Abstract:The usage of food-derived polyphenols with different polarities has been limited by their instability and incompatibility. Therefore, a biocarrier was developed by co-assembly of whey protein isolate (WPI) and hydrophilic proanthocyanidin (PC) for loading hydrophobic pterostilbene (PTE). Such biocarrier has superior affinity for PTE than WPI alone as determined by encapsulation efficiency (EE) and loading capacity (LC) assay, fluorescence quenching analysis and molecular docking, while assembly process was characterized by particle size and zeta-potential, 3D fluorescence and SEM. CD and FTIR spectra confirmed the α-helix to β-sheet and random coil transition of proteins during the nanocomplexes formation. WPI acted as a mediator through altering the binding mode of PC and PTE, allowing them to perform significant synergistic effects in enhancing ABTS and DPPH radical scavenging and reducing H2O2-induced cell damage. This research may serve to develop new protein/polyphenol co-loading systems and offer a reliable nutritional fortification.

Abstract:Phytosterol (PS)/whey protein isolate (WPI) self-assembly complex was formed with different PS:WPI mass ratios (from 1:1 to 1:15) to reveal the relationship of interaction mechanism in PS/WPI complex, WPI secondary structure, and PS digestion properties. The sample with 1:5 mass ratio had the strongest hydrogen bonds and the largest encapsulation efficiency (EE). Circular dichroism analysis indicated that the mass ratio of PS/WPI complex had a vital effect on WPI secondary structure. The content of β-sheet increased with the increase in PS ratio, which, in turn, inhibited the release rate and bioaccessibility of PS. The β-turn of WPI was positively correlated with hydrogen bonds in the complex, thus playing a significant role in increasing EE, promoting PS release, and even further enhancing PS bioaccessibility in digestion simulation. These findings appeared to be promising for future applications in enhancing the bioaccessibility of phytosterol through specific structure in high-protein food systems.

Abstract:Phytosterol (PS) is a steroid, and its bioavailability can be enhanced by interacting with protein in the C-24 hydroxyl group. The interaction between sterols and amino acid residues in proteins can be enhanced by enzymatic hydrolysis. Phytosterol and whey insulation hydrolysates (WPH1-4) fabricated by the Alcalase enzyme at different enzymatic hydrolysis times were selected as delivery systems to simulate sterol C-24 hydroxyl group interaction with protein. Increasing hydrolysis time can promote the production of beta-Lg, which raises the ratio of beta-turn in the secondary structure and promotes the formation of interaction between WPH and PS. The correlation coefficient between hydrogen bonds and encapsulation efficiency (EE) and bioaccessibility is 0.91 and 0.88 (P < 0.05), respectively, indicating that hydrogen bonds of two components significantly influenced the combination by concealing the hydrophobic amino acids and some residues, which improved PS EE and bioavailability by 3.03 and 2.84 times after PS was combined with the WPI hydrolysate. These findings are expected to enhance the absorption of PS and other macromolecules by protein enzymatic hydrolysis to broaden their applications for food.
Whey Protein Isolate/flavor Cinnamaldehyde Conjugates Prepared by Different Methods and the Stabilization Effects on Β-Carotene-loaded Emulsions

Abstract:Whey protein isolate (WPI) interacts with cinnamaldehyde (CA) to form conjugates with superior functional properties, which are affected by the conjugate structures formed by different food processing techniques, especially heating treatment. Initially, we compared three preparation methods of conjugates, including heating WPI before interacting with CA [(HWPI)/CA)], heating the resulting conjugates without heating WPI initially [H(WPI/CA)], and the normal conjugates without any heating process (WPI/CA). Subsequently, these conjugates were used to stabilize emulsions for the encapsulation and delivery of 13-carotene. Both the aforementioned heating methods increased the interaction and enhanced the conjugates' surface hydrophobicity. Synchronous fluorescence suggested that heating expanded the protein structure and exposed hydrophobic groups, while higher CA concentration had a shielding effect on this process. Moreover, H-(WPI/CA) conjugates reduced the interfacial tension to 4.38 mN/m, resulting in the best emulsification, which effectively stabilized emulsions and protected 13-carotene from the influence of temperature. H-(WPI/CA) conjugates helped to maintain the retention rate at more than 70% within 28 days and improved 13-carotene bioaccessibility during digestion. This knowledge provides insights into investigating the relationship between conjugates preparation methods and their properties, and is of great significance for developing a novel multifunctional food ingredient system based on protein modification.

Abstract:A glycosylated protein/procyanidin complex was prepared by self-assembly of glycosylated whey protein isolate and proanthocyanidins (PCs). The complex was characterized through endogenous fluorescence spectroscopy, polyacrylamide gel electrophoresis, Fourier infrared spectroscopy, oil–water interfacial tension, and transmission electron microscopy. The results showed that the degree of protein aggregation could be regulated by controlling the added amount of procyanidin, and the main interaction force between glycosylated protein and PCs was hydrogen bonding or hydrophobic interaction. The optimal binding ratio of protein:PCs was 1:1 (w/w), and the solution pH was 6.0. The resulting glycosylated protein/PC compounds had a particle size of about 119 nm. They exhibited excellent antioxidant and free radical-scavenging abilities. Moreover, the thermal denaturation temperature rose to 113.33 °C. Confocal laser scanning microscopy (CLSM) images show that the emulsion maintains a thick interface layer and improves oxidation resistance with the addition of PCs, increasing the application potential in the functional food industry.

Abstract:(−)-Epigallocatechin-3-gallate (EGCG) was conjugated to whey protein isolate (WPI) using alkaline treatment, free radical grafting and tyrosinase-catalyzed oxidation. The structure and properties of the obtained conjugates and their application as antioxidant emulsifiers in stabilizing β-carotene emulsion were systematically characterized. Fourier transform infrared spectroscopy and sodium dodecyl sulfate–polyacrylamide gel electrophoresis verified the covalent linking between WPI and EGCG. The highest grafting efficiency was obtained for enzyme conjugate, followed for alkaline conjugate. Conjugation of EGCG decreased the α-helix content and fluorescence intensity of protein, and these changes depend on both EGCG conjugation and the cross-linking methods. Due to their improved emulsifying properties and antioxidant activity, WPI-EGCG conjugates formed smaller emulsion droplets and protect the encapsulated β-carotene more effectively, and the protective property is positively correlated with EGCG content in conjugates.

Abstract:In this study, pectin (PEC) and pea protein isolate(PPI) was successfully used to create complexes as a novel delivery system for pterostilbene (PT). When the mass ratio of PEC to PPI was 0.5, the particle size and ζ-potential of PPI-PEC-PT were 119.41 ± 5.68 nm and –23.26 ± 0.61 mV, respectively, and the encapsulation efficiency (EE) of PT was 90.92 ± 2.08%. The photochemical stability of PT was enhanced after encapsulation. The results of the molecular docking and multispectral analysis demonstrated that the PPI and PT binding was spontaneous and mostly fueled by hydrophobic interactions. The hydrophobicity of PPI was significantly decreased and the emulsification activity and emulsion stability were significantly improved after production with PEC and PT. The best emulsification impact was demonstrated by the PPI-PEC-PT complex. PPI-PEC is an effective PT delivery material, and the PPI-PEC-PT complex is a new functional emulsification material with significant potential in liquid and semi-liquid food and health products.

Abstract:This study investigated the effects of the interaction of walnut protein isolate (WPI) with epigallocatechin gallate (EGCG), chlorogenic acid (CLA), (+)-catechin (CA), and ellagic acid (EA) on the structural and functional properties of proteins. The results for polyphenol binding equivalents and content of free amino and sulfhydryl groups as well as those from sodium dodecyl sulfate‒polyacrylamide gel electrophoresis confirmed the covalent interaction between WPI and the polyphenols. The binding capacities of the WPI-polyphenol mixtures and conjugates were as follows: WPI-EGCG > WPI-CLA > WPI-CA > WPI-EA. Fourier transform infrared spectroscopy (FTIR) and fluorescence spectrum analysis identified changes in the protein structure. The conjugation process obviously increased the polyphenols' antioxidant properties and the surface hydrophobicity was substantially reduced. WPI-EGCG conjugates had the best functional properties, followed by WPI-CLA, WPI-CA, and WPI-EA. Lycopene (LYC) was loaded into nanocarriers by WPI-EGCG self-assembly. These results indicated that WPI-polyphenol conjugates can be utilized to develop food-grade delivery systems to protect chemically lipophilic bioactive compounds.

Abstract:The co-encapsulation of multiple bioactive components in a carrier may produce synergistic effects and improve health benefits. In this study, the interactions of β-lactoglobulin (β-LG) with epigallocatechin-3-gallate (EGCG) and/or piceatannol (PIC)/oxyresveratrol (OXY) were investigated by multispectroscopic techniques, isothermal titration calorimetry, and molecular docking. The static quenching mechanism of β-LG by EGCG, PIC and OXY was confirmed by fluorescence spectroscopy and UV-vis absorption difference spectroscopy. The binding sites of these three polyphenols in β-LG were identified by site marking fluorescence experiments and molecular docking. The thermodynamic parameters of the β-LG + EGCG/PIC/OXY binary complex and β-LG + EGCG + PIC/OXY ternary complex were obtained from fluorescence data and used to analyze the main driving force for complex formation. The exothermic binding process was further confirmed by isothermal titration calorimetry. The α-helical content, particle size and morphology of free and ligand-bound β-LG were determined by circular dichroism spectroscopy, dynamic light scattering and transmission electron microscopy, respectively. The effect of EGCG, PIC and OXY on the conformation of β-LG was studied by Fourier transform infrared spectroscopy. In addition, the maximum synergistic antioxidant activity between EGCG and PIC/OXY was obtained by response surface analysis. The effects of β-LG in the binary and ternary systems on the antioxidant activity, stability, solubility and cytotoxicity of the polyphenols were also studied. Finally, the different cytotoxicities of the complexes and nanoparticles of the binary and ternary systems were compared. The results of this study are expected to provide a theoretical basis for the development of β-LG-based carriers co-encapsulating a variety of bioactive components.

Abstract:Free radical grafting is a green and expeditious tool that was used to modify whey protein isolate (WPI) to synthesize WPI-(−)-epigallocatechin-3-gallate (EGCG) and WPI-catechin (C) antioxidant emulsifier and improve its functional and interfacial properties. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and matrix assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF–MS) were used to verify the covalent bonding between polyphenols and WPI. Results revealed that at least two EGCG molecules and three catechin molecules were grafted onto WPI. Meanwhile, the grafting of tea polyphenols, especially EGCG, with WPI decreased their fluorescence intensity; α-helix content; and free amino, sulfhydryl, and tyrosine group contents. A lycopene nanoemulsion constructed with the WPI-EGCG conjugate had smaller changes in size and ζ-potential than that constructed with WPI during 30-day storage, which was attributed to its enhanced emulsifying activity, emulsification stability, and ability to reduce the interfacial tension of conjugates. The improvement in oxidation resistance and interfacial adsorption layer thickness helped the WPI-EGCG conjugate to significantly reduce the lycopene degradation rate during storage. Thus, WPI modified with polyphenol by free radical grafting can be used to construct an emulsion delivery system for functional nutrients with a stable interface and high retention rate.

Abstract:Background Recent studies have highlighted the potential of protein fibrils for the microencapsulation of lipophilic bioactive molecules. Given their biocompatible and biodegradable properties, as well as their enhanced material stability, proteins are ideal for this purpose. However, β-carotene, a common antioxidant, degrades in acidic environments. In light of this, the present study explored the stability and antioxidant properties of acidic/alkaline polysaccharide–whey protein isolate fibril (WPF) bilayer microcapsules containing β-carotene under acidic conditions. Methods WPFs were prepared and then homogenized with a β-carotene/limonene solution to form microcapsules. The addition of pectin and chitosan created polysaccharide–WPF bilayer microcapsules. Zetasizer was used to analyze the microcapsules’ size and stability, while confocal microscopy was used to observe their morphology under acidic conditions. The β-carotene concentration was determined via heptane/ethanol extraction. Significant findings The results show that the pectin–WPF bilayer microcapsules exhibited excellent stability under acidic conditions, which can enhance stability and retain the antioxidant activity of β-carotene. Thus, this work suggests pectin–WPF bilayer microcapsules to be promising carriers for lipophilic bioactive molecules due to enhancing their bioavailability.

Comparison of rapid detection methods for influenza A virus and their value in health-care management of institutionalized geriatric patients

Abstract:Respiratory specimens from 160 geriatric patients with suspected influenza illness were used to evaluate the abilities of two enzyme immunoassays (EIAs; Directigen FLU-A [Becton Dickinson Microbiology Systems, Cockeysville, Md.] and Prima EIA [Baxter/Bartels Diagnostics, Inc., Issaquah, Wash.]) and direct immunofluorescence testing (immunofluorescence assay [IFA]) to identify influenza A virus. In comparison with culture isolation, the sensitivities and specificities of the IFA, Directigen FLU-A, and Prima EIA were 92.5 and 97.2%, 86.8 and 99.1%, and 92.5 and 98.1%, respectively. In contrast to EIA, IFA was labor intensive and required a high degree of technical expertise, and the results of IFA were difficult to interpret. These factors may preclude the use of IFA for testing large numbers of specimens. A retrospective epidemiologic survey of influenza infection was done in six geriatric institutions which had used either rapid and culture testing or culture alone. Preventable cases of influenza A virus infection ranged from 9 to 38% of all cases in facilities which used culture testing only and which had not instituted amantadine prophylaxis. The use of direct specimen testing is recommended as an adjunct to culture isolation for the identification of influenza A virus. Use of a combination of these methods permits the timely administration of appropriate antiviral therapy and infection control measures, while it also permits the antigenic surveillance of circulating influenza strains, which is necessary for present vaccine efficacy evaluations and the creation of future effective vaccine formulations.

Abstract:When consumed at sufficiently high levels, polyphenols may provide health benefits, which is linked to their antidiabetic, antiinflamatory, antimicrobial, antioxidant, antitumor, and hypolipidemic properties. Moreover, certain polyphenol combinations exhibit synergistic effects when delivered together – the combined polyphenols have a higher biological activity than the sum of the individual ones. However, the commercial application of polyphenols as nutraceuticals is currently limited because of their poor solubility characteristics; instability when exposed to light, heat, and alkaline conditions; and, low and inconsistent oral bioavailability. Colloidal delivery systems are being developed to overcome these challenges. In this article, we review the design, fabrication, and utilization of food-grade biopolymer-based delivery systems for the encapsulation of one or more polyphenols. In particular, we focus on the creation of delivery systems constructed from edible proteins and polysaccharides. The optimization of biopolymer-based delivery systems may lead to the development of innovative polyphenol-enriched functional foods that can improve human health and wellbeing.

Abstract:Procyanidins (PC) are formed by the polymerisation of flavan-3-ol monomers, which have excellent bioactivity and present great health benefits. However, the application range of the PC is greatly limited due to its poor stability. Therefore, in this study, starch aerogels were used as carriers to encapsulate PC. After screening, wheat starch aerogel (WSA) was finally chosen to encapsulate PC. Afterward, characterization (SEM, XRD, FT-IR, TGA and BET), stability testing, investigations on antioxidant activity, and in vitro tests simulating digestion were carried out. The results of relevant characterization showed that the micro surface structure of the procyanidins wheat starch aerogel (PC-WSA) presented a network structure, and PC were encapsulated in WSA. In addition, PC-WSA allows PC to be released in the final stage of simulated digestion. Meanwhile, the results of antioxidant experiments indicate that PC-WSA exhibits antioxidant activity after encapsulating PC. Moreover, the retention of PC in PC-WSA was still 53.63 ± 3.23% when stored at 70°C for 20 days. After 6 days of light exposure, the retention of PC in PC-WSA under different light sources was all above 85%, which was higher than that of PC. In conclusion, the wheat starch aerogel could improve the stability of PC and has great potential for application.

Abstract:Houttuynia cordata polysaccharides (PSY) are known to exhibit a variety of beneficial activities, but these are currently not specifically utilized in food. Hence, using the two edible parts of Houttuynia cordata, a herbaceous plant native to Southeast Asia, this study developed polysaccharides of a stem (HCPS)-whey protein concentrate (WPC) complex and a leaf (HCPL)-WPC complex, and studied their stability, structure and antioxidant activity. The results showed that stability differed in complexes with different proportions, exhibiting only relative stability in the two complexes in which the ratio of HCPS-WPC and HCPL-WPC was 1:4, but increased stability in the HCPL-WPC complex (ζ-potential of HCPL-WPC: | -21.87 mv| >ζ-potential of HCPS-WPC: | -21.70 mv|). Structural characterization showed that there was electrostatic interaction between HCPS and WPC and between HCPL and WPC. The HCPL-WPC was found to have better antioxidant activity. The findings of this study, thus, provide a reference for the development of Houttuynia cordata polysaccharide applications in food.

Abstract:The ages of stellar populations in 52 elliptical and S0 galaxies in the Coma Cluster are investigated, using a new spectroscopic sample. More than 40% of the S0's are found to have undergone star formation in their central regions during the last ~5 Gyr, while such activity is absent in the ellipticals. Galaxies in this sample have absolute magnitudes in the range -20.5 < MB < -17.5, and the fraction of S0 galaxies with recent star formation is higher at fainter luminosities. The observed luminosity range of S0 galaxies with signs of recent star formation activity is consistent with them being the descendants of typical star-forming spirals at intermediate redshift whose star formation has been halted as a consequence of the dense environment.

Abstract:(-)-Epigallocatechin-3-gallate (EGCG) is the main active component of tea polyphenols, which endows tea with bitter and astringent taste and is susceptible to degradation in presence of light, heat and oxygen. In this study, whey protein isolate (WPI) and (3-cyclodextrin ((3-CD) were used to prepare the WPI-(3-CD nanocomplexes for masking the bitter taste and protecting EGCG. The encapsulation efficiency of EGCG by WPI-(3-CD nanocomplexes reached to 79.61%. Sensory evaluation indicated that the bitter and astringent taste of EGCG was masked. FTIR results suggested that the WPI/(3-CD-EGCG nanocomplexes were formed through electrostatic, hydrogen bonding and hydrophobic interactions among WPI, (3-CD and EGCG. The molecular mechanism of taste masking was preliminarily investigated by molecular docking, suggesting that the bitter and astringent taste moiety of EGCG was inserted into the hydrophobic cavity of (3-CD, thereby masking the bitter and astringent taste of EGCG. The results of in vitro gastrointestinal experiments and scanning electron microscopy indicated that the WPI-(3-CD nanocomplexes effectively encapsulated EGCG and achieved sustained release in simulated gastrointestinal conditions, thus showing potential application prospects in tea beverages and related areas.
High internal phase double emulsions stabilized by modified pea protein-alginate complexes: Application for co-encapsulation of riboflavin and β-carotene

Abstract:The application of many hydrophilic and hydrophobic nutraceuticals is limited by their poor solubility, chemical stability, and/or bioaccessibility. In this study, a novel Pickering high internal phase double emulsion co-stabilized by modified pea protein isolate (PPI) and sodium alginate (SA) was developed for the co-encapsulation of model hydrophilic (riboflavin) and hydrophobic (β-carotene) nutraceuticals. Initially, the effect of emulsifier type in the external water phase on emulsion formation and stability was examined, including commercial PPI (C-PPI), C-PPI-SA complex, homogenized and ultrasonicated PPI (HU-PPI), and HU-PPI-SA complex. The encapsulation and protective effects of these double emulsions on hydrophilic riboflavin and hydrophobic β-carotene were then evaluated. The results demonstrated that the thermal and storage stabilities of the double emulsion formulated from HU-PPI-SA were high, which was attributed to the formation of a thick biopolymer coating around the oil droplets, as well as thickening of the aqueous phase. Encapsulation significantly improved the photostability of the two nutraceuticals. The double emulsion formulated from HU-PPI-SA significantly improved the in vitro bioaccessibility of β-carotene, which was mainly attributed to inhibition of its chemical degradation under simulated acidic gastric conditions. The novel delivery system may therefore be used for the development of functional foods containing multiple nutraceuticals.

Abstract:Pomegranate peel polyphenols (PPPs) are recognized as promising food additives due to their diverse bioactivities; however, their application is limited by poor stability. To address this critical issue, three types of PPPs microcapsules were prepared using β-cyclodextrin (CD), whey protein isolate (WPI), and a composite material of CD-WPI through ultrasound treatment (US). Results revealed that ultrasound treatment can enhance the PPPs-wall material interaction, as evidenced by MD simulations. The encapsulation efficiency of CD-WPI-PPPs was 93.73 %, which was significantly higher than that of CD-PPPs and WPI-PPPs (p < 0.05). The degradation rate constant of CD-WPI-PPPs was reduced by 95.83 %, and its t1/2 was extended by 23-fold compared to that of unencapsulated PPPs. Furthermore, CD-WPI-PPPs exhibited greater DPPH scavenging activity and inhibited polyphenol release during oral and gastric digestion while promoting release during intestinal digestion. These outcomes were attributed to enhanced integrity and interactions between PPPs and composite materials in the microcapsules formed through ultrasound treatment, as supported by SEM images and FT-IR spectra. Consequently, the application of US in the preparation of PPPs microcapsules presents a promising strategy for developing natural nutrient additives for food applications, thereby enhancing the functional properties of food products.
