Abstract:Biofilms play a significant role in the existence of bacteria under adverse conditions and the pathogenesis of infections. They contribute to the creation of reservoirs of antibiotic-resistant pathogens. This determines the relevance of the search for biologically active substances that inhibit the formation of biofilms. In recent years, the objects of intensive study are fucoidans — sulfated polysaccharides from marine brown algae, which have a wide polyfunctional spectrum of action. Their influence on the formation of Yersinia pseudotuberculosis biofilm in a dynamic model simulating the natural conditions of ecosystems of fucoidans belonging to different structural groups was investigated in this study. They are synthesized by brown algae: Fucus evanescens , Saccharina cichorioides , and Saccharina japonica . The fucoidan isolated from F.evanescens have showed the most pronounced antibiotic activity. The authors developed a new approach that allows obtaining stable results of multidirectional action of sulfated polysaccharides on the biofilm formation of Y.pseudotuberculosis .
Investigation of antioxidant and anticancer potential of fucoidan (in-vitro & in-silico) from brown seaweed Padina boergesenii

Abstract:The present study was aimed to evaluate the antioxidant, anticancer activity and in-silico analysis of fucoidan extracted from the brown seaweed Padina boergesenii (SF-PB). The overall dry mass of the isolated SF-PB and sulphate residues was found to be 4.9 ± 0.12 % and 17.72 ± 0.25 % respectively. The Fourier Transform Infrared Spectroscopy (FTIR), X-Ray diffraction analysis (XRD), Nuclear Magnetic Resonance (NMR) Spectroscopy, High-performance liquid chromatography (HPLC), and Electrospray Ionisation Mass Spectrometry (ESI-MS) are used to further characterize the fucoidan. The antioxidant assays results showed significant antioxidant activity in a dose-dependent manner. The highest antioxidant activity was observed in the 2,2-diphenyl-1-picrylhydrazyl (DPPH)-76.75 %, Reducing power-66.55 %, 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) -(ABTS)-70.55 %, Hydrogen peroxide (H2O2)-59.9 %, Nitric oxide, and Total antioxidant scavenging (TAC) -68.92 % at 1000 μg/mL with an IC-50 value of 55.62, 59.70, 57.43, 64.40, 61.88 and 60.73 respectively. The cytotoxic effect of SF-PB on human cervical carcinoma cells (HeLa) demonstrated a significant percentage of inhibition 54 % at 40 μg/mL. Fucoidan IC 50 value against cancer cells was determined and found to be 38 μg/mL. Further, the apoptotic staining analysis depicted that fucoidan acts on HeLa cells and induces apoptosis through nuclear fragmentation, inhibition of the cell cycle at the G1 phase, and also with a high level of ROS production. Furthermore, Mesocyclops leuckarti copepod showed no toxicity when treatsed with fucoidans at a concentration of 250 μg/mL (100 % survival), demonstrating that fucoidan are not hazardous and are environmentally benign. In a docking investigation, fucoidan demonstrated the highest binding affinity against apoptotic proteins. Fucoidan may be an alternative for patients with cervical cancer. The findings showed that isolated fucoidan from P. boergesenii has substantial antioxidant and anticancer effects.

Abstract:Dental plaque biofilms cause various dental diseases; therefore, inhibiting the growths of the dental plaque bacteria which produce biofilms can be a strategy for preventing dental disease. Certain sulfated polysaccharides from marine algae exert antimicrobial activities against human bacterial pathogens in addition to their physiological benefits. On the basis of these observations, the antimicrobial and antibiofilm activities of sulfated polysaccharides from different marine algae were evaluated against dental plaque bacteria. Among the sulfated polysaccharides, a fucoidan from Fucus vesiculosus showed notable antimicrobial activities against the selected dental plaque bacteria, including some foodborne pathogenic bacteria. The minimum inhibitory concentrations were of 125 to 1000 μg mL−1. Regarding the antibiofilm activity, the fucoidan at the concentrations of above 250 μg mL−1 completely suppressed the biofilm formations and planktonic cell growths of Streptococcus mutans and S. sobrinus. However, no eliminative effect on the completed biofilm was observed. The fucoidan consisted of almost fucose base polysaccharide containing approximately 14.0% sulfate content. The average molecular weight of the fucoidan was changed by heat treatment (121 °C for 15 min) and it affected the antimicrobial activity.

Abstract:Fucoidans are a heterogenous class of fucose-rich sulfated carbohydrates which have attracted increasing attention in cancer and inflammation research due to their bioactive properties. There are reports that fucoidans may have direct antibacterial effects and synergy with antibiotics. However, the literature is conflicting, potentially due to the limited reporting of origin, characteristics, and extraction methods of the fucoidans tested. Here we report the results of 18 defined fucoidans screened for direct, indirect, and synergistic antibacterial effects. 15 distinct fucoidan fractions, isolated from Laminaria hyperborea using a solvent-free extraction process, were characterised for molecular weight, pH, viscosity, and sulfur content. These, together with three commercially available crude fractions, were assessed at concentrations from 0.03125-24 mg mL -1 for minimum inhibitory concentration against Staphylococcus aureus , Streptococcus mutans and Streptococcus sanguinis. Furthermore, we tested a selection of fucoidans for antibacterial synergy with vancomycin and indirect antibacterial effects in whole blood survival assays. Reverse-transcription quantitative polymerase chain reaction (RT-qPCR) was performed to assess the stress response in fucoidan-treated S. aureus cultures. We have identified one fucoidan fraction with bactericidal activity against diverse bacteria. This effect is dose-, fucoidan fraction- and bacteria-specific, and furthermore, not related to osmotic stress. No synergistic effects were observed with fucoidan in combination vancomycin. Fucoidans have exciting potential as antimicrobial agents. Further analysis is required to establish the precise molecular characteristics responsible for their potent bactericidal activity.

Potential prebiotic and antibacterial activities of fucoidan from Laminaria japonica

Abstract:Fucoidan from Laminaria japonica became sterilized with an autoclave and ultraviolet (UV) radiation. Potential prebiotic and antibacterial activities of sterilized fucoidans (SF) were the subject of investigation. Molecular weight, monosaccharide composition, FTIR, and NMR spectra of SF underwent evaluations to elucidate the relationship between the structure and activities of SF. The growth of Lactobacillus rhamnosus GG and L. acidophilus with autoclave sterilized fucoidan (ASF) and the growth of L. plantarum, L. gasseri, L. paracasei, and L. reuteri with UV sterilized fucoidan (USF) increased significantly. Also, fucoidan was vastly more effective than fructooligosaccharides in improving the growth of L. gasseri, L. reuteri, and L. paracasei. The growth of Escherichia coli and Bacillus cereus decreased at each SF concentration. ASF was more effective against E. coli, B. cereus, and Staphylococcus aureus than the USF efficiency. However, USF exhibited more inhibitory effects on the growth of Enterobacteriaceae compared to the ASF efficiency. When comparing the ASF and USF, autoclave caused a considerable decrease in molecular weight and uronic acid content, increased fucose and galactose, and made no significant changes in NMR spectra. Fucoidan effectively promoted probiotic bacterial growth and reduced pathogenic outbreaks in the medium. Therefore, it can occur as a new algal prebiotic and antibacterial agent.

Abstract:The marine macroalgae produce a collection of bioactive polysaccharides, of which the sulfated heteropolysaccharide fucoidan produced by brown algae of the class Phaeophyceae has received worldwide attention because of its particular biological actions that confer nutritional and health benefits to humans and animals. The biological actions of fucoidan are determined by their structure and chemical composition, which are largely influenced by the geographical location, harvest season, extraction process, etc. This review discusses the structure, chemical composition and physicochemical properties of fucoidan. The biological action of fucoidan and its applications for human health, tissue engineering, regenerative medicine and drug delivery are also addressed. The industrial scenario and prospects of research depicted would give an insight into developing fucoidan as a commercially viable and sustainable bioactive material in the nutritional and pharmacological sectors.
Optimizing the fucoidan extraction using Box-Behnken Design and its potential bioactivity

Abstract:Fucoidan is a sulfated polysaccharide that occurs naturally in the cell wall of brown seaweeds and has substantial biological efficacy. Optimizing the extraction of fucoidan from different brown seaweeds was the primary goal of this research. The optimization of fucoidan extraction was applied on the brown macroalga Turbinaria turbinata using a Box-Behnken Design (BBD) to inspect the impacts of different pH (3, 5, 7), temperature (70, 80, 90 °C) and extraction duration (60, 120, 180 min) on both the yield and sulfate content of fucoidan. The optimized parameters recorded to maximize the fucoidan yield and its sulfate content were a pH of 3.44 and a temperature of 82.26 °C for 60 min. The optimal conditions obtained from BBD were used for fucoidan extraction from T. turbinata, Sargassum cinereum, Padina pavonica, and Dictyota dichotoma. The highest average of fucoidan yield was derived from P. pavonica (40.76 ± 4.04 % DW). FTIR, 1H NMR, and HPLC were used to characterize extracted fucoidan. The extracted fucoidan's Physical characteristics, biochemical composition, antioxidant potential, antitumor effect against breast cancer cells (MCF-7), and antimicrobial and anticoagulant activity were assessed. The extracted fucoidan from D. dichotoma, followed by that extracted from S. cinereum, which had the highest sulphate content, depicted the highest antioxidant, anticancer, and anticoagulant activities. Fucoidan has demonstrated a strong antimicrobial action against some pathogenic microorganisms; Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Klebsiella pneumonia, and Candida albicans. The anticoagulant properties of fucoidan from D. dichotoma were stronger than those of fucoidan from S. cinereum, T. turbinata, and P. pavonica due to its higher sulphate content. These findings could be used for various biomedical applications to improve the pharmaceutical industry.

Abstract:Fucoidans are sulfated polysaccharides that are found in marine algae and have many useful activities, including antitumor effects, promotion of apoptosis of cancer cells, and antiviral, anti-inflammatory, and antiallergic actions. In oral medicine, several case reports have shown that fucoidan-containing creams and tablets markedly improved recurrent aphthous stomatitis, symptomatic inflamed tongue, and recurrent oral herpes labialis. The aim of this study was to examine the properties of fucoidans for use in oral healthcare. The antimicrobial, anti-adhesion, endotoxin-neutralizing, and cyclooxygenase (COX)-1 and COX-2 inhibitory activities of fucoidans were examined. Four key results were obtained: fucoidans showed strong antimicrobial activity against Candida albicans, Streptococcus mutans, and Porphyromonas gingivalis; significantly inhibited the adhesion of S. mutans to bovine teeth and porcelain; were suggested to bind to and neutralize endotoxin (lipopolysaccharide) in an LAL assay; and showed COX-1 and/or COX-2 inhibitory activity. These results suggested that fucoidans may be useful in the field of oral healthcare.

Abstract:Fucoidan, a sulphated polysaccharide from brown seaweed composed of several monosaccharides, has been stated to have several bioactive properties such as antioxidant, antiviral, anticancer, antithrombic, anti-inflammatory, and immunomodulatory effects. This paper provides research findings on green extraction methods, structural analysis of fucoidan, and its associated bioactivities. Fucoidans from brown seaweeds, Fucus vesiculosus and Ascophyllum nodosum, were extracted using green solvents such as citric acid (CA) followed by MWCO (molecular weight cut-off) filtration to obtain high-purity polysaccharides. The presence of functional groups typical to fucoidans, namely, fucose, sulfate, and glycosidic bonds, in the extracts were confirmed through the data obtained from FTIR (Fourier-transform infrared spectroscopy), TGA (thermogravimetric analysis), DSC (differential scanning calorimetry), and solid-state CP–MAS (cross-polarization magic angle spinning) analysis. The MWCO analysis identified that the >300 kDa fraction can have better content of fucoidan (FV-CA 79.16%, FV-HCl 63.59%, AN-CA 79.21%, AN-HCl 80.70%) than the conventional extraction process. Furthermore, the >300 kDa fraction showed significantly higher antioxidant activities compared to crude fucoidan extracts. Crude fucoidan extracts showed significant inhibition of cell viability in human lung (A459 lung carcinoma cells) and colorectal adenocarcinoma (Caco-2) cells at higher concentrations. The fucoidan extracted with green solvents and avoiding alcohol-based precipitation has substantial antioxidant/antitumor action, so, due to this activity, it can be employed as functional foods in food applications.

Abstract:A comparative study of the structural and physicochemical properties of commercial fucoidans produced by Sigma-Aldrich and fucoidans isolated by low-temperature extraction from the brown alga Fucus vesiculosus was performed. The structural features of fucoidans and their behavior in solutions and condensed phases (films) have been studied by various methods of physico-chemical analysis. It was shown that a water-soluble interpolymer complex was formed between fucoidan and alginate. The alginate/fucoidan complex was due to the formation of a cooperative system of hydrogen bonds between the ionogenic and hydroxyl groups of sodium alginate and fucoidan and hydrophobic interactions. The optimal conditions for the complex formation were realized at pH 6. The size of the complex particles was 0.5–1.7 nm. The film materials based on mixtures of fucoidan and sodium alginate were obtained. Using atomic force microscopy, it was shown that the morphology of the resulting alginate-fucoidan films was fine-grained, while the molecular weight of fucoidan did not affect the particle size of materials based on the alginate-fucoidan system. The particle sizes of the alginate/fucoidan interpolymer complex were significantly lower than those of the original polysaccharides. Testing of the microbiological activity of prepared alginate/fucoidan films against three Gram-negative bacteria Escherichia coli , Pseudomonas aeruginosa, and Proteus mirabilis and two Gram-positive bacteria Staphylococcus aureus and Bacillus subtilis showed the pronounced inhibitory effect. The results obtained could be used for the production new materials in pharmaceutical and medical applications.

Abstract:A biocompatible, heterogeneous, fucose-rich, sulfated polysaccharide (fucoidan) is biosynthesized in brown seaweed. In this study, fucoidan was isolated from Padina arborescens (PAC) using celluclast-assisted extraction, purified, and evaluated for its anti-inflammatory potential in lipopolysaccharide (LPS)-induced RAW 264.7 cells. Structural analyses were performed using Fourier transform infrared (FTIR) and scanning electron microscopy. Among the purified fucoidans, fucoidan fraction 5 (F5) exhibited strong inhibitory activity against LPS-induced nitric oxide (NO) production and pro-inflammatory cytokine generation through the regulation of iNOS/COX-2, MAPK, and NF-κB signaling in LPS-induced RAW 264.7 cells. Determination of the structural characteristics indicated that purified F5 exhibited characteristics similar to those of commercial fucoidan. In addition, further analyses suggested that F5 inhibits LPS-induced toxicity, cell death, and NO generation in zebrafish models. Taken together, these findings imply that P. arborescens fucoidans have exceptional anti-inflammatory action, both in vitro and in vivo, and that they may have prospective uses in the functional food sector.

Abstract:Fucoidans are sulfated, complex, fucose-rich polymers found in brown seaweeds. Fucoidans have been shown to have multiple bioactivities, including anti-inflammatory effects, and are known to inhibit inflammatory processes via a number of pathways such as selectin blockade and enzyme inhibition, and have demonstrated inhibition of inflammatory pathologies in vivo. In this current investigation, fucoidan extracts from Undaria pinnatifida, Fucus vesiculosus, Macrocystis pyrifera, Ascophyllum nodosum, and Laminaria japonica were assessed for modulation of pro-inflammatory cytokine production (TNF-α, IL-1β, and IL-6) by human peripheral blood mononuclear cells (PBMCs) and in a human macrophage line (THP-1). Fucoidan extracts exhibited no signs of cytotoxicity in THP-1 cells after incubation of 48 h. Additionally, all fucoidan extracts reduced cytokine production in LPS stimulated PBMCs and human THP-1 cells in a dose-dependent fashion. Notably, the 5–30 kDa subfraction from Macrocystis pyrifera was a highly effective inhibitor at lower concentrations. Fucoidan extracts from all species had significant anti-inflammatory effects, but the lowest molecular weight subfractions had maximal effects at low concentrations. These observations on various fucoidan extracts offer insight into strategies that improve their efficacy against inflammation-related pathology. Further studies should be conducted to elucidate the mechanism of action of these extracts.

Abstract:Marine seaweeds are rich source of polysaccharides present in their cell wall and are cultivated and consumed in China, Japan, Korea, and South Asian countries. Brown seaweeds (Phaeophyta) are rich source of polysaccharides such as Laminarin and Fucoidan. In present study, both the laminarin and fucoidan were isolated was yielded higher in PP ( Padina pavonica ) (4.36%) and STM ( Stoechospermum marginatum ) (2.32%), respectively. The carbohydrate content in laminarin and fucoidan was 86.91% and 87.36%, whereas the sulphate content in fucoidan was 20.68%. Glucose and mannose were the major monosaccharide units in laminarin (PP), however, fucose, galactose, and xylose in fucoidan (STM). FT-IR down peaks represent the carbohydrate of laminarin and fucoidan except, for 1219 cm −1 , and 843 cm −1 , illustrating the sulphate groups of fucoidan. The molecular weight of laminarin was 3–5 kDa, and the same for fucoidan was 2–6 kDa, respectively. Both the Fucoidan and Laminarin showed null cytotoxicity on Vero cells. Contrastingly, the fucoidan possess cytotoxic activity on human liver cancer cells (HepG2) (IC 50 —24.4 ± 1.5 μg/mL). Simultaneously, laminarin also shown cytotoxicity on human colon cancer cells (HT-29) (IC 50 —57 ± 1.2 μg/mL). The AO/EB (Acriding Orange/Ethidium Bromide) assay significantly resulted in apoptosis and necrosis upon laminarin and fucoidan treatments, respectively. The DNA fragmentation results support necrotic cancer cell death. Therefore, laminarin and fucoidan from PP and STM were potential bioactive compounds for anticancer therapy.

Abstract:Fucoidan is a sulphated polysaccharide with diversified biological activities commonly found in brown seaweeds. In our present study, fucoidan was extracted from Sargassum polycystum collected from Saint Martin's Island in three different extraction methods. The composition of the fucoidans was determined by measuring the total carbohydrate, sulfate, uronic acid, fucose, and protein contents. Fourier-transform infrared spectroscopy (FT-IR) and Proton nuclear magnetic resonance ( 1 H NMR) were studied to determine the structural groups of extracted fucoidans. Functional activities were analyzed by antioxidant activity and anti-diabetic activity assay. Fucoidan yield was obtained highest in the acid extraction method (0.061 ± 0.007 g/g), followed by the water extraction method (0.058 ± 0.005 g/g) and salt extraction method (0.041 ± 0.005 g/g). Water extracted fucoidan showed the highest carbohydrate content (62.97 %), uronic acid content (6.46 %), and protein content (2.62 %). Sulphate content (28.56 %) was highest in salt extracted fucoidan, whereas fucose content (24.0 %) was highest in water extracted fucoidan. FT-IR and 1 H NMR analysis confirm several characteristic picks for the extracted fucoidans. Extracted fucoidans presented comparable antioxidant activity, but salt-extracted fucoidan presented the highest DPPH radical scavenging assay (86.85 ± 0.74 %, IC 50 = 0.03 mg/mL), ABTS radical scavenging assay (65.13 ± 0.92 %, IC 50 = 1.94 mg/mL), Phosphomolybdenum assay (4.356 ± 0.11) and Nitric Oxide (NO) radical scavenging assay (57.08 ± 0.52 %, IC 50 = 3.73 mg/mL) in dose-dependency manner. Further anti-diabetic activity assay resulted in the highest α-amylase inhibition activity (62.82 ± 1.36 %, IC 50 = 2.30 mg/mL) and α-glucosidase inhibition activity (55.73 ± 0.84 %, IC 50 = 3.71 mg/mL) by salt-extracted fucoidan. These functional potentials of seaweed may add significant value to the development of functional food and pharmaceutical materials.

Abstract:Purpose: Biofilms, which are created by most microorganisms, are known for their widely developed drug resistance, even more than planktonic forms of microorganisms. The aim of the study was to assess the effectiveness of agents composed of farnesol and nanoparticles (silver, gold, copper, and zinc oxide) in the degradation of biofilms produced by pathogenic microorganisms. Methods: Escherichia coli, Enterococcus faecalis, Staphylococcus aureus, Pseudomonas aeruginosa, and Candida albicans were used to create the biofilm structure. Colloidal suspensions of silver, gold, copper, and zinc oxide (Ag, Au, Cu, ZnO) with the addition of farnesol (F) were used as the treatment factor. The size distribution of those composites was analyzed, their zeta potential was measured, and their structure was visualized by transmission electron microscopy. The viability of the microorganism strains was assessed by an XTT assay, the ability to form biofilms was analyzed by confocal microscopy, and the changes in biofilm structure were evaluated by scanning electron microscopy. The general toxicity toward the HFFF2 cell line was determined by a neutral red assay and a human inflammation antibody array. Results: The link between the two components (farnesol and nanoparticles) caused mutual stability of both components. Planktonic forms of the microorganisms were the most sensitive when exposed to AgF and CuF; however, the biofilm structure of all microorganism strains was the most disrupted (both inhibition of formation and changes within the structure) after AgF treatment. Composites were not toxic toward the HFFF2 cell line, although the expression of several cytokines was higher than in the not-treated group. Conclusion: The in vitro studies demonstrated antibiofilm properties of composites based on farnesol and nanoparticles. The greatest changes in biofilm structure were triggered by AgF, causing an alteration in the biofilm formation process as well as in the biofilm structure.

Abstract:The marine acid polysaccharide fucoidan has attracted attention from both the food and pharmaceutical industries due to its promising therapeutic effects. Fucoidan is a polysaccharide that mainly consists of L-fucose and sulphate groups. Its excellent biological function is attributed to its unique biological structure. Classical activities include antitumor, antioxidant, anticoagulant, antithrombotic, immunoregulatory, antiviral and anti-inflammatory effects. More recently, fucoidan has been shown to alleviate metabolic syndrome, protect the gastrointestinal tract, benefit angiogenesis and bone health. This review focuses on the progress in our understanding of the biological activities of fucoidan, highlighting its benefits for the treatment of human disease. We hope that this review can provide some theoretical basis and inspiration for the product development of fucoidan.

Abstract:Evolutionary changes in microorganisms, in response to the current environment, can contribute generically to adaptations that have harmful effects on human welfare. Antibacterial resistance is one of the results of genetic and evolutionary changes in microorganisms. Many bacteria have developed resistance to antibiotics. Recently, most researchers have been focused on overcoming this problem, relying on nanoparticle-based drug delivery. In this study, our aim is to synthesize silver-doped fucoidan nanoparticle using Tridax procumbens plant leaf aqueous extract and examine their biological efficacy. We synthesized silver-doped fucoidan with the aqueous extraction of Tridax procumbens using the titration method. The synthesized nanoparticles were characterized, including UV-spectroscopy, SEM, FT-IR, EDX, and XRD. Furthermore, we assessed the efficacy of the nanoparticles in terms of antioxidant activity using the DHHP assay, anti-inflammatory activity using the protein degradation assay, and antibacterial activity using well diffusion method. Our result revealed that the synthesized nanoparticles were doped with Ag 2+ in fucoidan using the plant extract. We observed a peak at 390 nm in the UV-spectra analysis, indicating the presence of silver nanoparticles. Further analysis, like SEM, FT-IR, EDX, and XRD showed the nanoparticle characterization. Moreover, these nanoparticles demonstrate good antioxidant and anti-inflammatory activity. Additionally, the nanoparticles exhibited antimicrobial activity against antibiotic-resistance bacteria. Finally, our synthesized silver-doped nanoparticles mediated by an aqueous extract of Tridax procumbens show potential in therapeutic aspects for bacterial infection.

Abstract:Fucoidan, a sulfated polysaccharide known for its anticancer, immunomodulant, antioxidant and antimicrobial activity is abundant in brown seaweeds. Sargassum ilicifolium , a brown alga, was not extensively explored for fucoidan extraction using different methods and assessments of their impact on its structural and therapeutic characteristics. This study aimed to compare the structural features, biological activity, solubility, and emulsification characteristics of fucoidan extracted from Sargassum ilicifolium using conventional and ultrasonic bath extraction. The ultrasonic bath extraction method was 2.5 times higher than the conventional method to extract fucoidan. The structural integrity of fucoidan obtained by both methods was compared using FTIR, 1 HNMR, XRD, FESEM, EDX, and DSC. The monosaccharide composition of fucoidan was determined by TLC. The IC 50 values for antioxidant, anti-inflammatory and antitumor effects on oral cancer cells were found to be significantly lesser for crude fucoidan of S. ilicifolium obtained by ultrasonic bath method than for conventional extraction. Furthermore, fucoidan from the alga has emulsifying and water-holding characteristics which supports its potential use in the food processing industries.
