Abstract:Research into cancer therapeutics has uncovered various potential medications based on metal-containing scaffolds after the discovery and clinical applications of cisplatin as an anti-cancer agent. This has resulted in many metallodrugs that can be put into medical applications. These metallodrugs have a wider variety of functions and mechanisms of action than pure organic molecules. Although platinum-based medicines are very efficient anti-cancer agents, they are often accompanied by significant side effects and toxicity and are limited by resistance. Some of the most studied and developed alternatives to platinum-based anti-cancer medications include metallodrugs based on ruthenium, gold, copper, iridium, and osmium, which showed effectiveness against many cancer cell lines. These metal-based medicines represent an exciting new category of potential cancer treatments and sparked a renewed interest in the search for effective anti-cancer therapies. Despite the widespread development of metal complexes touted as powerful and promising in vitro anti-cancer therapeutics, only a small percentage of these compounds have shown their worth in vivo models. Metallodrugs, which are more effective and less toxic than platinum-based drugs and can treat drug-resistant cancer cells, are the focus of this review. Here, we highlighted some of the most recently developed Pt, Ru, Au, Cu, Ir, and Os complexes that have shown significant in vivo antitumor properties between 2017 and 2023.

Abstract:Background: Cancer, a group of diseases, is a leading cause of death after heart disease. Many chemotherapies are available to treat cancer; however, several natural substances have been found to be effective for treating cancer, and flavonoids are one of them. Objective: Flavonoids are naturally occurring polyphenolic compounds with no or minimal toxicity, and have been utilized by people since ancient times. They produce several special therapeutic effects, such as anti-inflammatory, immune response modulating, and antioxidant effects, thereby supporting normal cellular functions. Flavonoids can be classified into six major subtypes or groups based on the degree of oxidation, chemical structure, and unsaturation in the linking chain. These are isoflavonoids, flavanones, flavanols, flavonols, flavones and anthocyanidins. Method: A literature search was conducted in Science Direct, Google Scholar, PubMed, Wiley Online Library, Springer, and Medline databases, for studies on flavonoids as anti-cancer agents. Results: The enhanced generation of reactive oxygen species (ROS) in the electron transport chain due to oxidative stress leads to inflammation, the development of many degenerative diseases, cancer, etc. Flavonoids help to relieve oxidative stress by regulating ROS homeostasis (scavenging ROS), triggering apoptotic pathways, and exerting a potent pro-oxidant effect (suppression of pro-oxidant enzymes) in cancerous cells (activation of antioxidant enzymes). Conclusion: In this review article, we have discussed the structure and classification of flavonoids along with their mechanism of action as anticancer agents and the challenges faced in developing flavonoids as anticancer agents.

Abstract:Natural drug-based chemotherapeutic have advanced at an unprecedented rate over the past decade. Owing to protective roles in vegetation, and exceptional antioxidant qualities, polyphenols and flavonoids have demonstrated promising clinical implications in malignancies amongst other plant metabolites. It is widely debated how dietary flavonoids and polyphenols can prevent cancer. Flavonoids are believed to execute a significant role in cancer prevention and treatment, as corroborated from experimental studies, epidemiological research, and human clinical trials. Carcinogen deactivation, antimutagenic, cell cycle arrest, apoptosis and differentiation, suppression of vasculature, oxidative stress, and reversion of multidrug resistance are among the many mechanisms of action that have been discovered. The present review outlines the classification of various polyphenols and flavonoids and their potential as anticancer therapeutic agents.

Abstract:Background: Designing and development of molecules for cancer treatment useful and with no side effects are a big challenge for the researchers in the field of drug discovery. The use of phytochemicals for chemoprevention is gaining more advantages, and intake of flavonoids has proved to reduce the occurrence of various cancers. Objectives: The present study was focused on selecting eight flavonoids and study them by in silico methods to analyse the interactions, affinity and pharmacophoric features that participate in the interactions between the flavonoid and the active sites of different cancer targets. Methods: The cancer targets were downloaded from the protein data bank, and flavonoids from PubChem and were docked by Glide XP molecular docking method to find the molecular interactions. The binding energy was calculated by Prime MM-GBSA application and ADMET analysis by Qikprop of Schrodinger. The anticancer potential of flavonoids screening was based on an online tool, Pass predictor. Phase module was used to find the common pharmacophore features that participate in essential interactions between the flavonoid and the active site. Results: In this study, myricetin has proved to be the best flavonoid for the treatment of breast and lung cancer with docking score of -11.50 kcal/mol and -10.56 kcal/mol respectively, whereas, quercetin has proved to be the best for prostate and colorectal cancer with docking score of -14.18 kcal/mol and -12.94 kcal/mol, respectively. The responsible forces for the interaction of these flavonoids are hydrogen bond, hydrophobic interactions, polar and pi-pi stackings. The PASS tool predicted the anticancer potential for the flavonoids, in particular, myricetin had responded highly active for most cancer cells. The hypothesis AADRR_1 has the highest survival score, which indicates the best alignment of the active ligands and represents the best pharmacophore model for anticancer activity. Conclusion: This work has screened eight flavonoids against various cancer targets and shown the binding interactions between them, stating that myricetin is the suitable lead candidate for breast and lung cancer; whereas, quercetin is the best lead for prostate and colorectal cancer. And these data are about the results obtained from PASS predictor. Moreover, the pharmacophore model has generated for the flavonoids, which correlate activities with the spatial arrangement of various chemical features. Therefore, this investigation strongly suggests that these flavonoids can be used as leads as anticancer agents.

Abstract:Abstract:: Flavonoids are a class of polyphenolic compounds that can be classified into six distinct categories, namely isoflavonoids, flavanones, flavanols, flavonols, flavones, and anthocyanidins. These compounds are naturally occurring and can be found in a diverse range of plant species. Flavonoids, a class of bioactive compounds, are mostly obtained through the consumption of vegetables, fruits and plant-derived beverages such as wine, cocoa-based products and green tea. Flavonoids have been demonstrated to exhibit a diverse range of anticancer properties. These include the modulation of activities of enzymes involved in scavenging reactive oxygen species, involvement in cell cycle arrest, induction of apoptosis and autophagy, as well as suppression of cancer cell proliferation and invasiveness. Flavonoids exhibit a dual role in maintaining reactive oxygen species balance. They function as antioxidants in regular physiological conditions, while also demonstrating significant pro-oxidant properties in cancer cells. This prooxidant activity induces apoptotic pathways and downregulates pro-inflammatory signalling pathways. The paper explores the biochemical characteristics, bioavailability, anticancer efficacy, and modes of action of flavonoids.

Advances in anticancer alkaloid-derived metallo-chemotherapeutic agents in the last decade: Mechanism of action and future prospects

Abstract:Metal-based complexes have occupied a pioneering niche in the treatment of many chronic diseases, including various types of cancers. Despite the phenomenal success of cisplatin for the treatment of many solid malignancies, a limited number of metallo-drugs are in clinical use against cancer chemotherapy till date. While many other prominent platinum and non‑platinum- based metallo-drugs (e.g. NAMI-A, KP1019, carboplatin, oxaliplatin, titanocene dichloride, casiopeinas® etc) have entered clinical trials, many have failed at later stages of R&D due to deleterious toxic effects, intrinsic resistance and poor pharmacokinetic response and low therapeutic efficacy. Nonetheless, research in the area of medicinal inorganic chemistry has been increasing exponentially over the years, employing novel target based drug design strategies aimed at improving pharmacological outcomes and at the same time mitigating the side-effects of these drug entities. Over the last few decades, natural products became one of the key structural motifs in the anticancer drug development. Many eminent researchers in the area of medicinal chemistry are devoted to develop new 3d-transition metal-based anticancer drugs/repurpose the existing bioactive compounds derived from myriad pharmacophores such as coumarins, flavonoids, chromones, alkaloids etc. Metal complexes of natural alkaloids and their analogs such as luotonin A, jatrorrhizine, berberine, oxoaporphine, 8-oxychinoline etc. have gained prominence in the anticancer drug development process as the naturally occurring alkaloids can be anti-proliferative, induce apoptosis and exhibit inhibition of angiogenesis with better healing effect. While some of them are inhibitors of ERK signal-regulated kinases, others show activity based on cyclooxygenases-2 (COX-2) and telomerase inhibition. However, the targets of these alkaloid complexes are still unclear, though it is well-established that they demonstrate anticancer potency by interfering with multiple pathways of tumorigenesis and tumor progression both in vitro and in vivo. Over the last decade, many significant advances have been made towards the development of natural alkaloid-based metallo-drug therapeutics for intervention in cancer chemotherapy that have been summarized below and reviewed in this article.

Abstract:Cancer is a group of diseases involving abnormal cell growth. The cells grow uncontrollably with the potential to invade and spread to other parts of the body. This disease is one of the principal death causes in the world, thus becoming a significant topic of scientific research. On the other hand, transition metals play a fundamental role in different living systems. In particular, Metallodrugs represent new and powerful tools for diverse therapeutic applications. To date, various metallodrugs display interesting biological activities for chemotherapy. In this field, cisplatin was the first inorganic compound with high relevance in cancer treatment. This compound was a leader agent in clinical use. Toxicity and resistance problems trigger the development of other platinum drugs with better clinical perspective and also raise the scientific interest for the putative antitumor properties of V, Ru and Cu compounds. Several scientific articles show that complexes of these metals are the new metal-based drugs used in the treatment of several cancers, such us, lung, colon, breast, bladder, etc. In this review we recapitulate current information and new advances on antitumor in vitro effects of several organic and inorganic compounds derived from copper, ruthenium and vanadium. These metal derived compounds targeting DNA or cell proteins involved in cell signaling pathways related to cancer. The mechanisms of cell death of these metallodrugs have also been comprehensibly reviewed. The knowledge of these mechanisms of death and the relationship between chemical structure and biological activity may be useful for the design of new metal-based drugs with promising pharmacologic applications as anticancer agents.

Abstract:Flavonoids, a ubiquitous group of naturally occurring polyphenolic compounds, have recently gained importance as anticancer agents. Unfortunately, due to low solubility, absorption, and rapid metabolism of dietary flavonoids, their anticancer potential is not sufficient. Nanocarriers can improve the bioavailability of flavonoids. In this review we aimed to evaluate studies on the anticancer activity of flavonoid nanoparticles. A review of English language articles published until 30 June 2020 was conducted, using PubMed (including MEDLINE), CINAHL Plus, Cochrane, and Web of Science data. Most studies determining the anticancer properties of flavonoid nanoparticles are preclinical. The potential anticancer activity focuses mainly on MCF-7 breast cancer cells, A549 lung cancer cells, HepG2 liver cancer cells, and melanoma cells. The flavonoid nanoparticles can also support the anti-tumour effect of drugs used in cancer therapy by enhancing the anti-tumour effect or reducing the systemic toxicity of drugs.

Anti-Cancer Agents in Medicinal Chemistry

Abstract:Cancer continues to be one of the major causes of death worldwide. Despite many advances in the understanding of this complex disease, new approaches are needed to improve the efficacy of current therapeutic treatments against aggressive tumors. Natural products are one of the most consistently successful sources of drug leads. In recent decades, research activity into the clinical potential of this class of compounds in cancer has increased. Furthermore, a highly promising field is the use of metals and their complexes in the design and development of metal-based drugs for the treatment of cancer. Metal complexes offer unique opportunities due to their ability to alter pharmacology, improving the efficacy and/or reducing the negative side effects of drug molecules. In addition, transition metals as copper, iron, and manganese, among others, can interact with active sites of enzymes, playing important roles in multiple biological processes. Thus, these complexes not only possess higher activities but also reach their targets more efficiently. This review article highlights recent advances on the emerging and expanding field of metal-based drugs. The emphasis is on new therapeutic strategies consisting of metal complexes with natural product like-compounds as a starting point for the rational design of new antitumor agents.

Abstract:Objective: Thiosemicarbazones are Schiff based ligands of significant biological importance and their biological relevance has been studied for a considerable amount of time period. When the thiosemicarbazones bind with metal ions, they have shown an array of potential anticancer, antimicrobial and antioxidant activities etc. They have also found numerous applications in Analytical Chemistry. This present review summarizes some of the medicinal benefits of thiosemicarbazone based complexes especially with transition metals. Data Sources: The studies cited in the present review were sourced from journals, books and conference proceedings preferentially written and published in English. Literature from the past 15 years or so has been included in the article. Papers indexed in known databases such as PUBMED, SCOPUS, INDEX COPERNICUS, CHEMICAL AND BIOLOGICAL ABSTRACTS, MEDLINE, EMBASE, EBSCO, DOAJ, and THOMSON REUTERS have been reviewed and included. Summary of the contents of the article: The study unravels the mechanistic action of these compounds on in vitro living beings viz. cell culture as well as animal models. An elaborate review of the available literature on thiosemicarbazones has shown that the modifications in the ligand moiety leads to enhancement in its activity and some of the best structural alterations have been cited in the review. Some of the potential future applications and uses of the complexes have also been discussed. Structure optimization of the compounds may result in path breaking finding of potential anticancer and antimicrobial drugs. Conclusion: Latest advances in medicinal inorganic chemistry have given considerable importance to the development of metal based drugs with thiosemicabazones. The presence of metal ion in the drug moiety usually mitigates the ill effects of the compounds, this may lead to the production of new metal based drugs. The key factors identified for their action has been the inhibition of RR, topo II and the production of ROS, but identification of other probable targets need to be explored.

Abstract:With the progression in the field of bioinorganic chemistry, the role of transition metal complexes as the most widely used therapeutics is becoming a more and more attractive research area. The complexes of transition metals possess a great variety of attractive pharmacological properties, including anticancer, anti-inflammatory, antioxidant, anti-infective, etc., activities. Transition metal complexes have proven to be potential alternatives to biologically active organic compounds, especially as antitumor agents. The performance of metal coordination compounds in living systems is anticipated to differ generally from the action of non-metal-containing drugs and may offer unique diagnostic and/or therapeutic opportunities. In this review, the rapid development and application of metallocenes and metal complexes of elements from Groups 4 to 7 in cancer diagnostics and therapy have been summarized. Most of the heavy metals discussed in the current review are newly discovered metals. That is why the use of their metal-based compounds has attracted a lot of attention concerning their organometallic and coordination chemistry. All of this imposes more systematic studies on their biological activity, biocompatibility, and toxicity and presupposes further investigations.

Diet-derived Flavonoids: Bridging Epidemiological Chemoprevention and Preclinical Anti-tumor Mechanisms in Clinical Oncology

Abstract:Flavonoids are an abundantly consumed group of dietary polyphenols present in fruits, vegetables, teas, herbs and other plant-derived foods composed of a diphenylpropane (C6-C3-C6) ring structure, allowing subclassification into flavonols, flavones, flavan-3-ols, anthocyanins and isoflavones based on substitutions on the heterocyclic C ring. Multiple case-control studies and prospective cohort analyses reveal higher intake of certain flavonoid subgroups associated with reduced risk of various epithelial cancers like lung, breast, pancreatic, oral and liver. In vitro studies across diverse human cancer cell lines and in vivo, animal models demonstrate anticancer effects of select flavonoids either directly or in synergy with chemotherapy by targeting hallmark capabilities that enable tumours including resisting cell death, sustaining proliferation, inducing angiogenesis, activating invasion and metastasis. The well-explored anticancer mechanisms range from direct antioxidant activity, quenching free radicals and bolstering endogenous defenses; to anti-inflammatory signalling via NF-κB and cytokine modulation; epigenetic alterations by chromatin remodeling; to direct regulation of cell cycle controllers (CDKs, cyclins) and apoptotic mediators (caspases, Bcl-2. Early human trials mostly indicate the safe use of certain flavonoids and subclasses at tested doses however, progression to therapeutic benefit faces challenges like suboptimal systemic availability upon metabolism, unclear metabolite activities and study design limitations regarding delivery methods, combination treatments and clinical priority. In essence, dietary flavonoids exhibit pleiotropic pharmacological strengths against cancer progression warranting expanded translational research and human trials to develop formulations/delivery systems and validate targeted clinical integration, especially alongside chemotherapy regimens.

Abstract:Cancer remains a global health problem that requires constant research for the development of new treatment strategies. Flavonoids, a diverse group of naturally occurring polyphenolic compounds abundant in fruits, vegetables, and other plant sources, have received considerable attention for their potential anticancer properties. This review aimed to provide a comprehensive overview of the current scientific literature on five specific natural flavonoids, namely quercetin, Epigallocatechin Gallate (EGCG), kaempferol, apigenin, and curcumin that have been widely reported in numerous carcinomas and evaluate their effectiveness and mechanisms in fighting different types of cancer. Known for its antioxidant and anti-inflammatory properties, quercetin has shown promise in inhibiting cancer cells and modulating key signaling pathways. EGCG, a prominent catechin found in green tea, has been extensively studied for its ability to induce apoptosis and inhibit angiogenesis, highlighting its potential as an anticancer agent. Kaempferol has antioxidant and anti-inflammatory effects and has shown anticancer potential by modulating cellular processes involved in tumor development. Apigenin, abundant in parsley and chamomile, has been shown to exert anticancer properties by interrupting the cell cycle and inducing apoptosis in cancer cells. Curcumin has shown several anticancer effects, including inhibiting cell proliferation, inducing apoptosis, and modulating inflammatory pathways. Despite these promising findings, it is essential to recognize the complexity of cancer biology and the need for further research to clarify the precise mechanisms of action of these natural flavonoids and optimize their therapeutic applications. Furthermore, understanding flavonoids' potential synergy and interactions with traditional cancer therapies is paramount for developing effective combinatorial strategies. This review thus aimed to summarize the current knowledge on these natural flavonoids and provide insight into their potential role as an adjunctive or stand-alone therapy in the fight against breast, prostate, colon, lung, skin, ovarian, liver, and pancreatic cancer.

Abstract:There is a great deal of research interest in the design of alternative metallodrugs to Pt(II)‐derivatives for cancer treatment. The low solubility of such drugs in biological mediums leading to poor bioavailability is the major hurdle of several metal‐based anticancer agents. These issues have recently been addressed by designing bio‐active ligands based on metal‐containing anticancer agents. Conjugating with bioactive ligands has significantly improved the bioavailability of the metallodrugs and their cancer cell targeting ability. One such naturally available bioactive ligand is curcumin. Until recently, several curcumin‐based anticancer metallodrugs have been developed and successfully demonstrated for their anticancer studies. In this article, we aim to highlight, the synthesis, structure, and anticancer properties of various Zn(II)‐curcumin‐based coordination complexes. The effect of introducing different functional groups, targeting ligands, and photo‐active ligands on the anticancer potential of such complexes has been mentioned in detail. The current status and future perspective on curcumin‐based metallodrugs for cancer treatment have also been stated.

Abstract:Flavonoids and their derivatives are polyphenolic secondary metabolites with an extensive spectrum of pharmacological activities, including antioxidants, antitumor, anti-inflammatory, and antiviral activities. These flavonoids can also act as chemopreventive agents by their interaction with different proteins and can play a vital role in chemotherapy, suggesting a positive correlation between a lower risk of cancer and a flavonoid-rich diet. These agents interfere with the main hallmarks of cancer by various individual mechanisms, such as inhibition of cell growth and proliferation by arresting the cell cycle, induction of apoptosis and differentiation, or a combination of these mechanisms. This review is an effort to highlight the therapeutic potential of natural and synthetic flavonoids as anticancer agents in leukemia treatment with respect to the structure-activity relationship (SAR) and their molecular mechanisms. Induction of cell death mechanisms, production of reactive oxygen species, and drug resistance mechanisms, including p-glycoprotein efflux, are among the best-described effects triggered by the flavonoid polyphenol family.

Abstract:This article provides an overview of the development, structure and activity of various metal complexes with anti-cancer activity. Chemical researchers continue to work on the development and synthesis of new molecules that could act as anti-tumor drugs to achieve more favorable therapies. It is therefore important to have information about the various chemotherapeutic substances and their mode of action. This review focuses on metallodrugs that contain a metal as a key structural fragment, with cisplatin paving the way for their chemotherapeutic application. The text also looks at ruthenium complexes, including the therapeutic applications of phosphorescent ruthenium(II) complexes, emphasizing their dual role in therapy and diagnostics. In addition, the antitumor activities of titanium and gold derivatives, their side effects, and ongoing research to improve their efficacy and reduce adverse effects are discussed. Metallization of host defense peptides (HDPs) with various metal ions is also highlighted as a strategy that significantly enhances their anticancer activity by broadening their mechanisms of action.

Anti-Cancer Agents in Medicinal Chemistry

Abstract:Over the last few decades, an alarming rise in the percentage of individuals with cancer and those with multi-resistant illnesses has forced researchers to explore possibilities for novel therapeutic approaches. Numerous medications currently exist to treat various disorders, and the development of small molecules as anticancer agents has considerable potential. However, the widespread prevalence of resistance to multiple drugs in cancer indicates that it is necessary to discover novel and promising compounds with ideal characteristics that could overcome the multidrug resistance issue. The utilisation of metallo-drugs has served as a productive anticancer chemotherapeutic method, and this approach may be implemented for combating multi-resistant tumours more successfully. Schiff bases have been receiving a lot of attention as a group of compounds due to their adaptable metal chelating abilities, innate biologic properties, and versatility to tweak the structure to optimise it for a specific biological purpose. The biological relevance of Schiff base and related complexes, notably their anticancer effects, has increased in their popularity as bio-inorganic chemistry has progressed. As a result of learning about Schiff bases antitumor efficacy against multiple cancer cell lines and their complexes, researchers are motivated to develop novel, side-effect-free anticancer treatments. According to study reports from the past ten years, we are still seeking a powerful anticancer contender. This study highlights the potential of Schiff bases, a broad class of chemical molecules, as potent anticancer agents. In combination with other anticancer strategies, they enhance the efficacy of treatment by elevating the cytotoxicity of chemotherapy, surmounting drug resistance, and promoting targeted therapy. Schiff bases also cause cancer cell DNA repair, improve immunotherapy, prevent angiogenesis, cause apoptosis, and lessen the side effects of chemotherapy. The present review explores the development of potential Schiff base and their d and f block metal complexes as anticancer agents against various cancer cell lines.
