Abstract:As important biomolecules, the deficiency or maladjustment of hydrogen sulfide (H2S) or ascorbic acid (AA) is associated with the symptoms of the same disease (e.g., cardiovascular disease or cancer). There is an urgent need to develop a fluorescent probe capable of distinguishing between H2S and AA simultaneously. Here, we report the syntheses, structure, and property of the first dual-detection fluorescent probe which can differentiate H2S or/and AA in aqueous media. Accordingly, a novel [EuCu(pydc)2(ox)0.5(H2O)3·1.5H2O]2 n (1, H2pydc = 2,3-pyridinedicarboxylic acid and ox = oxalic acid) for selective and sensitive detection of H2S and AA in a wide range has been constructed (H2S: [130 nM, +∞); AA: [55 nM, +∞)), exhibiting excellent catalytic activity comparable to horseradish peroxidase. In addition, the highly efficient detection in human serum sample also proves the potential application in medical diagnosis. Meanwhile, a combinatorial logic gate (AND(INH)-OR) based on activated 1 has also been constructed. Furthermore, this approach for simultaneous H2S and AA detection suggests that the current work will expand the potential application of metal-organic frameworks for dual or multiple detections in biomedical fields.

Abstract:Hydrogen sulfide (H 2 S) is an important sulfur-containing signaling molecule in the human body, as well as a highly toxic gas that is soluble in water. A variety of pathological processes are associated with abnormal metabolism of hydrogen sulfide, so it is very important to develop an assay that can rapidly and specifically detect H 2 S. In this work, two iridium(III) complexes were designed and synthesized using ester groups as reaction sites for recognition of H 2 S. As the H 2 S concentration increased, the photoluminescence of the probe was rapidly quenched within 5 seconds, with a detection limit as low as 0.45 μM. Among them, the probe Ir1 , whose luminescence was located in the visible region, had been successfully used for the detection of trace amounts of H 2 S in water samples with a recovery rate of 92.5 ~ 104.0% and a relative standard deviation (RSD, n=5) of < 2.8%. The luminescence of probe Ir2 was located in the near-infrared region, which could avoid the interference of biological autofluorescence and be successfully used for luminescence imaging of H 2 S in HeLa cells. This study provided a promising method for rapid detection of trace H 2 S in water samples and cellular microenvironments.

Abstract:As a crucial biological gasotransmitter, hydrogen sulfide (H2S) plays important roles in many pathological and physiological processes. Highly selective and sensitive detection of H2S is significant for the precise diagnosis and evaluation of diverse diseases. Nevertheless, challenges remain in view of the interference of autofluorescence in organisms and the stronger reactivity of H2S itself. Herein, we report the design and synthesis of a novel H2S-responsive β-diketonate-europium(III) complex-based probe, [Eu(DNB-Npketo)3(terpy)], for background-free time-gated luminescence (TGL) detection and imaging of H2S in autofluorescence-rich biological samples. The probe, consisting of a 2,4-dinitrobenzenesulfonyl (DNB) group coupled to a β-diketonate-europium(III) complex, shows almost no luminescence owing to the existence of intramolecular photoinduced electron transfer. The cleavage of the DNB group by a H2S-triggered reaction results in the recovery of the long-lived luminescence of the Eu3+ complex, allowing the detection of H2S in complicated biological samples to be performed in TGL mode. The probe showed a fast response, high specificity, and high sensitivity toward H2S, which enabled it to be successfully used for the quantitative TGL detection of H2S in tissue homogenates of mouse organs. Additionally, the low cytotoxicity of the probe allowed it to be further used for the TGL imaging of H2S in living cells and mice under different stimuli. All of the results suggested the potential of the probe for the investigation and diagnosis of H2S-related diseases.

Abstract:Hydrogen sulfide (H 2 S) is traditionally considered as an ecologically toxic gas, has been proven to be the third endogenous signaling gasotransmitter and plays important roles in physiological and pathophysiological conditions. Herein, we designed and synthesized a novel "turn off" fluorescent probe for the detection of hydrogen sulfide(H 2 S) in vitro. Probe A could detect H 2 S in CH 3 CN/PBS system with significant fluorescence quenching. Probe A also showed obviously aggregation- induced emission (AIE) effects. Fluorescence titration experiments revealed probe A combine with H 2 S in a 1: 1 ratio with a detection limit of 6.54 μM. Cell imaging results demonstrated the potential applicability of probe A for H 2 S detection in living cells. The supramolecular assembly of probe A and β-cyclodextrin further revealed the interaction with H 2 S.

Abstract:Hydrogen sulfide (H 2 S) is a common toxic gas that threatens the quality and safety of environmental water and food. Herein, a new near-infrared fluorescent probe DTCM was synthesized and characterized by single crystal X-ray diffraction for sensing H 2 S. It exhibited a remarkable "turn-on" near-infrared (NIR) emission response at 665 nm with a remarkably massive Stokes shift of 175 nm, super-rapid detection ability (within 30 s), excellent photostability, high selectivity and sensitivity (limit of detection, LOD = 58 nM). Additionally, the probe was successfully utilized for the detection of H 2 S in environmental water samples. The DTCM -loaded test papers enabled convenient and real-time monitoring of H 2 S produced by food spoilage.

Abstract:In this work, a novel Eu3+-DTPA-bis(AMC) complex with red luminescence was designed and synthesized for sequential detection of Cu2+ and biothiols (Cys/Hcy/GSH) based on the displacement strategy with the good selectivity, high sensitivity, and large Stokes shift (288 nm). The possible detection mechanism was verified by UV-vis, the high-resolution mass spectrometry, and the fluorescence decay curve. The experimental parameters, including the solution pH, the incubation time, the concentration ratio of Eu3+-DTPA-bis(AMC) to Cu2+ and biothiols concentration, were optimized. Under the optimal conditions, it shows a good linear relationship between the concentration (0-10 μM) of Cu2+ and the fluorescence intensity of Eu3+-DTPA-bis(AMC), with a low detection limit of 0.065 μM. The linear range and the limit of detection of the Eu3+-DTPA-bis(AMC)/Cu2+ system for Cys/Hcy/GSH were 2.5-22.5/5-45/5-50 μM and 0.11/0.07/0.05 μM, respectively. Surprisingly, the high or low concentration of Eu3+-DTPA-bis(AMC)/Cu2+ can significantly affect the selectivity of the sensing system to biothiols (Cys/GSH/Hcy). When the concentration of the Eu3+-DTPA-bis(AMC)/Cu2+ system is 10.0 μΜ, it could recognize biothiols (Cys/GSH/Hcy) from other substances, but when the concentration is as low as 3.3 μM, it could further specifically distinguished Cys from Hcy/GSH. Owing to the high anti-interference characteristics, accuracy and specificity, the sensing system was well applied to the cascade detection of Cu2+ in actual environmental samples and Cys in biological and food samples, including FBS, urine, milk, beverage, fresh juice with the satisfactory recoveries from 96.20 to 106.80 %.

Abstract:As the third gasotransmitter after NO and CO, hydrogen sulfide plays essential biological roles in living systems, and developing sensitive fluorescent probe for the rapid tracking of endogenous H2S is attracting much attention. A new H2S fluorescent probe was constructed in this study based on a copper complex formed by a new NBD fluorescent ligand, in which a Cu2+ chelator was incorporated. This probe displays very weak emission owing to the emission quenching effect of Cu2+, and it is able to sense H2S by exhibiting distinct emission enhancement upon H2S addition. The S2--induced Cu2+ removal from this complex was proposed as the sensing mechanism of this Cu2+ complex-based probe. Besides the quick response, the high sensing selectivity over other anion of this probe has also been confirmed, and its detection limit was estimated to be similar to 0.2 mu mol.L-1 and the linear range is 1 similar to 20 mu mol.L-1.

Abstract:As a signal transduction molecule in organism, H2S plays an important role in many physiological processes. In this paper, a novel H2S fluorescent probe SBD was designed based on 1,8-naphthalimide and dinitrobenzenesulfonyl group. In DMSO-PBS solution, probe SBD could recognize H2S and change the color of solution from yellow to pink, realizing the naked eye recognition for H2S. In addition, probe SBD displayed high fluorescence enhancement rate (45-fold), good selectivity, strong anti-interference ability and the detection limit was as low as 152 nM. Most importantly, probe SBD showed very low cytotoxicity and had been successfully applied to image exogenous H2S in living cells.

Abstract:H2S has been reported to play essential roles in a variety of physiological and pathological procedures. In this work, a novel fluorescent probe, Rho-HS, for detecting H2S was developed by introducing the ortho-halogen to activate the least reactive recognition group 2,4-dinitrophenyl moiety. In combination of the structures from both Rhodamine B and fluorescein, Rho-HS could generate both the colorimetric and fluorescent responses. This feature was not frequently achieved and could lead to the quantitative and convenient for the end-user. In comparison with recent probes for H2S, the major advantages of Rho-HS included suiting wide pH range (6.0-10.0), relatively rapid response (within 15 min) and the high selectivity among the competing species including the biothiols. With low cytoxicity, Rho-HS was further applied in the biological imaging in living MCF7 cells and Caenorhabditis elegans. We hope that the designing strategy in this work might provide useful information for more preferable implements in this field.

Abstract:Since Hydrogen Sulfide (H2S) was recognized as a gas transmitter, its detection and quantification have become a hot research topic among chemists and biologists. In this area, fluorescent probes have shown great advantages: fast and strong response, low detection limit and easy manipulation. Here we developed a new fluorescent probe that detected H2S selectively among various bioactive and inorganic salts. This probe was based on the core structure of fluorescein and reacted with H2S through azide-reduction. Great linearity was achieved correlating fluorescence intensity and H2S concentrations in solution. The detection of H2S in cancer cells was also achieved.

Abstract:Copper ions (Cu2+) and hydrogen sulfide (H2S) detection is of great significance in environmental monitoring and disease screening. Herein, a novel fluorescent probe (DAHK) was rationally designed and easily synthesized based on a copper peptide backbone labelled a dansyl fluorophore. DAHK exhibited large Stokes shift (210 nm), high sensitivity (38.9 nM) and high selectivity response to Cu2+ ions based on obvious fluorescence quenching in HEPES buffer (10.0 mM, pH 7.4) solutions. In addition, the in situ formed DAHK-Cu2+ ensemble was successfully used as a secondary probe for rapidly and sequentially detected H2S via fluorescence “off–on” response, and without interference by a range of other anions. The limit of detection (LOD) for 47.3 nM was determined, which was a much lower value than WHO and EPA guidelines (H2S) for drinking water. The reversibility of interaction of DAHK with Cu2+ and H2S was monitored using fluorescence emission spectra. Cells imaging experiments exhibited that DAHK had good membrane permeability and excellent low toxicity. Moreover, DAHK was successfully made into fluorescent test strips for visual detection and rapid analysis based on the exciting stability and excellent water solubility.

Abstract:8554 Background: Patients with peripheral T/NK cell lymphomas (PTCL) still have a dismal prognosis with 5-yr survival less than 30% in most cases. No alternative regimen has been proven superior to CHOP so far. This multicenter phase II study was carried out to assess efficacy and safety of bortezomib in combination with an intensified CHOP-like regimen. METHODS Pts aged 18 to 65 yrs with previously untreated PTCL were planned to receive 4 bi-monthly cycles of ACVBP (doxorubicine 75 mg/m2 D1, cyclophosphamide 1200 mg/m2 D1, vindesine 2 mg/m2 D1 and D5, bleomycine 10 mg D1 and D5 and prednisone D1 to D5) followed by a sequential consolidation consisting of HD methotrexate (2 courses), etoposide + ifosfamide (4 courses) and cytarabine (2 courses) at 2 weeks intervals. Bortezomib 1.5 mg/m2 was administered at D1 and D5 of each ACVBP cycle, and then at D1, D8 and D15 every 4 weeks during consolidation phase for a total of 20 injections during the whole treatment. RESULTS 57 eligible pts (M 38, F 19, median age 52.5 yrs) with mostly AITL and PTCL NOS subtypes were enrolled between January 2006 and November 2007; 78% had stage III-IV disease and 53% had aaIPI ≥ 2. Forty six pts (81%) have completed induction treatment with ACVBP and only 28 (49%) the consolidation phase, mainly for disease progression. The CR + CRu rate was 45% after induction and 46% after consolidation. As of November 14th, 2008, 22 pts (39%) have died, mostly from lymphoma. The median percentage of planned dose of bortezomib received was 98% during ACVBP induction where the vinca alkaloid used was vindesine, and ranged from 90 to 95% during the consolidation courses. The dose intensity of bortezomib was 84.3% during induction, similar to that of doxorubicine and cyclophosphamide. Thrombocytopenia was more pronounced than previously observed with ACVBP alone but no life-threatening hemorrhagic event occurred. CONCLUSIONS The combination of bortezomib with ACVBP is feasible without neurological or platelet unexpected toxicities. The response rate of such a regimen in PTCL does not appear higher than previously observed with ACVBP alone in our historical cohort. No significant financial relationships to disclose.

A novel structurally modified isophorone fluorescent probe for H2S detection

Abstract:Hydrogen sulfide (H2S) has a comprehensive contribution to the normal operation and stability of organisms and is also present in environmental water samples and food deterioration. Thus, it is exceedingly promising and significant to develop a highly sensitive detection technique for tracing H2S. Inspired by this, we designed and synthesized a new fluorescent probe 2-[3-[2-[3-bromo-4-(2,4- dinitrobenzenesulfonate)] ethenyl]-5,5-dimethyl-2-cyclohexen-1-ylidene]propanedinitrile (SP-Br) for hydrosulfide ion detection by introducing bromine atom. Compared with reported H2S probes based on the same fluorescent parent, SP-Br has longer fluorescence emission (λem = 670 nm), shorter response time (3 min), lower detection limit (149 nM), and wider detection range (0-30 nM). SP-Br can emit weak yellow fluorescence, and the emission intensity at 670 nm is considerably enhanced in the presence of hydrosulfide ions. The identification mechanism of hydrosulfide ion by SP-Br was verified by high-resolution mass spectrometry, fluorescence, and UV-vis absorption spectroscopy. In addition, SP-Br has been successfully applied to the monitoring of actual water samples and beer samples and has certain development prospects and value in the fields of environmental pollution and food quality analysis.

Abstract:A novel selective fluorescent chemosensor 1 has been synthesized with a phenanthrene-fused dipyrromethene structure. Selective binding of Cu2+ by 1 results in a complex that displays high selectivity and sensitivity for H2S. The signal transduction occurs via reversible formation-separation of the Cu-1 complex and CuS. The probes reviewed in this paper were categorized according to their chemical mechanism of sensing and were divided into three groups: H2S reducibility-based probes, H2S nucleophilicity-based probes, and metal sulfide precipitation-based probes. The structure of the probes, their sensing mechanism, and imaging results have been discussed in detail. Moreover, we also introduced some probes for hydrogen polysulfides.
