Abstract:In order to find new drugs with potent antiproliferative effect, a series of novel barbituric acid derivatives containing azoles at the C-5 position were designed, synthesized, and evaluated for antiproliferative activity against three human cancer cell lines (BEL-7402, MCF-7, and HCT-116) using MTT assay. Several of the synthesized compounds exhibited potent antiproliferative effects. The most promising compound was 5-((1-(4-(trifluoromethyl)phenyl)-1H-1,2,3-triazol-4-yl) methylene)pyrimidine-2,4,6(1H,3H,5H)-trione (3s), which showed considerably high antiproliferative activity in the BEL-7402 cell line, with a half-maximal inhibitory concentration of 4.02 µM and 20.45-fold higher selectivity for BEL-7402 cells than for normal L02 cells. The apoptosis experiment showed that compound 3s induced apoptosis and cell necrosis in a concentration-dependent manner and exert its anti-proliferative activity. Therefore, compound 3s exhibited better therapeutic activity and specificity compared with the positive control 5-fluorouracil.

Abstract:A novel class of acetylhydrazone derivatives (5a-x) containing 2-(phenylthiomethyl)-1H-benzo-[d]-imidazole moieties are synthesizer, and their antitumor activities against A549, HCT116, HepG2, PC-9, and A375 were determined by the MTT assay. Among them are N-(2,4-dihydroxybenzylidene)-2-(2-(phenylthiomethyl)-1H-benzo[d]-imidazol-1-yl)acetohydrazide (5a) and N-(5-bromo-2-hydroxy-benzylidene)-2-(2-(phenylthiomethyl)-1H-benzo[d]-imidazol-1-yl)acetohydrazide (5d) which displayed excellent cancer inhibitory activity against the tested cancer cells (IC(50) 4-17 μM), compared with 5-FU and SU11248. The others have moderate to weak inhibitory activity against the tested cancer cell lines.

Abstract:New phosphorus-containing isatin-3-hydrazones were synthesized by the condensation reaction of 5-substituted isatins with 2-chloroethyl [4-(dimethylamino)phenyl](2-hydrazinyl-2-oxoethyl)phosphinate (KAPAKh). The resulting compounds were shown to have high antitumor activity against duodenal adenocarcinoma (HuTu80) and cervical epithelioid carcinoma (M-HeLa) cell lines with low toxicity towards erythrocytes and normal human liver cells.

Abstract:A series of new isatin-3-hydrazones bearing different ammonium fragments was synthesized by a simple and easy work-up reaction of Girard's reagents analogs with 1-(3,5-di-tert-butyl-4-hydroxybenzyl)isatin. All derivatives have been shown to have antioxidant properties. In terms of bactericidal activity against gram-positive bacteria, including methicillin-resistant strains of Staphylococcus aureus, the best compounds are 3a, 3e, and 3m, bearing octyl, acetal, and brucine ammonium centers, respectively. In addition, brucine and quinine derivatives 3l, and 3j exhibit platelet antiaggregation activity at the level of acetylsalicylic acid, and this series of isatin derivatives does not adversely affect the hemostasis system as a whole. Thus, all the obtained results can lay the groundwork for future pharmaceutical developments for the creation of effective antibacterial drugs with reduced systemic toxicity due to the presence of antioxidant properties.

Abstract:Seventeen new flavone derivatives substituted at the 4′-OH position were designed, synthesized and evaluated for their anticancer and antibacterial activities. Among them, compounds 3, 4, 6f, 6e, 6b, 6c and 6k demonstrated the most potent antiproliferative activities against a human erythroleukemia cell line (HEL) and a prostate cancer cell line (PC3). The results also showed that the IC50 value of compounds 3, 4, 6f, 6e, 6b, 6c and 6k were close to that of the anticancer drug cisplatin (DDP) and lower than that of apigenin. All of the derivatives did not present antibacterial activities. The structure–activity relationships evaluation showed that the configuration of methyl amino acid might affect their biological activities.

Abstract:A series of novel mono and bishydrazones each bearing a 2-oxindole moiety along with substituted phenylaminopropanamide, pyrrolidin-2-one, benzimidazole, diphenylmethane, or diphenylamine fragments were synthesized, and their anticancer activities were tested by MTT assay against human melanoma A375 and colon adenocarcinoma HT-29 cell lines. In general, the synthesized compounds were more cytotoxic against HT-29 than A375. 3-((4-Methoxyphenyl)(3-oxo-3-(2-(2-oxoindolin-3-ylidene)hydrazinyl)propyl)amino)-N′-(2-oxoindolin-3-ylidene)propanehydrazide and (N′,N′′′)-1,1′-(methylenebis(4,1-phenylene))bis(5-oxo-N′-(2-oxoindolin-3-ylidene)pyrrolidine-3-carbohydrazide) were identified as the most active compounds against HT-29 in 2D and 3D cell cultures. The same compounds showed the highest antioxidant activity among the synthesized compounds screened by ferric reducing antioxidant power assay (FRAP). Their antioxidant activity is on par with that of a well-known antioxidant ascorbic acid.

Abstract:A large series of 2-arylchromen-4-ones containing from 1 to 3 fluorine atoms or a trifluoromethyl group in the structure was synthesized by condensation of fluorinated 2-hydroxyacetophenones with benzaldehydes in an alkaline medium and subsequent oxidative cyclization of the resulting 2'-hydroxychalcones by action of I2 in DMSO. The cytotoxicity of the obtained compounds was studied in glioblastoma cell line, SNB19, and in a monkey-derived normal kidney epithelium cell line, Vero. In addition, antiglycation activity of the obtained compounds was evaluated. The inhibitory activity of some fluorinated 2-arylchromen-4-ones against acetylcholinesterase, butyrylcholinesterase and carboxylesterase as well their primary antioxidant activity in ABTS and FRAP tests were investigated. Screening of the synthesized compounds for their inhibitory activity against influenza A virus A/Puerto Rico/8/34 (H1N1) in the MDCK cell culture revealed that fluorinated compounds 32, 31 and 39 showed manifest antiviral effects (with IS = 57, 38 and 25 correspondingly) that makes this series of new biologically attractive fluorinated heterocycles promising for further development and in-depth study.

Abstract:A new series of imidazolyl hydrazone derivatives IA ( 1-12 ) were prepared from a condensation reaction between indoline-2,3-dione (isatin) and 2-benzylidenehydrazinecarboximidamide derivatives. The structure of compounds was elucidated using various spectral techniques including FTIR, 1 H NMR, 13 C NMR, and HRMS. The proposed structure of IA-2 was determined by single-crystal X-ray analysis. Synthesized compounds were evaluated for their inhibitory action against carbonic anhydrase I and II isoenzymes (hCA I and hCA II), as well as cytotoxicity activity in a cancer cell line (HT-29) and a healthy cell line (NIH 3T3). Among them, some compounds exhibited remarkable CA inhibitory activities compared to a standard inhibitor with Ki values in the range of 13.434 ± 3.278-522.549 ± 360.720 nM for hCA I (Ki value for standard inhibitor = 271.15 ± 74.620 nM) and 41.108 ± 10.180-271.171 ± 65.293 nM for hCA II (Ki value for standard inhibitor = 113.07 ± 20.980 nM) and significant antiproliferative activity with less toxicity to a health cell line. In addition, the theoretical parameters of the bioactive molecules were calculated to establish their drug-likeness qualities and ADME/T analysis was carried out to examine the drug properties of the synthesized compounds.

Abstract:An approach to the synthesis of phosphoryl substituted spiro-1,3-dioxolane oxindoles was developed from the base-catalyzed reaction of various isatins with (3-hydroxyprop-1-yn-1-yl)phosphonates. It was found that various aryl-substituted and N-functionalized isatins with the formation of appropriate products with high yields and stereoselectivity when using t-BuOLi are able to react. Cytotoxic activity evaluation suggests that the most significant results in relation to the HuTu 80 cell line were shown by N-benzylated spirodioxolanes. 5-Cloro-N-unsubstituted spirooxindoles exhibit antiaggregational activity exceeding the values of acetylsalicylic acid.

Abstract:A new series of hydrazone derivatives 4 a–l incorporating spirocyclic pyrimidine with aryl alkanesulfonates were synthesized, and their antiproliferative activities were tested in vitro against NCI‐60 cell panel. Three compounds showed good activities against different kinds of cancers. A new series of hydrazone derivatives incorporating spirocyclic aminopyrimidine with aryl alkanesulfonates were synthesized. The targeted products were obtained in excellent yields via an acid catalyzed reaction of spirocyclic hydrazinopyrimidine with aryl aldehyde sulfonates. The structures of the synthesized hydrazones were studied by the different spectroscopic tools, in addition to the high resolution mass analysis. The antiproliferative activities of the compounds were tested in vitro against the National Cancer Institute (NCI) 60 cell panel by the Developmental Therapeutics Program. Amongst the tested hydrazone compounds, the methansulfonate derivatives showed the highest activities against lung, brain, kidney, and breast cancers.

Abstract:目的分析CD20在成人急性B淋巴细胞白血病（B-ALL）患者中的表达及CD20阳性B-ALL患者的临床特点和预后。方法回顾性分析郑州大学附属肿瘤医院血液科2007年5月至2011年7月收治的168例初治成人B-ALL患者，其中CD20阳性58例，阴性110例。对比分析两组患者的临床特点和生存情况。结果 CD20阳性与阴性组在性别、年龄分布及贫血、血小板减少、肝脾或淋巴结肿大、髓外浸润、Ph染色体阳性、髓系抗原表达、早期死亡、高危患者比例方面差异均无统计学意义（P值均>0.05），CD20阳性组患者初诊时中位白细胞计数（19.2×109/L）和高白细胞计数患者比例（37.9%）明显高于CD20阴性组（6.93×109/L和20.9%），差异有统计学意义（P值均<0.05）；两组诱导缓解治疗4周完全缓解率分别为78.6%和76.9%，差异无统计学意义（P>0.05），3年累积复发率分别为59.3%和42.5%，差异有统计学意义（P<0.05）。多因素分析显示CD20阳性是影响患者总生存（P=0.004）和疾病无进展生存（P=0.001）的独立预后不良因素。结论 CD20阳性表达与成人B-ALL患者外周血白细胞计数相关，且CD20阳性者预后不良。
Novel fluorinated thiazolidin-4-one derivatives: synthesis and anti-cancer potential against HepG2 and HCT116 cell lines

Abstract:A novel synthetic route has been designed to introduce fluorine functionality into a series of compounds containing thiazolidin-4-one rings. These compounds were synthesised from various aniline derivatives using a two-step approach: an addition reaction of ethyl isothiocyanate with different aromatic fluorinated anilines, followed by cyclisation to yield the final products. A total of 15 novel fluorinated thiazolidinone compounds were synthesised and characterised using 1 H NMR, 19 F NMR, Fourier transform–infrared, elemental analysis and liquid chromatography–mass spectrometry. Stereochemistry around the imine bond in the synthesised derivatives was determined using nuclear Overhauser effect spectroscopy. The in vitro anticancer potential of the compounds was tested against two human cancer cell lines, liver (HepG2) and colon (HCT116). The study revealed that the derivatives having fluorine functionality at both the m- positions in the aromatic ring showed promising anticancer potential, as compared to those at o- and p- positions.

Polycyclic Aromatic Compounds

Abstract:In this work, three N' -arylidene-5-phenyl- 1H -pyrazole-3-carbohydrazide ( 3a - 3c ), derivatives have been synthesized and fully characterized by IR, 1 H & 13 C NMR and ESI-MS using experimental and theoretical methods. Theoretical calculations were carried out by DFT/B3LYP method using 6-311++G(d,p) basis set. For theoretical IR, 1 H & 13 C NMR (with GIAO method), MEP, HOMO-LUMO energies analyses of compounds 3a-3c were performed over the optimized structures. All newly synthesized compounds were screened for their cytotoxicity, proylenpeptidase and α-glucosidase inhibitory activities. Compounds 3c showed significant anticancer activity against lung cancer cell line (H460) with the IC 50 value 0.15 ± 0.01 μM. The other compounds exhibited prolyl endopeptidase (PEP) activity. Compounds 3a and 3c have demonstrated a potent α-glucosidase inhibitory activity with the IC 50 value 360.4 ± 0.7 μM and 370.3 ± 1.17 μM, respectively. Furthermore, in-silico based molecular docking study was performed between the title ligands and lung cancer cell line-H460 protein: 1M17. The best affinity bond was observed between compound 3c and the protein 1M17 with an energy of −8.6 (kcal/mol), an inhibition constant of 0.496769 μM and an active hydrogen bond to three.

Synthesis, characterization, and biological properties of novel Schiff bases containing pentafluorophenyl hydrazine

Abstract:In the present study, new Schiff bases derived from pentafluorophenyl-hydrazine (L1, L2, L3, L4) were synthesized and their structures were characterized by 1 H nuclear magnetic resonance spectroscopy (NMR), 13 C NMR, 19 F NMR, fourier transform infrared spectroscopy, and elemental analysis methods. Then, the anticancer activities of the obtained compounds were investigated using three human cancer cell lines (A2780, over; Caco-2, colon; and HT-29 colon carcinoma cell lines). According to the obtained cytotoxicity results, compound number L4 was found to have the highest anticancer activity in A2780 (over) and Caco-2 (colon) cell lines. Furthermore, in silico, ADMET properties, where studies play an important role in the development and prediction of drug compounds, were calculated using web-based platforms. In addition, molecular docking studies were performed to evaluate the binding interactions between the synthesized pentafluorophenyl-hydrazone compounds and the MDM2 protein (4JSC). Both in vitro and in silico results showed that the synthesized compounds could act as potent anticancer agents.

Abstract:Novel 2-imino-6-(aryldiazenyl)-2 H -chromene-3-carboxamides 6a–e , 2-amino-4 H -cyclopenta or benzo[ b ]thiophene-3-carboxamides 10a,b , 2,7-diaminopyrazolo[1,5- a ]pyrimidine-6-carboxamides 13a–e , pyrimidine-5-carboxamides 14 , 15 and 3-amino-1 H -pyrazole-4-carboxamide 16 were synthesized from the reaction of 2-cyano- N -(1,3-dihydroxy-2-(hydroxyl-methyl) propan-2-yl) acetamide 2 with 4-arylazosalicylaldehydes 5a-e , cyclopentanone and/or cyclohexanone, guanidine derivatives and hydrazine hydrate, respectively. Some new compounds were evaluated for antibacterial activity in vitro , and exhibited good efficacy compared to gentamicin. Compound 4c showed greater activity against gram negative bacteria ( Klebsiella pneumonia and Pseudomonas aeruginosa ) than standard antibiotic. Compound 4c with two withdrawing groups also showed the higher activity (38.7 ± 0.6) against fungi ( Candida albicans ) than the Nystatin (20 ± 0.5). On the other hand, compounds 13a , 13c, and 13e have strong cytotoxic activity among the tested compounds in the three selected cancer cell lines (HePG2, MCF7 and Hela). Physicochemical characterization by Swiss ADME predication was also performed for some synthesized compounds exhibiting better biological and antimicrobial properties.

Abstract:A series of novel hybrid compounds containing benzofuroxan and 2-aminothiazole moieties are synthesized via aromatic nucleophilic substitution reaction. Possible reaction pathways have been considered quantum-chemically, which allowed us to suggest the most probable products. The quantum chemical results have been proved by X-ray data on one compound belonging to the synthesized series. It was shown that the introduction of substituents to both the thiazole and amine moieties of the compounds under study strongly influences their UV/Vis spectra. Initial substances and obtained hybrid compounds have been tested in vitro as anticancer agents. Target compounds showed selectivity towards M-HeLa tumor cell lines and were found to be more active than starting benzofuroxan and aminothiazoles. Furthermore, they are considerably less toxic to normal liver cells compared to Tamoxifen. The mechanism of action of the studied compounds can be associated with the induction of apoptosis, which proceeds along the mitochondrial pathway. Thus, new hybrids of benzofuroxan are promising candidates for further development as anticancer agents.
