oxidase-like activity ( K m was 0.105 mM and V max was 63.7 × 10-8 M S -1 ) and could catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to oxidized TMB (oxTMB), which was because that CMO can achieve the conversion of oxygen in solution to superoxide anion (O 2 •- ). In addition, it is interesting to note that oxTMB can be diazotized in the presence of nitrite under acidic condition, causing a shift in the ratio of nitrite concentration to the absorbance peaks at 450 and 652 nm ( A 450 /A 652 ). The ratio of A 450 /A 652 exhibited a positive linear relationship with the concentration of nitrite within the concentration range of 0.2–200 μM, with a detection limit of 0.094 μM. Simultaneously, this method was also successful in quantifying the nitrite produced by brined and pickled foods, as well as dynamic tracking the nitrite levels in various types of dishes. The analysis method based on the polymetallic oxide nanozyme catalysis integrated with the particular diazotization reaction not only offers dual-signal ratio sensing with high sensitivity, but also holds the benefit of outstanding selectivity for the use of the particular reaction, which has a wide range of application prospects in food safety management.

Abstract:A triple-mode colorimetric and fluorescent sensing scheme based on manganese dioxide nanoparticles (MnO2NPs) and carbon quantum dots (CQDs) were developed to determine nitrite. MnO2NPs can oxidize 3,3',5,5'-tetramethylbenzidine (TMB) into a blue oxidation product (TMBox), which is further oxidized into a yellow diimine derivative by nitrite. The ratio of absorbance at 652 nm to 452 nm was monitored as signal response for UV-vis detection mode. A "turn-off" CQDs fluorescence probe was also constructed for fluorescent detection mode. Smartphone tool kit was used to capture the color of sample for smartphone-based measurement. Various analytical performance under different detection modes were obtained and compared. The proposed methods were applied to food samples with satisfactory recoveries (83.3-106 %). The results were validated with AOAC standard spectrophotometric method. The current triple-mode detection were accurate, convenient, low-cost and fast for analyzing nitrite in foods and water samples on-site.

However, excessive addition of NO2- is detrimental to health, so accurate and portable detection of NO2- is critical for food quality control. Notably, the selectivity of most carbon dots (CDs)-based fluorescence sensors was not enough due to the nonspecific interaction mechanism of hydrogen bond, electrostatic interaction and inner filter effect etc. Herein, a novel fluorescence/UV-vis absorption (FL/UV-vis) dual-mode sensor was developed on basis of mC-CDs, which were prepared by simple solvothermal treatment of m-Phenylenediamine (m-PDA) and cyanidin cation (CC). The fluorescence of these mC-CDs could be selectively responded by NO2- through the specific diazo coupling reaction between NO2- and amino groups on the surface of mC-CDs, thus effectively improving the selectivity of NO2- detection. The CDs-based fluorescence sensor possessed a low detection limit of 0.091 μM and 0.143 μM for FL and UV-vis methods and the excellent linear range of 0.0-60.0 μM. Furthermore, the mC-CDs sensor was employed to detect NO2- in real samples with a recovery rate of 97.11 %-104.15 % for quantitative addition. Moreover, the smartphone-assisted fluorescence sensing platform developed could identify the subtle color changes that could not be distinguished by the naked eye, and had the advantages of fast detection speed and intelligence. More importantly, the portable solid phase sensor based on mC-CDs had been successfully applied to the specific fluorescence identification and concentration monitoring of NO2-. Accordingly, the designed sensor provided a new strategy for the highly selective and convenient sensing of NO2- in food substrates, and paved the way for the wide application of CDs-based nanomaterials in the detection of food safety.

Abstract:After fertilization most eggs become surrounded by a complex extracellular matrix. This study examines those matrix assembly processes that are triggered by fertilization of the sea urchin egg. The study uses antibodies that identify five different storage compartments in the egg. These compartments release their protein contents in a highly regulated fashion to assemble and modify the extraembryonic layers. The exocytosis sequence begins with a fertilization wave that progresses from the site of sperm entry and elevates the fertilization envelope above a water-filled perivitelline space. The immediate surface of the zygote then becomes covered by a newly secreted hyaline layer. Prior to fertilization some of the antigens are localized to cortical granules. Others are found in "basal laminar vesicles" that are released in a wave beginning at about 30 sec, or roughly at the same time as cortical granule exocytosis. The remaining antigens are exocytosed with a rather precise timing, but with a delay of several to tens of minutes relative to the first wave of exocytosis. "Apical vesicles," so named because antigens from this class are preferentially exocytosed toward the apical cell surface of polarized cells, include antigens that are exocytosed beginning at about 5 min postfertilization. The fourth compartment, named "echinonectin vesicles" release echinonectin, a protein that is deposited to the inner side of the hyaline layer. Surface staining of echinonectin is first detected about 10-15 min following sperm contact. Finally, maternal cadherin, which is stored in yet a fifth distinct compartment, is not detected on the surface until at least 30 min following fertilization. The data are also consistent with the notion that the tightly regulated timing of exocytosis contributes to the ordered assembly of the hyaline layer and elevation of the fertilization envelope. Finally, two of the vesicle classes continue to exocytose after the cells become polarized. In polarized cells apical and basal laminar antigens are trafficked toward opposite sides of the same cell after passing through the same trans-Golgi network-like compartment.

Abstract:A novel ratiometric fluorescent nanoprobe (Rh6G@UIO-66-NH2) was fabricated for efficient nitrite (NO2-) detection in the present study. When NO2- was introduced, it interacted with the amino groups on the surface of Rh6G@UIO-66-NH2, forming diazonium salts that led to the quenching of blue fluorescence. With this strategy, a good linear relationship between NO2- concentration and the fluorescent intensity ratio of the nanoprobe in the range of 1-100 μM was established, with a detection limit of 0.021 μM. This dual-readout nanosensor was applied to analyze the concentration of NO2- in real meat samples, achieving satisfactory recovery rates of 94.72-104.52%, highlighting the practical potential of this method. Furthermore, a portable Gel/Rh6G@UIO-66-NH2 hydrogel test kit was constructed for on-spot dual-mode detection of NO2-. This kit allows for convenient colorimetric analysis and fluorometric detection when used in conjunction with a smartphone. All the photos taken with the portable kit was converted into digital information using ImageJ software. It provides colorimetric and fluorescent visual detection of NO2- over a range of 0.1-1.5 mM, achieving a direct quantitative tool for NO2- identification. This methodology presents a promising strategy for NO2- detection and expands the application prospects for on-spot monitoring of food safety assessment.
