Abstract:In this paper, theaflavins extracts from black tea and tea polyphenols were used as control, composition of theaflavins in enzymatic oxidation products was analyzed by HPLC, and the antioxidant activity was investigated by the technique of chemiluminescence. Results showed that, TF2A was the main composition in enzymatic oxidation products. In theaflavins extracts from black tea, there mainly existed four kinds of theaflavins and their contents were not greatly fluctuated. By employing CuSO4-Vc-H2O2-yeast and Fe2+-H2O2-yeast systems, antioxidant activity of enzymatic oxidation products was better than theaflavins extracts from black tea and tea polyphenols.

Abstract:Tyrosinase catalyzes the oxidation of phenols and catechols (o-diphenols) to o-quinones. The reactivities of o-quinones thus generated are responsible for oxidative browning of plant products, sclerotization of insect cuticle, defense reaction in arthropods, tunichrome biochemistry in tunicates, production of mussel glue, and most importantly melanin biosynthesis in all organisms. These reactions also form a set of major reactions that are of nonenzymatic origin in nature. In this review, we summarized the chemical fates of o-quinones. Many of the reactions of o-quinones proceed extremely fast with a half-life of less than a second. As a result, the corresponding quinone production can only be detected through rapid scanning spectrophotometry. Michael-1,6-addition with thiols, intramolecular cyclization reaction with side chain amino groups, and the redox regeneration to original catechol represent some of the fast reactions exhibited by o-quinones, while, nucleophilic addition of carboxyl group, alcoholic group, and water are mostly slow reactions. A variety of catecholamines also exhibit side chain desaturation through tautomeric quinone methide formation. Therefore, quinone methide tautomers also play a pivotal role in the fate of numerous o-quinones. Armed with such wide and dangerous reactivity, o-quinones are capable of modifying the structure of important cellular components especially proteins and DNA and causing severe cytotoxicity and carcinogenic effects. The reactivities of different o-quinones involved in these processes along with special emphasis on mechanism of melanogenesis are discussed.

Oxidation of 4-alkylphenols and catechols by tyrosinase: ortho-substituents alter the mechanism of quinoid formation

Abstract:Numerous phenols and catechols are known to be substrates for tyrosinase. While the catalytic mechanism of phenol oxidation by tyrosinase has been well studied, little work has been done to determine the influence of substituents on the reaction. In the present investigation, we explored the effects of changing substituents at the 2 and 6 position on the mechanism of tyrosinase-catalyzed oxidation of 4-allyl and 4-propylphenols and catechols. We have previously demonstrated that tyrosinase initially oxidizes hydroxychavicol (4-allyl-catechol) to an o-quinone (3,5-cyclohexadien-1,2-dione) which because of the relatively acidic protons in the benzyl position, readily isomerizes to the tautomeric p-quinone methide (4-allylidene-2,5-cyclohexadien-1-one, QM) (Bolton et al., 1994). We have confirmed through GSH trapping studies that oxidation of 4-allylphenol by tyrosinase yields the same o-quinone GSH conjugates as hydroxychavicol. In contrast, the presence of additional ortho substituents dramatically alters the mechanism of tyrosinase-catalyzed oxidation of 4-alkylphenols. For example, eugenol (4-allyl-2-methoxyphenol), which possesses 1 ortho-methoxy substituents, is not oxidized to a o-quinone or a QM. However, when both ortho o-quinones or QMs which may be selectively toxic to the malignant melanocyte. Although mammalian tyrosinase is much more substrate specific compared to the mushroom tyrosinase used in this study [42], it should be possible to identify compounds which are substrates for the mammalian form but are otherwise oxidatively stable. In order to develop such target compounds an improved understanding of substituent effects on tyrosinase-catalyzed oxidation of catechols and phenols is necessary. This should for the development of strategies for therapeutic compounds that are selectively toxic toward melanoma.

Abstract:Hydroxytyrosol (HT) has outstanding antioxidant properties, however, the lack of information regarding its potential interactions with biological systems prevents the direct application of HT in the field of medicine. In particular, the interaction of HT with antioxidant defense systems based on tripeptides containing thiols (glutathione, GSH) remains elusive and requires further investigation. Herein, we investigated the electrochemical behavior of HT in Britton-Robinson buffer solution (B-R) in the absence and presence of GSH using bulk electrolysis (BE) and cyclic voltammetry (CV). The posssible reaction pathways of HT and oxidation products were identified by combining ultrahigh-performance liquid chromatography and mass spectrometry (UHPLC-MS). The findings of the study indicate that the phenolic ring of HT experiences a quasi-reversible oxidation process. This process is associated with the oxidation of the catechol moiety through the transfer of 2e − /2H + in the presence of existing GSH. As a result of this oxidation, a quinone is produced. The quinone molecule can undergo reduction, leading to the regeneration of the HT molecule or interacting with GSH, resulting in the formation of mono-GSH-HT, bi-GSH-HT, and bi-GSH-HT trimer conjugates. Using density functional theory (DFT), the ratio of disulfide, mono-GSH-HT, bi-GSH-HT, and bi-GSH-HT trimer formation energies ( E f ) was determined to be 4.5:1.8:1.1:1.4, which are consistent with UHPLC-MS measurement. Accordingly, the formation of the disulfide form of GSH and parent HT is preferred over the formation of other conjugates. The higher values of electrophilicity index ( ω ) for the HT trimer and quinone suggest a remarkable ability to interact with cells in biological systems.

Abstract:While a conserved tyrosine (Tyr) is found in oxidases, the roles of phenol ring pKa and reduction potential in O2 reduction have not been defined despite many years of research on numerous oxidases and their models. These issues represent major challenges in our understanding of O2 reduction mechanism in bioenergetics. Through genetic incorporation of unnatural amino acid analogs of Tyr, with progressively decreasing pKa of the phenol ring and increasing reduction potential, in the active site of a functional model of oxidase in myoglobin, a linear dependence of both the O2 reduction activity and the fraction of H2O formation with the pKa of the phenol ring has been established. By using these unnatural amino acids as spectroscopic probe, we have provided conclusive evidence for the location of a Tyr radical generated during reaction with H2O2, by the distinctive hyperfine splitting patterns of the halogenated tyrosines and one of its deuterated derivatives incorporated at the 33 position of the protein. These results demonstrate for the first time that enhancing the proton donation ability of the Tyr enhances the oxidase activity, allowing the Tyr analogs to augment enzymatic activity beyond that of natural Tyr.

Abstract:In this research, the inhibition mechanism of a pairs of homoisoflavonoid analogues (6-aldehydo-isoophiopogonanone A (AIO-A) and 6-aldehydo-isoophiopogonanone B (AIO-B)) on tyrosinase (Tyr) was investigated by multi-spectroscopic techniques combined with molecular docking. The results demonstrated that AIO-A suppressed the activity of Tyr in a reversible competitive-inhibition with a half inhibitory concentration (IC50) of (12.67 ± 2.52) × 10–5 molˑL−1, while AIO-B was a reversible mixed-inhibition with IC50 of (4.81 ± 0.74) × 10–5 molˑL−1. AIO-A/AIO-B bound to Tyr driven by the intermolecular interactions, including van der Waals interaction, hydrogen bonding and hydrophobic interaction, thereby resulted in a secondary structure change of Tyr and its intrinsic fluorescence quenching. The molecular docking revealed that an aldehyde oxygen atom in AIO-A and methoxyphenyl in AIO-B formed coordination with two Cu ions in the active center of Tyr protein. It can be deduced that the coordination with two Cu ions may prevent the entrance of substrate and then inhibit the catalytic activity of Tyr. The research might provide new perspectives on the inhibition mechanism of AIO-A and AIO-B on Tyr and functional research for the prevention and treatment of pigmented skin diseases and anti-browning of homoisoflavonoids as a food supplement.

Abstract:The pigmentation of the skin, modulated by different actors in melanogenesis, is mainly due to the melanins (protective pigments). In humans, these pigments' precursors are synthetized by an enzyme known as tyrosinase (TyH). The regulation of the enzyme activity by specific modulators (inhibitors or activators) can offer a means to fight hypo‐ and hyper‐pigmentations responsible for medical, psychological and societal handicaps. Herein, we report the investigation of phenylalanine derivatives as TyH modulators. Interacting with the binuclear copper active site of the enzyme, phenylalanine derivatives combine effects induced by combination with known resorcinol inhibitors and natural substrate/intermediate (amino acid part). Computational studies including docking, molecular dynamics and free energy calculations combined with biological activity assays on isolated TyH and in human melanoma MNT‐1 cells, and X‐ray crystallography analyses with the TyH analogue Tyrp1, provide conclusive evidence of the interactions of phenylalanine derivatives with human tyrosinase. In particular, our findings indicate that an analogue of L‐DOPA, namely (S)‐3‐amino‐tyrosine, stands out as an amino phenol derivative with inhibitory properties against TyH.

Abstract:α-Tocopheroxide is not readily reduced polarographically and in mixtures with α-tocopherol does not give a stable electromotive potential; thus α-tocopheroxide is not the electromotively active, bivalent oxidation product detectable polarographically. Various experiments are reported on the conversion of α-tocopheroxide to α-tocopherol and to α-tocopherylquinone; under weakly acidic conditions α-tocopheroxide readily forms α-tocopherylquinone, whereas under similar conditions with ascorbic acid present α-tocopherol is formed. The formation of a common intermediate with a hemi-ketal structure in both reactions is considered as a likely possibility. No intermediate was detected spectrophotometrically during the slow conversion of α-tocopheroxide to α-tocopheryl;quinone near neutral pH. Treatment of α-tocopherylquinone in absolute ethanol with ascorbic and hydrochloric acids converts the quinone principally to α-tocopherol, or a product closely resembling α-tocopherol, by a pathway not involving α-tocopherylhydroquinone; with some preparations considerable amounts of products of unknown structure are formed. Ascorbate and α-tocopherol act synergistically in the prevention of the acceleration by cytochrome c of the oxidation of linoleate near neutral pH. Neither α-tocopheroxide or α-tocopherylquinone are effective with or without ascorbate. The implications of these and other findings are discussed.

Abstract:Tyrosine is a conserved redox-active amino acid that plays important roles in heme-copper oxidases (HCO). Despite the widely proposed mechanism that involves a tyrosyl radical, its direct observation under O2 reduction conditions remains elusive. Using a functional oxidase model in myoglobin called F33Y-Cu(B)Mb that contains an engineered tyrosine, we report herein direct observation of a tyrosyl radical during both reactions of H2O2 with oxidized protein and O2 with reduced protein by electron paramagnetic resonance spectroscopy, providing a firm support for the tyrosyl radical in the HCO enzymatic mechanism.

Abstract:Tyrosinase is the central enzyme involved in the highly complex process of melanin formation, catalyzing the rate-limiting steps of this biosynthetic pathway. Due to such a preponderant role, it has become a major target in the treatment of undesired skin pigmentation conditions and also in the prevention of enzymatic food browning. Numerous phenolic-based structures from natural sources have been pointed out as potential tyrosinase inhibitors, including anthocyanins. The aim of the present study was to individually assess the tyrosinase inhibitory activity of eight purified compounds with a variable degree of structural complexity: native anthocyanins, deoxyanthocyanins, and pyranoanthocyanins. The latter two, the groups of anthocyanin-related compounds with enhanced stability, were tested for the first time. Compounds 1 to 4 (luteolinidin, deoxymalvidin, cyanidin-, and malvidin-3-O-glucoside) revealed to be the most effective inhibitors, and further kinetic studies suggested their inhibition mechanism to be of a competitive nature. Structure–activity relationships were proposed based on molecular docking studies conducted with mushroom tyrosinase (mTYR) and human tyrosinase-related protein 1 (hTYRP1) crystal structures, providing information about the binding affinity and the different types of interactions established with the enzyme’s active center which corroborated the findings of the inhibition and kinetic studies. Overall, these results support the applicability of these compounds as pigmentation modulators.

Abstract:Mushroom tyrosinase (EC 1.14.18.1) is a copper containing oxidase that catalyzes both the hydroxylation of tyrosine into o -diphenols and the oxidation of o -diphenols into o -quinones, and then forms brown or black pigments. In the present study, the effects of some flavonoids on the oxidation of L-3,4-dihydroxyphenylalanine (L-DOPA) have been studied. The results show that flavonoids can lead to reversible inhibition of the enzyme. A kinetic analysis showed that the flavonols are competitive inhibitors, whereas luteolin is an uncompetitive inhibitor. The rank order of inhibition was: quercetin > galangin > morin; fisetin > 3,7,4"-trihydroxyflavone; luteolin > apigenin > chrysin.

Abstract:The oxidation of natural flavonoid luteolin in aqueous solution is studied by electrochemical methods, electron paramagnetic resonance (EPR), spectroelectrochemistry and separation techniques HPLC-DAD and HPLC–MS/MS. The number of electrons involved in the oxidation of luteolin depends on the presence of its dissociation forms in solution. The study explains the differences in the number of electrons presented in the literature. The overall one electron oxidation mechanism of luteolin in alkaline solution is explained by the comproportionation reaction of resulting quinone, despite the fact that quinone is formed by two electron oxidation. Then a hydroxylation takes place. The EPR spectroelectrochemical study of the semiquinone radical anion formation as well as of the reaction steps following the electron transfer during the oxidation is presented. The novelty of this contribution consists in the additional temperature controlled semi-quantitative in situ EPR spectroelectrochemical experiment of the flavonoid oxidation. The data acquired by temperature controlled in situ EPR spectroelectrochemistry supports the comproportionation/disproportionation equilibria as well as the oxidative decomposition of luteolin and shows that the formation of a pi-dimer is less probable. The oxidation products hydroxy-luteolin and 3,5-dihydroxy-2-(2-oxoacetyl)phenyl-3,4-dihydroxybenzoate are not stable under ambient conditions and decompose to low molecular hydroxycompounds such as 3,4-dihydroxybenzoic acid and 2,5,7-trihydroxy-4H-1-benzopyran-4-one.

Synthesis, Identification, and Structure Elucidation of Adducts Formed by Reactions of Hydroxycinnamic Acids with Glutathione or Cysteinylglycine

Abstract:Grape polyphenols, especially hydroxycinnamic acids such as caftaric and caffeic acid, are prone to enzymatic oxidation reactions during the winemaking process, forming o-quinones and leading to color darkening. Glutathione is capable of trapping these o-quinones and thus limiting juice browning. In this study, the addition of glutathione or cysteinylglycine onto caftaric or caffeic acid o-quinones formed by polyphenoloxidase-catalyzed reactions was investigated by UPLC-DAD-ESIMS and NMR data analyses. Complete identification of adducts has been achieved via NMR data. The results confirmed that the favored reaction is the substitution of the sulfanyl group of cysteine at C-2 of the aromatic ring. Several minor isomers, namely, the cis-isomer of the 2-S adduct and trans-isomers of the 5-S and 6-S adducts, and the 2,5-di-S-glutathionyl adducts were also identified and quantified by qNMR. With the exception of 2-(S-glutathionyl)- and 2,5-di(S-glutathionyl)-trans-caftaric acid, these products had never been formally identified. In particular, the 5-S and 6-S derivatives are reported here for the first time. The first formal identification of 2-S cis-derivatives is also provided. Moreover, NMR and UPLC-DAD-ESIMS analysis showed that signature UV and MS spectra can serve as markers of the conformation and substitution position in the aromatic ring for each of the isomers.
Concentration Dependence of Anti- and Pro-Oxidant Activity of Polyphenols as Evaluated with a Light-Emitting Fe2+-Egta-H2O2 System

Abstract:Hydroxyl radical (•OH) scavenging and the regeneration of Fe2+ may inhibit or enhance peroxidative damage induced by a Fenton system, respectively. Plant polyphenols reveal the afore-mentioned activities, and their cumulative net effect may determine anti- or pro-oxidant actions. We investigated the influence of 17 phenolics on ultra-weak photon emission (UPE) from a modified Fenton system (92.6 μmol/L Fe2+, 185.2 μmol/L EGTA (ethylene glycol-bis(β-aminoethyl-ether)-N,N,N′,N,-tetraacetic acid) and 2.6 mmol/L H2O2 pH = 7.4). A total of 8 compounds inhibited (antioxidant effect), and 5 enhanced (pro-oxidant effect) UPE at all studied concentrations (5 to 50 μmol/L). A total of 4 compounds altered their activity from pro- to antioxidant (or vice versa) along with increasing concentrations. A total of 3 the most active of those (ferulic acid, chlorogenic acid and cyanidin 3-O-glucoside; mean UPE enhancement by 63%, 5% and 445% at 5 μmol/L; mean UPE inhibition by 28%, 94% and 24% at 50 μmol/L, respectively) contained catechol or methoxyphenol structures that are associated with effective •OH scavenging and Fe2+ regeneration. Most likely, these structures can determine the bidirectional, concentration-dependent activity of some phenolics under stable in vitro conditions. This is because the concentrations of the studied compounds are close to those occurring in human fluids, and this phenomenon should be considered in the case of dietary supplementation with isolated phenolics.

Abstract:Tyrosinase is generally known as a melanin-forming enzyme, facilitating monooxygenation of phenols, oxidation of catechols into quinones, and finally generating biological melanin. As a homologous form of tyrosinase in plants, plant polyphenol oxidases perform the same oxidation reactions specifically toward plant polyphenols. Recent studies reported synthetic strategies for large scale preparation of hydroxylated plant polyphenols, using bacterial tyrosinases rather than plant polyphenol oxidase or other monooxygenases, by leveraging its robust monophenolase activity and broad substrate specificity. Herein, we report a novel synthesis of functional plant polyphenols, especially quercetin and myricetin from kaempferol, using screened bacterial tyrosinases. The critical bottleneck of the biocatalysis was identified as instability of the catechol and gallol under neutral and basic conditions. To overcome such instability of the products, the tyrosinase reaction proceeded under acidic conditions. Under mild acidic conditions supplemented with reducing agents, a bacterial tyrosinase from Bacillus megaterium (BmTy) displayed efficient consecutive two-step monophenolase activities producing quercetin and myricetin from kaempferol. Furthermore, the broad substrate specificity of BmTy toward diverse polyphenols enabled us to achieve the first biosynthesis of tricetin and 3′-hydroxyeriodictyol from apigenin and naringenin, respectively. These results suggest that microbial tyrosinase is a useful biocatalyst to prepare plant polyphenolic catechols and gallols with high productivity, which were hardly achieved by using other monooxygenases such as cytochrome P450s.

Abstract:Catechin, epicatechin and chlorogenic acid are widely distributed in the plant kingdom. At present, influencing factors of phenol chemical oxidation is little research. In order to study non-enzymatic browning factors, this research utilized catechin, epicatechin and chlorogenic acid to establish simulation systems. The browning degree and products of flavanols were investigated by transmittance and high-performance liquid chromatography (HPLC). The main results and conclusions were follows. The brown generation is increased after phenols of chemical oxidation at 50 degrees C and at pH 3.7 phosphate buffered saline, the sequence of influencing factor of browning is pH > kind of phenol > temperature. Oxidation of compounds of catechin and epicatechin results in formation of their Methylene quinone or alpha-Quinones. In addition, oxidation products of catechin, epicatechin and chlorogenic acid were mixture of different molecular sizes. The research has showed that brown generation correlated well with chemical oxidation of phenols and chemical oxidation reaction generated larger molecular weight polymers.

Abstract:Flavonoids are naturally occurring phenol derivatives present in substantial amounts in a large variety of plants, fruits and vegetables daily eaten by humans. Most of these compounds exhibit several interesting biological activities, such as antiradical and antioxidant actions. Indeed, by complexation with specific enzymes, flavonoids are notably liable to metabolize molecular dioxygen. On the basis of experimental results describing oxygenolysis of the flavonoid quercetin, activated by the enzyme quercetin 2,3-dioxygenase (2,3-QD),ur attention has focused on the role of metal center in the activation of the substrate quercetin. Thus, in the present study, by means of DFT calculations at the B3LYP/ 6-31(+)G* level on model molecular systems, we describe different mechanisms for dioxygen metabolization by quercetin. Stationary points are described, and energetic and structural analyses along the reaction paths are reported. Our calculations show that the copper cation must act as an oxidant towards the substrate and that the reaction proceeds through a 1,3-cycloaddition.

Abstract:Tyrosinase, the most predominant member of the coupled binuclear copper protein family, binds and activates dioxygen towards the regioselective monooxygenation of monophenols to catechols in melanin biosynthesis. In this Perspective, we discuss past and current efforts to define the mechanism of this critical monooxygenation reaction, in the context of the recent elucidation of its ternary (tyrosinase/dioxygen/monophenol) intermediate. Tyrosinase is the most predominant member of the coupled binuclear copper (CBC) protein family. The recent trapping and spectroscopic definition of the elusive catalytic ternary intermediate (enzyme/O2/monophenol) of tyrosinase dictates a monooxygenation mechanism that revises previous proposals and involves cleavage of the μ‐η2:η2‐peroxide dicopper(II) O‐O bond to accept the phenolic proton, followed by monophenolate coordination to copper concomitant with aromatic hydroxylation by the non‐protonated μ‐oxo. Here, we compare and contrast previously proposed and current mechanistic models for monophenol monooxygenation of tyrosinase. Next, we discuss how these recent insights provide new opportunities towards uncovering structure‐function relationships in CBC enzymes, as well as understanding fundamental principles for O2 activation and reactivity by bioinorganic active sites.
