Abstract:Background Diabetes mellitus is becoming a global health issue which demands a transformation of the research and healthcare practice for better patient management. To this end, the abundance of data and advancements in technology and artificial intelligence provides opportunities for such an endeavour. Aims This review aims to provide an overview of diabetes, artificial intelligence and the current research in diabetes which involve artificial intelligence in risk prediction, diagnosis, prognosis and prediction of complications. Conclusion Artificial intelligence is transforming diabetes research in many technical and organizational aspects. Although its deployment is still limited and is facing many challenges, it will likely impact future patient care by providing automated and personalized healthcare for patients.

Abstract:Artificial intelligence (AI) use in diabetes care is increasingly being explored to personalise care for people with diabetes and adapt treatments for complex presentations. However, the rapid advancement of AI also introduces challenges such as potential biases, ethical considerations, and implementation challenges in ensuring that its deployment is equitable. Ensuring inclusive and ethical developments of AI technology can empower both health-care providers and people with diabetes in managing the condition. In this Review, we explore and summarise the current and future prospects of AI across the diabetes care continuum, from enhancing screening and diagnosis to optimising treatment and predicting and managing complications.
Transforming Diabetes Care Through Artificial Intelligence: The Future Is Here

Abstract:An estimated 425 million people globally have diabetes, accounting for 12% of the world's health expenditures, and yet 1 in 2 persons remain undiagnosed and untreated. Applications of artificial intelligence (AI) and cognitive computing offer promise in diabetes care. The purpose of this article is to better understand what AI advances may be relevant today to persons with diabetes (PWDs), their clinicians, family, and caregivers. The authors conducted a predefined, online PubMed search of publicly available sources of information from 2009 onward using the search terms "diabetes" and "artificial intelligence." The study included clinically-relevant, high-impact articles, and excluded articles whose purpose was technical in nature. A total of 450 published diabetes and AI articles met the inclusion criteria. The studies represent a diverse and complex set of innovative approaches that aim to transform diabetes care in 4 main areas: automated retinal screening, clinical decision support, predictive population risk stratification, and patient self-management tools. Many of these new AI-powered retinal imaging systems, predictive modeling programs, glucose sensors, insulin pumps, smartphone applications, and other decision-support aids are on the market today with more on the way. AI applications have the potential to transform diabetes care and help millions of PWDs to achieve better blood glucose control, reduce hypoglycemic episodes, and reduce diabetes comorbidities and complications. AI applications offer greater accuracy, efficiency, ease of use, and satisfaction for PWDs, their clinicians, family, and caregivers.

Abstract:Artificial intelligence is growing quickly, and its application in the global diabetes pandemic has the potential to completely change the way this chronic illness is identified and treated. Machine learning methods have been used to construct algorithms supporting predictive models for the risk of getting diabetes or its complications. Social media and Internet forums also increase patient participation in diabetes care. Diabetes resource usage optimisation has benefited from technological improvements. As a lifestyle therapy intervention, digital therapies have made a name for themselves in the treatment of diabetes. Artificial intelligence will cause a paradigm shift in diabetes care, moving away from current methods and toward the creation of focused, data-driven precision treatment.

The goal of this article was to help people with diabetes, their clinicians, families, and caregivers better understand what AI breakthroughs are important now. AI medical applications include diagnosis, categorization, therapy, and robotics, among others. We are no longer confined to descriptive data analysis because we may acquire a larger by recognizing and anticipating patterns coming from inductive reasoning, owing to medical learning. AI applications have the potential to revolutionize diabetic care by assisting millions of people with diabetes in achieving better blood glucose management, reducing hyperglycemic episodes, and lowering diabetic morbidity and complications. We can see the evolution of closed-loop insulin delivery systems with inbuilt AI algorithms to safeguard both hypoglycemia and hyperglycemia in Type 1 diabetes, which have been relatively few attempts in management techniques for diabetes excursions. The medication you choose and how much you take depend on a variety of factors, including your body mass index, which influences beta-cell activity and insulin resistance, among other things. There are great assessments of research that have employed an AI approach to treat diabetes. The only way to deal with vast, diverse datasets is to rely solely on quantitative data. At this point, there are numerous issues with absolute reliance on quantitative data, depending on the frequently poor quality of this sort of information, as well as the necessity to complement combining a quantitative and qualitative approach. Attempting toward the transformation of unstructured data into digitally processed information data is a domain of cognitive computing that is predicted to grow in importance. To make a substantial contribution to AI aside from the purpose, there are 48 other factors to consider.
Synaptojanin Inhibition of Phospholipase D Activity by Hydrolysis of Phosphatidylinositol 4,5-Bisphosphate*

Abstract:A 150-kDa protein that inhibits phospholipase D (PLD) activity stimulated by ADP-ribosylation factor and phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2) was previously purified from rat brain. The sequences of peptides derived from the purified PLD inhibitor now identify it as synaptojanin, a nerve terminal protein that has been implicated in the endocytosis of fused synaptic vesicles and shown to be a member of the inositol polyphosphate 5-phosphatase family. Further characterization of the enzymatic properties of synaptojanin now shows that it hydrolyzes only the 5-phosphate from inositol 1,4,5-trisphosphate (I(1,4,5)P3) and that it does not catalyze the dephosphorylation of either I(1,3,4)P3 or inositol 1,4-bisphosphate. However, synaptojanin hydrolyzes both the 4- and 5-phosphates of PI(4,5)P2 and the 4-phosphate of phosphatidylinositol 4-phosphate, converting both compounds to phosphatidylinositol. Magnesium is required for the hydrolysis of I(1,4,5)P3, but not for that of phosphoinositides, by synaptojanin. The inhibition of PLD by synaptojanin is attributable to its ability to hydrolyze PI(4,5)P2. Synaptojanin did not inhibit PLD in the absence of PI(4,5)P2, and the extent of PLD inhibition was related to the extent of PI(4,5)P2hydrolysis in substrate vesicles. It has been proposed that the biosynthesis of PI(4,5)P2 and the activation of PLD by ADP-ribosylation factor constitute a positive loop to increase rapidly the concentrations of PI(4,5)P2 and phosphatidic acid (PA) during membrane vesiculation. The PA thus produced, probably together with PI(4,5)P2, facilitates vesicle coat assembly. The hydrolysis of PI(4,5)P2, and consequent inhibition of PLD, by synaptojanin might therefore constitute a mechanism to halt the positive loop connecting PI(4,5)P2 and PA during the endocytotic cycle of synaptic vesicles and serve as a signal for uncoating.

Abstract:Abstract Purpose of Review Artificial intelligence (AI) can make advanced inferences based on a large amount of data. The mainstream technologies of the AI boom in 2021 are machine learning (ML) and deep learning, which have made significant progress due to the increase in computational resources accompanied by the dramatic improvement in computer performance. In this review, we introduce AI/ML-based medical devices and prediction models regarding diabetes. Recent Findings In the field of diabetes, several AI-/ML-based medical devices and regarding automatic retinal screening, clinical diagnosis support, and patient self-management tool have already been approved by the US Food and Drug Administration. As for new-onset diabetes prediction using ML methods, its performance is not superior to conventional risk stratification models that use statistical approaches so far. Summary Despite the current situation, it is expected that the predictive performance of AI will soon be maximized by a large amount of organized data and abundant computational resources, which will contribute to a dramatic improvement in the accuracy of disease prediction models for diabetes.

Abstract:The utilization of artificial intelligence (AI) in diabetes care has focused on early intervention and treatment management. Notably, usage has expanded to predict an individual's risk for developing type 2 diabetes. A scoping review of 40 studies by Mohsen et al. shows that while most studies used unimodal AI models, multimodal approaches were superior because they integrate multiple types of data. However, creating multimodal models and determining model performance are challenging tasks given the multi-factored nature of diabetes. For both unimodal and multimodal models, there are also concerns of bias with the lack of external validations and representation of race, age, and gender in training data. The barriers in data quality and evaluation standardization are ripe areas for developing new technologies, especially for entrepreneurs and innovators. Collaboration amongst providers, entrepreneurs, and researchers must be prioritized to ensure that AI in diabetes care is providing quality and equitable patient care.

Abstract:Background: Artificial intelligence methods in combination with the latest technologies, including medical devices, mobile computing, and sensor technologies, have the potential to enable the creation and delivery of better management services to deal with chronic diseases. One of the most lethal and prevalent chronic diseases is diabetes mellitus, which is characterized by dysfunction of glucose homeostasis. Objective: The objective of this paper is to review recent efforts to use artificial intelligence techniques to assist in the management of diabetes, along with the associated challenges. Methods: A review of the literature was conducted using PubMed and related bibliographic resources. Analyses of the literature from 2010 to 2018 yielded 1849 pertinent articles, of which we selected 141 for detailed review. Results: We propose a functional taxonomy for diabetes management and artificial intelligence. Additionally, a detailed analysis of each subject category was performed using related key outcomes. This approach revealed that the experiments and studies reviewed yielded encouraging results. Conclusions: We obtained evidence of an acceleration of research activity aimed at developing artificial intelligence-powered tools for prediction and prevention of complications associated with diabetes. Our results indicate that artificial intelligence methods are being progressively established as suitable for use in clinical daily practice, as well as for the self-management of diabetes. Consequently, these methods provide powerful tools for improving patients' quality of life.

Abstract:Context: The diagnostic methods for diabetes mellitus (DM), a chronic metabolic disorder characterized by elevated blood sugar levels, are rapidly evolving thanks to artificial intelligence (AI), particularly machine learning (ML) and deep learning (DL). This review explores the applications of AI in risk assessment and diagnosing different types of diabetes. Evidence Acquisition: The review highlights the effectiveness of various ML models, including support vector machines (SVMs), random forests (RFs), and DL techniques like convolutional neural networks (CNNs), in achieving high diagnostic accuracy. Challenges include limited data availability, interpretability of complex models, and the need for standardized performance metrics. Results: Machine learning methods like SVMs and RFs are highly effective at diagnosing different types of diabetes, and DL techniques like CNNs also show great promise. Conclusions: Overall, AI has immense potential to revolutionize diabetes diagnosis by facilitating risk assessment and early detection, improving treatment efficacy, and preventing severe complications.
Radioprotective effects of miso (fermented soy bean paste) against radiation in B6C3F1 mice: increased small intestinal crypt survival, crypt lengths and prolongation of average time to death.

Abstract:The radioprotective effect of miso, a fermentation product from soy bean, was investigated with reference to the survival time, crypt survival and jejunum crypt length in male B6C3F1 mice. Miso at three different fermentation stages (early-, medium- and long-term fermented miso) was mixed in MF diet into biscuits at 10% and was administered from 1 week before irradiation. Animal survival in the long-term fermented miso group was significantly prolonged as compared with the short-term fermented miso and MF cases after 8 Gy of 60Co-gamma-ray irradiation at a dose rate of 2Gy min(-1). Delay in mortality was evident in all three miso groups, with significantly increased survival. At doses of 10 and 12 Gy X-irradiation at a dose rate of 4 Gy min(-1), the treatment with long-term fermented miso significantly increased crypt survival. Also the protective influence against irradiation in terms of crypt lengths in the long-term fermented miso group was significantly greater than in the short-term or medium-term fermented miso and MF diet groups. Thus, prolonged fermentation appears to be very important for protection against radiation effects.

Abstract:Diabetes mellitus, stemming from either insulin resistance or inadequate insulin secretion, represents a complex ailment that results in prolonged hyperglycemia and severe complications. Patients endure severe ramifications such as kidney disease, vision impairment, cardiovascular disorders, and susceptibility to infections, leading to significant physical suffering and imposing substantial socio-economic burdens. This condition has evolved into an increasingly severe health crisis. There is an urgent need to develop new treatments with improved efficacy and fewer adverse effects to meet clinical demands. However, novel drug development is costly, time-consuming, and often associated with side effects and suboptimal efficacy, making it a major challenge. Artificial Intelligence (AI) and Machine Learning (ML) have revolutionized drug development across its comprehensive lifecycle, spanning drug discovery, preclinical studies, clinical trials, and post-market surveillance. These technologies have significantly accelerated the identification of promising therapeutic candidates, optimized trial designs, and enhanced post-approval safety monitoring. Recent advances in AI, including data augmentation, interpretable AI, and integration of AI with traditional experimental methods, offer promising strategies for overcoming the challenges inherent in AI-based drug discovery. Despite these advancements, there exists a notable gap in comprehensive reviews detailing AI and ML applications throughout the entirety of developing medications for diabetes mellitus. This review aims to fill this gap by evaluating the impact and potential of AI and ML technologies at various stages of diabetes mellitus drug development. It does that by synthesizing current research findings and technological advances so as to effectively control diabetes mellitus and mitigate its far-reaching social and economic impacts. Towards Personalized AI-Based Diabetes Therapy: A Review

Abstract:Insulin pumps and other smart devices have recently made significant advancements in the treatment of diabetes, a disorder that affects people all over the world. The development of medical AI has been influenced by AI methods designed to help physicians make diagnoses, choose a course of therapy, and predict outcomes. In this article, we thoroughly analyse how AI is being used to enhance and personalize diabetes treatment. The search turned up 77 original research papers, from which we've selected the most crucial information regarding the learning models employed, the data typology, the deployment stage, and the application domains. We identified two key trends, enabled mostly by AI: patient-based therapy personalization and therapeutic algorithm optimization. In the meanwhile, we point out various shortcomings in the existing literature, like a lack of multimodal database analysis or a lack of interpretability. The rapid improvements in AI and the expansion of the amount of data already available offer the possibility to overcome these difficulties shortly and enable a wider deployment of this technology in clinical settings.
Overview of Artificial Intelligence–Driven Wearable Devices for Diabetes: Scoping Review

Abstract:Background: Prevalence of diabetes has steadily increased over the last few decades with 1.5 million deaths reported in 2012 alone. Traditionally, analyzing patients with diabetes has remained a largely invasive approach. Wearable devices (WDs) make use of sensors historically reserved for hospital settings. WDs coupled with artificial intelligence (AI) algorithms show promise to help understand and conclude meaningful information from the gathered data and provide advanced and clinically meaningful analytics. Objective: This review aimed to provide an overview of AI-driven WD features for diabetes and their use in monitoring diabetes-related parameters. Methods: We searched 7 of the most popular bibliographic databases using 3 groups of search terms related to diabetes, WDs, and AI. A 2-stage process was followed for study selection: reading abstracts and titles followed by full-text screening. Two reviewers independently performed study selection and data extraction, and disagreements were resolved by consensus. A narrative approach was used to synthesize the data. Results: From an initial 3872 studies, we report the features from 37 studies post filtering according to our predefined inclusion criteria. Most of the studies targeted type 1 diabetes, type 2 diabetes, or both (21/37, 57%). Many studies (15/37, 41%) reported blood glucose as their main measurement. More than half of the studies (21/37, 57%) had the aim of estimation and prediction of glucose or glucose level monitoring. Over half of the reviewed studies looked at wrist-worn devices. Only 41% of the study devices were commercially available. We observed the use of multiple sensors with photoplethysmography sensors being most prevalent in 32% (12/37) of studies. Studies reported and compared >1 machine learning (ML) model with high levels of accuracy. Support vector machine was the most reported (13/37, 35%), followed by random forest (12/37, 32%). Conclusions: This review is the most extensive work, to date, summarizing WDs that use ML for people with diabetes, and provides research direction to those wanting to further contribute to this emerging field. Given the advancements in WD technologies replacing the need for invasive hospital setting devices, we see great advancement potential in this domain. Further work is needed to validate the ML approaches on clinical data from WDs and provide meaningful analytics that could serve as data gathering, monitoring, prediction, classification, and recommendation devices in the context of diabetes.

Abstract:Introduction Diabetic retinopathy is one of the leading causes of avoidable blindness among adults globally, and screening programmes can enable early diagnosis and prevention of progression. Artificial intelligence (AI) diagnostic solutions have been developed to diagnose diabetic retinopathy. The aim of this review is to identify ethical concerns related to AI‐enabled diabetic retinopathy diagnostics and enable future research to explore these issues further. Methods This is a narrative review that uses thematic analysis methods to develop key findings. We searched two databases, PubMed and Scopus, for papers focused on the intersection of AI, diagnostics, ethics, and diabetic retinopathy and conducted a citation search. Primary research articles published in English between 1 January 2013 and 14 June 2024 were included. From the 1878 papers that were screened, nine papers met inclusion and exclusion criteria and were selected for analysis. Results We found that existing literature highlights ensuring patient data has appropriate protection and ownership, that bias in algorithm training data is minimised, informed patient decision‐making is encouraged, and negative consequences in the context of clinical practice are mitigated. Conclusions While the technical developments in AI‐enabled diabetic retinopathy diagnostics receive the bulk of the research focus, we found that insufficient attention is paid to how this technology is accessed equitably in different settings and which safeguards are needed against exploitative practices. Such ethical issues merit additional exploration and practical problem‐solving through primary research. AI‐enabled diabetic retinopathy screening has the potential to enable screening at a scale that was previously not possible and could contribute to reducing preventable blindness. It will only achieve this if ethical issues are emphasised, understood, and addressed throughout the translation of this technology to clinical practice.

Abstract:Abstract Background and aim Artificial intelligence (AI) has emerged as a promising technology in the field of endocrinology, offering significant potential to revolutionize the diagnosis, treatment, and management of endocrine disorders. This comprehensive review aims to provide a concise overview of the current landscape of AI applications in endocrinology and metabolism, focusing on the fundamental concepts of AI, including machine learning algorithms and deep learning models. Methods The review explores various areas of endocrinology where AI has demonstrated its value, encompassing screening and diagnosis, risk prediction, translational research, and “pre-emptive medicine”. Within each domain, relevant studies are discussed, offering insights into the methodology and main findings of AI in the treatment of different pathologies, such as diabetes mellitus and related disorders, thyroid disorders, adrenal tumors, and bone and mineral disorders. Results Collectively, these studies show the valuable contributions of AI in optimizing healthcare outcomes and unveiling new understandings of the intricate mechanisms underlying endocrine disorders. Furthermore, AI-driven approaches facilitate the development of precision medicine strategies, enabling tailored interventions for patients based on their individual characteristics and needs. Conclusions By embracing AI in endocrinology, a future can be envisioned where medical professionals and AI systems synergistically collaborate, ultimately enhancing the lives of individuals affected by endocrine disorders.

Abstract:Artificial intelligence and machine learning are driving a paradigm shift in medicine, promising data-driven, personalized solutions for managing diabetes and the excess cardiovascular risk it poses. In this comprehensive review of machine learning applications in the care of patients with diabetes at increased cardiovascular risk, we offer a broad overview of various data-driven methods and how they may be leveraged in developing predictive models for personalized care. We review existing as well as expected artificial intelligence solutions in the context of diagnosis, prognostication, phenotyping, and treatment of diabetes and its cardiovascular complications. In addition to discussing the key properties of such models that enable their successful application in complex risk prediction, we define challenges that arise from their misuse and the role of methodological standards in overcoming these limitations. We also identify key issues in equity and bias mitigation in healthcare and discuss how the current regulatory framework should ensure the efficacy and safety of medical artificial intelligence products in transforming cardiovascular care and outcomes in diabetes.
