Abstract:Nitrogen (N) deposition is a significant aspect of global change and poses a threat to terrestrial biodiversity. The impact of plant-soil microbe relationships to N deposition has recently attracted considerable attention. Soil microorganisms have been proven to provide nutrients for specific plant growth, especially in nutrient-poor desert steppe ecosystems. However, the effects of N deposition on plant soil microbial community interactions in such ecosystems remain poorly understood. To investigate these effects, we conducted a 6-year N-addition field experiment in a Stipa breviflora Griseb. desert steppe in Inner Mongolia Autonomous Region, northern China. Four N treatment levels (N0, N30, N50, and N100, corresponding to 0, 30, 50, and 100 kg N/(hm 2 •a), respectively) were applied to simulate atmospheric N deposition. The results showed that N deposition did not significantly affect the aboveground biomass of desert steppe plants. N deposition did not significantly reduce the alfa-diversity of plant and microbial communities in desert steppe, and low and mediate N additions (30 and 50 kg N/(hm 2 •a)) had a promoting effect on them. The variation pattern of plant Shannon index was consistent with that of the soil bacterial Chao1 index. N deposition significantly affected the beta-diversity of plants and soil bacteria, but did not significantly affect fungal communities. In conclusion, N deposition led to co-evolution between desert steppe plants and soil bacterial communities, while fungal communities exhibited strong stability and did not undergo significant changes. These findings help clarify atmospheric N deposition effects on the ecological health and function of the desert steppe.

Abstract:Nitrogen (N) deposition is a worldwide issue caused by human activity. Long-term deposition of N strongly influences plant productivity and community composition. However, it is still unclear how the microbial community responds to long-term N addition in a desert ecosystem. Therefore, a long-term experiment was conducted in the Gurbantonggut Desert in northwestern China in 2015. Four N addition rates, 0 (CK), 5 (N1), 20 (N2), and 80 (N3) kg N ha −1 yr. −1 , were tested and the soil was sampled after 6 years of N addition. High-throughput sequencing (HTS) was used to analyze the soil microbial composition. The HTS results showed that N addition had no significant effect on the bacterial α-diversity and β-diversity ( p > 0.05) but significantly reduced the archaeal β-diversity ( p < 0.05). The fungal Chao1 and ACE indexes in the N2 treatment increased by 24.10 and 26.07%, respectively. In addition, N addition affected the bacterial and fungal community structures. For example, compared to CK, the relative abundance of Actinobacteria increased by 17.80%, and the relative abundance of Bacteroidetes was reduced by 44.46% under N3 treatment. Additionally, N addition also changed the bacterial and fungal community functions. The N3 treatment showed increased relative abundance of nitrate-reducing bacteria (27.06% higher than CK). The relative abundance of symbiotrophic fungi was increased in the N1 treatment (253.11% higher than CK). SOC and NH 4 + -N could explain 62% of the changes in the fungal community function. N addition can directly affect the bacterial community function or indirectly through NO 3 − -N. These results suggest that different microbial groups may have various responses to N addition. Compared with bacteria and fungi, the effect of N addition was less on the archaeal community. Meanwhile, N-mediated changes of the soil properties play an essential role in changes in the microbial community. The results in the present study provided a reliable basis for an understanding of how the microbial community in a desert ecosystem adapts to long-term N deposition.

Abstract:An arid ecosystem might be sensitive to nitrogen (N) deposition, but the associated ecosystem‐specific response of soil microbes is not well studied. To assess the N enrichment effects on plant and prokaryotic community diversity, we performed a two‐year NH4NO3 treatment in a desert steppe in northwestern China. Results showed that N addition increased plant aboveground biomass and decreased plant Shannon diversity. A C4 herb (Salsola collina) became dominant, and loss of legume species was observed. The concentrations of soil NH4+‐N, NO3−‐N, microbial biomass N, and the plant aboveground biomass N pool increased in contrast to total N, suggesting that the N input into the arid ecosystem might mainly be assimilated by plants and exit the ecosystem. Remarkably, the α‐diversity and structure of the soil prokaryotic community did not vary even at the highest N addition rate. Structural equation modeling further found that the plant aboveground N pool counteracted the acidification effect of N deposition and maintained soil pH thus partially stabilizing the composition of prokaryotic communities in a desert steppe. These findings suggested that the plants and N loss might contribute to the lack of responsiveness of soil prokaryotic community to N deposition in a desert steppe.This article is protected by copyright. Abstract:Human-caused nitrogen (N) deposition is a global environmental issue that can change community composition, functions, and ecosystem services. N deposition affects plants, soil, and microorganisms regionally and is linked to ecosystem, soil, and climate factors. We examined the effects of six N addition levels (0, 2.34 g, 4.67, 9.34,18.68, and 37.35 g N m−2 yr−1) on aboveground vegetation, surface soil properties, and microbial community. Alterations in microbial communities in response to N addition were monitored using 16S rRNA (16S ribosomal ribonucleic acid, where S donates a sedimentation coefficient) and ITS (internal transcribed spacer) regions for bacterial and fungal communities, respectively. N addition positively affected aboveground vegetation traits, such as biomass and community weighted mean of leaf nitrogen. N addition also limited phosphorus (P) availability and altered the microbial community assembly process from random processes to deterministic processes. The microbial community diversity and composition, however, were not sensitive to N addition. Partial least squares structural equation models showed that the composition of bacterial communities was mainly driven by the composition of plant communities and total nitrogen, while the composition of fungal communities was driven by soil pH and community weighted mean of leaf nitrogen. Taken together, the results of this research improved our understanding of the response of grassland ecosystems to N deposition and provided a theoretical basis for grassland utilization and management under N deposition.

Abstract:China has experienced a widespread increase in N deposition due to intensive anthropogenic activities, particularly in the subtropical regions. However, the effects of long-termN deposition on soil bacterial and fungal abundance, diversity, and community composition remain largely unclear. We assessed the effects of N deposition on soil microbial communities in summer and winter, using quantitative polymerase chain reaction and Illumina Miseq sequencing of bacterial 16S rRNA and fungal ITS genes fromsubtropical natural forest soils. The abundance of both soil bacteria and fungi exhibited a decreasing pattern with increasing N deposition rates. Nitrogen deposition increased bacterial diversity in both summer and winter, whereas fungal diversity was significantly decreased in summer, but greatly increased under the highest level of N deposition (150 kg N ha(-1) yr(-1)) in winter. Nitrogen deposition significantly increased the relative abundance of bacterial phyla Actinobacteria, Chloroflexi, and WPS-2, but decreased that of Acidobacteria and Verrucomicrobia. In addition, N deposition significantly decreased the relative abundance of Ascomycetes, but did not exert any significant effect on Basidiomycetes. The bacterial and fungal community compositions were greatly influenced by N deposition, with soil N availability and soil pH identified as the two most influential soil properties. This study demonstrates that the fungal community was more sensitive than the bacterial community to N deposition, and further emphasizes the importance of simultaneously evaluating soil bacterial and fungal communities in response to global environmental changes. Abstract:Context. Although nitrogen (N) deposition application has resulted in excess N entering grassland ecosystems, our understanding of how short-term N enrichment affects soil microbial community in a desert steppe is incomplete. Aims. This study assessed the effects of 2 years of N addition on structure and function of soil microbial community at the soil depth of 0-10 cm in a desert steppe.Methods. Six addition rates of N (equivalent to 0, 5, 10, 15, 20 and 40 g N m-2 year-1) were applied in a desert steppe of northern China, and the 16S rRNA and ITS 1 genes were sequenced for the prokaryotic and fungal communities, respectively.Key results. N addition altered prokaryotic taxa composition, but not fungal taxa composition and a-diversities of prokaryotic and fungal communities. N shifted microbial (3-diversities, and prokaryotic (3-diversity was more sensitive than fungal (3-diversity. Abundances of predicted prokaryotic functions decreased as N rates increased, whereas predicted fungal functions presented no obvious variations among N additions. Predicted enzyme activities related to N cycling in prokaryotes were similar to fungi, but variations of C and P cycling-related enzyme activities were opposite to fungi. Additionally, phylum abundance, a-diversity, and (3-diversity of prokaryotes were more closely influenced by soil and plant properties than fungi.Conclusions. Two years of N addition altered both microbial community structure and function, but the prokaryotic community responded more sensitive than the fungal community.

Abstract:Abstract Aims Nitrogen (N) deposition is a global environmental problem that can alter community compositions and functions, and consequently, the ecosystem services. In this study, we assessed the responses of aboveground vegetation, surface soil properties and microbial communities to N addition, and explored the drivers of microbial community in a semiarid steppe ecosystem in northwest of China. Methods Thirty-six 6×10-m 2 plots composed of six N addition levels and six replicates were distributed in six columns and six rows. Nine vegetation characteristics and seven soil properties were measured and calculated. Soil microbial characteristics were analyzed by 16S rRNA high-throughput sequencing. Results N addition positively affected aboveground vegetation traits such as the community weighted-mean of leaf nitrogen content (LN CWM ). High N inputs significantly altered the microbial community assembly process from random to deterministic. The microbial community diversity and composition, however, were not sensitive to N addition. A piecewise structural equation model (SEM) further showed that the microbial community composition was affected by both aboveground vegetation and soil properties. The composition of bacterial communities was mainly regulated by the composition of plant communities and soil total N. In contrast, the composition of fungal communities was driven by soil pH and the community weighted-mean of specific leaf area (SLA CWM ). Microbial diversity and composition remained unchanged because their drivers were not affected by N addition. The results of this research improved our understanding of the response of grassland ecosystems to N deposition, and provided a theoretical basis for grassland utilization and management under N deposition.
Differential Responses of Bacterial and Fungal Community Structure in Soil to Nitrogen Deposition in Two Planted Forests in Southwest China in Relation to pH

Abstract:Increased nitrogen deposition profoundly impacts ecosystem nutrient cycling and poses a significant ecological challenge. Soil microorganisms are vital for carbon and nutrient cycling in ecosystems; however, the response of soil microbial communities in subtropical planted coniferous forests to nitrogen deposition remains poorly understood. This study carried out a four-year nitrogen addition experiment in the subtropical montane forests of central Yunnan to explore the microbial community dynamics and the primary regulatory factors in two coniferous forests (P. yunnanensis Franch. and P. armandii Franch.) under prolonged nitrogen addition. We observed that nitrogen addition elicited different responses in soil bacterial and fungal communities between the two forest types. In P. yunnanensis Franch. plantations, nitrogen supplementation notably reduced soil bacterial α-diversity but increased fungal diversity. In contrast, P. armandii Franch. forests showed the opposite trends, indicating stand-specific differences. Nitrogen addition also led to significant changes in soil nutrient dynamics, increasing soil pH in P. yunnanensis Franch. forests and decreasing it in P. armandii Franch. forests. These changes in soil nutrients significantly affected the diversity, community structure, and network interactions of soil microbial communities, with distinct responses noted between stands. Specifically, nitrogen addition significantly influenced the β-diversity of fungal communities more than that of bacterial communities. It also reduced the complexity of bacterial interspecies interactions in P. yunnanensis Franch. forests while enhancing it in P. armandii Franch. forests. Conversely, low levels of nitrogen addition improved the stability of fungal networks in both forest types. Using random forest and structural equation modeling, soil pH, NH4+-N, and total nitrogen (TN) were identified as key factors regulating bacterial and fungal communities after nitrogen addition. The varied soil nutrient conditions led to different responses in microbial diversity to nitrogen deposition, with nitrogen treatments primarily shaping microbial communities through changes in soil pH and nitrogen availability. This study provides essential insights into the scientific and sustainable management of subtropical plantation forest ecosystems.

Abstract:Nitrogen (N) deposition and precipitation changes can strongly influence soil microbial properties in arid and semiarid regions. Here, we examined these effects on soil samples from the Inner Mongolia desert steppe of northern China after 7 yr of consecutive simulated N deposition by adding NH4NO3 and manipulation of precipitation, using a dilution plate method, PCR analysis, and 18S rRNA sequencing. The experimental treatments were as follows: control (CK), N addition (+N), N and water addition (+N+W), and N addition plus water reduction (+N-W). In this study, 14 genera and 32 fungal species were isolated, and Penicillium was determined to be the dominant fungal group. Treatment +N-W significantly increased (by 94.8%) the number of cultivable fungi as compared with CK. Compared with the CK community, fungal communities exposed to the three treatments, especially +N+W and +N-W, showed shifts in the relative abundances of cultivable fungi. Treatment +N-W significantly enhanced species richness compared with +N at the 02 cm soil depth. However, N addition and manipulation of precipitation did not influence species richness, the ShannonWeiner index, or evenness at the 030 cm soil depth. This study can provide insight into how fungal composition and diversity respond to climate change scenarios.

Abstract:Introduction: Continuous nitrogen deposition increases the nitrogen content of terrestrial ecosystem and affects the geochemical cycle of soil nitrogen. Forest-grassland ecotone is the interface area of forest and grassland and is sensitive to global climate change. However, the structure composition and diversity of soil microbial communities and their relationship with soil environmental factors at increasing nitrogen deposition have not been sufficiently studied in forest-grassland ecotone. Methods: In this study, experiments were carried out with four nitrogen addition treatments (0 kgN·hm −2 ·a −1 , 10 kgN·hm −2 ·a −1 , 20 kgN·hm −2 ·a −1 and 40 kgN·hm −2 ·a −1 ) to simulate nitrogen deposition in a forest-grassland ecotone in northwest Liaoning Province, China. High-throughput sequencing and qPCR technologies were used to analyze the composition, structure, and diversity characteristics of the soil microbial communities under different levels of nitrogen addition. Results and discussion: The results showed that soil pH decreased significantly at increasing nitrogen concentrations, and the total nitrogen and ammonium nitrogen contents first increased and then decreased, which were significantly higher in the N10 treatment than in other treatments (N:0.32 ~ 0.48 g/kg; NH 4 + -N: 11.54 ~ 13 mg/kg). With the increase in nitrogen concentration, the net nitrogen mineralization, nitrification, and ammoniation rates decreased. The addition of nitrogen had no significant effect on the diversity and structure of the fungal community, while the diversity of the bacterial community decreased significantly at increasing nitrogen concentrations. Ascomycetes and Actinomycetes were the dominant fungal and bacterial phyla, respectively. The relative abundance of Ascomycetes was negatively correlated with total nitrogen content, while that of Actinomycetes was positively correlated with soil pH. The fungal community diversity was significantly negatively correlated with nitrate nitrogen, while the diversity of the bacterial community was significantly positively correlated with soil pH. No significant differences in the abundance of functional genes related to soil nitrogen transformations under the different treatments were observed. Overall, the distribution pattern and driving factors were different in soil microbial communities in a forest-grassland ecotone in northwest Liaoning. Our study enriches research content related to factors that affect the forest-grassland ecotone.

Abstract:Desert microbes are expected to be substantially sensitive to global environmental changes, such as precipitation changes and elevated nitrogen deposition. However, the effects of precipitation changes and nitrogen enrichment on their diversity and community composition remain poorly understood. We conducted a field experiment over 2 years with multi-level precipitation and nitrogen addition in a desert shrubland of northern China, to examine the responses of soil bacteria and fungi in terms of diversity and community composition and to explore the roles of plant and soil factors in structuring microbial communities. Water addition significantly increased soil bacterial diversity and altered the community composition by increasing the relative abundances of stress-tolerant (dormant) taxa (e.g., Acidobacteria and Planctomycetes); however, nitrogen addition had no substantial effects. Increased precipitation and nitrogen did not impact soil fungal diversity, but significantly shifted the fungal community composition. Specifically, water addition reduced the relative abundances of drought-tolerant taxa (e.g., the orders Pezizales, Verrucariales, and Agaricales), whereas nitrogen enrichment decreased those of oligotrophic taxa (e.g., the orders Agaricales and Sordariales). Shifts in microbial community composition under water and nitrogen addition occurred primarily through changing resource availability rather than plant community. Our results suggest that water and nitrogen addition affected desert microbes in different ways, with watering shifting stress-tolerant traits and fertilization altering copiotrophic/oligotrophic traits of the microbial communities. These findings highlight the importance of resource availability in driving the desert microbial responses to short-term environmental changes.

Abstract:Introduction: Nitrogen (N) deposition seriously affects the function of carbon (C) and N cycling in terrestrial ecosystems by altering soil microbial communities, especially in desert steppe ecosystems. However, there is a need for a comprehensive understanding of how microorganisms involved in each C and N cycle process respond to N deposition. Methods: In this study, shotgun metagenome sequencing was used to investigate variations in soil C and N cycling-related genes in the desert steppe in northern China after 6 years of the following N deposition: N0 (control); N30 (N addition 30 kg ha −1 year −1 ): N50 (N addition 50 kg ha −1 year −1 ). Results: N deposition significantly increased the relative abundance of Actinobacteria ( P < 0.05) while significantly decreased the relative abundances of Proteobacteria and Acidobacteria ( P < 0.05). This significantly impacted the microbial community composition in desert steppe soils. The annual addition or deposition of 50 kg ha −1 year −1 for up to 6 years did not affect the C cycle gene abundance but changed the C cycle-related microorganism community structure. The process of the N cycle in the desert steppe was affected by N deposition (50 kg ha −1 year −1 ), which increased the abundance of the pmoA-amoA gene related to nitrification and the nirB gene associated with assimilation nitrite reductase. There may be a niche overlap between microorganisms involved in the same C and N cycling processes. Discussion: This study provides new insights into the effects of N deposition on soil microbial communities and functions in desert steppe and a better understanding of the ecological consequences of anthropogenic N addition.

Abstract:Currently, the impact of changes in precipitation and increased nitrogen(N) deposition on ecosystems has become a global problem. In this study, we conducted a 8-year field experiment to evaluate the effects of interaction between N deposition and precipitation change on soil bacterial communities in a desert steppe using high-throughput sequencing technology. The results revealed that soil bacterial communities were sensitive to precipitation addition but were highly tolerant to precipitation reduction. Reduced precipitation enhanced the competitive interactions of soil bacteria and made the ecological network more stable. Nitrogen addition weakened the effect of water addition in terms of soil bacterial diversity and community stability, and did not have an interactive influence. Moreover, decreased precipitation and increased N deposition did not have a superimposed effect on soil bacterial communities in the desert steppe. Soil pH, moisture content, and NH 4 + -N and total carbon were significantly related to the structure of bacterial communities in the desert steppe. Based on network analysis and relative abundance, we identified Actinobacteria, Proteobacteria, Acidobacteria and Cyanobacteria members as the most important keystone bacteria that responded to precipitation changes and N deposition in the soil of the desert steppe. In summary, we comprehensively analyzed the responses of the soil bacterial community to precipitation changes and N deposition in a desert steppe, which provides a model for studying the effects of ecological factors on bacterial communities worldwide.

Abstract:Increasing nitrogen (N) deposition profoundly affects nutrient cycling in soil, thereby influencing forest ecosystem productivity and function. Soil microorganisms are integral in driving nutrient turnover; the changes in microbial communities in response to N deposition and the associated soil nutrient availability, especially of limited nutrients, are far from clear. To explore the changes in soil bacterial and fungal communities and their key environmental drivers under N deposition, we conducted a multilevel field N addition experiment in a Castanopsis carlesii natural forest. Soil properties and bacterial and fungal communities were investigated. There were no significant changes in alpha diversities (presented as Chao1 and Shannon's indexes) and beta diversities of bacteria and fungi among the three treatments. Consistently, the relative abundances of dominant bacterial phyla (i.e., Proteobacteria, Acidobacteria, Actinobacteria, Chloroflexi, and Planctomycetes) and fungal phyla (i.e., Basidiomycota, Ascomycota, and Rozellomycota) did not change following N addition. These results suggest that N deposition did not alter microbial community diversity and structure. In addition, the results of the Mantel test showed that soil pH, NO3−-N, dissolved organic N (DON), and total phosphorus (TP) predominantly influenced the community diversity and structure in bacteria, but not in fungi. Meanwhile, the relative abundance of some sensitive microbial genera, such as Bryobacter, Bradyrhizobium, Sorangium, and Archaeorhizomyces, were significantly decreased. These results indicate a decreased microbial ability for N fixation and P mobilization induced by N deposition. Moreover, there were significant relationships between Bryobacter, Bradyrhizobium, and Archaeorhizomyces and NO3−-N and available P (AP), suggesting that the responses of sensitive microbial groups to N deposition likely depend on the changes in available nutrients in soil, especially limited N or P. Collectively, 6 years of N addition had no significant influence on microbial communities, but some sensitive microbial groups were associated with N or P turnover. This finding emphasizes the critical roles of sensitive microbial species in meditating limited nutrient cycling in soil under climate change.

Abstract:Although the effects of human-enhanced atmospheric nitrogen (N) deposition are well documented, the response of dryland soils to N deposition remains unclear owing to the divergence in hydrological outputs and soil heterogeneity. We selected a typical desert steppe in western China to simulate the effects of long-term N deposition by applying 0 (CK), 3.5, 7, and 14 g N m-2 yr-1 for 12 consecutive years. We found that, compared with the CK plots, the total N content of the upper (0-10 cm) and lower (10-20 cm) soil layers in fertilized plots increased by 8.3-14.6 % and 2.4-8.2 %, respectively. Correspondingly, the available, NH4+-, and NO3--N contents in the upper soil significantly increased by 25.5-68.3 %. However, in the lower soil, available and NO3--N contents were significantly lower than those in the CK plots, and their variation trend was opposite to that of NH4+-N, implying N turnover and leaching. As a result, the upper and lower soil pH in fertilized plots significantly decreased by 0.36-0.53 and 0.31-0.37 units; however, their CaCO3 content significantly increased by 9.8-22.8 % and 7.2-30.3 %, respectively. The total phosphorus (P) content in the upper and lower soil layers in fertilized plots significantly increased and decreased by 3.6-51.3 % and 16.7-62.5 %, respectively, however, both significantly decreased along the N fertilization gradient. Furthermore, the upper and lower soil organic carbon (SOC) content in the fertilized plots significantly increased by 57.7-78.1 % and 19.2-27.4 %, respectively. Pearson's correlation analysis revealed that available soil P was significantly negatively correlated with plant shoot Mn content (a proxy for rhizosphere carboxylates), whereas dissolved OC, SOC, and CaCO3 were significantly positively correlated, suggesting that Ca cycling is involved in P cycling and SOC sequestration. Our study suggests that long-term N input exacerbates P limitation in desert steppes, however, enhances SOC sequestration.

Abstract:Introduction: The impact of nitrogen (N) deposition on the soil N-transforming process in grasslands necessitates further investigation into how N input influences the structural composition and diversity of soil N-cycling microbial communities across different grassland types. Methods: In this study, we selected two types of grassland soils in northwest Liaoning, temperate steppe and warm-temperate shrub, and conducted short-term N addition experiments using organic N, ammonium N, and nitrate N as sources with three concentration gradients to simulate N deposition. Illumina MiSeq sequencing technology was employed to sequence genes associated with N-cycling microbes including N-fixing, ammonia-oxidizing and denitrifying bacteria, and ammonia-oxidizing archaea. Results and discussion: The results revealed significant alterations in the structural composition and diversity of the N-cycling microbial community due to N addition, but the response of soil microorganisms varied inconsistent among different grassland types. Ammonium transformation rates had a greater impact on soils from temperate steppes while nitrification rates were more influential for soils from warm-temperate shrubs. Furthermore, the influence of the type of N source on soil N-cycling microorganisms outweighed that of its quantity applied. The ammonium type of nitrogen source is considered the most influential driving factor affecting changes in the structure of the microbial community involved in nitrogen transformation, while the amount of low nitrogen applied primarily determines the composition of soil bacterial communities engaged in nitrogen fixation and nitrification. Different groups of N-cycling microorganisms exhibited distinct responses to varying levels of nitrogen addition with a positive correlation observed between their composition, diversity, and environmental factors examined. Overall findings suggest that short-term nitrogen deposition may sustain dominant processes such as soil-N fixation within grasslands over an extended period without causing significant negative effects on northwestern Liaoning's grassland ecosystems within the next decade.

Abstract:Investigating the response of soil microbial communities to nitrogen (N) deposition is critical to understanding biogeochemical processes and the sustainable development of forests. However, whether and to what extent different forms of N deposition affect soil microbial communities in temperate forests is not fully clear. In this work, a field experiment with three years of simulated nitrogen deposition was conducted in temperate forests. The glycine and urea were chosen as organic nitrogen (ON) source, while NH4NO3 was chosen as inorganic nitrogen (IN) source. Different ratios of ON to IN (CK = 0:0, Mix-1 = 10:0, Mix-2 = 7:3, Mix-3 = 5:5, Mix-4 = 3:7, Mix-5 = 0:10) were mixed and then used with equal total amounts of 10 kg·N·ha−1·a−1. We determined soil microbial diversity and community composition for bacteria and fungi (16S rRNA and ITS), and soil parameters. Different forms of N addition significantly changed the soil bacterial and fungal communities. Mixed N sources had a positive effect on soil bacterial diversity and a negative effect on fungal diversity. Bacterial and fungal community structures were significantly separated under different forms of N addition. Soil pH was the main factor affecting the change in fungal community structure, while bacterial community structure was mainly controlled by STN. We also found that Proteobacteria, Acidobacteriota, Basidiomycota and Ascomycota were the most abundant phyla, regardless of the form of N addition. RDA showed that C/P and NH4+ were the main factors driving the change in bacterial community composition, and C/P, pH and C/N were the main factors driving the change in fungal community composition. Our results indicate that different components of N deposition need to be considered when studying the effects of N deposition on soil microorganisms in terrestrial ecosystems.

Abstract:Understanding the response of the plant community to increasing nitrogen (N) deposition is helpful for improving pasture management in semi-arid areas. We implemented a 5-year N addition experiment in a Stipa krylovii steppe of Inner Mongolia, northern China. The aboveground biomass (AGB) and species richness were measured annually. Along with the N addition levels, the species richness declined significantly, and the species composition changed noticeably. However, the total AGB did not exhibit a noticeable increase. We found that compensatory effects of the AGB occurred not only between the grasses and the forbs but also among Gramineae species. The plant responses to N addition, from the community to species level, lessened in dry years compared to wet or normal years. The N addition intensified the reduction of community productivity in dry years. Our study indicated that the compensatory effects of the AGB among the species sustained the stability of grassland productivity. However, biodiversity loss resulting from increasing N deposition might lead the semi-arid grassland ecosystem to be unsustainable, especially in dry years.

Abstract:Nutrient addition can alter plant–soil‐microbe interactions. However, knowledge remains limited about how the soil biotic and abiotic properties drive changes in plant community composition and diversity after nitrogen (N) addition. In this study, four N addition rates (N0, N3, N6, and N9: 0, 3, 6, and 9 g N m −2 yr−1) were applied to a grassland community on the Loess Plateau (China) to evaluate the response of plant community biomass, evenness, and composition to N addition. Overall, N addition significantly increased plant shoot and root biomass by 27.75%–31.37% and 39.60%–65.42% compared with N0, but no significant difference was observed among N3, N6, and N9. N addition significantly decreased shoot/root biomass ratio by 7.17%–21.84%, and plant community evenness by 3.80%–8.80% compared with N0, particularly under N3 and N6 treatments. Furthermore, N addition enhanced the abundance of bacteria and fungi, but their composition was not altered. Responses of plant biomass and evenness to N addition were correlated to changes in NO3−‐N, soil total phosphorus, and the abundance of bacteria and fungi; additionally, the composition was primarily affected by the microbial abundance. Our research indicates that N addition can significantly alter plant community composition, particularly at low addition rates, and such alteration may be more affected by the soil microbial abundance. The findings of this study have important implications for the sustainable management of grassland ecosystems.
