Abstract:The quality and flavor of tea leaves are significantly influenced by chemical composition, with the content of free amino acids serving as a key indicator for assessing the quality of Tencha. Accurately and quickly measuring free amino acids during tea processing is crucial for monitoring and optimizing production processes. However, traditional chemical analysis methods are often time-consuming and costly, limiting their application in real-time quality control. Hyperspectral imaging (HSI) has shown significant effectiveness as a component detection tool in various agricultural applications. This study employs VNIR-HSI combined with machine learning algorithms to develop a model for visualizing the total free amino acid content in Tencha samples that have undergone different processing steps on the production line. Four pretreating methods were employed to preprocess the spectra, and partial least squares regression (PLSR) and least squares support vector machine regression (LS–SVR) models were established from the perspectives of individual processes and the entire process. Combining competitive adaptive reweighted sampling (CARS) and variable iterative space shrinkage approach (VISSA) methods for characteristic band selection, specific bands were chosen to predict the amino acid content. By comparing modeling evaluation indicators for each model, the optimal model was identified: the overall model CT+CARS+PLSR, with predictive indicators Rc2 = 0.9885, Rp2 = 0.9566, RMSEC = 0.0956, RMSEP = 0.1749, RPD = 4.8021, enabling the visualization of total free amino acid content in processed Tencha leaves. Here, we establish a benchmark for machine learning-based HSI, integrating this technology into the tea processing workflow to provide a real-time decision support tool for quality control, offering a novel method for the rapid and accurate prediction of free amino acids during tea processing. This achievement not only provides a scientific basis for the tea processing sector but also opens new avenues for the application of hyperspectral imaging technology in food science.
Quantitative Analysis by NIR Spectrum Technique for the Main Components of Savor in Green Tea

Abstract:Choosing some representative kinds of green tea as the materials, the experiment established the calibra- tion models of three main sapor components——tea polyphenols (TPP), amino acids (AA) and theine from the tea soup by using NIR Quantitative analysis. The tea polyphenols(TPP) model: the standard error of calibration(SEE), correlation coefficient(r)of calibration set(C-set), standard error of cross-validation (SEP) and validation set (V-set) were: 1.1455、 0.8792、1.0600、0.8623. The AA model: its SEE, SEE -r, SEP and SEP -r were: 0.2518、0.9728、0.2597、0.9722. The theine model: its SEE, SEE-r, SEP, and SEP-r were: 0.5553、0.9648、0.5315、0.9554. It indicated that by external cross validation, correlation coefficient(R) of true content of samples and forecasted content of samples: R of TPP was 0.8935, R of AA was 0.9541, R of theine was 0.9386. The models could satisfy the analysis requirements of tea components. The proposed method was proved to be a rapid method for mensurating components of tea soup.
Rapid Analysis of the In-Process Extract Solutions of Compound E Jiao Oral Liquid Using Near Infrared Spectroscopy and Partial Least-Squares Regression

Abstract:A rapid and simultaneous quantification of the main active compounds of the in-process extract solutions is beneficial to the process monitoring as well as ensuring quality consistency of the end products during the production of traditional Chinese medicine. Here we present a near infrared (NIR) spectroscopy-based method for rapid analysis of extract solutions in the production of compound E Jiao oral liquid. The partial least-squares regression (PLSR) models for four quality indicators (viz. total flavonoids, total saponins, total saccharides and soluble solid contents) were established and validated. The results showed that all of the four models exhibited satisfactory fitting and predictive capacity. The root mean squares error of prediction (RMSEP) was 0.0384 mg mL−1, 0.0154 mg mL−1, 3.80 mg mL−1 and 0.199% for total flavonoids, total saponins, total saccharides and soluble solid contents, respectively. This work here demonstrated that NIR spectroscopy coupled with PLSR calibration can offer a reliable and non-destructive alternative in the routine monitoring of the extraction process in the production of compound E Jiao oral liquid. The presented approach is expected to be equally applicable to the mixed decoction of other herbal medicines.
Intestinal Inflammation Modulates Expression of the Iron-Regulating Hormone Hepcidin Depending on Erythropoietic Activity and the Commensal Microbiota

Abstract:States of chronic inflammation such as inflammatory bowel disease are often associated with dysregulated iron metabolism and the consequent development of an anemia that is caused by maldistribution of iron. Abnormally elevated expression of the hormone hepcidin, the central regulator of systemic iron homeostasis, has been implicated in these abnormalities. However, the mechanisms that regulate hepcidin expression in conditions such as inflammatory bowel disease are not completely understood. To clarify this issue, we studied hepcidin expression in mouse models of colitis. We found that dextran sulfate sodium–induced colitis inhibited hepcidin expression in wild-type mice but upregulated it in IL-10–deficient animals. We identified two mechanisms contributing to this difference. Firstly, erythropoietic activity, as indicated by serum erythropoietin concentrations and splenic erythropoiesis, was higher in the wild-type mice, and pharmacologic inhibition of erythropoiesis prevented colitis-associated hepcidin downregulation in these animals. Secondly, the IL-10 knockout mice had higher expression of multiple inflammatory genes in the liver, including several controlled by STAT3, a key regulator of hepcidin. The results of cohousing and fecal transplantation experiments indicated that the microbiota was involved in modulating the expression of hepcidin and other STAT3-dependent hepatic genes in the context of intestinal inflammation. Our observations thus demonstrate the importance of erythropoietic activity and the microbiota in influencing hepcidin expression during colitis and provide insight into the dysregulated iron homeostasis seen in inflammatory diseases.
Establishment of Predictive Model for Quantitative Analysis of Major Components in Jasmine Tea by Near Infrared Spectroscopy(NIRS)

Abstract:Taking 112 representative jasmine tea as research material, the quantitative calibration models for determining the contents of 13 chemical compositions such as total tea polyphenols (TP), free amino acids (AA), caffeine (CAFF), water extract, GA, GC, EGC, C, EC, EGCG, GCG, ECG, CG were established by NIRS combined with chemometrics. Q-value taking all important statistical values into account can evaluate the quality of models. Results showed that almost all the Q-value of models was 0.8~0.9, except for CG with a Q-value of 0.7702. Each of the 13 predictive models was applied for external inspection. Coefficients of correlation (R) between predicted value by NIR and the actual value were exceed 0.9, except CAFF and GCG was 0.7~0.8 and GC、EGCG、ECG、CG was 0.8~0.9. The established models present high stability and predictive accuracy. This paper provided a simple and convenient method for measuring the contents of major quality components in jasmine tea.
Quantitative Evaluation of Taste-characteristic Attributes in Three White Tea Based on Near Infrared Spectroscopy

Abstract:In this study, tea taste-quality evaluations and attribution analysis were performed in white teas of different producing areas, tea types and storage times. The potential of near infrared spectroscopy (NIRS) in determining taste-attributes of white tea was also evaluated. The results showed that sweet, mellow and brisk tastes were the main aspects of taste-quality evaluation in white teas. Increasing of post-oxidation degrees could significantly enhance the sweet, mellow and brisk tastes of white tea with the same leave materials and processing technology. Near infrared spectroscopy (NIRS) was used to develop four models correlating spectral data to general taste-characteristics and its individual attributes (sweet, mellow, brisk tastes and post-oxidation characteristics). The four optimized models based on near infrared spectroscopy-partial least squares (NIRS-PLS) had considerable potential for taste-quality evaluation with correlation coefficient of calibration (Rc) ranging from 0.837 to 0.987 and that of prediction (Rp) ranging from 0.748 to 0.888, which suggested that fast taste-attribute evaluation of white tea could be achieved by NIRS-PLS.

Abstract:In order to improve the technical level of the rapid detection of liquor fermented grains, in this paper, use near infrared spectroscopy technology to quantitative analysis moisture, starch, acidity and alcohol of liquor fermented grains. Using CARS, iPLS and no information variable elimination method (UVE), realize the characteristics of spectral band selection. And use the multiple scattering correction (MSC), derivative and standard normal variable transformation (SNV) pretreatment method to optimize the models. Establish models of quantitative analysis of fermented grains by PLS, and in order to select the best modeling method, using R-2, RMSEP and optimal number of main factors to evaluate models. The results showed that the band selection is vital to optimize the model and CARS is the best optimization of the most significant effect. The calculation results showed that R2 of moisture, starch, acidity and alcohol were 0. 885, 0. 915, 0. 951, 0. 885 respectively and RMSEP of moisture, starch, acidity and alcohol were 0. 630, 0. 519, 0. 228, 0. 234 respectively. After optimization, the model prediction effect is good, the models can satisfy the requirement of the rapid detection of liquor fermented grains, which has certain reference value in the practical.

Abstract:The application of near infrared (NIR) spectroscopy for on-line quantitative monitoring of alcohol precipitation of the Danhong injection was investigated. For the NIR measurements, two fiber optic probes designed to transmit NIR radiation through a 2mm path length flow cell were applied to collect spectra in real-time. Particle swarm optimization- (PSO-) based least square support vector machines (LS-SVM) and partial least squares (PLS) models were developed for quantitative analysis of the critical intermediate quality attributes: the soluble solid content (SSC) and concentrations of danshensu (DSS), protocatechuic aldehyde (PA), hydroxysafflor yellow A (HSYA) and salvianolic acid B (SAB). The optimal models were then used for on-line quantitative monitoring of alcohol precipitation. The results showed that the PSO-based LS-SVM with a radial basis function (RBF) kernel was slightly better than the conventional PLS method, even though both methods exhibited satisfactory fitting results and predictive abilities. In this study, successful models were built and applied on-line; these models proffer real-time data and instant feedback about alcohol precipitation.
Rapid monitoring approaches for concentration process of lanqin oral solution by near-infrared spectroscopy and chemometric models

Abstract:Qualitative and quantitative detection methods based on near-infrared spectroscopy (NIRs) have been proposed in the process analysis of traditional Chinese medicine in recent years. In this study, rapid monitoring methods were developed for quality control of concentration process of lanqin oral solution (LOS). Partial least squares regression (PLSR) method was adopted to construct quantitative models for epigoitrin, geniposide, baicalin, berberine hydrochloride and density. Simultaneously, the genetic algorithm joint extreme learning machine (GA-ELM) was first applied in qualitative analysis of NIRs to distinguish end point of concentration process. Results of PLSR models were satisfactory with the relative standard error of calibration valued at 3.80%, 3.75%, 3.79%, 11.5% and 1.22% for epigoitrin, geniposide, baicalin, berberine hydrochloride and density respectively, and the residual predictive deviation values were higher than 3. For qualitative analysis, the GA-ELM model obtained 100% prediction accuracy. The PLSR quantitative models and the end point discrimination model constructed by GA-ELM correspond with the requirements of practical applications. The results indicate that NIRs in combination with chemometrics has great potential in improving the efficiency in production. (c) 2020 Elsevier B.V. All rights reserved.
Near-infrared Spectroscopy On-Line and Real-Time Monitoring of Extraction Process of Xuebijing Injection

Abstract:Objective:To establish a rapid quantitative analysis model for quality control of Xuebijing Injection extraction with application of near-infrared(NIR) spectroscopy,and quickly determine the end point of extraction process,so as to realize on-line monitoring of the extraction process of Xuebijing Injection.Methods: Taking mixed extraction process of Ligusticum chuanxiong Hort.,Salvia miltiorrhiza Bge.and Angelica sinensis(Oliv.) Diels for an example,partial least squares regression(PLSR) models were developed for the parameters of interest: total solid content and ferulic acid.The established PLS calibration models were used for on-line and real-time monitoring of the extraction process,and the moving block of standard deviation(MBSD) was used to identify the extraction end point.Furthermore,a model updating method was adopted to improve the stability and prediction accuracy of the calibration models.Results: The correlation coefficients of total solid content and ferulic acid models have reached 0.986.When the established models were applied to on-line monitoring,the correlation coefficients of prediction(r) and the residual predictive deviations(RPD) were above 0.967 and 3,respectively,and the relative standard errors of predictions(RSEP) were less than 10%.The result of MBSD was generally coincided with the variation of actual-measured ferulic acid concentration.In addition,the predictive accuracy was significantly improved after model updating with new samples.Conclusions: In this study,NIR spectroscopy was successfully applied to predicting total solid content and ferulic acid concentration in real-time and rapid determination of the extraction end point.Moreover,to solve the problem of calibration invalidation,the method of model updating was utilized to extend the use of established models and obtain better prediction results.

Abstract:Thirty-eight beers from different producing areas and/or makers were distinguished by principal component analysis (PCA) of the near infrared (NIR) spectra acquired by a portable NIR spectrometer. Classsification of Akita beers: beers locally produced in Akita prefecture, Japan, from other famous brand beers could be successfully performed, especially when the PCA was calculated on the standard normal variate (SNV) spectra. The classification equations use information related to water and CH2absorption that reflected the differences in chemical com position of beers due to different production processes. In addition, the compositions of total polyphenol and total nitrogen were estimated from NIR spectra by multiple linear regression (MLR). This study showed that NIR spectroscopy is promising for beer quality evaluation, both for identifying multifarious beers including Akita beers using PCA and for rapid in-line quality control and inspection for beer production using the quantitative MLR analysis.
Primary Exploration of Key Influential Factors Recognization Method of Tanreqing Injection

Abstract:To recognize the key influential factors during the liquid preparation process of Tanreqing injection, the near infrared(NIR) spectra of the raw materials and the operating parameters of 24 batches of physical manufacturing were recorded as independent variables, and the total soluble solids contents and the light inspection acceptance rate of the final products were collected as dependent variables. The calibration models were developed using the partial least-square regression (PLSR) method, and the correlation coefficients between the independent variables and the dependent variables were calculated. For the quantitative models, the correlation coefficients for the calibration and inner cross validation of total soluble solids contents and the light inspection acceptance rate reached 0.911 9, 0.724 2 and 0.873 8, 0.795 9, respectively. Using the correlation coefficients diagrams, several key influence factors were preliminarily determined, and the physical significance were analyzed combined with production experience. This work demonstrated that NIR spectroscopy with PLSR algorithm could be used for the key influential factors recognization during the liquid preparation process of Tanreqing injection and can be popularized to solve similar problems..
On-line Monitoring of Traditional Chinese Medicinal Powder Blending Process by Using Near-Infrared Spectroscopy

Abstract:OBJECTIVE To establish a rapid method based on near-infrared diffuse reflectance spectroscopy(NIR) to monitor the blending process of traditional Chinese medicinal powder on-line.METHODS NIR spectra were collected during the blending processes of mixtures consisting of Red Peony Root powder and defatted Spine Date Seed powder.Multivariate calibration models based on PLS regression were developed to correlate the collected spectra with the content of Red Peony Root powder.The optimal calibration model was applied to monitor the blending process in real time with noninterference.RESULTS After spectral pretreatment using multiplicative signal correction(MSC),first derivative and Savitzky-Golay filter,Red Peony Root powder contents were quantified using PLS algorithm in the spectral ranges of 5 900-5 950 cm-1,6 700-6 750 cm-1 and 6 950-7 100 cm-1.The correlation coefficient of the calibration model was 0.999 0.The RMSEC,RMSEP and RMSECV were 1.14,1.16 and 1.26,respectively.CONCLUSION It was demonstrated that a miniature near-infrared analyzer could be used to monitor the contents of the constituents and the blending homogeneity of traditional Chinese medicinal powder in real time,and to rapidly determine the end-point of blending process.In addition,it can be used to judge whether the physical state of powder mixture conforms to the standard.Therefore,NIR spectroscopy is considered to be an appropriate PAT tool for end-point control of blending process of traditional Chinese medicinal powder.
[Application of Feedforward Control Strategy Based on Spectra of Raw Materials to Optimize Alcohol Extraction Process of Panax Notoginseng].

Abstract:In this work, we obtained the quality information of Panax notoginseng with near infrared spectroscopy in an efficient way. By combining the quality information of raw materials with process parameters in experimental design, quantitative relationship models were established between material quality information, process parameters and product quality of P. notoginseng, thus realizing the feedforward control of the alcohol extraction process. The quantitative models established have high robustness and satisfactory forecasting ability. With new batches of P. notoginseng introduced for the alcohol extraction process, the quality information of raw materials was rapidly obtained. Based on the established models, the process parameters were optimized according to the expected critical quality attributes (CQA), thereby reducing the fluctuation of product quality caused by raw material variation.
A Feasibility Study on the Non-Invasive Analysis of Bottled Compound E Jiao Oral Liquid Using Near Infrared Spectroscopy

Abstract:The aim of this study was to explore the feasibility of near infrared (NIR) spectroscopy for the quantitative analysis of Compound E Jiao oral liquid, a popular oral TCM preparation, through intact glass vials. Transmission spectra were acquired with a sample holder tailored to the instrument's sample compartment. Total flavonoids, total saponins and soluble solid contents were considered as quality indicators of this oral preparation and calibration models were built using partial least-squares regression (PLSR) algorithm. The sample set was comprised of production samples as well as laboratory samples prepared by a concentrating-diluting method so as to extend the concentration range beyond the production variation. The developed models gave root mean square error of prediction (RMSEP) of 0.084 mg mL(-1) for total flavonoids, 0.014 mg mL(-1) for total saponins and 0.21% for soluble solid contents. To better evaluate the prediction performance, an independent set of ten production samples was introduced for the external validation. The models are compared with those built with the transflectance spectra collected in the destructive way. The results of paired t-test and F-test at a significance level of 0.05 revealed that the two sets of models exhibited comparable model performance. The overall results demonstrate that the proposed non-destructive method can be used as an advantageous alternative in the quality inspection of Compound E Jiao oral liquid production. Application of Near Infrared Spectroscopy Combined with Particle Swarm Optimization Based Least Square Support Vactor Machine to Rapid Quantitative Analysis of Corni Fructus

Abstract:A novel method was developed for the rapid determination of multi-indicators in corni fructus by means of near infrared (NIR) spectroscopy. Particle swarm optimization (PSO) based least squares support vector machine was investigated to increase the levels of quality control. The calibration models of moisture, extractum, morroniside and loganin were established using the PSO-LS-SVM algorithm. The performance of PSO-LS-SVM models was compared with partial least squares regression (PLSR) and back propagation artificial neural network (BP-ANN). The calibration and validation results of PSO-LS-SVM were superior to both PLS and BP-ANN. For PSO-LS-SVM models, the correlation coefficients (r) of calibrations were all above 0.942. The optimal prediction results were also achieved by PSO-LS-SVM models with the RMSEP (root mean square error of prediction) and RSEP (relative standard errors of prediction) less than 1.176 and 15.5% respectively. The results suggest that PSO-LS-SVM algorithm has a good model performance and high prediction accuracy. NIR has a potential value for rapid determination of multi-indicators in Corni Fructus.

Abstract:As a rapid and accurate technique, near infrared (NIR) spectroscopy has been widely used in process monitoring to improve efficiency and ensure quality consistency. In the present work, the application of NIR spectroscopy for on-line quantitative monitoring of the concentration process of Wangbi tablets was investigated. Partial least squares regression (PLSR) models based on the full-range spectra or the key wavelengths selected by the competitive adaptive reweighted sampling (CARS) method were established. The accuracy of the CARS-PLSR method was greater than that of full-spectrum PLSR for three quality parameters (soluble solid contents, paeoniflorin and icariin). For the prediction set samples using the CARS-PLSR models the coefficients of determination (r(2)) and root mean square error [RMSEP (%)] were 0.97 and 1.29 for soluble solid contents, 0.93 and 0.014 for paeoniflorin and 0.85 and 0.009 for icariin, respectively. Overall the results indicated that NIR spectroscopy coupled with CARS-PLSR calibration is a reliable and non-destructive alternative method for on-line monitoring of the concentration process of Wangbi tablets on an industrial scale.

Paeoniflorin, the main active component of Moluodan, was chosen as the quality index. Samples were partitioned into calibration and validation sets by random select and set partitioning based on joint X-Y distances algorithm (SPXY), respectively. Wavelengths for modeling were selected by manual method and competitive adaptive reweighted sampling (Cars) algorithm. Particle swarm optimizationbased least square support vector machines (PSO-LS-SVM) and partial least squares models were both established for quantitative analysis to determine the content of paeoniflorin online. At last, 8 models were obtained according to the combination of these algorithms and the SPXY-Cars-PSO-LS-SVM model had the best quantitative analysis performance compared with the other 7 models. Specifically, in the developed SPXY-Cars-PSO-LS-SVM model, the determination coefficients of the calibration R2c and validation R2v sets were 0.99 and 0.95, respectively, and the root mean square errors of the calibration and validation sets were 0.012 and 0.024 mg/ mL, respectively, and the relative standard errors of the calibration and validation sets were 2.84% and 6.34%, respectively. These results suggested that the appropriate sample partition, wavelength selection, and regression analysis methods in this study, namely, the SPXY-Cars-PSO-LS-SVM algorithm combined with NIR spectroscopy, could be an effective and real-time approach for online monitoring of the extraction process of Moluodan.
Rapid and Non-Destructive Discrimination of Special-Grade Flat Green Tea Using Near-infrared Spectroscopy

Abstract:Special-grade green tea is a premium tea product with the best rank and high value. Special-grade green tea is normally classified by panel sensory evaluation which is time and sample costly. Near-infrared spectroscopy is considered as a promising rapid and non-destructive analytical technique for food quality evaluation and grading. This study established a discrimination method of special-grade flat green tea using Near-infrared spectroscopy. Full spectrum was used for partial least squares (PLS) modelling to predict the sensory scores of green tea, while specific spectral regions were used for synergy interval-partial least squares (siPLS) modelling. The best performance was achieved by the siPLS model of MSC + Mean Centering pretreatments and subintervals from 15 intervals. The optimal model was used to discriminate special-grade flat green tea with the prediction accuracy of 97% and 93% in the cross-validation and external validation respectively. The chemical compositions of green tea samples were also analyzed, including polyphenols (total polyphenols, catechins and flavonol glycosides), alkaloids and amino acids. Principal components analysis result showed that there is potential correlation between specific spectral regions and the presence of polyphenols and alkaloids. Thus, NIR technique is a practical method for rapid and non-destructive discrimination of special-grade flat green tea with chemical support.
Application of Near Infrared Spectroscopy for Rapid Analysis of Intermediates of Tanreqing Injection.

Abstract:A method for rapid quantitative analysis of four kinds of Tanreqing injection intermediates was developed based on Fourier transform near infrared (FT-NIR) spectroscopy and partial least squares (PLS) algorithm. The NIR spectra of 120 samples were collected in transflective mode. The concentrations of chlorogenic acid, caffeic acid, luteoloside, baicalin, ursodesoxycholic acid (UDCA), and chenodeoxycholic acid (CDCA) were determined with the HPLC–DAD/ELSD as reference method. In the PLS calibration, the NIR spectra were pretreated with different methods and the number of PLS factors used in the model calibration was optimized by leave-one-out cross-validation. The performance of the final PLS models was evaluated according to the root mean square error of calibration (RMSEC), root mean square error of cross-validation (RMSECV), root mean square error of prediction (RMSEP), BIAS, standard error of prediction (SEP), and correlation coefficients (R). The R values in the prediction sets were all higher than 0.93, and the SEPs for the 6 compounds are 1.18, 6.02, 2.71, 155, 126, 30.0mg/l, respectively. The established models were used for the liquid preparation process analysis of Tanreqing injection in three batches, and a model updating method was proposed for the long-term usage of the established models. This work demonstrated that NIR spectroscopy is more rapid and convenient than the conventional methods to analyze the intermediates of Tanreqing injection, and the presented method is helpful to the implementation of process analytical technology (PAT) in pharmaceutical industry of Chinese Medicines Injections.
