Abstract:Abstract Due to their sessile nature, plants must respond to various environmental assaults in a coordinated manner. The endoplasmic reticulum is a central hub for plant responses to various stresses. We previously showed that Phytophthora utilizes effector PsAvh262-mediated binding immunoglobulin protein (BiP) accumulation for suppressing endoplasmic reticulum stress-triggered cell death. As a BiP binding partner, Bcl-2-associated athanogene 7 (BAG7) plays a crucial role in the maintenance of the unfolded protein response, but little is known about its role in plant immunity. In this work, we reveal a double-faced role of BAG7 in Arabidopsis–Phytophthora interaction in which it regulates endoplasmic reticulum stress-mediated immunity oppositely in different cellular compartments. In detail, it acts as a susceptibility factor in the endoplasmic reticulum, but plays a resistance role in the nucleus against Phytophthora. Phytophthora infection triggers the endoplasmic reticulum-to-nucleus translocation of BAG7, the same as abiotic heat stress; however, this process can be prevented by PsAvh262-mediated BiP accumulation. Moreover, the immunoglobulin/albumin-binding domain in PsAvh262 is essential for both pathogen virulence and BiP accumulation. Taken together, our study uncovers a double-faced role of BAG7; Phytophthora advances its colonization in planta by utilizing an effector to detain BAG7 in the endoplasmic reticulum.

Abstract:Heat Shock Protein 70 kDa (Hsp70) family molecular chaperones play critical roles in protein folding and trafficking in all eukaryotic cells. The mechanisms by which Hsp70 family chaperones are regulated, however, are only partly understood. BAG-1 binds the ATPase domains of Hsp70 and Hsc70, modulating their chaperone activity and functioning as a competitive antagonist of the co-chaperone Hip. We describe the identification of a family of BAG-1-related proteins from humans (BAG-2, BAG-3, BAG-4, BAG-5), the invertebrate Caenorhabditis elegans (BAG-1, BAG-2), and the fission yeast Schizosaccharomyces pombe (BAG-1A, BAG-1B). These proteins all contain a conserved approximately 45-amino acid region near their C termini (the BAG domain) that binds Hsc70/Hsp70, but they differ widely in their N-terminal domains. The human BAG-1, BAG-2, and BAG-3 proteins bind with high affinity (KD congruent with 1-10 nM) to the ATPase domain of Hsc70 and inhibit its chaperone activity in a Hip-repressible manner. The findings suggest opportunities for specification and diversification of Hsp70/Hsc70 chaperone functions through interactions with various BAG-family proteins.

Roles of six Hsp70 genes in virulence, cell wall integrity, antioxidant activity and multiple stress tolerance of Beauveria bassiana

Abstract:Functions of most Hsp70-coding genes remain unknown in filamentous fungi. Here, we show important roles of six Hsp70 homologs (Hsp70a-f) in Beauveria bassiana, a filamentous fungal insect pathogen that serves as a main source of wide-spectrum fungal insecticides. In phylogeny, Hsp70b-e, Hsp70g and Hsp70h are distinct from eight other partners that share the same clade with 14 Hsp70 proteins in model yeast. Among the 14 Hsp70-coding genes in B. bassiana, eight (hsp70g/hsp70h/ssa/ssb/ssc/kar/sse/ssz) were individually undeletable perhaps due to an essentiality of each for the fungal viability. Moderate defects in growth and conidiation occurred in the absence of hsp70e or hsp70f (lsh1). The disruption of hsp70f resulted in a reduction of 43% in conidial heat (45 degrees C) tolerance and of 97% in virulence, and the two reductions decreased to 19-27% and 18-45% in the disruption mutants of hsp70a to hsp70d respectively. Conidial UVB resistance decreased more in the absence of hsp70e (33%) than of hsp70f (25%), hsp70c (19%) or hsp70d (15%). All of these disruption mutants were differentially compromised in both cell wall integrity and antioxidant activity and also in cellular tolerance to Na+, Zn-2+, Mn2+, Cu2+ and/or Fe2+. Delayed acidification of submerged cultures and reduced secretion of organic acids occurred only in the absence of hsp70b or hsp70f. Our findings indicate that hsp70a-f are important for both host infection and multiple stress tolerance of B. bassiana, and functionally similar to or beyond the lsh1, ssb and ssz homologs characterized previously in some plant-pathogenic fungi.

Abstract:Plant stress responses control programmed cell death (PCD) pathways, potentially targeting crop improvement against environmental challenges, while maintaining intracellular protein homeostasis is crucial for survival. Because intracellular proteins are regularly damaged and lose functioning under stressful conditions, molecular chaperones serve as vital for life support because they maintain protein structure and function. Bcl-2-associated athanogenes (BAGs) are a multifunctional group chaperones distinguished by a common conserved BAG domain sequence. This review provides an extensive analysis of recent advancements in the study of plant BAGs , elucidating their versatile roles in plant biology. Plant BAGs are involved in many cellular processes, including stress response, cellular homeostasis, and plant development. Our perspective on plant BAGs has just been updated. Recently, we reported that AtBAG2 and AtBAG6 enable to tolerate Arabidopsis thaliana to multiple abiotic stresses. This review synthesizes the most recent research on the functional aspects of BAGs in different plant species, including Arabidopsis , tomato, rice, maize, wheat, chickpea, tobacco, banana, soybean, grapes, and mosses, highlighting their diverse interactions with signaling pathways and stress responses. This study explores the physiological functions of plant BAG proteins, highlighting challenges in unraveling BAG-mediated processes and the importance of advanced research approaches for sustainable agriculture, stress tolerance, and crop improvement. This study emphasizes the potential of plant BAG genes in addressing global food security and climate change, promoting additional research and discovery to open up new possibilities.

Characterization of the Hog1 MAPK pathway in the entomopathogenic fungus Beauveria bassiana.

Abstract:High-osmolarity glycerol (HOG) pathway required for yeast osmoregulation relies upon the mitogen-activated protein kinase (MAPK) Hog1 cascade that comprise the MAPKKKs Ssk2/Ssk22 and Ste11 converging on the MAPKK Pbs2. Here we show a Hog1 cascade with the unique MAPKKK Ssk2 acting in Beauveria bassiana. Hypersensitivity to high osmolarity and high resistance to fludioxonil fungicide appeared in ssk2, pbs2 and hog1 mutants whereas the two hallmark phenotypes were reversed in ste11. Increased sensitivity to heat shock and decreased sensitivity to cell wall perturbation also occurred in the three mutants but not in ste11 although antioxidant phenotypes were different in all deletion mutants. Intriguingly, signals of Hog1 phosphorylation induced by osmotic, oxidative and thermal cues were present in ste11 but absent in ssk2 and pbs2. Moreover, vegetative growth on minimal media with different carbon/nitrogen sources was much more suppressed in ste11 and ssk2 than in pbs2 and hog1 although all mutants suffered similar, but severe, conidiation defects on a standard medium. Normal host infection was abolished in ste11 while virulence was differentially attenuated in other mutants. Our findings exclude Ste11 from the Hog1 cascade that regulates multiple stress responses and environmental adaptation of B. bassiana and perhaps other filamentous fungi.

Abstract:Oxidation tolerance is an important determinant to predict the virulence and biocontrol potential of Beauveria bassiana , a well-known entomopathogenic fungus. As a transcriptional coactivator, multiprotein bridging factor 1 mediates the activity of transcription factor in diverse physiological processes, and its homolog in B. bassiana ( BbMBF1 ) contributes to fungal oxidation tolerance. In this study, the BbMBF1-interactomes under oxidative stress and normal growth condition were deciphered by mass spectrometry integrated with the immunoprecipitation. BbMBF1p factor has a broad interaction with proteins that are involved in various cellular processes, and this interaction is dynamically regulated by oxidative stress. Importantly, a B. bassiana homolog of yeast AP-1-like transcription factor ( BbAP - 1 ) was specifically associated with the BbMBF1-interactome under oxidation and significantly contributed to fungal oxidation tolerance. In addition, qPCR analysis revealed that several antioxidant genes are jointly controlled by BbAP - 1 and BbMBF1 . Conclusively, it is proposed that BbMBF1p protein mediates BbAP-1p factor to transcribe the downstream antioxidant genes in B. bassiana under oxidative stress. This study demonstrates for the first time a proteomic view of the MBF1-interactome in fungi, and presents an initial framework to probe the transcriptional mechanism involved in fungal response to oxidation, which will provide a new strategy to improve the biocontrol efficacy of B. bassiana .
Glc8, a Regulator of Protein Phosphatase Type 1, Mediates Oxidation Tolerance, Asexual Development and Virulence in Beauveria Bassiana, a Filamentous Entomopathogenic Fungus

Abstract:Protein phosphatase type 1 (PP1) plays an important role in cellular metabolism and development in yeast. In PP1 enzyme complex, Glc8 protein is a global regulatory subunit and regulates many physiological processes. However, its biological roles are unexplored in filamentous fungi. In this study, we characterized a yeast ortholog of Glc8 in Beauveria bassiana, a filamentous entomopathogenic fungus. Gene disruption of BbGlc8 had no significant effect on vegetative growth, but resulted in a significant reduction in conidiation (51%) and blastospore yield (55%) in the mutant. The ΔBbGlc8 mutant displayed an enhanced sensitivity to oxidative stress and a weakened virulence as indicated by cuticle infection and intrahemocoel injection assays. Transcriptomic analysis indicated that the genes regulated by BbGlc8 during conidiation were primarily associated with metabolism, cell rescue and cell wall formation. Notably, as a down-regulated gene in ΔBbGlc8 mutant, BbOsmC2 (a member of OsmC protein family) contributes to fungal resistance to salt stress, spore differentiation and virulence. Thus, BbOsmC2 functions as a down-stream target of BbGlc8 during spore differentiation, but not in stress response. Our findings indicate that BbGlc8 contributes to the biocontrol potential of B. bassiana by mediating comprehensive genetic pathways.
Genome-wide Identification of BCS1 Domain-Containing Proteins Reveals the Mitochondrial Bcs1 Essential for Growth, Stress Response, and Virulence of the Filamentous Entomopathogenic Fungus Beauveria Bassiana

Abstract:In yeasts, bcs1 is a mitochondrial AAA protein (ATPase associated with diverse cellular activities) and required for biogenesis of the complex III in mitochondrial electron transfer chain. However, the presence and biological roles of bcs1 remain largely unknown in the filamentous fungi. In present study, genome-wide identification revealed that there were six BCS1-domain containing proteins (Bbbcs1a through f) in the filamentous insect pathogenic fungus Beauveria bassiana, five of which (except for Bbbcs1f) were functionally analyzed. Phenotypic evaluation revealed that only Bbbcs1b and Bbbcs1c contributed to fungal physiologies, and they localized to nuclei and mitochondria, respectively. Hence, Bbbcs1c is considered as the ortholog of yeast bcs1 in B. bassiana. Ablation of Bbbcs1c did not affect biogenesis of mitochondria, but its loss significantly attenuated mitochondrial functionality (e.g., ATP synthesis and mitochondrial targeting of proteins) significantly. ΔBbbcs1c mutant displayed the impaired phenotypes in vegetative growth, stress response, development, and virulence. Notably, ΔBbbcs1c mutant displayed the increased sensitivity to linoleic acid (LA) stress and lost the intracellular fatty acid homeostasis. The Bbbcs1c loss compromised the mitochondrial membrane potential, and LA stress exacerbated this damage. These findings indicate that Bbbcs1c is a functional homolog of yeast bcs1 in B. bassiana and links mitochondrial functionality to unique lifestyle in the entomopathogenic fungi.
Three Mitogen-Activated Protein Kinases Required for Cell Wall Integrity Contribute Greatly to Biocontrol Potential of a Fungal Entomopathogen

Abstract:Bck1, Mkk1 and Slt2 are three mitogen-activated protein (MAP) kinases constituting cell wall integrity (CWI) pathway that may control multi-stress responses via crosstalk with high-osmolarity glycerol (HOG) pathway in budding yeast. In this study, Bck1, Mkk1 and Slt2 orthologues in Beauveria bassiana were confirmed as the three-module cascade essential for CWI because cell wall impairment occurred in the hyphae and conidia of Δbck1, Δmkk1 and Δslt2 examined in multiple experiments. Strikingly, all the deletion mutants became more sensitive to hyperosmotic NaCl and sorbitol with the Western blot of Hog1 phosphorylation being weakened in Δbck1 and absent in Δmkk1 and Δslt2. Apart from crossing responses to cell wall perturbation and high osmolarity, three deletion mutants exhibited faster growth and conidiation on nutrition-rich medium, much less virulence to Galleria mellonella larvae, and higher sensitivity to nutritional, fungicidal, thermal and UV-B irradiative stresses, accompanied with less accumulation of intracellular mannitol and trehalose. Moreover, Δmkk1 and Δslt2 were equally more sensitive to all the stresses of different types except wet-heat stress than wild type and more or less different from Δbck1 in sensitivity to most of the stresses despite their null responses to two oxidants. All the changes in three deletion mutants were restored by each targeted gene complementation. Taken together, the CWI-required Bck1, Mkk1 and Slt2 are all positive, but differential, regulators of multi-stress tolerance and virulence perhaps due to interplay with the HOG pathway essential for osmoregulation, thereby contributing greatly to the biocontrol potential of the fungal entomopathogen.
Divergent Physiological Functions of Four Atg22-like Proteins in Conidial Germination, Development, and Virulence of the Entomopathogenic Fungus Beauveria bassiana

Abstract:In yeast, Atg22 functions as a vacuolar efflux transporter to release the nutrients from the vacuole to the cytosol after the degradation of autophagic bodies. There are more than one Atg22 domain-containing proteins in filamentous fungi, but their physiological roles are largely unknown. In this study, four Atg22-like proteins (BbAtg22A through D) were functionally characterized in the filamentous entomopathogenic fungus Beauveria bassiana. These Atg22-like proteins exhibit different sub-cellular distributions. BbAtg22A localizes in lipid droplets. BbAtg22B and BbAtg22C are completely distributed in the vacuole, and BbAtg22D has an additional association with the cytomembrane. The ablation of Atg22-like proteins did not block autophagy. Four Atg22-like proteins systematically contribute to the fungal response to starvation and virulence in B. bassiana. With the exception of increment Bbatg22C, the other three proteins contribute to dimorphic transmission. Additionally, BbAtg22A and BbAtg22D are required for cytomembrane integrity. Meanwhile, four Atg22-like proteins contribute to conidiation. Therefore, Atg22-like proteins link distinct sub-cellular structures for the development and virulence in B. bassiana. Our findings provide a novel insight into the non-autophagic roles of autophagy-related genes in filamentous fungi.

Abstract:Autophagy is a conserved degradation system in eukaryotic cells that includes non-selective and selective processes. Selective autophagy functions as a selective degradation mechanism for specific substrates in which autophagy-related protein 11 (ATG11) acts as an essential scaffold protein. In B. bassiana, there is a unique ATG11 family protein, which is designated as BbATG11. Disruption of BbATG11 resulted in significantly reduced conidial germination under starvation stress. The mutant ΔBbATG11 displayed enhanced sensitivity to oxidative stress and impaired asexual reproduction. The conidial yield was reduced by approximately 75%, and this defective phenotype could be repressed by increasing exogenous nutrients. The virulence of the ΔBbATG11 mutant strain was significantly impaired as indicated in topical and intra-hemocoel injection bioassays, with a greater reduction in topical infection. Notably, BbATG11 was involved in pexophagy and mitophagy, but these two autophagic processes appeared in different fungal physiological aspects. Both pexophagy and mitophagy were associated with nutrient shift, starvation stress and growth in the host hemocoel, but only pexophagy appeared in both oxidation-stressed cells and aerial mycelia. This study highlights that BbATG11 mediates pexophagy and mitophagy in B. bassiana and links selective autophagy to the fungal stress response, conidiation and virulence.

Abstract:P-type Ca2+-ATPase (Pmr1) is a core element in calcium-calcineurin pathway and evidence for its cross-talk with other signalling pathways in filamentous fungi is of scarcity. Here, we characterized the striking functions of a Golgi Ca2+-ATPase (Bbpmr1) in Beauveria bassiana (fungal entomopathogen) by multi-phenotypic and transcriptional analyses under normal and stressful conditions. Bbpmr1 inactivation caused severe defects in nutritional uptake, growth, conidiation and germination under normal conditions, drastic reductions in cell tolerances to oxidative, hyperosmotic, cell wall disturbing and fungicidal stresses and toxic metal ions during colony growth and/or conidial germination, and half loss of the fungal biocontrol potential represented by conidial virulence, thermotolerance and UV-B resistance. Accompanied with the multi-phenotypic defects, four important genes associated with asexual development were repressed by 75% in Bbpmr1 versus wild type, and all or most of stress-responsive genes encoding 14 cascaded proteins in MAPK pathways, two Ras GTPases, two protein kinases, Ssk1-type response regulator, TOR signalling protein, and many downstream enzymes and proteins were greatly downregulated in Bbpmr1 under the chemical stresses. Conclusively, Bbpmr1 regulates positively fundamental aspects on B.bassiana biology and environmental adaptation through wide cross-talk with cellular signalling networks including MAPK cascades and those upstream or independent of the cascades.
The Autophagy-Related Genes BbATG1 and BbATG8 Have Different Functions in Differentiation, Stress Resistance and Virulence of Mycopathogen Beauveria Bassiana

Abstract:Autophagy-related proteins play significantly different roles in eukaryotes. In the entomopathogenic fungus Beauveria bassiana, autophagy is associated with fungal growth and development. BbATG1 (a serine/threonine protein kinase) and BbATG8 (a ubiquitin-like protein) have similar roles in autophagy, but different roles in other processes. Disruption mutants of BbATG1 and BbATG8 had impaired conidial germination under starvation stress. The mutant ΔBbATG8 exhibited enhanced sensitivity to oxidative stress, while a ΔBbATG1 mutant did not. BbATG1 and BbATG8 showed different roles in spore differentiation. The blastospore yield was reduced by 70% and 92% in ΔBbATG1 and ΔBbATG8 mutants, respectively, and the double mutant had a reduction of 95%. Conidial yield was reduced by approximately 90% and 50% in ΔBbATG1 and ΔBbATG8 mutants, respectively. A double mutant had a reduction similar to ΔBbATG1. Additionally, both BbATG1 and BbATG8 affected the levels of conidial protein BbCP15p required for conidiation. The virulence of each autophagy-deficient mutant was considerably weakened as indicated in topical and intrahemocoel injection assays, and showed a greater reduction in topical infection. However, BbATG1 and BbATG8 had different effects on fungal virulence. Our data indicate that these autophagy-related proteins have different functions in fungal stress response, asexual development and virulence.

Abstract:Phytopathogenic oomycetes, particularly Phytophthora capsici, the causal agent of Phytophthora blight disease in essential vegetables and fruit crops, remains a persistent challenge in the vegetable production industry. However, the core molecular regulators of the pathophysiology and broad-range host characteristics of P. capsici remain unknown. Here, we used transcriptomics and CRISPR-Cas9 technology to functionally characterize the contributions of a novel gene (PcTBCC1) coding for a hypothetical protein with a tubulin-binding cofactor C domain with a putative chloroplast-targeting peptide (cTP) to the pathophysiological development of P. capsici. We observed significant upregulation in the expression of PcTBCC1 during pathogen–host interactions. However, the vegetative growth of the ∆Pctbcc1 strains was not significantly different from the wild-type strains. PcTBCC1 gene replacement significantly compromised the sporulation, pathogenic differentiation, and virulence of P. capsici. At the same time, ∆Pctbcc1 strains were sensitive to cell wall stress-inducing osmolytes. These observations, coupled with the close evolutionary ties between PcTBCC1 and pathogenic oomycetes and algae, partly support the notion that PcTBCC1 is a conserved determinant of pathogenesis. This study provides insights into the significance of tubulin-binding cofactors in P. capsici and underscores the potential of PcTbcc1 as a durable target for developing anti-oomycides to control phytopathogenic oomycetes.

Abstract:Pil1A and Pil1B are two core eisosome proteins that are homologous to yeast Pil1/Lsp1 or filamentous fungal Pil1A/Pil1B but have been unexplored in entomopathogenic fungi. Here we examined subcellular localization and functions of Pil1A and Pil1B in Beauveria bassiana, a fungal insect pathogen. Either localization or co-localization experiments of the two proteins demonstrated that Pil1A and Pil1B were simultaneously localized at the periphery of hyphal cells for formation of stable, punctuate spots in B. bassiana. This is different from a reliance of proper Lsp1/Pil1B localization upon Pil1/Pil1A in other fungi. Deletions of pil1A and pil1B caused opposite changes in expression of many autophagy-related genes and formation of intravacuolar autophagosomes. Such opposite changes were restored to nearly normal status by exogenous rapamycin, implicating a link of Pil1A/B to the target of rapamycin signalling pathway. All single/double deletion mutants of pil1A and pil1B lost almost all pathogenicity due to reduced ability to secrete Pr1 proteases for cuticle degradation. They also showed differential changes in cell wall integrity and multiple stress responses. These findings unveil opposite roles for Pil1A and Pil1B in autophagic regulation and an essentiality of both for cell integrity, function and pathogenicity of the fungal entomopathogen.
A Mitochondrial Localized Chaperone Regulator OsBAG6 Functions in Saline-Alkaline Stress Tolerance in Rice

Abstract:B-cell lymphoma 2 (Bcl-2)-associated athanogene (BAG) family genes play prominent roles in regulating plant growth, development, and stress response. Although the molecular mechanism underlying BAG's response to abiotic stress has been studied in Arabidopsis, the function of OsBAG underlying saline-alkaline stress tolerance in rice remains unclear. In this study, OsBAG6, a chaperone regulator localized to mitochondria, was identified as a novel negative regulator of saline-alkaline stress tolerance in rice. The expression level of OsBAG6 was induced by high concentration of salt, high pH, heat and abscisic acid treatments. Overexpression of OsBAG6 in rice resulted in significantly reduced plant heights, grain size, grain weight, as well as higher sensitivity to saline-alkaline stress. By contrast, the osbag6 loss-of-function mutants exhibited decreased sensitivity to saline-alkaline stress. The transcriptomic analysis uncovered differentially expressed genes related to the function of "response to oxidative stress", "defense response", and "secondary metabolite biosynthetic process" in the shoots and roots of OsBAG6-overexpressing transgenic lines. Furthermore, cytoplasmic levels of Ca2+ increase rapidly in plants exposed to saline-alkaline stress. OsBAG6 bound to calcium sensor OsCaM1-1 under normal conditions, which was identified by comparative interactomics, but not in the presence of elevated Ca2+. Released OsCaM1-1 saturated with Ca2+ is then able to regulate downstream stress-responsive genes as part of the response to saline-alkaline stress. OsBAG6 also interacted with energy biosynthesis and metabolic pathway proteins that are involved in plant growth and saline-alkaline stress response mechanisms. This study reveals a novel function for mitochondrial localized OsBAG6 proteins in the saline-alkaline stress response alongside OsCaM1-1.

Abstract:The allosteric coupling of the highly conserved nucleotide- and substrate-binding domains of Hsp70 has been studied intensively. In contrast, the role of the disordered, highly variable C-terminal region of Hsp70 remains unclear. In many eukaryotic Hsp70s, the extreme C-terminal EEVD motif binds to tetratricopeptide-repeat domains of Hsp70 co-chaperones. Here, we discovered that the TVEEVD sequence of Saccharomyces cerevisiae cytoplasmic Hsp70 (Ssa1) functions as a SUMO-interacting motif. A second C-terminal motif of ~15 amino acids between the α-helical lid and the extreme C terminus, previously identified in bacterial and eukaryotic organellar Hsp70s, is known to enhance chaperone function by transiently interacting with folding clients. Using structural analysis, interaction studies, fibril formation assays, and in vivo functional assays, we investigated the individual contributions of the α-helical bundle and the C-terminal disordered region of Ssa1 in the inhibition of fibril formation of the prion protein Ure2. Our results revealed that although the α-helical bundle of the Ssa1 substrate-binding domain (SBDα) does not directly bind to Ure2, the SBDα enhances the ability of Hsp70 to inhibit fibril formation. We found that a 20-residue C-terminal motif in Ssa1, containing GGAP and GGAP-like tetra-peptide repeats, can directly bind to Ure2, the Hsp40 co-chaperone Ydj1, and α-synuclein, but not to the SUMO-like protein SMT3 or BSA. Deletion or substitution of the Ssa1 GGAP motif impaired yeast cell tolerance to temperature and cell wall damage stress. This study highlights that the C-terminal GGAP motif of Hsp70 is important for substrate recognition and mediation of the heat shock response.

Abstract:Hsp90 is a conserved molecular chaperone that assists in the folding and function of diverse cellular regulators, with a profound impact on biology, disease, and evolution. As a central hub of protein interaction networks, Hsp90 engages with hundreds of protein–protein interactions within eukaryotic cells. These interactions include client proteins, which physically interact with Hsp90 and depend on the chaperone for stability or function, as well as co-chaperones and partner proteins that modulate chaperone function. Currently, there are no methods to accurately predict Hsp90 interactors and there has been considerable network rewiring over evolutionary time, necessitating experimental approaches to define the Hsp90 network in the species of interest. This is a pressing challenge for fungal pathogens, for which Hsp90 is a key regulator of stress tolerance, drug resistance, and virulence traits. To address this challenge, we applied a novel biochemical fractionation and quantitative proteomic approach to examine alterations to the proteome upon perturbation of Hsp90 in a leading human fungal pathogen, Candida albicans. In parallel, we performed affinity purification coupled to mass spectrometry to define physical interacting partners for Hsp90 and the Hsp90 co-chaperones and identified 164 Hsp90-interacting proteins, including 111 that are specific to the pathogen. We performed the first analysis of the Hsp90 interactome upon antifungal drug stress and demonstrated that Hsp90 stabilizes processing body (P-body) and stress granule proteins that contribute to drug tolerance. We also describe novel roles for Hsp90 in regulating posttranslational modification of the Rvb1-Rvb2-Tah1-Pih1 (R2TP) complex and the formation of protein aggregates in response to thermal stress. This study provides a global view of the Hsp90 interactome in a fungal pathogen, demonstrates the dynamic role of Hsp90 in response to environmental perturbations, and highlights a novel connection between Hsp90 and the regulation of mRNA-associated protein granules.

Abstract:Oomycetes are filamentous microorganisms easily mistaken as fungi but vastly differ in physiology, biochemistry, and genetics. This commonly-held misconception lead to a reduced effectiveness by using conventional fungicides to control oomycetes, thus it demands the identification of novel functional genes as target for precisely design oomycetes-specific microbicide. The present study initially analyzed the available transcriptome data of the model oomycete pathogen, Phytophthora sojae , and constructed an expression matrix of 10,953 genes across the stages of asexual development and host infection. Hierarchical clustering, specificity, and diversity analyses revealed a more pronounced transcriptional plasticity during the stages of asexual development than that in host infection, which drew our attention by particularly focusing on transcripts in asexual development stage to eventually clustered them into 6 phase-specific expression modules. Three of which respectively possessing a serine/threonine phosphatase (PP2C) expressed during the mycelial and sporangium stages, a histidine kinase (HK) expressed during the zoospore and cyst stages, and a bZIP transcription factor (bZIP32) exclusive to the cyst germination stage were selected for down-stream functional validation. In this way, we demonstrated that PP2C, HK, and bZIP32 play significant roles in P . sojae asexual development and virulence. Thus, these findings provide a foundation for further gene functional annotation in oomycetes and crop disease management. Oomycetes are fungi-like microorganisms with differences in biology and genetics. Using fungicides to control the pathogenic oomycetes has limited effectiveness. Therefore, a detailed understanding of the oomycetes is necessary for identifying potential candidates as fungicide targets. The present study investigated the transcriptional changes in a model oomycete pathogen, Phytophthora sojae , during asexual development and plant infection. The analysis identified various types of genes with differences in expression patterns during development and infection. Further analysis focused on the transcriptome dynamics during asexual stages which exhibited more remarkable plasticity. Detailed analysis of the genes revealed stage-specific responses of a few genes and specifically identified three genes with functional relevance in different developmental stages of P . sojae , including a serine/threonine phosphatase in mycelial and sporangium stages, a histidine kinase in zoospore and cyst stages, and a bZIP transcription factor in cyst germination. Also these three genes function in different participate in multiple layers of the pathogenicity process. Thus, the study provides a reference for further research on the candidate genes, which may be used to defeat the pathogenic oomycetes.

Abstract:Abstract The basic leucine zipper (bZIP) transcription factor family, one of the largest and most diverse transcription factor families in eukaryotes, is associated with the development and stress responses of many eukaryotic organisms. However, their biological functions in oomycete plant pathogens are unclear. A genome-wide analysis of bZIP transcription factors in Phytophthora sojae showed that PsBZP32, which has a unique bZIP-PAS domain structure, exhibited a high transcription level during the early stages of P. sojae infection. We silenced PsBZP32 in P. sojae and found that the transformants showed defective cyst germination and pathogenicity. The transformants could not eliminate host-derived reactive oxygen species during infection and were more sensitive to oxidative stress. Their response to oxidative stress was independent of the PsBZP32 transcription level; however, subcellular localization and phosphorylation of PsBZP32 were affected by H 2 O 2 stress. These results indicate that PsBZP32 is involved in regulation of P. sojae cyst germination, oxidative stress response, and pathogenicity.
