Abstract:Antibiotic contamination has become a global environmental issue of widespread concern, among which oxytetracycline contamination is very severe. In this study, earthworm (Eisenia fetida) was exposed to oxytetracycline to study its impact on the soil environment. The total protein (TP), catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), malondialdehyde (MDA), glutathione S-transferase (GST), and glutathione peroxidase (GPX) oxidative stress indicators in earthworms were measured, and the integrated biomarker response (IBR) approach was used to evaluate the toxic effect of oxytetracycline on earthworms. A Technique for Order Preference by Similarity to an Ideal Solution (TOPSIS) and a path analysis model were used to explore the physiological and metabolic processes of earthworms after stress occurs. The results showed that SOD, GPX, and GST play important roles in resisting oxytetracycline stress. In addition, stress injury showed a good dose–effect relationship, and long-term stress from pollutants resulted in the most serious damage to the head tissue of earthworms. These results provide a theoretical basis for understanding the toxic effect of oxytetracycline on soil animals, monitoring the pollution status of oxytetracycline in soil, and conducting ecological security risk assessment.

Microbial Response to Repeated Treatments of Manure Containing Sulfadiazine and Chlortetracycline in Soil

Abstract:Substantive addition of antibiotic-contaminated manure to agricultural soil may lead to "persistent" residues of antibiotics and may affect soil health. Therefore, this study examines the effects of repeated manure treatments containing sulfadiazine (SDZ) and chlortetracycline (CTC) residues, both individually and combined, on the functional diversity and structure of soil microbial communities in the soils under laboratory conditions. The average well color development (AWCD), Simpson diversity index (1/D, dominant populations), Shannon-Wiener diversity index (H', richness), and McIntosh diversity index (U, evenness) in the antibiotics-treated soils decreased in the first 60-day treatment and then gradually recovered or even exceeded the initial level in the unamended soils with increasing treatment frequency. A total of 11 specific bands in temperature gradient gel electrophoresis (TGGE) profiles were observed and sequence analyzed for five repeated treatments, and most of them belonged to the phyla Firmicutes, Actinobacteria, and Proteobacteria. These results indicate that repeated treatments of manure containing SDZ and CTC residues can alter soil microbial community structure, although they have a temporary suppression effect on soil microbial functional diversity.

Abstract:Antibiotic contamination caused by the long–term use of organic manure (OM) in greenhouse agricultural soils poses potential detrimental effects to the soil environment. By applying OM containing chlortetracycline (CTC) and/or ciprofloxacin (CIP) ten times in soil under laboratory conditions, we investigated the dissipation and accumulation characteristics of CTC and CIP in the soil, the changes in the microbial pollution–induced community tolerance (PICT), and the diversity and abundance of antibiotic resistance genes (ARGs) in the soil microbiome. The dissipation of CTC was rapid while CIP was accumulated in repeatedly treated soils; further, CIP could inhibit the dissipation of CTC. Meanwhile, the PICT to CTC and/or CIP significantly increased up to 15.0–fold after ten successive treatments compared to that in the first treatment. As the treatment frequency increased, significant upward trends in the abundances of tetracycline resistance genes tetA(G), tetX2, tetX, tetG, tetA(33), tetA, tetW, and tetA(P), fluoroquinolone resistance gene qnrA6, and multiple resistance gene mexF were revealed by both metagenomic and qPCR analyses. The findings demonstrated that repeated treatments with CTC and/or CIP can alter the dissipation rate, promote an increase in PICT to CTC and/or CIP, and increase the ARGs abundance in steps.
Exploring Microbial Community Structure and Biological Function in Manured Soil During Ten Repeated Treatments with Chlortetracycline and Ciprofloxacin.

Abstract:The changes of enzyme activities, microbial community structure and function, and the diversity and resistance level of antibiotic-resistant bacteria (ARB) were studied in soil during ten repeated treatments with chlortetracycline (CTC) and/or ciprofloxacin (CIP) together with organic manure (OM) under laboratory conditions. The activities of neutral phosphatase (NPA) and catalase (CAT) displayed the suppression-recovery-stimulation trend in the OM&CTC treatment but the stimulation trend in the OM&CTC&CIP treatment. The NPA was stimulated but the CAT was little affected in the OM&CIP treatment. Soil microbial functional diversity displayed the suppression-recovery-stimulation trend in the OM&CTC and OM&CTC&CIP treatments and the stimulation-suppression trend in the OM&CIP treatment with the treatment frequency. Metagenomic analysis showed that the relative abundances of Actinobacteria and Firmicutes in the antibiotic treatment significantly increased by 0.5-235.6%, but that of Proteobacteria decreased by 0.2-27.3% compared to the control with the treatment frequency. Furthermore, the relative abundances of dominant bacterial genera including Streptomyces, Actinomadura, Mycobacterium, and Streptococcus in the antibiotic treatment significantly increased by 1.1-10433.3% compared to the control. Meanwhile, repeated antibiotic treatments induced a significant increase in the diversity and resistance level of ARB isolates, especially in the OM&CTC treatment. It is concluded that repeated treatments with CTC and/or CIP can alter enzyme activities, microbial community structure and function, and increase the diversity and resistance level of ARB isolates.

Abstract:The degradation characteristics of carbendazim (CBD) and chlortetracycline (CTC) in individual and combined treatments, and dynamics of soil microbial structural and functional diversity as well as their potential relations were studied during three repeated treatments using different concentrations. The results showed that the degradation half-life of CBD at concentrations of 3mg/kg and 6mg/kg obviously increased, but that of CTC at levels of 1mg/kg and 10mg/kg decreased with increasing treatment frequency. Soil microbial activity and functional diversity displayed the suppression trend in CBD treatment and the suppression-recovery-stimulation trend in CTC and CBD+CTC treatments, which were consistent with the findings of decreased degradation rate of CBD and increased degradation rate of CTC. 16S amplicon sequencing analysis revealed five potentially dominant CTC-resistant microbial genera including Bacillus, Actinobacillus, Pseudomonas, Mycobacterium, and Corynebacterium, which may mainly carry major facilitator superfamily transporter protein, ribosomal protection protein, and other proteins encoded by tetA, tetB, tetC, tetH, tetL, tetM, tetO, tetV, tetW, tetX, tetZ, tet33, and tet39. These five dominant genera may jointly contribute to the elevated bacterial community resistance to CTC. Our findings provided a better understanding of microbial community structure and function changes in repeatedly treated soils with CBD and CTC.
Combined Effects of Cadmium and Sulfamethoxazole on Eisenia Fetida: Insights into Accumulation, Subcellular Partitioning, Biomarkers and Toxicological Responses

Abstract:Cadmium (Cd) and sulfamethoxazole (SMX) frequently coexist in farmlands, yet their synergistic toxicological impacts on terrestrial invertebrates remain unexplored. In this study, earthworms were exposed to artificial soils percolated with Cd (5 mg/kg), SMX (5 mg/kg) or combination of them for 7 days, followed by a 12-day elimination phase in uncontaminated soil. The uptake of Cd and SMX by the earthworms, along with their subcellular distribution, was meticulously analyzed. Additionally, a suite of biomarkers-including superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA), and weight loss-were evaluated to assess the health status of the earthworms and the toxicological effects of the Cd and SMX mixture. Notably, the cotreatment with Cd and SMX resulted in a significantly higher weight loss in Eisenia fetida (41.25 %) compared to exposure to Cd alone (26.84 %). Moreover, the cotreatment group exhibited substantially higher concentrations of Cd in the total internal body, fraction C (cytosol), and fraction E (tissue fragments and cell membranes) in Eisenia fetida compared to Cd alone counterparts. The combined exposure also significantly elevated the SMX levels in the total body and fraction C compared with the SMX-only treated earthworms. Additionally, Eisenia fetida subjected to the combined treatment showed markedly increased activities of SOD, CAT, and MDA compared to those treated with Cd alone. The effect addition indices (EAIs), ranging from 1.00 to 2.23, unequivocally demonstrated a synergistic effect of the combined treatments. Interestingly, relocating the earthworms to clean soil did not mitigate the observed adverse effects. These findings underscore the increased risk posed by the Cd-SMX complex to terrestrial invertebrates in agricultural areas.
Study on the regulation of earthworm physiological function under cadmium stress based on a compound mathematical model

Abstract:A cadmium (Cd) stress test was carried out on Eisenia fetida in artificial soil. Six Cd concentration gradient solutions (0, 50, 100, 125, 250 and 500 mg/kg) were prepared. Two treatment groups, short-term stress and long-term stress, were established. The former lasted for 10 days, and the latter lasted for 30 days. The Biolog ECO-microplate culture method was used to determine the utilization of the 31 carbon sources by the microbes in earthworm homogenate. The total protein content (TP), peroxidase activity (POD), catalase activity (CAT), superoxide dismutase activity (SOD), glutathione peroxidase activity (GPX), glutathione-S-transferase activity (GST), malondialdehyde content (MDA) and acetylcholinesterase activity (AChE) in earthworm were determined in order to investigate the regulation of oxidative stress and the functional diversity of microbial communities in earthworms under Cd stress. By combining the entropy weight method (EW) and the technique for order preference by similarity to an ideal solution model (TOPSIS), the physiological functional indices of earthworms were assessed objectively and scientifically, and the physiological changes under the different stress periods were evaluated. The results showed that a Cd-tolerant dominant population appeared in the microbial community under Cd stress. In the short-term test, oxidative stress were more effective in coping with Cd stress than the microbial community, and oxidative stress regulated the microbial community functional diversity. Under long-term Cd stress, the regulatory effect was weak or non-existent. In this study, a new evaluation model was established to explore the regulation process of earthworm on its oxidation stress and the functional diversity of microbial communities under Cd stress, and provide a theoretical basis for revealing the detoxification mechanism of earthworms.
A Compositional Shift in the Soil Microbiome Induced by Tetracycline, Sulfamonomethoxine and Ciprofloxacin Entering a Plant-Soil System.

Abstract:Antibiotics entering the soil likely disturb the complex regulatory network of the soil microbiome, which is closely associated with soil quality and ecological function. This study investigated the effects of tetracycline (TC), sulfamonomethoxine (SMM), ciprofloxacin (CIP) and their combination (AM) on the bacterial community in a soil-microbe-plant system and identified the main bacterial responders. Antibiotic effects on the soil microbiome depended on antibiotic type and exposure time. TC resulted in an acute but more rapidly declining effect on soil microbiome while CIP and SMM led to a delayed antibiotic effect. The soil exposed to AM presented a highly similar bacterial structure to that exposed to TC rather than to SMM and CIP. TC, SMM and CIP had their own predominantly impacted taxonomic groups that include both resistance and sensitive bacteria. The antibiotic sensitive responders predominantly distributed within the phylum Proteobacteria. The potential bacteria resistant to each antibiotic exhibited phyla preference to some extent, particularly those resistant to TC. CIP and SMM resistance in soil was increased with exposure time while TC resistance gave the opposite result. Overall, the work extended the understanding of antibiotic effects on soil microbiome after introduced into the soil during greenhouse vegetable cultivation.

Abstract:This study investigated soil microbial responses to the application of tetracycline (TC), sulfamonomethoxine (SMM), and ciprofloxacin (CIP) alone and in combination in a soil culture pot experiment conducted at Hangzhou, China. Multiple approaches were applied for a better and complete depiction. Among the three antibiotics, SMM has a lowest dissipation and shows a most dramatic inhibition on microbial community and metabolism diversity. The combined application (AM) of SMM, CIP, and TC improved the dissipation of each antibiotic; similarly, SMM- and CIP-resistant bacteria showed larger populations in the AM than all single applications. Soils accumulated a large content of NO3–N at day 20 after multi-antibiotics perturbation. All antibiotics stimulated soil basal respirations and inhibited soil metabolism diversity, whereas the interruption exerted by SMM and AM lasted for a longer time. Six nitrogen-cycling genes including chiA, amoA, nifH, nirK, nirS, and narG were quantified and found to decrease owing to both single- and multi-antibiotics perturbation. Overall, AM was most interruptive for soils, followed by SMM perturbation, while other antibiotics could be less interruptive. These results provide systematic insights into how soil microbial systems would shift under each single- or multi-antibiotics perturbation.
The changes of antioxidant system and intestinal bacteria in earthworms (Metaphire guillelmi) on the enhanced degradation of tetracycline

Abstract:Tetracycline (TC) in soil severely imperils food security and ecosystem function. Metaphire guillelmi is a common species in farmland. It could impact the degradation of antibiotics. However, how it affects is rarely unknown. Hence, the present study aimed to investigate the effects of M. guillelmi on the TC degradation in soil and the changes of the antioxidant system and intestinal bacteria in M. guillelmi. The treatments that M. guillelmi was inoculated on soil contaminated with different TC concentrations were contrasted with those without M. guillelmi. After 21 days, the degradation rate of TC significantly increased by 13.70%, 18.14% and 29.01% at 10, 50 and 100 mg kg -1 TC dose, respectively, due to the inoculation of M. guillelmi. The half-life of TC was also shortened nearly by 1/3 to 2/3. Superoxide dismutase (SOD) increased in a dose-dependent manner with the increase of TC concentration on the 7th and 14th day. Catalase (CAT) and glutathione S-transferase (GST) presented an inverted U-shaped dose response on the 7th day, and the peak of enzyme activities occurred at TC concentration of 0.1, 1 mg kg -1 (CAT) and 0.1 mg kg -1 (GST). Malondialdehyde (MDA) contents did not change significantly. At the phylum level, only Verrucomicrobia significantly decreased under 1 mg kg -1 and 100 mg kg -1 TC dose. Genus Paracoccus, Singulisphaera, Acinetobacter and Bacillus significantly increased and became the dominant bacterium during the TC degradation. Overall, the antioxidant system and intestinal bacteria of M. guillelmi were affected by the different concentrations of TC pollution, which provided new ideas for the research of mechanism of TC degradation by earthworms in the future.

Abstract:Soil toxicological tests are commonly performed using Eisenia fetida as the standard earthworm species, but it is tolerant to a wide range of pollutants. Therefore, the inclusion of susceptible species is crucial for the accurate estimation of soil contamination. In this study, we examined the sensitivity to soil cadmium (Cd) of anecic Metaphire guillelmi and epigeic E. fetida by measuring multiple indexes of oxidative stress. Using subcellular partitioning analysis, we further elucidated the inherent mechanism underlying the species-specific sensitivity of the two earthworm species. Among the battery of biochemical indexes, reactive oxygen species and protein carbonyl groups served as sensitive biomarkers. According to their respective response thresholds, M. guillelmi was more sensitive than E. fetida and they differed in their dose-response relationships. In E. fetida, the activities of three antioxidant enzymes, superoxide dismutase (SOD), catalase (CAT) and glutathione S-transferase (GST), exhibited a hormesis-like U-shaped dose-response relationship, while in M. guillelmi SOD, glutathione peroxidase (an analogue of CAT) and GST showed an inverted U-shaped relationship. The concentrations of Cd in the subcellular fractions and whole body of the earthworms well fit (R2>0.9) a saturation model versus bioavailable Cd concentrations determined by the diffusive gradients in thin films technique. Despite the lower accumulation capacity of M. guillelmi, the Cd-binding capacity (Cmax) of its subcellular heat-stable protein fraction, the so-called biologically detoxified metal pool, was 2.7 times lower than that of E. fetida, whereas the Cd binding affinity (logK) of its heat-denatured protein fraction, i.e. the metal-sensitive fraction, was 3.0 times higher, which accounted for the high susceptibility of M. guillelmi to soil Cd. Our results suggest that because of their sensitivity, as exemplified by M. guillelmi, native earthworm species should be taken into account in soil risk assessments to avoid underestimation of the toxicity of various pollutants.
Effects of sulfamethazine and tetracycline at molecular, cellular and tissue levels in Eisenia fetida earthworms

Abstract:Soil contamination by antibiotics is a global issue of great concern that contributes to the rise of bacterial antibiotic resistance and can have toxic effects on non-target organisms. This study evaluated the variations of molecular, cellular, and histological parameters in Eisenia fetida earthworms exposed to sulfamethazine (SMZ) and tetracycline (TC), two antibiotics commonly found in agricultural soils. The earthworms were exposed for 14 days to a series of concentrations (0, 10, 100, and 1000 mg/kg) of both antibiotics. SMZ and TC did not affect the survival of E. fetida, however, other effects at different levels of biological complexity were detected. The two highest concentrations of SMZ reduced the viability of coelomocytes. At the highest TC concentration, there was a noticeable decline in cell viability, acetylcholinesterase activity (neurotoxicity), and the relative presence of mucopolysaccharides in the epidermis (mucous production). Glutathione S-transferase activity decreased in all TC treatments and at the highest SMZ concentration. However, levels of malondialdehyde and protein carbonyls did not change, suggesting an absence of oxidative stress. Tetracycline was neurotoxic to E. fetida and changed the integrity of the epidermis. Both antibiotics altered the intestinal microbiota of E. fetida, leading to a reduction in the relative abundance of bacteria from the phyla Proteobacteria and Bacteroidetes, while causing an increase in the phylum Actinobacteroidota. All observed changes indicate that both SMZ and TC can disrupt the earthworms' immune system and gut microbiome, while fostering the growth of bacteria that harbour antibiotic resistance genes. Finally, both antibiotics exerted additional metabolic and physiological effects that increased the vulnerability of E. fetida to pathogens.
Study on the regulatory mechanism of the earthworm microbial community in vitro and in vivo under cadmium stress

Abstract:In this paper, cadmium (Cd) stress tests were performed on Eisenia fetida in sterile artificial soil, and its regulatory mechanism between microbial communities in vivo and in vitro after Cd stress was explored. In the test, 0, 50, 100, 125, 250 and 500 mg.kg-1 Cd stress concentrations were implemented. After long-term and short-term stress, the microbes in the earthworms and the soil were cultured with ECO plates. The data statistics of carbon source utilization intensity were carried out using the method developed by our team. CCA was scientifically integrated into TOPSIS to establish a new data analysis model to find the regulatory nodes after stress[<a class="workspace-trigger" href="#bib1">1</a>]. Macro gene sequencing technology revealed that the species with the highest absolute abundance in the microbial communities in vivo and in vitro were all unnamed new species. It was confirmed that the HBA gene, NEUROD1 gene and ABCA3 gene were the regulatory genes of the microbial community in the earthworms under Cd stress, while the TC.FEV.OM gene and cheBR gene were the main regulatory genes of the microbial community in the soil. These results provide a scientific and theoretical reference and model basis for the bioremediation of Cd-contaminated soil and the detoxification mechanism of earthworms.

Abstract:Globally increasing trace metal contamination of soils requires a better mechanistic understanding of metal-stress impacts on microbially mediated nutrient cycling. Herein, a 5-month laboratory experiment was employed to assess the effects of cadmium (Cd) on soil microbial N-cycling processes and associated functional gene abundance, with and without urea amendment. In non-N-amended soils, Cd progressively stimulated microbial populations for N acquisition from initial dissolved organic N (DON) to later recalcitrant organic N. The acceleration of N catabolism was synchronously coupled with C catabolism resulting in increased CO2/N2O fluxes and adenosine triphosphate (ATP) contents. The abundance of microbes deemed inefficient in N catabolism was gradually repressed after an initial stimulation period. We posit that enhanced exergonic N processes diminished the need for endergonic activities as a survival strategy for N communities experiencing metal stress. With urea amendment, Cd exhibited an initial stimulation effect on soil nitrification and a later a promotion effect on mineralization, along with an increase in the associated microbial populations. In N-amended soils, Cd accelerated N/C transformation processes, but decreased N2O and CO2 fluxes by 19 and 14%, respectively. This implies that under eutrophic conditions, Cd synchronously altered microbial C/N metabolism from a dominance of catabolic to anabolic processes. Effects of Two Ecological Earthworm Species on Tetracycline Degradation Performance, Pathway and Bacterial Community Structure in Laterite Soil.

Abstract:This study explored the change of tetracycline degradation efficiency, metabolic pathway, soil physiochemical properties and degraders in vermiremediation by two earthworm species of epigeic Eisenia fetida and endogeic Amynthas robustus. We found a significant acceleration of tetracycline degradation in both earthworm treatments, and 4-epitetracycline dehydration pathway was remarkably enhanced only by vermiremediation. Tetracycline degraders from soils, earthworm intestines and casts were different. Ralstonia and Sphingomonas were potential tetracycline degraders in soils and metabolized tetracycline through direct dehydration pathway. Degraders in earthworm casts (Comamonas, Acinetobacter and Stenotrophomonas) and intestines (Pseudomonas and Arthrobacter) dehydrated 4-epitetracycline into 4-epianhydrotetracycline. More bacterial lineages resisting tetracycline were found in earthworm treatments, indicating the adaptation of soil and intestinal flora under tetracycline pressure. Earthworm amendment primarily enhanced tetracycline degradation by neutralizing soil pH and consuming organic matters, stimulating both direct dehydration and epimerization-dehydration pathways. Our findings proved that vermicomposting with earthworms is effective to alter soil microenvironment and accelerate tetracycline degradation, behaving as a potential approach in soil remediation at tetracycline contaminated sites.

Abstract:Trifloxystrobin is a new type of fungicide, which is extensively used due to its excellent antifungal activity. In this study, oxidative stress and DNA damage induced by trifloxystrobin exposure was evaluated using Eisenia fetida at subchronic toxicity concentrations in artificial soil and brown soil (0.1–2.5 mg/kg). Throughout the exposure period (days 7, 28 and 56), six biochemical indicators including reactive oxygen species (ROS), antioxidant enzymes (SOD and CAT), glutathione S-transferase (GST), lipid peroxidation and DNA damage (8-hydroxydeoxyguanosine) were measured. In addition, the integrated biomarker response (IBR) index was calculated to make comparison of toxicological response between artificial and brown soils. Results indicated that trifloxystrobin can induce oxidative stress and DNA damage to earthworms with subchronic toxicity greater in brown soil compared to artificial soil as determined through integrated calculations for six biochemical indicators. Trifloxystrobin toxicological experiments in artificial soil may not accurately evaluate its toxicity in natural soil ecosystems, as the toxicity of trifloxystrobin to Eisenia fetida was underestimated.

Abstract:Tetracycline pollution is common in Chinese arable soils, and vermicomposting is an effective approach to accelerate tetracycline bioremediation. However, current studies mainly focus on the impacts of soil physicochemical properties, microbial degraders and responsive degradation/resistance genes on tetracycline degradation efficiencies, and limited information is known about tetracycline speciation in vermicomposting. This study explored the roles of epigeic E. fetida and endogeic A. robustus in altering tetracycline speciation and accelerating tetracycline degradation in a laterite soil. Both earthworms significantly affected tetracycline profiles in soils by decreasing exchangeable and bound tetracycline but increasing water soluble tetracycline, thereby facilitating tetracycline degradation efficiencies. Although earthworms increased soil cation exchange capacity and enhanced tetracycline adsorption on soil particles, the significantly elevated soil pH and dissolved organic carbon benefited faster tetracycline degradation, attributing to the consumption of soil organic matter and humus by earthworms. Different from endogeic A. robustus which promoted both abiotic and biotic degradation of tetracycline, epigeic E. foetida preferently accelerated abiotic tetracyline degradation. Our findings described the change of tetracycline speciation during vermicompsiting process, unraveled the mechanisms of different earthworm types in tetracycline speciation and metabolisms, and offered clues for effective vermiremediation application at tetracycline contaminated sites.
Overlooked interconversion between tetracyclines and their 4-epimers in soil and effects on soil resistome and bacterial community

Abstract:With the widespread use of tetracycline antibiotics (TCs) and the application of manure fertilizer in farmland, TCs and their metabolites especially 4-epimers have been heavily detected in agricultural soil. However, existing studies have focused on the residual and environmental behavior of maternal TCs, and few studies have looked at the ecotoxicity of their 4-epimers in soil. In this study, the degradation and interconversion of tetracycline (TC), oxytetracycline (OTC) and their 4-epimers (4-epitetracycline, ETC; 4-epioxytetracycline, OTC) were revealed. Their effects on antibiotic resistance genes (ARGs), mobile genetic elements (MGEs) and bacterial community in soil were also investigated in comparison. The results showed that the 4-epimers could be substantially transformed to their parents and degraded as a whole. The degradation rates of four selected pollutants are followed: TC > OTC > ETC > EOTC. This indicated that when TCs entered the soil, part of TCs transformed into slower-degraded 4-epimers, and these 4-epimers could also be converted back to their antibiotic parents, causing the long-term residue of TCs in soil. When added to the soil alone, TC and OTC significantly promoted the proliferation of most ARGs and MGEs, among them, trb-C, IS1247 and IS1111 were the top three genes in abundance. ETC and EOTC had little effect at the beginning. However, as the 4-epimers continuously converted into their parents after one month of cultivation, ETC and EOTC treatments showed similar promoting effect on ARGs and MGEs, indicating that the effect of ETC and EOTC on soil resistome was lagged and mainly caused by their transformed parents. Nocardioides, unclassified_Rhizobiaceae, norank_Sericytochromatia, Microlunatus, Solirubrobacter and norank_67-14 were the most frequent hosts of ARGs, Most of which belong to the phylum Actinobacteria. Due to their large transformation to TCs, slow degradation rate and potential effects on soil microbes and ARGs, the harm of TCs' 4-epimers on soil ecosystem cannot be ignored.

Abstract:In this study, the changes in the enzymatic activity and bacterial communities inhabiting the earthworm gut were investigated to determine the correlation between feeding conditions and tetracycline (TC) degradation. Two TC concentrations (high and low) were fed to earthworms using either the sequential swallowing or repeated swallowing technique. The repeated swallowing feeding technique was found to have a better effect on the degradation rate of TC. The peroxidase and catalase activities in earthworms fed using the sequential swallowing technique were higher than those in earthworms fed using the repeated swallowing technique. When the sequential swallowing technique was used, the TC degradation rate was found to be negatively correlated with the abundance of Clostridium, Hyphomicrobium, and Streptomyces, whereas when the repeated swallowing technique was used for feeding the earthworms, the TC degradation rate was positively correlated with the abundance of 18 bacterial species, including Clostridium, Hyphomicrobium, and Fodinicola.
