Abstract:Aerobic CH4 oxidation coupled to denitrification (AME-D) can not only mitigate the emission of greenhouse gas (e.g., CH4) to the atmosphere, but also reduce NO3− and/or NO2− and alleviate nitrogen pollution. The effects of O2 tension on the community and functional gene expression of methanotrophs and denitrifiers were investigated in this study. Although higher CH4 oxidation occurred in the AME-D system with an initial O2 concentration of 21% (i.e., the O2-sufficient condition), more NO3−-N was removed at the initial O2 concentration of 10% (i.e., the O2-limited environment). Type I methanotrophs, including Methylocaldum, Methylobacter, Methylococcus, Methylomonas, and Methylomicrobium, and type II methanotrophs, including Methylocystis and Methylosinus, dominated in the AME-D systems. Compared with type II methanotrophs, type I methanotrophs were more abundant in the AME-D systems. Proteobacteria and Actinobacteria were the main denitrifiers in the AME-D systems, and their compositions varied with the O2 tension. Quantitative PCR of the pmoA, nirS, and 16S rRNA genes showed that methanotrophs and denitrifiers were the main microorganisms in the AME-D systems, accounting for 46.4% and 24.1% in the O2-limited environment, respectively. However, the relative transcripts of the functional genes including pmoA, mmoX, nirK, nirS, and norZ were all less than 1%, especially the functional genes involved in denitrification under the O2-sufficient condition, likely due to the majority of the denitrifiers being dormant or even nonviable. These findings indicated that an optimal O2 concentration should be used to optimize the activity and functional gene expression of aerobic methanotrophs and denitrifiers in AME-D systems.

Abstract:Aerobic methane (CH) oxidation coupled to denitrification (AME-D) can not only mitigate CH emission into the atmosphere, but also potentially alleviate nitrogen pollution in surface waters and engineered ecosystems, and it has attracted substantial research interest. O concentration plays a key role in AME-D, yet little is understood about how it impacts microbial interactions. Here, we applied isotopically labeled KNO and CH and metagenomic analyses to investigate the metabolic and microbial link of AME-D at different O levels. Among the four experimental O levels of 21%,10%, 5% and 2.5% and a CH concentration of 8% (i.e., the O/CH ratios of 2.62, 1.26, 0.63 and 0.31), the highest NO-N removal occurred in the AME-D system incubated at the O concentration of 10%. Methanol and acetate may serve as the trophic linkage between aerobic methanotrophs and denitrifers in the AME-D systems. Methylotrophs including Methylophilus, Methylovorus, Methyloversatilis and Methylotenera were abundant under the O-sufficient condition with the O concentration of 21%, while denitrifiers such as Azoarcus, Thauera and Thiobacillus dominated in the O-limited environment with the O concentration of 10%. The competition of denitrifiers and methylotrophs in the AME-D system for CH-derived carbon, such as methanol and acetate, might be influenced by chemotactic responses. More methane-derived carbon flowed into methylotrophs under the O-sufficient condition, while more methane-derived carbon was used for denitrification in the O-limited environment. These findings can aid in evaluating the distribution and contribution of AME-D and in developing strategies for mitigating CH emission and nitrogen pollution in natural and engineered ecosystems.

Abstract:Aerobic methane oxidation coupled with denitrification (AME-D) executed in membrane biofilm bioreactors (MBfRs) provides a high promise for simultaneously mitigating methane (CH 4 ) emissions and removing nitrate in wastewater. However, systematically experimental investigation on how oxygen partial pressure affects the development and characteristics of counter-diffusional biofilm, as well as its spatial stratification profiles, and the cooperative interaction of the biofilm microbes, is still absent. In this study, we combined Optical Coherence Tomography (OCT) with Confocal Laser Scanning Microscopy (CLSM) to in-situ characterize the development of counter-diffusion biofilm in the MBfR for the first time. It was revealed that oxygen partial pressure onto the MBfR was capable of manipulating biofilm thickness and spatial stratification, and then managing the distribution of functional microbes. With the optimized oxygen partial pressure of 5.5 psig (25% oxygen content), the manipulated counter-diffusional biofilm in the AME-D process obtained the highest denitrification efficiency, due mainly to that this biofilm had the proper dynamic balance between the aerobic-layer and anoxic-layer where suitable O 2 gradient and sufficient aerobic methanotrophs were achieved in aerobic-layer to favor methane oxidation, and complete O 2 depletion and accessible organic sources were kept to avoid constraining denitrification activity in anoxic-layer. By using metagenome analysis and Fluorescence in situ hybridization (FISH) staining, the spatial distribution of the functional microbes within counter-diffused biofilm was successfully evidenced, and Rhodocyclaceae , one typical aerobic denitrifier, was found to survive and gradually enriched in the aerobic layer and played a key role in denitrification aerobically. This in-situ biofilm visualization and characterization evidenced directly for the first time the cooperative path of denitrification for AME-D in the counter-diffused biofilm, which involved aerobic methanotrophs, heterotrophic aerobic denitrifiers, and heterotrophic anoxic denitrifiers.

Abstract:Previous work documented complete perchlorate reduction in a membrane biofilm reactor (MBfR) using methane as the sole electron donor and carbon source. This work explores how the biofilm’s microbial community evolved as the biofilm stage-wise reduced different combinations of perchlorate, nitrate, and nitrite. The initial inoculum, carrying out anaerobic methane oxidation coupled to denitrification (ANMO-D), was dominated by uncultured Anaerolineaceae and Ferruginibacter sp. The microbial community significantly changed after it was inoculated into the CH4-based MBfR and fed with a medium containing perchlorate and nitrite. Archaea were lost within the first 40 days, and the uncultured Anaerolineaceae and Ferruginibacter sp. also had significant losses. Replacing them were anoxic methanotrophs, especially Methylocystis, which accounted for more than 25 % of total bacteria. Once the methanotrophs became important, methanol-oxidizing denitrifying bacteria, namely, Methloversatilis and Methylophilus, became important in the biofilm, probably by utilizing organic matter generated by the metabolism of methanotrophs. When methane consumption was equal to the maximum-possible electron-donor supply, Methylomonas, also an anoxic methanotroph, accounted for >10 % of total bacteria and remained a major part of the community until the end of the experiments. We propose that aerobic methane oxidation coupled to denitrification and perchlorate reduction (AMO-D and AMO-PR) directly oxidized methane and reduced NO3 − to NO2 − or N2O under anoxic condition, producing organic matter for methanol-assimilating denitrification and perchlorate reduction (MA-D and MA-PR) to reduce NO3 −. Simultaneously, bacteria capable of anaerobic methane oxidation coupled to denitrification and perchlorate reduction (ANMO-D and ANMO-PR) used methane as the electron donor to respire NO3 − or ClO4 − directly.
Denitrifying Anaerobic Methane Oxidation: A Previously Overlooked Methane Sink in Intertidal Zone

Abstract:The intertidal zone is an open ecosystem rich in organic matter and plays an important role in global biogeochemical cycles. It was previously considered that methane was mainly removed by sulfate-dependent anaerobic methane oxidation (sulfate-AOM) process in marine ecosystems while other anaerobic methane oxidation processes were ignored. Recent researches have demonstrated that denitrifying anaerobic methane oxidation (DAMO), consisting of nitrite-dependent anaerobic methane oxidation (nitrite-AOM) and nitrate-dependent anaerobic methane oxidation (nitrate-AOM), can also oxidize methane. In this work, the community structure, quantity and potential methane oxidizing rate of DAMO archaea and bacteria in the intertidal zone were studied by high-throughput sequencing, qPCR and stable isotope tracing method. The results showed that nitrate-AOM and nitrite-AOM were both active in the intertidal zone and showed approximate methane oxidation rates. The copy number of 16S rRNA gene of DAMO archaea and DAMO bacteria were 104 ∼ 105 copies g-1 (dry sediment), whereas NC10 bacteria were slightly higher. The contribution rate of DAMO process to total anaerobic methane removal in the intertidal zone reached 65.6% ∼ 100%, which indicates that DAMO process is an important methane sink in intertidal ecosystem. Laboratory incubations also indicated that DAMO archaea were more sensitive to oxygen and preferred a more anoxic environment. These results help us draw a more complete picture of methane and nitrogen cycles in natural habitats.
Strategy for Denitrifying Anaerobic Methane-Oxidizing Bacteria Growing under the Oxygen-Present Condition.

Abstract:Denitrifying anaerobic methane oxidizing (DAMO) bacteria are newly discovered microorganisms that use methane as the electron donor to reduce nitrite into dinitrogen. They have potential value on nitrogen removal from wastewater. However, the oxygen exposure in engineering is considered one of the bottlenecks for DAMO engineering application. In this work, we cultured DAMO bacteria under oxic and anoxic conditions in a gas-lift sequencing batch reactor (GLSBR) to explore DAMO bacterial response to oxygen stress. Under oxic conditions (7.5-8mgO(2)/L), the extension of hydraulic retention time (HRT) from 2 days to 4 days increased DAMO bacterial abundance by 3.8 times. Under anoxic conditions (0.2-0.5 mg O-2/L), DAMO bacterial abundance increased by 30.1 times and were kept over 2.0 x 10(11) copies g(-1) wet sludge. During the enrichment, microbial aggregates were formed and DAMObacteria tended to be distributed inside the aggregates. Notably, aerobic methanotrophs existed in thewhole process, capable of consuming oxygen and providing a suitable environment for DAMO bacterial growth. Finally, DAMO bacteria were enriched and the relative abundance was 16.16%. This work provides new insights into DAMO bacterial enrichment and their application in wastewater treatment. Nitrate Effects on Chromate Reduction in a Methane-Based Biofilm.

Abstract:The effects of nitrate (NO3-) on chromate (Cr(VI)) reduction in a membrane biofilm reactor (MBfR) were studied when CH4 was the sole electron donor supplied with a non-limiting delivery capacity. A high surface loading of NO3- gave significant and irreversible inhibition of Cr(VI) reduction. At a surface loading of 500 mg Cr/m(2)-d, the Cr(VI)-removal percentage was 100% when NO3- was absent (Stage 1), but was dramatically lowered to < 25% with introduction of 280 mg N m(-2)-d NO3- (Stage 2). After similar to 50 days operation in Stage 2, the Cr(VI) reduction recovered to only similar to 70% in Stage 3, when NO3- was removed from the influent; thus, NO3- had a significant long-term inhibition effect on Cr(VI) reduction. Weighted PCoA and UniFrac analyses proved that the introduction of NO3- had a strong impact on the microbial community in the biofilms, and the changes possibly were linked to the irreversible inhibition of Cr(VI) reduction. For example, Meiothermus, the main genus involved in Cr(VI) reduction at first, declined with introduction of NO3-. The denitrifier Chitinophagaceae was enriched after the addition of NO3-, while Pelomonas became important when nitrate was removed, suggesting its potential role as a Cr(VI) reducer. Moreover, introducing NO3- led to a decrease in the number of genes predicted (by PICRUSt) to be related to chromate reduction, but genes predicted to be related to denitrification, methane oxidation, and fermentation increased. Intracellular electron competition in response to the oxygen pressure of the aerobic denitrification process in an O 2 -based membrane biofilm reactor (MBfR) for nitrate removal

Abstract:This study quantitatively investigated the effect of dissolved oxygen (DO) concentration on aerobic denitrification, and showed the mechanism of aerobic denitrification from the perspective of electron competition by cultivating Pseudomonas stutzeri T13, a typical aerobic denitrifier, in an oxygen-based membrane biofilm reactor (O 2 -based MBfR). The experiments showed that when the O 2 pressure increased from 2 to 10 psig , the average effluent DO concentration during steady-state phases increased from 0.02 to 4.23 mg/L, and the corresponding mean NO 3 − -N removal efficiency slightly decreased from 97.2 % to 90.9 %. Compared to the maximum theoretical flux of O 2 in various phases, the actual O 2 transfer flux increased from a limited status (2.07 e − eq m −2 d −1 at 2 psig) to an excessive status (5.58 e − eq m −2 d −1 at 10 psig). The increase of DO inhibited the electron availability for aerobic denitrification, which decreased from 23.97 % to 11.46 %, accompanying the increased electron availability for aerobic respiration from 15.87 % to 28.36 %. Unlike the napA and norB genes, the expression of the nirS and nosZ genes was significantly affected by DO, with the highest relative fold-changes of 6.5 and 6.13 at 4 psig O 2 , respectively. The results contribute to clarifying the mechanism of aerobic denitrification from the quantitative perspective of electron distribution and the qualitative perspective of gene expression, which benefits the control and practical application of aerobic denitrification for wastewater treatment.

Effect of oxygen gradients on the activity and microbial community structure of a nitrifying, membrane-aerated biofilm

Abstract:Shortcut nitrogen removal, that is, removal via formation and reduction of nitrite rather than nitrate, has been observed in membrane-aerated biofilms (MABs), but the extent, the controlling factors, and the kinetics of nitrite formation in MABs are poorly understood. We used a special MAB reactor to systematically study the effects of the dissolved oxygen (DO) concentration at the membrane surface, which is the biofilm base, on nitrification rates, extent of shortcut nitrification, and microbial community structure. The focus was on anoxic bulk liquids, which is typical in MAB used for total nitrogen (TN) removal, although aerobic bulk liquids were also studied. Nitrifying MABs were grown on a hollow-fiber membrane exposed to 3 mg N/L ammonium. The MAB intra-membrane air pressure was varied to achieve different DO concentrations at the biofilm base, and the bulk liquid was anoxic or with 2 g m(-3) DO. With 2.2 and 3.5 g m(-3) DO at the biofilm base, and with an anoxic bulk-liquid, the ammonium fluxes were 0.75 and 1.0 g N m(-2) day(-1), respectively, and nitrite was the main oxidized nitrogen product. However, with membrane DO of 5.5 g m(-3), and either zero or 2 g m(-3) DO in the bulk, the ammonium flux was around 1.3 g N m(-2) day(-1), and nitrate flux increased significantly. For all experiments, the cell density of ammonium oxidizing bacteria (AOB) was relatively uniform throughout the biofilm, but the density of nitrite oxidizing bacteria (NOB) decreased with decreasing biofilm DO. Among NOB, Nitrobacter spp. were dominant in biofilm regions with 2 g m(-3) DO or greater, while Nitrospira spp. were dominant in regions with less than 2 g m(-3) DO. A biofilm model, including AOB, Nitrobacter spp., and Nitrospira spp., was developed and calibrated with the experimental results. The model predicted the greatest extent of nitrite formation (95%) and the lowest ammonium oxidation flux (0.91 g N m(-2) day(-1)) when the membrane DO was 2 g m(-3) and the bulk liquid was anoxic. Conversely, the model predicted the lowest extent of nitrite formation (40%) and the highest ammonium oxidation flux (1.5 g N m(-2) day(-1)) when the membrane-DO and bulk-DO were 8 g m(-3) and 2 g m(-3), respectively. The estimated kinetic parameters for Nitrospira spp., revealed a high affinity for nitrite and oxygen. This explains the dominance of Nitrospira spp. over Nitrobacter spp. in regions with low nitrite and oxygen concentrations. Our results suggest that shortcut nitrification can effectively be controlled by manipulating the DO at the membrane surface. A tradeoff is made between increased nitrite accumulation at lower DO, and higher nitrification rates at higher DO.
Indigenous mixotrophic aerobic denitrifiers stimulated by oxygen micro/nanobubble-loaded microporous biochar

Abstract:The prevalence of hypoxia in surface sediment inhibits the growth of aerobic denitrifiers in natural waters. A novel oxygen micro/nanobubble-loaded microporous biochar (OMB) was developed to activate indigenous aerobic denitrifiers in this study. The results indicate a thin-layer OMB capping mitigates hypoxia effectively. Following a 30-day microcosm-based incubation, a 60 % decrease in total nitrogen concentration was observed, and the oxygen penetration depth in the sediment was increased from <4.0 mm to 38.4 mm. High-throughput sequencing revealed the stimulation of indigenous mixotrophic aerobic denitrifiers, including autotrophic denitrifiers such as Hydrogenophaga and Thiobacillus , heterotrophic denitrifiers like Limnobacter and unclassified_f_Methylophilaceae , and heterotrophic nitrification aerobic denitrification bacteria, including Shinella and Acidovorax , with total relative abundance reaching up to 38.1 %. Further analysis showed OMB enhanced the overall collaborative relationships among microorganisms and promoted the expression of nitrification- and denitrification-related genes. This study introduces an innovative strategy for stimulating indigenous aerobic denitrifiers in aquatic ecosystems.

Abstract:Nitrification, a process carried out by aerobic microorganisms that oxidizes ammonia to nitrate via nitrite, is an indispensable step in wastewater nitrogen removal. To facilitate energy and carbon savings, applying low dissolved oxygen (DO) is suggested to shortcut the conventional biological nitrogen removal pathway, however, the impact of low DO on nitrifying communities within activated sludge is not fully understood. This study used genome-resolved metagenomics to compare nitrifying communities under extremely low- and high-DO. Two bioreactors were parallelly operated to perform nitrification and DO was respectively provided by limited gas-liquid mass transfer from the atmosphere (AN reactor, DO 5.0 mg/L). Low DO was thought to limit nitrifiers growth; however, we demonstrated that complete nitrification could still be achieved under the extremely low-DO conditions, but with no nitrite accumulation observed. Kinetic analysis showed that after long-term exposure to low DO, nitrifiers had a higher oxygen affinity constant and could maintain a relatively high nitrification rate, particularly at low levels of DO (<0.2 mg/L). Community-level gene analysis indicated that low DO promoted enrichment of nitrifiers (the genera Nitrosomonas and Nitrospira , increased by 2.3- to 4.3-fold), and also harbored with 2.3 to 5.3 times higher of nitrification functional genes. Moreover, 46 high-quality (>90 % completeness and <5 % contamination) with 3 most abundant medium-quality metagenome-assembled genomes (MAGs) were retrieved using binning methods. Genome-level phylogenetic analysis revealed the species succession within nitrifying populations. Surprisingly, compared to DO-rich conditions, low-DO conditions were found to efficiently suppressed the ordinary heterotrophic microorganisms (e.g., the families Anaerolineales, Phycisphaerales , and Chitinophagales ), but selected for the specific candidate denitrifiers (within phylum Bacteroidota ). This study provides new microbial insights to demonstrate that low-DO favors the enrichment of autotrophic nitrifiers over heterotrophs with species-level successions, which would facilitate the optimization of energy and carbon management in wastewater treatment.

Abstract:Denitrifying anaerobic methane oxidation (DAMO) combining anaerobic ammonium oxidation (Anammox) process is a novel nitrogen removal technology. However, the roles of methane transfer (gas phase) and nitrogen transfer (liquid phase) in the heterogeneous process remain unclear. In this study, granular DAMO and Anammox co-cultures were inoculated from a hollow-fiber membrane bioreactor into a sequence batch reactor (SBR). Since the methane transfer became limited in SBR, the nitrate removal rate first decreased and then increased to 10 mg/(L∙day), while the ammonium removal rate did not recover and was around 2 mg/(L∙day). The activity of DAMO archaea and Anammox bacteria decreased noticeably. Furthermore, granular aggregates dispersed into small granules and ultimately became flocs with poor settleability in SBR. The content of extracellular polymeric substances decreased, especially that of proteins and humics. DAMO archaea decreased by 94.6% and Anammox bacteria decreased by 72%. In summary, the limitation of methane transfer affected DAMO and Anammox processes more notably than nitrogen transfer, resulting in lower nitrogen removal, granule disruption, and microbial community succession.

The results indicated that the high concentration of DO (1.0 mg/L) significantly inhibited the activity of anammox bacteria (AnAOB) while enhanced the activity of nitrite oxidation bacteria (NOB) in the PN/A reactor. 1.0 mg/L DO promoted the abundance of NOB (such as Nitrospira ) and some heterotrophic microorganisms (e.g., SBR1031 and OLB13), while decreased ammonia oxidizing bacteria (AOB, e.g., Nitrosomonas ) and AnAOB (Ca. Brocadia and Ca. Kuenenia ). 1.0 mg/L DO concentration driven a larger and more complex network with increased percentage of positive links, which might be a strategy for microbes to resist the adverse environment. Furthermore, the subnetworks of anammox bacteria and Nitrosomonas became smaller and simpler, whilst the subnetworks of heterotrophic bacteria (SBR1031) were more complex and connected. The decreased activity and abundance of AnAOB, together with the shifts of microbial interactions might result in the deterioration of nitrogen removal under 1.0 mg/L DO. This study provides a relatively comprehensive perspective for us to understand the effects of DO on the microbial interactions in the PN/A system.

Optimizing the Activity of Denitrifying Anaerobic Methane Oxidation Archaea by Tailoring Micronutrient Compositions: A Strategy for Enhanced Nitrate Reduction

Abstract:Metallic micronutrients play a crucial role in regulating the activity of microorganisms. Denitrifying anaerobic methane oxidation (DAMO) archaea possess all the canonical enzymes analogous to those involved in methanogenesis, which is notably rich in metalloenzymes. However, the relationship between metallic micronutrients and DAMO archaeal activity remains unexplored. Here we investigated the effect of micronutrients (Fe(II), Co(Ⅱ), Ni(Ⅱ), Zn(Ⅱ), Se(Ⅳ), W(Ⅵ) and Mo(Ⅵ)) on the DAMO archaeal activity. Batch assays indicated that among these micronutrients, Mo(Ⅵ), W(Ⅵ) and Se(Ⅳ) had a significant impact on DAMO archaeal activity, whereas the others showed minimal effects within their respective concentration ranges. Notably, the optimal concentrations of Mo(Ⅵ), W(Ⅵ) and Se(Ⅳ) substantially boosted DAMO archaeal activity, increasing nitrate conversion rates by 2.1, 1.45, and 1.84 times, respectively, compared to the rates at their lowest concentrations. An orthogonal experiment further confirmed the significant impact of Se(Ⅳ) and identified an optimal micronutrients composition. Subsequent long-term operation of a sequencing batch reactor (SBR) with this optimized medium demonstrated substantial improvements in DAMO performance. Specially, the nitrate removal rate increased to approximately 1.5 times higher than that of the original medium, and the nitrate consumption rate reached 1.98 mmol·d−1. In addition, microbial analysis indicated that both the abundance of Candidatus Methanoperedens and the key functional gene mcrA increased by 1.26 and 2.52 times, respectively, compared to that operated with the original medium, suggesting that the optimized medium enhanced both the metabolic activity and growth of DAMO archaea. These results underscore the pivotal role of micronutrients in modulating DAMO archaeal activity, thereby offering valuable insights for optimizing the DAMO process and advancing its practical application.
Anaerobic Oxidation of Methane Coupled with Dissimilatory Nitrate Reduction to Ammonium Fuels Anaerobic Ammonium Oxidation

Abstract:Nitrate/nitrite-dependent anaerobic methane oxidation (n-DAMO) is critical for mitigating methane emission and returning reactive nitrogen to the atmosphere. The genomes of n-DAMO archaea show that they have the potential to couple anaerobic oxidation of methane to dissimilatory nitrate reduction to ammonium (DNRA). However, physiological details of DNRA for n-DAMO archaea were not reported yet. This work demonstrated n-DAMO archaea coupling the anaerobic oxidation of methane to DNRA, which fueled Anammox in a methane-fed membrane biofilm reactor with nitrate as only electron acceptor. Microelectrode analysis revealed that ammonium accumulated where nitrite built up in the biofilm. Ammonium production and significant upregulation of gene expression for DNRA were detected in suspended n-DAMO culture with nitrite exposure, indicating that nitrite triggered DNRA by n-DAMO archaea. 15N-labeling batch experiments revealed that n-DAMO archaea produced ammonium from nitrate rather than from external nitrite. Localized gradients of nitrite produced by n-DAMO archaea in biofilms induced ammonium production via the DNRA process, which promoted nitrite consumption by Anammox bacteria and in turn helped n-DAMO archaea resist stress from nitrite. Abstract:Aerobic methane (CH4) oxidation coupled to denitrification (AME-D) can not only mitigate CH4 emission into the atmosphere, but also potentially alleviate nitrogen pollution in surface waters and engineered ecosystems, and it has attracted substantial research interest. O2 concentration plays a key role in AME-D, yet little is understood about how it impacts microbial interactions. Here, we applied isotopically labeled K15NO3 and 13CH4 and metagenomic analyses to investigate the metabolic and microbial link of AME-D at different O2 levels. Among the four experimental O2 levels of 21%,10%, 5% and 2.5% and a CH4 concentration of 8% (i.e., the O2/CH4 ratios of 2.62, 1.26, 0.63 and 0.31), the highest NO3--N removal occurred in the AME-D system incubated at the O2 concentration of 10%. Methanol and acetate may serve as the trophic linkage between aerobic methanotrophs and denitrifers in the AME-D systems. Methylotrophs including Methylophilus, Methylovorus, Methyloversatilis and Methylotenera were abundant under the O2-sufficient condition with the O2 concentration of 21%, while denitrifiers such as Azoarcus, Thauera and Thiobacillus dominated in the O2-limited environment with the O2 concentration of 10%. The competition of denitrifiers and methylotrophs in the AME-D system for CH4-derived carbon, such as methanol and acetate, might be influenced by chemotactic responses. More methane-derived carbon flowed into methylotrophs under the O2-sufficient condition, while more methane-derived carbon was used for denitrification in the O2-limited environment. These findings can aid in evaluating the distribution and contribution of AME-D and in developing strategies for mitigating CH4 emission and nitrogen pollution in natural and engineered ecosystems.

Abstract:The effects of the influent nitrate (NO 3 − -N) concentration on reactor performance and microbial communities of anaerobic methane oxidation coupled to denitrification in a membrane biofilm reactor were emphatically investigated after prolonged storage. The system was run for a period of 185 days including the storage period, reactivating process, and stable operation phase. The influent NO 3 − -N concentrations varied from 10, 20, and 30 mg/L, and the reactor was continuously operated for 135 days. The denitrification rate in the reactor was activated first and then inhibited, with the average value of NO 3 − -N consumed of 0.42 mg/L·h −1 at influent NO 3 − -N of 20 mg/L; there was no obvious nitrite and ammonia–nitrogen accumulation in the whole stage. Illumina MiSeq sequencing results showed that increases in NO 3 − -N loading could improve the microbial abundance in the system. The highest value for variety and the lowest value for the evenness of the microbial community corresponded to the ideal value of 20 mg/L. It speculated that promoting biofilm bacteria (PBB) became the key functional bacterial group after prolonged storage. Besides, the abundance of methane-oxidizing bacteria was significantly increased in synergy with PBB and denitrifiers to achieve reactivating and stable operation of anaerobic methane oxidation coupled to denitrification.

Abstract:Methane (CH 4 ) and nitrous oxide (N 2 O) are two important greenhouse gases (GHGs), and it is very important to achieve the reduction of both gases in the case of increasingly serious climate change. The study confirms the simultaneous mitigation of potent GHGs N 2 O and CH 4 in denitrifying anaerobic methane oxidation (DAMO) enrichment, supplementing the understanding that DAMO microorganisms do not produce and reduce N 2 O. We observed rapid accumulation and consumption of N 2 O in DAMO cultures subjected to high nitrate and nitrite loadings. Moreover, when N 2 O served as the sole electron acceptor, a significant correlation was found between the flux of N 2 O reduction and CH 4 oxidation. Inhibition of the particulate methane monooxygenase (pMMO) led to a notable decrease of 95 % in N 2 O reduction and 50 % in CH 4 oxidation. Metagenomic analysis revealed that Candidatus Methanoperedens and Candidatus Methylomirabilis were the dominant genera, with all functional genes related to denitrification and methane oxidation identified, including NO reductase ( nor ) of Ca. Methylomirabilis. Transcriptomic analysis further confirmed that the transcripts of nor were one order of magnitude higher than other functional genes. These results indicate that when the substrate NO 2 - surpasses the dismutation capacity of NO dismutase ( nod ), DAMO bacteria can alternatively induce nor to reduce NO to N 2 O. However, considering that N 2 O reductase ( nosZ ) is still missing in Ca . Methylomirabilis, the partner Methyloversatilis may be responsible for N 2 O reduction. Our results provide new insights into the link between GHG emissions in DAMO systems under anaerobic conditions.

Abstract:Simultaneous removal of ammonium and nitrate was achieved in a methane-fed moving bed biofilm reactor (MBBR). In the reactor, methanotrophic microorganisms oxidized methane under hypoxic conditions likely to methanol, hence providing an electron donor to denitrifiers to reduce nitrate to nitrite that then allowed anaerobic ammonium oxidizing bacteria (Anammox) to remove excess ammonium as N2. The ammonium and nitrate removal rates reached 72.09 ± 5.81 mgNH4+-N/L/d and 62.61 ± 4.17 mgNO3--N/L/d when the MBBR was operated in continuous mode. Nitrate removal by the methane-fed mixed consortia was confirmed in a batch test revealing a CH4/NO3- molar removal ratio of 1.15. The functional populations were unveiled by FISH analysis and 16S rRNA gene sequencing, which showed that the biofilm was dominated by Anammox bacteria (Candidatus Kuenenia) and diverse taxa associated with the capacity for denitrification: aerobic methanotrophs (Methylobacter, Methylomonas, and unclassified Methylococcaceae), methylotrophic denitrifiers (Opitutaceae and Methylophilaceae), and other heterotrophic denitrifiers (Ignavibacteriaceae, Anaerolineaceae, Comamonadaceae, Rhodocyclaceae and Thauera). Neither DAMO archaea nor DAMO bacteria were found in the sequencing analysis, indicating that more unknown community members possess the metabolic capacity of methanotrophic denitrification.

Abstract:The effect of dissolved oxygen (DO) on heterotrophic denitrification using poly(butylene succinate) as the carbon source and biofilm carrier was evaluated in a lab-scale experiment. Aerated, low-oxygen, and anoxic treatment groups were set up, which had average DO concentrations of 5.2±1.0, 1.4±1.2, and 0.5±0.3 mg L(-1), respectively. The NO3(-)-N and total nitrogen (TN) removal rates in the aerated group (37.44±0.24 and 36.24±0.48 g m(-3) d(-1), respectively) were higher than those in the other two groups. There was no significant difference between the low-oxygen and anoxic groups for the NO3(-)-N or TN removal rate. Accumulation of NO2(-)-N reached 5.0 mg L(-1) in the aerated group; no nitrite accumulation was found in the other two treatment groups. Bacterial communities of the low-oxygen and anoxic groups showed high similarity and were significantly different from those of the aerated group.
