Observations on genetic differentiation in Peromyscus.

Abstract:Macrogeographic patterns of allele frequencies revealed considerable variation among populations of Peromyscus polionotus (Selander et al., 1971), P. leucopus (Browne, 1977; Price and Kennedy, 1980), P. maniculatus (Avise et al., 1979) and P. californicus (Smith, 1979). Reported patterns in P. maniculatus and P. leucopus were similar although samples were from different regions of North America. We compare herein levels of genetic differentiation among populations of P. maniculatus and P. leucopus from the same geographic region, the northeastern United States, using standardized variance of allele frequencies (FsT; Wright, 1965). We also compare FsT for these two sets of samples to values of FsT for these species from elsewhere in their ranges and for other mice. Eighty-four P. maniculatus and 154 P. leucopus were live trapped from May to October 1976 at 13 deciduous forest sites from Vermont to West Virginia. These sites in order of descending latitude were: (1) 3 miles ESE Owls Head, Franklin Co., New York; (2) 4 W Chester, Rutland Co., Vermont; (3) 4 W Stratton, Windham Co., Vermont; (4) 2 SE Marilla, Erie Co., New York; (5) 1 E Cobleskill, Schoharie Co., New York; (6) 2 E Richmondville, Schoharie Co., New York; (7) 2 SW Dryden, Tompkins Co., New York; (8) 1 S Slaterville Springs, Tompkins Co., New York; (9) 2 E West Windsor, Broome Co., New York; (10) 4 S Binghamton, Broome Co., New York; (11) 1 SW St. Joseph, Susquehanna Co., Pennsylvania; (12) 2 W Starlight, Wayne Co., Pennsylvania; and (13) 5 NE Morgantown, Monongalia Co., West Virginia. Preparation of liver and blood samples, electrophoretic techniques and staining procedures were essentially the same as described by Selander et al. (1971) with the following exceptions: liver tissues were prepared by chopping with scissors and ESTERASE-I (ES-1) was stained with naphthol-AS-D acetate as the substrate (Van Deusen and Kaufman, 1978).
