Abstract:A new eremophilane sesquiterpene, capnodiumone (1), along with five known eremophilane sesquiterpenes (2-6) and eight other compounds (7-14), have been isolated from a mangrove-associated fungus Capnodium sp. SZ-F22. The chemical structures were elucidated on the basis of extensive spectroscopic analysis. The broth extract of the fungus exhibited a good inhibitory effect on the mycelium growth against Fusarium graminearum at 100g/mL, however, all the 14 compounds showed no expected antifungal activity. Abstract:From the fruits of Celastrus orbiculatus Thunb. a new beta-dihydroagarofuran sesquiterpene ester named 1beta,2beta,13-triacetoxy-9alpha-cinnamoyloxy-beta-dihydroagarofuran (1) has been isolated along with two known compounds, 6alpha-acetoxy-1beta, 9beta-dibenzoyloxy-8beta-hydroxy-beta-dihydroagarofuran (2), and 1beta, 6alpha-diacetoxy-9beta-benzoyloxy-8beta-hydroxy-beta-dihydroagarofuran ( 3). Their structures were elucidated on the basis of spectroscopic data.

Abstract:Ten novel (1, 2, 3a, 3b, 4a, 4b, 5a, 5b, 6a, and 6b) furancarboxylic acids including four pairs of epimers (3a, 3b; 4a, 4b; 5a, 5b; 6a, 6b), together with seven known analogues (7a, 7b, 8a, 8b, 9a, 9b, and 10), were isolated from the fermentation of the soil-derived fungus Penicillium sp. sb62. Their structures were established on the basis of spectroscopic data analysis, and the absolute configurations were determined by time-dependent density functional theory electronic circular dichroism calculations, comparison of the specific optical rotation values, and modified Mosher's method. Compounds 1-4 represent the first class of natural furancarboxylic acids featuring a thiophene moiety. Compounds 1-7 showed antimicrobial inhibitory activities against Escherichia coli, Staphylococcus aureus, and Candida albicans with MIC values ranging from 0.9 to 7.0 μg/mL, from 1.7 to 3.5 μg/mL, and from 3.3 to 7.0 μg/mL, respectively.

Abstract:Marine-derived fungi of the genus Penicillium represent a huge potential for synthesizing the secondary metabolites with structural and bioactive uniqueness and diversity. In this study, six previously undescribed compounds peniciphenalenins A-F and four known compounds (+)-sclerodin, (+)-scleroderolide, (+)-sclerodione, and physcion were isolated from the culture of a marine-derived fungus Penicillium sp. ZZ901. Structures of the isolated compounds were elucidated by a combination of extensive NMR spectroscopic analysis, HRESIMS data, optical rotation value, ECD calculation, and single crystal X-ray diffraction. Peniciphenalenins A-C are the second examples of the type of neoherqueinones. The possible biosynthetic route of nine phenalenone derivatives has been suggested. The known (+)-scleroderolide showed both antiproliferative activity against glioma cells with IC50 values of 23.24-37.26 mu M and antibacterial activity in suppressing the growth of methicillin-resistant Staphylococcus aureus and Escherichia call with MIC values of 7.0 and 9.0 mu g/mL, respectively. Abstract:Three new a-pyrones, scirpyrones AC (13), and four new pyranes, scirpyranes AD (47), were isolated from solid cultures of the plant pathogen Pestalotiopsis scirpina. Their structures were elucidated primarily by NMR experiments. The absolute configurations of C-7 and C-2' in 1, C-7 in 2, and C-6 and C-7 in 3 were deduced on the basis of circular dichroism (CD) data, whereas those of the secondary alcohols in 47 were determined by a modification of Mosher's method. Compound 1 is the first described 6-(5-oxotetrahydrofuran-2-yl)-2H-pyran-2-one. Compounds 46 showed significant cytotoxicity towards MCF-7 cells.

Abstract:Six new caryophyllene-based sesquiterpenoids named punctaporonins H-M (1-6), together with punctaporonin B (7) and humulane (8) were isolated from the fermentation broth of the sponge-derived fungus Hansfordia sinuosae. Their structures were determined by the extensive HRESIMS and NMR spectroscopic analysis, including the X-ray crystallographic data for the assignment of the absolute configurations of punctaporonins H-I (1-2). The isolated compounds were evaluated for antihyperlipidemic, cytotoxic and antimicrobial activities, and punctaporonin K (4) exhibited potent effects to reduce the triglycerides and total cholesterol in the intracellular levels.

Abstract:Two novel 3,5-dimethylorsellinic acid derived meroterpenoids, peniciterpenolides A (1) and B (2) individually containing a 2-oxaspiro[4,5]deca-6,8-diene-1,4-dione and a 6-oxabicyclic[3,2,1]oct-3-en-7-one motif, one new spongian-type diterpene, peniciterpenolide C (3), and one new drimane-type sesquiterpene, peniciterpenolide D (4), together with the known meroterpenoid atlantinone B (5) were isolated from Penicillium chrysogenum MT-40, an endophytic fungus identified from Huperzia serrata. All the five compounds additionally contain a unique 2-oxabicyclo[2.2.2]octan-3-one moiety formed by the annulation of the C-3 hydroxyl and the oxidized methyl on C-10. The structures were established by the analysis of their spectroscopic data, single-crystal X-ray diffraction, and quantum chemical calculations of electronic circular dichroism spectra. Compounds 1-5 showed inhibition of nitric oxide production in lipopolysaccharide-activated RAW 264.7 macrophage cells with IC50 values in the range of 20.6-46.9 mu M.

Abstract:Two new sesterterpenes together with related known terpestacin were isolated from the cultural broth of a marine fungal strain (5XNZ5-4) that lives in the gut of a marine crab. The new compounds were identified on the basis of analyses of NMR and mass spectral data. This is the first report on the oxidative derivatives of terpestacin. The new compounds showed no activity in antibacterial and cytotoxicity assays.

Abstract:Two pairs of new enantiomeric hydroxyphenylacetic acid derivatives, (±)‐corylophenols A and B ((±)‐1 and (±)‐2), a new α‐pyrone analogue, corylopyrone A (3), and six andrastin‐type meroterpenoids (4–9) were isolated and identified from the deep‐sea cold‐seep sediment‐derived fungus Penicillium corylophilum CS‐682. Their structures and stereo configurations were determined by detailed spectroscopic analysis of NMR and MS data, chiral HPLC analysis, J‐based configuration analysis, and quantum chemical calculations of ECD, specific rotation, and NMR (with DP4+ probability analysis). Compound 3 showed inhibitory activity against some strains of pathogenic bacteria.

Abstract:Four new polyketides including two arthrinic acid derivatives (1-2), one phenolic derivative (3) and (S)-3-hydroxy-6-(2-hydroxypropyl)-5-methyl-2H-pyran-2-one (4) along with one methyl ester of arthrinic acid (5) were isolated from the culture broth of Arthrinium sp., which was an entophytic fungus of clam worm. Their structures were identified on the basis of HR-ESI-MS and NMR spectral analyses together with advanced Mosher's method. In the assay of inhibiting the prostate cancer PC3 cell line, none of the isolated compounds showed significant cytotoxicity.

Abstract:Chemical examination of an unidentified sponge associated marine fungus Penicillium terrestre resulted in the isolation of eight compounds. Their structures are identified as cyclo (glycyl-D-proline) (1), cyclo (L-prolyl-L-alanine) (2), cyclo (L-prolylD-alanine) (3), cyclo (L-leucyl-trans-4-hydroxy-L-proline) (4), cyclo (L-leucyl-cis-4-hydroxy-D-proline) (5), cyclo (L-trans-(4-hydroxyprolinyl)-L-phenylalanine) (6), hexylitaconic acid (7), and hexylitaconic methylate (8), based on spectroscopic analysis (1D, 2D NMR). Compounds 7, 8 were obtained from the genus Penicillium for the first time, while compounds 1-6 were isolated from the fungus Penicillium terrestre for the first time.

Abstract:To study the chemical constituents of deep-sea fungus Penicillium sp. F00120,nine compounds were isolated from its Et OAc extract using silica gel,Sephadex LH-20,ODS and HPLC methods. Based on the spectroscopic analysis,their structures were identified as ergosterol(1),4-methyl-5,6-dihydro-2-pyranone(2),citreohybridonol(3),1-Linoleoylglycerol(4),5α,6α-Epoxy-(22 E,24R)-ergosta-8(14),22-diene-3β,7β-diol(5),macrophorin(6),purpurogemutantin(7),3-(2S,4S-dihydroxypentyl)-6,8-dihydroxyisocoumarin(8),5-hydroxy-7-(2’-hydroxypropyl)-2-methylchromone(9). Compounds 2-4,6-9 were isolated from Penicillium sp. F00120 for the first time.

Abstract:Penigrines A−E ( 1 − 5 ), five undescribed azepine-indole alkaloids, were isolated from the fungus Penicillium griseofulvum . Their structures with absolute configurations were determined by NMR, HRESIMS, ECD calculation, and X-ray diffraction experiments. Penigrine C ( 3 ) possesses an undescribed 6-oxa-8-azabicyclo[3.2.2]nonane-7,9-dione moiety that fused to an indole core, and penigrines D and E ( 4 and 5 ) are a pair of epimers. The plausible biosynthetic pathways of 1 − 5 are proposed. Penigrine A ( 1 ) shows the potential for heart failure treatment.

Abstract:Four unusual steckwaic acids E–H (1–4), possessing a rarely described acrylic acid unit at C-4 (1–3) or a double bond between C-12 and C-13 (4) are reported for the first time, along with four new analogues (5–8) and two known congeners (9 and 10). They were purified from the organic extract of Penicillium steckii AS-324, an endozoic fungus obtained from a deep-sea coral Acanthogorgiidae sp., which was collected from the Magellan Seamount at a depth of 1458 m. Their structures were determined by the interpretation of NMR and mass spectroscopic data. The relative and absolute configurations were determined by NOESY correlations, X-ray crystallographic analysis, and ECD calculations. All compounds were tested for their antimicrobial activities against human- and aquatic-pathogenic bacteria and plant-related pathogenic fungi.

Abstract:Heparin-induced thrombocytopenia (HIT) is a life-threatening, thrombotic disorder associated with development of anti-platelet factor 4 (anti-PF4)/heparin autoantibodies. Little is known about the antigenic and cellular requirements that initiate the immune response to these complexes. To begin to delineate mechanisms of autoantibody formation in HIT, we studied the immunizing effects of murine PF4 (mPF4)/heparin in mice with and without thymic function. Euthymic mice were injected with mPF4/heparin complexes, mPF4, heparin, or buffer. Mice injected with mPF4/heparin, but not mPF4 or heparin alone, developed heparin-dependent autoantibodies that shared serologic and functional characteristics of human HIT antibodies, including preferential binding to mPF4/heparin complexes and causing heparin- and FcRgammaIIA-dependent platelet activation. In contrast, athymic mice did not develop HIT-like antibodies. Taken together, these studies establish that PF4/heparin complexes are highly immunogenic and elicit self-reacting anti-PF4/heparin antibodies in a T cell-dependent manner.
[Quality control of blood glucose self-monitoring using the Glucosignal test strip and Haemo-Glucotest 20-800].

Abstract:In 157 capillary blood samples of type I diabetics parallel determinations of the blood sugar concentration were performed by means of the visual evaluation of the test strips Glucosignal and Haemo-Glucotest 20-800 in comparison to an enzymatic reference method by trained special personnel. On the basis of the correlation coefficients the two test strips showed a good accordance between visual evaluation and the reference method (r = 0.98 and 0.97, respectively). In comparison to the reference method in Glucosignal there is a tendency to underestimation (-0.67 mmol/l), on the other hand in the Haemo-Glucotest 20-800 the blood glucose concentrations are on an average +0.87 mmol/l above the reference value. In visual valuation of the blood glucose concentrations by means of Haemo-Glucotest 20-800 with increasing blood glucose concentration an increasing inexactness is to be stated, whereas Glucosignal is characterized by more favourable parameters of quality.
