Abstract:This study innovated constructed an activated carbon-loaded nano-zero-valent iron (nZVI-C) enhanced membrane aerated biofilm reactor (MABR) coupled partial nitritation/anammox (PN/A) system for optimizing nitrogen and antibiotics removal. Results showed that nitrogen and antibiotic removal efficiencies of 88.45 ± 0.14% and 89.90 ± 3.07% were obtained by nZVI-C, respectively. nZVI-C hastened Nitrosomonas enrichment (relative abundance raised from 2.85% to 12.28%) by increasing tryptophan content in EPS. Furthermore, nZVI-C proliferated amo gene by 3.92 times and directly generated electrons, stimulating Ammonia monooxygenase (AMO) co-metabolism activity. Concurrently, via antibiotic resistance genes (ARGs) horizontal transfer, Nitrosomonas synergized with Arenimonas and Comamonadaceae for efficient antibiotic removal. Moreover, nZVI-C mitigated antibiotics inhibition of electron transfer by proliferating genes for PN and anammox electron production (hao, hdh) and utilization (amo, hzs, nir). That facilitated electron transfer and synergistic substrate conversion between ammonia oxidizing bacteria (AOB) and anaerobic ammonia oxidizing bacteria (AnAOB). Finally, the high nitrogen removal efficiency of the MABR-PN/A system was achieved.

Abstract:A laboratory-scale membrane-aerated biofilm bioreactor (MABR) equipped with non-woven fabrics support around the gas-permeable carbon tube was developed for single-stage autotrophic nitrogen removal based on partial nitrification and anaerobic ammonium oxidization. This reactor allowed air to be supplied through the microporous carbon tube wall to the biofilm that was supported by non-woven fabrics. The partial nitrification and consumption of dissolved oxygen occurred in the inner layer and Anammox in the anoxic outer layer of the non-woven fabrics, thus realizing autotrophic nitrogen removal in a single reactor. After 116d of operation, the maximal nitrogen removal of 0.77kgNm(-3)d(-1) at a volumetric ammonium loading rate of 0.87kgNm(-3)d(-1) was achieved. The spatial profiles of the ammonia-oxidizing bacteria and Anammox bacteria were evaluated by fluorescence in situ hybridization. This study demonstrated that MABR was a very suitable experimental set-up for the operation of the single-stage autotrophic nitrogen removal process.

Abstract:Inhibition of nitrite oxidizing bacteria (NOB) is important for efficient wastewater treatment via anammox. Here, a novel strategy was demonstrated, which based on the detachment and reattachment of the biofilm to establish partial nitrification and anammox in a membrane aerated biofilm reactor (MABR). The results showed that using free ammonia as the sole inhibitor to start up partial nitrification took more time, and NOB acclimated to the free ammonia inhibition. Detaching the biofilm made it possible for free ammonia to contact NOB in deeper zones of the biofilm while at the same time washing NOB out of the reactor. Nitrite accumulation efficiency (NAE) increased to 70 % in 50 days when both high ammonia loading and biofilm reattachment were applied. With a short hydraulic retention time and high concentrations of ammonia and nitrite, anammox activity appeared in the reactor and resulted in a nitrogen removal efficiency of 60 %. Microbial community analysis indicated that Nitrosomonas , Nitrospira , and Candidatus Brocadia were the main functional taxa. Anaerobic ammonia oxidizing bacteria (AnAOB) proliferated within the biofilm, inhibiting NOB growth by competing for nitrite. Endogenous denitrifying bacteria also grew, coexisting with AnAOB. Predicted abundance of genes related to fermentation correlated with those involved in denitrification, indicating the possibility of heterotrophic denitrification. These findings help further our understanding of biofilm control-based strategies for NOB management and have implications for establishing autotrophic nitrogen removal using MABRs.

Abstract:This study is the first to report the start-up and the nitrogen removal performance of a single-stage PN/A/PD MABR without a heating system. It was found that the lumen pressure difference significantly affected the total nitrogen removal efficiency (TNRE), and the dissolved oxygen (DO) changed markedly with the lumen pressure difference. When the lumen pressure difference decreased, the DO tended to decrease, and (partial) denitrification can be enhanced, thus, the TNRE was further improved. And the highest removal efficiency of NH 4 + -N and TN can reach 100 % and 88.03 %, even at 8.7 °C-19.1 °C. At the low temperatures and C/N ratio 3-4, it was found that the relative abundance of Candidatus_Brocadia increased from 3.99 % to 8.64 % from day 45 to day 99 in the PN/A/PD MABR, showing Candidatus_Brocadias low-temperature adaptability, based on 16S rRNA and metagenomics technology. Carbohydrate metabolism and nitrogen metabolism were also analyzed according to the Kyoto Encyclopedia of Genes and Genomes (KEGG) metabolism pathway. The study achieved NO 2 − -N accumulation through partial nitrification and partial denitrification processes in a single-stage Anammox MABR with low-intensity substrates and had engineering application guiding value for mainstream Anammox MABR.

Abstract:Membrane aerated biofilm reactor (MABR) has attracted a lot of attention as an energy-efficient integrated nitrogen removing technology in recent years. However, it is lacking of understanding to realize stable partial nitrification in MABR because of its unique oxygen transfer mode and biofilm structure. In this study, free ammonia (FA) and free nitrous acid (FNA) based control strategies for partial nitrification with low NH4+-N concentration were proposed in a MABR of sequencing batch mode. The MABR was operated for over 500 days under different influent NH4+-N concentrations. With the influent NH4+-N of around 200 mg-N/L, partial nitrification could be established with relatively low concentration of FA (0.4-2.2 mg-N/L) which suppressed nitrite oxidizing bacteria (NOB) on the biofilm. With lower influent NH4+-N concentration of around 100 mg-N/L, the FA concentration was lower and strengthened suppression strategies based on FNA were needed. With the final pH of operating cycles below 5.0, the FNA produced in the sequencing batch MABR could stabilize partial nitrification by eliminating NOB on the biofilm. Since the activity of ammonia oxidizing bacteria (AOB) was lower without the blow-off of dissolved carbon dioxide in the bubbleless MABR, longer hydraulic retention time was required to reach a low pH for high concentration of FNA to suppress NOB. After exposures to FNA, the relative abundance of Nitrospira was decreased by 94.6%, while the abundance of Nitrosospira increased greatly which became another dominant AOB genus in addition to Nitrosomonas.

Abstract:Shortcut nitrogen removal holds significant economic appeal for mainstream wastewater treatment. Nevertheless, it is too difficult to achieve the stable suppression of nitrite-oxidizing bacteria (NOB), and simultaneously maintain the activity of ammonia-oxidizing bacteria (AOB). This study proposes to overcome this challenge by employing the novel acid-tolerant AOB, namely " Candidatus Nitrosoglobus", in a membrane-aerated biofilm reactor (MABR) Superior partial nitritation was demonstrated in low-strength wastewater from two aspects. First, the long-term operation (256 days) under the acidic pH range of 5.0 to 5.2 showed the successful NOB washout by the in situ free nitrous acid (FNA) of approximately 1 mg N/L. This was evidenced by the stable nitrite accumulation ratio (NAR) close to 100% and the disappearance of NOB shown by 16S rRNA gene amplicon sequencing and fluorescence in situ hybridization. Second, oxygen was sufficiently supplied in the MABR, leading to an unprecedentedly high ammonia oxidation rate (AOR) at 2.4 ± 0.1 kg N/(m 3 d) at a short hydraulic retention time (HRT) of a mere 30 minutes. Due to the counter diffusion of substrates, the present acidic MABR displayed a significantly higher apparent oxygen affinity (0.36 ± 0.03 mg O 2 /L), a marginally lower apparent ammonia affinity (14.9 ± 1.9 mg N/L), and a heightened sensitivity to FNA and pH variations, compared with counterparts determined by flocculant acid-tolerant AOB. Beyond supporting the potential application of shortcut nitrogen removal in mainstream wastewater, this study also offers the attractive prospect of intensifying wastewater treatment by markedly reducing the HRT of the aerobic unit.

Integration of Zeolite Membrane Bioreactor With Granular Sludge-Based Anammox in High-Efficiency Nitrogen Removal From Iron Oxide Red Wastewater

Abstract:Acquisition of stable nitritation and efficient anammox play a crucial role in partial nitritation (PN) combined with anammox for nitrogen removal from ammonium-rich wastewater. Due to the limitation of ammonia-oxidizing bacteria (AOB) enrichment and nitrite-oxidizing bacteria (NOB) control in traditional membrane biological reactor (MBR), it can result in a lower nitrite production rate (NPR) and unstable PN, eventually reducing the nitrogen removal rate (NRR) via PN-anammox. In this study, we developed a zeolite membrane biological reactor (ZMBR) to enhance the PN of iron oxide red wastewater (IORW), in which the biofilm derived from the zeolite surface can provide free ammonia (FA)-containing microenvironment for AOB enrichment and NOB inhibition. The results showed that ZMBR can tolerate a higher influent nitrogen loading rate (NLR) of 2.78 kg/(m3⋅day) in comparison to the traditional MBR [2.02 kg/(m3⋅day)] and the NPR in ZMBR and traditional MBR were 1.39 and 0.96 kg/(m3⋅day), respectively. The mass concentration ratio of NO 2 - -N/ NH 4 + -N ranged from 1.05 to 1.33 in ZMBR, suggesting a suitable condition for nitrogen removal via anammox. Subsequently, the domesticated granular sludge obtained from a paper-making wastewater treatment was used as the carrier of anammox bacteria to remove nitrogen. After 93 days of operation, the NRR was observed to be 2.33 kg/(m3⋅day) and high-throughput sequencing indicated that the relatively higher abundance (45.0%) of Candidatus Kuenenia stuttgartiensis was detected in the granular sludge of the bottom part of the reactor, which can produce more proteins and lipids, suggesting a good settleability. Overall, this study provides a high-efficient method to control PN and domesticate anammox for nitrogen removal from IORW.
SNAD for nitrogen removal using carbon tube membrane-aerated biofilm reactor

Abstract:A lab-scale membrane-aerated biofilm bioreactor(MABR) equipped with nonwoven fabrics support around the gas-permeable carbon tube was developed for the removal of ammonium based on simultaneous partial nitrification,anaerobic ammonium oxidization and denitrification(SNAD).The temperature,pH and HRT were controlled at 34±1 ℃,7.5~8.5 and 8 h.By decreasing the air pressure in the carbon tube gradually,DO in the reactor was decreased from 8 mg/L to 0.5 mg/L.Initially,the reactor was inoculated with nitrifying biomass.Subsequently,anammox biomass was inoculated to start up SNAD system in one reactor.The results showed that after 180 d of operation,93.4 % of NH+4-N removal rate,92.5 % of TN removal rate and 97.2 % of COD removal were achieved in MABR.The ammonium nitrogen removal rate was 0.6 kg N/(m3 ·d).The suitable C/N ratio is 0.2~0.6,and the higher COD will inhibit the Anammox bacteria and make the reactor performance worsen.

Abstract:An AnMBR-PN/A system was developed for mainstream sewage treatment. To verify the efficient methanation and subsequent chemolitrophic nitrogen removal, a long-term experiment and analysis of microbial activity were carried out. AnMBR performance was less affected by the change of hydraulic retention time (HRT), which could provide a stable influent for subsequent PN/A units. The COD removal efficiency of AnMBR was > 93% during the experiment, 85.5% of COD could be recovered in form of CH 4 . With the HRT of PN/A being shortened from 10 to 6 h, nitrogen removal efficiency (NRE) of PN/A increased from 60.5% to 80.4%, but decreased to 68.8% when the HRT PN/A further decreased to 4 h. Microbial analysis revealed that the highest specific ammonia oxidation activity (SAOA) and the ratio of SAOA to specific nitrate oxidation activity (SNOA) provide stable NO 2 − -N/NH 4 + -N for anammox, and anammox bacteria (mainly identified as Candidatus Brocadia ) enriched at the bottom of Anammox-UASB might play an important role in nitrogen removal. In addition, the decrease of COD in Anammox-UASB indicated partial denitrification occurred, which jointly promoted nitrogen removal with anammox.

Abstract:The anammox process is considered as a revolutionary new denitrification technology. In this study, the anammox process was started in a single-stage moving bed biofilm reactor (MBBR) and the mechanism of excess removal of ammonia nitrogen was studied. At stage I (day 0–51), anammox bacteria (AnAOB) was enriched by feeding synthetic sewage without adding organic carbon. The removal rate of ammonia nitrogen was maintained at about 54% and the removal rate of total inorganic nitrogen was maintained at about 62%. At stage II (day 52–91), internal circulation was added into the MBBR. After adding internal circulation, the ammonium removal efficiency reached about 96% (at day 56) and the total nitrogen removal efficiency reached about 86%. At day 90, the biofilm sample was drowned out for high-throughput sequencing. The results showed that the relative abundance of AnAOB was 23.23%. The dominant anammox genus was Candidatus Brocadia. The relative abundance of Nitrosomonas (ammonia oxidizing bacteria, AOB) was 0.63%. The excess ammonia nitrogen was removed by AOB and AnAOB through the partial nitrification and anammox (PNA) process.

Abstract:The partial nitritation-anammox process based on a membrane-aerated biofilm reactor (MABR) faces several challenges, such as difficulty in suppressing nitrite-oxidizing bacteria (NOB), excessive effluent nitrate, and ineffective synergy between denitrification and anammox bacteria. Therefore, a novel partitioned granular sludge coupling with MABR (G-MABR) was constructed. The chemical oxygen demand (COD) and nitrogen removal efficiency were 88.8 ± 1.8 %–92.6 ± 1.2 % and 88.8 ± 1.5 %–93.6 ± 0.7 %, respectively. The COD was mainly lowered in the lower granular sludge-zone, while nitrogen was removed in the upper MABR-zone. NOB was significantly suppressed in the MABR-zone due to competition for substrate with denitrifying bacteria and anammox bacteria. This partitioned configuration reduced the C/N ratio in the MABR-zone, thus facilitating autotrophic nitrogen removal. Both partial nitrification and denitrification provided nitrite for anammox bacteria in granular sludge, whereas partial nitrification mainly supplied nitrite to the anammox bacteria in membrane biofilms.

Abstract:In this study, an anaerobic membrane bioreactor coupled with a complete nitrification and partial denitrification–anammox process (AnMBR–NF–PDA) was developed to efficiently remove both chemical oxygen demand (COD) and nitrogen. Precise control of raw water ratios was utilized to adjust the ratio of COD/NO 3 − –N, resulting in maximum nitrogen removal efficiency of 90.14% at a ratio of 3.44. Initially, specific anammox activity (SAA) increased with the proportion of raw water, peaking at 17.83 mg-N/(g-VSS·d) in stage II before decreasing. This variation was attributed to the significant presence of filamentous bacteria, especially " Acinetobacter " (13.58%–31.59%), which facilitated nitrite generation, supporting the nitrous oxide hypothesis in partial denitrification processes and enabling cross-feeding with AnAOB. Additionally, the average particle size of granular sludge increased from 300 to 528 μm under the influence of filamentous bacteria. Metagenomic analysis revealed an upsurge in genes associated with partial denitrification (NarG and NapA) as the COD/NO 3 –N ratio rose. The abundance of genes closely correlated with anammox (Hzs and Hdh) peaked during stage II, indicating the beneficial role of filamentous bacteria in the stable conversion of nitrite in PDA system. This study offers valuable insights into optimizing the synergistic metabolism and granulation processes involving filamentous bacteria and AnAOB, thereby laying the groundwork for the practical application of AnMBR coupled with anammox processes in wastewater treatment.

Abstract:In order to reduce the resource and energy consumption of traditional biological nitrogen removal (BNR) process, heterotrophic nitrification-aerobic denitrification (HN-AD) bacteria was in situ enriched in membrane aerated biofilm reactor (MABR) by inoculating conventional activated sludge. Contaminants removal performance, EPS composition and microbial community were explored. The results indicated that the average removal efficiency of COD and TN under optimal condition reached 84.13% and 91.54%, respectively, which demonstrated that the reactor possessed excellent contaminants removal capacity. EPS analysis suggested that abundant protein especially tryptophan protein-like substance played a vital role in maintaining the microbial stability of biofilms. Multiple HN-AD genera, mainly Paracoccus, were detected with the highest relative abundance of 54.70%, which confirmed the successful enrichment of the HN-AD bacteria. Conventional nitrifiers and denitrifiers also thrived in biofilm, which demonstrated the synergistic nitrogen removal of multiple microorganisms. This study provided important insights into application of HN-AD bacteria and synergistic nitrogen removal in BNR system.

Abstract:The partial nitrification (PN)–Anammox process was considered as one of the most efficient and economical method to remove ammonia from wastewater without organic carbon consumption. In this process, PN is the key step since it should provide stable effluent with suitable ratio for the subsequent Anammox. In this study, PN was successfully started-up during 30 d at ambient temperature (25°C) in a continuous flow membrane bioreactor (MBR), by gradually increasing ammonia-loading rate (ALR) under oxygen-limited condition. Finally, the system achieved an ammonia oxidation rate of 0.52 kg m−3 d−1 and an excellent effluent with appropriate ratio as 1.1–1.3. ALR was indicated as the indispensable controlling factor, and the applied alkalinity with the combination of high ALR inhibition and oxygen limitation provided the selective washout of nitrite-oxidizing bacteria (NOB) and an active aerobic ammonia-oxidizing (AerAOB) population. Techniques of fluorescence in situ hybridization and clone sequencing were utilized simultaneously to study the microbial characteristics in the stable PN system. The results demonstrated that small amounts of NOB were present and not active, while AerAOB predominated in the MBR–PN system, and Nitrosomonas-related AerAOB were considered to be responsible for the satisfactory PN performance.

After 120 d of operation, 88.7% of NH4(+) -N removal rate, 48.65 mg/L of average TN in the effluent, 83.77% of maximal TN removal efficiency was achieved under the following conditions: air pressure of 0.015 MPa, HRT of 6 h, and influent NH4(+) -N of 200 mg/L +/- 10 mg/L.

The results indicated that the optimal DO, pH and C/N ratio were 1-2 mg/L, 9.0 and 4-7, respectively. The corresponding average organic removal rate (ORR), total nitrogen removal rate (TNRR) and nitrite accumulation rate (NAR) reached 324 gCOD・m-3・d-1, 48 gN・m-3・d-1 and 77.70%, respectively. Extracellular polymeric substance (EPS) content in biofilm was more abundant than that in inoculated sludge. Multiple aerobic denitrifiers were detected in the biofilm with the relative abundance of 11.19%-22.71%. AQUASIM simulation implied that the distribution and proportion of substrates and bacteria were significantly affected by DO and pH regulation. Overall, this study provided some important insights in the start-up and operation of synergistic nitrogen removal process in BNR system.

Abstract:Anaerobic technologies with downstream autotrophic nitrogen removal have been proposed to enhance bioenergy recovery and transform a wastewater treatment plant from an energy consumer to an energy exporter. However, approximately 20-50 % of the produced methane is dissolved in the anaerobically treated effluent and is easily stripped into the atmosphere in the downstream aerobic process, contributing to the release of greenhouse gas emissions. This study aims to develop a solution to beneficially utilize dissolved methane to support high-level nitrogen removal from anaerobically treated mainstream wastewater. A novel technology, integrating Partial Nitritation, Anammox and Methane-dependent nitrite/nitrate reduction (i.e. PNAM) was demonstrated in a membrane-aerated biofilm reactor (MABR). With the feeding of ∼50 mg NH4+-N/L and ∼20 mg/L dissolved methane at a hydraulic retention time of 15 h, around 90 % of nitrogen and ∼100 % of dissolved methane can be removed together in the MABR. Microbial community characterization revealed that ammonia-oxidizing bacteria (AOB), nitrite-oxidizing bacteria (NOB), anammox bacteria, nitrite/nitrate-dependent anaerobic methane oxidation microorganisms (n-DAMO bacteria and archaea) and aerobic methanotrophs co-existed in the established biofilm. Batch tests confirmed the active microbial pathways and showed that AOB, anammox bacteria and n-DAMO microbes were jointly responsible for the nitrogen removal, and dissolved methane was mainly removed by the n-DAMO process, with aerobic methane oxidation making a minor contribution. In addition, the established system was robust against dynamic changes in influent composition. The study provides a promising technology for the simultaneous removal of dissolved methane and nitrogen from domestic wastewater, which can support the transformation of wastewater treatment from an energy- and carbon-intensive process, to one that is energy- and carbon-neutral.

Abstract:Serious sludge washout is a bottleneck in the partial nitritation-anammox (PN/A) process that needs to be addressed urgently. A membrane bioreactor (MBR) is suitable for the PN/A process because of its 100 % biomass retention ability. Therefore, this study establishes a single-stage PN/A MBR and a PN/A moving bed membrane reactor (PN/A MB-MBR) for investigating the operation performance, membrane fouling behavior, and effect of biofilm carrier addition. The results demonstrate that the PN/A MB-MBR achieves a significantly shorter start-up time and substantially higher maximum nitrogen removal rate (NRR) of 17 days and 1.02 g N/L/d, respectively, compared to that of PN/A MBR (61 days and 0.19 g N/L/d, respectively). Further, the PN/A MB-MBR exhibits a lower membrane fouling of 0.023 kPa/L compared to that of 0.050 kPa/L in PN/A MBR because of the integration of the biofilm carrier. The biofilm in PN/A MB-MBR shows a higher relative abundance of Candidatus Kuenenia (18.90–25.10 %), which is a major AnAOB, compared to that observed in the suspended sludge of PN/A MB-MBR (10.30–14.32 %). Furthermore, the relative abundance of Candidatus Kuenenia absorbed on the membrane surface of PN/A MB-MBR (8.6 %) was lower than that on PN/A MBR (15.5 %). This study presented a promising method for enhancing the performance of PN/A MBR, which can serve as a valuable reference for its extensive implementation.

Abstract:Reactor configuration, control strategy and inoculation method were key factors affecting the rapid start-up of partial nitrification/anammox (PN/A) process and the efficient enrichment of functional bacteria (anammox and ammonia oxidizing bacteria). At present, PN/A process was generally operated through single factor rather than forming a system. In this study, a novel aerobic-biofilm/anaerobic-granular nitrogen removal system (OANRS) was constructed, which consisted of a multi-stage aerobic-biofilm/anaerobic-granular baffle reactor (MOABR) and a control strategy on pH/aeration time. PN process was started within 10d, and PN/A process was started on the basis of stable PN process within 41d. The simultaneous enrichment of functional bacteria was achieved by combining the advantages of single-stage and two-stage PN/A process. The results of high-throughput sequencing showed that Candidatus Kuenenia (20.42±15.88%) was highly enriched in each compartment at day 98, and the relative abundance of Candidatus Kuenenia in the anaerobic compartment R4 was as high as 43.13%.
