Abstract:To improve the viability of Lactobacillus plantarum (P) during digestion and storage, the probiotics were encapsulated by alginate (ALG) and alginate-gelatin (ALG-GE) hydrogels beads. ALG-P-GE showed much better physicochemical properties than ALG-P. The scanning electron microscopy (SEM) results validated the incor-poration of bacterial cells into the beads. ALG-P-GE exhibited good encapsulation efficiency of 97.7 %, and the storage and thermal stability of probiotic were increased by 15 % and 8 %, respectively, when comparing with ALG-P. ALG-P-GE beads could protect the probiotics from inactivation in simulated gastric fluid and then release it in simulated intestinal fluid. The protective mechanism of ALG-GE for probiotics was further studied by fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD) and found that ALG and GE can form gel network through hydrogen bonding and electrostatic interactions. In the mimic beverage systems, ALG-P-GE beads could protect the encapsulated probiotics and increase its viability. The storage, thermal, and digestion stability of encapsulated probiotic were significantly increased and showed high viability in the mimic beverage systems. ALG-P-GE beads have great potential for the protection and delivery of probiotics in food systems.

Abstract:Probiotics are used in food products, dietary supplements and pharmaceutical products because they may provide health-promoting effects in humans. To be efficacious, probiotics need to be viable at sufficient abundance within the large intestine. However, many commercial products containing probiotics suffer from a substantial loss of bacterial viability during shelf storage and during gastrointestinal transit. In this study, a probiotic (Lactobacillus salivarious Li01) was incorporated into either alginate or alginate-gelatin microgels. The morphology of the microgels was characterized by scanning electron microscopy, which indicated that they had a spherical shape and that almost all of the bacterial cells were encapsulated inside them. Probiotic viability was determined under aerobic conditions, heat treatment, and simulated gastrointestinal conditions. Encapsulation significantly enhanced the viability of the probiotic during aerobic storage. The microgels maintained their structures under simulated gastric conditions, but either eroded or swelled under simulated small intestine conditions. The alginate-gelatin microgels were the most effective at protecting the bacteria under simulated gastrointestinal conditions when compared to the alginate microgels and non-encapsulated bacteria. In conclusion, alginate-gelatin microgels have great potential for the protection and delivery of probiotics in food, supplement, and pharmaceutical products.

Abstract:The major reduction in probiotic viability that occurs during food storage and gastrointestinal transit limits their potential health benefits. Microencapsulation is one of the most effective methods to protect probiotics from various harsh conditions. In this study, a model probiotic (Pediococcus pentosaceus Li05) was encapsulated in an alginate-gelatin microgels in the absence and presence of magnesium oxide (MgO) nanoparticles (NPs). The morphology and surface properties of the encapsulation systems were characterized by transmission electron microscopy (TEM) and atomic force microscopy (AFM), which showed that both the probiotics and NPs were successfully incorporated into spherical microgels. The viability of the probiotic was evaluated after exposure to different conditions: long-term storage in an aerobic environment; heat treatment; and gastrointestinal transit. Encapsulation of the probiotics significantly enhanced their viability under these different conditions. Probiotics encapsulated in MgO-loaded microgels were more stable than free bacterial cells or those encapsulated in microgels alone: less than 2 log(10) CFU reduction after 40 min incubation in gastric fluids versus 5 log(10) CFU reduction in the first 10 min for free cells. The SEM images indicated that the NPs may lead to enhanced probiotic viability by filling pores inside the microgels, which may have inhibited the ability of oxygen and hydrogen ions to access the probiotics. Moreover, the MgO NPs neutralized the hydrogen ions in the gastric fluids, thereby reducing acid-induced degradation of the probiotics. These results demonstrate that MgO-loaded microgels may be a promising encapsulation and delivery system for improving the efficacy of orally administered probiotics by protecting them from the harsh conditions during storage and in the gastrointestinal tract.
Preparation of alginate-whey protein isolate and alginate-pectin-whey protein isolate composites for protection and delivery of Lactobacillus plantarum

Abstract:Probiotics are sensitive to external conditions, resulting in low survival rates after being ingested or during food production, transportation and storage. In order to improve the survival rate of Lactobacillus plantarum (LP) during gastrointestinal digestion, storage, and freeze-drying, alginate-whey protein isolate (ALG-WPI) and alginate-pectin-whey protein isolate (ALG-PEC-WPI) composites were employed to encapsulate LP. The encapsulation efficiency of ALG-WPI-LP and ALG-PEC-WPI-LP beads both reached more than 99 %. Scanning electron microscopy (SEM) indicated that dense and rough aggregates were formed on the surface of both composites, and attached LP cells could be observed inside the beads. The ALG-WPI and ALG-PEC-WPI composites can protect the viability of LP in simulated gastric fluid (SGF) and release the probiotics in simulated intestinal fluid (SIF). The storage stability of LP at 4 °C was improved by about 15 % in comparison with bare LP and the survival rates of LP in ALG-WPI-LP and ALG-PEC-WPI-LP powders after freeze-drying were increased by 65.37 % and 72.06 %, respectively. The formation mechanism of ALG-WPI and ALG-PEC-WPI composites was further explored by fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD) and thermogravimetric analysis (TGA). The ALG-WPI and ALG-PEC-WPI composites have great potential to protect and deliver probiotics in food systems.

Abstract:Probiotics have considerable interest due to their inseparable link to human health. However, probiotic products are seriously challenged during processing, preservation, and intake. Food-grade probiotic delivery systems need to be further explored as an effective way to enhance cell viability. In this study, water-in-oil (W/O) Pickering emulsions were fabricated by adding candelilla wax (CLW) as a network stabilizer based on acylated EGCG derivatives in the crystalline form as a Pickering stabilizer. The effects of acylated EGCG derivatives' concentration, CLW concentration, and oil phase volume fraction on the droplet size distribution, microstructure, and physical stability of Pickering emulsions were explored. The presence of CLW reduced the particle size and improved the physical stability of acylated EGCG-based emulsions, and the effect was more positive with increasing concentration. The protective effect of emulsions with different oil phase volume fractions on Lactobacillus plantarum during freeze-thaw cycles, storage, and gastrointestinal digestion was evaluated. The outer-phase physical barrier of W/O emulsions co-stabilized with acylated EGCG derivatives and CLW facilitated the sensitivity of probiotics to ice crystal growth, temperature changes, acidic environments, and digestive enzymes. The emulsions formulated with 40% oil phase volume fractions allowed Lactobacillus plantarum to survive up to 7.75 log CFU g −1 in the harsh gastrointestinal environment. The results offer promising strategies for applying W/O emulsion probiotic delivery systems in food processing, storage, and oral administration.

Abstract:Biopolymer microgels (hydrogel beads) can be used to improve probiotic stability during food storage and gastrointestinal passage. In this study, the impact of including natural polymeric and colloidal fillers on the viability of Lactobacillus casei encapsulated in calcium alginate microgels was examined. The polymeric filler was polysaccharides isolated from Choerospondias axillaris fruit peel (CP) and the colloidal filler was oil-in-water emulsion stabilized by CP. Results showed that addition of CP or CP coated lipid droplets significantly increased the antioxidant activity but reduced the apparent viscosity of the alginate solutions. Encapsulation of the probiotics in pure calcium alginate microgels reduced their viability (≈ 0 log CFU g−1 after 24 h). The inclusion of the polymeric or colloidal fillers within the microgels eliminated this negative effect. Moreover, they enhanced probiotic viability under simulated gastrointestinal tract conditions (> 7.0 log CFU g−1). The calcium alginate microgels containing CP had the highest probiotic viability (7.42 ± 0.08 log CFU g−1) after 20 days storage. Our results indicated that simple calcium alginate microgels are not effective at protecting Lactobacillus casei during storage or gastrointestinal passage. However, incorporation of suitable polymeric or colloidal fillers can significantly enhance their viability.

Abstract:In this study, we developed a novel delivery system using carboxymethyl konjac glucomannan-chitosan (CMKGM-CS) nanogels stabilized single and double emulsion incorporated into alginate hydrogel as microcapsule matrix for intestinal-targeted delivery of probiotics. Through in vitro experiments, it was demonstrated that alginate hydrogel provided favorable biocompatible growth conditions for the proliferation of Lactobacillus reuteri (LR). The alginate hydrogel containing single (ASE) or double emulsions (ACG) enhanced the resistance of LR to various adverse environments. Simulated gastrointestinal digestion experiments revealed that the survivability of LR in free, CON, ASE and ACG group decreased by 6.45 log CFU/g, 4.21 log CFU/g, 1.26 log CFU/g and 0.65 log CFU/g, respectively. In vivo studies conducted in mice showed that ACG maintained its integrity during passage through the stomach and released the probiotics in the targeted intestinal area, whereas the pure alginate hydrogels (CON) were prematurely released in the gastrointestinal tract. Moreover, the viable counts of ACG in different intestinal segments (jejunum, ileum, cecum, and colon) were increased by 1.11, 1.42, 1.68, and 1.89 log CFU/g, respectively, after 72 h of oral administration compared to the CON group. This research contributed valuable insights into the development of an effective microbial delivery system with potential applications in the biopharmaceutical and food industries.

Abstract:Web based musical discussions are sometimes limited by the fact that most of the applications used (forums, guestbooks etc.) only support textual messages. Although systems with advanced functionalities like file attachments or images and sounds inline inserts are technically possible, very few if any of the existing "off the shelf", freely available systems support them. We propose a generalized procedure based on client side programming that can easily "enhance" any existing discussion system to allow people to include a variety of musically interesting "objects" including, but not limited to, sound and voice clips, midi files, partitures and tablatures. No client side browser plugin is required by the visitor to view or insert these "attachments". The resulting system poses no additional security threat and works with any modern generation browser.

Abstract:Polyglycerol polyricinoleate (PGPR) was used as the oil-soluble surfactant and beeswax was used as the oil phase to formulate a water-in-oil (W/O) high internal phase emulsions (HIPEs) for the encapsulation and protection of probiotics. The physicochemical properties of the W/O HIPEs and the survival of the encapsulated probiotics when exposed to acidic conditions and in vitro digestion were investigated. The viability of the probiotics decreased slightly when exposed to high-speed shearing. The rheological analysis, microstructural images, physicochemical stability showed that the W/O HIPEs remained relatively stable. The survival of the probiotics loaded in the SK-HIPEs (prepared with skim milk) was much higher than in the NS-HIPEs (prepared with normal saline) during storage at 4 ?. An in vitro gastrointestinal model showed that encapsulation of the probiotics enhanced their survival. This study provides useful insights into the utilization of W/O HIPEs to improve the efficacy of probiotics in the food industry.

Abstract:To improve the survivability of probiotics, Lactobacillus plantarum was microencapsulated using pufferfish skin gelatin (PSG)-based wall materials by spray-drying. This work investigated the protective effect of three different pH-dependent proteins (sodium caseinate (SC), soy protein isolate (SPI), and whey protein isolate (WPI)) combined with PSG on L. plantarum. The experimental results of spray-drying with an inlet temperature of 120 °C and an outlet temperature of 80 °C, storage at 4 °C for 6 months, simulated digestion, and turbidity indicated that PSG/SC had better stability and encapsulation effects and was more suitable to encapsulate L. plantarum than PSG/SPI and PSG/WPI. The optimum preparation conditions for L. plantarum microcapsules were a PSG/SC mass ratio of 2:1, an SC concentration of 20 g/L, and a cell concentration of 10 g/L. The encapsulation efficiency of the obtained microcapsules was 95.0%, and the survival rate was 94.2% in simulated gastric fluid for 2 h and 98.0% in simulated intestinal fluid for 2 h. Amino acid composition analysis exhibited that the imino acid and aspartic acid contents of PSG were 27.98 and 26.16 g/100 g protein, respectively, which was much higher than commercial bovine gelatin. This characteristic was favorable to the high encapsulation efficiency and stability of microcapsules. In vitro release experiments showed that the PSG/SC microcapsules did not disintegrate in simulated gastric fluid for 2 h but could completely release in simulated intestinal fluid for 2 h, which can maintain the high survivability of L. plantarum in simulated digestion. In general, this study demonstrated that microcapsules using PSG/SC as wall materials can effectively improve the survivability of probiotics and have great potential for application in probiotic products.

Automatic Extraction of Axioms for Planning

Abstract: Axioms can be used to model derived predicates in domain-independent planning models. Formulating models which use axioms can sometimes result in problems with much smaller search spaces than the original model. We propose a method for automatically extracting a particular class of axioms from standard STRIPS PDDL models. More specifically, we identify operators whose effects become irrelevant given some other operator, and generate axioms that capture this relationship. We show that this algorithm can be used to successfully extract axioms from standard IPC benchmark instances, and show that the extracted axioms can be used to significantly improve the performance of satisficing planners.

Abstract:In this study, we developed (W1/O/W2) double emulsions encapsulated by calcium-alginate hydrogel beads system (ACGs) for intestinal-targeted delivery of probiotics (Lactobacillus reuteri). Firstly, the carboxymethyl konjac glucomannan-chitosan (CMKGM-CS) nanogels were successfully fabricated by EDC/NHS initiated crosslinking, as concluded from the TEM images, FTIR spectra, XRD, etc. Then, double emulsions were prepared and encapsulated with various concentrations of alginate to form ACGs hydrogel beads. In vitro probiotic release experiments showed the lyophilized ACG-2 and ACG-3 hydrogel beads had a sustained release in simulated intestinal fluid (SIF), the viability of cells exceeded 107 CFU/mL at 6 h. The lyophilized ACG-3 hydrogel beads exhibited the viable release amount of 8.4 × 107 CFU/mL after storage for 90 d at 4 °C. Besides, the alginate concentration in the ACGs hydrogel beads influences the swelling behavior and structure of hydrogel beads by affecting the hydrogen bonds between alginate and CMKGM-CS, thereby mediating the release of probiotics.

Abstract:Hypothesis The strategies for stabilizing water-in-water (W/W) emulsions include the adsorption of solid particles at the water-water interface and the generation of interfacial films. We hypothesize that if sodium alginate is crosslinked at the water-water interface of W/W Pickering emulsions, the microstructure and rheological properties of the emulsions could be improved, thus enhancing the activity of probiotics in simulated gastrointestinal digestion. Experiments The W/W Pickering emulsions consisted of a dispersed maltodextrin (MD) phase in a continuous hydroxypropyl methylcellulose (HPMC) phase. The crosslinking W/W Pickering emulsion with fine-tuned internal structure was designed by leaching the CaCO 3 particles packed in the dispersed phase to release Ca 2+ crosslinked with sodium alginate. Findings Confocal laser scanning microscope results revealed sodium alginate crosslinked with calcium ions at the W/W interface. The rheological results of the crosslinking W/W Pickering emulsions suggested that the loss modulus (G′′) was higher than the energy storage modulus (G'). The microstructure indicated that the emulsions formed a dense porous network structure after crosslinking conditions. The viable cell count of Lactobacillus helveticus CICC 22536 (LC) encapsulated in crosslinking W/W Pickering emulsion after simulated gastrointestinal digestion was 7.563 × 10 7 CFU/mL, which was three orders of magnitude higher than that of naked cells.

Abstract:Probiotics serve a very important role in human health. However, probiotics have poor stability during processing, storage, and gastrointestinal digestion. The gellan gum (GG) is less susceptible to enzymatic degradation and resistant to thermal and acidic environments. This study investigated the effect of casein (CS)-GG emulsions to encapsulate Lactiplantibacillus plantarum CICC 6002 (L. plantarum CICC 6002) on its storage stability, thermal stability, and gastrointestinal digestion. L. plantarum CICC 6002 was suspended in palm oil and emulsions were prepared using CS or CS-GG complexes. We found the CS-GG emulsions improved the viability of L. plantarum CICC 6002 after storage, pasteurization, and digestion compared to the CS emulsions. In addition, we investigated the influence of the gellan gum concentration on emulsion stability, and the optimal stability was observed in the emulsion prepared by CS-0.8% GG complex. This study provided a new strategy for the protection of probiotics based on CS-GG delivery system.
Ultrasound-assisted multilayer Pickering emulsion fabricated by WPI-EGCG covalent conjugates for encapsulating probiotics in colon-targeted release

Abstract:This study demonstrated the influences of ultrasound-assisted multilayer Pickering double emulsion capsules on the pasteurization and gastrointestinal digestive viability of probiotic (L. plantarum) strain liquid. Firstly, the role of ultrasonic homogenization on the morphology of W1/O/W2 double emulsions were studied. The double emulsion formed by ultrasonic intensity at 285 W had a single and narrow distribution with smallest droplet size. The double emulsion particles were then coated with chitosan(Chi), alginate (Alg), and CaCl2(Ca). The multilayer emulsion after pasteurization and gastrointestinal digestion both had the highest viability at 5 coating layers, but its particle size (108.65 μm) exceeded the limit of human oral sensory (80 μm). It could be noted that the deposition of 3-4 layers of coating had similar activity after pasteurization/GIT digestion. And droplets with 3 layers of coating were the minimum and most available formulation for encapsulated probiotics (L. plantarum). Hence, the results suggest that the use of ultrasound-assisted multilayer emulsions encapsulated with probiotics in granular food and pharmaceutical applications is a promising strategy.

Abstract:In this study, the whey protein concentrate and xanthan gum complex obtained by specific pH treatment, along with κ-carrageenan (KC), were used to encapsulate Lactobacillus acidophilus JYLA-191 in an emulsion gel system. The effects of crosslinking and KC concentration on the visual characteristics, stability, mechanical properties, and formation mechanism of emulsion gels were investigated. The results of optical imaging, particle size distribution, and rheology exhibited that with the addition of crosslinking agents, denser and more homogeneous emulsion gels were formed, along with a relative decrease in the droplet size and a gradual increase in viscosity. Especially when the concentration of citric acid (CA) was 0.09 wt%, KC was 0.8 wt%, and K + was present in the system, the double-network emulsion gel was stable at high temperatures and in freezing environments, and the swelling ratio was the lowest (9.41 %). Gastrointestinal tract digestive treatments and pasteurization revealed that the probiotics encapsulated in the double-network emulsion gel had a higher survival rate, which was attributed to the synergistic cross-linking of CA and K + biopolymers to construct the emulsion gels. Overall, this study highlights the potential of emulsion gels to maintain probiotic vitality and provides valuable insights for developing inventive functional foods.

Abstract:The relationship of plasma levels of high density lipoprotein (HDL) cholesterol, apolipoproteins A-I and A-II (the major apolipoproteins in HDL), low density lipoprotein (LDL) cholesterol, triglyceride, and glucose to microangiopathy was evaluated in 49 insulin-dependent diabetic subjects. Although the HDL cholesterol/LDL cholesterol ratio (a risk determinant for macroangiopathy) was lower in women with proteinuria, no other relationships between HDL cholesterol or the A apolipoproteins and renal microangiography were found. The only independent association between HDL and retinal microangiopathy was found in women, where an inverse correlation was found between the apo A-I/apo A-II ratio and the number of microaneurysms (rs = −0.561, P < 0.05). Men showed strong relationships of glucose, triglyceride, cholesterol, and LDL cholesterol to renal microangiopathy whereas women, in general, had stronger correlations of these variables with retinal microangiopathy. Thus, several alterations in lipoprotein cholesterol distribution and HDL composition are associated with diabetic microangiopathy. In addition, differences between sexes suggest that previously undescribed hormonal factors may influence the severity of this process.
Combinedly increased viability of Lactiplantibacillus plantarum grx16 by co‐encapsulation of cryoprotectants and porous starch within calcium alginate capsules

Abstract:Summary Alginate‐based encapsulation is an effective probiotics delivery system in the intestine. However, due to calcium ions and gastric juice damaging to cell wall/membrane integrity during freeze‐drying and gastrointestinal ingestion, lots of cell loss their viability. This study, taking Lactiplantibacillus plantarum grx16 as an example, indicated that co‐encapsulation of cryoprotectants and porous starch within calcium alginate capsules could combinedly improve viability of probiotics encapsulated. Compared with calcium alginate‐protectant capsule (CAC‐Pro) (50.67%), combination of porous starch increased the encapsulation rate of PS0.8‐CAC‐Pro capsules to 82.68%. Meanwhile, the death rate constants of PS0.8‐CAC‐Pro capsules decreased by 31.3% (‐20 °C), 13.5% (4 °C), and 7.5% (25 °C) respectively. The survival rate of PS0.8‐CAC‐Pro capsules was 81.94% after continuous simulated gastrointestinal digestion, which was significantly higher than that of calcium alginate capsules (69.46%) and CAC‐Pro capsules (76.92%). This study provided an effective approach for modifying alginate capsule capsules to reduce the loss of probiotics viability.

Enhancement of gum Arabic/casein microencapsulation on the survival of Lactiplantibacillus plantarum in the stimulated gastrointestinal conditions

Abstract:Probiotic products that contain lactobacilli have long histories of safe use as Lactobacillus strains have many physiological functions in the gastrointestinal tract (GIT). However, the viability of probiotics can be affected by food processing and the adverse environment. This study investigated the O/W (Oil-in-water emulsions) emulsions formed by coagulation of casein/GA (Gum Arabic) complexes for Lactiplantibacillus plantarum microencapsulation, and the stability of the strains during gastrointestinal environment were also determined. The results showed that the particle size of the emulsion decreased from 9.72 μm to 5.48 μm when the GA concentration increased from 0 to 2 ( w / v ), and the emulsion particles were found to be more uniform as observed by CLSM (Confocal Laser Scanning Microscope). The surface of this microencapsulated casein/GA composite forms smooth, dense agglomerates and has high viscoelasticity, which effectively improved casein's emulsifying activity (8.66 ± 0.17 m 2 /g). After the casein/GA complexes microencapsulation, a higher viable count was detected after gastrointestinal digestion in vitro , and the activity of L. plantarum is more stable (about 7.51 log CFU/mL) during 35 days of storage at 4 °C. The results of study will help to design lactic acid bacteria encapsulation systems based on the GIT environment for the oral delivery strategy.

Abstract:Hybrid alginate-soy protein isolate-based hydrogel beads were prepared and evaluated to enhance the survival of the encapsulated probiotics (Lactobacillus plantarum) during heat processing to incorporate in mango juice. The solutions of sodium alginate-soy protein isolate (SA-SPI) with probiotic cells were dropped into the gelation bath containing calcium chloride (3% w/v) solution to develop various types of hydrogel beads. The level of survival of probiotics in encapsulated beads under acidic conditions (pH 2, 3, and 6.5) and bile salt (0.5 and 1.0% w/v) was evaluated. The survival of the encapsulated probiotics to thermal processing was evaluated by treating the beads in saline solution (0.9% w/v) at 30, 50, 63, and 72°C. The encapsulated probiotic bacteria were found alive even after treatment at 72°C for 90 s. Most of the free cells did not survive at the temperature higher than 50°C and very low pH (pH 2 and 3). The survival of probiotic cells was found higher with the hybrid hydrogel beads containing alginate and soy protein isolate (1:8 w/w). Furthermore, mango juice fortified with encapsulated L. plantarum in hydrogel beads was subjected to thermal pasteurization at 72°C for 90 s.
