Abstract:To evaluate the characteristics of chicken interleukin-18 (ChIL-18) in different forms in vitro, the ChIL-18 full-length gene (ChIL-18-F) and the ChIL-18 presumed mature protein gene (ChIL-18-M) were cloned and inserted into the eukaryotic expression vector pCI, to construct recombinant pCI-ChIL-18-F and pCI-ChIL-18-M. The recombinant plasmids were then transferred into chicken splenic lymphocytes (CSLs). Western blot showed that ChIL-18-F, with a molecular weight of 23.0 kDa, was produced in CSLs transfected by pCI-ChIL-18-F; ChIL-18-M, with a molecular weight of 19.5 kDa, was produced in CSLs transfected by pCI-ChIL-18-M. The nitric oxide (NO) level in the transfected CSLs and the culture medium at different time points was further examined under confocal microscopy using 4,5-diaminofluorescein staining. The results showed that both pCI-ChIL-18-F and pCI-ChIL-18-M groups showed significant increase in intracellular and extracellular NO production compared with pCI transfected control cells. These results suggest that both ChIL-18-F and ChIL-18-M could stimulate NO secretion in CSLs. To characterize the intracellular distribution of ChIL-18, ChIL-18-F and ChIL-18-M were each fused to the enhanced green fluorescent protein gene, and expressed in Vero cells. The results showed that the ChIL-18-F tended to the membranous region in Vero cells, while ChIL-18-M did not. This indicates that the N-terminal 27 amino acid peptide helped ChIL-18 target to Vero cell membranes.

Abstract:The distribution of neuronal nitric oxide synthase (nNOS) in the process of normal mouse brain growth from embryonic (E) Day 11 to postnatal (P) Day 1 was investigated by means of immunohistochemical and immunofluorescent methods. Our results demonstrated that nNOS positive neurons appeared early in superficial cortex at E11. At E13, nNOS positive neurons were located in lateral hypothalamus and amygdala, and temporarily in medullar and ventral hypothalamic neuroepithelia. From E15 to P0, nNOS positive neurons were distributed in superior and inferior colliculi, positive staining could also be seen in superior and inferior tectal neuroepithelium at E15. From E17 to birth, the medial geniculate nucleus had a high density of nNOS labeling. The distribution of nNOS gradually increased and extended laterally in embryo brain, which in turn implies that NO might be involved in the development of mouse brain. Anat Rec, 2012. Abstract:The relations between innateness and heritability of behaviour, environmental heterogeneity, genetic diversity of populations, and rates of evolution of behaviour are discussed on the basis of a simple quantitative model. An index I of innateness is proposed which can be related to the heritability index h2. I and h2 are positively related but the degree of correlation depends on the genetic variability of the population, the heterogeneity of the conditioning environment, and the level of spontaneous behavioural variability. The influence of innateness on rates of evolution is moderate. The frequency distributions of I and h2 are unimodal, there is no evidence for an innate-acquired dichotomy.

Abstract:Abstract. The nitric oxide (NO) signaling pathway is a major nonadrenergic-noncholinergic transmitter mechanism in the enteric nervous system. Our aim was to localize the enzymes in question, i.e., neuronal nitric oxide synthase (nNOS), soluble guanylate cyclase (sGC), and cGMP-dependent kinase type I (cGK-I) in rat small intestine by indirect immunofluorescence. nNOS staining was found in neurons of the myenteric plexus and in varicose nerve fibers mainly in the circular muscle layer. The cells positive for neurokinin-1 (NK-1) receptor and c-kit (interstitial cells of Cajal, ICC) in the deep muscular plexus (DMP) did not show nNOS reactivity, but nNOS-positive nerve fibers were directly adjacent to them. sGC was found in flattened cells surrounding myenteric ganglia (periganglionic cells, PGC), in ICC of the DMP, faintly in smooth muscle cells (SMC), and in cells perivascularly scattered throughout the circular muscle layer. cGK-I immunoreactivity was found abundantly in PGC (which presumably are ICC), in ICC of DMP, in SMC of the innermost circular and longitudinal muscle layers, but less intensively in the outer circular layer. Weak cGK-I staining occurred in nerve cells within the myenteric and submucosal plexus. Conclusively the key enzymes of the NO signaling pathway are differentially distributed: Occurrence of nNOS exclusively in neurons and the presence of sGC and cGK-I predominantly in ICC suggest a sequence of neuronal NO release, activation of ICC, and consecutive smooth muscle relaxation. ICC of the DMP seem to be the primary targets for neurally released NO.

Abstract:The present study attempts to review presently known data regarding the distribution of nitric oxide (NO) synthase and the function of NO in invertebrate species. NO is synthesized from L-arginine by the enzyme NO-synthase, and activates guanylate cyclase which in turn leads to an increase in levels of cGMP in target cells. Major contributions to the knowledge of NO as a messenger molecule in invertebrates have been made by NADPH-diaphorase histochemistry and biochemical assays. These techniques suggest the presence of a L-arginine/NO pathway in a variety of tissues, thus implicating multiple roles for NO in invertebrates.

Abstract:Nitric oxide synthase (NOS) produces nitric oxide, a mediator of potential importance in numerous physiologic and inflammatory processes in the lung. We localized constitutive NOS (c-NOS) and inducible NOS (i-NOS) within lung tissue by immunoperoxidase labeling with specific antibodies or by histochemical demonstration of the characteristic NADPH diaphorase activity of NOS. We analyzed human airway (n = 4) or parenchyma (n = 10) specimens obtained from uninvolved areas of surgical tumor resections. We also studied human fetal lung samples (n = 6) and normal or inflamed (16 h after intratracheal LPS instillation) rat lung tissue. Immunostaining with anti-c-NOS identified c-NOS antigen in rat lung nerves, endothelium, and airway epithelium. Normal or inflamed rat macrophages were not stained. Human nerve elements and large-vessel endothelium showed immunostaining with the anti-c-NOS, but no labeling of the airway or alveolar epithelium was seen. Immunostaining with anti-i-NOS showed strong labeling of rat macrophages after LPS treatment, in vivo or in vitro, while normals were negative. Human alveolar macrophages were occasionally positive for i-NOS, especially in areas of chronic inflammation, which also showed focal immunolabeling of endothelium. Uniform labeling of epithelium in large, cartilaginous airways was found with anti-i-NOS in both human bronchi and normal rat trachea samples, suggesting a constitutive role for a NOS that shares epitope(s) with or is highly homologous to the inducible, macrophage type of NOS. Abstract:Inducible nitric oxide synthase (iNOS), which produce large amounts of nitric oxide (NO), is induced in macrophages and microglia in response to inflammatory mediators such as LPS and cytokines. Although iNOS is mainly expressed by microglia that become activated in different pathological and experimental situations, it was recently reported that undifferentiated amoeboid microglia can also express iNOS during normal development. The aim of this study was to investigate the pattern of iNOS expression in microglial cells during normal development and after their activation with LPS by using the quail retina as model. iNOS expression was analyzed by iNOS immunolabeling, western-blot, and RT-PCR. NO production was determined by using DAR-4M AM, a reliable fluorescent indicator of subcellular NO production by iNOS. Embryonic, postnatal, and adult in situ quail retinas were used to analyze the pattern of iNOS expression in microglial cells during normal development. iNOS expression and NO production in LPS-treated microglial cells were investigated by an in vitro approach based on organotypic cultures of E8 retinas, in which microglial cell behavior is similar to that of the in situ retina, as previously demonstrated in our laboratory. We show here that amoeboid microglia in the quail retina express iNOS during normal development. This expression is stronger in microglial cells migrating tangentially in the vitreal part of the retina and is downregulated, albeit maintained, when microglia differentiate and become ramified. LPS treatment of retina explants also induces changes in the morphology of amoeboid microglia compatible with their activation, increasing their lysosomal compartment and upregulating iNOS expression with a concomitant production of NO. Taken together, our findings demonstrate that immature microglial cells express iNOS during normal development, suggesting a certain degree of activation. Furthermore, LPS treatment induces overactivation of amoeboid microglia, resulting in a significant iNOS upregulation.

Abstract:Nitric oxide (NO) is a key signaling molecule in almost all organisms and is active in a variety of physiological and pathological processes. Our understanding of the peculiarities and functions of this simple gas has increased considerably by extending studies to non-mammal vertebrates and invertebrates. In this review, we report the nitric oxide synthase (Nos) genes so far characterized in chordates and provide an extensive, detailed, and comparative analysis of the function of NO in the aquatic chordates tunicates, cephalochordates, teleost fishes, and amphibians. This comprehensive set of data adds new elements to our understanding of Nos evolution, from the single gene commonly found in invertebrates to the three genes present in vertebrates.

Abstract:Nitric oxide (NO) produced by vascular endothelial cells is an important determinant of the basal tone of small arteries and arterioles. Impaired endothelial NO production has been implicated in the pathophysiology of pulmonary hypertension in humans. Available data suggest that reduction of endothelial NO synthesis, with evidence of reduced endothelial NO synthase expression in pulmonary arterioles, is associated with increased pulmonary vasomotor tone and vascular remodelling in hypertensive broilers. Supplemental L-arginine, a precursor of NO, has been shown to induce flow-dependent pulmonary vasodilation, to prevent reduced endothelial NO synthase expression and to inhibit vascular remodelling in broilers with pulmonary hypertension. Nevertheless, its effect on pulmonary hypertension syndrome incidence is limited. It appears that impaired production of NO is a secondary rather than a causative factor in the pathogenesis of pulmonary hypertension in broilers.
Distribution of NOS in the Cochlea of Guinea Pig

Abstract:Objective　The purpose of this investigation was to identify the distribution of nitric oxide synthases(NOS) in the cochlea of the guinea pig.Methods　The animals were fixed by cardiac perfusion with 4%paraformaldehyde in 0.1M phosphate buffer solution. After decalcification with 3% EDTA solution. Cryostat(10 μm) sections were prepared. Sections were incubated with NADPH incubation solution(β-NADPH,Tetranitro blue tetrazolium,Triton X-100) for 60 mins at 37 ℃.Results　The staining for NADPH-diaphorase in frozen sections of decalcified inner ear showed a strong reactivity in the inner and outer hair cells, inner and outer pillar cells, supporting cells, stria vascularis, spiral ganglion cells and glomeruli. The intense staining was detected in the inner hair cell and outer hair cells-cochlear nerve synapse and the glomeruli.Conclusion　NO is involved in neurotransmission according to the intense staining in the hair cells-cochlear nerve synapse. The intense reactivity of endothelial cells of glomeruli indicates the vasodilation action of NO.

Abstract:Recent studies have revealed a non-homogeneous distribution of nitric oxide synthase (NOS) in neurons. However, it is not yet clear whether the intracellular distribution of NOS represents the intracellular nitric oxide (NO) distribution. In the present study, software developed in our laboratory was applied to the reconstructed image obtained from confocal slice images in order to project the 3-D reconstructed images in any direction and to cut the neuron in different sections. This enabled the spatial distribution of NO to be visualized in any direction and section. In single neurons, NO distribution was seen to be heterogeneous. After stimulation with glutamate, the spatial changes in different areas of the neuron were different. These findings are consistent with immunocytochemical data on the intracellular localization of nNOS in hippocampus neurons, and will help to elucidate the specificity of nitric oxide signaling. Finally, the administration of SNAP and l-NAME was used to examine DAF-2 distribution in the neurons. The results showed this distribution to be homogenous; therefore, it did not account for the NO distribution results.

Abstract:Objective: To investigate nitric oxide synthase (NOS) expression in cochlea of guinea pig, and define the effect of NO as neurotransmitter and endothelium derived relaxing factor (EDRF) in auditory physiology and pathophysiology of inner ear diseases. Methods: Thirty five white red eyes guinea pigs were used, and expression of NOS in cochlea of guinea pig was studied with NADPH diaphorase histochemistry and in situ hybridization technology. Results: NOS mainly distributed in inner hair cell (IHC), outer hair cell (OHC) of Corti's organ, the cells of spiral ganglion (SG) and the marginal cells of stria vascularis (SV) in the guinea pig cochlea. The NOS in the IHC, OHC and spiral ganglion cell was different from the NOS in the marginal cells of SV. It submitted to brain NOS or endothelium NOS. Conclusion: Expression of NOS in cochlea was detected. It showed NO was neurotransmitter or modulator in cochlea afferent and joined self regulation of microcirculation of the cochlea.

Determination of catecholamines and the total antioxidant potential of blood plasma by use of an improved RPHPLC-ED assay

Abstract:Preliminary results are presented from application of reversed-phase HPLC with electrochemical detection to analysis of L-dopa and total antioxidant potential. It was possible to perform both analyses by using the same chromatographic conditions. The method was used to estimate their daily profiles in the blood plasma of patients treated with different types of drug.

Abstract:The Escherichia coli (E. coli) nirC gene encodes a nitrite transporter, which involved in transporting toxic nitrite (NO2−) from the environment into the bacteria. Although the deletion of nirC gene could cause changes in motility, adhesion in the previous study, and the virulence involved in the specified mechanism for pathogenic E. coli remains to be known. In the present work, we aimed to evaluate the role of NirC in a serotype O2:K1:H7 avian pathogenic Escherichia coli (APEC) strain. For this purpose, we generated a NirC-deficient mutant of APEC XM strain and examined its biological characteristics. The nirC gene deletion mutant enhanced ability of motility, decreased in biofilm formation, and it markedly reduced ability to adhere mouse brain microvascular endothelial cell b.End3 cells. For understanding its mechanism, sequentially we detected and found the stress regulator rpoS and its downstream genes csrA were up-regulated in NirC-deficient mutant while diguanylate cyclase gene dgcT was down-regulated. By high-performance liquid chromatography (HPLC) experiment, we demonstrated the concentration of intracellular 3′,5′-cyclic diguanosine monophosphate (c-di-GMP) significantly decrease in nirC gene deletion mutant. Taken data together, we may make a conclusion with a possible signal pathway clue, due to NirC mutation, environmental NO2− accumulation leads to nitrite stress and inactivates c-di-GMP synthesis by stimulating the stress regulator RpoS, resulting in changes of biological characteristics.

Role of nitric oxide synthase on nerve conduction and capillary blood flow in guinea pig cochlea

Abstract:AIM: To explore the effect and mechanism of nitric oxide synthase(NOS) in the regulation of nerve conduction and capillary blood flow in guinea pig cochlea.METHODS: After paraformaldehyde perfusion of the heart in 6 guinea pigs, the cochleas taken to prepared frozen sections following decalcification with edetic acid. The sections were incubated in nicotinamide adenine dinucleotide phosphate hydrogen(NADPH) for charactering the distribution and activity of NADPH-diaphorase by means of enzyme-linked immunosorbent assay(ELISA).RESULTS: Obvious NADPH-diaphorase activity was found in the guinea pig cochlea, cochlea nerve ending underlying the outer hair cells, and the endothelial cells of the capillary glomus. In addition, NADPH-diaphorase activity was also detected in the outer and inner pillar cells, supporting cells,spiral ligaments, and spiral ganglion cells.CONCLUSION: Nitric oxide is crucial in maintaining normal nerve conduction and capillary blood flow in the cochlea of guinea pigs.
Catalytic Nitric Oxide Synthase Activity in the White and Gray Matter Regions of the Spinal Cord of Rabbits

Abstract:The latest research reveals that nitric oxide as a gas messenger may diffuse into the surrounding extracellular fluid and act locally upon neighboring target cells. However, several observations raise the possibility that nitric oxide may also be released at a greater distance from the neuronal cell body. The catalytic nitric oxide synthase (cNOS) activity was therefore studied in the cervicothoracic and lumbosacral segments of the spinal cord of rabbits, including the white matter of dorsal columns (DC), lateral columns (LC) and ventral columns (VC), as well as the gray matter of dorsal horns (DH), intermediate zone (IZ) and ventral horns (VH). Lower cNOS activity was found in the white matter of both cervicothoracic (47 %) and lumbosacral (30 %) regions, whereas that detected in the gray matter of the lumbosacral part of the spinal cord was considerably higher (70 %). Enzyme activity varied from 43.4 to 77.2 dpm/μg protein in the cervicothoracic segments of the gray matter in the descending order: DH>VH>IZ. Similar cNOS activity was found in the white matter of the cervicothoracic segments (42.1-62.8 dpm/μg protein). When the activity of cNOS was compared in the lumbosacral segments, the highest enzyme activity was found in DH of the gray matter (198.7 dpm/μg protein) and the lowest cNOS in DC (45.8 dpm/μg protein) of the white matter. It was concluded that the white matter of the spinal cord contains similar cNOS activity in comparison to the gray matter.

Abstract:Objectives: Our purpose was to investigate the role of the nitric oxide (NO) signaling pathway in the regulation of ciliary beat frequency (CBF) in mouse nasal and tracheal epithelial cells.Methods: We studied the effects of the NO donor L-arginine (L-Arg) and specific inhibitors of the NO signaling pathway on CBF of both nasal and tracheal epithelial cells by using high-speed digital microscopy. We also examined eNOS, sGC beta, PKG I and acetylated a tubulin expression in native mouse nasal and tracheal epithelium using immunohistochemical methods.Results: L-Arg significantly increased CBF of cultured nasal and tracheal epithelial cells, and the effects were blocked by pretreatment with N-G-nitro-L-arginine methyl ester CL-NAME), a NOS inhibitor, with LY-83583, a sGC inhibitor, or with KT-5823, a PKG inhibitor. Positive immunostaining for NO signaling molecules including eNOS, sGC beta and PKG I was observed in either nasal or tracheal ciliated epithelium.Conclusion: NO plays a role in regulating CBF of mouse respiratory epithelial cells via a eN0S-NO-sGC beta-cGMP-PKG I pathway. Abstract:Collaria Provancher (Mirinae: Stenodemini) is a genus of grass-feeding plant bugs with 14 recognized species. The present work presents a taxonomic revision of the genus, including redescriptions of all species and the description of C. schwartzi sp. nov. from Central and Eastern Africa. The female genitalia of C. boliviana and C. villiersi, and the male and female genitalia of C. improvisa and C. obscuricornis are described for the first time. New distributional records for the Neotropical region are provided. A key to separate all the species and illustrations of the male and female genitalia of almost all the species are included. We highlight the need to produce better series of specimens with accurate host-plant data.
