Abstract:Four experimental diets were formulated to evaluate the effect of coated lysine (Lys) and methionine (Met) supplemented in mixture protein diets of juvenile black sea bream, Acanthopagrus schlegelii. Fish meal (FM) was the main protein source in control diet, and 21.2% FM was replaced by the mixture of soybean protein concentrate, meat and bone meal, and monosodium glutamate protein in replacement diets with coating Lys and Met were suppled at none (0%, 0%), low (1.7%, 1.1%) and high (5.0%, 3.4%) level (designated as NLM, LLM and HLM diet, respectively). Each diet was fed to triplicate groups of 20 fish (initial mean weight, 4.13 +/- 0.16 g) twice a day to apparent satiation for 8 weeks, in a flow-through system. Results showed that amino acid supplementation significantly improved weight gain and specific growth rate in these mixture protein diets groups, and the fish fed with HLM diet showed comparable growth with the control group. Feed efficiency ratio (FER), protein efficiency ratio (PER), and crude protein and Met content in dorsal muscle were significantly affected by diet amino acid level. The highest apparent digestibility coefficients appeared in HLM group but the values showed insignificant difference with the control group. Serum activities of aspertate aminotransferase and alanine aminotransferase in control group were lower than those of in other treatments. Fish fed with NLM and LLM diets had higher triglycerides concentration than those of fed the HLM and FM diets. Furthermore, the feed cost of the various feed treatments were evaluated, and the three test diets were all lower than the control diet. The present study indicated that 21.2% FM in black sea bream diet can be successfully replaced by mixture protein with sufficient coated Lys (5.0%) and Met (3.4%) supplemented.

Abstract:A feeding trial of eight weeks was conducted to examine the influence of food supplementation with lauric acid (LA) on Acanthopagrus schlegelii (juvenile black sea bream). A 24 percent fish meal baseline diet was created, while the other two diets were generated with dietary supplementation of graded points of LA at 0.1 percent and 0.8 percent, respectively. Each diet was given a triplicate tank with 20 fish weighing 6.22 ± 0.19 g. In comparison with the control group, the weight gain rate, growth rate, as well as feed efficiency of fish fed of 0.1 percent diet of LA were considerably (P < 0.05) greater. The total body and dorsal muscle proximate compositions did not change significantly between groups (P > 0.05). Triglyceride (TG) content was considerably (P < 0.05) greater in the LA-supplemented meals eating group in comparison with the control group. In the group eating LA-supplemented meals, the height of villus and the number of goblet cells/villus were considerably (P < 0.05) larger. The microbial makeup of the gut was also studied. The differences in phyla, class, and family level were not statistically significant (P > 0.05). Firmicutes in the phylum, Betaproteobacteri, Gammaproteobacteria, and Clostridia in the class, and Clostridiaceae in the family were all substantially increased with higher levels of LA supplementation (P < 0.05). According to the findings of this study, an LA-supplemented diet improves fish development, antioxidative capability, gut microbiota and intestinal health.

Abstract:The effects of diets fermented with compound probiotics, namely Lactobacillus acidophilus, Limosilactobacillus reuteri and Lactiplantibacillus plantarum, on the growth performance, physiological and biochemical indexes, fatty acid composition and intestinal health of juvenile largemouth bass (Micropterus salmoides) were investigated. Three hundred healthy juvenile M. salmoides (5.29 ± 0.02 g) were selected and randomly divided into two groups with triplicates for each. The basic diet was set as the control group (CON), and fermentation of the basic diet with a mixed bacterial solution (1.8 × 109 cfu/mL, L. acidophilus:L. reuteri:L. plantarum = 1:1:1) was set as the fermentation group (FER). Fish were hand fed to satiation for 56 days and two-thirds of the culture water was renewed every 3 days. The results showed that feed intake of fish in the FER group was significantly lowered, thereby increasing feed efficiency (FE) and protein efficiency (PER) (p < 0.05). Serum alanine aminotransferase (ALT) activity was significantly decreased, and catalase (CAT) activity was significantly increased in the FER group (p < 0.05). The liver superoxide dismutase activity (SOD) was significantly enhanced, and intestinal trypsin was significantly increased in the FER group (p < 0.05). Being fed with the fermentation diet significantly increased the content of n-3 polyunsaturated fatty acids (PUFA), docosahexaenoic acid (DHA) and the n-3/n-6 PUFAs ratios in the liver (p < 0.05). Intestinal histology showed that villus height and width of the intestine and the number of goblet cells were significantly increased in the FER group (p < 0.05). Those fed with fermentation diets had limited diversity of gut microbiota. Compared to the CON group, the relative abundance of Aeromonas decreased significantly (p < 0.05), while the relative abundance of Fusobacteria, Cetobacteria and Lactobacillusis in FER increased greatly in the gut microbiota of the FER group. In conclusion, fermented feed with the three probiotics effectively improved the feed utilization and antioxidant capacity, promoted digestion and absorption of dietary protein, improved the ability of synthesize DHA and n-3 PUFAs in the liver and reduced the abundance of pathogenic bacteria in the gut. Therefore, the present research provided a new way of co-fermented feed with three probiotics for the aquaculture of M. salmoides.

Abstract:Introduction: This research aimed to assess the impact of Fermented Chinese herbs (FCHM) on the intestinal barrier, immunity, and protection against Aeromonas hydrophila (A. hydrophila) infection in largemouth bass ( Micropterus salmoides ). Methods: Four experimental diets were formulated, including H0 (basal diet), H1, H2, and H3, which contained 1%, 3%, and 5% FCHM added to the basal diet, respectively. The fish were randomly allocated to four treatment groups, each with 3 parallel per treatment, consisting of 20 fish per replicate and were raised for 56 days. Results and discussion: The experiment revealed that: compared with the control group, adding 1% FCHM significantly improved the weight gain rate (WGR) and specific growth rate (SGR) of the juvenile largemouth bass ( P< 0.05 ). The ingestion of FCHM substantially elevated the activities of CAT, SOD, GSH-PX, APK, ACP, and LZM, and T-AOC level in the gut region of largemouth bass, while decreasing the MDA content in intestine ( P< 0.05 ). Supplementation with FCHM enhanced the intestinal villus height and relative mRNA expression of intestinal barrier genes ZO-1 , Claudin , and Occludin in juvenile largemouth bass. After injecting A.hydrophila , all groups of largemouth bass experienced mortality, but the consumption of FCHM resulted in a decrease in cumulative mortality. After infected with A.hydrophila , the antioxidant enzymes and immune enzymes activities of all test groups were enhanced compared to those before infection, and the antioxidant enzymes and immune enzymes activities of all groups were considerably higher than the control after feeding FCHM ( P< 0.05 ). After infected with A.hydrophila , the intestinal MDA content of largemouth bass was higher compared with that before infection in all cases ( P< 0.05) , but after feeding FCHM, the MDA content was lower than the control ( P< 0.05 ). Upon consuming FCHM, the mRNA relative expressions of pro-inflammatory biomarkers IL-1β , TNF-α, IL-15 and IL-8 in largemouth bass infected with A.hydrophila were decreased in comparison to the control group. In contrast, the mRNA expression of the anti-inflammatory biomarkers TGF-β and IL-10 were significantly elevated ( P< 0.05 ). In summary, FCHM could improve the intestinal morphology, immunity, and antioxidant capacity of juvenile largemouth bass, and enhance it against A.hydrophila , with a better effect at 1% addition.

Modulation of growth performance, antioxidant capacity, non-specific immunity and disease resistance in largemouth bass (Micropterus salmoides) upon compound probiotic cultures inclusion

Abstract:An 8-week feeding trial was conducted to evaluate the effects of dietary supplementation of compound probiotic cultures (CPC; Bacillus subtilis, Lactobacillus plantarum and Saccharomyces cerevisiae) on the growth performance, antioxidant capacity, non-specific immunity and disease resistance of juvenile largemouth bass. Triplicate groups of largemouth bass (average weight 42.05 ± 0.02 g), with a destiny of 30 individuals per tank, were fed diets supplemented with different concentration of compound probiotic cultures (CPC) (0%, CPC (0.0); 0.5%, CPC (0.5); 1.0%, CPC (1.0); 2.0%, CPC (2.0)). After the feeding trial, tissue samples of largemouth bass were collected and the challenge test with Aeromonas hydrophila was performed. Results indicated that the CPC supplementation produced no significant difference on the growth performance, feed utilization and body composition of largemouth bass, while significantly increased the cumulative survival rate in the Aeromonas hydrophila challenge test. Meanwhile, the inclusion of CPC elevated the hepatic antioxidant capacity, and the highest activity of antioxidant enzymes, including T-AOC, CAT, GPx and T-SOD, was observed in the CPC (2.0) group. Meanwhile, the transcription of Nrf2/keap1 and antioxidant related genes, including CAT, GPx, GST, SOD1 and SOD2, was significantly elevated with the inclusion of CPC. In addition, the inclusion of CPC improved the non-specific immunity of largemouth bass. The activity of serum lysozyme was significantly elevated in the CPC (2.0) group, while the transcription of RelA and pro-inflammatory factors, including TNF-α and IL-1β, was inhibited with the inclusion of CPC. Meanwhile, related genes potentially linked to RelA, including TLR2 and p38 MAPK, were detected that their relative expression was significantly inhibited with the inclusion of CPC. The current findings indicated that the inclusion of 2% CPC improved the antioxidant capacity, non-specific immunity and disease resistance of juvenile largemouth bass, and suggested that 2% CPC as a functional additive could be applied to the diet of juvenile largemouth bass in aquaculture practice.
An Evaluation of Laminarin Additive in The Diets of Juvenile Largemouth Bass (Micropterus salmoides): Growth, Antioxidant Capacity, Immune Response and Intestinal Microbiota

Abstract:A 28 day feeding trial was conducted to investigate the growth performance, immune response and intestinal microbiota of laminarin (LAM) supplemented diets in juvenile largemouth bass (Micropterus salmoides). Four hundred and eighty fish (initial average weight: 0.72 ± 0.04 g) were randomly divided into four groups (40 fish per tank with three replicates in each group) Four diets were prepared with LAM supplementation at the doses of 0 (control), 5 g Kg−1 (LL), 10 g Kg−1 (ML) and 15 g Kg−1 (HL), respectively. No significant difference in the specific growth rate (SGR) and hepatosomatic index (HSI) was observed in fish among the four groups, or in the lipid and ash content of fish flesh. In addition, fish in the LL group exhibited much higher antioxidant capacity (p 0.05). Dietary LAM at the dose of 5 g Kg−1 inhibited the transcription of interleukin-1β (il-1β) and tumor necrosis factor-α (tnf-α), while promoting the expression of transforming growth factor-β (tgf-β) in fish intestine. Moreover, the beneficial intestinal bacteria Bacteroide, Comamonas and Mycoplasma abundance significantly increased in fish from the LL group, while the content of opportunistic pathogens Plesiomonas, Aeromonas and Brevinema in fish of the HL group was substantially higher than the control group. Overall, the appropriate dose of supplemented LAM in the diet was 5 g Kg−1, while an excessive supplementation of LAM in the diet led to microbial community instability in largemouth bass.

Abstract:The present study was conducted to investigate the effects of dietary inclusion of protein hydrolysates on growth performance, digestive enzyme activities, protein metabolism, and intestinal health in larval largemouth bass (Micropterus salmoides). The experimental feeding trial presented in this study was based on five isonitrogenous and isolipidic diets formulated with graded inclusion levels of protein hydrolysates, and it showed that protein hydrolysates improved growth performance, reduced larval deformity rate, and increased the activity of digestive enzymes, including pepsin and trypsin. Gene expression results revealed that the supplementation of protein hydrolysates upregulated the expression of intestinal amino acid transporters LAT2 and peptide transporter 2 (PepT2), as well as the amino acid transporters LAT1 in muscle. Dietary provision of protein hydrolysates activated the target of rapamycin (TOR) pathway including the up-regulation of TOR and AKT1, and down-regulation of 4EBP1. Additionally, the expression of genes involved in the amino acids response (AAR) pathway, ATF4 and REDD1, were inhibited. Protein hydrolysates inhibited the transcription of some pro-inflammatory cytokines, including IL-8 and 5-LOX, but promoted the expression of anti-inflammatory cytokines TGF-beta and IL-10. The 16S rRNA analysis, using V3-V4 region, indicated that dietary protein hydrolysates supplementation reduced the diversity of the intestine microbial community, increased the enrichment of Plesiomonas and reduced the enrichment of Staphylococcus at the genus level. In summary, protein hydrolysates have been shown to be an active and useful supplement to positively complement other protein sources in the diets for largemouth bass larvae, and this study provided novel insights on the beneficial roles and possible mechanisms of action of dietary protein hydrolysates in improving the overall performance of fish larvae.

Abstract:Introduction As the aquaculture industry intensifies to enhance production efficiency and capacity, the risk of disease outbreaks in high-density systems, such as those for largemouth bass, has escalated. This necessitates the exploration of novel strategies for disease prevention and control. Studies have shown that Atractylodes macrocephala polysaccharide (AMP) possesses the functions of promoting growth and enhancing immune capacity, making it a potential feed additive in animal production. Methods This study aimed to assess the impact of AMP on the growth performance, serum biochemical indices, and intestinal flora structure of largemouth bass. A total of 360 healthy largemouth bass (mean weight: 15.25 ± 1.29 g) were randomly assigned to four groups, each with three replicates. The groups were fed diets supplemented with 0% (Group D, control group), 0.4% (Group A), 0.8% (Group B), or 1.2% (Group C) AMP for 42 days. Results The results showed that the addition of an appropriate amount of AMP in the feed significantly improved the weight gain rate (WGR) and specific growth rate (SGR) of largemouth bass, reduced the feed conversion ratio (FCR), and enhanced growth performance. AMP reduced the levels of AST and ALT, indicating a hepatoprotective effect on largemouth bass, with significant differences from the control group (P < 0.05). AMP also improved the intestinal microbiota composition of largemouth bass, positively affecting intestinal health. The analysis of intestinal microbiota revealed 1,288 amplicon sequence variants (ASVs) in the intestine of largemouth bass in Group B, dominated by Firmicutes and Bacteroidota. In contrast, Group C (1.2%) had 920 ASVs, with Fusobacteriota and Firmicutes as the major components, while the relative abundance of Firmicutes was lower, showing significant differences from the control Group D. The functional analysis of intestinal microbiota based on KEGG showed significant differences (P < 0.05) among the four groups in Carbon metabolism, Biosynthesis of amino acids, and Metabolic pathways. Discussion The study concludes that the inclusion of AMP at 0.8% – 1.2% in the feed can enhance the growth performance and intestinal health of largemouth bass, offering a promising strategy for disease prevention and control in intensive aquaculture settings.

Abstract:The active ingredients extracted from yeast are important for regulating animal health. The aim of the current research was to explore the impacts of dietary yeast glycoprotein (YG) on the growth performance, intestinal morphology, antioxidant capacity, immunity and disease resistance of largemouth bass ( Micropterus salmoides ). A total of 375 juvenile fish (6.00 ± 0.03 g) were allocated into 15 fiberglass tanks. Triplicate tanks were assigned to each diet. The dietary YG inclusion was as follows: the first group was given a high fishmeal diet (40% fishmeal, 0% YG) (FM) and the second group was given a low fishmeal diet (30% fishmeal and 15% soybean meal, 0% YG) (LFM). The fish in the third, fourth and fifth groups were fed the LFM diet supplemented with 0.5% (LFM+YG0.5), 1.0% (LFM+YG1.0) and 2.0% (LFM+YG2.0) YG, respectively. After a 60- day feeding trial, a challenge test using A. hydrophila was carried out. The results showed that the final body weight (FBW) and weight gain rate (WGR) in the LFM+YG2.0 group were significantly higher than those in the LFM group and were no significantly different from those in the FM group. This may be partially related to the activation of the target of rapamycin (TOR) signaling pathway. Dietary YG supplementation enhanced intestinal physical barriers by upregulating the intestinal tight junction protein related genes ( claudin1 , occludin and zo2 ) and improving the structural integrity of the gut, which may be partially associated with AMPK signaling pathway. Moreover, dietary YG increased the antioxidant capacity in the gut, upregulated intestinal anti-inflammatory factors ( il-10 , il1-1β and tgf-β ) and downregulated proinflammatory factors ( il-1β and il-8 ), which may be partially related to the Nrf2/Keap1 signaling pathways. The results of the challenge test indicated that dietary supplementation with 0.5 or 1.0% YG can increase the disease tolerance of largemouth bass against A. hydrophila . In conclusion, the present results indicated that dietary supplementation with YG promotes the growth performance, intestinal immunity, physical barriers and antioxidant capacity of largemouth bass. In addition, 1.0% of dietary YG is recommended for largemouth bass based on the present results.

Abstract:The present study was aimed to explore the influence of graded replacing fish meal (0.00%, 14.29%, 28.57%, 42.86%, and 57.14%) with spray dried chicken plasma (SDCP) on growth performance, feed utilization, and antioxidant capacity of largemouth bass. Triplicate groups of fish (initial weight: 10.05 ± 0.05 g) were fed experimental diets twice daily for 8 weeks. Results indicated that specific growth rate and feed intake of fish were not remarkably influenced with the replacement of fish meal. Meanwhile, no significant difference was observed in feed intake and feed efficiency ratio among treatments. The inclusion of SDCP significantly elevated crude protein content of muscle, liver and whole fish body, which resulted in the increased protein retention rate. Meanwhile, the lipid retention rate was also significantly increased with the replacement of fish meal. The replacement of 57.14% fish meal with SDCP significantly reduced the content of lipid peroxidation product malondialdehyde. Meanwhile, the total antioxidant capacity and catalase activity was significantly higher in fish fed the diet with replacement of 57.14% fish meal compared to the control group. In above, 57.14% fish meal could be substituted by SDCP in diets for largemouth bass under this experimental condition.

Abstract:The present study investigated the effects of Astragalus membranaceus extract (AME) on growth performance, immune response, and energy metabolism of juvenile largemouth bass (Micropterus salmoides). Seven diets containing 0%, 0.1%, 0.2%, 0.3%, 0.4%, 0.5%, and 0.6% AME (Con, AME0.1, AME0.2, AME0.3, AME0.4, AME0.5, and AME0.6 groups) were formulated and fed to M. salmoides for 8 weeks. Final body weight (FBW), feed intake (FI), weight gain (WG), and specific growth rate (SGR) were all significantly higher in AME0.4 group than in Con group (P < 0.05). Feed conversion rate (FCR) was significantly improved in AME0.5 group compared with Con group (P < 0.05). Whole-body crude protein contents were significantly increased in AME0.2 group (P < 0.05). Whole-body crude lipid contents were significantly lower in AME0.2 and AME0.3 groups, while muscle lipid was upregulated by dietary AME (P < 0.05). Hepatic malondialdehyde (MDA) contents were significantly lowered in AME0.3 and AME0.4 groups, and catalase (CAT) activities were significantly increased in AME0.1 and AME0.2 groups (P < 0.05). Plasma aspartate aminotransferase (AST) level was significantly lowered in AME0.5, and AME0.6 groups, and alanine aminotransferase (ALT) level was lowered in AME0.5 groups (P < 0.05). Plasma triglyceride was declined in AME0.6 group, and glucose was decreased by 0.3%-0.5% AME (P < 0.05). Significantly higher hepatocyte diameter, lamina propria width, and submucosal layer thickness were recorded in AME0.6 groups, while the longest villi height was obtained in AME0.2 and AME0.3 groups (P < 0.05). The mRNA expression levels of insulin-like growth factor 1 (igf1) revealed the growth-promoting effect of AME. The anti-inflammatory and antiapoptotic effects of AME were demonstrated by transcription levels of interleukin 8 (il-8), tumor necrosis factor-alpha (tnf-a), caspase, B-cell lymphoma-xl (Bcl-xl), bcl-2 associated x (Bax), and bcl-2-associated death protein (Bad). The transcription levels of lipid metabolism and gluconeogenesis related genes, including acetyl-CoA carboxylase alpha (acc1), fatty acid synthase (fasn), fatty acid binding protein 1 (fabp1), phosphoenolpyruvate carboxykinase 2 (pepck2), and glucose-6-phosphatase catalytic subunit 1a (g6pc), were reduced by AME treatment, while the levels of glycolysis-related genes, including glucokinase (gck) and pyruvate kinase (pk), were the highest in AME0.2 and AME0.3 groups (P < 0.05). According to polynomial regression analysis of SGR, WG, FCR, whole-body crude lipid, MDA, and ALT, the optimal AME supplementation level was estimated to be 0.320%-0.429% of the diet. These results provided insights into the roles of AME in regulating immunity and metabolism, which highly indicated its potential as immunostimulants and metabolic regulators in diverse aquatic animals.

Abstract:Abstract Background: The intestine is the first line of defense against bacteria, toxins and antigens presents in the lumen of the instine. Aeromonas veronii is an opportunistic pathogen that has been related to mortality in fish. However, the protective mechanisms of intestinal damage caused by Aeromonas veronii are poorly defined. Thus, the objetive of this study was to evaluate the influence of dietary valine (Val) on growth, antioxidant capacity, and endoplasmic reticulum stress-mediated apoptosis in intestines of the juvenile largemouth bass Micropterus punctulatus after infection with Aeromonas veronii (A. veronii). Methods: A total of 720 fish (37.99 ± 0.01 g) were randomly allocated to six groups with three replicates each group, with different levels of Val (11.0, 14.0, 17.0, 20.1, 23.1, and 26.1 g/kg diet, respectively) for 77 d, and then challenged with A. veronii. Results: In the present study, we demonstrated that dietary Val: (1) increased the percent weight gain (PWG), specific growth rate (SGR), feed intake (FI), feed efficiency (FE), protein efficiency ratio (PER), protein production value (PPV), lipid production value (LPV), and ash production value (APV) (P< 0.05); (2) improved survival rate (SR) and alleviated intestinal injury; (3) reduced the levels of reactive oxygen species (ROS), malondialdehyde (MDA), and protein carbonyl (PC) (P< 0.05), and improved the intestinal antioxidant capacity by enhancing the mRNA levels of Cu/Zn superoxide dismutase (SOD1), catalase (CAT), glutathione-S-transferase (GST), glutathione peroxidase (GPx), glutathione reductase (GR), and γ-glutamyl cysteine ligase catalytic subunit (GCLC) (P< 0.05); (4) increased nuclear NFE2-related factor 2 (Nrf2) mRNA and protein levels (P< 0.05); (5) reduced intestinal DNA fragments, and down-regulated the mRNA levels of 78 KD glucose regulated protein (GRP78), activating transcription factor 6 (ATF6), inositol requiring enzyme-1 (IRE1), protein kinase RNA-dependent-like endoplasmic reticulum kinase (PERK), eukaryotic initiation factor 2α (eIF2α), activating transcription factor 4 (ATF4), C/EBP homologous protein (CHOP), cysteine-aspartic protease 9 (caspase 9), and cysteine-aspartic protease 3 (caspase 3), while up-regulating that of B-cell lymphoma-2 (Bcl2) (P< 0.05); (6) decreased phosphorylation of PERK (p-PERK) and phosphorylation of eIF2α (p-eIF2α), caspase 9, and caspase 3 protein levels. Conclusion: Dietary optimal Val level enhanced the growth performance and the intestinal antioxidant capacity via the Nrf2 signaling pathway after infection with A. veronii. Furthermore, this study is the first to demonstrate the role of Val in reducing ERS-mediated apoptosis through modulation of the PERK/eIF2α signaling pathway. Finally, Quadratic regression analysis of the SGR indicated that the dietary Val requirement of largemouth bass (37.99-162.92 g) was 21.50 g/kg diet, corresponding to 46.64 g/kg protein.

Abstract:The first measurements of the line-integrated electron density in a coaxial microsecond-conduction-time plasma opening switch during switch operation are presented. Current conduction is observed to cause a radial redistribution of the switch plasma, with a large decrease in axial line density over most of the radial extent of the switch. A local reduction in line density of more than an order of magnitude occurs by the time opening begins. Abstract:This experiment evaluated the function of chlorogenic acid (CGA) in the growth, health status, and inflammation of largemouth bass (Micropterus salmoides). Over eight weeks, CGA supplementation was designed at five levels: 0, 60, 120, 180, and 240 mg/kg. The 180 and 240 mg/kg CGA-supplemented groups showed significant improvements in the FBW, SGR, and WGR compared to the control group (0 mg/kg) (p < 0.05). All the CGA-supplemented groups exhibited a significant reduction in the FCR (p < 0.05), with the 180 mg/kg CGA group showing the lowest FCR. Nonetheless, there were no appreciable differences in the plasma concentrations of TP, ALT, or AST among the treatments (p > 0.05). Compared to the control group, the 180 mg/kg CGA group exhibited significantly lower TC and TG levels (p < 0.05). The ALP levels showed no significant differences from the control group (p > 0.05). In terms of antioxidant parameters, CGA supplementation considerably reduced the MDA content (p < 0.05) and increased the GSH levels, while decreasing the CAT, SOD, and GPx activity levels Meanwhile, CGA supplementation resulted in reduced mRNA levels of SOD, CAT, Nrf2, Keap1, and NF-κB compared to the control group. In contrast, the mRNA levels of GPx, IL-8, TLR2, and RelA were elevated in the liver. Our findings indicated that CGA supplementation improved the growth performance and antioxidant status and weakened the inflammatory response of largemouth bass. These findings suggest that CGA could be a valuable dietary supplement for enhancing the health and growth of this species.

Abstract:The improper components of formulated feed can cause the intestinal dysbiosis of juvenile largemouth bass and further affect fish health. A 28 day feeding trial was conducted to investigate the effect of partially replacing fish meal (FM) with autolyzed Yarrowia lipolytica (YL) on juvenile largemouth bass (Micropterus salmoides). We considered four diets—control, YL25, YL50, and YL75—in which 0%, 25%, 50%, and 75% of the FM content, respectively, was replaced with YL. According to results, the weight gain rate (WGR) and specific growth rate (SGR) of the fish with the YL25 and YL50 diets were significantly higher than the WGR and SGR with the control diet, while the YL75 diet significantly reduced fish growth and antioxidant enzymes activities, and shortened the villus height in the intestinal mucosa. The 16S rRNA analysis of the intestinal microbiota showed that the relative abundance of Mycoplasma was significantly increased with the YL25 and YL50 diets, while the Enterobacteriacea content was increased with the YL75 diet. Moreover, our transcriptome analysis revealed that certain differentially expressed genes (DEGs) that are associated with growth, metabolism, and immunity were modulated by YL inclusion treatment. Dietary YL25 and YL50 significantly reduced the mRNA level of ERBB receptor feedback inhibitor 1 (errfi1) and dual-specificity phosphatases (dusp), while the expression of the suppressor of cytokine signaling 1 (socs1), the transporter associated with antigen processing 2 subunit type a (tap2a), and the major histocompatibility complex class I-related gene (MHC-I-l) were sharply increased with YL75 treatment. We determined that the optimum dose of dietary YL required for maximum growth without any adverse influence on intestinal health was 189.82 g/kg (with 31.63% of the fishmeal replaced by YL), while an excessive substitution of YL for fishmeal led to suppressed growth and antioxidant capacity, as well as intestinal damage for juvenile largemouth bass.

Abstract:The cultivation of largemouth bass ( Micropterus salmoides ), a species of significant economic value in aquaculture, has experienced notable growth recently. However, the deterioration of water quality seriously affects the metabolic responses of M. salmoides . While compound microbial agent (CMA) is widely utilized for ecological rehabilitation and water filtration, its application in M. salmoides has not been reported. Here, based on physio-biochemical tests and 16S rRNA sequencing, we investigated the effects of CMA (yeast, Bacillus subtilis , and lactic acid bacteria) on the water quality within the recirculating aquaculture system, along with physiological indices and gut microbiota of M. salmoides . Compared to the control and single microbial agent (yeast), CMA treatment improved the water quality by improving the dissolved oxygen and delaying the increase of pH, total nitrogen, total phosphorus, ammonia nitrogen, and nitrite. The 16s rRNA gene sequencing revealed that the water treated with CMA exhibited elevated levels of chao1, Shannon, Pd, and a larger population of dominant bacterial. Besides, higher values of ACE, chao1, Shannon, and OTU level, and lower Simpson index were found in CMA treated M. salmoides samples, suggesting that CMA treatment enhanced the species richness and diversity of gut microbiota of M. salmoides . Furthermore, CMA treatment hindered the generation and proliferation of harmful bacteria, such as the Mycoplasma mobile 163K species and the Erysipelotrichaceae family, which was associated with enhanced antioxidant enzymatic activity and decreased MDA level in both the serum and liver. These findings shed light on the essential roles of CMA in M. salmoides culturing and introduce an innovative approach to enhance the aquatic environment.

Abstract:Probiotics play an essential role in the largemouth bass (Micropterus salmoides) aquaculture sector. They aid the fish in sickness prevention, intestinal structure improvement, food absorption, and immune system strengthening. In this experiment, Bacillus subtilis (BS, 107 CFU/g) and Lactobacillus reuteri (LR, 107 CFU/g) were added to the feed and then fed to M. salmoides for 35 days. The effects of two probiotics on the growth, immunity, and metabolism of M. salmoides organisms were studied. The results revealed that the BS group significantly increased the growth rate and specific growth rate of M. salmoides, while both the BS and LR groups significantly increase the length of villi M. salmoides intestines. The BS group significantly increased the levels of AKP, T-AOC, and CAT in the blood of M. salmoides, as well as AKP levels in the intestine. Furthermore, the BS group significantly increased the expression of intestinal genes Nrf2, SOD1, GPX, and CAT, while significantly decreasing the expression of the keap1 gene. M. salmoides gut microbial analysis showed that the abundance of Planctomycetota was significantly different in both control and experimental groups. Analyzed at the genus level, the abundance of Citrobacter, Paracoccus, Luedemannella， Sphingomonas, Streptomyces and Xanthomonas in the both control and experimental groups were significantly different. The BS group's differentially expressed genes were predominantly enriched in oxidative phosphorylation pathways in the intestine, indicating that they had a good influence on intestinal metabolism and inflammation suppression. In contrast, differentially expressed genes in the LR group were primarily enriched in the insulin signaling and linoleic acid metabolism pathways, indicating improved intestine metabolic performance. In conclusion, B. subtilis and L. reuteri improve the growth and health of M. salmoides, indicating tremendous potential for enhancing intestinal metabolism and providing significant application value.

Abstract:This study aimed to evaluate the effects of fermented Chinese herbal medicines (FCHM) on growth performance, digestive enzyme activity, antioxidant capacity and intestinal inflammatory gene expression of juvenile largemouth bass (Micropterus salmoides). Four trial diets H0 (basal diet), H1, H2, and H3 (added 1 %, 3 %, and 5 % FCHM to the basal diet, respectively) were prepared. Fish were randomly divided into 4 treatment groups of 3 replicates per treatment (20 fish/replicate) and reared for 8 weeks. The results of the test showed that: 1) Feeding 1 % FCHM significantly increased the weight gain rate (P < 0.01) and specific growth rate (P 0.05) but could result in an overall decreasing trend in muscle drip loss, steaming loss, and freezing loss. 3) FCHM significantly increased the activity of SOD, GSH-PX and CAT in the liver of juvenile largemouth bass and decreased the MDA compared to that of the control group (P < 0.01). (4) Compared with the control group, FCHM significantly increased the relative mRNA expression of the intestinal anti-inflammatory factors TGF-β and IL-10 (P < 0.01) and decreased the pro-inflammatory factors IL-8, IL-15 (P < 0.01) and TNF-α (P < 0.05) of juvenile Largemouth bass. In summary, a 1 % FCHM addition amount can improve the growth performance, liver antioxidant and immune capacity of juvenile largemouth bass.

Abstract:The present research examined the impact of L-glutamic acid (Glu) supplementation on the growth performance, muscle composition, gene expression correlated with muscle growth, and intestinal health of largemouth bass. There were 525 fish in total, which were distributed randomly into five groups. Each group had three replicates, and each replicate consisted of 35 fish. Groups with control and experimental diets were assigned glutamic acid amounts of 0.2%, 0.4%, 0.6%, and 0.8%. The findings demonstrated that glutamic acid supplementation enhanced growth performance, feed intake (FI), and condition factor (CF), with the best value being attained at 0.4% Glu. The mean muscle fiber area was increased and the muscle fiber density was decreased in the 0.6% Glu group. The levels of total amino acids and specific amino acids, such as glutamic acid, aspartic acid, leucine, valine, alanine, and glycine, were shown to be higher in the 0.6% Glu group. In the 0.6% Glu group, the mRNA expression levels of atrogin-1, murf-1, foxo3a, and 4e-bp1 were decreased compared to the control group. Conversely, the mRNA expression levels of myf5, myog, myod, s6k1, tor, akt, and pi3k were increased in the 0.6% Glu group compared to the control group. The 0.4% Glu group had higher intestinal amylase, lipase, and protease activities and greater villus height, villus width, and muscle thickness. In summary, Glu can support largemouth bass growth, muscular development, intestinal digestion, and absorption.
