Abstract:In recent years, extensive research has focused on cannabidiol (CBD), a well-studied non-psychoactive component of the plant-derived cannabinoids. CBD has shown significant therapeutic potential for treating various diseases and disorders, including antioxidants and anti-inflammatory effects. Due to the promising therapeutic effect of CBD in a wide variety of diseases, synthetic derivatization of this compound has attracted the attention of drug discovery in both industry and academia. In the current research, we focused on the derivatization of CBD by introducing Schiff base moieties, particularly (thio)-semicarbazide and aminoguanidine motifs, at the 3-position of the olivetolic ring. We have designed, synthesized, and characterized new derivatives based on CBD's framework, specifically aminoguanylhydrazone- and (thio)-semicarbazones-CBD-aldehyde compounds. Their antioxidant potential was assessed using FRAP and DPPH assays, alongside an evaluation of their effect on LDL oxidation induced by Cu2+ and AAPH. Our findings suggest that incorporating the thiosemicarbazide motif into the CBD framework produces a potent antioxidant, warranting further investigation.
An Improved Iodine-Catalyzed Aromatization Reaction and Its Application in the Synthesis of a Key Intermediate of Cannabidiol

Abstract:In this study, the development of an improved process for the synthesis of a key cannabidiol intermediate, methyl olivetolate, is described. The process involves an improvement of the iodine-catalyzed aromatization of cyclohexanone using potassium persulfate as an oxidant. This approach enabled for the efficient synthesis of methyl olivetolate with a 90% yield and 99.84% HPLC purity on a 5 kg scale. Additionally, a total of 19 cyclohexanone substrates afforded higher yields (71–92%) of m -diphenol compounds compared to the established methods. Abstract:Cannabidiol (CBD) is garnering increasing interest due to its significant biological activity. This natural compound is one of the major cannabinoids in Cannabis sativa L. In this work, we describe the encapsulation of CBD in solid and hollow pH‐sensitive poly(4‐vinylpyridine) (solid@p4VP and hollow@p4VP) nanoparticles, and temperature‐sensitive poly(N‐isopropylacrylamide) (solid@pNIPAM and hollow@pNIPAM) nanoparticles for transport and release CBD in a controlled manner. The CBD loading into these smart polymeric systems was effective and their release profiles, solubility and resistance to stomach and intestinal conditions were evaluated, showing satisfactory properties and improved bioavailability with respect to free CBD. Finally, the A549 human lung cancer cell line was used as lung adenocarcinoma epithelial cellular model to carry out preliminary assays of the in vitro activity of the vehiculized CBD. For all these studies, synthetic CBD was employed, for which a new efficient and scalable synthesis of cannabinoids has been developed.

Abstract:The comprehensive anti-cancer activities of cannabinoid acids and non-cannabinoids are not fully explored. Herein we report simple extraction, faster bioassay-guided fractionation, and HPLC-assisted purification of bioactive secondary metabolite and their identification. Anti-proliferative activity of six cannabinoid acids and eleven non-cannabinoids from the leaves of Cannabis sativa was evaluated. HPLC- assisted purification from the bioactive fractions leads to isolate one new compound Methoxy Canniprene (methoxy isoprenyl bibenzyl), including all six CBDA's eleven non-cannabinoids in a single step, and NMR, HR-ESI-MS studies, and comparison with the literature data authenticated their structures. The crude extract (IC 50 range: 18.0–37.2 μg/mL), Fraction A to D (11.9–222 μg/mL), and purified different cannabinoid and non cannabinoids (IC 50 range: 0.5–100 μM) displayed potent anti-proliferative activities against almost all the tested five human cancer cell lines. Computational analysis based on molecular docking studies and absolute binding free energy calculation have identified targets for various cancer-associated proteins, including Aldehyde dehydrogenase 1 A1, Topoisomerase I, GSK-3 Beta, Protein Kinase C beta II, and Neurotrophic Tyrosine Kinase receptor type 1.

Abstract:Cannabidiol (CBD) is a nonpsychoactive phytocannabinoid that can be found in Cannabis sativa and possesses numerous pharmacological effects. Due to these promising effects, CBD can be used in a wide variety of diseases, for instance cardiovascular diseases. However, CBD, like tetrahydrocannabinol (THC), has low bioavailability, poor water solubility, and a variable pharmacokinetic profile, which hinders its therapeutic use. Chemical derivatization of CBD offers us potential ways to overcome these issues. We prepared three new CBD derivatives substituted on the aromatic ring by Mannich-type reactions, which have not been described so far for the modification of cannabinoids, and studied the protective effect they have on cardiomyocytes exposed to oxidative stress and hypoxia/reoxygenation (H/R) compared to the parent compound. An MTT assay was performed to determine the viability of rat cardiomyocytes treated with test compounds. Trypan blue exclusion and lactate dehydrogenase (LDH) release assays were carried out to study the effect of the new compounds in cells exposed to H2O2 or hypoxia/reoxygenation (H/R). Direct antioxidant activity was evaluated by a total antioxidant capacity (TAC) assay. To study antioxidant protein levels, HO-1, SOD, catalase, and Western blot analysis were carried out. pIC50 (the negative log of the IC50) values were as follows: CBD1: 4.113, CBD2: 3.995, CBD3: 4.190, and CBD: 4.671. The newly synthesized CBD derivatives prevented cell death induced by H/R, especially CBD2. CBD has the largest direct antioxidant activity. The levels of antioxidant proteins were increased differently after pretreatment with synthetic CBD derivatives and CBD. Taken together, our newly synthesized CBD derivatives are able to decrease cytotoxicity during oxidative stress and H/R. The compounds have similar or better effects than CBD on H9c2 cells.

Abstract:In recent years, the interest in cannabidiol (CBD) has increased because of the lack of psychoactive properties. However, CBD has low solubility and bioavailability, variable pharmacokinetics profiles, poor stability, and a pronounced presystemic metabolism. CBD nanoformulations include nanosuspensions, polymeric micelles and nanoparticles, hybrid nanoparticles jelled in cross-linked chitosan, and numerous nanosized lipid formulations, including nanostructured lipid carriers, vesicles, SNEEDS, nanoemulsions, and microemulsions. Nanoformulations have resulted in high CBD solubility, encapsulation efficiency, and stability, and sustained CBD release. Some studies assessed the increased Cmax and AUC and decreased Tmax. A rational evaluation of the studies reported in this review evidences how some of them are very preliminary and should be completed before performing clinical trials. Almost all the developed nanoparticles have simple architectures, are well-known and safe nanocarriers, or are even simple nanosuspensions. In addition, the conventional routes of administration are generally investigated. As a consequence, many of these studies are almost ready for forthcoming clinical translations. Some of the developed nanosystems are very promising for a plethora of therapeutic opportunities because of the versatility in terms of the release, the crossing of physiological barriers, and the number of possible routes of administration.
Isolation, Purification, and Antimicrobial Characterization of Cannabidiolic Acid and Cannabidiol from Cannabis sativa L.

Abstract:The emergence of multi-drug resistant bacteria such as methicillin-resistant Staphylococcus aureus (MRSA) causes a major threat to public health due to its limited therapeutic options. There is an urgent need for the development of new effective antimicrobial agents and alternative strategies that are effective against resistant bacteria. The parallel legalization of cannabis and its products has fueled research into its many therapeutic avenues in many countries around the world. This study aimed at the development of a reliable method for the extraction, purification, characterization, and quantification of cannabidiolic acid (CBDA) and its decarboxylated form cannabidiol (CBD) present in the fiber type Cannabis sativa L. The two compounds were extracted by ethanol, purified on a C18 sep-pack column, and the extracts were analyzed by high performance liquid chromatography coupled with ultraviolet (UV)–vis and ESI-MS (electrospray ionization mass spectrometry) detection. The antimicrobial effect of CBDA and CBD was also evaluated. CBD displayed a substantial inhibitory effect on Gram-positive bacteria with minimal inhibitory concentrations ranging from 1 to 2 μg/mL. Time kill analysis and minimal bactericidal concentration revealed potential bactericidal activity of CBDA and CBD. While cannabinoids showed a significant antimicrobial effect on the Gram-positive S. aureus and Staphylococcus epidermidis, no activity was noticed on Gram-negative Escherichia coli and Pseudomonas aeruginosa. CBDA presented a two-fold lower antimicrobial activity than its decarboxylated form, suggesting that the antimicrobial pharmacophore of the analyzed cannabinoids falls in the ability for permeabilizing the bacterial cell membrane and acting as a detergent-like agent. A synergy test performed on MRSA with CBD and a range of antibiotics did not indicate a synergetic effect, but noteworthy no antagonist influence either. CBD and CBDA manifested low hemolytic activity on human red blood cells. Likewise, the safety of CBD toward human keratinocyte cells presents no toxicity at a concentration of up to seven-fold higher than the antibacterial minimal inhibitory concentration. Similarly, both CBD and CBDA are well tolerated by mammals, including humans, and conserve a safe value limits for blood-contacting drug development. Overall, CBD exhibited a strong antimicrobial effect against Gram-positive strains and could serve as an alternative drug for tackling MRSA.

Abstract:Phenolic acids are attractive plant-derived bioactive substances owing to their wide range of pharmaceutical properties for the prevention of various human diseases as well as their applications in adjuvant therapy. However, phenolic acids exhibit poor water solubility, which limits their bioavailability and applications. In this study, phenolic acids were combined with matrine to form salts. Matrine is an alkaloid from a traditional Chinese herb that also has abundant pharmaceutical features and was employed to enhance the water solubility and pharmaceutical applicability of phenolic acids. Five bioactive salts, i.e., matrinium syringate, matrinium vanillate, matrinium protocatechuate, matrinium caffeinate, and matrinium gallate, were synthesized and characterized. The results show that all the salts have a hydrogen-bonding-stabilized orthorhombic crystal structure and exhibit a synergistic effect between the anions and cations. Compared with their raw materials, the salts display reduced melting points, higher water solubility (several to hundreds of times greater), lower cytotoxicity for normal human cells, as well as better anticancer, antibacterial, antioxidant, and anti-inflammatory capabilities, accompanied by relatively high thermal stability (>140 °C). This indicates that formation of salts by combining phenolic acids and matrine is an effective method for enhancing pharmaceutical applicability, which may be further used to develop new nutrients or drugs.

Abstract:Background: Cannabidiol (CBD) is an approved treatment for childhood epilepsies and a candidate treatment for several other CNS disorders. However, it has poor oral bioavailability. We investigated the effect of a novel lipid formulation on its absorption in humans and on its tissue distribution in mice. Methods: In a double-blind crossover study in fasting healthy volunteers, we compared the pharmacokinetics of a single dose of 1000 mg of CBD in the lipid formulation and in a powder formulation (ClinicalTrials.gov: NCT05032807). In a second study, male CD1 mice were administered CBD in either the lipid formulation or dissolved in water, via oral gavage (n = 1 per timepoint). The tissue distribution of CBD was assessed using matrix-assisted laser desorption/ionization mass spectrometric imaging. Results: Plasma exposure (AUC0–48) of CBD was nine times greater for the lipid formulation than the powder formulation (611.1 ng·h/mL [coefficient of variation {CV%}: 104.6] and 66.8 ng·h/mL [CV%: 50.7], respectively). With the powder formulation, the AUC0–48 was related to the concentration of specific gastrointestinal bacteria and bile acids. These associations were attenuated with the lipid formulation. In the animal study, after treatment with the lipid formulation, measurable concentrations of CBD were identified in all organs. For the aqueous formulation, tissue concentrations of CBD were below the limit of quantification. Conclusions: Administering oral CBD in a lipid formulation was associated with an increase in its gastrointestinal absorption, as well as an attenuation of the relationship between its absorption and features of the gut microbiome.

Abstract:2-Hydroxybenzylamine (2-HOBA) is a drug used to effectively treat oxidative stress. To improve its aqueous solubility and thermal stability, salt screening and synthesis was carried out. The conductor-like screening model for the real solvents model (COSMO-RS) was applied to virtual screening of coformers among 200 commonly used candidates for salification of 2-HOBA. As a result, 40 hit compounds were subjected to experimental liquid-assisted grinding (LAG) with 2-HOBA, then 21 systems were characterized as new solid phases by PXRD. Nine multicomponent single crystals of 2-HOBA with succinic acid, p-aminobenzoic acid, p-nitrobenzoic acid, o-nitrobenzoic acid, p-toluic acid, 2,3-dihydroxybenzoic acid, 3,4-dihydroxybenzoic acid, p-nitrophenol, and 5-hydroxyisophthalic acid were obtained and characterized by single-crystal X-ray diffraction, powder X-ray diffraction, differential scanning calorimetry, and thermogravimetric analysis. All of them were salts and exhibited higher decomposition temperatures compared with pure 2-HOBA. The apparent aqueous solubility of three new salts, i.e., those with succinic acid, p-aminobenzoic acid, and p-nitrophenol were higher than the equilibrium solubility of 2-HOBA. The accelerated stability test indicated that all salts show excellent stability under conditions (40 °C and 75% RH) for 4 weeks. Overall, this work introduced a protocol that combined the virtual screening tool based on the COSMO-RS model and the experimental LAG method to screen new salts for a target compound. The feasibility of this protocol was confirmed in the case of 2-HOBA whose new salts were successfully obtained and represented an improvement for aqueous solubility and thermal stability.

Molecular Interactions Underlying Dissolution Trends in Cannabidiol-Polymer Amorphous Solid Dispersions

Abstract:Cannabidiol (CBD) is viewed as promising therapeutic agent against a variety of health ailments; however, its efficacy is limited by poor aqueous solubility. Amorphous solid dispersions (ASDs) can enhance the solubility of therapeutics by distributing it throughout a polymer matrix. In consideration of ASD formulations with CBD, we investigate the interactions of CBD with various polymers: polyvinylpyrrolidone (PVP), polyvinylpyrrolidone/vinyl acetate (PVP/VA) copolymer, hydroxypropyl methylcel- lulose phthalate (HPMCP), hydroxypropyl methylcellulose acetate succinate (HPM- CAS), and poly(methyl methacrylate) (PMMA). Both experiment and molecular dynamics simulation reveal diverse mixing behavior amongst the set of polymers. Detailed structural and nano-scale interaction analyses suggest that positive deviations from ideal-mixing behavior arise from the formation of stable polymer-CBD hydrogen bonds whereas negative deviations are associated with disruptions to the polymer- polymer hydrogen-bond network. Polymer-water interaction analyses indicate the significance of polymer hydrophobicity that can lead to poor dissolution of CBD. These results have implications for drug dissolution rates based on how CBD and water interacts with each polymer. Furthermore, these insights may be used to guide ASD formulations for CBD or other small-molecule therapeutic agents.

Abstract:Cannabidiol (CBD) is a high-value natural compound of Cannabis Sativa plant. It is a non-psychotropic phytocannabinoid, attracting significant attention as a multifunctional active ingredient for topical applications. Although it is demonstrated that CBD can be used for specific dermatological ailments, reliable data on functionalities are limited. The present study aimed to investigate the structural stability, biofunctionality, and antimicrobial characteristics of CBD isolate to assist in the design of various topical formulations. The stability of CBD in solid and solubilized states was assessed to establish storage and formulation conditions. The performance of CBD solubilized in organic and aqueous media was evaluated for free radical scavenging, tyrosinase, and collagenase enzyme inhibition, which showed good prospects for the ingredient. The antimicrobial activity of solubilized CBD was evaluated against Gram-negative (E. coli, P. aeruginosa), Gram-positive bacterial strains (S. aureus, S. epidermidis, C. acnes), and fungal strains (C. albicans, M. furfur) using agar well diffusion and broth microdilution methods. Due to the presence of surfactants in CBD aqueous solution, it displayed a lack of antimicrobial activity against all the tested microorganisms. CBD solubilized in an organic medium showed no activity against Gram-negative bacterial strains but higher activity against tested Gram-positive bacterial and fungal strains.

Abstract:Much confusion exists about the chemical composition of widely sold Cannabis sativa products that utilize the cannabidiol (CBD) acronym and related terms such as "CBD oil", "CBD plus hemp oil", "full spectrum CBD", "broad spectrum CBD", and "cannabinoids". Their rational chemical and subsequent biological assessment requires both knowledge of the chemical complexity and the characterization of significant individual constituents. Applicable to hemp preparations in general, this study demonstrates how the combination of liquid-liquid-based separation techniques, NMR analysis, and quantum mechanical-based NMR interpretation facilitates the process of natural product composition analysis by allowing specific structural characterization and absolute quantitation of cannabinoids present in such products with a large dynamic range. Countercurrent separation of a commercial "CBD oil" yielded high-purity CBD plus a more polar cannabinoid fraction containing cannabigerol and cannabidivarin, as well as a less polar cannabinoid fraction containing cannabichromene, trans-Δ9-tetrahydrocannabinol, cis-Δ9-tetrahydrocannabinol, and cannabinol. Representatives of six cannabinoid classes were identified within a narrow range of polarity, which underscores the relevance of residual complexity in biomedical research on cannabinoids. Characterization of the individual components and their quantitation in mixed fractions were undertaken by TLC, HPLC, 1H (q)NMR spectroscopy, 1H iterative full spin analysis (HiFSA), 13C NMR, and 2D NMR. The developed workflow and resulting analytical data enhance the reproducible evaluation of "CBD et al." products, which inevitably represent complex mixtures of varying molecular populations, structures, abundances, and polarity features.
Development and characterization of a zeolite based drug delivery system: Application to cannabidiol oral delivery

Abstract:The growing interest in the therapeutic potential of cannabidiol (CBD) has led to the need for effective and reliable delivery methods that overcome its low oral absorption. Zeolites, a class of porous nanoparticles, offer unique advantages as drug carriers due to their high surface area and adjustable pore size. In this study, a zeolite-based drug delivery system was developed for the encapsulation of CBD. The zeolite particles were characterized using various techniques such as Scanning Electron Microscopy (SEM), N2 adsorption analysis, Solid-state Fourier Transform Infrared (FTIR), Direct Light Scattering (DLS), X-ray diffraction (XRD) and thermogravimetric analysis (TGA) before and after the loading. The drug encapsulation efficiency, and the release profile of CBD from the zeolite matrix were evaluated in addition to in vitro dissolution experiments in the intestinal and gastric simulated fluids. The results showed that the loaded zeolite particles exhibited high encapsulation efficiency of 73.5 %. XRD analysis proved that the USY structure remained intact after loading with CBD. DLS and N2 adsorption analysis indicated that CBD was successfully loaded into the zeolite matrix. When compared to CBD containing particles in a commercialized capsule, the in-vitro dissolution rate of CBD loaded zeolite was significantly higher after 30 min in the simulated stomach (pH 1.8) and the intestinal (pH 6.8) fluids, 67.8 % versus 43.6 % and 62.6 % vs 38.4 % respectively. Our findings open new avenues for the use of zeolites as an efficient drug delivery system for drugs with low bioavailability like CBD.
[Canine leishmaniosis in Central Europe: retrospective survey and serological study of imported and travelling dogs].

Abstract:Canine leishmaniosis is a common parasitic disease in Central Europe affecting dogs imported or returning from endemic countries around the Mediterranean basin. Through an internet discussion forum owners of dogs with suspected or proven leishmaniosis living in Central Europe (D, A, CH), were questioned about the dog's history. Additionally, serologic examinations of the dogs for anti-Leishmania antibodies (ELISA using antigen of promastigote stages) were offered to the participants. From February to October 2003, 291 dogs imported or returning from Southern Europe (Spain, Italy, Greece, Turkey, France, Malta, Portugal and others) were analysed. Serologically, 111 dogs (38%) were classified positive; 103 being imported and eight travelling dogs. The majority of seropositive dogs originated from Spain (67%). No significant correlation could be established between race, sex and age and the incidence of the disease. The clinical symptoms in the seropositive dogs varied widely and ranged from mild general symptoms to visceral manifestations with chronic renal failure. Skin disorders were found in 78% of the seropositive, symptomatic animals. In the country of origin or after import or return, 174 out of 291 dogs (60%) had been tested for the presence of anti-Leishmania antibodies by different immunofluorescence antibody tests (IFAT). Discrepancies between the ELISA and the various IFATs used were noted in 55 cases (32%), especially in cases of low IFAT titers. Most of the seropositive dogs (80%) had been treated against leishmaniosis. In 91% of these cases, Allopurinol as monotherapy or in combination with Glucantime had been used. For diagnostics and therapy, dog owners had spent an average of 1,100 euros (median 900 euros, maximum 5,800 euros).
Salts and Cocrystals of Benzocaine with Increased Dissolution Rate and Permeability Open New Avenues for Enhancing the Duration of Action

Abstract:Benzocaine, a widely used local anesthetic, has some usage limitations due to its short duration of action, resulting in inconsistent clinical outcomes. In this study, the formation of salts and cocrystals is explored as a strategy to enhance the pharmacokinetic profile of benzocaine, aiming for a higher dissolution rate and permeability in vitro as well as to potentially increase its pharmacological effect and duration of action in vivo. A new cocrystal and nine new salts with two different categories of acids (carboxylic and sulfonic) were prepared and characterized: BH oxalate, BH naphthalenesulfonate, BH camphorsulfonate, BH maleate, BH mesylate, BH tartrate, BH benzenesulfonate, BH p-toluenesulfonate, BH esylate (BH: protonated benzocaine), and benzocaine–ligustrazine (cocrystal). Dissolution rate and permeability were improved for all newly synthesized forms. Indeed, every system was completely dissolved within 1 h, while benzocaine was dissolved at 54%, and it was still present in suspension after 4 h. Furthermore, permeability was increased 2-fold for benzocaine maleate and up to seven times for benzocaine camphorsulfonate, highlighting the potential of salification and cocrystallization to increase the in vivo performances of benzocaine.

Structural and Property Analysis of Three Novel Pharmaceutical Salt Solvates Synthesized from Paliperidone and Aromatic Carboxylic Acids

Abstract:Paliperidone (PLPT) is a pharmaceutical compound widely used in the treatment of psychiatric disorders. However, its limited solubility poses challenges for its formulation and bioavailability.In this study, three previously unreported salt solvates of PLPT were successfully synthesized: PLPT·PCBA·H2O (PCBA, para-chlorobenzoic acid), PLPT·NA·2H2O·2MeOH (NA, niacin acid), and PLPT·GA·2EtOH (GA, gallic acid). Considering the requirement of drying in actual production, the test samples were dried in a 333.15 K overnight and then used for TGA-DSC (thermogravimetric analysis-differential scanning calorimetry) testing. The results indicate that the thermal stability of PLPT·NA·2H2O·2MeOH was enhanced by approximately 40 K. The salt-like nature of these compounds was confirmed through ΔpKa calculations and single-crystal structure analysis, which revealed the formation of charge-assisted hydrogen bonds (N+-H···O−). Significant hydrogen bonding and π-π interactions were observed, as evidenced by 3D dnorm Hirshfeld surfaces and 2D fingerprint plots. They were characterized by predominant H···H, O···H, and N···H interactions, contributing to their structural stability. PXRD (powder X-ray diffraction) and IR (infrared spectroscopy) characterization provided further confirmation of the solvate formation. Notably, solubility experiments demonstrated a remarkable increase in the solubility of PLPT·NA·2H2O·2MeOH in simulated human body environments, exhibiting solubility enhancements of approximately 883.78 times, 166.57 times, 5.18 times, and 6.37 times across the range from pure water to pH 1.2. The remaining two salt solvates exhibit a notable enhancement in solubility under different pH conditions. An anomaly is observed only at pH 4, which is associated with the degree of protonation of PLPT and the intrinsic solubility of GA and PCBA. Dissolution rates indicate a significant improvement in all three salt solvates under various simulated human physiological environments, particularly at pH 6.8.These findings highlight the potential of these solvates as promising candidates for enhanced PLPT formulations. Dissolution rate experiments also indicate that the formation of salts significantly enhances the drug's dissolution rate. This study represents the first report on salt solvates of PLPT and provides valuable insights for the improvement of PLPT and other poorly soluble drug formulations.
Bioactive Properties and In Vitro Digestive Release of Cannabidiol (CBD) from Tailored Composites Based on Carbon Materials

Abstract:The use of carriers to improve cannabidiol (CBD) bioavailability during digestion is at the forefront of research. The main objective of this research was to evaluate CBD bioactivity and develop CBD composites based on tailored carbon support to improve availability under digestive conditions. The antioxidant capacity of CBD was evaluated using spectrophotometric methods, and anti-proliferative assays were carried out using human colon carcinoma cells (SW480). Twenty-four composites of CBD + carbon supports were developed, and CBD desorption tests were carried out under simulated digestive conditions. The antioxidant capacity of CBD was comparable to and superior to Butylhydrox-ytoluene (BHT), a commercial antioxidant. CBD reflected an IC-50 of 10,000 mg/L against SW480 cancer cells. CBD in biological systems can increase the shelf life of lipid and protein foods by 7 and 470 days, respectively. Finally, acid carbons showed major CBD adsorption related to electrostatic interactions, but basic carbons showed better delivery properties related to electrostatic repulsion. A tailored composite was achieved with a CBD load of 27 mg/g with the capacity to deliver 1.1 mg, 21.8 mg, and 4 mg to the mouth, stomach, and duodenum during 18 h, respectively. This is a pioneering study since the carriers were intelligently developed to improve CBD release.
