Abstract:Aflatoxin B1 (AFB1) is a potent carcinogen, and is among the most hazardous mycotoxins in agricultural products. Therefore, the development of sensitive and convenient detection methods for AFB1 is significant for food safety against mycotoxins. Herein, a bioluminescent enzyme immunoassay (BLEIA) was developed for ultrasensitive detection of AFB1, based on the novel Fc-specific antibody-nanoluciferase (Ab-Nluc) conjugates which were fabricated using an IgG-binding protein-assisted photo-conjugation strategy. In indirect competitive immunoassay format, the proposed BLEIA exhibited the detection limit of 0.0232 ng mL-1, which was 37.4-fold lower than that obtained using the classical enzyme-linked immunosorbent assay (ELISA) based on Ab-horseradish peroxidase (Ab-HRP) chemical conjugates (0.868 ng mL-1). Meanwhile, the BLEIA exhibited high accuracy and precision. Thus, the proposed Fc-specific Ab-Nluc conjugates-based BLEIA provides an ultrasensitive and reliable method for detecting toxins and has potential for use in food safety monitoring.

Abstract:Edible oil safety impacts food safety and consumer health. The typical pollutants—aflatoxin B1 (AFB1) and benzo(α)pyrene (BaP), and kitchen waste oil—are significant hazards in edible oil consumption. Herein, we developed a dual-readout lateral flow immunoassay (tdLFIA) for the multi-quantitative detection of AFB1, BaP and capsaicin (CAP). A novel monoclonal antibody against BaP was developed with a sensitivity of 1.92 ng/mL. Subsequently, gold nanoparticles and Eu3+ labelled fluorescent nanospheres were synthesized as colorimetric and fluorescent sensors. A rapid synchronous pretreatment method based on immunomagnetic beads(IMAB)combined with a molecularly imprinted solid phase extraction (MISPE) was developed. The tdLFIA enabled a rapid response of 7 min for AFB1, BaP and CAP detection, with quantitative limits of detection of 0.003, 0.6, and 0.01 ng/mL, respectively. The proposed strategy indicated reliability and has been applied in real samples, providing useful products for evaluating edible oil quality and safety.
Chemiluminescent Enzyme Immunoassay and Bioluminescent Enzyme Immunoassay for Tenuazonic Acid Mycotoxin by Exploitation of Nanobody and Nanobody–Nanoluciferase Fusion

Abstract:The isolation of nanobodies (Nbs) from phage display libraries is an increasingly effective approach for the generation of new biorecognition elements, which can be used to develop immunoassays. In this study, highly specific Nbs against the Alternaria mycotoxin tenuazonic acid (TeA) were isolated from an immune nanobody phage display library using a stringent biopanning strategy. The obtained Nbs were characterized by classical enzyme-linked immunosorbent assay (ELISA), and the best one Nb-3F9 was fused with nanoluciferase to prepare an advanced bifunctional fusion named nanobody–nanoluciferase (Nb–Nluc). In order to improve the sensitivity and reduce the assay time, two different kinds of luminescent strategies including chemiluminescent enzyme immunoassay (CLEIA) and bioluminescent enzyme immunoassay (BLEIA) were established, respectively, on the basis of the single Nb and the fusion protein Nb–Nluc for TeA detection. The two-step CLEIA was developed on the basis of the same nanobody as ELISA, only with simple substrate replacement from 3,3′,5,5′-tetramethylbenzidine (TMB) to luminol. In contrast with CLEIA, the novel BLEIA was conducted in one-step new strategy on the basis of Nb–Nluc and bioluminescent substrate coelenterazine-h (CTZ-h). Their half maximal inhibitory concentration (IC50) values were similar to 8.6 ng/mL for CLEIA and 9.3 ng/mL for BLEIA, which was a 6-fold improvement in sensitivity compared with that of ELISA (IC50 of 54.8 ng/mL). Both of the two assays provided satisfactory recoveries ranging from 80.1%–113.5% in real samples, which showed better selectivity for TeA analogues and other common mycotoxins. Abstract:Aflatoxin B1 (AFB1), one of the most toxic mycotoxins, has undergone a wide range of studies over the years. The development of rapid, simple, and sensitive analytical methods remains a major challenge for the accurate detection of AFB1 in foodstuffs. In this study, we designed an enhanced and stable ingenious electrochemiluminescence bioaptasensor (IEC-BA) for ultrasensitive detection of AFB1 based on the synergistic effects and enzyme-driven programmable assembled 3D DNA nanoflowers (EPDNs). This synergistic effect was comprised by the competitive impact on auxiliary probes (AP) and the cutting effect of the Hae III. Compared to the traditional aptamer direct-competition method, the synergistic effects ensured that the aptamer was more efficiently and adequately competed away by the target. Also, the redundant double-stranded probes were removed, which greatly facilitates simple, quick, and sensitive detection of AFB1. Besides, a large chunk of positively charged Ru(II) complexes (Ru(bpy)32+) was accumulated by the utilization of EPDNs, which resulted in tremendous improvement of the sensitivity of the designed method. Thus, even in the presence of trace amounts of AFB1, a sharply visual electrochemiluminescent signal was generated. The proposed method can realize the quantification of AFB1 with a good linear range from 1 ppt (pg mL–1) to 5 ppb (ng mL–1) with a detection limit of 0.27 ppt. In addition, it can also be successfully applied for the analysis of AFB1 in a peanut and wheat, with total recoveries ranging from 93.7 to 106.6%. A versatile CuCo@PDA nanozyme-based aptamer-mediated lateral flow assay for highly sensitive, on-site and dual-readout detection of Aflatoxin B1

Abstract:Mycotoxin contaminations in food and environment seriously harms human health. Constructing sensitive and point-of-test early-warning tools for mycotoxin determination is in high demand. In this study, a CuCo@PDA nanozyme-based aptamer-mediated lateral flow assay (Apt-LFA) has been elaborately designed for on-site and sensitive determination of mycotoxin Aflatoxin B1 (AFB1). Benefiting from the rich functional groups and excellent peroxidase-like activity, the CuCo@PDA with original dark color can be conjugated with the specific recognition probe (i.e., aptamer), generating colorimetric signal on the test lines of Apt-LFA via a competitive sensing strategy. The signal can further be amplified in-situ by catalytic chromogenic reaction. Therefore, a visual and dual-readout detection of AFB1 has been realized. The developed Apt-LFA provides a flexible detection mode for qualitative and quantitative analysis of AFB1 by naked-eyes observation or smartphone readout. The smartphone-based LFA platform shows a reliable and ultrasensitive determination of AFB1 with the limit of detection (LOD) of 2.2 pg/mL. The recoveries in the real samples are in the range of 95.11-113.77% with coefficients of variations less than 9.84%. This study provides a new approach to realize point-of-test and sensitive detection of mycotoxins in food and environment using nanozyme-based Apt-LFAs.

Abstract:Traditional lateral flow immunoassays (TLFIAs) are valued for their simplicity, speed, and user-friendliness. However, the specificity of conventional test strips often necessitates large quantities of antigen-protein conjugates for target detection, which can be resource-intensive. Here, we present a strategy aimed at enhancing the universality of test strips while reducing the consumption of antigen-protein conjugates, without compromising sensitivity. By coating streptavidin on the test line and employing bifunctional antigen-protein conjugates (competitor to target and immunoprobe linker), the test strip was thus served as a universal module. We developed three universal lateral flow immunoassays (ULFIAs) for the detection of aflatoxin B1 (AFB1), carbendazim (CBZ), and enrofloxacin (ENR). Compared to traditional methods based on the same aggregation-induced emission fluorescent microspheres, the proposed ULFIAs demonstrated a significant increase in sensitivity, with enhancements of 11.0-fold for AFB1, 10.9-fold for CBZ, and 4.1-fold for ENR. Additionally, this approach substantially reduced the consumption of antigen-protein conjugates by 19.1-fold, 40.9-fold, and 23.8-fold, respectively, thereby promoting greener detection methods. This bifunctional antigen conjugate strategy offers a promising pathway for the sensitive detection of small molecule hazards.

Abstract:Traditional enzyme-linked immunosorbent assay (ELISA) suffers from the limitations of relatively low sensitivity and stability, and enzyme-labelled antibodies are hard to be prepared and purified. Based on a nanozyme, an aptamer and Fe3O4 magnetic nanoparticles (MNP), a nanozyme and aptamer-based immunosorbent assay (NAISA) was developed for aflatoxin B1 (AFB1) detection with simpler operation and separation. In this work, mesoporous SiO2/Au-Pt (m-SAP) were prepared to act as signal labels, which showed high catalase-like activity and was denoted as nanozyme. Aptamer was adopted to specifically recognize with AFB1, and MNP facilitated to realize magnetic separation. To verify the performance of NAISA, traditional ELISA (t-ELISA) and enhanced ELISA (e-ELISA) using MNP and m-SAP nanozyme were applied in AFB1 detection. The NAISA method showed the lowest limit of detection (LOD) with 5 pg mL−1 (n = 3, ±4.2 %), 600 and 12-fold lower than that of t-ELISA (3 ng mL−1) and e-ELISA (0.06 ng mL−1), respectively. In the interference tests, AFB1 can be identified among six different interfering substances. The NAISA method, thus, can be of great importance as it allows selective and sensitive AFB1 detection, while providing the simplicity of use and need for screening hazardous materials.

A lateral flow immunochromatographic assay based on nanobody-oriented coupling strategy for aflatoxin B1 detection

Abstract:Nonoriented coupling of nanobodies (Nb) to nanomaterials can considerably reduce epitope recognition or induce other dysfunctions, thereby restricting the application of Nb in field detection. Herein, using Avi-tag/streptavidin (Avi/SA)-oriented coupling strategy, we developed a lateral-flow immunochromatographic assay (LFIA) based on Nb-Avi/SA@quantum dots (QDs) probes (Nb-LFIA) for Aflatoxin B1 (AFB1) detection. Compared with nonoriented coupled Nb@QD probes, the detection sensitivity and stability of Nb-Avi/SA@QD probes were greatly retained. Furthermore, Nb showed higher tolerance to the extreme detection environments compared with monoclonal antibodies in traditional enzyme-linked immunosorbent assay and LFIA. After optimization, the limit of detection of Nb-LFIA was 0.095 ng mL−1, half-maximal inhibitory concentration was 0.85 ng mL−1, and the visual cut-off level was 1.25 ng mL−1. Nb-Avi/SA@gold nanoparticle probes were assessed for the wide applicability of the Avi/SA strategy, and they showed excellent performance (coefficient of determination = 0.999). Finally, the analysis performance of probes was assessed in oat samples, revealing a recovery rate of 88.8–116.7%, which was consistent with the results of high-pressure liquid chromatography coupled with tandem mass spectrometry. Altogether, our study provides a convenient and sensitive tool for AFB1 detection, and presents a viable path for efficient Nb application.

Abstract:Background: The utilization of inner filter effect (IFE) brings more opportunities for construction of fluorescence immunoassays but remains a great challenge, especially how to select best donor in the face of extensive fluorescent nanomaterials. Aflatoxin B1 possesses high toxicity among mycotoxins and is frequently found in agricultural products that may significantly threaten to human health. Therefore, with the help of signal transduction mechanism of IFE to develop a convenient and sensitive approach for AFB1 detection is of great significance in ensuring food safety. Results: Herein, the classical alkaline phosphatase (ALP) catalyzes hydrolysis of p-nitrophenylphosphate to produce p-nitrophenol (PNP) was employed as a model reaction, which intends to explore tunable multicolor fluorescence of gold nanoclusters (AuNCs) for matching PNP to maximize IFE efficiency. The luminescent green-emitting AuNCs were selected as an optimal donor in terms of excellent spectral overlap, high photoluminescence, and adequate system adaptability, thus achieving a 22-fold increase in sensitivity improvement compared to colorimetric method for ALP detection. The fluorescence quenching mechanism between PNP and AuNCs was validated as IFE by studying ultraviolet absorption, zeta potentials and fluorescence lifetime. In light of this, we integrated a highly specific antibody-antigen recognition system, efficient enzymatic reaction and excellent optical characteristics of AuNCs to develop dual-mode immunoassay for AFB1 monitoring. The sensitivity of fluorometric immunoassay was lower to 0.06 ng/mL, which obtained a 3.5-fold improvement compared to "gold standard" ELISA. Their practicability and applicability were confirmed in the tap water, corn, wheat and peanuts samples. Significance: This work provides an easy-to-understand screening procedure to select optimal donor-acceptor pairs in IFE analysis. Furthermore, we expect that integration of IFE-based signal conversion strategy into mature immunoassay not only extends the signal types, simplifies signal amplification steps, and reduces the false-positive/false-negative rates, but also provides a simple, convenient, and versatile strategy for monitoring of trace other contaminants.

Abstract:Mycotoxins posit serious threats to human and animal health, and numerous efforts have been performed to detect the multiple toxins by a single diagnostic approach. To best of our knowledge, for the first time, we synthesized an aptamer induced "turn on" fluorescence resonance energy transfer (FRET) biosensor using dual-color gold nanoclusters (AuNCs), L-proline, and BSA synthesized AuNCs (Lp-AuNCs and BSA-AuNCs), with WS2 nanosheet for simultaneous recognition of aflatoxinB(1) (AFB(1)) and zearalenone (ZEN) by single excitation. Here, AFB(1) aptamer stabilized blue-emitting AuNCs (AFB'apt-Lp-AuNCs) (at 442 nm) and ZEN aptamer functionalized with red-colored AuNCs (ZEN-apt-BSA-AuNCs) (at 650 nm) were employed as an energy donor and WS2 nanosheet as a fluorescence quencher. With the addition of AFB(1) and ZEN, the change in fluorescence intensity (F.I) was recorded at 442 and 650 nm and can be used for simultaneous recognition with a detection limit of 0.34 pg mL(-1) (R-2 = 0.9931) and 0.53 pg mL(-1) (R-2 = 0.9934), respectively. Most importantly, the semiquantitative determination of AFB(1) and ZEN can also be realized through photovisualization. The current approach paves a new way to develop sensitive, selective, and convenient metal nanocluster-based fluorescent "switch-on" probes with potential applications in multipurpose biosensing.
Mycotoxin Detection through Colorimetric Immunoprobing with Gold Nanoparticle Antibody Conjugates

Abstract:Driven by their exceptional optical characteristics, robust chemical stability, and facile bioconjugation, gold nanoparticles (AuNPs) have emerged as a preferred material for detection and biosensing applications in scientific research. This study involves the development of a simple, rapid, and cost-effective colorimetric immuno-sensing probe to detect aflatoxin B1 and zearalenone using AuNP antibody (AuNP-mAb) conjugates. Anti-toxin antibodies were attached to the AuNPs by using the physical adsorption method. The colorimetric immunosensor developed operates on the principle that the optical properties of the AuNP are very sensitive to aggregation, which can be induced by a critical high salt concentration. Although the presence of antibodies on the AuNP surface inhibits the aggregation, these antibodies bind to the toxin with higher affinity, which leads to exposure of the surface of AuNPs and aggregation in a salt environment. The aggregation triggers a noticeable but variable alteration in color from red to purple and blueish gray, as a result of a red shift in the surface plasmon resonance band of the AuNPs. The extent of the shift is dependent on the toxin exposure dose and can be quantified using a calibration curve through UV–Visible–NIR spectroscopy. The limit of detection using this assay was determined to be as low as 0.15 ng/mL for both zearalenone and aflatoxin B1. The specificity of the prepared immunoprobe was analyzed for a particular mycotoxin in the presence of other mycotoxins. The developed immunoprobe was evaluated for real-world applicability using artificially spiked samples. This colorimetric immunoprobe based on localized surface plasmon resonance (LSPR) has a reduced detection limit compared to other immunoassays, a rapid readout, low cost, and facile fabrication.

Abstract:Aflatoxin B1 (AFB1) is one of the most hazardous mycotoxins and lead to a deadly danger to animal and human health. The development of portable, low-cost, and easy to operate analytical tools is essential for AFB1 detection, especially in resource-limited regions. Herein, a dual-mode microfluidic paper-based analytical device (μPAD) with electrochemical and colorimetric readout was constructed for AFB1 detection using Au NPs anchored Ni-Co layered double hydroxides nanocages (Au/Ni-Co LDH NCs) as a signal amplifier. In this design, tetrahedral DNA nanostructures (TDNs) were employed as a scaffold to anchor “bottom-up” aptamers (Apt) for improving capture efficiency of AFB1. Au/Ni-Co LDH NCs with excellent peroxidase (POD)-like activity were bound on the TDNs through hybridization of Apt and complementary DNA (cDNA), a significant electrochemical signal of H2O2 reduction was generated. Meanwhile, the·OH from H2O2 decomposition catalyzed the oxidation of 3,3′,5,5′-tetramethylbenzidine and resulted in a colorimetric signal. The binding of target AFB1 to Apt triggered the dissociation of Apt/cDNA, causing the release of the signal amplifier from the sensing interface and signal decrease. Consequently, a detection range of 0.2 pg/mL-100 ng/mL for electrochemical and 50 pg/mL-100 ng/mL for colorimetric were achieved for AFB1, holding promising application in the fields of food safety.

Abstract:We describe a new fluorescence turn-on sensor for homogeneous detection of aflatoxin B1 (AFB1), a potent low molecular weight mycotoxin. A key innovation is the binding-induced intramolecular interaction involving the following two sets of probes: (1) a gold nanoparticle (AuNP) immobilized with hundreds of assistant oligonucleotides (AO) and dozens of anti-AFB1 monoclonal antibodies (mAb), and (2) the AFB1-BSA antigen conjugated with fluorophore-labeled signal oligonucleotides (SO) that contained a short sequence complementary to AO. Specific binding of AFB1-BSA to the antibody brought the fluorophore very close to the surface of the AuNP through a stable intramolecular hybridization between AO and SO, resulting in efficient quenching of fluorescence. The improved fluorescence quenching substantially reduced the background, due to the binding-induced intramolecular hybridization, and improved the signal-to-background ratio by 390%. In the presence of AFB1 in a sample, competitive binding of AFB1 in the sample to the antibodies immobilized on the AuNP caused the release of the fluorophore-labeled AFB1-BSA from the AuNP, turning on fluorescence. A detection limit of 2.3 nM was achieved, which meets the requirement for AFB1 detection at regulatory levels. Analyses of rice samples using this assay showed recoveries of 86-102%. Incorporating appropriate antibody probes could extend the assay to the detection of other small molecules.

Abstract:Aflatoxin B1 (AFB1) is a prominent food contaminant known for its high toxicity. Rapid, low-cost, and sensitive detection of AFB1 is an urgent demand for food safety risk control. In the traditional enzyme-linked immunosorbent assay (ELISA) method for AFB1 detection, horseradish peroxide (HRP) is high-cost and possesses poor environmental stability. In this study, a direct competitive single-atom nanozymes (SAzymes) -linked immunosorbent assay was developed based on Fe-N-C SAzymes. The proposed methods exhibited excellent sensitivity, rapid detection, fabrication, and low cost. Under the optimal conditions, the limit of detection of the new assay was 3.3 pg mL−1, with a quantitative relationship between 0.0084 and 0.358 ng mL−1. In addition, the developed method presented good recoveries in spiked samples, ranging from 84.50% and 103.75%, with coefficients of variation below 10%. Furthermore, the developed detection method showed good consistency (R2 = 0.9852) with the reference method, liquid chromatography/mass spectrometry (LC-MS/MS). To our knowledge, this is the first report about single-atom nanozymes-based ELISA for AFB1 determination. This work provides a feasible strategy for the rapid and sensitive detection of food contaminants using SAzymes.

Abstract:This work describes a novel "off-on" biosensor for super-sensitive and selective detection of aflatoxin B1 (AFB1). In the biosensor, electrochemiluminescence (ECL) and aggregation-induced emission (AIE) work together to enhance ECL performance by a dibenzo-[b,d]thiophene sulfone-based conjugated microporous polymer, and hairpin DNA1 (H1) and ferrocene (Fc)-apt act as the switches for the "off-on" mode. In the absence of target AFB1, the ECL signal is inhibited because of the steric-hindrance effect, and the biosensor remains in the "off" state. In the presence of AFB1, ECL signal recovery is observed caused by the specific recognition between the AFB1 and Fc-apt, i.e., the biosensor changes to the "on" state. Under optimal experimental conditions, AFB1 can be quantified from 1 pg·mL −1 to 1 μg·mL −1 with an ultralow detection limit of 0.46 pg·mL −1 by the "off-on" biosensor. The biosensor exhibited good practicability for AFB1 assay in corn samples, thus providing a simple and sensitive method for monitoring food safety.

Abstract:Aflatoxin B 1 (AFB 1 ) is one of the most prevalent and dangerous biotoxin in crops and feedstuff, which poses a great threat to human health and also cause significant financial losses. Therefore, there is an urgent need to develop an effective method for AFB 1 detection. In this work, we developed an automatic reaction equipment and nanozyme-enhanced immunosorbent assay (Auto-NEISA) for sensitive and accurate detection of AFB 1 by combining the highly effective signal probes with a self-designed automated immunoreactive equipment. Wherein, polystyrene (PS) nanoparticles were used as signal carriers for loading a massive in situ-synthesized platinum and palladium bimetallic nanozyme, which could enrich horseradish peroxidase-labeled goat anti-mouse antibody (HRP-Ab 2 ) on the nanozyme surface to form a bimetallic nanozyme–bioenzyme hybrid material for multiple signal amplification. The entire reaction could be automatically completed by the self-developed immunoreactive equipment. The Auto-NEISA method realized the sensitive detection of AFB 1 with a wide linear detection range of 10–10 4 pg/mL, at a low limit of detection (LOD) of 5.52 pg/mL. The LOD was 65-fold lower than that of the enzyme-linked immunosorbent assay (ELISA). Additionally, Auto-NEISA was successfully applied to detect AFB 1 in real food samples, demonstrating that it has considerable potential for detecting food contaminants with high accuracy and efficiency.

Abstract:Immunoassays based on enzyme-labeled antibodies have been widely used in the food safety field. However, the production process of enzyme-labeled antibodies is complicated and the low storage stability limits their application. Herein, antibody-horseradish peroxidase (HRP) co-assembled nanocomposites (AHC NCs) with outstanding advantages such as enhanced stability, lower cost, and substrate affinity were successfully prepared via a one-pot green method. Then the AHC NCs were employed as an alternative to traditional enzyme-labeled antibodies to develop a chemiluminescence enzyme immunoassay (CLEIA) toward Escherichia coli (E. coli) O157:H7. Under optimal conditions, E. coli O157:H7 can be detected in a linear range from 1 × 103 CFU mL-1 to 5 × 106 CFU mL-1, while the limit of detection (LOD) is as low as 2.2 × 102 CFU mL-1 (3σ). A series of repeatability studies showed reproducible results with a coefficient of variation of less than 7%. In addition, the proposed CLEIA was successfully applied to the analysis of spiked samples (tap water) and gave quantitative recoveries from 93.72% to 100.72%. This work demonstrates that the developed CLEIA can be applied as a universal platform for specific detection of diversified analytes.
