Abstract:The sensory features of white peony teas (WPTs) significantly change with storage age; however, their comprehensive associations with composition are still unclear. This study aimed to clarify the sensory qualityrelated chemical changes in WPTs during storage. Liquid chromatography-tandem mass spectrometry based on widely targeted metabolomics analysis was performed on WPTs of 1-13 years storage ages. Weighted gene coexpression network analysis (WGCNA) was used to correlate metabolites with sensory traits including color difference values and taste attributes. 323 sensory trait-related metabolites were obtained from six key modules via WGCNA, verified by multiple factor analysis. The decline and transformation of abundant flavonoids, tannins and amino acids were related to the reduced astringency, umami and increased browning of tea infusions. In contrast, the total contents of phenolic acids and organic acids increased with storage. This study provides a highthroughput method for the association of chemical compounds with various sensory traits of foods.

Abstract:The aim of this study was to reveal the molecular basis of aroma changes during storage of An tea (AT). The key volatile compounds in AT were screened using SPME-GC-MS and SPE-GC-MS analytical techniques in combination with odor activity value (OAV) and flavor dilution factor (FD). The results showed that with the increase of storage time the stale and woody aromas were revealed. Esters, acids and hydrocarbons are the main types of volatile compounds in AT, and their content accounts for 52.69%-61.29% of the total volatile compounds. The key volatile compounds with stale and woody aromas during AT storage were obtained by OAV value and FD value, namely ketoisophorone (flavor dilution factor, FD=64), linalool oxide C (FD=64), 1-octen-3-ol (OAV>1, FD=32), 1,2-dimethoxybenzene (FD=16), naphthalene (OAV>1, FD=32), 3,4-dimethoxytoluene (FD=16), and 1,2,3-trimethoxybenzene (FD=8). Our research provides a scientific basis and insights for the improvement of quality during the storage of dark tea.

Abstract:The fresh aroma of Longjing tea is vulnerable to unfavorable storage conditions. However, limited research has addressed effective solutions apart from low-temperature storage. This study aimed to investigate the impact of oxygen scavenger on aroma quality of packaged Longjing tea samples at elevated storage temperatures. As a result, the utilization of oxygen scavenger could effectively mitigate aroma deterioration of Longjing tea caused by elevated temperature during storage, resulting in a decrease in the stale odor scores by more than 3.0. The utilization of oxygen scavenger achieved aroma-preserving effect by inhibiting the accumulation of key stale odor compounds and maintaining the levels of volatiles related to freshness aroma. The key volatile contributors to stale odor are primarily ketones and alcohols resulting from thermal carotenoids degradation/lipid oxidation, which exhibit woody or fatty odors. These findings provide essential theoretical principles for improving Longjing tea preservation technology.
Study on Variation of the Content of Main Chemical Components and Quality of Puer-tea during Storage

Abstract:The quality and the content of main chemical components in puer-tea under different storage condition were analyzed. Results indicated that the period showing best quality was advanced with the rising of temperature and water-content during storage. The best quality of 55℃/12% treatment was formed at 45 days storage and the best quality of 37℃/9% treatment was formed at 135 days storage. The contents of tea polyphenols and soluble sugar were decreased with the increasing of time and temperature of storage. The content of amino acid reduced with the rising of temperature and time, but had no related with the water content in Puer-tea.

Abstract:The increasing demand for tea consumption calls for the development of more products with distinct characteristics. The sensory quality of tencha is significantly determined by innate differences among tea cultivars. However, the correlations between the chemical composition and sensory traits of tencha are still unclear. To enhance the understanding of the flavor formation mechanism in tencha and further to develop new cultivars resources, we investigated non-volatiles and volatile metabolites as well as sensory traits in tencha from different tea cultivars (Camellia sinensis cv. Yabukita, Longjing 43 and Baiye 1); the relationships between the flavor traits and non-volatiles/volatiles were further evaluated by partial least squares - discriminate analysis (PLS-DA), multiple factor analysis (MFA) and multidimensional alignment (MDA) analysis. A total of 64 non-volatiles and 116 volatiles were detected in all samples, among which 71 metabolites were identified as key flavor-chemical contributors involving amino acids, flavonol glycosides, flavones, catechins, ketones, alcohols, hydrocarbons, aldehydes, esters and acids. The levels of taste-related amino acids, flavonol glycosides and gallic acid varied significantly among the tencha samples made from different tea cultivars. All the samples exhibited typical quality characteristics of tencha. The tencha from Camellia sinensis cv. Longjing 43 and Camellia sinensis cv. Baiye 1 (cultivated in the open) exhibited higher levels of amino acids and gallic acid, which were associated with the umami taste and mellow taste of tea infusion. Abundant flavonol glycosides were related to the astringency, while partial tri-glycosides specifically quercetin-3-O-galactoside-rhamnoside-glucoside and total of flavonol galactoside-rhamnoside-glucoside were associated with mellow taste. The floral alcohols were identified as significant contributors to the refreshing aroma traits of tencha. The green, almond-like, acidic and fruity odorants were associated with a green and fresh aroma, while the green, cheesy and waxy odorants such as ketones, esters, acids and hydrocarbons were associated with seaweed-like aroma. This study provides insight into sensory-related chemical profiles of tencha from different tea cultivars, supplying valuable information on flavor and quality identification for tencha.

Abstract:In this study, ripened Pu-erh teas were obtained by different mix strains pile co-fermentation. A total of 135 volatile compounds were annotated using HS-SPME-GC-MS, of which the highest content was alcohols (54.26%), followed by esters (16.73%) and methoxybenzenes (12.69%). The total content of these compounds increased with the duration of fermentation and the diversity of the mixed strains. The flavor profiles of 27 volatile compounds with rOAVs > 1 were analyzed by relative odor activity values (rOAVs). Alcohols in ripened Pu-erh tea mainly contribute flower and fruit sweet flavors, while methoxybenzenes mainly contribute musty and stale flavors. The ripened Pu-erh tea fermented by Saccharomyces: Rhizopus: Aspergillus niger mixed in the ratio of 1:1:1 presented the remarkable flavor characteristics of flower and fruit sweet flavor, and having better coordination with musty and stale flavor. This study demonstrated the content changes of ripened Pu-erh tea's flavor compounds in the fermentation process, and revealed the optimal fermentation time. This will be helpful to further understand the formation mechanism of the characteristic flavor of ripened Pu-erh tea, and guide the optimization of the fermentation process of ripened Pu-erh tea.

Abstract:The aroma of Keemun black tea (KBT) changes during storage. We investigated key aroma volatiles of fresh KBT (FKBT) and KBT stored for 1 year. Through gas chromatography–olfactometry–mass spectrometry/aroma extract dilution analysis (GC-O-MS/AEDA), 27 aroma volatiles with a flavor dilution (FD) value ≥16 were quantitated. In odor activity value (OAV) analysis, the two samples had nearly the same key aroma volatiles; (Z)-methyl epijasmonate was the exception. Dimethyl sulfide, 3-methylbutanal, 2-methylpropanal, and linalool had especially high OAVs. Except for β-damascenone, volatiles with OAVs > 1 had higher concentrations in FKBT, which revealed that most key aroma compounds were lost during storage. Sweet, malty, floral, and green/grassy aromas corresponded directly to certain compounds. Lastly, the addition test indicated that the addition of several key aroma volatiles decreasing during storage could enhance the freshness of KBT aroma, which may be a potential to control the aroma style of KBT or other teas in industry.

Abstract:Crude Pu-erh tea (CPET) is distinct from ripened Pu-erh tea, and usually needs a long-term aging under natural conditions, which can make it taste more mellow. In the present study, metabolomics integrated with sensory evaluation was used to investigate the key astringency compounds of CPET during storage. Metabolomics results showed that these CPET samples could be classified into two groups, one was storage from 1 to 5 years, and the other was 6–8 years. Meanwhile, non-galloylated catechins, hydroxycinnamic acids, flavonol glycosides, and gallotannins were marker compounds, which were responsible for distinguishing short and long aging time. Correlation coefficients between astringency scores and (–)-epigallocatechin, (–)-epicatechin, quercetin-glucose, myricetin-glucose, and (–)-gallocatechin gallate were 0.826, 0.821, 0.815, 0.811, and 0.718, respectively. The astringency contribution of galloylated catechins and flavonol glycosides was also investigated by treating CPET infusion with tannase and β-glycosidase. The tannase treatment greatly reduced the contents of galloylated catechins, and correspondingly decreased the astringency scores. β-Glycosidase could hydrolyze the flavonol glycosides into flavonol aglycones, but the astringency intensity of CPET infusion was not affected. These results suggested hydrolysis of galloylated catechins was the key reason for astringency decrease during storage of CPET, while flavonol glycosides comparatively contributed less to the astringency.

Abstract:This paper studied the influence of storage duration on the flavor profile of white tea in detail, with samples produced between 2020 and 2023. Sensory evaluation was performed by quantitative descriptive analysis (QDA), followed by an in-depth aroma components analysis employing an electronic nose (E-nose), headspace gas chromatography-ion mobility spectrometry (HS-GC-IMS), and headspace solid-phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS). The QDA findings revealed a gradual transition in the flavor profile of white tea during storage, shifting from sweet, fruity, and floral to more herbal and stale characteristics. E-nose could well distinguish white tea with different storage times. A total of 55 and 53 volatile compounds were identified by HS-GC-IMS and HS-SPME-GC-MS, respectively. The orthogonal partial least squares-discriminant analysis models, based on HS-GC-IMS (R2Y = 0.998, Q2 = 0.987) and HS-SPME-GC-MS (R2Y = 0.984, Q2 = 0.993), successfully distinguished white tea samples stored for different storage times. Furthermore, 14 and 8 key compounds were screened based on the double variable criterion of one-way analysis of variance (p < 0.05) and variable importance in projection (VIP) >1.2, and their content changes were also compared. It is the gradual decrease of important aroma components such as 2-hexenal, 2-methyl-2-hepten-6-one, linalool, and geraniol, which are positively correlated with sweet, fruity, and floral aromas, and the gradual increase of hexanoic acid, thiophene, propanoic acid, dimethyl disulfide, and borneyl acetate, which are positively correlated with herbal and stale flavor, that leads to the changes in flavor and aroma of white tea during storage. The results of the study provided a reference for elucidating the aroma characteristics of white tea at different storage times as well as a theoretical basis for the quality control of white tea.
The flavor substances changes in Fuliang green tea during storage monitoring by GC–MS and GC-IMS

Abstract:To study the effect of storage (for 0, 3, 6, and 12 months) on the flavor of green tea (GT), we monitored the volatile organic compounds (VOCs) in GT through gas chromatography (GC) combined with ion mobility spectrometry and headspace solid-phase micro extraction, GC–MS (mass spectrometry). Then, relative odor activity value (ROAV) was applied to analyze the aroma contribution of the VOCs. During storage, the polyphenol and caffeine contents gradually decreased from 22.38 % to 18.51 % and from 4.37 % to 3.74 %, respectively, and the total soluble sugar first increased and then decreased (from 4.89 % to 7.16 % and then 5.02 %). Although the total free amino acid contents showed a fluctuating trend, the content of cysteamine increased gradually. The contents of VOCs with positive contribution to GT aroma, including linalool, geraniol, nonanal, and 6-methyl-5-hepten-2-one, decreased. They also contributed less in the ROAV after storage. The ROAVs of nonanal, linalool, and geraniol decreased from 3.37 to 0.79, from 100 to 38.21, and from 2.98 to 1.8, respectively, after 12 months of storage. Principal component analysis can be used to identify the samples with different storage durations based on these data. Given the increase in amount of cysteamine and decrease in that of linalool oxide, oxidation may be not the only factor responsible for tea quality in storage.

Abstract:Compressed white tea (CWT) is a reprocessed tea of white tea. Long-term storage has greatly changed its aroma characteristics, but the material basis and transformation mechanism of its unique aroma are still unclear. In this study, flavor wheel, headspace gas chromatography ion mobility spectroscopy, chemometrics and microbiomics were applied to study the flavor evolution and important aroma components during long-term storage of CWT, and core functional bacteria were screened. During long-term storage, the aroma of CWT gradually changed from sweet, fruity and floral to stale flavor, woody and herbal. A total of 56 volatile organic compounds (VOCs) were identified, 54 of which were significantly differences during storage. The alcohols content was the highest during 1-5 years of storage, the esters content was the highest during 7-13 years of storage, and the aldehydes content was the highest during 16 years of storage. Twenty-nine VOCs were identified as important aroma components, which were significantly correlated with 6 aroma sub-attributes (P < 0.05). The functional prediction of bacterial community reminded that bacterial community could participate in the transformation of VOCs during storage of CWT. Twenty-four core functional bacteria were screened, which were significantly associated with 29 VOCs. Finally, 23 characteristic differential VOCs were excavated, which could be used to identify CWT in different storage years. Taken together, these findings provided new insights into the changes in aroma characteristics during storage of CWT and increased the understanding of the mechanism of characteristic aroma formation during storage.

Abstract:We aim to improve the product quality of GABA raw Pu-erh tea during development and processing. In this study, headspace solid-phase microextraction gas chromatography–mass spectrometry technology combined with relative odor activity evaluations was used to compare the volatile compounds of GABA sun-dried green tea and GABA raw Pu-erh tea. Sensory evaluation showed a higher aroma score of GABA raw Pu-erh tea than that of GABA sun-dried green tea, with significant differences in aroma type and purity. A total of 147 volatile compounds of 13 categories were detected, which differed in composition and quantity between the two teas. 2-Buten-1-one,1-(2,6,6-trimethyl-1,3-cyclohexadien-1-yl)-,(E)- and beta.-myrcene largely contributed to the aroma formation of both teas. Five volatile compounds were screened as potential markers for tea aroma. Metabolic pathway analysis showed that monoterpenoid biosynthesis may be beneficial to the formation of flowery and fruity aromas in the teas. We suggest that the findings of this study may provide important guidance for the processing and optimization of GABA tea.

Impact of storage time on non-volatile metabolites and fungal communities in Liupao tea using LC-MS based non-targeted metabolomics and high-throughput sequencing

Abstract:Long-term storage of Liupao tea (LPT) is usually believed to enhance its quality and commercial value. The non-volatile metabolites variations and the fungal succession play a key role for organoleptic qualities during the storage procedure. To gain in-depth understanding the impact of storage time on the quality of LPT, two different brands of LPT with different storage time, including Maosheng LPTs (MS) with 0, 5, 10 and 15 years and Tianyu LPTs (TY) with 0, 3, 5, 8 and 10 years, were resorted to investigate the changes of non-volatile metabolites and fungi as well as their correlation by multi-omics. A total of 154 and 119 differential metabolites were identified in these two different brands of MS and TY, respectively, with the aid of high-performance liquid chromatography with quadrupole-time-of-flight mass spectrometry. In both categories of LPTs, the transformation of differential metabolites in the various stages referred to the formation of alkaloids, increase of organic acids, biosynthesis of terpenoids as well as glycosylation and methylation of flavonoids. Thereinto, glycosylation and methylation of flavonoids were the critical stages for distinguishing MS and TY, which were discovered in MS and TY stored for about 10 and 8 years, respectively. Moreover, the results of high-throughput sequencing showed that the key fungal genera in the storage of LPTs consisted of Eurotium, Aspergillus, Blastobotrys, Talaromyces, Thermomyces and Trichomonascus. It was confirmed on the basis of multivariate analysis that the specific fungal genera promoted the transformation of metabolites, affecting the tea quality to some extent. Therefore, this study provided a theoretical basis for the process optimization of LPT storage.

Abstract:BackgroundPu-erh tea aroma is the comprehensive result of various volatile compounds (VCs). More than 1000 VCs were reported in Pu-erh tea. Pu-erh tea aroma was widely studied using sensory evaluation, instrumental analysis, odor activity value (OAV), aroma recombination, and omission test. The activities of microorganism is the key to the formation of Pu-erh tea aroma.Scope and approachThe main purpose of this review is to elaborate on the volatile components contributing to the unique aroma, methods used for VCs extraction and evaluation, and metabolic mechanism of key odor-active compounds of Pu-erh tea. The reported VCs, odor-active compounds, and their extraction methods were summarized. Sensory evaluation, extraction methods, and analytical methods were elaborated concerning aroma analysis. The possible metabolic pathways of key odor-active compounds and related microorganisms were combed.Key findings and conclusionsThe composition of VCs varies greatly with different extraction methods. Combining multiple extraction methods will be an effective strategy to fully extract the VCs of Pu-erh tea. Sensory evaluation provides a fast and intuitive strategy to determine the aroma profile or aroma attributes. GC–O–MS combines with OAV, aroma recombination, and omission test can not only realize quantitative and qualitative detection but also realize the screening of key aroma compounds. E-nose combined with multivariate statistical provides an easier strategy for distinguishing Pu-erh tea aroma. Most VCs are formed by the activities of microorganisms, but their exact metabolic mechanisms are still unclear, which needs further exploration.

Abstract:An tea (AT) is a dark tea with a unique flavor that affects the quality of storage time. However, changes in the sensory quality and chemical composition of AT during storage are unknown. The present study performed sensory evaluation and metabolomic analysis of AT at seven storage time points. The results indicated that the tea liquor color changed from orange to dark red during storage, and the astringency intensity gradually decreased concomitant with the appearance of a stale taste. Seventy differential metabolites were identified by untargeted liquid chromatography coupled with mass spectrometry (LC–MS) including catechins, flavonols and their glycosides, free amino acids, alkaloids, and phenolic acids and their derivatives. Targeted analysis demonstrated that quercetin, quercetin 3-O-glucuronide, quercetin 3-O-rutinoside, glycine, aspartic acid, alanine, serine, arginine, threonine, tyrosine, theanine, GABA and isoleucine were highly correlated with storage time (r < −0.8 or r > 0.8). Furthermore, linear regression analysis confirmed that these 13 substances were good predictors of AT storage time (correlation coefficient of 0.8767). The present study reveals the changes in the components during storage of AT and provides a theoretical basis for quality improvement of stored dark tea.

Abstract:The differences of volatile substances of 4 kinds of An-tea samples, made in 2007 (high-aging sample, HAS), 2012 (middle-aging sample, MAS), 2017 (low-aging sample, LAS) and 2018 (control check, CK) were studied by headspace solid phase microextraction combined with gas chromatography-mass spectrometry. A total of 64 compounds, including 13 ketones, 12 aldehydes, 4 alcohols, 8 esters, 2 phenols, 13 hydrocarbon compounds, 7 nitrogenous compounds, 2 ethers and 3 heterocyclic compounds, were identified. Among them, there were 34 compounds in HAS, 41 compounds in MAS, 39 compounds in LAS, and 31 compounds in CK, indicating that different kinds and contents of volatile aroma components contribute significantly to the quality of tea, which leads to the unique aroma quality of tea samples with different storage time. Linalool and β-ionone are the common aroma components of 4 kinds of An-tea, and play an important role in the formation of An-teas’ aroma. In addition, with the extension of storage time, the relative content of alcohols in An-tea decreased gradually, while the relative content of esters increased, which is probably due to the esterification reaction between alcohols and acids.
Characterisation of the volatile compounds profile of Chinese pan-fried green tea in comparison with baked green tea, steamed green tea, and sun-dried green tea using approaches of molecular sensory science

Abstract:Pan-fried green tea (PGT) is an easily acceptable tea drink for general consumers. In this study, volatile profiles and characteristic aroma of 22 representative Chinese PGT samples were extracted using stir bar sorptive extraction (SBSE) and analysed by gas chromatography-mass spectrometry (GC-MS), gas chromatography-olfactometry (GC-O) analysis, and odour activity value (OAV) calculations. In total, 88 volatile compounds were identified. Alcohols (45%), esters (19%), and ketones (16%) were the dominant volatiles, and geraniol (484.8 μg/kg) was the most abundant volatile component in PGT, followed by trans-β-ionone and linalool. In addition, the differences of aroma characteristics among PGT and other three types of green tea, namely baked green tea, steamed green tea, and sun-dried green tea, were also observed using partial least squares discriminant analysis (PLS-DA) and heatmap analysis, and it was found that β-myrcene, methyl salicylate, ( E )-nerolidol, geraniol, methyl jasmonate were generally present at higher content in PGT. This is the first comprehensive report describing the volatile profiles of Chinese PGT, and the findings from this study can advance our understanding of PGT aroma quality, and provide important theoretical basis for processing and quality control of green tea products.
