Abstract:Methionine dipeptide (Met-Met) could improve milk protein synthesis in bovine epithelia mammary cells and lactating mice, while the effects of Met-Met on lactation performance, rumen fermentation and microbiota profile in lactating dairy cows have not been explored. For this reason, 60 Chinese lactating Holstein cows were allocated into three treatment groups: control group (CON), 6 g/d methionine dipeptide group (MM), and 6.12 g/d rumen-protected methionine dipeptide group (RPMM). The experiment lasted for 10 weeks to monitor lactation performance, plasma amino acid profile and rumen fermentation parameters and microbiota profile. Results showed that MM increased the energy-corrected milk (ECM), and RPMM increased both milk yield and ECM (p < 0.05). The milk protein concentration and yield were increased by MM and RPMM (p < 0.05). The rumen fermentation showed that RPMM increased total volatile fatty acids, acetate and valerate concentrations (p < 0.05). The relative abundance of Firmicutes, including Succiniclasticum, Selenomonas and Clostridium_XlVa, were enriched and the Prevotella was decreased by RPMM (p < 0.05). In summary, daily supplementing with 6 g of MM or RPMM in lactating dairy cows could improve milk yield and both percentage and yield of milk protein, and RPMM benefited the rumen fermentation and altered the bacterial composition. These results provided the first evidence that Met-Met supplementation can improve lactation performance of dairy cows.
Integrated Transcriptomic Analyses of Liver and Mammary Gland Tissues Reveals the Regulatory Mechanism Underlying Dairy Goats at Late Lactation when Feeding Rumen-Protected Lysine

Abstract:Although low-protein diets can improve the nitrogen utilization efficiency and alleviate economic pressures in ruminants, they may also negatively impact dairy performance. Rumen-protected lysine (RPL) supplementation can improve the health status and growth performance of ruminants without compromising nitrogen utilization efficiency and feed intake. In this study, a total of thirty-three multiparous dairy goats in the late-lactation period were randomly divided into three groups that were separately fed the control diet (namely the protein-adequacy group), the low-protein diet (namely the protein-deficient group), and the RPL-supplemented protein-deficient diet (namely RPL-supplementation group) for five weeks. Here, we investigated the molecular mechanisms regarding how low-protein diets with RPL supplementation compromise lactation phenotypes in dairy goats through cross-tissue transcriptomic analyses. Dietary protein deficiency caused an imbalance in amino acid (AA) intake, disrupted hepatic function, and impaired milk synthesis. Transcriptomic analyses further showed that RPL supplementation exhibited some beneficial effects, like mitigating abnormal lipid and energy metabolism in the liver, elevating hepatic resistance to oxidative stress, improving the mammary absorption of AAs, as well as activating mammary lipid and protein anabolism primarily through peroxisome proliferator-activated receptor (PPAR) and janus kinase-signal transducer (JAK)—signal transducer and activator of transcription (STAT) signaling, respectively. RPL supplementation of a low-protein diet contributes to maintaining late lactation in dairy goats primarily through mitigating hepatic energy disturbances and activating both lipid and protein metabolism in the mammary glands. Since RPL supplementation initiated a series of comprised events on mammary protein and lipid metabolism as well as the hepatic function and energy generation in dairy goats under protein deficiency during late lactation, these findings thus provide some insights into how RPL supplementation helps maintain milk production and health in dairy mammals especially at late lactation.
Regulation of Nutritional Metabolism in Transition Dairy Cows: Energy Homeostasis and Health in Response to Post-Ruminal Choline and Methionine

Abstract:This study investigated the effects of rumen-protected methionine (RPM) and rumen-protected choline (RPC) on energy balance, postpartum lactation performance, antioxidant capacity and immune response in transition dairy cows. Forty-eight multiparous transition cows were matched and divided into four groups: control, 15 g/d RPC, 15 g/d RPM or 15 g/d RPC + 15 g/d RPM. Diet samples were collected daily before feeding, and blood samples were collected weekly from the jugular vein before morning feeding from 21 days prepartum to 21 days postpartum. Postpartum dry matter intake (DMI) was increased by both additives (P < 0.05), and energy balance values in supplemented cows were improved after parturition (P < 0.05). Both RPC and RPM decreased the plasma concentrations of non-esterified fatty acids (NEFA), β-hydroxybutyric acid (BHBA), total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) (P < 0.05), but increased the plasma levels of glucose, very-low-density lipoprotein (VLDL) and apolipoprotein B100 (ApoB 100, P < 0.05). The supplements improved milk production (P < 0.05), and increased (P < 0.05) or tended to increase (0.05 < P < 0.10) the contents of milk fat and protein. The post-ruminal choline and methionine elevated the blood antioxidant status, as indicated by total antioxidant capacity (T-AOC), glutathione peroxidase (GSH-Px) activity and the vitamin E concentration (P < 0.05), and reduced the plasma malondialdehyde (MDA) level (P < 0.05). Furthermore, RPM and RPC elevated the plasma interleukin 2 (IL-2) concentration and the CD4+/CD8+ T lymphocyte ratio in peripheral blood (P < 0.05). Alternatively, the levels of tumor necrosis factor-α (TNF-α) and IL-6 were decreased by RPM and RPC (P < 0.05). Overall, the regulatory responses of RPC and RPM were highly correlated with time and were more effective in the postpartum cows. The results demonstrated that dietary supplementation with RPC and RPM promoted energy balance by increasing postpartal DMI and regulating hepatic lipid metabolism, improved postpartum lactation performance and enhanced antioxidant capacity and immune function of transition dairy cows.

Abstract:The objectives of this study were to evaluate the effects of dietary supplementation of ruminally protected lysine (RPL), or methionine (RPM), and their combination (RPML) on the production efficiency of transition cows. A total of 120 pre-partum multiparous Holstein cows were assigned to four treatments based on previous lactation milk production, days (d) of pregnancy, lactation, and body condition score (BCS). Cows were fed a basal diet [pre-calving: 1.53 Mcal/kg dry matter (DM) and post-calving: 1.70 Mcal/kg DM] with or without supplemental ruminally protected amino acids (RPAA). Treatments were the basal diets without supplemental amino acids (CONTROL, n = 30), with supplemental methionine (RPM, pre-calving at 0.16% of DM and post-calving at 0.12% of DM, n = 30), with supplemental lysine (RPL, pre-calving at 0.33% of DM and post-calving at 0.24% DM, n = 30), and the combination (RPML, pre-calving at 0.16% RPM + 0.33% RPL of DM and post-calving at 0.12% RPM + 0.24 % RPL DM, n = 30). The dietary content of lysine was balanced to be within 6.157.2% metabolizable protein (MP)–lysine and that of methionine was balanced within 2.1–2.35% MP-methionine. Dry matter intake (DMI) was measured daily. Milk samples were taken on d 7, 14, and 21 days relative to calving (DRC), and milk yields were measured daily. Blood samples were taken on d −21, −14, −7 before expected calving and d 0, 7, 14, and 21 DRC. Data were analyzed using SAS software. There were significant Trt × time interactions ( P < 0.01) for DMI pre- and post-calving period. The CON cows had lower DMI than RPM, RPL, and RPML, both pre-calving ( P < 0.01) and post-calving periods ( P < 0.01). Energy-corrected milk ( P < 0.01), milk fat ( P < 0.01), protein ( P = 0.02), and lactose ( P < 0.01) percentage levels were greater for RPM, RPL, and RPML cows compared to CON. Supplementing RPAA assisted in maintaining BCS post-calving than CON ( P < 0.01). Blood concentrations of β-hydroxybutyrate decreased with RPM or RPL or the combination pre-calving ( P < 0.01) and tended to decrease post-calving ( P = 0.10). These results demonstrated that feeding RPL and RPM improved DMI and milk production efficiency, maintained BCS, and reduced β-hydroxybutyrate concentrations of transition cows.

Effects of rumen-encapsulated methionine and lysine supplementation and low dietary protein on nitrogen efficiency and lactation performance of dairy cows

Abstract:Low crude protein (CP) diets might be fed to dairy cows without affecting productivity if the balance of absorbed AA were improved, which would decrease the environmental effect of dairy farms. The aim of this study was to investigate the effects of supplementing ruminally protected Lys (RPL) and Met (RPM) at 2 levels of dietary CP on nutrient intake, milk production, milk composition, milk N efficiency (MNE), and plasma concentrations of AA in lactating Holstein cows and to evaluate these effects against the predictions of the new NASEM (2021) model. Fifteen multiparous cows were used in a replicated 3 × 3 Latin square design with 21-d periods. The 3 treatments were (1) a high-protein (HP) basal diet containing 16.4% CP (metabolizable protein [MP] balance of -130 g/d; 95% of target values), (2) a medium-protein diet containing 15% CP plus RPL (60 g/cow per day) and RPM (25 g/cow per day; MPLM; MP balance of -314 g/d; 87% of target values), and (3) a low-protein diet containing 13.6% CP plus RPL (60 g/cow per day) and RPM (25 g/cow per day; LPLM; MP balance of -479 g/d; 80% of target values). Dry matter intake was less for cows fed MPLM and LPLM diets compared with those fed the HP diet. Compared with the HP diet, the intake of CP, neutral detergent fiber, acid detergent fiber, and organic matter, but not starch, was lower for cows fed MPLM and LPLM diets. Milk production and composition were not affected by MPLM or LPLM diets relative to the HP diet. Milk urea N concentrations were reduced for the MPLM and LPLM diets compared with the HP diet, indicating that providing a low-protein diet supplemented with rumen-protected AA led to greater N efficiency. There was no significant effect of treatment on plasma AA concentrations except for proline, which significantly increased for the MPLM treatment compared with the other 2 treatments. Overall, the results supported the concept that milk performance might be maintained when feeding lactating dairy cows with low CP diets if the absorbed AA balance is maintained through RPL and RPM feeding. Further investigations are needed to evaluate responses over a longer time period with consideration of all AA rather than on the more aggregated MP and the ratio between Lys and Met.
Effects of Pelletizing and Supplementary Methionine, Lysine, and Choline on the Performance of Periparturient Dairy Cows

Abstract:This study was conducted to investigate the effects of pelletizing a portion of concentrate, and additional supplementary methionine (Met) and lysine (Lys) and choline on milk performance of periparturient cows. Animals were allocated to one of four treatments: basal unpellitized concentrate diet (control) and three diets in which a portion of the concentrate in the basal diet (6.72% DM prepartum; 15.7% DM postpartum) was pelletized and amino acids were added as follows: basal pellet (PEL), PEL plus lysine and methionine (PAA), and PAA plus choline (PAC). Pelletizing a portion of the diet (PEL) results in a significant increase in milk yield compared with the unpellitized control and addition of Lys and Met (PAA) but not choline (PAC) increased the yield further. Milk composition did not differ among treatments, but their yields increased by inclusion of Lys and Met-containing pelletized concentrate. Feeding Lys and Met-containing pelletized concentrate resulted in higher N conversion and fat content of milk compared with control and PEL. Cows on pelletized concentrate containing Lys and Met with or without choline tended to have lower serum non-esterified fatty acid, but no differences were observed in other parameters. It is concluded that pelletizing of a portion of concentrate can improve milk production and there is an additional beneficial effect of Lys and Met supplementation on yield and N and fat content of milk, but no additional effect of adding choline was observed.
Supplementation with rumen-protected methionine or choline during the transition period influences whole-blood immune response in periparturient dairy cows

Abstract:Methionine, together with Lys, is the most limiting AA for milk production in dairy cows. Besides its crucial role in milk production, Met and its derivate metabolites (e.g., glutathione, taurine, polyamines) are well-known immunonutrients in nonruminants, helping support and boost immune function and activity. In the present study, the effects of Met or choline, as its precursor, were investigated using an ex vivo whole blood challenge. The study involved 33 multiparous Holstein cows (from a larger cohort with a factorial arrangement of treatments) assigned from d -21 to +30 relative to parturition to a basal control (CON) diet, CON plus rumen-protected Met (MET, Smartamine M, Adisseo NA, Alpharetta, GA) at a rate of 0.08% of dry matter, or CON plus rumen-protected choline (CHOL, ReaShure, Balchem Inc., New Hampton, NY) at 60 g/d. Blood was sampled on d -15, -7, 2, 7, and 20 for ex vivo lipopolysaccharide (LPS) challenge, and on d 1, 4, 14, and 28 relative to parturition for phagocytosis and oxidative burst assays. The MET cows had greater energy-corrected milk production and milk protein content. Overall, IL-6 response to LPS increased around parturition, whereas IL-1β remained constant, casting doubt on the existence of systemic immunosuppression in the peripartal period. Supplementation with MET dampened the postpartal blood response to LPS (lower IL-1β), while improving postpartum neutrophil and monocyte phagocytosis capacity and oxidative burst activity. In contrast, CHOL supplementation increased monocyte phagocytosis capacity. Overall, the data revealed a peripartal immune hyper-response, which appeared to have been mitigated by MET supplementation. Both MET and CHOL effectively improved immune function; however, MET affected the immune and antioxidant status before parturition, which might have been beneficial to prepare the cow to respond to metabolic challenges after parturition. These results provide insights on potential differences in the immunomodulatory action of methionine and choline in dairy cows. As such, the effects observed could have implications for ration formulation and dietary strategies.

Abstract:One of the most important aspects of livestock is the supply of essential quantities of balanced nutrients manufacture. In several physiological functions of nutrients, concerning organisms, protein has an important role. Ruminants are not wholly protected of amino acids by microflora degradation of animal feed sources and by maintaining proteins from the breakdown in the rumen where both high and medium yielding ruminant amino acids are required, they must be met; therefore, rumen-protected protein (RPP) dietary supplementation, including primarily rumen-protected protein (RPP), methionine (RPMeth), protected from rumen flora, it was necessary. Numerous scientists are attracted in the study of the character of (RPMeth) in a dairy cow about it’s an impact on milk produce, growth rate, N- efficiency, coefficient digestibility, intake of DM. Unluckily, the findings obtained from many RPMeth investigations showed excessive fluctuation results in ruminant nutrition between its helpful and useless influence, specifically during early and late lactation periods. Thus, for ruminants, when they use this review post, farm owners may be careful when they are plan to apply RPMeth to the diet of animals. In summary, supplementation RPMeth also has a positive balanced effect, even with no detrimental effect on the yield of milk, animal productivity and hemato-biochemical parameters, especially during the early lactation when used with a ruminant diet low in crude protein
Enhancing methionine supply alleviates inflammation and oxidative stress and improves liver function during the peripartum period in dairy cows

Abstract:During the transition period, which comprises three weeks prepartum until 3 weeks postpartum, dairy cows experience a state of systemic inflammation and oxidative stress. Because the energy consumed by the cow is not enough to meet the energy required for milk production, adipose tissue is mobilized and provides a source of energy for tissues like the liver. However, excessive release of fatty acids from adipose tissues impairs liver function. Furthermore, high plasma concentrations of fatty acids (NEFA) contribute to oxidative stress and uncontrolled inflammatory responses often leading to health problems. Besides being an important building block for milk protein synthesis, methionine is a precursor for synthesis of VLDL, antioxidants and other methyl‐group requiring compounds. Thus, we hypothesized that increasing the intestinal supply of methionine could help alleviate inflammation and oxidative stress, and improves liver function during the peripartum period in dairy cows. Sixty‐one multiparous Holstein cows were used in a randomized complete block design and were fed the experimental treatments consisting of a basal control (CON; n=31) diet with no methionine supplementation or the basal diet plus rumen‐protected methionine (MET; Mepron®, Evonik Nutrition & Care GmbH, Germany; n=30). Methionine was supplied from ‐28 days to 60 days relative to parturition at a rate of 0.09% and 0.10% of dry matter (DM) during the prepartum and postpartum period, respectively. From −45 to −29 days before expected parturition, all cows received the same diet [1.33 Mcal/kg DM and 13.9% crude protein (CP)] with no MET. The basal “close‐up” (from −28 days to parturition), early lactation (from 1 to 30 days) and high‐production (from 31 to 60 d) diets contained 1.47 Mcal/kg DM and 15.3% CP, 1.67 Mcal/kg DM and 17.7% CP, and 1.61 Mcal/kg DM and 17.4% CP, respectively. Blood samples from 15 cows/group without clinical diseases after calving were collected at −30, −14, 1, 7, 21, 30 and 60 days relative to parturition and analyzed for biomarkers of energy balance, liver function, inflammation and oxidative stress. The indicators of energy balance β‐hydroxybutyrate, glucose and NEFA were not affected by supply of methionine. Among the five biomarkers of liver function analyzed, MET led to overall greater (P = 0.01; 94.8 vs 84.0 U/mL) paraoxonase (negative acute‐phase protein) and lower (P = 0.01; 15.1 vs 22.9 U/L) γ‐glutamyl transferase compared with CON. For cholesterol, a treatment × time (P = 0.07; 4.0 vs 3.5 mmol/L) was observed due to a greater increase over time in MET than CON cows. Aspartate aminotransferase and bilirubin were not affected by supply of methionine. Among inflammation biomarkers measured, MET led to greater (P = 0.01; 38.1 vs 37.1 g/L) albumin (negative acute‐phase protein) while alkaline phosphatase, ceruloplasmin (positive acute phase protein), and myeloperoxidase were not affected by methionine supply. The lower concentrations of reactive oxygen metabolites (P = 0.07; 15.2 vs 16.8 mg of H 2 O/100 mL) in MET compared with CON cows indicated a state of reduced oxidative stress. Overall, the results indicate that increasing methionine supply by feeding rumen‐protected methionine is an effective approach to help mitigate inflammation and oxidative stress as well as to enhance liver function during the peripartum period in dairy cows.

Abstract:Researchers have reported the benefits of feeding rumen-protected methionine (RPM) during the peripartum on the health parameters of dairy cows. Rumen-protected Met has reportedly improved milk yield, milk components, and liver health, but the literature is scarce on its effects in commercial herds. Therefore, we aimed to determine the effects of feeding RPM (Smartamine M, Adisseo Inc., Antony, France) prepartum (8 g/cow per day) and postpartum (15 g/cow per day) on performance, metabolic profile, and culling rate of Holstein cows in a commercial herd. One hundred sixty-six (n = 166) Holstein cows, 58 nulliparous and 108 parous, were randomly assigned to 1 of 2 dietary treatments, consisting of TMR top-dressed with RPM (RPMet; 2.35% and 2.24% Met of MP for close-up and fresh cows, respectively) or without (control [CON] 2.03% and 1.89% Met of MP for close-up and fresh cows, respectively), fed from 21 ± 6 d prepartum until 16 ± 5 d postpartum. From 17 DIM until dry-off, all cows received RPMet. Daily milk yield was recorded, and milk samples were collected in the first and second weeks after calving to determine their composition. Blood samples were collected before the morning feeding on -14, -7, +1, +7, and +14 d relative to calving. Mortality and morbidity were recorded during the first 60 DIM. Cows supplemented with RPMet had greater milk yield during the first 16 DIM (31.76 vs. 30.37 kg/d; SEM = 1.04, respectively), and had greater milk fat content (4.45 vs. 4.10%; SEM = 0.11, respectively), but not milk total protein (3.47 vs. 3.39%; SEM = 0.04, respectively) and casein contents (2.74 vs. 2.66%; SEM = 0.04, respectively) than CON cows. Cows in RPMet had increased plasma Met concentrations than cows in CON (24.9 vs. 21.0 µmol/L; SEM = 1.2, respectively). Although morbidity was similar between treatments, the culling rate from calving until 60 DIM was lower for RPMet cows than for CON cows (2.4% vs. 12.1%; SEM = 0.02). In conclusion, cows receiving RPMet have greater milk yield, improved milk fat content, and a lower culling rate at 60 DIM than CON cows.

Abstract:This experiment was conducted to investigate the effects of dietary rumen-protected betaine (RPB) supplementation, as partial replacement for methionine, on the lactation performance of mid-lactation dairy cows. A total of 36 Holstein dairy cows were randomly assigned to three groups [control, 20 g/day RPB, or 15 g/day rumen-protected methionine (RPM)]. The experiment was conducted over 9 weeks, with the first week for adaptation. Blood metabolites were analyzed with metabolomics in the control and RPB groups. The results revealed that the milk yield and milk protein content were higher in cows fed RPB and RPM compared to those in the control group. Concentrations of nine metabolites differed between cows in the RPB and control groups. These metabolites were mainly concentrated in six pathways, such as arginine synthesis and proline degradation and cyanoamino acid synthesis. This study revealed that RPB can spare methionine and improve lactation performance of dairy cows fed with diets moderately deficient in methionine.
Effects of rumen-protected methionine supplementation on the performance of high production dairy cows in the tropics

Abstract:Abstract Increasing methionine availability in dairy cow diets during the first third of lactation may enhance their performance and health. The aim of this study was to determine the effect of supplementing rumen-protected methionine (RPMet) in a lactation diet containing adequate protein and energy levels. Seventy-six multiparous Holstein cows (39.1 ± 6.8 kg of milk/d and 65 ± 28 DIM) were assigned to 1 of 2 dietary treatments (38/treatment) according to a randomized complete block design with a 2-wk (covariate) and 10-wk experimental period. Treatments were a basal diet (CON; 3.77 Lys:1Met); and CON + 14 g RPMet (2.97 Lys:1 Met). Individual milk samples were taken every 2 weeks to determine milk composition. Blood was collected from 24 cows on d+30 d to measure plasma AA levels. Body weight and body condition score (BCS) were measured at the beginning and the end of the experiment. The SM diet promoted higher milk yield (41.7 vs. 40.1 kg/d; P = 0.026), energy-corrected milk yield (41.0 vs. 38.0 kg/d; P ≤ 0.01), milk protein yield (1.30 vs. 1.18 kg/d; P ≤ 0.01), and milk fat yield (1.42 vs. 1.29 kg/d; P = 0.02). Higher milk protein (3.14% vs. 2.97%; P ≤ 0.01) and casein (2.39% vs. 2.28%; P ≤ 0.01) and a trend ( P = 0.06) for higher milk fat % (3.41% vs. 3.21%) were also observed. Both diets resulted in similar body weight, but CON-fed cows tended ( P = 0.08) to have higher BCS. Higher plasma methionine levels were determined with SM compared with CON (29.6 vs. 18.4 μ M ; P < 0.01), but lysine and histidine were not different. Dietary supplementation of RPM improved productive performance by increasing milk yield and milk components yields, suggesting better dietary AA utilization when Met levels are adjusted in Lys-adequate lactation diets.

Abstract:Providing essential amounts of balanced nutrients is one of the most vital aspects of livestock production. Among nutrients, protein has an essential role in many physiological functions of animals. Amino acids in needs for both high and medium yielding ruminant animals are not fully covered by microbial degraded feed sources in the rumen of animals, and they must be met by protecting the proteins from being broken down in the rumen; hence, the dietary supplementation of rumen-protected proteins (RPP), including mainly rumen-protected methionine (RPM), became imperative. Many researchers are interested in studying the role of (RPM) in ruminant animals concerning its effect on milk yield, growth performance, digestibility, dry matter intake and nitrogen utilization efficiency. Unfortunately, results obtained from several investigations regarding RPM indicated great fluctuation between its useful and useless effects in ruminant nutrition particularly during early and late lactation period; therefore, this review article may be helpful for ruminant farm owners when they decide to supplement RPM in animal's diet. Conclusively, supplementation of RPM often has a balanced positive influence, without any reported negative impact on milk yield, growth performance and blood parameters especially in early lactating ruminant animals and when used with the low crude protein diet.

Abstract:During the transition period, fatty liver syndrome may be caused in cows undergo negative energy balance, ketosis or hypocalcemia, retained placenta or mastitis problems. During the transition stage, movement of non-esterified fatty acids (NEFA) increases into blood which declines the hepatic metabolism or reproduction and consequently, lactation performance of dairy cows deteriorates. Most of studies documented that, choline is an essential nutrient which plays a key role to decrease fatty liver, NEFA proportion, improve synthesis of phosphatidylcholine, maintain lactation or physiological function and work as anti-oxidant in the transition period of dairy cows. Also, it has a role in the regulation of homocysteine absorption through betaine metabolite which significantly improves plasma α-tocopherol and interaction among choline, methionine and vitamin E. Many studies reported that, supplementation of rumen protected form of choline during transition time is a sustainable method as rumen protected choline (RPC) perform diverse functions like, increase glucose level or energy balance, fertility or milk production, methyl group metabolism, or signaling of cell methionine expansion or methylation reactions, neurotransmitter synthesis or betaine methylation, increase transport of lipids or lipoproteins efficiency and reduce NEFA or triacylglycerol, clinical or sub clinical mastitis and general morbidity in the transition dairy cows. The purpose of this review is that to elucidate the choline importance and functions in the transition period of dairy cows and deal all morbidity during transition or lactation period. Furthermore, further work is needed to conduct more studies on RPC requirements in dairy cows ration under different feeding conditions and also to elucidate the genetic and molecular mechanisms of choline in ruminants industry.

Abstract:Mastitis, the inflammatory condition of mammary glands, has been closely associated with immune suppression and imbalances between antioxidants and free radicals in cattle. During the periparturient period, dairy cows experience negative energy balance (NEB) due to metabolic stress, leading to elevated oxidative stress and compromised immunity. The resulting abnormal regulation of reactive oxygen species (ROS) and reactive nitrogen species (RNS), along with increased non-esterified fatty acids (NEFA) and β-hydroxybutyric acid (BHBA) are the key factors associated with suppressed immunity thereby increases susceptibility of dairy cattle to infections, including mastitis. Metabolic diseases such as ketosis and hypocalcemia indirectly contribute to mastitis vulnerability, exacerbated by compromised immune function and exposure to physical injuries. Oxidative stress, arising from disrupted balance between ROS generation and antioxidant availability during pregnancy and calving, further contributes to mastitis susceptibility. Metabolic stress, marked by excessive lipid mobilization, exacerbates immune depression and oxidative stress. These factors collectively compromise animal health, productive efficiency, and udder health during periparturient phases. Numerous studies have investigated nutrition-based strategies to counter these challenges. Specifically, amino acids, trace minerals, and vitamins have emerged as crucial contributors to udder health. This review comprehensively examines their roles in promoting udder health during the periparturient phase. Trace minerals like copper, selenium, and calcium, as well as vitamins; have demonstrated significant impacts on immune regulation and antioxidant defense. Vitamin B12 and vitamin E have shown promise in improving metabolic function and reducing oxidative stress followed by enhanced immunity. Additionally, amino acids play a pivotal role in maintaining cellular oxidative balance through their involvement in vital biosynthesis pathways. In conclusion, addressing periparturient mastitis requires a holistic understanding of the interplay between metabolic stress, immune regulation, and oxidative balance. The supplementation of essential amino acids, trace minerals, and vitamins emerges as a promising avenue to enhance udder health and overall productivity during this critical phase. This comprehensive review underscores the potential of nutritional interventions in mitigating periparturient bovine mastitis and lays the foundation for future research in this domain.

Abstract:This work was conducted to study the effect of rumen-protected choline (RPC) addition oil milk performance and blood metabolic parameters in transition dairy cows. In Experiment 1, fourteen Chinese Holstein dairy cows were Supplemented with 0 or 20 g/d of RPC from 7 d before expected calving to 21 d post partum. Feeding of RPC tended to increase milk yield and milk protein percentage. while milk fat and lactose percentage were not changed, Plasma concentrations of glucose tended to increase as cows consumed RPC. while plasma concentrations of triglycerides. very low density lipoproteins. cholesterol and nonesterified fatty acids were not significantly different between the two groups, In Experiment 2, thirty-six Chinese Holstein dairy cows were supplemented with 0, 30, 60 or 90 g/d RPC from 15 d before expected calving to 15 d post partum. Feeding of RPC tended to increase yield of milk and 4% fat-corrected milk for all the lactating cows, and milk composition was similar among the four groups, Plasma concentratiom of glucose were remained at a higher level in 30 or 60 g/d RPC-supplemented groups, and nonesterified fatty acids v ere decreased ill the 30 g/d group. Concentrations of triglycerides tended to reduce in 30 and 90 g/d RPC-supplemented animals. and cholesterol was reduced in 0 or 30 g/d group. These results suggest that RPC addition tended to increase milk field and improve blood metabolic parameters during transition dairy Cows, and feeding 30 g/d of RPC may be the optimal.

Abstract:Research articles about the effects of rumen-protected choline (RPC) supplementation on metabolic response, and reproductive and productive performances in transitional dairy cows were reviewed and presented. Analysis was conducted on 32 research papers that were published. The papers examined treatments without RPC supplementation and RPC supplementation levels varying from 10 to 100 gm/day. The feeding duration of RPC started from 40 to 140 days prepartum and continued until 20 to 140 days postpartum in multiparous dairy cows. Studies indicated that adding herbal choline (Cho) to the diet of transition dairy cows resulted in increased milk production and improved milk quality, leading to enhanced energy balance and reduced oxidative stress. The concentration and yield of IgG in colostrum provide passive immunity to dairy newborns and can be enhanced by dietary Cho supplementation. The inconsistent effects of RPC supplementation on reproduction may be due to several factors such as heat stress, genetics, and management. RPC supplementation improved the transition dairy cows’ milk yield and quality, but dosage response was not observed as in the prior publication. Remarkably, the length of RPC supplementation had a positive correlation with an increase in milk yield. Based on this review, 45–50 gm/day dietary RPC supplementation between 3 weeks pre-calving to 8 weeks post-calving is suggested to increase at least 10% milk yield in dairy cows.

Abstract:10050 Background: The incidence of colorectal cancer (CRC), one of the commonest malignancies worldwide, is still increasing. Despite advances in the diagnostic procedure, a large number of patients with CRC still presents with advanced disease. More sensitive and efficient diagnostic technique would make an impact on the prognosis of CRC patients through improving their compliance to the screening program, hence allowing earlier detection of the disease. To test the hypothesis that p16 methylation may serve as a marker for the diagnosis of CRC, we quantified the methylation levels of p16 in the serum DNA of CRC patients. METHODS Fresh specimens of 168 CRC and corresponding noncancerous tissues were obtained surgically at the Department of Surgery II, Nagoya University Graduate School of Medicine, together with the corresponding serum samples obtained 1 week prior to surgery. We defined the p16 methylation rate (p16 MR, in %) as follows: CM/(CM + CU) × 100%. CM is the concentration of methylated p16 sequences and CU is the concentration of unmethylated p16 sequences measured by quantitative methylation-specific PCR (Q-MSP) after bisulfite conversion. The relationship between the p16 MR and clinicopathologic findings were evaluated. RESULTS Aberrant p16 promoter methylation was found in 59% (99 of 168) of surgically resected CRC tissues. None of the corresponding normal tissues had methylated p16 sequences. 37% (37 of 99) serum samples of the CRC patients with tumoral p16 methylation had the same alterations. p16 MRs of 99 serum samples with tumoral p16 methylation were 0 to 68.9 (mean was 5.43±11.01). p16 MRs were significantly correlated with lymph node metastasis (P=0.001), lymphatic invasion (P=0.001) and venous invasion (P=0.020) of CRCs. p16 MRs increased significantly with tumor stage [stage I : 0.94 ± 1.68, stage II : 2.33 ± 5.19, stage III : 8.49 ± 14.22, stage IV : 10.03 ± 14.27 (P = 0.021)]. Moreover, the survival of patients with low p16 MR was significantly superior to those with high p16 MR (P=0.006). CONCLUSIONS These results suggest that quantification of p16 methylation in the serum DNA might be a novel diagnostic tool and a prognostic factor for CRC. Abstract:Overproduction of reactive oxygen species (ROS) is a well-known phenomenon experienced by ruminants, especially during the transition from late gestation to successful lactation. This overproduction of ROS may lead to oxidative stress (OS), which compromises the immune and anti-inflammatory systems of animals, thus predisposing them to health issues. Besides, during the periparturient period, metabolic stress is developed due to a negative energy balance, which is followed by excessive fat mobilization and poor production performance. Excessive lipolysis causes immune suppression, abnormal regulation of inflammation, and enhanced oxidative stress. Indeed, OS plays a key role in regulating the metabolic activity of various organs and the productivity of farm animals. For example, rapid fetal growth and the production of large amounts of colostrum and milk, as well as an increase in both maternal and fetal metabolism, result in increased ROS production and an increased need for micronutrients, including antioxidants, during the last trimester of pregnancy and at the start of lactation. Oxidative stress is generally neutralized by the natural antioxidant system in the body. However, in some special phases, such as the periparturient period, the animal's natural antioxidant system is unable to cope with the situation. The effect of rumen-protected limiting amino acids and choline on the regulation of immunity, antioxidative, and anti-inflammatory status and milk production performance, has been widely studied in ruminants. Thus, in the current review, we gathered and interpreted the data on this topic, especially during the perinatal and lactational stages.
