Abstract:The porous rice starch (PS) and Xanthan gum (XG) are mixed at different ratios to embed the curcumin. The morphology and physicochemical properties of the microcapsules are characterized with scanning electron microscopy (SEM), X-ray diffraction (XRD), differential scanning calorimeter (DSC), and fourier transform infrared spectroscopy (FT-IR); the release of curcumin is evaluated with a simulated gastric-small intestine model. Results indicate that all the microcapsules present high encapsulation efficiency (>90%) with an average particle size of around 3.30 mu m. The curcumin loaded in PS and PS/XG microcapsules forms inclusion complexes rather than simple physical mixtures. The release mechanism of curcumin from microcapsules fits first-order (R-2> 0.974) and Higuchi (R-2> 0.969) models well in gastric and intestinal models, respectively, indicating that encapsulation of curcumin within PS/XG enables controlled release of curcumin. PS/XG wall materials show great potential for encapsulation and delivery of curcumin.

Abstract:Aim: In this study, a nanosponge structure was synthesised with capability of encapsulating curcumin as a model polyphenolic compound and one of the herbal remedies that have widely been considered due to its ability to treat cancer.Methods: FTIR, DSC and XRD techniques were performed to confirm the formation of the inclusion complex of the nanosponge-drug.Results: DSC and XRD patterns showed an increasing stability and a decreasing crystallinity of curcumin after formation of inclusion complex. Encapsulation efficiency was 98% (w/w) and a significant increase was observed in loading capacity (184% w/w). The results of cytotoxicity assessments demonstrated no cell toxicity on the healthy cell line, while being toxic against cancer cells. Haemolysis test was performed to evaluate the blood-compatibility characteristic of nanosponge and complex and the results showed 0.54% haemolysis in the lowest complex concentration (50μgml−1) and 5.09% at the highest concentration (200μgml−1).Conclusions: Thus, the introduced system could be widely considered in cancer treatment as a drug delivery system.

Influence of molecular interactions on structure, controlled release and cytotoxicity of curcumin encapsulated chitosan - Silica nanostructured microspheres

Abstract:Curcumin possesses numerous medicinal benefits including anti-cancer and anti-viral properties. However, its wide scale use as a drug is often hindered owing to the dearth of suitable drug-delivery systems which can solubilise it for long-term sustained-release and safeguard its beneficial properties. In this work, a fast, one-step method, employing evaporation induced assembly of colloids, has been employed for the synthesis of curcumin encapsulated organic-inorganic hybrid micron-sized spheres. Detailed physical properties of the microspheres, with scaffolds of silica nanoparticles (∼8.5 nm) cross linked by chitosan, are studied to trace the underlying mechanism of structural assembly in such systems, by tuning the polymer matrix with solubilizing agents, DMSO and Tween 20. A systematic modification in the hydrogen bonding network, conformations and interactions between macromolecules is revealed upon tuning the organic matrix. This in turn is found to control the assembly vis-à-vis the granular morphology, drug entrapment and packing fraction of nanoparticles in the microspheres, which have direct influence on the biological properties. Consequently, the microspheres are found to follow a first order drug release kinetics with a tunable rate constant which follows the ordering of packing fraction of silica nanoparticles in the micro-granules. A sustained curcumin release for a period extending up to 24 h has been achieved. Further studies using human lung and cervical cancer cell lines assert good anti-cancer properties of these nanostructured microspheres in cancer cells, while showing no toxicity towards normal cells. Thus, such hybrid organic-inorganic formulations achieved using multi-component colloidal assembly approach, with enhanced stability against degradation, are promising candidates for future clinical applications of water-insoluble drugs.

Abstract:Curcumin, a natural chemical compound found in Curcuma longa that has been used in antitumor and anti-inflammation applications, exhibits very limited water solubility and rapid in vivo degradation, which limits its clinical application. To overcome these limitations, niosome nanoparticles were prepared by microfluidic mixing for curcumin encapsulation. Niosome nanoparticles are lipid-based, and composed of non-ionic surfactants with cholesterol orientated into a membrane bilayer structure. Two different non-ionic surfactants were used and the mixing parameters were varied to optimize the characteristics of the prepared niosomes. The prepared niosomes had an average particle size of 70–230 nm depending on the type of non-ionic surfactant used and the mixing parameter. Moreover, all prepared niosomes were monodisperse with an average polydispersity index ranging from 0.07 to 0.3. All prepared niosomes were spherical as demonstrated by transmission electron microscopy. Curcumin was encapsulated with a maximum encapsulation efficiency of around 60% using Tween 85 as the non-ionic surfactant. Niosomes prepared by microfluidic mixing provided a controlled release of curcumin, as indicated by the release profile of curcumin, improving its therapeutic capability. These results demonstrate that niosomes prepared by microfluidic mixing to encapsulate curcumin are a promising delivery system to reach target cells.

Abstract:Novel engineered microgels with amphipathic network structures were designed and synthesized by copolymerizing N-isopropylacrylamide, 1-vinylimidazole, and 2-(cinnamoyloxy)ethyl methacrylate in the presence of 1,6-dibromohexane. The engineered microgels possess hydrophilic quaternization cross-linking structures and hydrophobic cross-linking inner nanodomains, which are suitable for loading and simultaneous release of hydrophilic nonsteroidal anti-inflammatory drug diclofenac sodium (DS) and hydrophobic antic cancer drug doxorubicin (DOX), respectively. The engineered microgels exhibited excellent stability, low cytotoxicity, and long blood circulation time and could be uptaken into the cytoplasm of cells, metabolized, and excreted from the living body by the kidney and the liver. In vivo experiments showed that with injection of DS and DOX dual-drug-loaded microgels, simultaneous antitumor treatment and inflammation depression were achieved along with high antitumor efficacy and low drug-related toxicity. Such microgels with amphipathic network structures have promising applications for combination therapy.

Abstract:Transdermal drug delivery system delivers a drug through skin to the desired cells. Its use is limited due to the poor drug diffusion through the outer skin layer. The aim of this work was to fabricate and characterize the gelatin cryogel matrices for the targeted transdermal curcumin delivery via iontophoresis for the breast cancer treatment. The gelatin cryogels were fabricated at various concentrations under the freeze-drying method. FTIR, SEM, STA, and BET were utilized to characterize the gelatin cryogels and curcumin loaded gelatin cryogels. The curcumin release behavior was investigated under the effects of gelatin concentrations, electric fields, pig skin permeation, and pHs. The gelatin cryogel matrices possessed the interconnected pore sizes between 56.61 μm and 129.67 μm, in which the pore size increased with increasing gelatin concentration. The curcumin percentage released from the 1.5%v/v PSG cryogel was 89.7 %, relatively higher than the 0.8%v/v PSG cryogels. Under the effect of electric fields of 3V and 6V, the curcumin release percentages from the 1.5%v/v PSG cryogel were increased to reach 94.7 % and 96.0 %, respectively. For the release-permeation of curcumin through the pig skin, the curcumin release percentages from the 1.5%v/v PSG cryogel under the electric fields of 0V, 3V, and 6V became lower at 38.45 %, 46.33 %, and 72.33 %, respectively, as the pig skin was the obstacle in the curcumin diffusion. Slightly higher release rates and amounts of curcumin from the 1.5%v/v PSG cryogel were observed at the of pH 5.5 relative to the pH of 7.4, and at the electric field of 6V relative to 0V. The effect of applied electric field was shown to shorten the delivery time from the PSG cryogel. The 1.5%v/v PSG cryogel under applied electric field 6V was shown here as a potential biobased matrix to be used in the curcumin transdermal delivery via iontophoresis for cancer treatment. Abstract:Curcumin (Cur) is a beneficial ingredient with numerous bioactivities. However, due to its low solubility and poor bioavailability, its therapeutic application is limited. In this work, we prepared poly-N-isopropylacrylamide p(NIPAm) and polyallylamine p(Am)-based nanogel (p(NIPAm-co-Am)) NG for a dual pH- and temperature-sensitive copolymer system for drug delivery application. In this copolymer system, the p(NIPAm) segment was incorporated to introduce thermoresponsive behavior and the p(Am) segment was incorporated to introduce drug binding sites (amine groups) in the resulting (p(NIPAm-co-Am)) NG system. Various instrumental characterizations including 1H nuclear magnetic resonance (1H NMR) spectroscopy, Fourier transform infrared (FT-IR) analysis, scanning electron microscopy (SEM), zeta potential, and particle size analysis were performed to confirm the copolymer synthesis. Curcumin (Cur), an anticancer bioactive substance, was employed to assess the in vitro drug loading and release performance of the resulting copolymer nanogels system at varied pH levels (pH 7.2, 6.5, and 4.0) and temperatures (25 °C, 37 °C, and 42 °C). The cytocompatibility of the p(NIPAm-co-Am) NG sample was also tested on MDA-MB-231 cells at various sample concentrations. All the study results indicate that the p(NIPAm-co-Am) NG produced might be effective for drug loading and release under pH and temperature dual-stimuli conditions. As a result, the p(NIPAm-co-Am) NG system has the potential to be beneficial in the use of drug delivery applications in cancer therapy.

Abstract:Curcumin is an extract of turmeric (Curcuma longa) which possesses anti-inflammatory, anti-cancer and wound-healing effects and has been used as an active compound in biomedical research for many years. However, its poor solubility presents challenges for its use in drug delivery systems. A modified nanogel delivery system, with PNIPAM and β-cyclodextrin grafted onto hyaluronic acid (PNCDHA), was utilized to enhance the solubility. The polymer was characterized by NMR, and the inclusion complex between curcumin and β-cyclodextrin was confirmed by FTIR. The potential of this PNCDHA polymer complex as a drug delivery vehicle was supported by a curcumin encapsulation efficiency of 93.14 ± 5.6% and the release of encapsulated curcumin at 37 °C. At a concentration of 0.5% w/v in water, PNCDHA nanogels were biocompatible with fibroblast cell line (L929) up to a curcumin concentration of 50 μM. There was a direct concentration between curcumin loading and cellular internalization. A more detailed study of the cellular internalization of PNCDHA nanogel should be considered in order to clarify cellular delivery mechanisms and to assess how its viability as a carrier may be optimized.

Abstract:Introduction: Most of the cytotoxic anticancer drugs belong to substances with both low solubility in aqueous fluids and poor cellular uptake, which lead to lack of specific therapy with apparent side effects. Therefore, there is a need to develop a novel drug delivery that can remotely and selectively release their payload. Curcumin(CUR) is an antibiotic, also a powerful inhibitor of the proliferation of several tumor cells. Aims: The aim of the present work is to highlight and discussed hyaluronic acid (HA ) to be grafted with Mono-6-deoxyl-6-ethylenediamino-γ-cyclodextrin (ɣ-CD-EDA) to shape a hydrogel that ought to structure inclusion complexes with curcumin, bettering its water-solubility and serving as a model drug delivery system Methods: Distinct copolymers had been organized HA grafted with ɣ-cyclodextrin (ɣ-CD-EDA) to form a hydrogel with various HA: ɣ-CD-EDA ratios and characterized, by means of 1H-NMR spectroscopy, zeta potential, Thermogravimetry analysis) TGA) Differential scanning calorimetry (DSC), and transmission electron microscopy (TEM). Encapsulation of CUR into polymer increased its selectivity, distribution, and accumulation into the cancer cells. HA-CD-EDA1conjugated curcumin if incorporated in suitable matrix has a potential utility for treatment of wound, and down regulation in THP-1 cells.
Temperature/pH/reduction-sensitive multiple stimulus-responsive nanogels synthesized by one-pot method for design as curcumin carriers

Abstract:Curcumin (Cur) is of interest due to its widely reported pharmacology, which is limited by its low water solubility and low bioavailability of clinical use. Here, we report the characterization and application of a multiple stimulus-responsive nanogel loaded with Cur. Sensitive P(AM-co-NIPAM) nanogels for pH, temperature, and reduction stimuli were prepared by reflux precipitation polymerization method using AM and NIPAM as raw materials. DLS and SEM analyses showed that the fabricated nanogels had a uniform size distribution with spherical and nucleated-shell structures. By adjusting the feeding amounts of reactive monomers, cross-linkers, and initiators, a series of monodisperse nanogels with different particle sizes could be obtained. The representative nanogels have different Cur-loaded behaviors at different temperatures, times, and initial concentrations of the drug, as well as a sensitive release to changes in pH, temperature, or reducing conditions. Cur-P(AM-co-NIPAM) nanogels showed superior antioxidant activity over the free Cur in vitro. Meanwhile, it inhibited LPS and H2O2-induced oxidative stress, which could effectively inhibit the cellular production of excessive reactive oxygen species and had a protective effect against LPS-induced cellular damage. This nanogel controlled release system significantly improved the safety limit of Cur without hemolytic behavior and had good cytocompatibility. The successful development of such Cur nanogels will lead to advanced materials that can be functionalized and optimized for the controlled delivery of small molecules or biomolecules for targeted therapeutic and biomedical applications.

Abstract:The oral bioavailability of curcumin is limited, attributed to its low solubility or dissolution and poor absorption. Herein, the study describes formulation of curcumin-loaded mixed micelles of Gelucire® 48/16 and TPGS for its dissolution rate enhancement. Curcumin was dispersed in these molten lipidic surfactants which was then adsorbed on carrier and formulated as pellets by extrusion spheronization. Critical micelle concentration (CMC) of binary mixture of Gelucire® 48/16 and TPGS was lower than their individual CMC demonstrating the synergistic behavior of mixture. Thermodynamic parameters like partition coefficient and Gibbs free energy of solubilization indicated that mixed micelles were more efficient than micelles of its individual components in curcumin solubilization. Dynamic light scattering (DLS) suggested slight increase in micellar size of mixed micelles than its components suggesting curcumin loading in mixed micelles. Fourier transform infrared spectroscopy (FTIR) revealed that phenolic hydroxyl group interacts with lipids which contribute to its enhanced solubility. Furthermore, the differential scanning calorimetry (DSC) and X-ray diffraction (XRD) study indicated the conversion of crystalline curcumin into amorphous form. In the pellet formulation, Gelucire® 48/16 acted as a binder and eliminated the requirement of additional binder. Microcrystalline cellulose (MCC) forms wet mass and retards the release of curcumin from pellets. Increase in concentration of water-soluble diluent increased drug release. The optimized formulation released more than 90% drug and maintains supersaturation level of curcumin for 2 h. Thus, mixed micellar system was effective delivery system for curcumin while pellet formulation is an interesting formulation strategy consisting semi-solid lipids.Graphical

Abstract:The rapid growth and diversification of drug delivery systems have been significantly supported by advancements in micro- and nano-technologies, alongside the adoption of biodegradable polymeric materials like poly(lactic-co-glycolic acid) (PLGA) as microcarriers. These developments aim to reduce toxicity and enhance target specificity in drug delivery. The use of in silico methods, particularly molecular dynamics (MD) simulations, has emerged as a pivotal tool for predicting the dynamics of species within these systems. This approach aids in investigating drug delivery mechanisms, thereby reducing the costs associated with design and prototyping. In this study, we focus on elucidating the diffusion mechanisms in curcumin-loaded PLGA particles, which are critical for optimizing drug release and efficacy in therapeutic applications.

Abstract:Curcumin has a broad-spectrum anti-tumor effect and has no toxic side effects. However, the unique diketone structure of curcumin will undergo diketo-enol tautomerism under different acid-base conditions, resulting in its instability under physiological conditions. In addition, the low biocompatibility and absorption rate of curcumin also limit the use of curcumin drugs. In this paper, curcumin was modified by substitution of acryloyl and acrylsulfonyl groups, and four kinds of nanoparticles with regular morphology were prepared using non-toxic and non-irritating acrylic resin as coating material to improve the stability and bioavailability of the compounds. Zeta potential testing shows that the composites surface carries positive charges and have good stability. In the release experiment, four complexes have the potential for slow and controlled release. Imaging of Hela cells with different channels was performed, and the imaging results showed that the complexes could enter the cells and be absorbed by them, demonstrating good imaging performance. MTT experiments have shown that the complexes have certain anti-tumor activity and low cytotoxicity. In general, the complexes synthesized in this paper have potential in the field of drug fluorescence imaging detection. At the same time, this experiment provides a new idea for the design of slow and controlled release of drugs.

Amphiphilic Curcumin Conjugate-Forming Nanoparticles As Anticancer Prodrug and Drug Carriers: in Vitro and in Vivo Effects

Abstract:Curcumin has been shown to have high cytotoxicity towards various cancer cell lines, but its water insolubility and instability make its bioavailability exceedingly low and, thus, it is generally inactive in in vivo anticancer tests. Here, we report an intracellular-labile amphiphilic surfactant-like curcumin prodrug curcumin conjugated with two short oligo(ethylene glycol) (Curc-OEG) chains via beta-thioester bonds that are labile in the presence of intracellular glutathione and esterase. Curc-OEG formed stable nanoparticles in aqueous conditions and served two roles - as an anticancer prodrug and a drug carrier. As an anticancer prodrug, the formed nanoparticles had a high and fixed curcumin-loading content of 25.3 wt%, and released active curcumin in the intracellular environment. Curc-OEG had high inhibition ability to several cancer cell lines due to apoptosis. Intravenously injected Curc-OEG significantly reduced the tumor weights and tumor numbers in the athymic mice xenografted with intraperitoneal SKOV-3 tumors and subcutaneous (mammary fat pad) MDA-MB-468 tumors. Preliminary systemic toxicity studies found that Curc-OEG did not cause acute and subchronic toxicities to mouse visceral organs at high doses. As drug carriers, Curc-OEG nanoparticles could carry other anticancer drugs, such as doxorubicin and camptothecin, and ship them into drug-resistant cells, greatly enhancing the cytotoxicity of the loaded drug. Thus, Curc-OEG is a promising prototype that merits further study for cancer therapy.

Abstract:Modifying positive surface charge and reducing bulk size of nanoparticles has been proven beneficial to cancer cellular delivery, but meanwhile results in fast clearance and unspecific distribution in body after intravenous injection. How to balance these problems is still a challenge to construct an ideal nanoscaled drug delivery system in cancer treatment. Herein, we developed a multistage drug delivery system to enhance anticancer efficacy of curcumin (CUR), which could intelligently alter its size and surface charge after long-circulation and extravasation from leaky blood vessels at tumor sites. This micellar system was constructed by amphiphilic and pH-sensitive methoxy poly(ethylene glycol)-poly(lactide)-poly(beta-amino ester) (MPEG-PLA-PAE) copolymers. As compared with MPEG-PLA micelles, MPEG-PLA-PAE micelles displayed several advantageous characteristics for drug delivery and treatment. We found that CUR-loaded MPEG-PLA-PAE micelles remained stable in murine plasma at 37 degrees C even with high drug loading. More interestingly, when the media pH decreased from 7.4 to 5.5, the micelles shrank from 171.0 nm to 22.6 nm and their surface charge increased to 24.8 mV meanwhile, which resulted in the significantly improved cell uptake of CUR by human breast cancer MCF-7 cells. Using indocyanine green (ICG) as a fluorescence probe, it was observed that MPEG-PLA-PAE micelles experienced longer circulation than MPEG-PLA micelles followed by accumulation at tumors with stronger fluorescence intensity. Consequently, MPEG-PLA-PAE micelles achieved enhanced cancer growth inhibition of 65.6% in vivo. All these findings demonstrated the potential of size/charge-changing MPEG-PLA-PAE micelles as a promising drug delivery system for tumor-targeted therapy. (C) 2014 Elsevier Ltd. All rights reserved.
Elaboration and Characterization of Curcumin-Loaded Tri-CL-mPEG Three-Arm Copolymeric Nanoparticles by a Microchannel Technology

Abstract:Purpose: Clinical applications of curcumin (Cur) have been greatly restricted due to its low solubility and poor systemic bioavailability. Three-arm amphiphilic copolymer tricarballylic acid-poly (ε-caprolactone)-methoxypolyethylene glycol (Tri-CL-mPEG) nanoparticles (NPs) were designed to improve the solubility and bioavailability of Cur. The present study adopted a microchannel system to precisely control the preparation of self-assembly polymeric NPs via liquid flow-focusing and gas displacing method. Methods: The amphiphilic three-arm copolymer Tri-CL-mPEG was synthesized and self-assembled into nearly spherical NPs, yielding Cur encapsulated into NP cores (Cur-NPs). The obtained NPs were evaluated for physicochemical properties, morphology, toxicity, cellular uptake by A549 cells, release in vitro, biodistribution, and pharmacokinetics in vivo. Results: Rapidly fabricated and isodispersed Cur-NPs prepared by this method had an average diameter of 116±3 nm and a polydispersity index of 0.197±0.008. The drug loading capacity and entrapment efficiency of Cur-NPs were 5.58±0.23% and 91.42±0.39%, respectively. In vitro release experiments showed sustained release of Cur, with cumulative release values of 40.1% and 66.1% at pH 7.4 and pH 5.0, respectively, after 10 days post-incubation. The results of cellular uptake, biodistribution, and in vivo pharmacokinetics experiments demonstrated that Cur-NPs exhibited better biocompatibility and bioavailability, while additionally enabling greater cellular uptake and prolonged circulation with possible spleen, lung, and kidney targeting effects when compared to the properties of free Cur. Conclusion: These results indicate that Tri-CL-mPEG NPs are promising in clinical applications as a controllable delivery system for hydrophobic drugs.

Abstract:Curcumin, a polyphenol with a rich history spanning two centuries, has emerged as a promising therapeutic agent targeting multiple signaling pathways and exhibiting cellular-level activities that contribute to its diverse health benefits. Extensive preclinical and clinical studies have demonstrated its ability to enhance the therapeutic potential of various bioactive compounds. While its reported therapeutic advantages are manifold, predominantly attributed to its antioxidant and anti-inflammatory properties, its efficacy is hindered by poor bioavailability stemming from inadequate absorption, rapid metabolism, and elimination. To address this challenge, nanodelivery systems have emerged as a promising approach, offering enhanced solubility, biocompatibility, and therapeutic effects for curcumin. We have analyzed the knowledge on curcumin nanoencapsulation and its synergistic effects with other compounds, extracted from electronic databases. We discuss the pharmacokinetic profile of curcumin, current advancements in nanoencapsulation techniques, and the combined effects of curcumin with other agents across various disorders. By unifying existing knowledge, this analysis intends to provide insights into the potential of nanoencapsulation technologies to overcome constraints associated with curcumin treatments, emphasizing the importance of combinatorial approaches in improving therapeutic efficacy. Finally, this compilation of study data aims to inform and inspire future research into encapsulating drugs with poor pharmacokinetic characteristics and investigating innovative drug combinations to improve bioavailability and therapeutic outcomes.

Facile preparation of pH-sensitive chitosan microspheres for delivery of curcumin; characterization, drug release kinetics and evaluation of anticancer activity

Abstract:Curcumin (CUR) is a lowly water-soluble natural polyphenol with chemopreventive and chemotherapeutic activities. Hence, to achieve the system with good CUR loading ability, porous MIL-88 (Fe) was prepared in the presence of the presynthesized graphene quantum dots (GQDs) (GQDs@MIL-88 (Fe)). In the following, CUR loaded in the fabricated GQDs@MIL-88 (Fe) nanohybrid. The characterization techniques; Fourier transform infrared (FT-IR), X-ray powder diffraction (XRD), scanning electron microscope (SEM), photoluminescence (PL), and Brunauer-Emmett-Teller (BET) analysis showed success in the synthesis of GQDs@MIL-88 (Fe). Moreover, the FT-IR analysis displayed the loading of CUR and the formation of CUR@GQDs@MIL-88(Fe). Chitosan (CS) was used as a green coating to enhance the biocompatibility of the prepared system (CS/CUR@GQDs@MIL-88(Fe). The fabricated microspheres showed pH-sensitive swelling behavior and released 38.3% of CUR in pH 5.0 which is better fitted with the First-order kinetic model (R2 = 0.9726). In comparison with CUR@GQDs@MIL-88(Fe), the MTT and DAPI assay exhibited less toxic effect for CS/CUR@GQDs@MIL-88(Fe) against MDA-MB 231 cells. Moreover, the safety of the CS/CUR@GQDs@MIL-88(Fe) confirmed after incubation against MCF 10A as a model of the normal cell line. The results conveyed a new concept that the CS/CUR@GQDs@MIL-88(Fe) is a potential candidate for using as a biocompatible carrier with controlled drug delivery ability.
