Abstract:Microplastics are ubiquitous in the world's oceans and pose serious environmental concerns, including their ingestion and the release of potentially toxic mixtures of intrinsic and extrinsic chemical compounds (i.e. leachates; MPLs). Mussels, as key intertidal bioengineers and filter-feeders are particularly susceptible to both exposure pathways. While the effects of microplastic ingestion have been widely investigated, research on the impacts of MPLs has only recently begun. This study examined the influence of MPLs derived from beached pellets collected in two separate regions, namely France and Portugal, on the respiration rates of two key ecosystem engineers, Mytilus edulis and Mytilus galloprovincialis . Possibly due to distinct mixtures of leached chemicals, unlike Portuguese-MPLs, exposure to French-MPLs significantly decreased the respiration rate of both mussel species. This research provides new insights into the physiological impacts of MPLs on bioengineer species, highlighting the importance of MP source and potential cascading effects at the ecosystem level. While we reported significant effects on mussel respiration after acute MPL exposure, future research should investigate long-term impacts and potential detoxification mechanisms to clarify the effects of MPs on mussel physiological performance and their potential consequences on specie fitness.

Abstract:Self-organized spatial patterns are increasingly recognized for their contribution to ecosystem functioning. They can improve the ecosystem's ability to respond to perturbation and thus increase its resilience to environmental stress. Plastic pollution has now emerged as major threat to aquatic and terrestrial biota. Under laboratory conditions, we tested whether plastic leachates from pellets collected in the intertidal can impair small-scale, spatial self-organization and byssal threads production of intertidal mussels and whether the effect varied depending on where the pellets come from. Specifically, leachates originating from plastic pellets collected from relatively pristine and polluted areas respectively impaired and inhibited the ability of mussels to self-organize at small-scale and to produce byssal threads compared to control conditions (i.e., seawater without leaching solution). Limitations to natural self-organizing processes and threads formation may translate to a declined capacity of natural ecosystems to avoid tipping points and to a reduced restoration success of disturbed ecosystems.

Abstract:Microplastics (MPs) disturb the normal activity of aquatic organisms at different levels, causing physiological stress and altering feeding, growth, and reproduction. Alterations of epigenetic patterns due to exposure to MPs have scarcely been studied in invertebrates. In this study, Mytilus galloprovincialis mussels (N = 61) were intermittently exposed to different concentrations of pure polystyrene microbeads for three weeks. The concentrations used in this research were similar to those currently found in certain polluted environments (E1), as well as higher doses to which mussels could be further exposed (E2 and E3). After exposure period, the global methylation patterns were investigated using Amplified Fragment Length Polymorphism (AFLPs). Significantly lower methylation was found in exposed groups compared to the control group. The level of hypomethylation increased with the concentration of microbeads. Similar results were found from field samples inhabiting two sites differentially MPs-polluted. The implications of this discovery were analysed and discussed, noting the already known effects of MPs on metabolism and cell division. Further studies on this and other sentinel organisms are recommended to understand the response of the aquatic species to the currently increasing MPs pollution.

Abstract:The presence and persistence of microplastics (MPs) in diverse aquatic environments are of global concern. Microplastics can impact marine organisms via direct physical interaction and the release of potentially harmful chemical additives incorporated into the plastic. These chemicals are physically bound to the plastic matrix and can leach out. The hazards associated with chemical additives to exposed organisms is not well characterized. We investigated the hazards of plastic additives leaching from plastic. We used the common plasticizer dibutyl phthalate (DBP) as a chemical additive proxy and the New Zealand green‐lipped mussel (Perna canaliculus) as a model. We used early‐adult P. canaliculus exposed to combinations of virgin and DBP‐spiked polyvinyl chloride (PVC), MPs, and DBP alone for 7 days. Whole transcriptome sequencing (RNA‐seq) was conducted to assess whether leaching of DBP from MPs poses a hazard. The differences between groups were evaluated using pairwise permutational multivariate analysis of variance (PERMANOVA), and all treatments were significantly different from controls. In addition, a significant difference was seen between DBP and PVC MP treatment. Transcriptome analysis revealed that mussels exposed to DBP alone had the most differentially expressed genes (914), followed by PVC MP + DBP (448), and PVC MP (250). Gene ontology functional analysis revealed that the most enriched pathway types were in cellular metabolism, immune response, and endocrine disruption. Microplastic treatments enriched numerous pathways related to cellular metabolism and immune response. The combined exposure of PVC MP + DBP appears to cause combined effects, suggesting that DBP is bioavailable to the exposed mussels in the PVC MP + DBP treatment. Metabolic responses of the marine mussel Mytilus galloprovincialis after exposure to microplastics of different shapes and sizes

Abstract:Microplastics (MP) are ubiquitous pollutants with diverse shapes, sizes, and characteristics that pose critical risks to marine organisms and the environment. In this study, we used the Mediterranean mussel Mytilus galloprovincialis as a marine benthic organism model to investigate the metabolic consequences of exposure to different polyethylene terephthalate MP sizes and shapes: round (27-32 μm), small fibers (200-400 μm), large fibers (3000 μm), small fragments (20 μm), medium fragments (45-75 μm), and large fragments (>150 μm). After exposure to high concentrations (100 mg L-1) of MP for 14 days, round and small fiber-type MP were highly accumulated in mussels. Metabolomic analysis revealed that exposure to round and small fiber-type MP induced significant changes in 150 metabolites. Partial least squares-discriminate analysis (PLS-DA) showed that the round and small fiber MP treatment groups displayed similar cluster patterns that differed from those of the control group. In addition, only 22 annotated metabolites related to histidine, valine, leucine, and isoleucine degradation/biosynthesis and vitamin B6 and aminoacyl-tRNA biosynthesis were significantly affected by round or small fiber-type MP. Among the histidine metabolites, round and small fiber-type MP upregulated the levels of L-histidine, L-glutamate, carnosine, imidazole-4-acetaldehyde, 4-imidazolone-5-propanoate, and methylimidazole acetaldehyde and downregulated methylimidazole acetic acid and N-formimino-L-glutamate. These results suggest novel insights into the potential pathways through which MP of specific sizes and shapes affect metabolic processes in mussels.

Abstract:Microplastic contamination is a growing marine environmental issue with possible consequences for seafood safety. Filter feeders are the target species for microplastic (MPs) pollution because they filter large quantities of seawater to feed. In the present study, an experimental contamination of Mytilus galloprovincialis was conducted using a mixture of the main types of MPs usually present in the seawater column (53% filaments, 30% fragments, 3% granules) in order to test the purification process as a potential method for removing these contaminants from bivalves intended for human consumption. A set of molecular biomarkers was also evaluated in order to detect any variations in the expression levels of some genes associated with biotransformation and detoxification, DNA repair, cellular response, and the immune system. Abstract:The ocean contamination caused by micro- and nano-sized plastics is a matter of increasing concern regarding their potential effects on marine organisms. This study compared the effects of a 21-day exposure to 1.5, 15, and 150 ng/L of polystyrene microplastics (PS-MP, 3-μm) and nanoplastics (PS-NP, 50-nm) on a suite of biomarkers measured in the Mediterranean mussel Mytilus galloprovincialis. Endpoints encompassed immunological/lysosomal responses, oxidative stress/detoxification parameters, and neurotoxicological markers. Compared to PS-MP, PS-NP induced higher effects on lysosomal parameters of general stress. Exposures to both particle sizes increased lipid peroxidation and catalase activity in gills; PS-NP elicited greater effects on the phase-II metabolism enzyme glutathione S-transferase and on lysozyme activity, while only PS-MP inhibited the hemocyte phagocytosis, suggesting a major role of PS particle size in modulating immunological/detoxification pathways. A decreased acetylcholinesterase activity was induced by PS-NP, indicating their potential to impair neurological functions in mussels. Biomarker data integration in the Mussel Expert System identified an overall greater health status alteration in mussels exposed to PS-NP compared to PS-MP. This study shows that increasing concentrations of nanoplastics may induce higher effects than microplastics on the mussel’s lysosomal, metabolic, and neurological functions, eventually resulting in a greater impact on their overall fitness.

Exposure to Polymethylmethacrylate Microplastics Induces a Particle Size-Dependent Immune Response in Mediterranean Mussel Mytilus galloprovincialis

Abstract:The widespread occurrence of plastic has become a significant problem in the natural environment and may give rise to a range of deleterious impacts in biota, particularly where plastic particles overlap in size with that of the particles that are naturally ingested by filter-feeders. In this context, the effects of two particle sizes (10 μm and 50 μm) of polymethylmethacrylate (PMMA) on ecologically and commercially significant mussel Mytilus galloprovincialis have been investigated. Mussel health status was evaluated by measuring the condition index and stress-on-stress test during and after 72 h exposure to PMMA microparticles in the 0.1–10 mg L−1 concentration range. The decreased condition and fitness indices in the exposed mussels pointed to significant physiological effects at the entire organism level. The 10 μm and 50 μm PMMA particles were noted to rapidly increase the total haemocytes count in haemolymph, and significantly reduce cell viability at higher concentrations of both particle sizes. The results also indicated a significant increase in levels of vacuolised haemocytes as a result of PMMA exposure. While both microparticles were detected in the haemolymph, only the 10 μm PMMA was observed in the gill tissue and digestive gland by histological cryosections, indicating their rapid uptake, transport, and accumulation in tissue. Lack of accumulation of 50 μm microparticles in tissue may be related to a combination of chemical identity and size considerations, enabling more efficient depuration of microparticles in pseudofaeces. The PMMA particles did not induce significant changes in activity of a range of enzymes involved in neurotransmission and responses to oxidative stress.

Abstract:The rise of plastic production has triggered a surge in plastic waste, overwhelming marine ecosystems with microplastics. The effects of climate change, notably changing salinity, have shaped the dynamics of coastal lagoons. Thus, understanding the combined impact of these phenomena on marine organisms becomes increasingly crucial. To address these knowledge gaps, we investigated for the first time the interactive effects of environmental microplastics (EMPs) and increased salinity on the early development of Mytilus galloprovincialis larvae. Morphological assessments using the larval embryotoxicity test revealed larval anomalies and developmental arrests induced by EMPs and increased salinity. Transcriptomic analyses targeting 12 genes involved in oxidative stress, apoptosis, DNA repair, shell formation, and stress proteins were conducted on D-larvae uncovered the potential effects of EMPs on shell biomineralization, highlighting the role of Histidine Rich Glycoproteine (HRG) and tubulin as crucial adaptive mechanisms in Mytilus sp. in response to environmental shifts. Furthermore, we explored oxidative stress and neurotoxicity using biochemical assays. Our findings revealed a potential interaction between EMPs and increased salinity, impacting multiple physiological processes in mussel larvae. Our data contribute to understanding the cumulative effects of emerging anthropogenic pollutants and environmental stressors, emphasizing the need for a holistic approach to assessing their impact on marine ecosystems.

Abstract:Plastic pollution and invasive species are recognised as pervasive threats to marine biodiversity. However, despite the extensive on-going research on microplastics’ effects in the biota, knowledge on their combination with additional stressors is still limited. This study investigates the effects of polyamide microplastics (PA-MPs, 1 mg/L), alone and in combination with the toxic exudate from the invasive red seaweed Asparagopsis armata (2%), after a 96 h exposure, in the mussel Mytilus galloprovincialis. Biochemical responses associated with oxidative stress and damage, neurotoxicity, and energy metabolism were evaluated in different tissues (gills, digestive gland, and muscle). Byssus production and PA-MP accumulation were also assessed. Results demonstrated that PA-MPs accumulated the most in the digestive gland of mussels under PA-MP and exudate co-exposure. Furthermore, the combination of stressors also resulted in oxidative damage at the protein level in the gills as well as in a significant reduction in byssus production. Metabolic capacity increased in both PA-MP treatments, consequently affecting the energy balance in mussels under combined stress. Overall, results show a potential increase of PA-MPs toxicity in the presence of A. armata exudate, highlighting the importance of assessing the impact of microplastics in realistic scenarios, specifically in combination with co-occurring stressors, such as invasive species.

Abstract:Electrocardiograms (ECGs) are widely used by clinicians to identify the functional status of the heart. Thus, there is considerable interest in automated systems for real-time monitoring of arrhythmia. However, intra- and inter-patient variability as well as the computational limits of real-time monitoring poses significant challenges for practical implementations. The former requires that the classification model be adjusted continuously, and the latter requires a reduction in the number and types of ECG features, and thus, the computational burden, necessary to classify different arrhythmias. We propose the use of adaptive learning to automatically train the classifier on up-to-date ECG data, and employ adaptive feature selection to define unique feature subsets pertinent to different types of arrhythmia. Experimental results show that this hybrid technique outperforms conventional approaches and is therefore a promising new intelligent diagnostic tool.

Abstract:Microplastic (MP) contamination in the aquatic environment is a cause of concern worldwide since MP can be taken up by different organisms, altering different biological functions. In particular, evidence is accumulating that MP can affect the relationship between the host and its associated microbial communities (the microbiome), with potentially negative health consequences. Synthetic microfibers (MFs) represent one of the main MPs in the marine environment, which can be accumulated by filter-feeding invertebrates, such as bivalves, with consequent negative effects and transfer through the food chain. In the mussel Mytilus galloprovincialis, polyethylene terephthalate (PET) MFs, with a size distribution resembling that of an MF released from textile washing, have been previously shown to induce multiple stress responses. In this work, in the same experimental conditions, the effects of exposure to PET-MF (96 h, 10, and 100 μg/L) on mussel hemolymph microbiome were evaluated by 16S rRNA gene amplification and sequencing. The results show that PET-MF affects the composition of bacterial communities at the phylum, family and genus level, with stronger effects at the lowest concentration tested. The relationship between MF-induced changes in hemolymph microbial communities and responses observed at the whole organism level are discussed.
Quantifying Effects and Ingestion of Several Pristine Microplastics in Two Early Life Stages of Freshwater Mussels

Abstract:Microplastics have been found in freshwater systems, and in turn have been detected in freshwater bivalves. However, there is limited research that defines the toxicity of bicroplastics to native freshwater bivalves that have long been imperiled in North America. Our objective was to determine whether a suite of pristine microplastics has an adverse effect on two early life stages of unionid freshwater mussels. Glochidia of Lampsilis fasciola (a Canadian species at risk) and Lampsilis siliquoidea (widespread across Canada) were individually exposed to spheres of polystyrene (6 and 90 μm), polyethylene (28, 90, and 1000 μm), and cellulose acetate (1000 μm), as well as fibers of polyethylene terephthalate (60 μm). After 24 h, there was no significant decrease in glochidia viability in either species. Juvenile L. siliquoidea mussels were also exposed to spheres of polystyrene (6 and 90 μm) and polyethylene (28 μm), and fibers of polyethylene terephthalate (60 μm) in individual 28‐day subchronic tests followed by a 7‐day depuration period. Burial was assessed weekly, and ingestion of each microplastic was compared in nondepurated and depurated mussels. There was no sustained effect on juvenile burial with any microplastic tested. Ingestion of microplastics was concentration dependent, and depuration occurred for all particles and size ranges tested. The results suggest that pristine microplastics were not acutely toxic to the early life stages of these freshwater mussels, but that the energetic costs associated with particle uptake and depuration, which were not measured in our study, may have an impact on fitness that warrants further investigation. Abstract:The ubiquitous occurrence of microplastics (MPs) in the marine environment is raising concern for interactions with marine organisms. These particles efficiently adsorb persistent organic pollutants from surrounding environment and, due to the small size, they are easily available for ingestion at all trophic levels. Once ingested, MPs can induce mechanical damage, sub-lethal effects, and various cellular responses, further modulated by possible release of adsorbed chemicals or additives. In this study, ecotoxicological effects of MPs and their interactions with benzo(a)pyrene (BaP), chosen as a model compound for polycyclic aromatic hydrocarbons (PAHs) were investigated in Mediterranean mussels, Mytilus galloprovincialis. Organisms were exposed for 4 weeks to 10 mg/L of low-density polyethylene (LDPE) microparticles (2.34 * 107 particles/L, size range 20–25 μm), both virgin and pre-contaminated with BaP (15 μg/g). Organisms were also exposed for comparison to BaP dosed alone at 150 ng/L, corresponding to the amount adsorbed on microplastics. Tissue localization of microplastics was histologically evaluated; chemical analyses and a wide battery of biomarkers covering molecular, biochemical and cellular levels allowed to evaluate BaP bioaccumulation, alterations of immune system, antioxidant defenses, onset of oxidative stress, peroxisomal proliferation, genotoxicity, and neurotoxicity. Obtained data were elaborated within a quantitative weight of evidence (WOE) model which, using weighted criteria, provided synthetic hazard indices, for both chemical and cellular results, before their integration in a combined index. Microplastics were localized in hemolymph, gills, and especially digestive tissues where a potential transfer of BaP from MPs was also observed. Significant alterations were measured on the immune system, while more limited effects occurred on the oxidative status, neurotoxicity, and genotoxicity, with a different susceptibility of analyzed pathways, depending on tissue, time, and typology of exposure. Molecular analyses confirmed the general lack of significant transcriptional variations of antioxidant and stress genes. The overall results suggest that microplastics induce a slight cellular toxicity under short-term (28 days) exposure conditions. However, modulation of immune responses, along with bioaccumulation of BaP, pose the still unexplored risk that these particles, under conditions of more chronic exposure (months to years) or interacting with other stressors, may provoke long-term, subtle effects on organisms' health status.

In this study, the response of multiple biomarkers (i.e., total antioxidant capacity, the activity of acetylcholine, ion content and transport enzyme, metabolic enzymes, and lipids content) in mussels Mytilus galloprovincialis exposed to polystyrene nanoplastics (PS-NPs) for 7 days were evaluated. Significant inductions of total antioxidant capacity (T-AOC) and inhibition of acetylcholine (AChE) activity were detected after 7 days of PS-NPs exposure. PS-NPs also triggered significant alteration in ion content (Na + and K + ) and suppressed the activities of the ion transport enzyme (Na + /K + -ATPase). Moreover, we found the activity of metabolic enzymes (succinate dehydrogenase and pyruvate kinase) and lipids content (triacylglycerol and cholesterol) were significantly altered, suggesting the interference of PS-NPs on energy metabolism and lipid metabolism. This investigation provides substantial information to understand the physical responses of invertebrate gills to PS-NPs exposure. Given the crucial ecological roles of invertebrates, the presence of PS-NPs in the marine environment may have far-reaching impacts on population abundance, biodiversity, and stability of the marine ecosystem.

Abstract:Microplastic continues to be an environmental concern, especially for filter feeding bivalves known to ingest these particles. It is important to understand the effects of microplastic particles on the physiological performance of these bivalves and many studies have investigated their impact on various physiological processes. This study investigated the effects of microplastic (10 μm) on digestive enzyme (amylase) activity of Mytilus galloprovincialis at 55,000 and 110,000 microplastic particles/L under laboratory conditions. Additionally, our study measured the expression of an isoform of Hsp70 in the gills to assess whether or not these particles may cause protein denaturation. Results revealed that this regime negatively affect the ability of M . galloprovincialis to digest starch under high food conditions but not low food conditions. Exposure to extreme levels of microplastic raised amylase activity. Furthermore, Hsp70 transcript abundance was not elevated in treatment mussels. These results show that mussels may be resilient to current microplastic pollution levels in nature.

Abstract:The oxidation of benzaldehyde and p -methylbenzaldehyde solubilized in various polyoxyethylene glycol ether nonionic surfactants has been measured manometrically. Evidence is presented to show that rates of oxidation are related to the degree of saturation of the dispersion where this is expressed as the ratio of the amount of aldehyde present to its solubility in the surfactant solution. The effect on oxidation of variation in chain length of the hydrophilic and hydrocarbon parts of the surfactant molecule is discussed.

Abstract:Growing micro- and nano-plastic (MNPs) pollution in the environment poses a threat to marine animals. Due to their excellent filtration capacity, bivalves can easily ingest MNPs, which could be translocated to open circulation system with potential risks. In the present study, the accumulation and elimination of MNPs (200 nm and 1 µm) in the mussel hemolymph serum and hemocytes were firstly quantified, and the differential sensitiveresponses of two subpopulations of hemocytes were then explored by in vivo exposure under environmentally relevant concentration of MNPs (200 µg/L). We demonstrated that MNPs were readily translocated into hemolymph serum, but were immediately followed by efficient internalization by hemocytes. Remarkably, concentrations of MNPs in hemolymph were only 0.63 and 0.39 times lower than the ambient exposure concentration. Granulocytes displayed a much higher potential of accumulating MNPs than the agranulocytes. MPs were more readily internalized by granulocytes, with their estimated maximum bioaccumulation factor (BCF) of 0.29 L/g. Due to the primary function of phagocytic encapsulation of MNPs by granulocytes, lysosome features especially the decline of subsequent lysosome membrane potential could be a potential sensitive biomarker in response to MNPs exposure. Our results provided insights on the bioaccumulation of MNPs at the cellular levels in marine bivalves.

Abstract:<p>Plastic particles cause toxic effects on marine organisms but whether food particles can affect the toxic effects of plastic particles on filter feeding animals remains unknown. To evaluate the intake and physiological effects of different size particles and their exposure ways, the thick shell mussels <em>Mytilus coruscus</em> were exposed to polystyrene (PS) nanoplastics (NPs, 70 nm) and microplastics (MPs, 10 µm) respectively for two weeks by mixing NPs/MPs with microalgae or exposed to MNPs individually after feeding. Intake of particles and their physiological effects including energy budget, digestive enzymes and oxidative responses were assessed after exposure. Results indicated food presence mediate the effects while MPs decrease the energy budget and increase the catalase activity and malondialdehyde levels. Moreover, exposure way significantly affected energy budget and size of particle had a significant impact on enzyme activities. Our results showed MPs induce more significant effects than NPs on mussels, emphasized the importance of particle exposure way and suggested that mixture exposure with microalgae alleviate the influences on mussels caused by plastic particles alone. This study emphasized that we need to take the food particles into account for evaluating the toxic effects of plastic particles on filter feeding animals in the natural environment.</p>
