Abstract:The time-course of plasma amino acid ( AA) following response to early weaning was investigated over a 30 d period in weaned piglets using AA analyzer. Eight 21-day-old "Duroc × Landrace × Large York-shire" weaned piglets were selected and their blood samples were collected on days 0, 1, 3, 5, 7, 10, 15, and 30 post-weaning. The results showed as follows: plasma contents of arginine and its immediate precursors ( citrulline and ornithine) decreased ( P<0.05) from days 0 to 5 post-weaning, and increased from days 7 to 30 post-weaning. Plasma contents of leucine, proline, tyrosine and taurine decreased ( P<0.05) on days 1 and 3 post-weaning, and increased to the level of day 0 post-weaning on day 30. Plasma contents of isoleucine, ly-sine, glycine and serine were the lowest on day 5 post-weaning; plasma contents of histidine, phenylalanine, valine, aspartic acid, cysteine were the lowest on day 10 post-weaning, which then increased thereafter. Plas-ma content of glutamic acid was significantly higher on days 0, 1, 10 post-weaning than that of the others ( P<0.05). Total AA content was significantly decreased from day 1 to day 5 post-weaning(P<0.05), then in-creased on day 7 and keep constant thereafter with no significant difference to day 30 post-weaning ( P>0.05) . In score plot of principal component analysis, the profiles of AA were gradually away from the profile of AA on day 0 post-weaning, and then were closed to that, and tended to be stable. On days 3 and 5 post-weaning, the profiles of AA were robustly altered. The pattern of partial least-square discriminant analysis demonstrated that glutamic acid played the most important role in differentiating the profiles of AA at different time points post-weaning. It is concluded that early weaning changes the metabolism of protein and amino acid in the body of piglets, especially on days 3 and 5 post-weaning.
Mechanisms of Endothelin-1 Elevation in Chronic Obstructive Pulmonary Disease Patients with Nocturnal Oxyhemoglobin Desaturation

Abstract:Background: Nonapneic, oxyhemoglobin desaturation associated with sleep has been described in patients with chronic obstructive pulmonary disease (COPD). Hypoxemia stimulates endothelin-1 (ET-1) secretion. Once released, ET-1 can act locally to elicit sustained pulmonary artery vasoconstriction, bronchoconstriction and activation of alveolar macrophages. Objective: The aim of this study was to examine a possible correlation between ET-1 levels and nocturnal, nonapneic, oxyhemoglobin desaturation during sleep, in patients with COPD. Methods: We examined 48 COPD patients with formal polysomnography (EEG, ECG, airflow, respiratory muscle movement, oximeter) to detect the presence of nocturnal, nonapneic, oxyhemoglobin desaturation. Twelve of them were disqualified because of inadequate sleep or sleep apnea syndrome. Nineteen of them desaturated below a baseline sleep saturation of 90% for 5 min or more, reaching a nadir saturation of at least 85%. We collected arterial samples to measure ET-1 levels, after 5 min of the first period of desaturation, in each of the 19 patients. We also collected arterial samples in the morning, before the study, to measure baseline ET-1 levels in all patients. Results: Baseline arterial ET-1 levels during the day were very significantly higher in ‘desaturator’ COPD patients (2.058 ± 0.252 pg/ml) compared to ‘non-desaturator’ COPD patients (1.382 ± 0.159 pg/ml; p < 0.001). Also in ‘desaturator’ COPD patients ET-1, levels during the night were significantly higher (4.297 ± 1.107 pg/ml) compared to those during the day (p ≤ 0.001) and a significant negative correlation was observed between ET-1 levels and degree of desaturation (p < 0.0001, r = 0.9305). Conclusions: According to our study we can conclude that (1) ET-1 levels are significantly higher in ‘desaturator’ COPD patients both during the day and during the night, and (2) ET-1 levels correlate negatively significant with the degree of the oxyhemoglobin desaturation. These findings are consistent with the hypothesis that ET-1 plays a very important role in the pathophysiological manifestations of COPD patients.
African-derived mitochondria in South American native cattle breeds (Bos taurus): evidence of a new taurine mitochondrial lineage.

Abstract:This article reports the nucleotide diversity within the control region of 42 mitochondrial chromosomes belonging to five South American native cattle breeds (Bos taurus). Analysis of these data in conjunction with B. taurus and B. indicus sequences from Africa, Europe, the Near East, India, and Japan allowed the recognition of eight new mitochondrial haplotypes and their relative positions in a phylogenetic network. The structure of genetic variation among different hypothetical groupings was tested through the molecular variance decomposition, which was best explained by haplotype group components. Haplotypes surveyed were classified as European-related and African-related. Unexpectedly, two haplotypes within the African cluster were more divergent from the African consensus than the latter from the European consensus. A neighbor-joining tree shows the position of two haplotypes compared to European/African mitochondrial lineage splitting. This different and putatively ancestral mitochondrial lineage (AA) is supported by the calibration of sequence divergence based on the Bos-Bison separation. The European/African mitochondria divergence might be subsequent (67,100 years before present) to that between AA and Africans (84,700 years before present), also preceding domestication times. These genetic data could reflect the haplotype distribution of Iberian cattle five centuries ago.

Abstract:1. Six 45 kg pigs with re-entrant ileal cannulas were used in two 3 × 3 Latin-square design experiments to study the site of absorption of protein and amino acids. Semi-purified diets containing soya-bean meal (SBM), rapeseed meal (RSM) or no protein source (protein-free) were offered at the rate of 1 kg dry matter/d. 2. Flow-rates of ileal contents for 24 h collection periods, corrected for recovery of marker, were 3135, 3127 and 1243 ml (SE 390) for SBM, RSM and protein-free diets respectively. 3. Amounts of dry matter digested in the small intestine were 730, 669 and 809 g/d for SBM, RSM and protein-free diets respectively, all values being significantly different (P < 0·001). 4. Nitrogen intakes were 32·6, 29·9 and 5·9 g/d, and amounts digested in the small intestine were 25·7, 20·2 and 1·6 g/d for SBM, RSM and protein-free diets respectively, all values being significantly different (P < 0·001). Amounts digested in the large intestine were 2·6, 3·7 and 0·7 g/d. 5. Total amino acid intakes and amounts collected at the ileum and in the faeces were (g/d): SBM, 177, 24 and 18; RSM, 149, 28 and 22; protein-free 3, 9 and 12. Digestibility in the small intestine was higher for SBM than RSM for seventeen of the eighteen amino acids estimated. Greater quantities of arginine, methionine, cystine and tyrosine were voided in the faeces than passed through the ileal cannulas for pigs receiving the SBM and RSM diets. For those receiving the protein-free diet this was true for each amino acid except proline. 6. Significant differences were found between all diets in the concentration of some amino acids in ileal and faecal amino-N, and endogenous protein secretions did not mask the differences between diets. 7. Differences in digestibility between SBM and RSM were greater at the ileum than in the faeces. Amino acid fermentation in the large intestine obscured or reduced differences between SBM and RSM.

Effects of Different Protein Sources on Amino Acid Absorption and Plasma Appearance of Tryptophan, Large Neutral Amino Acids, and Tryptophan Metabolites in Pigs

Abstract:Background: Absorption of tryptophan (TRP) across the gut epithelium is potentially modulated by competing large neutral amino acids (LNAAs), which could affect the appearance of TRP and its metabolites in the bloodstream. Objectives: This study aimed to determine, in a growing pig model of an adult human, the absorption of TRP and other LNAAs from the gastrointestinal tract, and plasma appearance of TRP, LNAAs, and TRP metabolites, in response to dietary proteins varying in TRP content. Methods: Pigs were adapted for 7 d to each of 4 diets that differed in their protein source and TRP content: 1) alpha-lactalbumin (AL; 9.95 mg TRP/g diet DM), 2) whey protein (6.59 mg TRP/g), 3) casein (3.73 mg TRP/g), or 4) zein (0.14 mg TRP/g). On day 8, pigs were euthanised after a 12-h fast (baseline), or 1, 2, 3, 4, or 6 h after they received a test meal consisting of 45 g protein, or a protein-free meal (n = 6 pigs at each time in each meal group). Tryptophan and LNAA absorption from the small intestine, and appearance of TRP, LNAAs, and TRP metabolites (melatonin, serotonin, kynurenine pathway metabolites), in the portal vein and systemic circulation, were determined. Results: AL intake resulted in sustained elevated plasma TRP concentrations after an overnight fast. The amount of TRP absorbed was dose-dependently related to protein TRP content (P = 0.028), with fastest rates for pigs fed AL (371 mg/h). Portal and systemic plasma TRP, TRP/LNAA, and the TRP metabolites were highest (P ≤ 0.05) after AL intake, and remained above baseline levels for ∼4 h postprandially. Absorption rates of TRP correlated with postprandial plasma TRP and TRP metabolites (P ≤ 0.05). Conclusions: In adult humans, postprandial plasma TRP and TRP metabolite concentrations can likely be modulated by the TRP content of the meal.
Free Amino Acid–Enriched Diets Containing Rapidly but Not Slowly Digested Carbohydrate Promote Amino Acid Absorption from Intestine and Net Fluxes across Skeletal Muscle of Pigs

Abstract:BACKGROUND: The approach to matching appropriate carbohydrates alongside free amino acids to achieve optimal muscle growth remains unclear.OBJECTIVES: We investigated whether the consumption of a diet containing rapidly digested carbohydrate and free amino acids can enhance intestinal absorption and muscular uptake of amino acids in pigs.METHOD: Twelve barrows (28 kg; 11 wk old) with catheters installed in the portal vein, mesenteric vein, femoral artery, and femoral vein were randomly assigned to consume 1 of 2 free amino acid-enriched diets (3.34%) containing rapidly [waxy corn starch (WCS)] or slowly [pea starch (PS)] digested carbohydrate for 27 d. Blood was collected to determine the fluxes of plasma glucose and amino acids across the portal vein and the hindlimb muscle. Dietary in vitro carbohydrate digestive rates were also determined. Data were analyzed using repeated-measures (time × group) ANOVA.RESULTS: Carbohydrate in vitro cumulative digestibility at 30 and 240 min was 69.00% and 95.25% for WCS and 23.25% and 81.15% for PS, respectively. The animal experiment presented WCS increased individual amino acids (lysine, 0.67 compared with 0.53 mmol/min; threonine, 0.40 compared with 0.29 mmol/min; isoleucine, 0.33 compared with 0.22 mmol/min; glutamate, 0.51 compared with 0.35 mmol/min; and proline, 0.51 compared with 0.27 mmol/min), essential amino acid (EAA; 3.26 compared with 2.65 mmol/min), and branched-chain amino acid (BCAA; 0.86 compared with 0.65 mmol/min) fluxes across the portal vein during 8 h postprandial, as well as individual amino acids (isoleucine, 0.08 compared with 0.02 mmol/min; leucine, 0.06 compared with 0.02 mmol/min; and glutamine, 0.44 compared with 0.25 mmol/min), EAA (0.50 compared with 0.21 mmol/min), and BCAA (0.17 compared with 0.06 mmol/min) net fluxes across the hindlimb muscle during 8 h postprandial compared with PS (P < 0.05).CONCLUSIONS: A diet containing rapidly digested carbohydrate and free amino acids can promote intestinal absorption and net fluxes across hindlimb muscle of amino acids in pigs.

Relationship between dietary amino acid release kinetics and nitrogen deposition in growing pigs

Abstract:Although the protein content of swine diets is formulated based on the ileal digestibility of protein and amino acids (AA) under current nutrition requirements, the nitrogen utilization efficiency of swine varies based on protein source, which may be related to AA release kinetics. In this experiment, a 2 × 2 factorial arrangement with casein (CAS)-enriched or corn gluten meal (CGM)-enriched protein sources at different digestible crude protein levels (normal [N], 13%; and low [L], 11%) were applied to 24 crossbred (Duroc × Landrace × Yorkshire) growing pigs (average body weight = 43.3 ± 3.5 kg) in 4 treatments (N.CAS, L.CAS, N.CGM, L.CGM, respectively) to investigate the effects of AA release kinetics on nitrogen deposition in growing pigs. Standardized ileal digestible AA in all diets were balanced by adding individual AA to meet the nutrient requirements. The AA release kinetics were detected in vitro by measuring the hydrolysis of various protein diets under pepsin and trypsin conditions. The results demonstrated that the time of AA release peak in the CGM diet was 12 h later than that in the CAS diet. The synchronization indices of dietary AA release in N.CAS, N.CGM, L.CAS, and L.CGM were 23.73%, 29.37%, 23.40%, and 26.07%, respectively. The N.CGM had the poorest AA release synchronism while the N.CAS had the greatest among the 4 diets. However, within the pigs, L.CAS and N.CGM showed the highest (81.08%) and lowest (73.54%) nitrogen biological values, respectively, despite the standard ileal digestible AA levels being equal for all diets. These results indicate that the release kinetics of dietary AA had great effect on nitrogen deposition. To optimize nitrogen deposition, AA release kinetics and composition should be taken into consideration when formulating diets for growing pigs.

Effects of Dietary Glutamine Supplementation on Nutrient Absorption and Activity of Enzymes Involved in Glutamine Metabolism and Energy Production in the Jejunum of Weaned Piglets

Abstract:Glutamine has an essential role with a beneficial function in improving the nutrition status of young mammals. The influence of L-Glutamine (GM) on the Coefficient of Total Tract Apparent Digestibility (CTTAD) and Apparent Ileal Digestibility (AID), the jejunal enzyme activity associated with nutrient absorption and the energy production in weaned piglets has not been sufficiently studied. The aim of the present study is to provide a profile of the effects of Gln on CTTAD, AID, the activities of jejunal enzymes in connection with nutrient digestion and absorption and energy production. The piglets were weaned at 21 days of age. There were two groups in Experiment 1 and 2 representing supplementation with 0 or 1% Gln to the basal diet. In Experiment 1, the CTTAD of the dietary components and energy was assessed at 3, 5, 10, 15 and 30 days after weaning. In Experiment 2, productive performance, AID, jejunal enzyme activities and expression of Peroxisome Proliferator-Activated Receptor gamma (PPAR gamma) were measured at 10 and 30 days post-weaning. Results showed that dietary Gln supplementation significantly improved the CTTAD of DM, OM, GE and AA and the CTTAD increased significantly with the extension of days after weaning. For the entire experiment, the average daily gain increased by 12.40% (p = 0.049) in the Gin group. Dietary Gln supplementation increased the AID of GE, Leu, Lys, Cys and Pro by 12.50 (p = 0.047), 7.03% (p = 0.041), 5.95% (p = 0.036), 9.30 (p = 0.025), 11.17% (p = 0.009), respectively at 10 days post-weaning; Pro by 6.11% (p = 0.044) at 30 days post-weaning. Jejunal brush border membrane-bound alkaline phosphatase activity increased in the Gln-supplemented pigs by 30.36% (p = 0.048) and 6.21% (p = 0.30) at 10 and 30 days post-weaning, respectively. Compared with the control pigs, the mRNA level of PPAR gamma decreased by 10.85% (p = 0.14) and 41.88% (p = 0.023) after the administration of 1% Gln for 10 and 30 days, respectively. The activity of glutamine synthetase decreased by 48.89% (p = 0.044) at 10 days post-weaning and pyruvate kinase by 13.13% (p = 0.036) at 30 days post-weaning in the Gln-supplemented pigs. In conclusion, 1% Gln supplementation to the post-weaned piglet diet enhanced the CTTAD and AID of diet, improved intestinal absorption and modified jejunal enzyme activities related to Gin metabolism and energy production.
Effects of Dietary Lysine Levels on Apparent Nutrient Digestibility and Serum Amino Acid Absorption Mode in Growing Pigs

Abstract:Two experiments were conducted to determine the effects of different dietary lysine levels on the apparent nutrient digestibility, the serum amino acid (AA) concentration, and the biochemical parameters of the precaval and portal vein blood in growing pigs. In Experiment 1, 15 noncannulated pigs received diets with different lysine densities (0.65%, 0.95%, and 1.25% lysine) for 13 d. A total collection digestion test was performed, and blood samples were collected from the precaval vein at the end of the experiment. In Experiment 2, four cannulated pigs were fed the same diets of Experiment 1. The experiment used a self-control experimental design and was divided into three periods. On d 5 of each period, at 0.5 h before feeding and hourly up to 8 h after feeding, single blood samples were collected from catheters placed in the portal vein. In Experiment 1, some serum AAs (including lysine), serum urinary nitrogen (SUN), and total protein (TP) concentrations were significantly affected by the dietary lysine levels (p<0.05). Moreover, the 0.65% lysine treatment showed a significant lower apparent digestibility of gross energy, dry matter, crude protein, and phosphorus than the other treatments (p<0.05). In Experiment 2, serum lysine, histidine, phenylalanine, threonine, valine, isoleucine (p = 0.0588), triglyceride, and SUN (p = 0.0572) concentrations were significantly affected by the dietary lysine levels (p<0.05). Additionally, almost all of the determined serum AA and total AA concentrations reached their lowest values at 0.5 h before feeding and their highest values at 2 h after feeding (p<0.05). These findings indicate that the greatest absorption of AA occurred at 2 h after feeding and that the dynamic profile of serum AA is affected by the dietary lysine levels. Moreover, when the dietary lysine content was 0.95%, the growing pigs achieved a better nutrient digestibility and serum metabolites levels.

Response to Dietary L-glutamine Supplementation in Weaned Piglets: a Serum Metabolomic Comparison and Hepatic Metabolic Regulation Analysis.

Abstract:A novel metabolomic method based on gas chromatography-mass spectrometry was applied to investigate serum metabolites in response to dietary Gln supplementation in piglets. Sixteen, 21-d-old pigs were weaned and assigned randomly to 2 isonitrogenous diets: 1) Gln diet, which contained 1% L-Gln (as-fed basis), and 2) control diet, which contained L-Ala to make this diet isonitrogenous with the Gln diet. Serum samples were collected to characterize metabolites after a 30-d treatment. in addition, 4 liver samples per treatment were collected to examine enzyme activity and gene expression involved in metabolic regulation. Results indicated that 12 metabolites were altered (P < 0.05) by Gln treatment, including carbohydrates, AA, and fatty acids. A leave-one-out cross validation of random forest analysis indicated that Pro was most important among the 12 metabolites. Thus, these data demonstrate that the control and Gln-supplemented pigs differed (P < 0.05) in terms of metabolism of carbohydrates, Pro, Tyr, and glycerophospholipids. Principal component analysis yielded separate clusters of profiles between the Gln and control groups. Metabolic enzyme activities of Ala aminotransferase and hexokinase increased by 26.8% (P = 0.026) and 26.2% (P = 0.004) in the liver of Gln-supplemented pigs vs. control, respectively, whereas pyruvate kinase (PK) activity decreased by 29.1% (P = 0.001). The gene expression of PK in the liver decreased by 66.1% (P = 0.034) by Gln treatment for 30 d. No differences were observed for the mRNA abundance of mammalian target of rapamycin and PPARγ. On the basis of these data, Gln treatment affected carbohydrate, lipid, and AA metabolism in the whole body of the early weaned piglets. These findings provide insight into specific metabolic pathways and lay the groundwork for the complex metabolic alteration in response to dietary Gln supplementation of pigs.

Abstract:The dietary contents of crude protein and free amino acids (AA) may affect the protein digestion and AA absorption in pigs. Trypsin and chymotrypsin activities, AA serum concentrations and expression of AA transporters in the small intestine of pigs fed a low protein, AA-supplemented (19.2%, LPAA) or a high protein (28.1%, HP), wheat-soybean meal diet were measured in two 14-d trials. The LPAA diet contained free L-Lys, L-Thr, DL-Met, L-Leu, L-Ile, L-Val, L-His, L-Trp and L-Phe. All pigs were fed the same amount of feed (890 and 800 g/d for trial 1 and 2 respectively). In trial 1, samples of mucosa (duodenum, jejunum and ileum) and digesta (duodenum and jejunum) were collected from 14 pigs (17.2 ± 0.4 kg); in trial 2, blood samples were collected from 12 pigs (12.7 ± 0.3 kg). The trypsin and chymotrypsin activities in both intestinal segments were higher in pigs fed the HP diet (p < 0.01). Trypsin activity was higher in jejunum than in duodenum regardless the dietary treatment (p < 0.05). Pigs fed the LPAA diet expressed more b0,+ AT in duodenum, B0 AT1 in ileum (p < 0.05), and tended to express more y+ LAT1 in duodenum (p = 0.10). In pigs fed the LPAA diet, the expression of b0,+ AT was higher in duodenum than in jejunum and ileum (p < 0.01), but no difference was observed in pigs fed the HP diet. Ileum had the lowest b0,+ AT expression regardless the diet. The serum concentrations of Lys, Thr and Met were higher in LPAA pigs while serum Arg was higher in HP pigs (p < 0.05). Serum concentrations of AA appear to reflect the AA absorption. In conclusion, these data indicate that the dietary protein contents affect the extent of protein digestion and that supplemental free AA may influence the intestinal site of AA release and absorption, which may impact their availability for growth of young pigs.

Abstract:Abstract The impact of individual amino acids (AA) on gut hormone secretion and appetite regulation in pigs remains largely unknown. The aim of the present study was to determine the effect of the 20 proteinogenic AA on the release of the anorexigenic hormones cholecystokinin (CCK) and glucagon-like peptide 1 (GLP-1) in postweaning pigs. Six 25-d-old male piglets (Domestic Landrace × Large White; body weight = 6.94 ± 0.29 kg) were humanely killed for the collection of intestinal segments from the duodenum, jejunum, and ileum. Tissue samples from the three intestinal segments were used to determine which of the regions were more relevant for the analysis of gut peptides. Only the segments with the highest CCK and GLP-1 secretion and expression levels were evaluated with the 20 individual AA. Tissue segments were cut open, cleaned, and stripped of their muscle layer before identical circular samples were collected and incubated in 24-well plates for 1 h (37 °C, 5% v/v CO2). The culture broth consisted of a glucose-free KRB buffer containing no added AA (control) or with the addition of 10 mM of 1 of the 20 proteinogenic AA. Following incubation, tissues and supernatant were collected for gene expression and secretion analysis of CCK and GLP-1 levels. CCK secretion and mRNA expression were higher (P < 0.05) in duodenum when compared with proximal jejunum or ileum, whereas GLP-1/proglucagon levels were higher in ileum vs. duodenum (P < 0.05) and jejunum (P < 0.05, for GLP-1 only) in postweaning pigs. Based on these results, the effect of AA on CCK and GLP-1 secretion was studied in the duodenum and ileum, respectively. None of the AA tested stimulated both anorexigenic hormones. Of all the essential AA, Ile, Leu, Met, and Trp significantly (P < 0.05) stimulated GLP-1 from the ileum, while only Phe stimulated CCK from the duodenum. Of the nonessential AA, amide AA (Gln and Asn) caused the release of CCK, while Glu and Arg increased the release of GLP-1 from the ileum. Interpreting the results in the context of the digestion and absorption dynamics, non-bound AA are quickly absorbed and have their effect on gut peptide secretion limited to the proximal small intestine (i.e., duodenum), thus, mainly CCK. In contrast, protein-bound AA would only stimulate CCK release from the duodenum through feedback mechanisms (such as through GLP-1 secreted mainly in the ileum).

Response to Weaning and Dietary L-glutamine Supplementation: Metabolomic Analysis in Piglets by Gas Chromatography/mass Spectrometry.

Abstract:A novel metabolomic method based on gas chromatography/mass spectrometry (GC-MS) was applied to determine the metabolites in the serum of piglets in response to weaning and dietary L-glutamine (Gln) supplementation. Thirty-six 21-d-old piglets were randomly assigned into three groups. One group continued to suckle from the sows (suckling group), whereas the other two groups were weaned and their diets were supplemented with 1% (w/w) Gln or isonitrogenous L-alanine, respectively, representing Gln group or control group. Serum samples were collected to characterize metabolites after a 7-d treatment. Results showed that twenty metabolites were down-regulated significantly (P<0.05) in control piglets compared with suckling ones. These data demonstrated that early weaning causes a wide range of metabolic changes across arginine and proline metabolism, aminosugar and nucleotide metabolism, galactose metabolism, glycerophospholipid metabolism, biosynthesis of unsaturated fatty acid, and fatty acid metabolism. Dietary Gln supplementation increased the levels of creatinine, D-xylose, 2-hydroxybutyric acid, palmitelaidic acid, and α-L-galactofuranose (P<0.05) in early weaned piglets, and were involved in the arginine and proline metabolism, carbohydrate metabolism, and fatty acid metabolism. A leave-one-out cross-validation of random forest analysis indicated that creatinine was the most important metabolite among the three groups. Notably, the concentration of creatinine in control piglets was decreased (P=0.00001) compared to the suckling piglets, and increased (P=0.0003) in Gln-supplemented piglets. A correlation network for weaned and suckling piglets revealed that early weaning changed the metabolic pathways, leading to the abnormality of carbohydrate metabolism, amino acid metabolism, and lipid metabolism, which could be partially improved by dietary Gln supplementation. These findings provide fresh insight into the complex metabolic changes in response to early weaning and dietary Gln supplementation in piglets.
Apparent Precaecal Digestibility of Nutrients and Level of Endogenous Nitrogen in Digesta of the Small Intestine of Growing Pigs as Affected by Various Digesta Viscosities

Abstract:Sixteen male growing pigs of about 24 kg BW were fitted with both a duodenal re-entrant and a post-valve T-shaped cannula inserted in the caecum. The animals were divided into four groups. Each group received one of the following diets: corn starch-soybean protein isolate-based diet without (diet C) and with carboxymethylcellulose (diet CMC) or a rye-wheat-based diet without (diet RW) and with xylanase addition (diet RWX). The diets provided similar levels of apparent precaecal digestible crude protein (CP), lysine, methionine + cystine, threonine and tryptophan. Additionally, [15N]-yeast was given with the diets during the first 10 days of the experiment. For estimation of digesta viscosity, N-flow of dietary and endogenous origin, apparent precaecal digestibilities of dry matter (DM), CP, amino acids and non starch polysaccharides (NSP) (only in pigs fed diets RW and RWX), ileal and duodenal digesta were quantitatively collected on day 16 and 17, respectively. The endogenous N-proportion was measured by the ratio of 15N enrichment in the digesta and urine. The duodenal and ileal digesta supernatant viscosity increased as carboxymethylcellulose was included into the diet. Xylanase addition to the rye-wheat based diet reduced the viscosity in the ileal digesta. There were no differences in precaecal digestibilities of DM, CP and amino acids between diet C and CMC. The precaecal digestibilities of DM and soluble and insoluble NSP increased from 69.5% to 73.9%, from 1.3% to 47.9% and from 17.0% to 35.4%, respectively, as xylanase was added to the rye-wheat-based diet. The apparent precaecal digestibility of most essential amino acids increased by 2 to 5 percent units. The amounts of endogenous N at the duodenal level were estimated to be 158, 233, 313 and 276 mg per 12 h per kg0.75 BW of pigs fed diets C, CMC, RW and RWX, respectively. The corresponding values at the ileal level were 95, 107, 164 and 150 mg per 12 h per kg0.75 BW. For endogenous N amounts, significant differences were observed between diets C and CMC (duodenum) and also between semi-purified and cereal-based diets (duodenum and ileum). Methodological aspects for the estimation of endogenous N using the isotope dilution technique are discussed. Obviously, the digesta viscosity per se does not affect the nutrient absorption and endogenous N flow within the small intestine of pigs. Other properties of complex dietary fibre, digesta passage rate or bacterial activity probably contribute to the observed changes.
Amino acid digestibility in plant protein sources fed to growing pigs

Abstract:OBJECTIVE: The objective was to determine standardized ileal digestibility (SID) of amino acids (AA) in 11 plant protein sources fed to growing pigs.METHODS: Eleven feed ingredients used were sesame meal, two sources of soybean meal (SBM) produced in the Republic of Korea, a source of SBM produced in India, high-protein distillers dried grains (HPDDG), perilla meal, canola meal, copra meal, corn germ meal, palm kernel expeller, and tapioca distillers dried grains (TDDG). Experimental diets were prepared to contain each test ingredient as a sole source of AA, and a nitrogen-free diet was also prepared to estimate the basal ileal endogenous losses of AA. Twelve barrows surgically fitted with T-cannulas the distal ileum with an initial body weight of 29.0 kg (standard deviation = 3.0) were individually housed in metabolism crates equipped with a feeder and a nipple drinker. A 12 × 9 incomplete Latin square design was employed with 12 experimental diets, 12 animals, and 9 periods. After a 5-d adaptation period, ileal digesta were collected on d 6 and 7 in each experimental period.RESULTS: Values for apparent ileal digestibility of most indispensable AA in three sources of SBM were greater compared with other test ingredients except HPDDG and canola meal (p<0.05). Pigs fed diets containing SBM sources had also greater SID of most indispensable AA compared with those fed diets containing other test ingredients (p<0.05) except for HPDDG and canola meal. There was no difference in the apparent ileal digestibility and SID of AA among sources of SBM. The TDDG had the least value for the SID of methionine among test ingredients (p<0.05).CONCLUSION: Standardized ileal digestibility of most amino acids in soybean meal, high-protein distillers dried grains, and canola meal were greater than those in sesame meal, perilla meal, copra meal, and tapioca distillers dried grains.

Abstract:Abstract Background Determination of nutrient requirements in the late gestating and lactating sows is essential to optimize sow productivity. The objectives of the present study were to quantify amino acid (AA) fluxes and heat production across portal-drained viscera (PDV) and liver in multiparous sows during transition and lactation. Methods Eight second parity sows were fitted with indwelling catheters in the femoral artery and in the mesenteric, portal and hepatic veins. Eight hourly sets of blood samples were taken starting 0.5 h before feeding at − 10, − 3, + 3, and + 17 d in milk (DIM). Blood gases, plasma metabolites and apparent total tract digestibility (ATTD) of nutrients were measured. Results Feed intake, the ATTD of DM, energy, nitrogen, fat and crude fiber changed with DIM ( P < 0.001). Except for Glu, O 2 , and urea, all net portal fluxes were positive, and all were affected by DIM ( P < 0.05) and by sampling time ( P < 0.01). Compared with pre partum levels, net portal uptake of AA was 3-63% lower at + 3 DIM but 40-100% higher at + 17 DIM. Net portal fluxes of AA peaked at 1.5 to 2.5 h after feeding except for Glu, and they were positively correlated with changes in sow feed intake across DIM. The net portal recovery was low for Met (49%), Thr (54%), and His (54%) and high for the remaining essential AA (63-69%) and none of them differed across DIM. Net hepatic uptake (i.e. hepatic oxidation) of Lys, Thr, Ile, Leu and Phe peaked at 0.5 to 2.5 h after feeding, whereas uptake of Trp, Val, and His was constant, while that of Met was close to zero. Conclusion The net portal recovery was substantially lower for Met, Thr, and His than the remaining essential AA. Hepatic AA oxidation peaks 0.5 to 2.5 h after feeding. The heat production in PDV and liver was approximately two-fold higher at peak lactation compared to other stages. The study suggests that lysine was the limiting AA in peak lactation but not in early lactation.

Abstract:The traditionally classified nutritionally non-essential amino acids are now known to be insufficiently synthesized for maximal growth and optimal health in piglets. This study determined the effects of dietary supplementation with an amino acid blend (AAB; glutamate:glutamine:glycine:arginine:N-acetylcysteine = 5:2:2:1:0.5) on piglet growth performance and intestinal functions. Sixteen piglets (24-day-old) were randomly assigned to a corn and soybean meal-based diet supplemented with 0.99% alanine (isonitrogenous control) or 1% AAB. On day 20 of the trial, blood and intestinal tissue samples were obtained from piglets. Compared with the control, AAB supplementation reduced (P < 0.05) diarrhoea incidence; plasma alanine aminotransferase and diamine oxidase activities; intestinal concentrations of hydrogen peroxide, malondialdehyde, and heat shock protein-70, and intestinal mRNA levels for interleukin-1β, interferon-γ, and chemokine (C–X–C motif) ligand-9; and the numbers of Enterobacterium family, Enterococcus genus and Clostridium coccoides in the colon digesta. Furthermore, AAB supplementation enhanced (P < 0.05): the plasma concentrations of serine, aspartate, glutamate, cysteine, tyrosine, phenylalanine, tryptophan, lysine, arginine, citrulline, ornithine, taurine, and γ-aminobutyric acid; intestinal villus height and surface area, villus height/crypt depth ratio, antioxidative enzyme activities, and mRNA levels for porcine β-defensin-1, sodium-independent amino acid transporters (b0,+AT and y+LAT1), aquaporin (AQP) 3, AQP8, AQP10, nuclear factor erythroid 2-related factor 2 and glutathione S-transferase omega-2, and protein abundances of AQP3, AQP4, claudin-1, occludin and myxovirus resistance 1; and the numbers of Bifidobacterium genus and Lactobacillus genus in the colon digesta. Collectively, these comprehensive results indicate that dietary AAB supplementation plays an important role in improving piglet growth and intestinal function.

Abstract:Background: Methionine (Met) is a popular nutritional supplement in humans and animals. It is routinely supplemented to pigs as L-Met, DL-Met, or DL-2-hydroxy-4-(methylthio) butanoic acid (DL-HMTBA). Methods: We investigated the effect of these Met supplements on jejunal amino acid (AA) transport in male castrated Piétrain × Danbred pigs, also including a non-supplemented group. The mucosal-to-serosal flux of ten [14C]-labeled AAs (L-glutamine, glycine, L-leucine, L-lysine, L-Met, L-serine, L-threonine, L-tryptophan, L-tyrosine and L-valine) was investigated at two concentrations (50 μM and 5 mM). Inhibition of apical uptake by mucosal L-Met was also measured for these AAs. The intestinal expression of apical AA transporters, angiotensin-converting enzyme II and inflammation-related genes were compared with those of a previous study. Results: Except for tryptophan and lysine at 5 mM, all AA fluxes were Na+-dependent (p ≤ 0.05), and the uptake of most AAs, except glycine and lysine, was inhibited by L-Met (p < 0.001). A correlation network existed between Na+-dependent fluxes of most AAs (except tryptophan and partly glycine). We observed the upregulation of B0AT1 (SLC6A19) (p < 0.001), the downregulation of ATB0,+ (SLC6A14) (p < 0.001) and a lower expression of CASP1, IL1β, IL8, TGFβ and TNFα in the present vs. the previous study (p < 0.001). Conclusions: The correlating AAs likely share the same Na+-dependent transporter(s). A varying effect of the Met supplement type on AA transport in the two studies might be related to a different level of supplementation or a different inflammatory status of the small intestine.

Effect of the essential amino acid-nitrogen to total nitrogen ratio on lysine requirement for nitrogen retention in growing pigs

Abstract:Low protein diets supplemented with essential amino acids (EAA) fed to pigs reduce the excess supply of EAA and nitrogen (N). However, low protein diets may become limiting in non-essential amino acids (NEAA) and N, thus affecting the utilization of EAA for N retention. It has been suggested that the EAA-N:total N (E:T) ratio can give an indication of dietary N sufficiency. An N-balance study was conducted to determine the effect of E:T ratio on the Lys requirement for maximum N retention. A total of 80 growing barrows (19.3 ± 0.21 kg initial body weight) were randomly assigned to 1 of 10 diets (n = 8) in 8 blocks in a 2 × 5 factorial arrangement. Diets consisted of a low ratio (LR; E:T of 0.33) or a high ratio (HR; E:T of 0.36) with graded Lys content (0.82%, 0.92%, 1.02%, 1.12%, and 1.22% standardized ileal digestible [SID]). After a 7-d adaptation, a 4-d N-balance collection was conducted. Blood samples were obtained on d 2 of the collection period 2 h after the morning meal for plasma urea N (PUN) analysis. Data were analyzed using the MIXED model procedure with fixed effects of ratio (n = 2), Lys (n = 5), and their interactions. The experimental block (room) was included as a random effect (n = 8). The SID Lys requirement was estimated using PROC NLIN linear broken-line breakpoint model. There was a significant interaction between E:T ratio and Lys (P < 0.01), where LR diets had a higher N retention than HR diets, while increasing Lys linearly increased N retention (P = 0.01) in both HR and LR diets. The marginal efficiency of utilizing SID Lys (P < 0.01) reduced with increasing Lys content, while the efficiency of utilizing N (P < 0.05) increased as Lys increased. The SID Lys required to maximize N retention of pigs fed HR diets was estimated at 1.08% (R2 = 0.61) and LR diets at 1.21% (R2 = 0.80). The current results indicate that N may be limiting in diets with a high E:T ratio, limiting N retention. Supplying additional dietary N, as intact protein, can increase N retention, resulting in a greater Lys requirement.
Plasma amino acid uptake by the mammary gland of the lactating sow

Abstract:The objectives of this study were to measure arteriovenous concentration (A-V) differences and estimate uptake of amino acids across the mammary gland in lactating sows. Four sows were used in Trial 1 and nine sows in Trial 2. Cannulas were fitted in the right anterior mammary vein and the carotid artery around d 7 of lactation. Arteriovenous samples were obtained on d 11, 14, 17, and 20. Litters were separated from the sows for 90 min, after which the first blood samples were drawn. Samples were taken at 20-min intervals for the next two consecutive hours. Milk production measurements and milk samples were obtained between d 11 and 19. Liters of plasma to liters of milk ratio (conversion coefficient) was estimated with the Fick method using A-V difference for lysine and milk lysine concentration. The conversion coefficient and the daily plasma flow were 541.41 +/- 35.72 L of plasma per liter of milk and 4,275 +/- 386 L of plasma, respectively. The A-V differences and the plasma flow were used to quantify daily amino acid uptake by the mammary gland. Uptake estimates (grams/day) were 36.51 +/- 4.05 (leucine), 31.24 +/- 3.64 (arginine), 23.39 +/- 2.97 (lysine), 21.22 +/- 1.96 (valine), 18.36 +/- 1.92 (isoleucine), 15.9 +/- 1.9 (threonine), 15.46 +/- 1.58 (phenylalanine), 7.61 +/- 1.12 (histidine), and 6.54 +/- 2.01 (methionine). The uptakes of arginine, leucine, isoleucine, valine, phenylalanine, and threonine significantly exceeded output of these amino acids in the milk. These results indicate that the sow mammary gland retains specific amino acids above requirements for milk protein synthesis.
