Abstract:Introduction: Alzheimer's disease (AD) causes progressive loss of cognitive function and synaptic plasticity, which is the most common form of dementia. The present study was designed to scrutinize the effects of cacao on passive avoidance memory function and to identify the roles of hippocampal synaptic plasticity and oxidative stress in an AD rat model induced by unilateral intracerebroventricular (UICV) injection of amyloid-beta (Aβ). Methods: Oral administration of cacao (500 mg/kg/ day) was given for 2 consecutive months. A memory retention test was conducted 24 h after passive avoidance training was completed. Subsequently, the amplitude of population spike (PS) and slope of field excitatory postsynaptic potentials (fEPSPs) were assessed at hippocampal long-term potentiation (LTP) in perforant pathway–dentate gyrus (PP-DG) synapses. Moreover, total thiol group (TTG) and malondialdehyde (MDA) concentrations were evaluated in the plasma. Furthermore, compact Aβ plaques were detected in the hippocampal DG by performing Congo red staining. Results: As a result of AD induction, passive avoidance memory was impaired; also, reduced fEPSP slopes, PS amplitudes, and content of TTG, and increase in MDA levels in the rats were observed. In contrast, cacao treatment ameliorated passive avoidance memory impairment, improved hippocampal LTP impairment, modulated oxidative–antioxidative status, and delayed Aβ plaques production in AD rats. Disscussion: Conclusively, cacao alleviates Aβ-induced cognitive deficit, probably by the amelioration of hippocampal LTP impairment, modulation of oxidative–antioxidative status, and inhibition of Aβ plaque accumulation

Abstract:Alzheimer's disease (AD) is the most common form of dementia and has no cure. Therapeutic strategies focusing on the reduction of oxidative stress, modulation of amyloid-beta (Aβ) toxicity and inhibition of tau protein hyperphosphorylation are warranted to avoid the development and progression of AD. The aim of this study was to screen the crude extracts (CEs) and ethyl-acetate fractions (EAFs) of Guazuma ulmifolia, Limonium brasiliense, Paullinia cupana, Poincianella pluviosa, Stryphnodendron adstringens and Trichilia catigua using preliminary in vitro bioassays (acetylcholinesterase inhibition, antioxidant activity and total polyphenol content) to select extracts/fractions and assess their protective effects against Aβ25-35 toxicity in SH-SY5Y cells. The effect of the EAF of S. adstringens on mitochondrial membrane potential, lipid peroxidation, superoxide production and mRNA expression of 10 genes related to AD was also evaluated and the electropherogram fingerprints of EAFs were established by capillary electrophoresis. Chemometric tools were used to correlate the in vitro activities of the samples with their potential to be evaluated against AD and to divide extracts/fractions into four clusters. Pretreatment with the EAFs grouped in cluster 1 (S. adstringens, P. pluviosa and L. brasiliense) protected SH-SY5Y cells from Aβ25-35-induced toxicity. The EAF of S. adstringens at 15.62 μg/mL was able completely to inhibit the mitochondrial depolarization (69%), superoxide production (49%) and Aβ25-35-induced lipid peroxidation (35%). With respect to mRNA expression, the EAF of S. adstringens also prevented the MAPT mRNA overexpression (expression ratio of 2.387x) induced by Aβ25-35, which may be related to tau protein hyperphosphorylation. This is the first time that the neuroprotective effects of these fractions have been demonstrated and that the electropherogram fingerprints for the EAFs of G. ulmifolia, L. brasiliense, P. cupana, P. pluviosa and S. adstringens have been established. The study expands knowledge of the in vitro protective effects and quality control of the evaluated fractions.

Determination of total 3 alpha-hydroxy bile acids in serum.

Abstract:The usual techniques for determination to total 3 alpha-hydroxy bile acids in serum involving liquid-solid extraction of the bile acids with the adsorbent XAD-2 and fluorimetric measurement of NADH generated from the reaction with a NAD-linked 3 alpha-hydroxysteroid dehydrogenase are evaluated and improved. The influence of different types of enzyme preparations on the results is examined. The results with the improved technique are compared to the results obtained with another method, avoiding extraction of the bile acids before the enzymatic reaction which is followed by fluorimetric measurement of resorufin, produced by transfer of the hydrogen of the generated NADH by diaphorase to resazurin. No significant difference between the results with the two types of methods was found. The concentration of total 3 alpha-hydroxy bile acids in serum of 46 fasting 'healthy' individuals aged 17 to 82 years is estimated. 30 were females, of whom 10 were taking estrogen-containing oral contraceptives, and 16 were males. Mean +/- standard deviation in all the females was 3.0 +/- 1.1 micromol/l, and in the males 4.0 +/- 1.9 micromol/l. There was no significant difference between any of the groups.
Effect and Mechanism of Auricularia Auriculajudae Polysaccharide in Hippocampus of Streptozotocin‐induced Diabetic Cognitive Impairment Rats

Abstract:AimTo investigate the effect and mechanism of auricularia auriculajudae polysaccharide(APP) in the hippocampus of streptozotocin‐induced diabetic cognitive impairment rats.MethodsAPP group was treated with intragastric administration of APP (150mg/day) for 4 weeks after intraperitoneal streptozotocin(STZ) injection (60 mg/kg). Their capacity for learning and memory were detected by the Morris water maze. Microstructural and ultrastructural changes were also analysed by transmission electron microscope (TEM). Thrombospondin‐1 (TSP‐1) expression in the CA1 and dentate gyrus (DG) hippocampal area was examined by immunohistochemistry and RT‐PCR.ResultsFour weeks after STZ induction, the diabetic rats showed decreased capacity for learning and memory. However, the cognitive impairment was significantly less severe in APP group as indicated by decrease in the time spent in the target quadrant (P < 0.05) and the number of crossings of the previous location of the platform (P < 0.05). TEM also revealed that APP group had less swollen of mitochondria, decreased heterochromatin accumulation and increased synaptic contacts when compared with the diabetic group. Morever, TSP‐1 expressions in the CA1 and DG were significantly lower in APP group than in diabetic group.ConclusionAPP could alleviate the impairment of memory and cognition in diabetic rat, which may be mediated by decreasing the expression of TSP‐1 in the hippocampus.
Supplementation with γ-glutamylcysteine (γ-GC) lessens oxidative stress, brain inflammation and amyloid pathology and improves spatial memory in a murine model of AD

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Introduction</h3><p>The accumulation of oxidative stress, neuroinflammation and abnormal aggregation of amyloid β-peptide (Aβ) have been shown to induce synaptic dysfunction and memory deficits in Alzheimer's disease (AD). Cellular depletion of the major endogenous antioxidant Glutathione (GSH) has been linked to cognitive decline and the development of AD pathology. Supplementation with γ-glutamylcysteine (γ-GC), the immediate precursor and the limiting substrate for GSH biosynthesis, can transiently augment cellular GSH levels by bypassing the regulation of GSH homeostasis.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Methods</h3><p>In the present study, we investigated the effect of dietary supplementation of γ-GC on oxidative stress and Aβ pathology in the brains of APP/PS1 mice. The APP/PS1 mice were fed γ-GC from 3 months of age with biomarkers of apoptosis and cell death, oxidative stress, neuroinflammation and Aβ load being assessed at 6 months of age.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>Our data showed that supplementation with γ-GC lowered the levels of brain lipid peroxidation, protein carbonyls and apoptosis, increased both total GSH and the glutathione/glutathione disulphide (GSH/GSSG) ratio and replenished ATP and the activities of the antioxidant enzymes (superoxide dismutase (SOD), catalase, glutamine synthetase and glutathione peroxidase (GPX)), the latter being a key regulator of ferroptosis. Brain Aβ load was lower and acetylcholinesterase (AChE) activity was markedly improved compared to APP/PS1 mice fed a standard chow diet. Alteration in brain cytokine levels and matrix metalloproteinase enzymes MMP-2 and MMP-9 suggested that γ-GC may lower inflammation and enhance Aβ plaque clearance <em>in vivo</em>. Spatial memory was also improved by γ-GC as determined using the Morris water maze.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusion</h3><p>Our data collectively suggested that supplementation with γ-GC may represent a novel strategy for the treatment and/or prevention of cognitive impairment and neurodegeneration.</p>

Nanoencapsulated Curcumin: Enhanced Efficacy in Reversing Memory Loss in An Alzheimer Disease Model

Abstract:Investigating new drugs or formulations that target Alzheimer disease (AD) is critical for advancing therapeutic interventions. Therefore, this study aimed to assess the effectiveness of nanoencapsulated curcumin (NC Curc) in alleviating memory impairment, oxidative stress, and neuroinflammation in a validated AD model. Male Wistar rats were given bilateral intracerebroventricular injections of either saline or streptozotocin (STZ) (3 mg/3 μL/site) to establish the AD model (day 0). On day 22, daily oral administrations of curcumin (6 mg/kg), NC Curc (6 mg/kg), or a vehicle (unloaded NC) were initiated and continued for 14 days. NC Curc significantly reversed memory deficits in object recognition and inhibitory avoidance tests induced by STZ. Both formulations of curcumin attenuated elevated acetylcholinesterase activity caused by STZ. Importantly, NC Curc alone effectively mitigated STZ-induced oxidative stress. Additionally, NC Curc treatment normalized GFAP levels, suggesting a potential reduction in neuroinflammation in STZ-treated rats. Our findings indicate that NC Curc improves memory in an AD rat model, highlighting its enhanced therapeutic effects compared to unencapsulated curcumin. This research significantly contributes to understanding the therapeutic and neurorestorative potential of NC Curc in AD, particularly in reversing pathophysiological changes.

Block Success Rate: A Question of Target and Definition

Abstract:To the Editor: We read with interest the article by Pakala et al describing an institutionspecific retrospective review of persistent phrenic nerve paresis (PPNP) after interscalene nerve block (ISB). The authors provide a convincing rationale for thorough evaluation of ISB candidates with coexistent cervical spine disease. As Pakala et al note, PPNP is rare,with an institution-specific incidence of 0.048% during a 9-year span, with more than 18,000 ISBs performed during that time. We offer the following comments to facilitate clinicians’ understanding of the etiology of this rare complication and possibly diminish its occurrence in the future. The authors note that, although the exact mechanism of phrenic nerve injury is uncertain, needle trauma or intraneuronal injection in the setting of cervicomechanically compromised nerves may lend credence to the “double crush” hypothesis of injury. In their table of PPNP cases (6 by paresthesia and 3 by nerve stimulator techniques), it is noteworthy that all 9 patients received volumes of local anesthetic that are considerably higher (range, 45–70mL) than volumes being used in the current ultrasound era. It would seem that the compressive force of this exceedingly large volume of local anesthetic within the tight confines of the fascial planes of the anterolateral neck applied a “triple crush” to the phrenic nerve, resulting in PPNP. Ultrasound guidance has delivered the ability to perform perineural blockade in a reliable, safe, and expeditious manner. We no longer require large volumes of anesthetic to ensure block onset or duration, and we are beginning to minimize side effects previously thought to be unavoidable, such as transient ipsilateral diaphragmatic paralysis. It will be interesting to follow the phenomenon of PPNP during the next several years to determine if the combination of ultrasound guidance and lower volumes of local anesthetic hastens the disappearance of this rare event entirely.

Abstract:To the Editor: Hereditary hemorrhagic telangiectasia (HHT) is an inherited disorder characterized by vascular ectasias in different organs, including gastrointestinal (GI) tract telangiectases. Officially classified as a rare disorder, the incidence in the United States is estimated to be 10 to 20 per 100,000 people with over 50,000 affected Americans. GI bleeding affects 25% to 30% of patients with HHT, usually over the age of 40 years. Of these approximately 25% will have significant GI bleeding requiring multiple blood transfusions often weekly for many years. There have been several cases of HHT patients requiring over 100U of blood during their lifetime from GI blood loss. There is very little data to guide physicians in treating this very problematic group of patients. A gastroenterologist’s initial reaction when GI telangiectases are encountered in a patient with anemia and GI bleeding is to attempt to cauterize them. This can be particularly problematic in HHT patients where the telangiectases are multiple and extend distally beyond the reach of a standard endoscope. Our group published the largest case series to date in 2003, with 40 patients with HHT and GI bleeding who were evaluated for response to medical therapy. Some patients responded extraordinarily well to hormonal therapy whereas others did not. The literature on estrogen therapy in HHT patients with bleeding GI lesions is very sparse with a few case reports and a single randomized trial studying the long-term effect of hormonal therapy on bleeding GI vascular malformations. In a study published in 1993, 7 of 8 patients with HHT had no evidence of bleeding while taking ethinylestradiol 0.05mg and norethisterone 1mg (Ovcon 50) for 6 months versus placebo and were, therefore, considered treatment successes. Because there is insufficient evidence to recommend systemic therapy or local endoscopic therapy for patients with HHT and GI bleeding, our group undertook a pilot study to evaluate ethinylestradiol 0.05mg and norethisterone 1mg (Ovcon 50) versus local endoscopic argon plasma coagulation therapy in patients with HHT and GI bleeding. We report the first patient who entered the trial on the hormonal arm and completed 12 months of therapy. This study was approved by the Yale University School of Medicine Human Investigation Committee. The patient is a 51-year-old woman with HHT and transfusiondependent GI bleeding. During the 5 years before study entry, she had required more than 150U of packed red blood cell transfusions and the year before study entry, she had required 79U of packed red blood cell transfusions. She was hepatitis C virus-positive and had been treated with interferon 10 years ago. She had 2 pulmonary arteriovenous malformations that had been closed just before study entry. She was receiving intravenous iron 3 times per week with her hematocrit ranging from 17% to 31%. An enteroscopy revealed 8 telangiectases in her stomach; 53 in her duodenum; and 79 in the first 60 cm of her jejunum. A colonoscopy revealed 10 telangiectases scattered throughout her colon. She was randomized to receive daily oral ethinylestradiol 0.050mg and norethindrone 1.0mg (Ovcon 50) for 1 year. Over the next 4 months, her transfusion requirements slowly and steadily markedly decreased so that she required only 10U over this time. Her last transfusion was 4 months after starting combination estrogen and progesterone therapy and since then she has maintained a hematocrit over 35%. She has now completed 14 months of hormonal therapy and has had no adverse effects. She has remained on intravenous iron, but decreased her dose to twice weekly. This case report illustrates that patients with HHT and severe anemia owing to GI telangiectases should be considered for combination estrogen and progesterone therapy as second line therapy if they are unable to maintain an adequate hematocrit level on oral and/or intravenous iron therapy.
Supracostal approach--an excellent exposure for renal and adrenal surgery.

Abstract:The supracostal extraperitoneal and extrapleural approach provides wide exposure during renal and adrenal operations without damaging the neurovascular bundles and spares the partial resection of rib. We performed renal and adrenal operations on 33 patients by this approach. The procedures were accomplished smoothly obtaining a broad surgical field without costal resection. Complications were rare and postoperative pain seemed to be mild. Hence, the supracostal approach is considered as a better method with excellent exposure to the kidney and adrenal and is anatomically comprehensive.
Cuscutae Japonicae Semen Ameliorates Memory Dysfunction by Rescuing Synaptic Damage in Alzheimer’s Disease Models

Abstract:Alzheimer’s disease (AD) is the most common type of dementia in the elderly. It is characterized by the accumulation of amyloid-beta (Aβ) and progressive cognitive impairment. To alleviate the symptoms of AD, functional foods and nutrients have been used for centuries. In this study, we investigated whether Cuscutae Japonicae Semen (CJS), a medicinal food traditionally used in East Asia, has effects on memory improvement and synapse protection in AD. We orally administered CJS to 5x familiar AD (5xFAD) transgenic mice and performed the Morris water maze test. The results showed that CJS treatment ameliorated the decline of memory function. Then, we demonstrated that CJS attenuated the degeneration of pre- and post-synaptic proteins in the hippocampi of 5xFAD mice. To demonstrate the effects of CJS in vitro, we treated Aβ in primary neuronal culture with CJS and observed that CJS rescued the loss of functional synapses. The protective effects of CJS on the synapse were due to the inhibition of activated caspase-3 expression. Additionally, CJS inhibited the phosphorylation of glycogen synthase kinase-3β and tau proteins, which contribute to synaptic dysfunction. Taken together, our results suggest that CJS is efficient in alleviating memory loss by rescuing caspase-3-mediated synaptic damage in AD treatment.

Neuroprotective Effect of Chlorogenic Acid in an Animal Model of Sporadic Alzheimer's Disease Induced by Streptozotocin

Abstract:Alzheimer's Disease is a degenerative neurological condition which leads to a decline in memory and cognitive function. Chlorogenic Acid (CGA) presents properties including neuroprotective, antioxidant and anti-inflammatory. The aim of this study was to examine the impact of CGA on cognitive impairments, neuroinflammation and neuronal damage in mice submitted to an experimental model of Sporadic Alzheimer Disease (SAD) induced by intracerebroventricular administration of streptozotocin (ICV-STZ). Male Swiss mice received bilateral ICV-STZ injections (3 mg/Kg) on days 1 and 3. The treatment with CGA (5 mg/Kg, orally) or vehicle (water, orally), was initiated and continued for 26 days, starting 2 h after the second induction procedure. At first, there was no change in serum glucose levels after SAD induction. ICV-STZ induces impairments in aversive, recognition, and spatial memory, while CGA treatment significantly alleviated these memory deficits. Furthermore, locomotor activity, working memory, and anxiety-related activities remained unaffected by the treatments. CGA treatment protects against ICV-STZ-induced increase in the nitrite/nitrate and TBARS levels. ICV-STZ induced a reduction in viable cells, depletion of BDNF, and triggered astrogliosis and microgliosis in the cortex and hippocampus. Treatment with CGA preserves viable cell count in the prefrontal cortex, CA1, and CA3 regions of the hippocampus. Additionally, it prevented BDNF depletion in the prefrontal cortex and hippocampus (CA1, CA3, and DG regions), and mitigated astrogliosis and microgliosis in the prefrontal cortex and hippocampus (CA1, CA3, and DG regions). These findings indicate the neuroprotective effects of CGA, underscoring their potential as therapeutic agents or adjuncts in the treatment of SAD.

Abstract:Natural products offer promising potential for the development of new therapies for Alzheimer's disease (AD). Blackberry fruits are rich in phytochemical compounds capable of modulating pathways involved in neuroprotection. Additionally, drug repurposing and repositioning could also accelerate the development of news treatments for AD. In light of the reduced brain glucose metabolism in AD, an alternative approach has been the use of the drug metformin. Thus, the aim of this study was to evaluate the effect of treatment with blackberry extract in a model of AD induced by streptozotocin (STZ) and compare it with metformin treatment. Male rats were divided into groups: I - Control; II - STZ; III - STZ + blackberry extract (100 mg/kg); IV - STZ + blackberry extract (200 mg/kg) and V - STZ + metformin (150 mg/kg). The animals received intracerebroventricular injection of STZ or buffer. Seven days after the surgical procedure, the animals were treated orally with blackberry extract or metformin for 21 days. Blackberry extract and metformin prevented the memory impairment induced by STZ. In animals of group II, an increase in acetylcholinesterase activity, phosphorylated tau protein, IL-6, oxidative damage, and gene expression of GSK-3β and Nrf2 was observed in the hippocampus. STZ induced a decrease in IL-10 levels and down-regulated the gene expression of Akt1, IRS-1 and FOXO3a. Blackberry extract and metformin prevented the alterations in acetylcholinesterase activity, IL-6, GSK3β, Nrf2, and oxidative damage. In conclusion, blackberry extract exhibits multi-target actions in a model of AD, suggesting new therapeutic potentials for this neurodegenerative disease.

Abstract:Alzheimer’s Disease (AD) is characterized by progressive cognitive decline and synaptic dysfunction, often associated with amyloid-beta accumulation and microglial alterations. Here, we investigate the role of the Sodium-dependent Vitamin C Transporter 2 (SVCT2) in microglia to modulate AD-like pathology in mice. Using a combination of RNA sequencing, advanced quantitative proteomics, electrophysiology, behavioral tests, high-throughput imaging, and microglial viral gene delivery, we explore the interplay between SVCT2 expression in microglia, amyloid-beta load, synaptic proteome changes, and synaptic plasticity. Our results demonstrate that SVCT2 expression in microglia decreases with age in the 5xFAD mice, correlating with memory deficits and alterations in synaptic mitochondrial proteome. Importantly, overexpression of SVCT2 in microglia leads to enhanced clearance of amyloid plaques and reconfiguration of the mitochondrial proteome landscape in the synapses, improving synaptic long-term plasticity (LTP) and memory performance. Our findings underscore the SVCT2 overexpression in microglia as a potent strategy to simultaneously decrease amyloid pathology and enhance synaptic plasticity and memory performance, offering new avenues for therapeutic interventions in AD.

Abstract:ABSTRACT Conclusion The study results indicate that children with a history of otitis media with effusion (OME) suffer from auditory processing disorder to some degree. The findings support the hypothesis that fluctuating hearing loss may affect central auditory processing during critical periods. Objectives Evidence suggests that prolonged OME in children can result in an auditory processing disorder, presumably because hearing has been disrupted during an important developmental period. A lack of auditory stimulation leads to the abnormal development of the hearing pathways in the brain. The aim of the present study was to determine the effects of OME on binaural auditory function and auditory temporal processing. Method In the present study, the dichotic digit test (DDT) was used for binaural hearing, and the gap in noise (GIN) test was used to evaluate temporal hearing processing. Results The average values of GIN differed significantly between children with a history of OME and normal controls (p < 0.001). The mean values of the DDT score were significantly different between the two groups (p = 0.002).

Abstract:Abstract Alzheimer's disease (AD) is a chronic and progressive neurodegenerative condition that is influenced by multiple factors along with neuroinflammation and oxidative stress. Our previous study proved that Lithocarpus polystachyus Rehd. aqueous extract (sweet tea aqueous extract, STAE) effectively inhibits hydrogen peroxide‐induced neuronal cell injury. However, it is not clear whether STAE can protect against AD, and its underlying mechanisms are still uncertain. Therefore, the present study was designed to evaluate the possible behavioral and neurochemical effects of STAE on A β 25‐35 ‐induced AD rats administered STAE (20, 40, 80 mg/mL) for 14 days. We showed that STAE administration significantly and dose‐dependently ameliorated the cognitive deficits in the AD rat models, assessed in the Morris water maze (MWM) test, Y‐maze test, and novel object recognition (NOR) test. The results of hematoxylin and eosin (H&E) staining and Nissl staining showed that after treatment with STAE, the pathological damage to the hippocampal CA1, CA3, and dentate gyrus (DG) neurons of rats was significantly improved. Furthermore, STAE dose‐dependently inhibited microglia and astrocyte activation in the hippocampus of rats accompanied by increased protein expression of silent mating‐type information regulation 2 homolog 6 (SIRT6) and decreased protein expression of nod‐like receptor thermal protein domain‐associated protein 3 (NLRP3) and its downstream pyroptosis‐related genes after following A β 25‐35 . In summary, our findings indicate that STAE effectively inhibits A β 25‐35 ‐induced learning and memory impairment in rats, and the mechanism is, at least partially, related to the regulation of SIRT6/NLRP3 signaling pathway.
Protective Effects of 6-(Methylsulfinyl)hexyl Isothiocyanate on Aβ1-42-Induced Cognitive Deficit, Oxidative Stress, Inflammation, and Apoptosis in Mice

Abstract:Alzheimer’s disease (AD) is the most common form of dementia among older people. Although soluble amyloid species are recognized triggers of the disease, no therapeutic approach is able to stop it. 6-(Methylsulfinyl)hexyl isothiocyanate (6-MSITC) is a major bioactive compound in Wasabia japonica, which is a typical Japanese pungent spice. Recently, in vivo and in vitro studies demonstrated that 6-MSITC has several biological properties. The aim of the present study was to investigate the neuroprotective activity of 6-MSITC in a murine AD model, induced by intracerebroventricular injection of β-amyloid oligomers (Aβ1-42O). The treatment with 6-MSITC started 1 h after the surgery for the next 10 days. Behavioral analysis showed that 6-MSITC ameliorated Aβ1-42O-induced memory impairments. The decrease of glutathione levels and increase of reactive oxygen species in hippocampal tissues following Aβ1-42O injection were reduced by 6-MSITC. Moreover, activation of caspases, increase of inflammatory factors, and phosphorylation of ERK and GSK3 were inhibited by 6-MSITC. These results highlighted an interesting neuroprotective activity of 6-MSITC, which was able to restore a physiological oxidative status, interfere positively with Nrf2-pathway, decrease apoptosis and neuroinflammation and contribute to behavioral recovery. Taken together, these findings demonstrated that 6-MSITC could be a promising complement for AD therapy.

Abstract:The accumulation of amyloid-beta (Aβ) peptides is a crucial factor in the neuronal degeneration of Alzheimer's disease (AD). The current study investigated the underlying neuroprotective mechanisms of shrimp shell extract (SSE) and liposome-encapsulated SSE (SSE/L) against Aβ 1-42 -induced neuronal damage and death in rats. Intracerebroventricular infusion of Aβ 1-42 effectively induced memory decline, as observed in a reduction of the rat's discriminating ability in the novel object recognition and novel object location tasks. Oral pretreatment with 100 mg/kg of SSE demonstrated no preventive effect on the memory decline induced by Aβ 1-42 infusion. However, treatment with SSE/L 100 mg/kg BW effectively attenuated memory deficits in both behavioral assessments following two and four weeks after Aβ 1-42 infusion. Moreover, SSE/L exerted neuroprotective effects by reducing lipid peroxidation and increasing Nrf2/HO-1 expression. There was a significant decrease in Iba1 and GFAP (biomarkers of microglia and astrocyte activity, respectively), as well as a decrease in the levels of NF-κB expression and the inflammatory cytokines TNF-α and IL-6 in the cortical and hippocampal tissues. Treatment with SSE/L also reduced the pro-apoptotic proteins Bax and cleaved caspase-3 while raising the anti-apoptotic protein Bcl2. In addition, the beneficial effects of SSE/L were along with the effects of a positive control commercial astaxanthin (AST). The findings of this study indicated that SSE/L provided neuroprotective effects on Aβ 1-42 -induced AD rats by ameliorating oxidative stress, neuroinflammation and apoptotic cell death. Therefore, SSE/L might be employed to prevent and mitigate Aβ accumulation-induced neurotoxicity in AD.

Abstract:A mode-matching DeltaSigma closed-loop vibratory-gyroscope readout interface fabricated in a 0.35 mum CMOS process is presented. This paper describes the architecture and circuits used to exploit mode matching effectively to attain order-of-magnitude noise and power dissipation reductions over state-of-the-art implementations without sacrificing other aspects of system performance. As micromachined vibratory gyroscopes push to lower noise floors, mode matching is necessary to moderate interface power dissipation. Our design overcomes these limitations by using force feedback to achieve a stable scale factor, a well defined phase relationship, and a bandwidth well in excess of 50Hz, commensurate with the requirements of automotive and consumer applications.
Supplementation of Medium-Chain Triglycerides Combined with Docosahexaenoic Acid Inhibits Amyloid Beta Protein Deposition by Improving Brain Glucose Metabolism in APP/PS1 Mice

Abstract:The deterioration of brain glucose metabolism predates the clinical onset of Alzheimer's disease (AD). Medium-chain triglycerides (MCTs) and docosahexaenoic acid (DHA) positively improve brain glucose metabolism and decrease the expression of AD-related proteins. However, the effects of the combined intervention are unclear. The present study explored the effects of the supplementation of MCTs combined with DHA in improving brain glucose metabolism and decreasing AD-related protein expression levels in APP/PS1 mice. The mice were assigned into four dietary treatment groups: the control group, MCTs group, DHA group, and MCTs + DHA group. The corresponding diet of the respective groups was fed to mice from the age of 3 to 11 months. The results showed that the supplementation of MCTs combined with DHA could increase serum octanoic acid (C8:0), decanoic acid (C10:0), DHA, and β-hydroxybutyrate (β-HB) levels; improve glucose metabolism; and reduce nerve cell apoptosis in the brain. Moreover, it also aided with decreasing the expression levels of amyloid beta protein (Aβ), amyloid precursor protein (APP), β-site APP cleaving enzyme-1 (BACE1), and presenilin-1 (PS1) in the brain. Furthermore, the supplementation of MCTs + DHA was significantly more beneficial than that of MCTs or DHA alone. In conclusion, the supplementation of MCTs combined with DHA could improve energy metabolism in the brain of APP/PS1 mice, thus decreasing nerve cell apoptosis and inhibiting the expression of Aβ.

Abstract:For Ionospheric clutter is non-stationary and very strong, targets submerged in it is too difficult to extract. The detection performance of High Frequency Surface Wave Radar (HFSWR) is influenced. Ridgelet analysis is a tool for analyzing non-stationary signals and can separate lines with different directions and scales in a 2D image. Therefore, this paper presents a new method which applies ridgelet analysis to time-frequency image of radar signal to mitigate the clutter. The results show that this method can mitigate the clutter effectively, and the target submerged in clutter can be extracted well. When the target submerged in clutter and have low signal-to-clutter ratio, about 15∼20dB improvement can be get after the processing.
