Abstract:An NHC-catalyzed annulation reaction of 3-halopropenals with nitrosobenzene derivatives, providing access to 2,3-disubstituted isoxazol-5(2H)-ones under mild conditions is described. This procedure represents a metal-free approach to the alternative equivalent of beta-acylvinyl anions for the formation of heterocyclic compounds.

Abstract:Over the past decade, our group has developed a series of synthetic methods for the preparation of heterocycles by using two different approaches: transition-metal-catalyzed cyclization and iodine/oxidant-catalyzed oxidative cyclization. Transition-metal salts of palladium or iron and nanoparticulate copper(II) oxide have been used as catalysts for the synthesis of heterocyclic compounds. Palladium-catalyzed Wacker-type oxidative heterocyclic cyclization, iron-catalyzed benzylation and tandem dehydration annulation, and nanoparticulate copper(II) oxide-catalyzed heterogeneous oxidative cyclizations have been developed in our laboratory. For iodine-catalyzed reactions, we have developed an oxidative tandem cyclization, an oxidative decarboxylative coupling cyclization, a direct Mannich-like reaction, and an oxidative amination catalyzed by diiodine and tert-butyl hydroperoxide under mild condition. These methods have enriched the range of synthetic strategies available for the construction of heterocyclic skeletons and they have potential applications in the fields of medicinal chemistry and materials science.

Abstract:Abstract: The reactivity of aldehydes and ketones carries great potential for multicomponent heterocyclizations. These reactions are convergent and highly versatile in the development of synthetic methodologies for compound families, displaying variations in substituents in their structures. Therefore, they have been regarded as an important tool in the field of Green Chemistry. Furthermore, they prove to be very useful in studies of biological activity, where small structural modifications can result in significant differences. Many heterocyclizations date back to the mid-19th and early 20th centuries. In this review, we aim to demonstrate, through some of these reactions, their continuously growing potential and improvements concerning synthetic development. Additionally, we present the original studies as reported, enabling us to appreciate the evolution of chemical representations over the years until reaching the standardization we have today.

Abstract:Recent advances in the environmentally benign synthesis of aromatic N-heterocycles are reviewed, focusing primarily on the application of catalytic methods and non-traditional activation. This account features two main parts: the preparation of single ring N-heterocycles, and their condensed analogs. Both groups include compounds with one, two and more N-atoms. Due to the large number of protocols, this account focuses on providing representative examples to feature the available methods.

Abstract:The role of allylic substitution reactions is crucial in organic synthesis. Through these reactions, the development of numerous molecular structures with a wide range of applications can be achieved in diverse fields. They also provide an efficient methodology for synthesizing novel heterocyclic compounds with potential as drugs, natural products, and bioactive structures. This review summarizes the research progress in cyclization reactions via catalytic allylic substitution for the asymmetric synthesis of heterocyclic compounds over the past five years. The discussion focused on catalytic asymmetric allylic substitutions with nitrogen, oxygen and carbon nucleophiles.

Abstract:The enzymatic synthesis of heterocycles is an emerging biotechnology for the sustainable construction of societally important molecules. Herein, we describe an enzyme-mediated strategy for the oxidative dimerization of thioamides enabled by enzymatic halide recycling by vanadium-dependent haloperoxidase enzymes. This approach allows for intermolecular biocatalytic bond formation using a catalytic quantity of halide salt and hydrogen peroxide as the terminal oxidant. The established method is applied to a diverse range of thioamides to generate the corresponding 1,2,4-thiadiazoles in moderate to high yield and with excellent chemoselectivity. Mechanistic experiments suggest that the reaction proceeds through two distinct enzyme-mediated sulfur halogen- ation events that are critical for heterocycle formation. Molecular docking experiments provide insight into reactivity differences between biocatalysts used in the study. Finally, the developed biocatalytic oxidative dimerization is applied to a preparative scale chemoenzymatic synthesis of the anticancer agent penicilliumthiamine B. These studies demonstrate that enzymatic halide recycling is a promising platform for new-to-nature intermolecular bond formation.

Abstract:Heterocycles are important compounds existed in diverse advanced chemicals, functional materials, bioactive molecules and natural products. Different strategies through transition metal‐catalyzed alkyne cyclization have been developed to synthesize various heterocycles. The merger of alkyne cyclization and metal nanoparticle catalysis has emerged as an important approach to diverse heterocycles. By using different kinds of metal nanoparticle catalysts, such as gold, silver, copper and palladium nanoparticles, a number of heterocycles are prepared with high yield and selectivity in a green, sustainable and highly efficient manner. This review summarizes and discusses recent achievements in metal nanoparticle‐catalyzed alkyne cyclization for the synthesis of heterocycles.

Abstract:N-arylated heterocycles are a significant class of core scaffolds in medicinal chemistry, materials science, and agrochemistry, highlighting their importance in various fields. The development of innovative methodologies for synthesizing these fundamental structures has been a central focus in organic synthesis. Over the past few decades, numerous approaches have been established to synthesize N-aryl heterocycles efficiently. Among these methods, the direct N-arylation of N-H heterocycles stands out as one of the most straightforward and robust strategies for accessing N-arylated heterocycles. This review provides a comprehensive review of the recent advances in the synthesis of N-arylated heterocycles, encompassing the relevant literature from the past decade. The review summarizes the N-arylation of N-H heterocycles using various catalytic systems, including palladium, nickel, copper, visible light-induced metal-catalyzed, and metal-free catalyzed methodologies. These advances highlighted the continuous evolution and optimization of synthetic strategies to create diverse and complex N-arylated heterocycles, which are pivotal for furthering research and development in multiple scientific domains.

Abstract:Heteroarene 1, n-zwitterions are powerful and versatile building blocks in the construction of heterocycles and have received increasing attention in recent years. In particular, pyridinium and quinolinium 1,4-zwitterions have been widely studied and used in a variety of cyclization reactions due to their air stability, ease of use, and high efficiency. Sulfur- and nitrogen-based pyridinium and quinolinium 1,4-zwitterions, types of emerging heteroatom-containing synthons, have attracted much attention from chemists. These 1,4-zwitterions, which contain multiple reaction sites, have been successfully used in the synthesis of three- to eight-membered cyclic compounds over the last decade. In this review, we present the exciting progress made in the field of cyclization reactions of sulfur- and nitrogen-based pyridinium and quinolinium 1,4-zwitterions. Moreover, the mechanistic insights, the transition states, some synthetic applications, and the challenges and opportunities are also discussed. We hope to provide an overview for synthetic chemists who are interested in the heterocycle synthesis from cyclization reaction with pyridinium and quinolinium 1,4-zwitterions pyridinium and quinolinium 1,4-zwitterions.

Abstract:During the past few decades, there has been considerable growth in the development of denitrogenative reactions of triazole skeletons to construct valuable cyclic compounds, particularly heterocycles. Despite the inherent difficulty of the ring-opening of triazole derivatives, many novel and efficient approaches have arisen in this area mainly with the use of transition metal (such as rhodium, palladium, silver, copper) catalysis, photolysis, or free radical mediated reactions. Generally, these procedures begin with the ring-opening of 1,2,3-triazoles or benzotriazoles followed by N2 extrusion and subsequent diverse transformations, which enable the rapid synthesis of various heterocycles in a single step. To avoid overlap with other related reviews, this minireview covers the recent advances in the denitrogenative cyclization of 1,2,3-triazoles since 2016 and the denitrogenative cyclization of benzotriazoles since 2012.

Abstract:This present review delineates the repertoire of vinyl cyclopropanes and their stuctural analogues to accomplish a wide array of oxa-cycles, aza-cycles, and thia-cycles under transition metal catalysis and metal-free approaches from early 2019 to the present date. The generation of electrophilic π-allyl intermediates and 1–3/1–5-dipolarophile chemistry originating from VCPs are always green, step- and atom-economical and sustainable strategies in comparsion with prefunctionalized and/or C–H activation protocols. Here, the strained ring-system extends its applicability by relieving the strain to undergo a ring-expansion reaction to accomplish 5–9 membered carbo- and heterocyclic systems. The availability of chiral ligands in the ring-expansion reaction of VCPs and their analogues has paved the way to realizing asymmetric synthetic transformations.

Abstract:Polynitrogen heterocycles are readily available and have recently arisen as versatile synthons for the formation of various C–C and C–X bonds, and medicinally active nitrogen-containing heterocycles. Several cascade reactions, including annulation, radical cascade, and borylation reactions, have been reported in which polynitrogen heterocycles are applied as arylation reagents. The success of these exceptional reactions illustrates the great synthetic potential of polynitrogen heterocycles, which provides a direct and useful approach to arylation reactions and the synthesis of nitrogen-containing heterocycles. The use of photocatalysts to effectively transfer energy from visible light to non-absorbing compounds has gained increasing attention as this method allows for the mild and efficient generation of radicals in a controlled manner. This approach has thus led to new methods that involve unique bond formation reactions. In addition, the use of free radical intermediates stabilized by transition metal catalysts is a powerful way to construct new chemical bonds. The aim of this review is to highlight the rapidly expanding area of radical-initiated denitrogenative cascade reactions of polynitrogen heterocycles and elaborate on their mechanisms from a new perspective by using photocatalysis and metal-based catalysis.This review provides a thorough overview of radical-initiated denitrogenative cascade reactions of polynitrogen heterocycles in the formation of various C–X bonds and medicinally active nitrogen-containing heterocycles.

Abstract:This review concentrates on recent developments from our laboratory concerning the Rh-catalyzed carbonylative C-C bond activation of cyclopropanes. Specifically, we have found that N-based directing groups are effective at controlling the regioselectivity of C-C bond activation during the formation of rhodacyclopentanone intermediates. These engage tethered π-unsaturated components (e.g. alkenes) or conventional nucleophiles in cycloaddition and heterocyclization processes, respectively. Using this approach, direct and modular access to a wide range of complex heterocyclic ring systems is achieved. The review summarizes the scope of our methodologies and outlines key mechanistic features.

Abstract:5-endo-trig radical cyclization has long been recognized as one of the most straightforward ways for the construction of densely functionalized five-membered rings. Nevertheless, according to Baldwin's rules, the 5-endo-trig radical cyclization is kinetically disfavored due to stereoelectronic effects and thus usually proceeds via a slow rate, which renders its application a challenging task. In recent years, with the emergence of efficient radical generation methods and effective cyclization strategies, 5-endo-trig radical cyclization has been successfully accelerated to a synthetically useful rate and has been utilized in the access of diverse five-membered carbo- and heterocyclic compounds. This review comprehensively summarizes the methodologies involving the 5-endo-trig radical cyclization process, with particular emphasis on the elucidation of the promoting strategies, which include the polar effect, geometrical constraints, spin delocalization effect, and persistent radical effect. Each of these strategies is discussed in detail, with illustrative examples from recent literature studies to highlight their practical applications and effectiveness. It is anticipated that the in-depth understanding of the 5-endo-trig radical cyclization provided by this review would inspire further innovation of this privileged reaction mode and expand its applications. Moreover, the potent ring-closure-promoting strategies revealed herein would also contribute to achieving other challenges of cyclizations with particular significance for organic synthesis.

Abstract:Background: Heterocyclic scaffolds widely occur in natural products and pharmaceutical molecules and because of their broad biological activities, the synthesis of such frameworks with diverse approaches is one of the most attractive fields in modern organic chemistry. Recently, cascade reactions catalyzed by transition metals made astounding advances in the synthesis of heterocyclic compounds for their step economy. Among them, gold-catalyzed cascade reactions emerged as important tools to construct complicated heterocyclic architecture from simple precursors in the past decade. Objective: This review presents an overview of recent advances in gold-catalyzed cascade reactions to generate diverse heterocyclic scaffolds and its applications in total synthesis of natural products.

Abstract:The development of highly efficient methodologies for constructing heterocyclic compounds is a major challenge in organic chemistry. This Account presents the recent progresses on cascade strategies for heterocyclic synthesis based on ketenimine intermediates. Three types of cascade reactions for the synthesis of a variety of heterocycles, involving nucleophilic addition to the central carbon of ketenimine, radical addition to the central carbon of ketenimine, and pericyclic reaction of ketenimine, are highlighted in this Account. A brief discussion on the mechanism of copper-catalyzed azide-alkyne cycloaddition (CuAAC) providing ketenimines is also included.

Abstract:Sequential Pd-catalyzed one-pot synthetic methodologies have emerged as a powerful and versatile approach in organic synthesis, enabling the construction of complex heterocyclic architectures with high efficiency, selectivity, and atom economy. This review discusses key advancements in multistep, sequentially Pd-catalyzed one-pot processes for accessing heterocyclic derivatives, focusing on classic reactions like Suzuki–Miyaura, Sonogashira, Heck, and hydroamination and extending to specialized techniques such as directed C-H activation. The concatenation of these steps has advanced the scope of one-pot strategies. A section is dedicated to exploring the cooperative use of palladium with other metals, particularly copper, ruthenium, and gold, which has broadened the range of accessible heterocyclic derivatives. Highlighted applications include the synthesis of biologically and pharmaceutically relevant compounds, such as tris(hetero)aryl systems, spiro-oxindoles, and indole derivatives. These one-pot strategies not only streamline synthesis but also align with green chemistry principles by minimizing purification steps and reducing waste and energy consumption. The review also addresses current challenges and limitations in these methodologies, offering insights into ongoing efforts to optimize reaction conditions and expand the applicability of sequential Pd-catalyzed processes.

Abstract:Carbon dioxide (CO2) has emerged as a sustainable, feasible, abundant one-carbon synthon and displays great potential in the synthesis of heterocycles such as lactones, lactams, and 2-oxazolidinones, which are privileged motifs in pharmaceutical chemistry demonstrating bioactivities. Although the fixation of CO2 is restricted due to its thermodynamic stability and kinetic inertness, multiple breakthroughs have been realized in annulation chemistry. This review concentrates on the advances made in the last five years in CO2-incorporated cyclization triggered by N-, O-, and C-nucleophiles. Three transformation modes of CO2 including carboxylative cyclization, carbonylative cyclization, and reductive cyclization have been summarized. Moreover, typical mechanisms and significant applications of these reactions are also described.
