Abstract:Colloidal delivery systems have been widely used as carriers for controlled delivery of pesticides to improve the efficacy and photostability of natural and semi-synthetic pesticides. In this study, we have synthesized emamectin benzoate nanoformulations (EB + NFs) depending on polymeric nanocapsules (PNC) and two types of the nanosilica, mesoporous nanosilica (MCM-48) and silicon dioxide nanoparticles (SNPs) as carriers for the emamectin benzoate (EB). The fabricated nanoformulations were characterized by using X-ray diffraction analysis, Fourier transform infrared spectroscopy, particle size, zeta potential, morphology, absolute recovery (AR), entrapment efficiency (EE), UV stability and release kinetics. The obtained results showed that the carriers had a remarkable loading ability for EB and improved the EB photostability. The EE% of nanoformulations were 92.84%, 87.45% and 71.19% for emamectin benzoate polymeric nanocapsules (EB + PNC), emamectin benzoate SNPs (EB + SNPs) and emamectin benzoate MCM-48 (EB + MCM-48) respectively. The insecticidal activity of EB + NFs against Plutella xylostella showed that the EB + SNPs was more effective than other EB + NFs and EB alone. The LC50 values were 0.18, 4.03, 8.49 and 11.06 mg L-1 for EB + SNPs, EB + MCM-48, EB + PNC and EB respectively. The obtained results suggest the colloidal delivery systems that used in this study could improve the efficacy and photostability for EB, and they are able to overcome the disadvantage of the natural and semi-synthetic pesticides such as environmental sensitivity and to increase the efficacy of pesticides, which eventually leads to reduce the dosage of pesticides needed, reducing the number of applications required in comparison to conventional formulations.
Synthesis, Characterization, and Pesticidal Activity of Emamectin Benzoate Nanoformulations Against Phenacoccus Solenopsis Tinsley (hemiptera: Pseudococcidae)

Abstract:Using nanotechnology to develop new formulations of pesticides is considered a possible option in enhancing the efficiency, safety, and photostability of pesticides under various climatic conditions. In the present study, two novel nanoformulations (NFs) were successfully prepared based on nano-delivery systems for emamectin benzoate (EMB) by loading it on cellulose nanocrystals (CNCs) and silicon dioxide nanoparticles (SNPs) as carriers through a freeze-drying method. The synthesized nanoformulations were examined using field emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), and dynamic light scattering (DLS). The results showed that SNPs and CNCs had a loading efficiency of 43.31% and 15.04% (w/w) for EMB, respectively, and could effectively protect EMB from photolysis under UV radiation. The LC50 values for EMB + SNPs, EMB + CNCs, and EMB commercial formulation against Phenacoccus solenopsis were 0.01, 0.05, and 0.31 μg/mL, respectively, indicating that both NFs were more effective than the EMB commercial formulation. This work seeks to develop new nano-carriers for potential applications of pesticides in plant protection, which will reduce the recommended dose of pesticides and thereby decrease the amount of pesticide residue in food and the environment.
Cellulose Nanocrystals Loaded with Thiamethoxam: Fabrication, Characterization, and Evaluation of Insecticidal Activity Against Phenacoccus Solenopsis Tinsley (hemiptera: Pseudococcidae).

Abstract:Using smart nanopesticide formulations based on nanomaterials can offer promising potential applications for decreasing pesticide residues and their effects on human health and the environment. In this study, a novel nanoformulation (NF) of thiamethoxam (TMX) was fabricated using the solvent evaporation method through loading TMX on cellulose nanocrystals (CNCs) as the carrier. The synthesized TMX-CNCs was investigated through different techniques, such as Fourier transform infrared spectrometer (FT-IR), X-ray diffraction (XRD), transmission electron microscopy (TEM), dynamic light scattering (DLS), and thermogravimetric analysis (TGA). The results revealed that the loading efficiency and entrapment efficiency were 18.7% and 83.7 ± 1.8% for TMX, respectively. The prepared nanoformulation (TMX-CNCs) had a width of 7–14 nm and a length of 85–214 nm with a zeta potential of −23.6 ± 0.3 mV. The drug release behavior study exhibited that the release of TMX from TMX-loaded CNCs was good and sustained. Furthermore, bioassay results showed that the insecticidal activity of TMX-CNCs against Phenacoccus solenopsis was significantly superior to that of the technical and commercial formulation, as indicated by the lower LC50 value. The results indicate that the TMX nanoformulation has great potential for application in agriculture for pest control.

Abstract:Currently, the inefficient application of pesticides results in substantial cost wastage and severe environmental contamination. The rapid advancement of nanomaterials and controlled/sustained-release technologies offer a promising guidance for reducing pesticide usage and enhancing pesticide bioavailability. In this study, naturally derived Zein and β-cyclodextrin (β-CD) polysaccharide were employed for synergistic encapsulation of emamectin benzoate (EB) pesticide molecule. Specifically, amino-functionalized Zein (AM-Zein) was effectively self-assembled onto β-cyclodextrin (β-CD) through electrostatic and hydrogen bonding interactions, with simultaneous encapsulation of EB through hydrophobic interactions, thereby establishing a core-double shell structured nano pesticide delivery system (EB@β-CD/AM-Zein). Compared to the single AM-Zein and β-CD, the double shell structured β-CD/AM-Zein demonstrated a notably higher encapsulation efficiency for EB. Importantly, the half-life period (t 0.5 ) under UV irradiation of EB for EB@β-CD/AM-Zein was almost three times longer than that of EB alone. Enzyme-responsive controlled-release tests for EB@β-CD/AM-Zein revealed that the majority of EB was encapsulated within the inner β-CD layer. Specifically, the degradation of β-CD by α-amylase enabled the intelligent responsive release feature of EB. Moreover, the insecticidal activity of EB@β-CD/AM-Zein remained stable. This research offers valuable insights for developing effective and environmentally safe nano pesticide systems.

Abstract:Environmentally friendly pesticide delivery system has drawn extensive attention in recent years, which shows great promise in sustainable development of agriculture. We herein report a multifunctional nanoplatform, carboxymethyl chitosan modified carbon nanoparticles (CMC@CNP), as the carrier for emamectin benzoate (EB, a widely used insecticide), and investigate its sustainable anti-pest activity. EB was loaded on CMC@CNP nanocarrier via simple physisorption process, with a high loading ratio of 55.56 %. The EB@CMC@CNP nanoformulation showed improved solubility and dispersion stability in aqueous solution, which is of vital importance to its practical application. Different from free EB, EB@CMC@CNP exhibited pH-responsive controlled release performance, leading to sustained and steady EB release and prolonged persistence time. In addition, the significantly enhanced anti-UV property of EB@CMC@CNP further ensured its anti-pest activity. Therefore, EB@CMC@CNP exhibited superior pest control performance than free EB. In consideration of its low cost, easy preparation, free of organic solution, and enhanced bioactivity, we expect, CMC@CNP holds brilliant future in pest control and green agriculture.

Abstract:Self‐assembling of alginate with quaternary ammonium for instant nanoformulation. To prompt the nanopesticides from laboratory investigation to agricultural application and reduce the use of pesticides and the impacts on the environment, sodium alginate (SA) was self‐assembled with cetyl trimethyl ammonium bromide (CTAB) into stable and uniform nanoparticle under mild conditions. The structure of the nanoparticle was investigated by dynamic light scattering (DLS), ζ‐potential measurement, Fourier transform infrared spectroscopy (FTIR) and transmission electron microscopy (TEM) and related to the loading and release properties. It was found that the nanoparticle size increased and ζ‐potential decreased with increasing concentration of SA, leading to higher loading efficiency and sustained release efficacy towards emamectin benzoate (EB). The amount of CTAB had little effects on the size of the nanoparticle, however made a competition to EB and reduced the loading and sustaining efficacy of the nanocarrier. The release of EB from SA‐CTAB nanoparticle was pH‐sensitive and was slower in the medium of higher pH value, due to the increased interaction with the nanocarrier. The entrapment into the nanoparticle greatly increased the insecticidal toxicity of EB due to the interaction between the carriers and cell membrane. The median lethal concentration of typical nanoformulation against Mythimna separata larvae was 30% of that of technical EB, after feeding to the insect for 48 h.

Abstract:The self‐assembly of lignin and cationic surfactants into nanoparticles loaded with emamectin benzoate exhibits good sustained release effect, pH sensitive release attributes, and a better mortality rate against model pests compared to commercial emulsions. Nanoformulation was an important strategy to reduce the use of pesticides and the impacts on the environment. However, complex process and high cost hurdled the application in agriculture. In this work, sodium lignosulfonate (SL) and cetyltrimethyl ammonium chloride (CTAC) were combined into nanoparticles and utilized as carriers for emamectin benzoate (EB), using a one‐pot method at mild conditions. The structure of nanoparticle was investigated by dynamic light scattering, ζ‐potential measurement, Fourier transform infrared spectroscopy, and transmission electron microscopy, and related to the loading and release properties. The mass ratio of SL and CTAC had great impacts on the structure of nanoparticle. The release of EB from SL‐CTAC nanoparticles was pH‐triggered. The retarding efficacy was enhanced in the medium of higher pH, due to the increased interaction between the EB and carrier. The incorporation into the nanoparticles greatly increased the insecticidal toxicity of EB. The median lethal concentration, against Spodoptera litura Fabricius larvae, of typical nanoformulation was 55% and 63% of that of conventional formulation emulsifiable concentrates after feeding for 24 and 48 h, respectively. The results indicated SL‐CTAC nanoparticles were good candidates of nanovehicles with high efficiency and low‐cost, which was of great significance to prompt the nanopesticides from laboratory investigation to agricultural application.

Characterization of an antibacterial peptide from indian cobra (Naja naja) venom

Abstract:Due to the development of antibiotic resistance in microorganisms, antimicrobial peptides from natural sources have attracted attention in recent times. Several antimicrobial peptides have been isolated from a wide range of animal sources, particularly snake venoms. Naja naja venom showed antibacterial as well as direct and indirect hemolytic activities, and an antibacterial peptide was purified through gel permeation and ion exchange chromatography. Its molecular mass was 2491Da, which was determined using Matrix Assisted Laser Desorption/Ionization-Time-of-Flight (MALDI-TOF) mass spectrometry and the amino acids sequence of the N-terminus was DEQSTHGAYVWKL. The purified peptide showed potent antibacterial activity against Gram-negative and Gram-positive bacterial strains like Escherichia coli, Pseudomonas aeruginosa and Vibrio cholerae, and Staphylococcus aureus, Enterococcus faecalis, Streptococcus pneumoniae, Streptococcus pyogenes, Bacillus subtilis, respectively. The most potent activity was towards Gram-negative bacteria. Activity was retained at concentrations as low as 100µg/ml. Minimum inhibitory concentrations (MIC; in mg) of Naja Antibacterial Peptide (NAP) and known antibiotics against Gram-positive and Gram-negative bacteria were determined using microdilution susceptibility test in sterile 96-well microdilution plates. However, the peptide did not show direct or indirect hemolytic activity.
Size-dependent Effect on Foliar Utilization and Biocontrol Efficacy of Emamectin Benzoate Delivery Systems

Abstract:The development of nanopesticides provides new avenues for pesticide reduction and efficiency improvement. However, the size effect of nanopesticides remains unclear, and its underlying mechanisms of influence have become a major obstacle in the design and application of pesticide nanoformulations. In this research, the noncarrier-coated emamectin benzoate (EB) solid dispersions (Micro-EB and Nano-EB) were produced under a constant surfactant-to-active ingredient ratio by a self-emulsifying-carrier solidification technique. The particle size of Micro-EB was 162 times that of spherical Nano-EB. The small size and large specific surface area of Nano-EB facilitated the adsorption of surfactants on the surface of the particles, thereby improving its dispersibility, suspensibility, and stability. The pinning effect of nanoparticles significantly suppressed droplet retraction and rebounding. Moreover, Nano-EB exhibited a 25% higher retention of the active ingredient on cabbage leaves and a 70% higher washing resistance than Micro-EB, and both were significantly different. The improvement of abilities in wetting, spreading, and retention of Nano-EB on crop leaves contributed to the increase in foliar utilization, which further resulted in a 1.6-fold enhancement of bioactivity against target Spodoptera exigua compared to Micro-EB. Especially, Nano-EB did not exacerbate the safety risk to the nontarget organism zebrafish with no significant difference. This study elaborates the size effect on the effectiveness and safety of pesticide formulations and lays a theoretical foundation for the development and rational utilization of efficient and environmentally friendly nanopesticides.

Abstract:Lepidopteran insects constitute one of the largest fractions of animals on earth, but most are considered pests in their relationship with humans. Especially, the midgut is the primary site of digestion and absorption, which is alkaline and contains many alkaline proteases. In current pesticide preparation, zein can be used as an ideal carrier material due to its antioxidation property, biocompatibility, low costs, and adhesion. Moreover, zein is stable in gastric acid but can be soluble in alkaline solution or degraded by alkaline protease in the intestine. Herein, a pH/enzyme dual-responsive Zein-EMB-Nanocapsule (Zein-EMB-NC) system was prepared to enhance the pesticide's adhesion, target wettability stability, anti-photolysis, and stimuli-responsive controlled release property. Scanning electron microscopy, transmission electron microscopy, and dynamic light scattering were applied to investigate their structural characteristics and stability, from which it was found that Zein-EMB-NC formed uniform nanoparticles with an average diameter of 156.9 ± 0.6 nm and had excellent stability in aqueous solution. As confirmed by ultraviolet-visible spectroscopy, the anti-UV property of Zein-EMB-NC revealed that the presence of zein could protect EMB from UV photolysis. The foliar surface adhesion and wettability performance of Zein-EMB-NC are significantly improved through hydrophobic interactions and hydrogen bonding. The toxicity of Zein-EMB-NC to Spodoptera exigua (Hübner) was significantly higher than that of the three commercially available formulations tested. The application of foliar spraying on peanuts against Spodoptera exigua (Hübne) shows that a 60% dose of Zein-EMB-NC retained the control efficacy similar to that of commercialized samples. Overall, this novel Zein-EMB-NC has a broad prospect in crop protection for improving insecticidal activity and protecting the pesticide from loss.

Abstract:BACKGROUNDAs the global population grows, large quantities of agrochemicals are used to secure food demand and there is an urgent need to develop efficient and environmentally friendly pesticides and fertilizers. Compared with traditional pesticides, nanopesticide formulations can achieve better foliar wettability, chemical stability, and water dispersibility without or with the minimal use of organic solvents, in line with the development of sustainable agriculture. RESULTSIn this research, glycidyl methacrylate (GMA) was used as an intermediate and glycine methyl ester (GLY) was used as an organic nitrogen source to modify carboxymethyl cellulose (CMC) to synthesize a pesticide nanocarrier CMC-PGMA-GLY. A nanopesticide EB@CMC-PGMA-GLY (EB@CPG) was formed by the hydrophobic and electrostatic interactions of emamectin benzoate (EB), which had good water dispersion. The good affinity of the composite towards leaves led to the effective moistening of the leaf surface by the pesticide and through the pH control pesticide release. Biological experiments show that nanopesticides can maintain high insecticidal activity and have no toxicity to seed germination. The results of pot experiment showed that the nanocarriers could be used as an organic nitrogen fertilizer to increase the fresh weight of plants by 39.77%. CONCLUSIONThis environmentally friendly pesticide carrier can be used as a fertilizer to provide sufficient nutrients for crops, opening a new method for the development of sustainable agriculture.

Abstract:Pesticides play an important role in agricultural disease and pest control. However, low utilization efficiency and environmentally unfriendly disadvantages of conventional pesticide formulations cause substantial environmental and ecological damage. Constructing intelligent controlled-release pesticide system by nanotechnology is a feasible way to overcome these defects. In this research, an emamectin benzoate-loaded liposome nano-vesicle (EB-Lip-NV) with multicompartment structure and thermo-responsive characteristic was developed to accurately control nocturnal pests and improve insecticidal activity. EB-Lip-NV is an unusual low-temperature rapid-release mode based on phase transitions of liposome membrane. Compared with conventional water soluble granule (SG), the EB-Lip-NV exhibited higher control activity on Spodoptera exigua. More importantly, the control efficacy of Spodoptera exigua at 20°C was around 1.4 times than that at 40°C because of low temperature-induced rapid release. This controlled-release behavior of EB-Lip-NV responding to temperature change could effectively control population of nocturnal pests. In addition, the toxicity of the EB-Lip-NV towards zebrafish was lower than that of SG by above 50%. This study provides a new strategy to construct intelligent controlled-release pesticide systems with improving utilization rate and reducing harm to environment and non-target organisms.

Abstract:Controlled release formulations (CRFs) are considered an effective way to solve the low bioavailability of traditional pesticides. However, CRFs prepared by coating or encapsulation has the disadvantage of explosive release of the ingredients. Sustained-release pesticides prepared by coupling with a carrier can overcome this shortcoming. In the present study, an emamectin-lignin sulfonic acid conjugate (EB-SL), in which emamectin was connected via sulfonamide bonds with lignin, was prepared using sodium lignosulfonate as the carrier. The structure of the conjugate was characterized by IR, 1HNMR, and elemental analysis. The sustained-release results showed that EB-SL maintained its original structure when released in pure water and soil columns, and the sulfamide bond did not break. The photolysis test displayed that the photolysis half-life T 0.5 of EB-SL was increased by 1.5 times compared with the emamectin suspending concentrate (EB-SC). Bioactivity tests in the greenhouse showed that EB-SL not only had similar insecticidal toxicity to emamectin emulsion concentrate (EB-EC) against Ostrinia nubilalis but also displayed a longer duration. The lethality of EB-SL on O. nubilalis was maintained at more than 70% across 19 days, whereas EB-EC as the control was less than 50% after 11 days of application.

Abstract:<p>Most traditional pesticide formulations contain environmentally unfriendly components and suffer from low utilization efficiency due to the loss, decomposition and burst release of active ingredients. Encapsulating pesticides using nanocarriers is a feasible approach to overcome these defects. In this work, the emamectin benzoate-loaded zein nanoparticles were fabricated by an antisolvent precipitation method combined with orthogonal experimental design. The mean size and zeta potential of the nanoparticles were 168 nm and 11.8 mV, respectively. The nano-delivery system could continuously release active ingredients, and exhibited the improved retention and bioavailability compared to traditional water dispersible granules. The one-step coprecipitation technique is simple, scalable and almost applicable to encapsulate all poorly soluble pesticides that have a certain solubility in ethanol solutions. The effective, sustained release and green nano-delivery system could significantly reduce the dosage and environmental pollution of pesticide, thereby improving ecological and economic effects.</p>

Abstract:Pesticide slow-release formulations provide a way to increase the efficiency of active components by reducing the amount of pesticide that needs to be applied. Slow-release formulations also increase the stability and prolong the control effect of photosensitive pesticides. Surfactants are an indispensable part of pesticide formulations, and the choice of surfactant can strongly affect formulation performance. In this study, emamectin-benzoate (EMB) slow-release microspheres were prepared by the microemulsion polymerization method. We explored the effect of different surfactants on the particle size and dispersity of EMB in slow-release microspheres. The results indicated that the samples had uniform spherical shapes with an average diameter of 320.5 ±5.24 nm and good dispersity in the optimal formulation with the polymeric stabilizer polyvinyl alcohol (PVA) and composite non-ionic surfactant polyoxyethylene castor oil (EL-40). The optimal EMB pesticide slow-release microspheres had excellent anti-photolysis performance, stability, controlled release properties, and good leaf distribution. These results demonstrated that EMB slow-release microspheres are an attractive candidate for improving pesticide efficacy and prolonging the control effect of EMB in the environment.

Abstract:Nanotechnology has provided a novel approach for the preparation of a safe and highly effective pesticide formulation. Thiazole-Zn, a widely used bactericide, was successfully prepared at nanoscale by an innovative approach of final synthesis process control. Its plausible formation mechanism based on restricted particle aggregation in a nanoreactor was elucidated. Then in order to assess the application performance of thiazole-Zn nanoparticle, the nanoformulation (NPF) was conveniently formulated. Interestingly, the physicochemical properties of NPF showed better than that of the commercial pesticide formulation (CPF) in dispersibility, wettability, spreadability, and stability. At the same time, the in vitro bioassay showed that the minimum inhibitory concentrations (MICs) of NPF against Xanthomonas oryzae pv Oryzae (XOO), Xanthomonas oryzae pv Oryzicola (XOC), Erwinia carotovora subsp. Carotovora (Jones) Holland (ECC), and Erwinia chrysanthemi pv Zeae (ECZ) were 46.88, 93.75, 93.75, and 375.00 mg/L, respectively, whereas those of CPF were 93.75, 375.00, 375.00, and 875.00 mg/L, respectively. Therefore, NPF exhibited stronger antibacterial activity against the above-mentioned pathogens. Moreover, NPF was more effective to bacterial blight of rice than CPF in field trial. As a conclusion, nanotechnology for pesticides by synthesis process control will have a potential in improving the utilization efficiency and relieving the corresponding environmental pollution.

Scores ranged from 0 to 14 (greater risk of developing RA).
