Abstract:Melatonin is a multi-functional molecule widely employed in order to mitigate abiotic stress factors, in general and salt stress in particular. Even though previous reports revealed that melatonin could exhibit roles in promoting seed germination and protecting plants during various developmental stages of several plant species under salt stress, no reports are available with respect to the regulatory acts of melatonin on the physiological and biochemical status as well as the expression levels of defense- and secondary metabolism-related related transcripts in bitter melon subjected to the salt stress.

Abstract:Melatonin has been found to be crucial in the growth and development of plants under stress conditions. In this study, the effects of melatonin and nano melatonin regarding the growth and development of tomato plants, along with their photosynthetic pigment, phenol, and antioxidant activity, were investigated under saline conditions. The study was conducted using a completely randomized design with three replications, and the applied treatments were salt stress and foliar spraying of melatonin at a concentration of 0 (control), melatonin (Mel), and nano capsule-melatonin (Nano-Mel) at 500 µM. Salinity treatments included application of sodium chloride with two concentration of 0 mM NaCl (S1) and 50 mM NaCl (S2). Under saline conditions, Mel and Nano-Mel increased both shoot and root fresh and dry weights, improved relative water content (RWC), and enhanced antioxidant activity and phenolic content. Salinity elevated leaf ABA content, unaffected by Mel or Nano-Mel. Chlorophyll fluorescence and SPAD values demonstrated resilience to salinity with Mel and Nano-Mel applications. Nano-Mel notably mitigated Na + accumulation in leaves under salinity, helping maintain K + homeostasis. Proline levels rise due to salinity but decreased with Mel and Nano-Mel treatments. Electrolyte leakage (EL) increased under salinity but is significantly reduced by Mel, indicating enhanced membrane stability. The findings reveal that salinity stress significantly reduced plasma membrane intrinsic protein (PIP) expression in roots and leaves, whereas Mel and Nano-Mel treatments enhance PIP expression, particularly in roots. The study concludes that Mel and Nano-Mel effectively alleviate salinity-induced stress, promoting growth and maintaining physiological homeostasis in tomato plants.
Melatonin-Mediated Alleviation of Soil Salinity Stress by Modulation of Redox Reactions and Phytochemical Status in Guar (Cyamopsis tetragonoloba L.)

Abstract:Salinity is one of the major environmental problems threatening crops production, especially in semiarid and arid areas of the world. Melatonin, a growth regulator, has recently been receiving significant attention for its critical role in improving plant salt tolerance. In this experiment, a 2-year field trial was conducted to examine the influence of four exogenous melatonin application rates (0, 100, 200, and 400 μmol L −1 ) on various physiological, biochemical, and yield characteristics of guar plants grown under four different NaCl salinity levels (0, 5, 10, and 15 dS m −1 ) conditions. According to the results, severe salinity stress caused reductions in leaf relative water content (RWC), total chlorophyll, carbohydrates, and phenol contents (by 9.8, 13.6, 42.1, and 25.3%), and stimulated proline level, superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), ascorbate peroxidase (APX), and glutathione reductase (GR) activities (by 54.8, 60.6, 25.1, 51.9, 51.2, and 60.5%) in guar leaves, respectively. In addition, photosynthesis (P n ) and transpiration (T r ) values showed a significant decline with increasing salinity levels up to 45.9 and 77.2%, respectively. However, exogenous melatonin application, particularly at 200 μmol L −1 , relieved physiological impacts of salt stress through increased proline (6–80%), phenol (8–27%), and chlorophyll (7–16%) contents and improved hydraulic functions in guar plants. Furthermore, melatonin-applied plants showed the greatest improvement in yield characteristics including biomass (14–106%), root length (8–13%), seeds per plants (8–36%), and seed gum (11–23%), and maintained higher viscosity and contents of protein and fat in the guar gum relative to untreated plants across four salinity levels. These findings suggested that melatonin improves plant resistance to salinity stress in guar and hence could be utilized to sustain crop yields in various regions affected by soil salinity.

Abstract:Soil salinization is a serious threat to agricultural production because it limits the growth and development of crops. Melatonin is a pleiotropic molecule widely used to alleviate abiotic stressors, including salt stress. The objective of the present study was to investigate the function of melatonin in metabolic processes related to the mitigation of salt-induced growth inhibition in tomato seedlings. The hydroponically grown tomato seedlings were foliar sprayed with 100 μM melatonin and salt (NaCl) treatment was applied in tomato roots for 7 days. The experiment was composed of four treatments: (1) control; (2) melatonin (MT, 100 μM); (3) salt (125 mM NaCl); and melatonin + NaCl (MT + salt, 100 μM + 125 mM NaCl). The results revealed that melatonin treatment significantly mitigated salt-induced adverse effects as evidenced by higher biomass production, greater accumulation of K + , and lower content of Na + in salt-stressed tomato leaves and roots, resulting in lower Na + /K + ratio as compared with only salt-stressed seedlings. Melatonin application enhanced photosynthetic efficiency through the elevation of gas exchange and chlorophyll fluorescence parameters values, along with the increment of carbon assimilation enzymes activity. Moreover, sucrose content and sucrose phosphate synthase (SPS) activity were increased in melatonin-received tomato seedlings over to salt-stressed plants. Melatonin also regulates gene expression related to sugar transporters and carbohydrate metabolism in plants exposed to salinity stress. Additionally, plants treated with melatonin showed higher nitrate reductase and nitrate reductase activity by 78% and 62%, respectively, compared with plants under salt stress. Similarly, the nitrogen assimilation capacity (higher nitrate, NO 3 − level) was increased in melatonin treatment plants. Taken together, the present research findings indicate that melatonin attenuates salinity-caused growth inhibition through the coordination action of carbohydrate and nitrogen metabolism and the balancing of Na + /K + homeostasis in tomato seedlings.

Abstract:Plants face many stresses in their natural environment. Different types of phytohormones help the plant to adapt to stressful conditions. Melatonin is a newly known substance in the plant kingdom, that is effective in relieving stress for plants. In this study, the effect of melatonin on Phaseolus vulgaris L. cv. Pak that was exposed to salinity stress at concentrations of 100 and 200 mM NaCl was investigated. Treatment of stressed plants and control group (0 mM NaCl) with concentrations of 100, 200 μM melatonin improved effects of salinity on the dry weight of shoot, the root and the photosynthetic pigments, net photosynthesis rate, leaf stomatal conductance, transpiration rate, K+, Na+, and Ca2+ content, and the ratio of K+/Na+. Melatonin application increased proline and sugar content while decreasing malondialdehyde and H2O2 content that were increased by salinity stress. Also, melatonin increased the activity of antioxidant enzymes of catalase, peroxidase, ascorbate peroxidase, polyphenol oxidase, and superoxide dismutase. Overall, lower concentrations of melatonin had significant ameliorating effects on salinity stress-induced damage to the bean plant. While, the use of 400 μM melatonin could somewhat exacerbate the effects of salinity in P. vulgaris L. cv. Pak.

Abstract:Salt accumulation in irrigated soils is a worldwide hazard to plants, which adversely reduces crop productivity. Broccoli is considered moderately tolerant to salinity. Melatonin is a multifunctional organic biomolecule with significant importance in plants against abiotic stresses. In this experiment, we studied the role of exogenously applied melatonin on broccoli cultivars under salinity. For this experiment, two popular cultivars (Marathon and Greenport) were selected. Three levels of melatonin (control, 50, & 100 μM) were sprayed on broccoli cultivars exposed to salt stress (0, 40, 80, & 120 mM) and under non-stressed conditions. The results showed that exogenous melatonin application improved morphological characteristics of broccoli, i.e., plant height, the number of leaves, head weight, head diameter, root length, shoot length, leaf relative water content, the number of florets, shoot fresh weight, root fresh, and dry weight. The application of 50-μM melatonin as foliar spray increased TSS, Acidity, Vitamin C, β-carotene, and total phenolics in the broccoli head. Additionally, the foliar spray of melatonin enhanced chlorophyll (a & b) in the head and leaves and reduced antioxidant damage under salinity by enhancing the activity of antioxidant enzymes, such as superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT). Lower malondialdehyde (MDA), hydrogen peroxide (H 2 O 2 ) , and higher Proline and glycine betaine (GB) contents were observed in exogenously applied melatonin than in controlled plants under salinity stress in both cultivars of the broccoli. The exposure to salinity (NaCl) determined an increase in Na + and K + concentrations in plant tissues. However, exogenously applied melatonin reduced the uptake of Na + from the soil. Among the cultivars, Marathon showed more tolerance against salt stress than Greenport. Collectively, our data provide evidence that the exogenous application of melatonin may ameliorate the adverse effects of salinity on broccoli plants.

Abstract:Melatonin, a natural agent, has multiple functions in animals as well as in plants. However, its possible roles in plants under abiotic stress are not clear. Nowadays, soil salinity is a major threat to global agriculture because a high soil salt content causes multiple stresses (hyperosmotic, ionic, and oxidative). Therefore, the aim of the present study was to explore: (1) the involvement of melatonin in biosynthesis of photosynthetic pigments and in regulation of photosynthetic enzymes, such as carbonic anhydrase (CA) and ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco); (2) the role of melatonin in osmoregulation by proline and carbohydrate metabolism; and (3) the function of melatonin in the antioxidant defense system under salinity. Outcomes of the study reveal that under non-saline conditions, application of melatonin (20 and 50 μM) improved plant growth, viz. shoot length, root length, shoot fresh weight (FW), root FW, shoot dry weight (DW), root DW and leaf area and physio-biochemical parameters [chlorophyll (Chl) a and b, proline (Pro) and total soluble carbohydrates (TSC) content, and increased the activity of CA and Rubisco]. However, tomato seedlings treated with NaCl exhibited enhanced Chl degradation, electrolyte leakage (EL), malondialdehyde (MDA) and reactive oxygen species (ROS; superoxide and hydrogen peroxide). ROS were detected in leaf and root. Interestingly, application of melatonin improved plant growth and reduced EL, MDA and ROS levels through upregulation of photosynthesis enzymes (CA, Rubisco), antioxidant enzymes (superoxide dismutase, catalase, glutathione reductase and ascorbate reductase) and levels of non-enzymatic antioxidants [ascorbate (ASC) and reduced glutathione (GSH)], as well as by affecting the ASC—GSH cycle. Additionally, exogenous melatonin also improved osmoregulation by increasing the content of TSC, Pro and Δ1-pyrroline-5-carboxylate synthetase activity. These results suggest that melatonin has beneficial effects on tomato seedlings growth under both stress and non-stress conditions. Melatonin’s role in tolerance to salt stress may be associated with the regulation of enzymes involved in photosynthesis, the antioxidant system, metabolism of proline and carbohydrate, and the ASC—GSH cycle. Also, melatonin could be responsible for maintaining the high ratios of GSH/GSSG and ASC/DHA.

Abstract:Melatonin is a potentially active signaling molecule and plays a crucial role in regulating the growth and development of plants under stress conditions, alleviating oxidative damage, enhancing antioxidant defence mechanisms and regulating ion homeostasis. This study examined the effects of exogenous melatonin application on leaf biomass, ion concentrations, betalains, phenolic acid and endogenous melatonin contents comparing red beet (Beta vulgaris L. 'Ruby Queen' and 'Scarlet Supreme') and white beet ('Rodeo' and 'Ansa') cultivars under increasing salinity levels of 50, 150, and 250 mM NaCl. Exogenous melatonin increased salinity‐induced reductions in fresh and dry weights and osmotic potential in leaves. Na+ concentrations rose significantly with increasing salinity, but cultivar‐specific decreases were observed in K+ and Ca2+ concentrations. Additionally, melatonin application improved betalain, betanin and neobetanin contents induced by salt stress. Furthermore, melatonin application caused salt stress and cultivar‐specific changes in phenolic acid contents e.g., ferulic acid, sinapic acid, or m‐coumaric acid, in soluble free, ester‐ and glycoside‐conjugated and cell wall‐bound forms. In addition, antioxidant enzyme activities and compound contents increased significantly in the beets and were subsequently lowered in a cultivar‐specific manner by salt stress + melatonin treatment. The current findings indicate that exogenous melatonin improved plant stress tolerance suppressing reactive oxygen species levels, increasing the antioxidant enzyme activities and compound contents and reducing the levels of Na+, maintaining an ionic homeostasis in the selected red and white sugar beet cultivars. It appears that melatonin application may help improve cultivar‐specific salt tolerance by enhancing ion homeostasis and betalain and phenolic acid production levels in beets.

Mitigation of salinity impact in spearmint plants through the application of engineered chitosan-melatonin nanoparticles

Abstract:High soil salinity represents a critical environmental constraint to crop production. In order to ameliorate the effects of salinity, a plethora of molecules have been applied and promising outcomes have been noted. The beneficial effects of chitosan (CTS) and melatonin (Mel) application, separately, have been previously recorded with respect to plant growth and productivity, leading to the hypothesis that their conjugation in the form of chitosan-melatonin nanoparticles (CTS-HPMC-Mel NPs) could lead to further enhanced performance of plants under control and stress conditions. In this regard, novel CTS-HPMC-Mel NPs were synthesized, characterized and then employed as a chemical priming agent in spearmint (Mentha spicata L.) plants 24 h prior to salinity stress imposition. As expected, salt stress negatively affected morphophysiological attributes such as plant height, leaf number, leaf fresh weight, leaf dry weight, photosynthetic pigments, Fv/Fo, and Fv/Fm. On the other hand, stress-related attributes, such as content of proline, MDA and H2O2, as well as activity of APX and GP enzymes were increased in response to salt stress. However, adverse effects of salt stress were ameliorated with Mel and CTS-HPMC-Mel NP treatments by enhancing morphological traits, proline, antioxidant enzymatic activities, as well as content of dominant constituents of essential oil profile. It is worth noting that conjugated form of Mel with chitosan, in comparison with solo treatment of Mel, was more effective in combating stress effects. To our knowledge, this is the first report to demonstrate that engineered CTS-HPMC-Mel NPs could be applied as an innovative protective agent to mitigate the effects of salinity in crop plants.
Effects of melatonin on lettuce plant growth, antioxidant enzymes and photosynthetic pigments under salinity stress conditions

Abstract:ABSTRACT Salinity is one of the most important abiotic stresses that significantly decreases the productivity of agricultural crops. Melatonin (MT) acts as an antioxidant and plays a vital role in overcoming oxidative damage. However, previous literature has not provided a clear understanding of the impact of MT on lettuce plants under salinity stress. So, we investigated the effect of exogenous MT at 0 μM, 50 μM, 100 μM and 150 μM on lettuce plants grown under salinity stress (0 mM NaCl, 50 mM NaCl and 100 mM NaCl) with respect to vegetative growth, photosynthetic pigments, relative water content (RWC), electrolyte leakage (EL), malondialdehyde (MDA), H 2 O 2 , O 2 •- and antioxidants enzymes. Results showed that NaCl stress significantly decreased vegetative growth, RWC and photosynthetic pigments and in contrast enhanced dry matter, EL, MDA, H 2 O 2 , O 2 •- , Na + , Cl-, peroxidase (POD), superoxide dismutase (SOD) and glutathione reductase (GR) of lettuce plants compared to non-salinized control. The results demonstrated that under salinity conditions, foliar applications of MT significantly alleviated the harmful effects of salinity and increased number of leaves, leaf area, fresh weight, chlorophyll (a), chlorophyll (b), total chlorophyll, carotenoids and RWC in comparison to untreated plants (control). Meanwhile, dry matter, MDA, H 2 O 2 , O 2 •- , Na + , Cl-, POD, SOD and GR were significantly decreased compared to untreated lettuce plants. In this respect, spraying MT at 150 μM ranked the first, then 100 μM, compared to the lower concentration (50 μM). In conclusion, MT application can be used to alleviate harmful effects of salinity stress.

Abstract:Melatonin (Mel) is a multifunctional signaling molecule, quantified in a wide range of higher plants. It acts as a pleiotropic molecule and is involved in numerous signaling and physiological processes, particularly under harsh environmental conditions in plants. In the present study, the defensive role of Mel was explored in salt (NaCl)-stressed tomato (Solanum lycopersicum) plants. Mel (50 or 100 µM) was supplied to the foliage of tomato plants at 30 days after transplantation (DAT), grown without stress, and in the presence of NaCl (100 or 150 mM) stress. Salt stress negatively affected photosynthetic efficiency, cell viability, plant growth, fruit quality, and impaired root morphology, and stomatal physiology, but induced the accumulation of reactive oxygen species ((H2O2 and O2·−), malondialdehyde (MDA) content, and electrolyte leakage. Interestingly, foliar supplementation of Mel to stressed and non-stressed plants lowered MDA levels, reduced oxidative stress and promoted photosynthesis, improved cell viability, stomatal behavior, and plant growth, and boosted the activity of catalase (CAT), peroxidase (POX), and superoxide dismutase (SOD), and elevated proline content which bestowed endurance and reduced the salt-induced oxidative damage.

Abstract:Soil salinisation is an important abiotic stress faced in grape cultivating, leading to weakened plant vigour and reduced fruit quality. Melatonin as a novel hormone has shown positive exogenous application value. Therefore, this study used wine grape ( Vitis vinifera ) 'Pinot Noir' as a test material to investigate the changes of foliar spraying with different concentrations of melatonin on the physiology and fruit quality of wine grapes in a field under simulated salt stress (200 mmol L −1 NaCl). The results showed that foliar spraying of melatonin significantly increased the intercellular CO 2 concentration, maximum photochemical quantum yield of PSII, relative chlorophyll and ascorbic acid content of the leaves, as well as the single spike weight, 100-grain weight, transverse and longitudinal diameters, malic acid, α-amino nitrogen and ammonia content of fruits, and decreased the initial fluorescence value of leaves, ascorbate peroxidase activity, glutathione content, fruit transverse to longitudinal ratio and tartaric acid content of plants under salt stress. Results of the comprehensive evaluation of the affiliation function indicated that 100 μmol L −1 melatonin treatment had the best effect on reducing salt stress in grapes. In summary, melatonin application could enhance the salt tolerance of grapes by improving the photosynthetic capacity of grape plants under salt stress and promoting fruit development and quality formation, and these results provide new insights into the involvement of melatonin in the improvement of salt tolerance in crop, as well as some theoretical basis for the development and industrialisation of stress-resistant cultivation techniques for wine grapes.

Abstract:Cucumber ( Cucumis sativus L.) is sensitive to the salinization of environment. Soil salinization in facility environment affects the yield and quality of cucumber. Melatonin (N-acetyl-5-methoxytryptamine) is an effective free radical scavenger in plants and could effectively alleviate the abiotic stress injury of plants. In this study, we investigated the effect of different concentrations of exogenous melatonin in mitigating salt stress in cucumber and some underlying molecular mechanisms. The results indicated that 100 μmol/L melatonin-treated plants increased significantly the content of the content of ascorbic acid (ASA), proline, glutathione (GSH), polyphenol and flavonoid, and superoxide dismutase (SOD) and peroxidase (POD) activity, soluble protein and sugars. We utilized RNA-seq technology to analyse the transcriptome profile of leaf tissues under M0 and M2 treatments. The analysis identified 699 differentially expressed genes (DEGs) with 506 up-regulated and 193 down-regulated genes. Most of the transcription factors identified were also involved in plant stress defense. Gene Ontology (GO) enrichment analysis of the DEGs showed significant enrichment of 328 GO terms ( p ≤ 0.05). Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that the DEGs were significantly enriched in 63 KEGG pathways ( p ≤ 0.05). Taken together, these results provided a valuable reference for exploring the mechanism of action of exogenous melatonin in cucumber in saline environment.

Abstract:Melatonin (N-acetyl-5-methoxytryptamine) is an important molecule in plants that regulates growth and development and responds to abiotic stresses. In this study, we studied the role of exogenous melatonin in alleviating salt stress in cucumber. The results showed that exogenous melatonin pretreatment could improve cell viability, protect photosynthesis, increase antioxidant enzyme activity, inhibit active oxygen explosion, reduce malondialdehyde (MDA) content and relative conductivity in cucumber Seedlings under salt stress compared with salt stress alone. Gene expression analysis showed that melatonin significantly increased the expression of antioxidant enzyme gene, nicotinamide adenine dinucleotide phosphate (NADPH) oxidase genes, mitogen-activated protein kinase (MAPK) genes (MAPK3, MAPK4, MAPK6) and salt overly sensitive (SOS) genes (SOS1, SOS2, SOS3) under salt stress. diphenyleneiodonium (DPI) or dimethylthiourea (DMTU) significantly reduced melatonin-induced defense responses, including decreased antioxidant enzyme activity and photosynthesis protection, as well as the expression of stress-related genes. Similar to DPI and DMTU, U0126 (MEK inhibitor) also reduces melatonin-induced defense responses. In conclusion, our study shows that exogenous melatonin treatment can effectively alleviate the damage of salt stress on cucumber seedlings, and the alleviation effect is related to hydrogen peroxide (H2O2). And there may be interactions between melatonin-mediated salt tolerance and the MAP kinase signaling pathway.

Abstract:Autotoxicity and soil salinization are the main causes of continuous cropping obstacles of melon, which often occur at the same time and aggravate with the extension of cultivation years. Melatonin (MT) has a broad application prospect in improving plant resistance. The effects of exogenous MT on seed germination of melon ( Cucumis melo L.) under autotoxicity and saline-alkali combined stress were observed in this paper. Starch and reactive oxygen species metabolism were determined to analyze the physiological and biochemical basis of MT function. Cinnamic acid was used to simulate melon autotoxicity. The mixed solution of NaCl and NaHCO 3 was used to simulate saline-alkali stress. The effect of MT showed a concentration-dependent manner. 16 μM exogenous MT can effectively improve the germination rate, germination potential, and vigor index of melon seeds under combined stress, and increase radicle length and fresh weight. Seed vigor index was the most sensitive index, and MT treatment increased by 102.22% compared with combined stress. However, at high concentration, this stress relief effect can be offset or even reversed. MT can significantly shorten the time of seed breaking through the seed coat and improve the root growth inhibited by stress. MT enhanced α-amylase and β-amylase gene expression, antioxidant enzyme, and amylase activity. The contents of starch and soluble sugar changed accordingly. At the same time, the contents of malondialdehyde and reactive oxygen species were reduced. The expression analysis of 10 MT synthesis genes showed that MT treatment up-regulated eight genes. In general, by up-regulating the expression of amylase and some MT synthesis genes, enhancing the activities of antioxidant enzymes and amylase, reducing the content of malondialdehyde and reactive oxygen species, exogenous MT effectively increased the germination rate and germination potential of melon seeds under autotoxicity and saline-alkali combined stress, and significantly improved the quality of seed germination.

Abstract:Melatonin is a ubiquitous signaling plant hormone that plays a crucial role in regulating the growth and development of plants under stress conditions. Since a few species have been investigated to unveil the effect of exogenous melatonin on salt stress, in the current research study, we investigated the effects of melatonin by measuring the photosynthetic characteristics and antioxidant defense system of maize seedling under salt stress (200 mM NaCl), along with different concentrations of melatonin (MT1—30, MT2–60, and MT3–90 µM) treatments. Salt stress reduced the plant growth characteristics and photosynthetic efficiency by increasing the ROS accumulation and reducing the antioxidant enzyme activities of maize seedling. However, pretreatment with melatonin on roots alleviated NaCl-induced decrease in photosynthetic rate and oxidative stress in a dose-dependent manner. Our results revealed that exogenous application of melatonin at an optimum concentration (60 µM) under salt stress conditions significantly increased the growth of plants, chlorophyll content, photosynthetic efficiency, antioxidant enzymes activities, i.e., superoxide dismutase (SOD), peroxide (POD), catalase (CAT) and ascorbate peroxidase (APX), and reduced the reactive oxygen species (ROS). This study unraveled the crucial role of melatonin in salt stress mitigation and thus can be implicated in the management of salinity in maize seedling.

Abstract:Abstract Background Soil salinization is becoming an increasingly serious problem worldwide, resulting in cultivated land loss and desertification, as well as having a serious impact on agriculture and the economy. The indoleamine melatonin (N-acetyl-5-methoxytryptamine) has a wide array of biological roles in plants, including acting as an auxin analog and an antioxidant. Previous studies have shown that exogenous melatonin application alleviates the salt-induced growth inhibition in non-halophyte plants; however, to our knowledge, melatonin effects have not been examined on halophytes, and it is unclear whether melatonin provides similar protection to salt-exposed halophytic plants. Results We exposed the halophyte Limonium bicolor to salt stress (300 mM) and concomitantly treated the plants with 5 μM melatonin to examine the effect of melatonin on salt tolerance. Exogenous melatonin treatment promoted the growth of L. bicolor under salt stress, as reflected by increasing its fresh weight and leaf area. This increased growth was caused by an increase in net photosynthetic rate and water use efficiency. Treatment of salt-stressed L. bicolor seedlings with 5 μM melatonin also enhanced the activities of antioxidants (superoxide dismutase [SOD], peroxidase [POD], catalase [CAT], and ascorbate peroxidase [APX]), while significantly decreasing the contents of hydrogen peroxide (H 2 O 2 ), superoxide anion (O 2 •− ), and malondialdehyde (MDA). To screen for L. bicolor genes involved in the above physiological processes, high-throughput RNA sequencing was conducted. A gene ontology enrichment analysis indicated that genes related to photosynthesis, reactive oxygen species scavenging, the auxin-dependent signaling pathway and mitogen-activated protein kinase ( MAPK ) were highly expressed under melatonin treatment. These data indicated that melatonin improved photosynthesis, decreased reactive oxygen species (ROS) and activated MAPK-mediated antioxidant responses, triggering a downstream MAPK cascade that upregulated the expression of antioxidant-related genes. Thus, melatonin improves the salt tolerance of L. bicolor by increasing photosynthesis and improving cellular redox homeostasis under salt stress. Conclusions Our results showed that melatonin can upregulate the expression of genes related to photosynthesis, reactive oxygen species scavenging and mitogen-activated protein kinase ( MAPK ) of L. bicolor under salt stress, which can improve photosynthesis and antioxidant enzyme activities. Thus melatonin can promote the growth of the species and maintain the homeostasis of reactive oxygen species to alleviate salt stress.

Abstract:Root systems are the key organs through which plants absorb water and nutrients and perceive the soil environment and thus are easily damaged by salt stress. Melatonin can alleviate stress-induced damage to roots. The present study investigated the effects of exogenous melatonin on the root physiology, transcriptome and metabolome of cotton seedlings under salt stress. Salt stress was observed to damage the cell structure and disorder the physiological system of cotton seedling roots. After subjecting melatonin-soaked seeds to salt stress, the activities of SOD, CAT and POD in cotton seedling roots increased by 10–25%, 50–60% and 50–60%, respectively. The accumulation of H2O2 and MDA were significantly decreased by 30–60% and 30–50%, respectively. The contents of soluble sugar, soluble protein and K+ increased by 15–30%, 15–30% and 20–50%, respectively, while the Na+ content was significantly reduced. Melatonin also increased auxin (by 20–40%), brassinosteroids (by 5–40%) and gibberellin (by 5–35%) and promoted melatonin content and root activity. Exogenous melatonin maintained the integrity of root cells and increased the number of organelles. Transcriptomic and metabolomic results showed that exogenous melatonin could mitigate the salt-stress-induced inhibition of plant root development by regulating the reactive oxygen species scavenging system; ABC transporter synthesis; plant hormone signal transduction, endogenous melatonin gene expression; and the expression of the transcription factors MYB, TGA and WRKY33. These results provide a new direction and empirical basis for improving crop salt tolerance with melatonin.

Abstract:Melatonin (N-acetyl-5-methoxytryptamine) as a natural biostimulating substance provides a number of benefits in stimulating plant growth in stress situations due to its natural antioxidant capacity. Rhizobia also play crucial roles in supporting plant growth under environmental stress conditions. The overall goal of this research is to study the possible positive effects of melatonin and rhizobacterium in enhancing the growth and salinity tolerance of common bean. To accomplish this objective, we conducted in vitro experiment to select the optimal melatonin concentration and treatment time of seed priming for the best germination. Also, a greenhouse experiment was performed to investigate the effect of melatonin pre-treatment applied before rhizobial inoculation to improve the fitness of common bean under salinity stress. The experiment was conducted using a completely randomized factorial design with six replications and three treatments: priming treatments (melatonin priming (PM100), hydro priming (PH) and dry (PD)), salinity (0, 4, 8, 10 and 16 dS m−1) and Rhizobium strain (inoculated (RS +) and uninoculated (RS−)). Our results showed that melatonin priming promoted bacterial colony size in Petri-dishes. The interactive effects of melatonin and RS + was found to alleviate reactive oxygen species (ROS) burst, and hence protect common bean chlorophylls a, b and carotenoid and photosynthetic activity and decrease malondialdehyde content through activation of antioxidant enzymes (superoxide dismutase, peroxidase, catalase and ascorbate peroxidase), facilitation of soluble protein synthesis, maintenance of Na+ and K+ homeostasis, and finally increase shoot dry weight (33.2, 39.5 and 31.5%) and seed yield (78.6, 91 and 54.2%) compared to the combination of PD and RS- treatments under 0, 4 and 8 dS m−1 salinity levels, respectively. Thus, our findings suggest that seed priming with melatonin, especially 100 µM melatonin is an effective strategy that can be used to enhance salt tolerance in common bean.

Abstract:Salt stress severely reduces growth and yield of plants. Considering the positive effects of selenium (Se) and chitosan (Cs) separately against abiotic stress, in these experiments, we synthesized chitosan–selenium nanoparticles (Cs–Se NPs) and investigated their ability to reduce the negative effects of salt stress on growth and some biochemical parameters of bitter melon (Momordica charantia). Bitter melon plants were grown at three NaCl salinity levels (0, 50, and 100 mM) and a foliar spray of Cs–Se NPs (0, 10, and 20 mg L−1) was applied. Some key morphological, biochemical, and physiological parameters in leaf samples and essential oil from fruit were measured at harvest. Salinity decreased growth and yield while foliar application of Cs–Se NPs increased these critical parameters. Furthermore, Cs–Se NPs enhanced bitter melon tolerance to salinity by increasing antioxidant enzyme activity, proline concentration, relative water content, and K+, and decreasing MDA and H2O2 oxidants and Na aggregation in plant tissues. Yield was also improved, as the highest amount of essential oils was produced by plants treated with Cs–Se NPs. Generally, the greatest improvement in measured parameters under saline conditions was obtained by treating plants with 20 mg L−1 Cs–Se NPs, which significantly increased salinity tolerance in bitter melon plants.
