Abstract:Maize is an important cereal crop worldwide and is sensitive to abiotic stresses in fluctuant environments that seriously affect its growth, yield, and quality. The small heat shock protein (HSP20) plays a crucial role in protecting plants from abiotic stress. However, little is known about HSP20 in maize (ZmHSP20). In this study, 44 ZmHSP20s were identified, which were unequally distributed over 10 chromosomes, and 6 pairs of ZmHSP20s were tandemly presented. The gene structure of ZmHSP20s was highly conserved, with 95% (42) of the genes having no more than one intron. The analysis of the cis-element in ZmHSP20s promoter demonstrated large amounts of elements related to hormonal and abiotic stress responses, including abscisic acid (ABA), high temperature, and hypoxia. The ZmHSP20s protein had more than two conserved motifs that were predictably localized in the cytoplasm, nucleus, endoplasmic reticulum, peroxisome, mitochondria, and plasma. Phylogenetic analysis using HSP20s in Arabidopsis, rice, maize, and Solanum tuberosum indicated that ZmHSP20s were classified into 11 categories, of which each category had unique subcellular localization. Approximately 80% (35) of ZmHSP20 were upregulated under heat stress at the maize seedling stage, whereas the opposite expression profiling of 10 genes under 37 and 48 °C was detected. A total of 20 genes were randomly selected to investigate their expression under treatments of ABA, gibberellin (GA), ethylene, low temperature, drought, and waterlogging, and the results displayed that more than half of these genes were downregulated while ZmHSP20-3, ZmHSP20-7, ZmHSP20-24, and ZmHSP20-44 were upregulated under 1 h treatment of ethylene. A yeast-one-hybrid experiment was conducted to analyze the binding of four heat stress transcription factors (ZmHSFs) with eight of the ZmHSP20s promoter sequences, in which ZmHSF3, ZmHSF13, and ZmHSF17 can bind to most of these selected ZmHSP20s promoters. Our results provided a valuable resource for studying HSP20s function and offering candidates for genetic improvement under abiotic stress.
Pan-Genome-Wide Investigation and Co-Expression Network Analysis of HSP20 Gene Family in Maize

Abstract:Heat shock protein 20 (HSP20) is a diverse and functionally important protein family that plays a crucial role in plants' tolerance to various abiotic stresses. In this study, we systematically analyzed the structural and functional characteristics of the HSP20 gene family within the Zea pan-genome. By identifying 56 HSP20 pan-genes, we revealed the variation in the number of these genes across different maize inbreds or relatives. Among those 56 genes, only 31 are present in more than 52 inbreds or relatives. Further phylogenetic analysis classified these genes into four major groups (Class A, B, C, D) and explored their diversity in subcellular localization, physicochemical properties, and the terminal structures of those HSP20s. Through collinearity analysis and Ka/Ks ratio calculations, we found that most HSP20 genes underwent purifying selection during maize domestication, although a few genes showed signs of positive selection pressure. Additionally, expression analysis showed that several HSP20 genes were significantly upregulated under high temperatures, particularly in tassels and leaves. Co-expression network analysis revealed that HSP20 genes were significantly enriched in GO terms related to environmental stress responses, suggesting that HSP20 genes not only play key roles in heat stress but may also be involved in regulating various other biological processes, such as secondary metabolism and developmental processes. These findings expand our understanding of the functions of the maize HSP20 family and provide new insights for further research into maize's response mechanisms to environmental stresses.

Abstract:Abstract Heat shock transcription factor (Hsf) plays a transcriptional regulatory role in plants during heat stress and other abiotic stresses. 31 non-redundant ZmHsf genes from maize were identified and clustered in the reference genome sequenced by Single Molecule Real Time (SMRT). The amino acid length, chromosome location, and presence of functional domains and motifs of all ZmHsfs sequences were analyzed and determined. Phylogenetics and collinearity analyses reveal gene duplication events in Hsf family and collinearity blocks shared by maize, rice and sorghum. The results of RNA-Seq analysis of anthesis and post-anthesis periods in maize show different expression patterns of ZmHsf family members. Specially, ZmHsf26 of A2 subclass and ZmHsf23 of A6 subclass were distinctly up-regulated after heat shock (HS) at post-anthesis stage. Nanopore transcriptome sequencing of maize seedlings showed that alternative splicing (AS) events occur in ZmHsf04 and ZmHsf17 which belong to subclass A2 after heat shock. Through sequence alignment, semi-quantitative and quantitative RT-PCR, we found that intron retention events occur in response to heat shock, and newly splice isoforms, ZmHsf04-II and ZmHsf17-II , were transcribed. Both new isoforms contain several premature termination codons in their introns which may lead to early termination of translation. The ZmHsf04 expression was highly increased than that of ZmHsf17 , and the up-regulation of ZmHsf04-I transcription level were significantly higher than that of ZmHsf04-II after HS.

Characterization of Small Heat Shock Proteins Associated with Maize Tolerance to Combined Drought and Heat Stress

Abstract:To investigate how the mechanisms of small heat shock proteins (sHSPs) in regulating maize leaves respond to the combination of drought and heat stress, leaf protein patterns were monitored using a proteomic approach in maize plants exposed to combined drought and heat stress. Two-dimensional electrophoresis (2-DE) was used to identify combined drought- and heat-responsive protein spots in maize leaves. After Coomassie brilliant blue staining, approximately 450 protein spots were reproducibly detected on each gel, wherein 7 protein spots were expressed only under heat and combined drought and heat stress but were almost undetected under control and drought. Using MALDI-TOF mass spectrometry, a total of seven proteins were identified, including cytochrome b6-f complex iron-sulfur subunit, sHSP17.4, sHSP17.2, sHSP26, guanine nucleotide-binding protein β-subunit-like protein, putative uncharacterized protein, and granule-bound starch synthase IIa. Moreover, the gene expression of three sHSPs was analyzed at the transcriptional level and indicated that all three sHSPs were expressed under several treatments although their expression levels were obviously more enhanced by heat and combined drought and heat stress than by control and drought. In investigations of the effect of abscisic acid (ABA) on the three sHSPs, pretreatment with 100 μM ABA enhanced substantially the expression of the three sHSPs at the protein level, but only slightly at the mRNA level. These results show that transcription levels are not completely concomitant with translation and suggest that ABA induces the post-transcriptional regulation of sHSP17.2, sHSP17.4, and sHSP26 expression, which can lead to a better understanding of the mechanisms of plant response to the combination of drought and heat stress.

Abstract:Heat stress has become one of the abiotic stresses that pose an increasing threat to maize production due to global warming. The Hsp20 gene family confers tolerance to various abiotic stresses in plants. However, very few Hsp20 s have been identified in relation to maize thermotolerance. In this study, we conducted a comprehensive study of Hsp20 s involved in thermotolerance in maize. A total of 33 maize Hsp20 genes (Zm Hsp20s ) were identified through scanning for a conserved α-crystalline domain (ACD), and they were categorized into 14 subfamilies based on phylogenetic analysis. These genes are distributed across all maize chromosomes and nine of them are in regions previously identified as heat-tolerance quantitative trait loci (hrQTL). These hrQTL-associated Zm Hsp20s show variation in tissue-specific expression profiles under normal conditions, and seven of them possess 1–5 heat stress elements in their promoters. The integration of RNA-seq data with real-time RT-PCR analysis indicated that ZmHsp23.4 , ZmHsp22.8B and ZmHsp18 were dramatically induced under heat stress. Additionally, these genes exhibited co-expression patterns with key ZmHsfs , which are crucial in the heat tolerance pathway. When a null mutant carrying a frame-shifted ZmHsp18 gene was subjected to heat stress, its survival rate decreased significantly, indicating a critical role of ZmHsp18 in maize thermotolerance. Our study lays the groundwork for further research into the roles of ZmHsp20s in enhancing maize's thermotolerance.

Abstract:Abiotic stresses substantially threaten global crop productivity, triggering a proactive response in plants to anticipate and counter temperature fluctuations. In this adaptive strategy, plants accumulate heat-shock proteins (HSPs) as a crucial mechanism to acquire tolerance for abiotic stress. These specialized proteins function as molecular chaperones, playing a pivotal role in stabilizing cellular proteins and maintaining cellular homeostasis. Their significance extends across diverse organisms, emphasizing the evolutionary conservation of HSPs. Within the realm of plant biology, various classes of HSPs, such as HSP60, HSP70, and HSP100, exhibit distinct roles in cellular processes and influence everything from mitochondrial functions to chloroplast development. Recent advancements in understanding the crucial role of HSPs in abiotic stress tolerance are the focus of this review. Our data mining analysis consisting of 300 published manuscripts in recent years on HSP expression under abiotic stress conditions suggests small HSPs are most abundant, followed by HSP70s. HSPs are most responsive to heat stress (HS) and respond minimally to flood and cold stress. Studies have unveiled the multifaceted involvement of HSPs in stress response and the intricate regulation of tolerance mechanisms. As climate change intensifies, crops demonstrate a specific expression of HSPs to effectively cope with diverse stressors, including elevated temperature and drought. The review also delves into ongoing research exploring the potential of molecular markers and epigenetics in breeding programs, aiming to enhance stress tolerance in crops. Integrating omics data provides valuable insights for genetic modification, promising improved resilience in plants facing environmental challenges. In the context of rising global temperatures, a nuanced understanding of the intricate roles played by HSPs becomes crucial. Implementing advanced technologies, informed by this understanding, is imperative for safeguarding agricultural productivity against the evolving backdrop of environmental challenges.

The alternative splicing of ZmHsf23 regulates heat stress tolerance in maize

Abstract:Heat stress is one of the major threats to maize (Zea mays) production globally. Heat shock transcription factors (HSFs) play vital roles in plant heat stress responses. However, the molecular and genetic basis of HSFs in maize thermotolerance remain largely unknown. In this study, we reveal that the alternative splicing of Hsf23 in maize modulates heat stress tolerance. Hsf23 produces two functional transcripts, the full-length transcript Hsf23b and the heat-inducible transcript Hsf23a. The two Hsf23 transcripts differ by the presence of a cryptic mini exon in Hsf23a, which is spliced out in Hsf23b. Both Hsf23a and Hsf23b were intensely expressed in response to heat stress. The overexpression of Hsf23b, not Hsf23a, enhanced heat stress tolerance, while loss-of-function mutations of Hsf23a and Hsf23b exhibited remarkably increased sensitivity to heat stress. Transcriptome analysis revealed that Hsf23b activates broader heat-responsive genes than Hsf23a, and Hsf23a and Hsf23b modulate heat stress response through different downstream targets. Furthermore, Hsf23a physically interacted with Hsf23b and promotes Hsf23b-regulated expression of sHSP genes. Together, our finding provides new insights into the roles of ZmHsf23 in the heat tolerance in maize, and presents an important candidate for the genetic improvement of heat-tolerant maize varieties.

Abstract:Heat shock proteins (Hsps), acting as molecular chaperones, play a pivotal role in plant responses to environmental stress. In this study, we found a total of 192 genes encoding Hsps, which are distributed across all 12 chromosomes, with higher concentrations on chromosomes 1, 2, 3, and 5. These Hsps can be divided into six subfamilies (sHsp, Hsp40, Hsp60, Hsp70, Hsp90, and Hsp100) based on molecular weight and homology. Expression pattern data indicated that these Hsp genes can be categorized into three groups: generally high expression in almost all tissues, high tissue-specific expression, and low expression in all tissues. Further analysis of 15 representative genes found that the expression of 14 Hsp genes was upregulated by high temperatures. Subcellular localization analysis revealed seven proteins localized to the endoplasmic reticulum, while others localized to the mitochondria, chloroplasts, and nucleus. We successfully obtained the knockout mutants of above 15 Hsps by the CRISPR/Cas9 gene editing system. Under natural high-temperature conditions, the mutants of eight Hsps showed reduced yield mainly due to the seed setting rate or grain weight. Moreover, the rice quality of most of these mutants also changed, including increased grain chalkiness, decreased amylose content, and elevated total protein content, and the expressions of starch metabolism-related genes in the endosperm of these mutants were disturbed compared to the wild type under natural high-temperature conditions. In conclusion, our study provided new insights into the HSP gene family and found that it plays an important role in the formation of rice quality and yield.

Genome-wide identification and analysis of biotic and abiotic stress regulation of small heat shock protein (HSP20) family genes in bread wheat

Abstract:Small Heat Shock Proteins (sHSPs)/HSP20 are molecular chaperones that protect plants by preventing protein aggregation during abiotic stress conditions, especially heat stress. Due to global climate change, high temperature is emerging as a major threat to wheat productivity. Thus, the identification of HSP20 and analysis of HSP transcriptional regulation under different abiotic stresses in wheat would help in understanding the role of these proteins in abiotic stress tolerance. We used sequences of known rice and Arabidopsis HSP20 HMM profiles as queries against publicly available wheat genome and wheat full length cDNA databases (TriFLDB) to identify the respective orthologues from wheat. 163 TaHSP20 (including 109 sHSP and 54 ACD) genes were identified and classified according to the sub-cellular localization and phylogenetic relationship with sequenced grass genomes (Oryza sativa, Sorghum bicolor, Zea mays, Brachypodium distachyon and Setaria italica). Spatio-temporal, biotic and abiotic stress-specific expression patterns in normalized RNA seq and wheat array datasets revealed constitutive as well as inductive responses of HSP20 in different tissues and developmental stages of wheat. Promoter analysis of TaHSP20 genes showed the presence of tissue-specific, biotic, abiotic, light-responsive, circadian and cell cycle-responsive cis-regulatory elements. 14 TaHSP20 family genes were under the regulation of 8 TamiRNA genes. The expression levels of twelve HSP20 genes were studied under abiotic stress conditions in the drought- and heat-tolerant wheat genotype C306. Of the 13 TaHSP20 genes, TaHSP16.9H-CI showed high constitutive expression with upregulation only under salt stress. Both heat and salt stresses upregulated the expression of TaHSP17.4-CI, TaHSP17.7A-CI, TaHSP19.1-CIII, TaACD20.0B-CII and TaACD20.6C-CIV, while TaHSP23.7-MTI was specifically induced only under heat stress. Our results showed that the identified TaHSP20 genes play an important role under different abiotic stress conditions. Thus, the results illustrate the complexity of the TaHSP20 gene family and its stress regulation in wheat, and suggest that sHSPs as attractive breeding targets for improvement of the heat tolerance of wheat.
Small heat shock protein (Hsp20) gene family in Phaseolus vulgaris L.: Genome-wide identification, evolutionary and expression analysis

Abstract:Understanding the mechanisms responsible for plant tolerance under various abiotic stresses is linked to understanding heat shock proteins (Hsps). The Hsp20 subfamily is the main family of heat shock proteins, but little is known about this family in common bean (Phaseolus vulgaris L.), an important agricultural crop sensitive to temperature and salt stress. We identified 41 candidate common bean Hsp20 genes (PvHsp20). We mapped the PvHsp20 genes according to their chromosomal locations and found 49 duplications, 35 segmental and 14 tandem. Subsequent gene structure analysis identified regions of exons, introns, and conserved motifs. In the phylogenetic analysis, all PvHsp20 genes were grouped into 11 subfamilies, except for two genes that could not be classified. Synteny analysis revealed that Hsp20 genes from the common bean are highly related to soybean. Promoter analysis of PvHsp20 genes showed the presence of hormone-related, light-responsive and stress-responsive cis-regulatory elements. 8 PvHsp20 genes were under the regulation of 8 PvmiRNA genes. The in silico expression profile showed that PvHsp20 was differentially expressed. The gene expression of the PvHsp20-31, PvHsp20-27, PvHsp-2, and PvHsp20-39 genes in the roots of the Ispir genotype suggested that these genes might play a vital role in the salt tolerance mechanism. Our results provide new information, contribute to understanding PvHsp20 genes, and validate their role in mitigating the adverse effects triggered by abiotic stresses.
The Heat shock factor 20–HSF4–Cellulose synthase A2 module regulates heat stress tolerance in maize

Abstract:Temperature shapes the geographical distribution and behavior of plants. Understanding the regulatory mechanisms underlying the plant heat response is important for developing climate-resilient crops, including maize (Zea mays). To identify transcription factors that may contribute to the maize heat response, we generated a dataset of short- and long-term transcriptome changes following a heat treatment time course in the inbred line B73. Co-expression network analysis highlighted several transcription factors, including the class B2a heat shock factor (HSF) ZmHSF20. Zmhsf20 mutant seedlings exhibited enhanced tolerance to heat stress. Furthermore, DNA affinity purification sequencing and Cleavage Under Targets and Tagmentation (CUT&Tag) assays demonstrated that ZmHSF20 binds to the promoters of Cellulose synthase A2 (ZmCesA2) and three class A Hsf genes, including ZmHsf4, repressing their transcription. We showed that ZmCesA2 and ZmHSF4 promote the heat response, with ZmHSF4 directly activating ZmCesA2 transcription. In agreement with the transcriptome analysis, ZmHSF20 inhibited cellulose accumulation and repressed the expression of cell wall–related genes. Importantly, the Zmhsf20 Zmhsf4 double mutant exhibited decreased thermotolerance, placing ZmHsf4 downstream of ZmHsf20. We proposed an expanded model of the heat stress response in maize, whereby ZmHSF20 lowers seedling heat tolerance by repressing ZmHsf4 and ZmCesA2, thus balancing seedling growth and defense.

Abstract:Abstract Background Common bean ( Phaseolus vulgaris ) is an essential crop with high economic value. The growth of this plant is sensitive to environmental stress. Heat shock factor (Hsf) is a family of antiretroviral transcription factors that regulate plant defense system against biotic and abiotic stress. To date, few studies have identified and bio-analyzed Hsfs in common bean. Results In this study, 30 Hsf transcription factors (PvHsf1–30) were identified from the PFAM database. The PvHsf1–30 belonged to 14 subfamilies with similar motifs, gene structure and cis -acting elements. The Hsf members in Arabidopsis , rice ( Oryza sativa ), maize ( Zea mays ) and common bean were classified into 14 subfamilies. Collinearity analysis showed that PvHsfs played a role in the regulation of responses to abiotic stress. The expression of PvHsfs varied across different tissues. Moreover, quantitative real-time PCR (qRT-PCR) revealed that most PvHsfs were differentially expressed under cold, heat, salt and heavy metal stress, indicating that PvHsfs might play different functions depending on the type of abiotic stress. Conclusions In this study, we identified 30 Hsf transcription factors and determined their location, motifs, gene structure, cis -elements, collinearity and expression patterns. It was found that PvHsfs regulates responses to abiotic stress in common bean. Thus, this study provides a basis for further analysis of the function of PvHsfs in the regulation of abiotic stress in common bean.

A Novel Heat Shock Transcription Factor (ZmHsf08) Negatively Regulates Salt and Drought Stress Responses in Maize

Abstract:Heat shock transcription factors (HSFs) play important roles in plant growth, development, and stress responses. However, the function of these transcription factors in abiotic stress responses in maize (Zea mays) remains largely unknown. In this study, we characterized a novel HSF transcription factor gene, ZmHsf08, from maize. ZmHsf08 was highly homologous to SbHsfB1, BdHsfB1, and OsHsfB1, and has no transcriptional activation activity. The expression profiles demonstrated that ZmHsf08 was differentially expressed in various organs of maize and was induced by salt, drought, and abscisic acid (ABA) treatment. Moreover, the overexpression of ZmHsf08 in maize resulted in enhanced sensitivity to salt and drought stresses, displaying lower survival rates, higher reactive oxygen species (ROS) levels, and increased malondialdehyde (MDA) contents compared with wild-type (WT) plants. Furthermore, RT-qPCR analyses revealed that ZmHsf08 negatively regulates a number of stress/ABA-responsive genes under salt and drought stress conditions. Collectively, these results indicate that ZmHsf08 plays a negative role in response to salt and drought stresses in maize.

Abstract:In our previous study, 25 Hsf gene homologues were identified and analyzed in maize. However, the exact functions of these gene family members are unclear at present. Here, we report the isolation and characterization of a strongly heat-induced HsfA2 gene from maize, ZmHsf04, and show its function in the heat/salt-stress response. Subcellular location assays showed that the ZmHsf04 protein is localized to the nucleus. Transcription activation data demonstrate that an AHA2 domain protein is necessary for the transcriptional activity of ZmHsf04. Furthermore, the function of ZmHsf04 was characterized in detail in transgenic plants of Arabidopsis thaliana Col-0. Overexpression of ZmHsf04 in Arabidopsis enhanced thermotolerance and positively regulated short-term-acquired thermotolerance. In addition, ZmHsf04 significantly improved salt tolerance and increased the sensitivity to abscisic acid in transgenic Arabidopsis. Overexpression of ZmHsf04 in Col-0 plants significantly up-regulated the expression of heat-specific HSP genes (AtHsp25.3-P, AtHsp18.2-CI, and AtHsp70B) and stress-related genes (AtAPX2 and AtGolS1) compared to the wild type. In summary, we have characterized a transcriptional regulator, ZmHsf04, which has important functions in enhancing heat and salt tolerance in plants.

Abstract:DnaJ, or heat shock protein 40 (HSP40), is a conserved protein that functions as a molecular chaperone with a strong cytoprotective effect, participating in plant signal transduction, growth, development, and the response to heat stress and other adverse environmental conditions. Although functional characteristics of DnaJ/HSP40 have been described in several plant species, the underlying mechanisms are not completely understood. In this study, 99 putative maize ( Zea mays L.) DnaJ genes ( ZmDnaJ01–99 ) randomly distributed on 10 chromosomes were identified in the whole genome using bioinformatics approaches and classified into three groups according to protein structure. The conservation of the gene structure was closely related to the evolutionary tree. Collinearity analysis of DnaJ family genes among species revealed a close evolutionary relationship between maize and sorghum. Transposon analysis revealed helitrons inserted in 49 ZmDnaJs. Multiple stress-related cis -elements were found in the promoter regions of ZmDnaJ genes, suggesting a role in gene expression regulation in response to abiotic stress. The co-expression network screened 12 ZmDnaJ genes that could be induced by stress (drought, high temperature, and salt) and regulated by abscisic acid. ZmDnaJ96 overexpression increased antioxidant enzyme activity and reduced the damage caused by drought and high-temperature stress in Arabidopsis chloroplasts. Virus-induced silencing of ZmDnaJ 96 reduced drought and heat tolerance in maize by reducing antioxidant enzyme activity. These results provide genetic resources for maize stress resistance molecular breeding, enhance knowledge of DnaJ gene function in maize, and offer a solid foundation for further in-depth study of the relationship between maize DnaJ / HSP40 members.

Comprehensive Identification and Functional Analysis of Stress-Associated Protein (SAP) Genes in Osmotic Stress in Maize

Abstract:Stress-associated proteins (SAPs) are a kind of zinc finger protein with an A20/AN1 domain and contribute to plants' adaption to various abiotic and biological stimuli. However, little is known about the SAP genes in maize (Zea mays L.). In the present study, the SAP genes were identified from the maize genome. Subsequently, the protein properties, gene structure and duplication, chromosomal location, and cis-acting elements were analyzed by bioinformatic methods. Finally, their expression profiles under osmotic stresses, including drought and salinity, as well as ABA, and overexpression in Saccharomyces cerevisiae W303a cells, were performed to uncover the potential function. The results showed that a total of 10 SAP genes were identified and named ZmSAP1 to ZmSAP10 in maize, which was unevenly distributed on six of the ten maize chromosomes. The ZmSAP1, ZmSAP4, ZmSAP5, ZmSAP6, ZmSAP7, ZmSAP8 and ZmSAP10 had an A20 domain at N terminus and AN1 domain at C terminus, respectively. Only ZmSAP2 possessed a single AN1 domain at the N terminus. ZmSAP3 and ZmSAP9 both contained two AN1 domains without an A20 domain. Most ZmSAP genes lost introns and had abundant stress- and hormone-responsive cis-elements in their promoter region. The results of quantitative real-time PCR showed that all ZmSAP genes were regulated by drought and saline stresses, as well as ABA induction. Moreover, heterologous expression of ZmSAP2 and ZmSAP7 significantly improved the saline tolerance of yeast cells. The study provides insights into further underlying the function of ZmSAPs in regulating stress response in maize.

[Effect of gum guar in the diet on the utilization of 14C-labeled protein and protein hydrolysates in young rats].

Abstract:Beginning on the 25th day of life and for 2 weeks therafter Sprague-Dawley rats were fed a casein diet containing 10% of gum guar. The animals were given. 14C algal protein or 14C algal protein hydrolysate by stomach tube and the radioactivity in some tissues and tissue proteins was determined 3 h after the application. Radioactivity in blood and in respiratory CO2 was measured up to 3 h and 6 h, respectively, after the application. The results indicate no effect of gum guar on the utilization of the protein or of the amino acids of the protein hydrolysate.
Heat shock factor ZmHsf17 positively regulates phosphatidic acid phosphohydrolase ZmPAH1 and enhances maize thermotolerance

Abstract:Heat stress (HS) adversely impacts plant growth, development and grain yield. Heat shock factors (Hsf), especially HsfA2 subclass, play a pivotal role in the transcriptional regulation of genes in response to HS. In this study, the coding sequence of maize ZmHsf17 was cloned. ZmHsf17 contains conserved domains: DNA binding, oligomerization and transcriptional activation. The protein was nuclear localized and had transcription activation activity. Yeast two hybrid and split luciferase complementary assays confirmed the interaction of ZmHsf17 with members of the maize HsfA2 subclass. Overexpression of ZmHsf17 in maize significantly increased chlorophyll content and net photosynthesis rate of maize leaves, and enhanced the stability of cellular membranes. Through integrative analysis of ChIP-seq and RNA-seq datasets, ZmPAH1, encoding phosphatidic acid phosphohydrolase of lipid metabolic pathways, was identified as a target gene of ZmHsf17. The promoter fragment of ZmPAH1 was bound by ZmHsf17 in protein-DNA interaction experiments in vivo and in vitro. Lipidomic data also indicates that the overexpression of ZmHsf17 increased levels of some critical membrane lipid components of maize leaves under HS. This research provides new insights into the role of the ZmHsf17-ZmPAH1 module in regulating thermotolerance in maize.

Abstract:Plants respond to various stresses by triggering the expression of genes that encode proteins involved in plant growth, fruit ripening, cellular protein homeostasis, and tolerance systems. sHSPs, a subfamily of heat shock proteins (HSPs), can be expressed in plants to inhibit abnormal aggregation of proteins and protect normal proteins by interacting with folding target proteins, protect cell integrity, and improve resistance under various adverse conditions. Thus, sHSPs have significant influences on seed germination and plant development. In this review, the classification, structure, and functions of sHSP family members in plants are systematically summarized, with emphasis on their roles in promoting fruit ripening and plant growth by reducing the accumulation of ROS, improving the survival rate of plants and the antioxidant activity, and protecting photosynthesis under biotic and abiotic stresses. Meanwhile, the production and regulatory mechanisms of sHSPs are described in detail. Heat shock factors, long non-coding RNA (lncRNAs), microRNA (miRNAs), and FK506 binding proteins are related to the production process of sHSPs. Molecular chaperone complex HSP70/100, plastidic proteins, and abscisic acid (ABA) are involved in the regulatory mechanisms of sHSPs. Besides, scientific efforts and practices for improving plant stress resistance have carried out the constitutive expression of sHSPs in transgenic plants in recent years. It is a powerful path for inducing the protective mechanisms of plants under various stresses. Therefore, exploring the role of sHSPs in the plant defense system paves a way for comprehensively unraveling plant tolerance in response to biotic and abiotic stress.
