Abstract:Ocean acidification (OA) can affect marine bivalves at various levels of biological organization. Yet, little effort has been devoted to understanding how OA affects the reproductive events of marine bivalves during multiple cycles of maturation. Here, we tested sex-specific reproductive responses of Manila clams (Ruditapes philippinarum) to OA during gonadal rematuration. Under acidified conditions, both male and female clams exhibited delayed gonadal rematuration following spawning and impairments in gonadal tissues, which can be likely ascribed to lowered concentrations of hormones and vitellogenin. The findings indicate that marine bivalves experience significant declines in reproductive capacity as a result of OA during their reproductive cycles, with clear sex-specific differences. Consequently, it is essential to consider sex-specific reproduction responses of marine bivalves to OA when developing conservation strategies and forecasting population sustainability in a rapidly acidifying marine environment.

Editorial Review of Personal Epistemologies and the Learning Paradox in Teacher Education: A Neglected Dilemma

Abstract:In this chapter, we discuss the role that personal epistemologies play in teacher education, particularly with respect to the potential problems and roadblocks they may present. We suggest that epistemological beliefs govern the kind of knowledge that preservice teachers consider to be legitimate and worthwhile learning in their programs, regulate the ways in which they make choices among competing knowledge claims and justify their own choices. A body of research has examined epistemic cognition as a dimension of the cognitive growth that occurs during the college years (e.g., BaxterMagolda, 2002; King & Kitchener, 2002; Perry, 1970), and recently, the role of personal epistemologies of preservice teachers, teachers, and teacher education professors in teacher education has been investigated (Schraw, Bendixen, & Dunckle, 2002; Joram, 2007: Shepard, 2000). However, relatively less work has been devoted to developing ways to modify preservice teachers’ personal epistemologies. Notwithstanding the few studies that have been conducted in this area, we argue that teacher educators need to design learning activities that may help change epistemic orientations that have potentially negative consequences for teacher education. We suggest that examining literature on the learning paradox, with particular attention to the role of metaphor in learning, offers inroads to accomplishing these goals. Drawing on this theoretical background, we identify four approaches teacher educators can take to teach more effectively by taking their students’ personal epistemologies into account. Assessment of the juvenile vulnerability of symbiont-bearing giant clams to ocean acidification

Abstract:Ocean acidification (OA) severely affects marine bivalves, especially their calcification processes. However, very little is known about the fate of symbiont-bearing giant clams in the acidified oceans, which hinders our ability to develop strategies to protect this ecologically and economically important group in coral reef ecosystems. Here, we explored the integrated juvenile responses of fluted giant clam Tridacna squamosa (Lamarck, 1819) to acidified seawater at different levels of biological organization. Our results revealed that OA did not cause a significant reduction in survival and shell growth performance, indicating that T. squamosa juveniles are tolerated to moderate acidification. Yet, significantly reduced net calcification rate demonstrated the calcifying physiology sensitivity to OA, in line with significant declines in symbiont photosynthetic yield and zooxanthellae density which in turn lowered the amount of energy supply for energetically expensive calcification processes. Subsequent transcriptome sequencing and comparative analysis of differentially expressed genes revealed that the regulation of calcification processes, such as transport of calcification substrates, acid-base regulation, synthesis of organic matrix in the calcifying fluid, as well as metabolic depression were the major response to OA. Taken together, the integration of physiological and molecular responses can provide a comprehensive understanding of how the early life history stages of giant clams respond to OA and make an important leap forward in assessing their fate under future ocean conditions.

Abstract:Ocean acidification and hypoxia have become increasingly severe in coastal areas, and their co-occurrence poses emerging threats to coastal ecosystems. Here, we investigated the combined effects of ocean acidification and hypoxia on the reproductive capacity of the thick-shelled mussel Mytilus coruscus. Our results demonstrated low pH but not low oxygen induced decreased gonadosomatic index (GSI) in mussels. Male mussels had a lower level of sex steroids (estradiol, testosterone, and progesterone) when kept at low pH. Expression of genes related to reproduction were also impacted by low pH with a downregulation of genes involved in gonad development in males (β-catenin and Wnt-7b involved in males) and an upregulation of testosterone synthesis inhibition-related gene (Wnt-4) in females. Overall, our results suggest that ocean acidification has an impact on the gonadal development through an alternation of gene expression and level of steroids while hypoxia had no significant effect.

Abstract:Whether sex determination of marine organisms can be altered by ocean acidification and warming during this century remains a significant, unanswered question. Here, we show that exposure of the protandric hermaphrodite oyster, Saccostrea glomerata to ocean acidification, but not warming, alters sex determination resulting in changes in sex ratios. After just one reproductive cycle there were 16% more females than males. The rate of gametogenesis, gonad area, fecundity, shell length, extracellular pH and survival decreased in response to ocean acidification. Warming as a sole stressor slightly increased the rate of gametogenesis, gonad area and fecundity, but this increase was masked by the impact of ocean acidification at a level predicted for this century. Alterations to sex determination, sex ratios and reproductive capacity will have flow on effects to reduce larval supply and population size of oysters and potentially other marine organisms.

Abstract:Ocean acidification (OA) can have widespread implications for marine bivalves. While our current understanding of OA effects on the physiological performance is increasing, very little is known about the physiology of digestive gland of marine bivalves in response to OA. Here, we examined how the digestive system of razor clams ( Sinonovacula constricta ) responded to OA. Following 35-day exposure to CO 2 -driven seawater acidification, no significant decreases in phenotypic traits, such as dry body weight gain, specific growth rate, condition index and survival rate, as well physiological functions, such as activities of antioxidant and digestive enzymes, were observed, demonstrating the resistance of razor clams under acidified conditions. Histological results showed that some direct damages on the structure of digestive gland was observed, including degradation of digestive tubular, atrophy of epithelial cells, loose cell arrangement, even diffuse. This study provides insights into the digestive performance of marine bivalves in a rapidly acidifying ocean.

Abstract:The Manila clam ( Ruditapes philippinarum ) is a commercially important marine bivalve, which inhabits the estuarine and mudflat areas. The osmoregulation is of great significance for molluscs adaptation to salinity fluctuations. In this study, we investigated the effects of low salinity (10 psu) and high salinity (40 psu) stress on survival and osmoregulation of the R. philippinarum . The results of physiological parameters showed that the ion (Na + , K + , Cl − ) concentrations and Na + /K + -ATPase (NKA) activity of R. philippinarum decreased significantly under low salinity stress, but increased significantly under high salinity stress, indicating that there are differences in physiological adaptation of osmoregulation of R. philippinarum. In addition, we conducted the transcriptome analysis in the gills of R. philippinarum exposed to low (10 psu) and high (40 psu) salinity challenge for 48 h using RNA-seq technology. A total of 153 and 640 differentially expressed genes (DEGs) were identified in the low salinity (LS) group and high salinity (HS) group, respectively. The immune ( IAP, TLR6, C1QL4 , Ank3 ), ion transport ( Slc34a2 , SLC39A14 ), energy metabolism ( PCK1 , LDLRA , ACOX1 ) and DNA damage repair-related genes ( Gadd45g , HSP70B2 , GATA4 ) as well as FoxO, protein processing in endoplasmic reticulum and endocytosis pathways were involved in osmoregulation under low salinity stress of R. philippinarum . Conversely, the ion transport ( SLC6A7 , SLC6A9 , SLC6A14 , TRPM2 ), amino acid metabolism ( GS , TauD , ABAT , ALDH4A1 ) and immune-related genes ( MAP2K6 , BIRC7A , CTSK , GVIN1 ), and amino acid metabolism pathways (beta-Alanine, Alanine, aspartate and glutamate, Glutathione) were involved in the process of osmoregulation under high salinity stress. The results obtained here revealed the difference of osmoregulation mechanism of R. philippinarum under low and high salinity stress through physiological and molecular levels. This study contributes to the assessment of salinity adaptation of bivalves in the context of climate change and provides useful information for marine resource conservation and aquaculture.

Abstract:In the past decade, the frequency, intensity and duration of marine heatwaves (MHWs) in the South China Sea have been increasing strikingly, resulting in serious impacts on intertidal bivalves and their ecosystems. The Manila clam, Ruditapes philippinarum, is one of the most ecologically and economically important bivalve species in the South China Sea, yet very little is known about its fate under intensifying MHWs events. Here, we examined how R. philippinarum responded to two consecutive scenarios of MHWs, with each composed of 4 °C and 8 °C rises of seawater temperatures, respectively. Up to 87% of Manila clams survived recurrent MHWs events, and significant increases in standard metabolic rate occurred predominantly under extreme conditions (+8 °C), indicating that the clams could trigger compensatory mechanisms to mitigate MHWs-induced thermal stress. Following acute and repeated exposures to MHWs, Manila clams showed similar responses in enzymes underpinning energy metabolism (NKA, CMA, and T-ATP), antioxidant defence (SOD, CAT, and MDA), and biomineralization (AKP and ACP), most of which exhibited significantly increasing and then decreasing trends with the intensification of MHWs. Of eight genes associated with physiological tolerance and fitness, ATAD3A, PFK, SOD, and C3 were significantly down-regulated in response to recurrent MHWs events, demonstrating the certain resistance to MHWs. These findings provide a better understanding that marine bivalves hold the potential to acclimate simulated MHWs events from the physiological and molecular processes.

Abstract:Gender differences in the hemocyte immune response of Hong Kong oyster Crassostrea hongkongensis to Vibrio harveyi and lipopolysaccharide (LPS) infection exist. To determine if a gender difference also exists, we use a 1H NMR-based metabolomics method to investigate responses in C. hongkongensis hepatopancreas tissues to V. harveyi and LPS infection. Both infections induced pronounced gender- and immune-specific metabolic responses in hepatopancreas tissues. Responses are mainly presented in changes in substances involved in energy metabolism (decreased glucose, ATP, and AMP in males and increased ATP and AMP in LPS-infected females), oxidative stress (decreased glutathione in males and decreased tryptophan and phenylalanine and increased choline and proline in LPS-infected females), tricarboxylic acid (TCA) cycle (decreased α-ketoglutarate acid and increased fumarate in LPS-infected males, and decreased fumarate in LPS-infected females), and osmotic regulation (decreased trigonelline and increased taurine in V. harveyi-infected males and decreased betaine in V. harveyi-infected females). Results suggest that post-spawning-phase male oysters have a more significant energy metabolic response and greater ability to cope with oxidative stress than female oysters. We propose that the impact of oyster gender should be taken into consideration in the aftermath of oyster farming or oyster disease in natural seas.

Abstract:Ocean acidification (OA), which reduces ocean pH and leads to substantial changes in seawater carbonate chemistry, may strongly impact organisms, especially those with carbonate skeletons. In marine molluscs, while the physiological effects of OA are well known, with a reduction of growth and shell calcification, there are few studies on behavioural effects. A large marine gastropod, Haliotis tuberculata, was exposed to ambient (pH T 8.0) or low pH (pH T 7.7) during a 5-month experiment. Because animal fitness can be affected through various behavioural changes, a broad spectrum of behavioural parameters was investigated, including situations involving no stress, responses to predators, righting to evaluate indirectly the level of energy reserves, and finally, reproductive behaviour. In addition, we measured the expression profile of the GABA A-like and serotonin receptor genes, often described as central neuromodulators of sensory performance and behaviour and known to be affected by OA in molluscs. No significant effect of low pH as compared to ambient pH was observed on abalone behaviour for any of these behavioural traits or gene expressions after either one week or several months of exposure to OA. The significance tests were corroborated by estimating the size of pH effects. The behaviour of this mollusc appears not to be affected by pH decrease expected by the end of the century, suggesting some resilience of the species to OA at the adult stage. This is probably related to the ecological niche of this abalone, where important pH variations can be observed at tidal, diurnal or seasonal scales.

Abstract:With the unprecedented environmental changes caused by climate change including ocean acidification, it has become crucial to understand the responses and adaptive capacity of fish to better predict directional changes in the ecological landscape of the future. We conducted a systematic literature review to examine if simulated ocean acidification (sOA) could influence growth and reproduction in fish within the dynamic energy budget theory framework. As such, we chose to examine metabolic rate, locomotion, food assimilation and growth in early life stages (i.e. larvae and juvenile) and adults. Our goal was to evaluate if acclimatization to sOA has any directional changes in these traits and to explore potential implications for energetic trade-offs in these for growth and reproduction. We found that sOA had negligible effects on energetic expenditure for maintenance and aerobic metabolism due to the robust physiological capacity regulating acid–base and ion perturbations but substantive effects on locomotion, food assimilation and growth. We demonstrated evidence that sOA significantly reduced growth performance of fish in early life stages, which may have resulted from reduced food intake and digestion efficiency. Also, our results showed that sOA may enhance reproduction with increased numbers of offspring although this may come at the cost of altered reproductive behaviours or offspring fitness. While these results indicate evidence for changes in energy budgets because of physiological acclimatization to sOA, the heterogeneity of results in the literature suggests that physiological and neural mechanisms need to be clearly elucidated in future studies. Lastly, most studies on sOA have been conducted on early life stages, which necessitates that more studies should be conducted on adults to understand reproductive success and thus better predict cohort and population dynamics under ongoing climate change. Ocean acidification (OA) could alter energy budgets of marine fish. OA had negligible effects on aerobic metabolism, but substantive effects on locomotion, food assimilation and growth, suggesting a potential shift in energy budgets in the future. Also, OA may enhance reproduction potentially at the cost of altered reproductive behaviours or offspring fitness.

Abstract:Abstract Marine bivalves secrete calcified shells to protect their soft bodies from predation and damages, which is of great importance for their survival, and for the safety of the coastal ecosystem. In recent years, larval shell formation of marine bivalves has been severely affected by ocean acidification (OA), and previous study indicated that OA might affect such process by disrupting endogenous energy metabolism. Developmental stages from trochophore to D-shape larvae are extremely important for initial shell formation in oyster since a calcified shell was formed to cover the chitin one. In the present study, metabolomic and transcriptomic approaches were employed to investigate the energy metabolism of oyster larvae during initial shell (prodissoconch I, PDI shell) formation and under experimental OA treatment. Totally 230 chemical compounds were identified from the present dataset, most of which were highly expressed in the “middle” stage (early D-shape larvae) which was critical for PDI shell formation since a calcified shell was formed to cover the chitin one. Several compounds such as glucose, glutarylcarnitine (C5), β-hydroxyisovaleroylcarnitine, 5-methylthioadenosine (MTA), myristoleate (14:1n5) and palmitoleate (16:1n7) were identified, which were involved in energy metabolic processes including amino acid oxidation, glycolysis, pentose phosphate pathway and fatty acid metabolism. In addition, mRNA expressions of genes related to protein metabolism, glycolysis, lipid degradation, calcium transport and organic matrix formation activities were significantly down-regulated upon experimental OA. These results collectively suggested that formation of the initial shell in oyster larvae required endogenous energy coming from amino acid oxidation, glycolysis, pentose phosphate pathway and fatty acid metabolism. These metabolic activities could be severely inhibited by experimental OA, which might alter the allocation of endogenous energy. Insufficient endogenous energy supply then suppressed the mobilization of calcium and resulted in a failure or delay in PDI shell formation.

Abstract:The combined effects of emerging pollutants and ocean acidification (OA) on marine organisms and marine ecosystems have attracted increasing attention. However, the combined effects of tralopyril and OA on marine organisms and marine ecosystems remain unclear. In this study, Crassostrea gigas (C. gigas) were exposed to tralopyril (1 μg/L) and/or OA (PH = 7.7) for 21 days and a 14-day recovery acclimation. To investigate the stress response and potential molecular mechanisms of C. gigas to OA and tralopyril exposure alone or in combination, as well as the effects of OA and/or tralopyril on bivalve biomineralization and marine carbon cycling. The results showed that the combined toxicity was between that of acidification and tralopyril alone. Single or combined exposure activated the general stress defense responses of C. gigas mantle, affected energy metabolism and biomineralization of the organism and the carbon cycle of the marine ecosystem. Moreover, acidification-induced and tralopyril-induced toxicity showed potential recoverability at molecular and biochemical levels. This study provides a new perspective on the molecular mechanisms of tralopyril toxicity to bivalve shellfish and reveals the potential role of tralopyril and OA on marine carbon cycling.

Abstract:Increasing temperature is causing rapid changes in oceanic environments. Understanding the responses of marine animals to such changes remains a major challenge, yet it is key to predicting future biodiversity loss or gain. In this study, we investigated molecular mechanisms of adaptation to acute temperature change in low and high latitude populations of the Manila clam Ruditapes philippinarum. Using transcriptome analysis, we studied temperature-responsive genes and pathways in clams cultured under acute temperature stress (−1°C and 28°C) and identified an extensive set of genes and pathways that respond to acute temperature stress. Physiological changes at the molecular level were compared to explore potential differences in the response of clams and their adaptation to temperature change and to uncover the underlying mechanisms of resilience or sensitivity to stress conditions in the two clam populations. The enrichment of genes involved in protein processing in the endoplasmic reticulum was integral to the adaptation of clams to stressful conditions. Furthermore, phagosome-related genes and pathways were significantly enriched under acute high temperature stress. The upregulation of xenobiotic metabolism by cytochrome P450 showed a population-specific response in the low-latitude clams under both heat and cold stress but not in the high-latitude clams, whereas genes encoding peroxisomes were upregulated in high-latitude clams under heat stress but not in low-latitude clams. This study provides useful information related to the response of R. philippinarum to temperature stress, which could be helpful for improving the temperature stress resistance of these clams in the aquaculture industry. Our results may also help predict future biodiversity trends under temperature change scenarios.

Abstract:Intertidal organisms, especially the sessile species, often experience long-term periodic air exposure during their lives. Learning the biochemical and physiological responses of intertidal organisms to long-term periodic air exposure and the relationship to duration of air exposure provides insight into adaptation to this variably stressful environment. We studied the Manila clam, Ruditapes philippinarum, an important species in world aquaculture, as a model to evaluate survival, growth, lipid composition, oxygen consumption, oxidative damage, and antioxidant enzyme activity in relation to the duration of air exposure in a long-term (60 days) laboratory study of varying durations of periodic emersion and re-immersion. Our results show: (1) clams undergoing a longer period of air exposure had lower survival and growth compared to those given a shorter exposure, (2) levels of oxidative damage and activities of antioxidant enzymes were higher in all air exposure treatments, but did not increase with duration of air exposure, and (3) the content of docosahexaenoic acid increased with duration of air exposure. Our results can largely be interpreted in the context of the energy expenditure by the clams caused by aerobic metabolism during the daily cycle of emersion and re-immersion and the roles of docosahexaenoic acid against oxidative stress.

Abstract:Rapidly increasing concentration of carbon dioxide (CO 2 ) in the atmosphere not only results in global warming , but also drives increasing seawater acidification. Infaunal bivalves play critical roles in benthic-pelagic coupling, but little is known about their behavioral responses to compound climate events. Here, we tested how heatwaves and acidification affected the behavior of Manila clams ( Ruditapes philippinarum ). Under acidified conditions, the clams remained capable of burrowing into sediments. Yet, when heatwaves attacked, significant decreases in burrowing ability occurred. Following two consecutive events of heatwaves, the clams exhibited rapid behavioral acclimation. The present study showed that the behavior of R. philippinarum is more sensitive to heatwaves than acidification. Given that the behavior can act as an early and sensitive indicator of the fitness of intertidal bivalves, whether, and to what extent, behavioral acclimation can persist under scenarios of intensifying heatwaves in the context of ocean acidification deserve further investigations.

Abstract:Salinity is an important environmental factor affecting the growth and survival of marine organisms. Marine shellfish possess the efficient osmoregulatory mechanism to cope with either hypo- or hyperosmotic stress. However, little is known about the molecular and metabolic adaptation involved in the osmotic regulation of these intertidal habitants. The Manila clam Ruditapes philippinarum is a worldwide cultured intertidal shellfish, which usually encounters a great risk of salinity stress in its coastal habitats. In the present study, the integrated analysis of transcriptomics and metabolomics was performed to investigate the transcriptional and metabolic profiles of the clams in response to abrupt salinity decrease. The comparative transcriptome analysis uncovers the changes of genes involved in ion channel activation, amino acid metabolism, and energy metabolism in the salinity-stressed condition. The metabolomics analysis reveals that the glucose-succinate pathway may play a critical role in energy metabolism during the osmotic stress. The multi-omics analysis reveals that neural adaptation to salinity stress may be potentially regulated by arachidonic acid metabolism and glutathione metabolism. The important metabolites (12-HPETE, PGEs, and spermine) may be served as the signaling molecules in response to acute salinity stress in the intertidal shellfish. These findings suggest that clams may get through the osmotic stress by ion channel activation, neural adaptation, and anaerobic metabolism to maintain their energy homeostasis for maximum survival. The present study provides great insights into the molecular and metabolic strategies of intertidal shellfish in response to salinity stress, and uncovers the new potential signaling metabolites to assess the health states of molluscs in aquaculture.

Abstract:Understanding the mechanisms by which individual organisms respond and populations adapt to global climate change is a critical challenge. The role of plasticity and acclimation, within and across generations, may be essential given the pace of change. We investigated plasticity across generations and life stages in response to ocean acidification (OA), which poses a growing threat to both wild populations and the sustainable aquaculture of shellfish. Most studies of OA on shellfish focus on acute effects, and less is known regarding the longer term carryover effects that may manifest within or across generations. We assessed these longer term effects in red abalone (Haliotis rufescens) using a multi-generational split-brood experiment. We spawned adults raised in ambient conditions to create offspring that we then exposed to high pCO2 (1180 μatm; simulating OA) or low pCO2 (450 μatm; control or ambient conditions) during the first 3 months of life. We then allowed these animals to reach maturity in ambient common garden conditions for 4 years before returning the adults into high or low pCO2 treatments for 11 months and measuring growth and reproductive potential. Early-life exposure to OA in the F1 generation decreased adult growth rate even after 5 years especially when abalone were re-exposed to OA as adults. Adult but not early-life exposure to OA negatively impacted fecundity. We then exposed the F2 offspring to high or low pCO2 treatments for the first 3 months of life in a fully factorial, split-brood design. We found negative transgenerational effects of parental OA exposure on survival and growth of F2 offspring, in addition to significant direct effects of OA on F2 survival. These results show that the negative impacts of OA can last within and across generations, but that buffering against OA conditions at critical life-history windows can mitigate these effects.

Abstract:The magnitude of the work of the great students of the Compositae in the 19 and early 20 centuries is astonishing, both in the quality and scope of their research, as well as in the great number of systematic treatments and floras they produced. Their achievements go beyond exceptional, however, when they are seen in the context of the technology that these workers had available at the time. What follows is an attempt to gather and tie together many scattered pieces of information on their lives, work, and ideas, and to pay tribute to those individuals who have made outstanding contributions to the field. We are grateful to these men of science; their work has enabled us to see and understand the most diverse and successful plant family on Earth. This chapter covers notable synantherologists up to the 1975 conference held at Reading; a two-volume work that resulted from this meeting was published in 1977 (Heywood et al. 1977). The scientists are discussed in chronological order of the date of their major contribution^) to our understanding of Compositae.
