Abstract:In this paper, a fluorescent sensing method based on functional CdSe/ZnS quantum dots (QDs) was investigated for rapid detection of acrylamide. The functional QDs were prepared by binding them with N-acryloxysuccinimide (NAS). Then NAS-QDs complexes were mixed with acrylamide solutions, and polymerization occurred in the presence of photo-initiator under UV irradiation. The reaction increased the distance between QDs, leading to the increase of fluorescence intensity compared with that no acrylamide existed. The detection limit and linear range using this method were 5 x 10(-7) mol L-1 and 5 x 10(-7) to 1 x 10(-2) mol L-1, respectively. Compared with LC-MS/MS method, this method could be applied to food detection with similar sensitivity and selectivity but less time and cost.

Abstract:Background: Maillard reaction involves the polymerization, condensation, and other reactions between compounds containing free amino groups and reducing sugars or carbonyl compounds during heat processing. This process endows unique flavors and colors to food, while it can also produce numerous hazards. Acrylamide (AAm) is one of Maillard's hazards with neurotoxicity and carcinogenicity, these effects can trigger mutations and alternations in gene expression in human cells and accelerate organ aging. An accurate and reliable acrylamide detection method with high sensitivity and specificity for future regulatory activities is urgently needed. Results: Herein, we constructed a colorimetric aptasensor with the hybridization of MIL-glucose oxidase (MGzyme)-cDNA and magnetic nanoparticle-aptamer (MNP-Apt) to specifically detect AAm. The incorporation of MB-Apt and AAm released MGzyme-cDNA in the supernatant, took the supernatant out, with the addition of glucose and TMB, MGzyme would oxidize glucose, the resulting center dot OH facilitated the oxidation of colorless TMB to blue ox-TMB. The absorbance value at 652 nm, which indicates the characteristic absorption peak of ox-TMB, exhibited a proportion to the concentration of AAm. MGzyme avoided the addition of harmful intermediate H2O2 and created an acid microenvironment for the catalytic reaction. MNP-Apt possessed the advantages of high specificity and simplified separation. Under optimal conditions, this method displayed a linear range of 0.01-100 mu M with the limit of detection of 1.53 nM. With the spiked analysis data cross-verified by ELISA kit, this aptasensor was proven to specifically detect AAm at low concentrations.
Development of bacterial biosensor for sensitive and selective detection of acetaldehyde.

Abstract:Acetaldehyde is a human carcinogen and widely existed in alcoholic beverages and polluted air. In this study, a simple, fast, convenient and sensitive acetaldehyde biosensor was developed based on an acetaldehyde dehydrogenase (AldDH) bacteria surface display system. The whole-cell catalyst facilitated the dehydrogenation of acetaldehyde, while coenzyme NAD+ was reduced and the resultant NADH can be detected spectrometrically at 340 nm. The correct location of AldDH on the bacteria surface was confirmed by the subcellular fraction and immunofluorescence analysis. By comparing the fusion protein expression level and whole-cell activity, the proper display system for anchoring AldDH on the cell surface was obtained. The results of kinetics analysis towards both surface-displayed AldDH and intracellular expressed AldDH demonstrated that the mass-transport resistance was dramatically alleviated by cell-surface display strategy. Under optimal conditions, AldDH-surface display strain with the highest whole-cell activity (3.41 ± 0.3 mU/OD600) was applied to spectrophotometry acetaldehyde detection system. An excellent linear relationship between the increases of absorbance at 340 nm and acetaldehyde concentration over the range from 1 μM to 300 μM was reached. The proposed approach offered adequate sensitivity for the detection of acetaldehyde at 0.33 μM. Most importantly, the developed biosensor showed the narrowest substrate specificity towards acetaldehyde, which has been employed for quick determination of acetaldehyde in real samples with good accuracy. The total detection time was within 20 min. The method reported here provided a simple, rapid, and low-cost strategy for the sensitive and selective measurement of acetaldehyde. Therefore, genetically engineered cells may find broad application in biosensors and biocatalysts.
Enzyme-Induced Silver Deposition on Gold Nanorods for Naked-Eye and Smartphone Detection of Acrylamide in Food

Abstract:Acrylamide is a probable human carcinogen with a wide occurrence in commonly consumed food. Because of the lack of specific antibodies against acrylamide, immunoassays usually measure acrylamide derivates, which require long derivatization times at elevated temperatures. One exception is xanthylacrylamide (XAA), which can be derived from acrylamide at room temperature within 30 min. Here, we present the development of a monoclonal antibody against XAA and a competitive enzyme-linked immunosorbent assay (ELISA) based on the multicolor detection of gold nanorods (AuNRs). The biotinylated monoclonal antibodies bound streptavidin alkaline phosphatase, which induced XAA concentration-dependent silver deposition on the AuNRs. The resulting blue shift of the AuNR surface plasmon resonance peak translated into a strong gray-to-orange color change that could be conveniently distinguished by the naked eye or smartphone color detection. The assay showed a broad linear range (0.3–17.2 ng/mL), a low limit of detection (0.06 ng/mL), and a 3.6-fold sensitivity improvement compared to those of conventional tetramethylbenzidine-based ELISA. The analysis of several different food products showed excellent acrylamide concentration recoveries (84–102%) and confirmed its applicability as an effective tool for routine monitoring of acrylamide in food. Rapid and simple color analysis demonstrated a strong potential for implementation into direct consumer product testing.

Abstract:As one of Maillard's hazards, acrylamide (AAm) can induce neurotoxicity, mutagenicity, and genotoxicity, and it's urgent to construct a detection method for sensitive and specific monitoring of AAm. Herein, an aptamer-gated colorimetric sensor was developed to detect AAm in food. UiO-66-NH 2 was selected to combine acetylcholinesterase (AChE) and the aptamer of AAm to construct the aptamer-gated probe UAzyme-Apt. The addition of AAm would detach Apt and expose the active sites of UAzyme, resulting in the transformation of acetylthiocholine (ATCh) into thiocholine (TCh) that could inhibit the oxidase mimic activity of MIL-53 (Fe/Mn). Consequently, the concentration of AAm was inversely proportional to the absorbance at 652 nm, thus providing a qualitative and quantitative detection of AAm. The aptamer-gated probe can recognize AAm to improve specificity and stability and regulate the enzyme activity of UAzyme. This aptasensor enhances the detection methods of AAm and broadens the scope of applications for AChE inhibition detection, and targets are not limited to the substrate or organophosphorus pesticides. The linear range and limit of detection were 0.05–100 μM (R 2 = 0.992) and 9.94 nM, and spiked analysis revealed the efficiency and robustness of the developed assembly. The colorimetric aptasensor can specifically detect AAm at low concentrations, and will be a promising tool to ensure food safety in the coming future.

Abstract:In order to protect human health from the threat of excessive acrylamide in heat-processed foods, swift, uncomplicated, highly selective and sensitive online monitoring methods for acrylamide are urgently needed. In this study, a rare earth metal/transition metal-organic frameworks Eu/Zr-MOFs fluorescence sensor with dual-band luminescence was prepared by the hydrothermal synthesis for detection of acrylamide in fried potatoes and cookie crumbs. This sensor exhibited two different fluorescence emission peaks at 430 and 615 nm under excitation of 365 nm, and the ratio of fluorescence intensity was linear with the experimental concentration of acrylamide in the range of 1 × 10−4 to 1 × 10−3 M. Under the optimal conditions, the proposed fluorescence analysis method showed that the detection limit of acrylamide was as low as 2.4 × 10−8 M, and the satisfactory recoveries of 94.05–104.04% were obtained for various spiked samples. More importantly, the sensor has ultra-high selectivity for the detection of acrylamide while other small organic molecules hardly interfere with it in the measurement environment. These studies confirmed that the ratiometric fluorescence sensing method was expected to achieve on-line rapid detection of acrylamide in real foods. In addition, the mechanism of Eu/Zr-MOFs sensing acrylamide was analyzed and discussed in depth, and the analog computation was carried out by Materials Studio software, and the entire sensing system was studied at the micro level in combination with density functional theory.

Abstract:Acrylamide is a neurotoxin and potential carcinogen, but is found in various thermally processed foods such as potato chips, biscuits, and coffee. Simple and sensitive methods for on-line detection of acrylamide are needed to ensure food safety. In this paper, a novel fluorescent sensing method based on acrylamide polymerization-induced distance increase between quantum dots (QDs) was proposed for detecting acrylamide in potato chips. The functional QDs were prepared by their binding with N-acryloxysuccinimide (NAS), which was characterized by Fourier transform infrared (FR-IR) spectra. The carbon–carbon double bonds of NAS modified QDs polymerized with assistance of photo initiator under UV irradiation, leading to QDs getting closer along with fluorescence intensity decreasing. Acrylamide in the sample participated in the polymerization and induced an increase of fluorescence intensity. This method possessed a linear range from 3.5×10−5 to 3.5gL−1 (r2=0.94) and a limit of detection of 3.5×10−5gL−1. Although the sensitivity and specificity cannot be compared with standard LC-MS/MS analysis, this new method requires much less time and cost, which is promising for on-line rapid detection of acrylamide in food processing.

Abstract:Acrylamide (AA), a detrimental aliphatic amide, is generated during food thermal processing as a result of the Maillard reaction occurring between asparagine and reducing sugars. WaveFlex fiber is a fiber-optic flexible sensor designed to enhance the evanescent wave (EW) for effectively exciting the localized surface plasmon resonance (LSPR) phenomenon, and in this study, the authors use this type of sensor to detect varying levels of AA. By immobilizing gold nanoparticles (AuNPs) on the probe's surface, the evanescent field has the capability to trigger the activation of AuNPs and induce the occurrence of LSPR phenomenon. In order to excite LSPR better and generate a stronger evanescent field, the WaveFlex biosensor is fabricated using a single-mode fiber (SMF). Cerium oxide nanorods (CeO2-NRs) and MXene are used to modulate the LSPR phenomenon by altering the dielectric environment of AuNPs. As a result, the sensing performance of the sensor is improved and the surface area of the fiber probe is increased, providing an additional attachment point for the AA antibody. The AA sensor's specific selectivity is enhanced by functionalizing the surface of the sensing probe with AA antibody. The sensor has a linear range of 3– g/mL with a detection limit of g/mL. Finally, the recoveries of AA in biscuits and chips are 82.8% % and 83.7% %, respectively. Furthermore, the probe's potential for detecting AA levels is demonstrated through rigorous testing of its reusability, reproducibility, stability, pH, and selectivity. The obtained results indicate a promising application scope for this WaveFlex biosensor.

Abstract:Acrylamide (AA), an odorless and colorless organic small-molecule compound found generally in thermally processed foods, possesses potential carcinogenic, neurotoxic, reproductive, and developmental toxicity. Compared with conventional methods for AA detection, bio/chemical sensors have attracted much interest in recent years owing to their reliability, sensitivity, selectivity, convenience, and low cost. This paper provides a comprehensive review of bio/chemical sensors utilized for the detection of AA over the past decade. Specifically, the content is concluded and systematically organized from the perspective of the sensing mechanism, state of selectivity, linear range, detection limits, and robustness. Subsequently, an analysis of the strengths and limitations of diverse analytical technologies ensues, contributing to a thorough discussion about the potential developments in point-of-care (POC) for AA detection in thermally processed foods at the conclusion of this review.

Abstract:Herein, a sensitive and simple tag-free aptasensor was employed for acrylamide determination via the fluorescent dye SYBR Green I (SGI) with the role of signal indicator. The foundation of this platform was built upon the establishment of a four-way junction structure, which resulted from the hybridization between terminus regions of aptamer and complementary (CP) strands. The preferential interaction of the aptamer sequence with acrylamide led to the disintegration of the four-way junction structure into the single strands, and consequently, a significant decrease in the output fluorescence signal. Under the optimal conditions, the proposed aptasensor was shown to detect acrylamide, with low detection limit (LOD) of 0.28 μmol/L in the linear dynamic range of 1 to 60 μmol/L with high selectivity. The aptasensor possesses remarkable proficiency in detecting acrylamide levels within coffee samples, rendering it as a highly effective tool for ensuring the safety and quality control of food products.

Abstract:The ferric ion-polydopamine coordination hedgehog-like superstructure (hedgehog-like Fe-PHS) was fabricated using the metal–ligand crosslinking strategy, which was used as a colorimetric sensor for acrylamide (AA) visual detection. The hedgehog-like Fe-PHS exhibited outstanding peroxidase-like activity toward the decomposition of H 2 O 2 and production of hydroxyl radical (OH), and oxidized colorless 3,3′,5,5′-tetramethylbenzidine (TMB) to blue oxTMB. The TMB oxidation was quenched by glutathione and then recovered via a thiol-ene Michael addition reaction between glutathione and AA. The increase in AA concentrations accelerated the TMB oxidation and recovered the dark blue color. Thus, the hedgehog-like Fe-PHS was used as a colorimetric sensor for detecting AA. A wide linear range from 0.75 to 36.00 μM ( R 2 = 0.9950) and a low limit of detection of 0.27 μM with excellent selectivity were obtained. AA was visually detected with "naked eye" using hedgehog-like Fe-PHS. Meanwhile, the hedgehog-like Fe-PHS colorimetric sensor was successfully applied for detecting AA in thermally processed foods, and its practical feasibility was verified using the HPLC method.

Abstract:The residual acrylamide in food paper packaging can be transferred into water and food, which will cause harmful effects on human beings. In this paper, a rapid and easily available fluorescent probe based on carboxymethyl cellulose (CMC)/Eu (III) nanocomposites was designed to detect the residue acrylamide with high sensibility. The probe could respond in 1 min. The concentration of acrylamide was linearly correlated to the fluorescence intensity of the probe at the emission wavelength of 615 nm in the concentration range of 0.1–100 μmol/L. The limit of detection (LOD) of the probe was 0.085 μg/L, which is lower than the guideline value of the European Union, the U.S. EPA, and the WHO. An experiment was performed to simulate the acrylamide migrating from food-contacting paper materials to different foods, including waterborne food, alcohol beverage, acidic food, and greasy food. The recoveries and RSDs of acrylamide in all samples indicated that the CMC/Eu (III) fluorescent probe was efficient for acrylamide detection. The possible mechanism of the probe for acrylamide detection involved both dynamically quenching and static quenching by forming of non-fluorescent substances.

Abstract:A portable colorimetric aptasensor was developed on the surface of a plastic sheet for on-site detection of acrylamide. The mechanism of aptasensor is based on the disruption of the aptamer complex with its complementary strand on the sensing zone, and subsequently, the catalytic activity of gold nanoparticles (AuNPs) for the reduction process of 4-nitrophenol (4-NP) in the presence of sodium borohydride (NaBH4). A yellow-to-colorless change of the sample solution revealed the target presence, easily discernible by the naked eye. The acrylamide quantification was accomplished using the smartphone imaging readout technique. The aptasensor detected the acrylamide concentration in the range of 0.01-500 nmol L-1 with a detection limit of 0.0024 nmol L-1. Coffee, potato chips, bread, and lake water samples were successfully analyzed by the aptasensor for their acrylamide content. The introduced aptasensor can pave a facile, cost-effective, portable, and user-friendly sensing tool for food safety control and environmental monitoring.
Antigen synthetic strategy and immunoassay development for detection of acrylamide in foods.

Abstract:Acrylamide, a toxic and carcinogenic compound, has been found to be present in a range of processed starchy foods. To prepare an effective immunogen compound for acrylamide, N-acryloxysuccinimide (NAS) was conjugated to bovine serum albumin (BSA) at a high molar ratio of 21.2 : 1. Antisera were obtained by immunization of rabbits with additional booster injections of the NAS-BSA conjugate after the regular process. The IgGs purified by an ammonium sulfate precipitation method were further fractionated with a BSA-immobilized immunoaffinity column. The affinity constant between the collected antibody and coated antigen (NAS-ovalbumin) is found to be 6.7 x 10(7) L mol(-1). Asparagine, the key precursor of acrylamide formation in foods, showed negligible cross-reactivity to the antibody. A biotin-avidin enzyme-linked immunosorbent assay (BA-ELISA) was developed and the optimum assay medium was found to be 0.1 mol L(-1) NaHCO(3) (pH 8.3, containing 0.5 mol L(-1) NaCl). The BA-ELISA afforded a practical sensitivity with a working range of 10-100 000 ng mL(-1) and a detection limit of 6 ng mL(-1). The assay was applied to detect acrylamide in potato fries and biscuits and the quantitative results were in good agreement with those obtained by the high-performance liquid chromatography method. This immunoassay will be very useful for monitoring acrylamide in food samples.

Abstract:Acrylamide (AA), a neurotoxin and potential carcinogen, has been found in various thermally processed foods such as potato chips, biscuits, and coffee. LC–MS/MS and GC–MS as standard detection methods show high sensitivity, selectivity, stability, and repeatability. However, these methods require expensive instruments, skilled technicians in laboratories, and high testing costs, and cannot meet the needs for real-time and on-line detection of AA in foods. Therefore, rapid detection methods with merits of simplicity and portability such as computer vision, ELISA, electrochemical biosensing, and fluorescent biosensing have obtained an increasing amount of attention. Reported research on rapid methods has shown similar sensitivity and selectivity, but requires less time and cost in comparison with standard methods through the use of nanomaterials and biomolecules with high affinity to AA. These improvements show great promise for high-throughput, real-time, and on-line detection of AA. This paper provides a comprehensive overview of rapid detection methods for AA in foods with comparison between rapid and standard methods. Meanwhile, suggestions for further research on rapid methods for detecting AA are also discussed based on technical challenges and industry needs.

Abstract:Abstract Acrylamide (AA) is a carcinogenic, neurotoxin, and pregnancy‐harmful agent produced in food thermal processing. A specified excellent analytical technique performance is desired for detecting low AA concentration for human protection from its adverse impacts. A photoluminescent sensor APTES capped zinc oxide quantum dots (APTES/ZnO QDs) has been synthesized for AA quantification. Different capping agent's effects, for instance, triethanolamine (TEA), oleic acid (OA), tetraethyl orthosilicate (TEOS), polyvinyl pyrrolidone (PVP), 3‐aminopropyl triethoxysilane (APTES), and ethyl acetate (EA) on ZnO QDs are examined. The photoluminescence (PL) response of pristine ZnO QDs and APTES/ZnO QDs probe toward AA concentrations is investigated. This developed sensor exhibits selectivity as well as sensitivity for detecting AA ranging from 0.01 to 8.0 mM with a strong correlation coefficient (R 2 ) of 0.9998 and a limit of detection (LOD) of 0.0019 mM, which is lesser than the WHO and European Union's guideline value. The possible photoluminescence enhancement mechanism of the APTES/ZnO QDs probe for AA detection is studied. Additionally, the reliability and performance of this facile, rapid, and precise analytical technique for quantifying AA in potato chip samples is confirmed and revealed satisfactory recoveries from 99.30 to 101.69% with relative standard deviations (RSD) from 1.4 to 1.6%.

Abstract:In this review, the mechanisms of acrylamide formation in food, along with aspects related to its toxicity and associated consumption risks, are investigated, highlighting the potential impact on human health. European regulations regarding acrylamide content in food products are also addressed, emphasizing the importance of monitoring and detecting this substance in nutrition, by public health protection measures. The primary objective of the research is to explore and analyze innovative methods for detecting acrylamide in food, with a particular focus on electrochemical biosensors. This research direction is motivated by the need to develop rapid, sensitive, and efficient monitoring techniques for this toxic compound in food products, considering the associated consumption risks. The research has revealed several significant results. Studies have shown that electrochemical biosensors based on hemoglobin exhibited increased sensitivity and low detection limits, capable of detecting very low concentrations of acrylamide in processed foods. Additionally, it has been found that the use of functionalized nanomaterials, such as carbon nanotubes and gold nanoparticles, has led to the improvement of electrochemical biosensor performance in acrylamide detection. The integration of these technological innovations and functionalization strategies has enhanced the sensitivity, specificity, and stability of biosensors in measuring acrylamides. Thus, the results of this research offer promising perspectives for the development of precise and efficient methods for monitoring acrylamides in food, contributing to the improvement of food quality control and the protection of consumer health.

Abstract:According to the classification of the International Agency for Research on Cancer (IARC), acrylamide is a compound in "probably carcinogenic for humans" class (Group 2A). Acrylamide is produced through the reactions induced by heating food within the amino group asparagine and the carbonyl group decreasing sugars along side the thermal treatment of initial Maillard reaction products. The purpose of this work is to present and enhance an innovative technique for acrylamide determination in potato chips through CdTe surface-functionalized quantum dots as a reagent for the determination of acrylamide by fluorescence spectroscopy. For this purpose, fluorescence emission spectroscopy was used instead of conventional HPLC/GC methods and data of merit of the suggested technique were assessed. The acrylamide quantity in 4 potato chips specimens, prepared from a local market in Tehran, was determined through utilizing the suggested process. Effective parameters in the process were optimized using the one-factor-at-a-time (OFAT) technique. The optimal quantities of effective parameters such as pH absorption, temperature, and emission wavelength were determined. Comparison between two methods, namely HPLC and fluorescence spectroscopy, was also described. The merit figures for the suggested approach were within the idyllic range. The developed methods showed a high correlation coefficient (0.991), high sensitivity, and repeatability. Results of the fluorescence emission spectroscopy and its comparison with Mass/HPLC revealed the high reliability and performance of the recommended method as an efficient, simple, and quick procedure with reduced cost and time in the determination of acrylamide in potato chip specimens.

Abstract:A highly sensitive quenching molecular imprinting (MIP) photoelectrochemical (PEC) sensor was proposed to detect acrylamide (AM) by using the photoactive composite of ZnO and polypyrrole (PPy) as the PEC signal probe. ZnO, with high electron mobility, excellent chemical and thermal stability as well as good biocompatibility, was selected as the photoelectrically active material. A polypyrrole film was formed on the nanodisk ZnO by electrochemical polymerization, and the recognition site of AM was left on the surface of the PPy film by elution, enabling the specific detection of AM. The transfer of electrons will be hindered when AM is adsorbed on the ZnO/PPy nanocomposites surface, which results in the decrease of photocurrent signal. The proposed molecularly imprinted PEC sensor exhibits significant detection performance of AM in the range of 10−1 M-2.5 × 10−9 M with a LOD of 2.147 × 10−9 M (S/N = 3). The use of photoelectrochemical technology combined with molecular imprinting technology enables the PEC sensor to have excellent selectivity, superior repeatability, preferable stability, low cost, and easy construction, providing a new method for the detection of AM. The high recovery rate in the detection of real samples of potato chips and biscuits indicates that the proposed PEC sensor has potential in monitoring the emerging food safety risks.
