Abstract:The sewer system is a significant source of hydrogen sulfide (H2S) and greenhouse gases which has attracted extensive interest from researchers. In this study, a novel combined dosing strategy using nitrate and calcium peroxide (CaO2) was proposed to simultaneously control sulfide and greenhouse gases, and its performance was evaluated in laboratory-scale reactors. Results suggested that the addition of nitrate and CaO2 improved the effectiveness of sulfide control. And the combination index method further proved that nitrate and CaO2 were synergistic in controlling sulfide. Meanwhile, the combination of nitrate and CaO2 substantially reduced greenhouse gas emissions, especially the carbon dioxide (CO2) and methane (CH4). The microbial analysis revealed that the combined addition greatly stimulated the accumulation of nitrate reducing-sulfide oxidizing bacteria (NR-SOB) that participate in anoxic nitrate-dependent sulfide oxidation, while the abundance of heterotrophic denitrification bacteria (hNRB) was reduced significantly. Moreover, the presence of oxygen and alkaline chemicals generated by CaO2 facilitated the inhibition of sulfate-reducing bacteria (SRB) activities. Therefore, the nitrate dosage was diminished significantly. On the other hand, the generated alkaline chemicals promoted CO2 elimination and inhibited the activities of methanogens, leading to a decrease of CO2 and CH4 fluxes, which facilitated elimination of greenhouse effects. The intermittent dosing test showed that the nitrate and CaO2 could be applied intermittently for sulfide removal. And the chemical cost of intermittent dosing strategy was reduced by 85 % compared to the continuous dosing nitrate strategy. Therefore, intermittent dosing nitrate combined with CaO2 is probably an effective and economical approach to control sulfide and greenhouse gases in sewer systems.
Demonstration Research on Treatment of Domestic Sewage from Tourism-Oriented Rural Towns and Villages with Multi-Soil-Layering (MSL) System.

Abstract:The rapid development of rural tourism has resulted in water pollution in the countryside.Rural tourist spots in Anji County,Zhejiang Province are mostly located in remote hilly areas,where no large piece of land for construction of wastewater treatment facilities is available.To solve this problem,a multi-soil-layering(MSL) technology featuring low investment,low operation cost,simple operation and management,and small land occupation was introduced.The treatment capacity of the MSL system is about 60 m3·d-1.After half a year of stable operation,monitoring shows that the effluent of the system is 43.8,5.1,3.1,0.6 and 7.0 mg·L-1 in average concentration of COD,TN,NH+4-N,TP and SS,indicating that their average removal rate being 82.7%,92.2%,94.1%,89.4% and 97.1%,respectively.The quality of its effluent may be stably kept up to the Criteria of Grade I of the Standard for Discharge of Pollutants From Urban Sewage Treatment Facilities(GB 18918—2002).
Enhanced Treatment of Decentralized Domestic Sewage Using Gravity-Flow Multi-Soil-Layering Systems Coupled with Iron-Carbon Microelectrolysis

Abstract:Soil-based decentralized treatment technology has become increasingly popular as an ideal solution for water pollution control in rural areas. It is very necessary to optimize the removal mechanisms and performance of such technologies on rural domestic sewage treatment. This was the first study of a gravity-flow multi-soil-layering (MSL) system coupled with iron-carbon microelectrolysis (ICM). Influent COD/TN (C/N) ratio and bottommost soil mixture block (SMB) submersion were selected as the operating factors relevant to the ICM in MSL systems. Such two key factors were investigated in the factorial experiment. The removal efficiencies of COD, TP, NH3-N, NO3−-N, and TN could be reached up to 96.3, 100, 95.4, 93.8, and 79.6%, respectively. Different levels of factors could comprehensively drive the performance variation. The factorial analysis indicated that the bottommost SMB submersion had the most significant and dominant negative effects on aerobic processes. The ideal TP removal attributed to the presence of the bottommost SMB submersion. It played the dominant role for the bottommost SMB submersion in facilitating an electrochemical reaction through the ICM. Zero-valent iron or ferrous ions could be transformed to final ferric ions more efficiently during the period of the ICM reactions. The ICM could promote the capability of a SMB for removing nutrients in sewage, especially provide electron donors to denitrifying bacteria in MSL systems. However, there were non-significant effects of the influent C/N ratio on the removal performance of MSL systems. This study can help enrich the pollutant removal mechanisms in MSL systems.

Abstract:In this study, an integrated multi-soil-layering and subsurface wastewater infiltration (MSL-SWI) system was developed for decentralized domestic sewage treatment under high hydraulic loading rates (HLRs). To improve sustainable nitrogen removal, the influence of intermittent operation and shunt distributing wastewater on the performance of MSL-SWI systems was investigated. The optimal performance—with removal efficiencies of 93.41% for chemical oxygen demand, 97.91% for total phosphorus, 74.02% for ammonia nitrogen, and 73.56% for total nitrogen—was achieved using both intermittent operation and shunt distributing wastewater under an HLR of 0.3 m3 m−2 d−1. The activity of microbial nitrogen functional genes (i.e., amoA, nirK, nirS, nosZ, and anammox 16S rRNA) and their relationships with nitrogen transformation rates were further analyzed in different layers of the system. The results imply that nitrification and anaerobic ammonium oxidation in the MSL section coupled with nitrification and denitrification in the SWI section contribute to main the mechanisms of sustainable nitrogen removal. In summary, MSL-SWI systems not only operate with high efficiency under high HLRs, but the contaminant removal is also stable and sustainable, which are promising properties for domestic sewage treatment in areas where land resources are limited.

Influence of COD/sulfate ratios on the integrated reactor system for simultaneous removal of carbon, sulfur and nitrogen.

Abstract:An integrated reactor system was developed for the simultaneous removal of carbon, sulfur and nitrogen from sulfate-laden wastewater and for elemental sulfur (S-0) reclamation. The system mainly consisted of an expanded granular sludge bed (EGSB) for sulfate reduction and organic carbon removal (SR-CR), an EGSB for denitrifying sulfide removal (DSR), a biological aerated filter for nitrification and a sedimentation tank for sulfur reclamation. This work investigated the influence of chemical oxygen demand (COD)/sulfate ratios on the performance of the system. Influent sulfate and ammonium were fixed to the level of 600 mg SO42- L-1 and 120 mg NH4+ L-1, respectively. Lactate was introduced to generate COD/SO42- = 0.5:1, 1:1, 1.5:1, 2:1, 3:1, 3.5:1 and 4:1. The experimental results indicated that sulfate could be efficiently reduced in the SR-CR unit when the COD/SO42- ratio was between 1:1 and 3:1, and sulfate reduction was inhibited by the growth of methanogenic bacteria when the COD/SO42- ratio was between 3.5:1 and 4:1. Meanwhile, the Org-C/S2-/NO3- ratios affected the S-0 reclamation efficiency in the DSR unit. When the influent COD/SO42- ratio was between 1:1 and 3:1, appropriate Org-C/S2-/NO3- ratios could be achieved to obtain a maximum S-0 recovery in the DSR unit. For the microbial community of the SR-CR unit at different COD/SO42- ratios, 16S rRNA gene-based high throughput Illumina MiSeq sequencing was used to analyze the diversity and potential function of the dominant species.
Nitrogen removal in improved subsurface wastewater infiltration system: Mechanism, microbial indicators and the limitation of phosphorus

Abstract:To enhance the nitrogen removal capacity, scrap iron filings and Si-Al porous clay mineral material (PCMW) was used to improve a subsurface wastewater infiltration system (SWIS). The results showed TN and NH4+-N removal efficiencies of improved SWIS were 20.72% and 5.49% higher than those of the control SWIS, respectively. Based on the response of the removal performance, microbial community and function analysis of 16s rRNA amplicon sequencing results, the amending soil matrix substantially enriched the nitrogen removal bacteria (Rhizobiales_Incertae_Sedis and Gemmatimonadaceae), and significantly improved the activities of key enzymes (Hao, NasAB, NarGHI, NirK, NorBC, NirA and NirBD), particularly at co-occurrence zone of nitrification and denitrification (70-130 cm depth). The amending soil matrix not only extended the growth space of microbes, but also provided additional electrons and carbon sources for denitrifying bacteria by regulating the structure and function of the microbial community. In addition, amending soil matrix could enhance phosphate metabolism genes and phosphate solubilizing microbes in the denitrification zone by increasing the phosphorus source, thus strengthening nitrogen metabolism. Nitrospiraceae, Rhizobiales_Incertae_Sedis and Gemmatimonadaceae related to nitrogen removal and Bacillaceae with phosphate-solubilizing ability could be used as microbial indicators of nitrogen removal in SWISs. The reciprocal action of environmental on microbial characteristics exhibited microbial functional were related to DO, Fe2+, TOC, TP, TN, NH4+-N and NO3--N. Those could be used as physicochemical and biological indicators for application and monitoring of SWIS. In conclusion, this study provided a low-cost and efficient enhancement approach for the application of SWIS in decentralized domestic sewage treatment, and furnished theoretical support for subsequent applications.

Abstract:A sewage-integrated treatment system (SITS) for the treatment of municipal wastewater, consisting of an expanded granular sludge bed (EGSB) reactor to remove soluble organic matter and an electrochemical (EC) reactor to oxidize the NH(3)-N, was evaluated. The performance of the EGSB reactor was monitored for 12 months in a pilot-scale plant. Iron shavings were added to the EGSB reactor on the sixtieth day to improve the removal efficiency of the chemical oxygen demand (COD), suspended solids (SS) and total phosphorus (TP). After the iron shavings were added, the effluent COD, SS and TP decreased from 104 to 46 mg L(-1), 21 to 8.6 mg L(-1) and 3.62 to 1.36 mg L(-1), respectively. Moreover, in the EC reactor, which was equipped with IrO(2)/Ti anodes, the NH(3)-N and total nitrogen (TN) concentrations decreased from 25 to 12 mg L(-1) and 29 to 15 mg L(-1), respectively. The NH(3)-N was directly oxidized to N(2), resulting in no secondary pollution. The results demonstrated the possibility of removing carbon and nutrients in a SITS with high efficiency. The system runs efficiently and with a flexible operation, making it suitable for low-strength wastewater. The results and parameters presented here can provide references for the practical project.

Abstract:The widely spread rural unsanitary landfills in South China pose an environmental threat to water bodies and soil. Although various processing technologies have been utilized for treatment of landfill leachate, their application to the landfills in rural areas is restricted by the availability of skilled professionals and high operation costs. In this experiment, four MSLs with altered soil mixed block (SMB) and different hydraulic load rate (HLR) were applied in the experiment to investigate the treatment of the landfill leachate without aeration or under low aeration supply. The experiment results showed that the improved MSL could effectively treat the chemical oxygen demand (COD), NH4-N and P. COD and NH4-N removal efficiencies of MSL were 97.4%, 82.4% and 72.0%, 62.0%, respectively under HLRs of 200 and 400 L/(m(2).d) without aeration; COD and NH4-N removal efficiencies of M800 and M1600 were 62.3%, 53.4% and 45.3%, 35.3% respectively under intermittent aeration. N removal efficiency was low due to a strong nitrification effect, and the nitrogen removal capacity of the MSL was greatly reduced at the end of the experiment. P removal efficiency of MSL was 75.6 to 91.9% under HLR 200 and 400 L/(m(2).d). The intermittent aeration was helpful to remove the clogging of MSLs, after they were clogged under HLRs of 800 and 1600 L/(m(2).d). MSL is promising as an appealing nitrifying biofilm reactor. Abstract:Adding zero‐valent iron and increasing C load are effective methods to improve denitrification efficiency and reduce greenhouse gas emissions in subsurface wastewater infiltration systems. Subsurface wastewater infiltration system (SWIS) has been recognized as a simple operation and environmentally friendly technology for wastewater purification. However, effectively removing nitrogen (N) remains a challenge, hindering the widespread application of SWIS. In this study, zero‐valent iron (ZVI) and porous mineral material (PMM) were applied in SWIS to improve the soil matrix. Our results suggested that the addition of ZVI and PMM could simultaneously enhance N removal efficiency and reduce nitrous oxide emissions. This could be attributed to the abundant electrons generated by ZVI alleviating the electronic limitation of denitrification and the porous structure of PMM providing solid phase support for microbial growth. In addition, the abundance of microbial functional genes increased in modified SWIS, which could further explain the higher pollutant removal efficiency. Overall, this study provides new insights into the mitigation of wastewater pollution and greenhouse gas emissions in SWIS. Practitioner Points ZVI and PMM can adapt to different C loads and enhance pollutant removal efficiency in SWIS. Increasing C–N ratios positively affected the nitrate removal performance and negatively affected ammonium removal performance in SWIS. The amending soil matrix promoted the reduction of the N2O to N2 and greenhouse gas emissions were well controlled. The abundance of microbial functional genes increased with the improvement of the soil matrix.

Abstract:A pilot plant of subsurface wastewater infiltration system (SWIS) filled with red clay was tested to treat rural sewage with hydraulic loading of 2 cm/d. The experiment results showed that average removal rates of COD, NH4(+)-N, total phosphorus, and total nitrogen were 84.7%, 70.0%, 98.0% and 77.7%, with average effluent concentrations of COD, NH4(+)-N, total phosphorus, and total nitrogen being 11.7 mg/L, 4.0 mg/L, 0.04 mg/L and 4.7 mg/L respectively, which met the standard for water reuse issued by the Ministry of Construction of China. Nitrogen removal mechanism analysis revealed that nitrogen in the influent was mainly eliminated through biological removal via nitrification/denitrification processes. In the pilot system, denitrification was well proceeded but nitrification was not satisfactory. How to improve nitrification performance through change of soil environment was the key to enhance nitrogen removal rate. The measurement results of redox potential in the filled soil showed that reductive property of soil was main hindrance to nitrification process.
Disentangling microbial coupled fillers mechanisms for the permeable layer optimization process in multi-soil-layering systems

Abstract:The multi-soil-layering (MSL) systems is an emerging solution for environmentally-friendly and cost-effective treatment of decentralized rural domestic wastewater. However, the role of the seemingly simple permeable layer has been overlooked, potentially holding the breakthroughs or directions to addressing suboptimal nitrogen removal performance in MSL systems. In this paper, the mechanism among diverse substrates (zeolite, green zeolite and biological ceramsite) coupled microorganisms in different systems (activated bacterial powder and activated sludge) for rural domestic wastewater purification was investigated. The removal efficiencies performed by zeolite coupled with microorganisms within 3 days were 93.8% for COD, 97.1% for TP, and 98.8% for NH4+-N. Notably, activated sludge showed better nitrification and comprehensive performance than specialized nitrifying bacteria powder. Zeolite attained an impressive 89.4% NH4+-N desorption efficiency, with a substantive fraction of NH4+-N manifesting as exchanged ammonium. High-throughput 16S rRNA gene sequencing revealed that aerobic and parthenogenetic anaerobic bacteria dominated the reactor, with anaerobic bacteria conspicuously absent. And the heterotrophic nitrification-aerobic denitrification (HN-AD) process was significant, with the presence of denitrifying phosphorus-accumulating organisms (DPAOs) for simultaneous nitrogen and phosphorus removal. This study not only raises awareness about the importance of the permeable layer and enhances comprehension of the HN-AD mechanism in MSL systems, but also provides valuable insights for optimizing MSL system construction, operation, and rural domestic wastewater treatment.

Abstract:This paper proposed a novel simultaneous denitrification and biogas desulfurization (SDBD) process for domestic sewage treatment and biogas upgrading. The process consisted of an expanded granular sludge bed (EGSB) for integrated autotrophic and heterotrophic denitrification (IAHD) and a biological aerated filter (BAF) for nitrification. The experiments were firstly conducted to investigate the performance of the lab-scale SDBD system with real domestic sewage at various hydraulic retention times (HRT). The system successfully achieved 83.0 ± 1.8% chemical oxygen demand (COD), 96.2 ± 1.8% ammonia (NH4+-N) and 67.3 ± 4.1% total nitrogen (TN) removal efficiency without biogas introduction, and the optimal HRT of EGSB and BAF was determined to be 6 h and 4 h, respectively. Then, biogas with three different hydrogen sulfide concentrations (1.5%, 1.2% and 0.9%) was introduced into the system. Almost complete removal of hydrogen sulfide in biogas was achieved in all the tested scenarios and the removal efficiency of COD, NH4+-N and TN maintained at around 86.8%, 98.4% and 83.4%, respectively. Microbial community analysis results showed that biogas introduction notably increased the bacterial diversity and particularly enriched the nitrate-reducing, sulfide-oxidizing bacteria, which forms the major contribution to the hydrogen sulfide removal in the system. Finally, we put the SDBD system in the whole treatment system of domestic sewage and discussed its economics. The results indicated that the total operating cost of sewage treatment plant could be saved to 0.59 RMB/m3, which proved SDBD system to be a promising alternative way in biological nitrogen removal and biogas desulfurization.

Abstract:Simultaneous nitrification and denitrification (SND) is a promising denitrification method. But so far, there have been few reports on the SND and denitrification mechanism of the coupled system with sponge iron (SI). In this paper, the denitrification performance of the mono-sludge system (only activated sludge), mono-carrier system (activated sludge + polyurethane foam) and coupled system (activated sludge + polyurethane foam and SI) was investigated. The study found that compared with the mono-sludge system, the TN removal efficiencies of the mono-carrier system and coupled system increased by 24% and 29.2%, respectively. The change of dissolved oxygen in the carrier of the coupled system showed that the time of forming anoxic and anaerobic micro-environment in one cycle decreased continuously until it reached zero. The nitrate-dependent ferric oxidizing bacteria Acidovorax and Aquabacterium appeared in the coupled system. In addition, it was found that the intervention of SI significantly increased the activity of membrane nitrate reductase (NAR). This study provides practical and useful technology for the treatment of domestic sewage.

Manganese sulfide-sulfur and limestone autotrophic denitrification system for deep and efficient nitrate removal: Feasibility, performance and mechanism

Abstract:Despite the great potential of sulfur-based autotrophic denitrification, an improvement in nitrate removal rate is still needed. This study used the desulfurized products of Mn ore to develop the MnS-S0-limestone autotrophic denitrification system (MSLAD). The feasibility of MSLAD for denitrification was explored and the possible mechanism was proposed. The nitrate (100 mg/L) was almost removed within 24 h in batch experiment in MSLAD. Also, an average TN removal of 98 % (472.0 mg/L/d) at hydraulic retention time of 1.5 h in column experiment (30 mg/L) was achieved. MnS and S0 could act as coupled electron donors and show synergistic effects for nitrate removal. γ-MnS with smaller particle size and lower crystallinity was more readily utilized by the bacterium and had higher nitrate removal efficiency than that of α-MnS. Thiobacillus and Sulfurimonas were the core functional bacterium in denitrification. Therefore, MnS-S0-limestone bio-denitrification provides an efficient alternative method for nitrate removal in wastewater.

Abstract:Nitrate has been used for nearly 80 years as a chemical to control problematic gases in the sewer system. However, few studies have explored simultaneous control in biofilm and sediment using different strategies. Here, we introduced a nitrate dosing method involving an initial high shock followed by low level dosing, tested at two distinct frequencies in a lab-scale sewer reactor <110 days. Long-term investigation revealed that the more frequent 20 min interval dosing slightly surpassed the 1 h interval method when applying the same hourly dose of 30 mg N/L (sulfide control: 98.3 ± 1.7 % vs 97.9 ± 1.5 %; methane control: 89.8 ± 4.5 % vs 83.4 ± 6.7 %). 16 s rRNA gene amplicon sequencing revealed biofilm detachment and sediment stratification, which can be attributed to the differing effects of nitrate dosing on biofilm and sedimentary microbial interactions. Dominant bacteria such as Thauera and Thiobacillus performed autotrophic denitrification and nitrate-reducing sulfide-oxidation in conjunction with methane oxidizers. These microbes collaboratively control sulfide and methane emissions from sediment. Our findings suggest that nitrate supports the diversity and versatility of their metabolism in the sewer system.

Nitrite-dependent Microbial Utilization for Simultaneous Removal of Sulfide and Methane in Sewers

Abstract:Chemicals are commonly dosed in sewer systems to reduce the emission of hydrogen sulfide (H2S) and methane (CH4), incurring high costs and environmental concerns. Nitrite dosing is a promising approach as nitrite can be produced from urine wastewater, which is a feasible integrated water management strategy. However, nitrite dosing usually requires strict conditions, e.g., relatively high nitrite concentration (e.g., ∼200 mg N/L) and acidic environment, to inhibit microorganisms. In contrast to “microbial inhibition”, this study proposes “microbial utilization” concept, i.e., utilizing nitrite as a substrate for H2S and CH4 consumption in sewer. In a laboratory-scale sewer reactor, nitrite at a relatively low concentrations of 25–48 mg N/L was continuously dosed. Two nitrite-dependent microbial utilization processes, i.e., nitrite-dependent anaerobic methane oxidation (n-DAMO) and microbial sulfide oxidation, successfully occurred in conjunction with nitrite reduction. The occurrence of both processes achieved a 58 % reduction in dissolved methane and over 90 % sulfide removal in the sewer reactor, with microbial activities measured as 15.6 mg CH4/(L·h) and 29.4 mg S/(L·h), respectively. High copy numbers of n-DAMO bacteria and sulfide-oxidizing bacteria (SOB) were detected in both sewer biofilms and sediments. Mechanism analysis confirmed that the dosed nitrite at a relatively low level did not cause the inhibition of sulfidogenic process due to the downward migration of activity zones in sewer sediments. Therefore, the proposed “microbial utilization” concept offers a new alternative for simultaneous removal of sulfide and methane in sewers.

Abstract:In this work, two deep subsurface wastewater infiltration systems (SWISs) were constructed and fed with domestic sewage (control system, S1) and mixed wastewater consisting of old landfill leachate and domestic sewage (experimental system, S2). S1 and S2 exhibited favorable removal efficiencies, with TP (98.8%, 98.7%), COD (87.6%, 86.9%), NH4+-N (99.8%, 99.9%) and TN (99.2%, 98.9%). Even when increasing the pollutant load in S2 by adding old landfill leachate, the almost complete removal performance could be maintained in terms of low effluent concentrations and even increased in terms of load removal capabilities, which included COD (19.4, 25.9 g·m-2·d-1), NH4+-N (8.2, 19.9 g·m-2·d-1), TN (8.9, 20.6 g·m-2·d-1). To investigate the transformation of dissolved organic matter along depth, Three-Dimensional Excitation Emission Matrix fluorescence spectroscopy combined with Fluorescence Regional Integration analysis was applied. The results showed that PI,n and PII,n (the proportions of biodegradable fractions) increased gradually from 6.59% and 21.8% at S2_20 to 10.8% and 27.7% at S2_110, but PIII,n and PV,n (the proportions of refractory organics) declined from 23.1% and 27.8% at S2_20 to 21.1% and 16.4% at S2_110, respectively. In addition, high-throughput sequencing technology was employed to observe the bacterial community at different depths, and the predicted functional potential of the bacterial community was analyzed by PICRUSt. The results showed that the genera Flavobacterium, Pseudomonas, Vogesella, Acinetobacter and Aquabacterium might be responsible for refractory organic degradation and that their products might serve as the carbon source for denitrifiers to achieve simultaneous nitrate and refractory organic removal. PICRUSt further demonstrated that there was a mutual response between refractory organic degradation and denitrification. Overall, the combined treatment of domestic sewage and old leachate in rural areas by SWIS is a promising approach to achieve comprehensive treatment.

Study into a land infiltration system for treating night soil sewage

Abstract:In this study, a land infiltration system with an area of 570 m 2 and a treatment ability of 15 m 3/d was constructed near a public lavatory, with the objective of treating the night soil sewage and to enhance the TN (Total Nitrogen) removal with biological denitrification. It was observed that the land infiltration system is efficient and economic in treating the night soil sewage. The oxidative condition of the medium was enough for ammonia removal, which is favorable for the biological nitrification process; and the high influent COD (Chemical Oxygen Demand) concentration in the present treatment is favorable for biological denitrification process. The average COD, SS(Suspended solids), NH 3-N (Ammonium nitrate) and TP (Total Phosphate) of the influent were 530.0 mg/L, 94.5 mg/L, 136.1 mg/L and 8.10 mg/L, respectively. After the treatment, the average COD, SS, NH 3-N and TP of the effluent reduced to 49.7 mg/L, 8.0 mg/L, 16.4 mg/L and 0.39 mg/L, respectively, which meets the standard for urban miscellaneous water consumption in China. The energy consumption was 0.03 USD/m3, which is relatively low and the system is easy to operate and maintain. After sterilization, the effluent was reused in washing toilets. Sludge generated in the septic tank and anaerobic tank were applied to farmland as fertilizer, and grass cut from the land were fed to the fishes in the pond nearby. Therefore, it is an eco-system in which no wastes are discharged and all substances are recycled. © IJIB, All rights reserved.

Abstract:Subsurface wastewater infiltration system (SWIS) is widely used in the treatment of rural domestic sewage. However, the nitrogen removal effect is unsatisfactory. In this study, laboratory-scale SWIS was built, and the nitrogen removal capacity of the system was optimized by a combination of intermittent operation and the shunt method. The optimal total nitrogen (TN) removal rate (56.53%) is obtained when the wet-dry ratio (Rwd) is 1:2 and the shunt ratio is 3:1. The optimal ammonia nitrogen (NH4+-N) removal rate (54.97%) is obtained when Rwd is 1:3 and shunt ratio is 2:1. In addition, SWIS achieved a good COD removal rate (>75%) and TP removal rate (>95%) during the whole operation stage. Proteobacteria, Acidobateria and Bacteroidetes are high abundance phyla in SWIS. Bacteroidetes, Chloroflexi and Nitrospirae associated with nitrogen removal are abundant at the shunt ratio of 2:1 and 3:1. At the genus level, Bosea, Bradyrhizobium and Phenylobacterium were functional genera associated with denitrification, enriching the substratum of SWIS at shunt ratios of 1:1, 2:1 and 3:1. Nitrogen (N) forms and ORP analysis showed that under the appropriate shunt ratio and Rwd, the SWIS matrix has a suitable redox microenvironment, and thus creates a well-defined nitrification area and denitrification area. This research provides a reliable theoretical basis for the construction and operation of SWIS in rural areas.
