Abstract:Prebiotics, probiotics, and synbiotics have been successfully used in dietary supplements to achieve aquatic animal health and therefore increase the quality and sustainability of fish production. In the present study, four hundred and twenty common carps, Cyprinus carpio ( 25.37 ± 0.22 g ; mean ± SE ) were randomly attributed to seven treatments, fed with diets containing PrimaLac (probiotic), inulin (prebiotic), and Biomin Imbo (synbiotic), 1 and 2 g/kg for each supplement. After 60 days of feeding, an increase in final weight (FW), weight gain (WG), specific growth rate (SGR), and survival rate (SR) was recorded in the treatment groups compared to that of the control ( P < 0.05 ). The food conversion ratio (FCR) in the treatment groups significantly decreased ( P < 0.05 ). The treated groups showed significant improvements in serum immune parameters: lysozyme (LYZ), alternative complement (ACH50), total immunoglobulin (total Ig), and myeloperoxidase (MPO) ( P < 0.05 ). Feeding fish with supplemented diets significantly showed enhanced antioxidant status: catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GPx). Malondialdehyde (MDA) activity was significantly lower in fish fed dietary additives ( P < 0.05 ). Compared with the control group, enzyme parameters revealed that supplementation could significantly decrease alanine transaminase (ALT), alkaline phosphatase (ALP), and aspartate transaminase (AST) ( P < 0.05 ). The biochemical parameters including triglyceride (TRIG), cholesterol (CHO), glucose (GLU), and cortisol (CORT) decreased with dietary supplementation ( P < 0.05 ). Total protein (TP) increased in fish fed experimental diets ( P < 0.05 ). Fish fed pro, pre, and synbiotic exhibited significantly higher digestive enzymes (amylase, lipase, and protease) ( P < 0.05 ). Skin mucus parameters (total Ig, ACH50, LYZ, protease, and ALP) were significantly enhanced in groups fed dietary additives ( P < 0.05 ). Thus, the best recommended inclusion level of supplementation is 2 g/kg for inulin and 2 g/kg for PrimaLac and doses of 1 or 2 g/kg for Biomin Imbo. Dietary PrimaLac, inulin, and Biomin Imbo could be recommended as beneficial feed additives to enhance growth performance, innate immune and antioxidant systems, and promoted biochemical parameters and digestion of common carp; also, the effect of dietary synbiotic was superior to that of prebiotic and probiotic.

Abstract:The effects of dietary Bacillus coagulans (MTCC 9872), Bacillus licheniformis (MTCC 6824) and Paenibacillus polymyxa (MTCC 122) supplementation on growth performance, non-specific immunity and protection against Aeromonas hydrophila infection were evaluated in common carp, Cyprinus carpio fry. Laboratory maintained B. coagulans, B. licheniformis and P. polymyxa were used to study antagonistic activity against fish pathogenic bacteria by agar well diffusion assay. Healthy fish fry were challenged by this bacterium for determination of its safety. Fish were fed for 80 days with control basal diet (B0) and experimental diets containing B. coagulans (B1), B. licheniformis (B2) and P. polymyxa (B3) at 10(9) CFU/g diet. Fish fry (mean weight 0.329 ± 0.01 g) were fed these diets and growth performance, various non-specific immune parameters and disease resistance study were conducted at 80 days post-feeding. The antagonism study showed inhibition zone against A. hydrophila and Vibrio harveyi. All the probiotic bacterial strains were harmless to fish fry as neither mortality nor morbidities were observed of the challenge. The growth-promoting influences of probiotic supplemented dietary treatments were observed with fish fry and the optimum survival, growth and feed utilization were obtained with P. polymyxa (B3) supplemented diet. Study of different non-specific innate immunological parameters viz. lysozyme activity, respiratory burst assay and myeloperoxidase content showed significant (p < 0.05) higher values in fish fry fed B3 diet at 10(9) CFU/g. The challenge test showed dietary supplementation of B. coagulans, B. licheniformis and P. polymyxa significantly (p < 0.05) enhanced the resistance of fish fry against bacterial challenge. These results collectively suggests that P. polymyxa is a potential probiotic species and can be used in aquaculture to improve growth, feed utilization, non-specific immune responses and disease resistance of fry common carp, C. carpio.

Abstract:The present trial investigates the effects of soil-derived Streptomyces chartreusis KU324443 as a probiotic on growth performance, mucosal and serum immune parameters, and immune and antioxidant-associated gene expression in common carp (Cyprinus carpio). In a two-month experiment, carps were fed with varying levels of S. chartreusis KU324443 (0 (control), 105 (S1), 106 (S2), and 107 (S3) CFU/g). Then, performance, skin mucus, and serum immune parameters besides immune and antioxidant-related gene expression (IL1 and Lyz, CAT and SOD) were measured. Fish fed S. chartreusis-supplemented diet showed a significant increase in growth performance parameters ( P < 0.05 ) regardless of inclusion levels. Including different doses of S. chartreusis significantly increased serum total Ig and lysozyme activity compared to those fed the control diet ( P < 0.05 ). While no significant difference was noticed in the case of skin mucus total Ig ( P > 0.05 ), lysozyme activity showed a significant increase in S. chartreusis-treated carps ( P < 0.05 ). No significant differences ( P > 0.05 ) were noticed in the case of serum antioxidant enzyme activity (CAT, SOD, and GPx) of S. chartreusis-fed carps and control. However, fish fed the control diet recorded the highest SOD and GPx enzyme activity in skin mucus of common carp, while no significant differences were noticed in the case of mucus CAT enzyme activity in common carp ( P > 0.05 ). Gene expression studies revealed noticeable alterations between treated fish and control. Fish in S3 treatment had significantly higher CAT, SOD, Lyz, and IL1 expression ( P < 0.05 ). These results confirmed the beneficial effects of a soil-derived probiotic (S. chartreusis) on the performance and health of common carp.

Modulatory effects of laurel-leaf cistus (Cistus laurifolius) ethanolic extract on innate immune responses and disease resistance in common carp (Cyprinus carpio)

Abstract:Medicinal herbs are used for growth promotion, disease control and other health benefits in aquaculture industry. Here, we examined the effect of dietary laurel-leaf cistus (Cistus laurifolius) ethanolic extract on growth performance, digestive enzyme activity, haematological profile and nonspecific immune responses in common carp (Cyprinus carpio). In addition, resistance against Aeromonas hydrophila infection was examined. Common carp was fed diets containing 0 (Control), 0.1 (CL0.1), 0.5 (CL0.5) and 1 (CL1) g kg-1 laurel-leaf cistus extract for 45 days. After 30 days, superoxide anion production (SAP) increased in CL0.1 and CL0.5 fish groups and at the end of the study all experimental fish groups had higher SAP compared to that of the control (P ˂ 0.05). Lysozyme activity (LA) was elevated in CL0.5 and CL1 treated groups on 30th day (P < 0.05), and this increase was only observed in C0.1 fish group at the end of study compared to control (P ˂ 0.05). Myeloperoxidase activity was significantly increased in CL0.5 and CL1 fish groups at the end of study. IL-1βgene expression was significantly increased in treated fish in a dose-depended manner. Similar results were observed for transcription of IL-6 and IL-8 (P < 0.05). Anti-inflammatory cytokines, IL-10 and TGF-β were highly up-regulated in the intestine and head kidney of CL treated fish groups compared to control (P < 0.05). At the end of experiment, significantly higher final body weight, weight gain, and specific growth rate were obtained in CL0.1 treated fish group compared to control. However, growth was negatively affected in CL1 fish group (P < 0.05). CL1 fish group had also a significantly higher FCR. Amylase activity was significantly increased in all experimental fish groups compared to control (P ˂ 0.05). Trypsin activity was decreased in CL0.1 and CL1 fish groups (P ˂ 0.05). WBC and RBC were significantly increased (P ˂ 0.05) in CL0.5 and CL1 fish groups, whereas haemoglobin, haematocrit, mean cell, mean cell haemoglobin contents were no significantly changed among control and treatment groups. Result of challenge test with A. hydrophila exhibited that survival rate in all treatment groups was significantly higher than that of control. These findings demonstrated that laurel-leaf cistus at 0.1 g kg-1 can be a suitable candidate for growth promotion, immune system induction and infection control in fish.
Bradykinin B 1 receptor blocks PDGF-induced mitogenesis by prolonging ERK activation and increasing p 27 Kip 1

First published March 6, 2002; 10.1152/ ajpcell.00289.2001.—The mechanism by which the bradykinin B1 receptor (B1R) inhibits platelet-derived growth factor (PDGF)-stimulated proliferation was investigated in cultured rat mesenteric arterial smooth muscle cells. The B1R agonist des-Arg9-bradykinin (DABK) was found to inhibit PDGF-mediated activation of the cyclin E-cyclin-dependent kinase 2 (Cdk2) complex and to prevent hyperphosphorylation of retinoblastoma protein. DABK did not inhibit upregulation of cyclin E expression but increased expression of the Cdk2 inhibitor p27Kip1 and the association of p27Kip1 with the cyclin E-Cdk2 complex. In addition, DABK inhibited the PDGF-stimulated expression of cyclin D that would otherwise siphon p27Kip1 away from inhibition of cyclin E-Cdk2. The signaling mechanism by which DABK regulated p27Kip1 was explored. DABK was found to stimulate the activity of mitogen-activated protein kinase kinase (MEK) and extracellular signal-regulated kinase (ERK) and to prolong activation of MEK and ERK by PDGF. Inhibition of ERK activation with the MEK inhibitors PD-98059 and U-0126 as well as the Src family kinase inhibitor PP2 completely blocked the effect of DABK to increase p27Kip1 and partially reversed the DABK-mediated inhibition of PDGF-stimulated proliferation. These studies demonstrate that the B1R inhibits PDGF-stimulated mitogenesis in part by prolonged activation of ERK leading to increased expression of p27Kip1.

Abstract:Natural products have gained considerable attention for improving fish growth performance and immunity to enhance disease resistance. This study evaluated the effect of dandelion polysaccharides (DP) on skin mucosal immune parameters, immune-related gene expression, and susceptibility to pathogen challenge in the Common carp Cyprinus carpio. Diets containing four different concentrations of DP (g Kg−1):0 g [basal diet], 0.5 g [D1], 1.5 g [D2], 2.5 g [D3], and 4.0 g [D4] were fed to the carp (average weight: 13.92 ± 0.83 g) for eight weeks. Growth parameters were analyzed four and eight weeks after feeding. Immunological, hematological, and antioxidant parameters were examined eight weeks post-feeding. Growth performance was significantly higher on D3, with a final weight gain of 71.48 ± 1.57 g and a specific growth rate of 3.06 ± 0.12. Among hematological parameters examined, erythrocyte, hematocrit, and mean corpuscular volume (MCV) levels were significantly higher in D3. Skin mucosal immune parameters, such as lysozyme (31.04 ± 1.02 Unit mL−1), alkaline phosphatase (122.6 ± 3.8 IU L-1), and protein level (10.6 ± 0.74 mg mL−1) were significantly higher in D3, while peroxidase activity was higher in D4. Furthermore, SOD activity was higher in D2–D3, whereas catalase activity was higher in D2–D4 (P < 0.05) than in the control. Malondialdehyde level decreased significantly in D3 (5.43 ± 0.36 nmol mL−1); whereas, serum ALT and AST levels were significantly lower on D2-D4. Intestinal tight-junction-related genes ZO-1 and Claudin 7 were significantly higher in the DP-fed groups; however, DP had no significant effect on claudin 3. Occludin expression was higher (p < 0.05) on D3 only. Pro-inflammatory cytokines (IL-1β and TNF-α) and IFN–γ strongly upregulated in the head kidney at D3. Conversely, the expression of the anti-inflammatory cytokine interleukin-10, HSP70, and TOR were considerably downregulated in D3. Fish from D3 exhibited markedly higher relative post-challenge survival (66.67%) against Aeromonas hydrophila challenge. The results of the present study suggest that dietary supplements of DP at 2.5 g kg−1 can significantly improve the growth performance, skin mucosal, and serum antioxidant parameters, and strengthen the immunity of C. carpio. Therefore, DP is a promising food additive for carp aquaculture.

Influence of age on Cu/Zn-superoxide dismutase and indole 2,3-dioxygenase activities in rat tissues.

Abstract:The aim of this study was to investigate in vitro the variations with age of the activities of the two antioxidant enzymes Cu/Zn-superoxide dismutase (SOD) and indole 2,3-dioxygenase (IDO) in metabolically active tissues of rats of various ages. In rats aged one week and 2-3 months the highest Cu/Zn-SOD activity was found in the liver and the lowest in the small intestine. At 12 and 18 months of age, the activity was higher in the brain and kidneys, when compared to the small intestine, lungs and liver. Cu/Zn-SOD activity decreased significantly after 2-3 months of age with advancing age in all tissues examined. In newborn rats IDO activity was present only in the small intestine. In the group of rats aged 2-3 months, the highest specific activity was observed in the small intestine and the lowest in the lungs and kidneys, whereas at 12 months of age, the highest IDO activity was found in the brain, with kidneys presenting the lowest activity. At 18 months, IDO returned to be more elevated in the small intestine. At 12 months of age the values of IDO in the tissues varied slightly, while at 18 months similar activities were found between the lungs and brain and between the small intestine and kidneys. In relation to age, IDO specific activity declined in the small intestine, after 2-3 months of age. In the lungs, the activity remained unchanged; in the brain and in the kidneys activity decreased significantly from 2-3 to 18 months of age. In conclusion, this study demonstrates an age-related decline in Cu/Zn-SOD and IDO activities, the two enzymes responsible for scavenging O2*-.
Dietary Lactobacillus helveticus and Gum Arabic improves growth indices, digestive enzyme activities, intestinal microbiota, innate immunological parameters, antioxidant capacity, and disease resistance in common carp

Abstract:The present study aimed at determining the effects of Lactobacillus helveticus (LH), Gum Arabic (GA; natural prebiotic), and their combination as synbiotic on growth performance, digestive enzymes activity, gut microbiota, innate immunity status, antioxidant capacity, and disease resistance against Aeromonas hydrophyla in common carp, Cyprinus carpio for 8 weeks. For this, 735 common carp juveniles (Mean ± standard deviation; 22.51 ± 0.40 g) were fed with 7 different diets including basal diet (C), LH1 (1 × 107 CFU/g), LH2 (1 × 109 CFU/g), GA1 (0.5%), GA2 (1%), LH1+GA1 (1 × 107 CFU/g + 0.5%), and LH2+GA2 (1 × 109 CFU/g + 1%) for 8 weeks. Dietary supplementation with GA and/or LH significantly increased growth performance, WBC, serum total immunoglobulin, superoxide dismutase and catalase activities, skin mucus lysozyme and total immunoglobulin and intestinal lactic acid bacteria. Whereas there were significant improvements in various parameters tested in different treatments, the highest improvement in growth performance, WBC, monocyte/neutrophil percentages, serum lysozyme, alternative complement, glutathione peroxidase and malondialdehyde, skin mucosal alkaline phosphatase, protease, and immunoglobulin, intestinal total bacterial count, protease and amylase activities were observed in the synbiotic treatments, particularly LH1+GA1. After an experimental infection with Aeromonas hydrophila, all experimental treatments exhibited significantly higher survival, compared to the control treatment. The highest survival was related to the synbiotic (particularly LH1+GA1), followed by prebiotic, and probiotic treatments. Overall, synbiotic containing 1 × 107 CFU/g LH + 0.5% GA can improve growth rate and feed efficiency in common carp. Moreover, the synbiotic can improve the antioxidant/innate immune systems and dominate lactic acid bacteria in the fish intestine that may be the reasons of the highest resistance against A. hydrophila infection.

Abstract:In this study, the alleviative effects of poly-β-hydroxybutyrate (PHB) in bioflocs on oxidative stress, inflammation and apoptosis of common carp ( Cyprinus carpio ) induced by lipopolysaccharide (LPS) were evaluated. Common carp were irregularity divided into 5 groups and fed five diets with 0 % (CK), 2 %, 4 %, 6 % and 8 % PHB. After 8-week feeding trial, LPS challenge was executed. Results showed that appropriate level of PHB enhanced serum immune function by reversing LPS-induced the decrease of C3, C4, IgM, AKP, ACP and LZM in serum, alleviated LPS-induced intestinal barrier dysfunction by decreasing the levels of 5-HT, D-LA, ET-1 and DAO in serum, increasing ZO-1 , Occludin , Claudin-3 and Claudin-7 mRNA, improving intestinal morphology. Moreover, dietary PHB reversed LPS-induced the decrease of AST and ALT in hepatopancreas, while in serum exhibited the opposite trend. Suitable level of PHB reversed LPS-induced the reduction of GSH-PX, CAT, T-SOD and T-AOC in intestines and hepatopancreas, whereas MDA showed the opposite result. PHB alleviated LPS-induced the decrease of Nrf2 , HO-1 , CAT , SOD and GSH-PX mRNA, the increase of Keap1 mRNA. Appropriate level of PHB alleviated LPS-induced inflammation and apoptosis by up-regulating TGF-β , IL-10 and Bcl-2 mRNA, down-regulating NF-κB , TNF-α , IL-6 , Bax , Caspase-3 , Caspase-8 and Caspase-9 mRNA. Furthermore, PHB inhibited activation of NLRP3 inflammasomes by reducing the levels of NLRP3, Caspase-1, ASC, IL-1β and IL-18 mRNA and protein. In addition, the increases of dietary PHB linearly and quadratically affected LPS-induced adverse effects on common carp. Summary, this study suggested that appropriate level of dietary PHB alleviated LPS-induced oxidative stress, inflammation, apoptosis and the activation of NLRP3 inflammasome in common carp. And the appropriate level of PHB in common carp diets was 4 %.

Effect of dietary heat-killed Lactiplantibacillus plantarum VSG3 on growth, immunity, antioxidant status, and immune-related gene expression in pathogen-aggravated Cyprinus carpio

Abstract:Heat-killed probiotics offer an alternative approach to enhance growth and disease resistance in farmed fish. In this study, we isolated Lactiplantibacillus plantarum VSG3 from the gut of Labeo rohita to investigate the effects of heat-killed L. plantarum (HK-LP) on the health and growth performance of Cyprinus carpio fingerlings. Different concentrations of HK-LP (0, 50, 100, 200, 300, and 400 mg/kg) were administered to the fish, followed by a challenge with Aeromonas hydrophila after 8 weeks of feeding. Notably, the LP200 group exhibited significantly improved percentage weight gain and specific growth rate, accompanied by the lowest feed conversion ratio. Post-challenge survival rates were considerably enhanced in the LP200 group, reaching 60.65%. Moreover, serum analysis indicated significantly higher levels of total protein and albumin in the LP200 group than in the control group. Although HK-LP had no substantial impact on certain serum parameters (glucose, total cholesterol, cortisol, and alanine aminotransferase), aspartate aminotransferase levels were considerably low in the LP200 group. Intestinal protease and trypsin activities significantly increased in the LP200 group, while no significant changes were observed in lipase and amylase activities post-pathogen challenge. Serum immunological indices, including lysozyme, alternative complement pathway, and phagocytic activity, improved considerably in the LP200 group. Additionally, serum antioxidant enzyme activities (superoxide dismutase [SOD], glutathione peroxidase [GPx], catalase [CAT], and myeloperoxidase) were significantly elevated in the LP200 group, while malondialdehyde level was reduced. Gene expression analysis in liver tissue indicated strong upregulation of antioxidant-related genes (SOD, CAT, nuclear factor erythroid 2 [NFE2]-related factor 2 [Nrf2], Kelch-like ECH-associated protein 1[Keap1]) in the LP100 and LP200 groups. Pro-inflammatory cytokines (IL-1β and TNF-α) were considerably downregulated in the kidneys of the LP200 post-challenged fish, although the anti-inflammatory cytokine IL-10 showed an increased expression. Quadratic regression analysis identified the optimal dietary HK-LP level for maximizing growth and immune performance (200.381-270.003 mg/kg). In summary, our findings underscore the potential of HK-LP as a valuable dietary supplement for enhancing carp aquaculture, particularly at the appropriate concentration.
Effects of antimicrobial peptides (AMPs) on blood biochemical parameters, antioxidase activity, and immune function in the common carp (Cyprinus carpio)

Abstract:Antibiotic use in livestock feed additives has resulted in harmful residue accumulation and spread of drug-resistance. We examined the use of antimicrobial peptides (AMPs) as a safer alternative to antibiotics in feeding the common carp. AMPs were added to common carp basal diets (Control) as additives at four concentrations: 100 mg kg(-1) (B1), 200 mg kg(-1) (B2), 400 mg kg(-1) (B3), 600 mg kg(-1) (B4) by dry weight of basal diet. After a 60-day feeding experiment, the final weight, DG and SGR of carps on B1, B2 and B3 diet were significantly higher than the control (p < 0.05). The FCR of carps on B1, B2 and B3 diet were significantly lower than the control (p < 0.05). Carps on B2, B3, and B4 diets showed significantly lower (p < 0.05) levels of triglyceride than the control. B4-fed carps showed significantly lower (p < 0.05) levels of total protein, albumin, and total cholesterol than the control. However there was no remarkable difference (p > 0.05) in levels of uric ammonia, globulin, glutamic-pyruvic transaminase, glutamic-oxalacetic transaminase, lactic dehydrogenase and blood glucose in all groups. The serum superoxide dismutase and catalase activity of B1-fed carps was significantly higher (p < 0.05) than the control and B4-fed carps. The serum alkaline phosphate activity of carps on B1 diets was significantly higher (p < 0.05) than B4-fed carps. The serum acid phosphatase activity of B1-fed carps was significantly higher (p < 0.05) than the control and other antimicrobial peptide-fed groups. The serum lysozyme activity of carps on B1, B2, and B3 diets was significantly higher (p < 0.05) than the control- and B4-fed carps. Regarding immune factors in serum, the levels of immunoglobulin (Ig) and interleukin (IL)-1β in B1-fed carps were significantly higher (p < 0.05) than the control and other groups, while IL-1α levels in B1-fed carps was significantly higher (p < 0.05) than the control-, B2-, and B3-fed carps. Furthermore, there were no significant differences in the content of MHC among the five groups. In conclusion, antimicrobial peptide can reduce triglyceride levels in serum, enrich oxidation resistance, and improve immunity of the common carp. It showed that appropriate concentration of antibacterial peptide as supplements in diets for common carp increased the final weight, DG, SGR and decreased the FCR.

Effects of Probiotics and Its Extracellular Products on the Growth Performance, Immune Response, and Aeromonas hydrophila Resistance of Grass Carp

Abstract:The goal of this study was to research the effects of Lactobacillus buchneri L3‐9 and its extracellular products on growth, immunity, intestinal microorganisms, and disease resistance to Aeromonas hydrophila in grass carp. A total of 120 fish (80–90 g) were distributed into 12 aquariums (10 fish/aquarium) in three replicates and fed 1% of their body weight for 21 days. The grass carp were fed four different diets: a control (group C), a diet supplemented with 1 × 106 cell g−1 of L. buchneri L3‐9 (group B), a diet supplemented with extracellular products of L. buchneri L3‐9 (group E), and a diet supplemented with L. buchneri L3‐9 and the extracellular products (group BE). Compared with the control diet, the B, E, and BE diets increased the weight gain rate by 4.49%, 2.59%, and 4.38%, respectively (p < 0.05), and significantly decreased the feed conversion ratio by 0.72, 0.57, and 0.76, respectively (p < 0.05). Groups B, BE, and E showed significant decreases in mortality, with values of 26.67%, 16.67%, and 23.33%, respectively (p < 0.05). Group B showed a significant increase in superoxide dismutase (SOD) activity and total antioxidant capacity (T‐AOC) and decrease in malondialdehyde (MDA) content compared to the control group (p < 0.05). The highest catalase (CAT) activity was found in group E (p < 0.05). In group B, the expression of the gut proinflammatory gene TNF-α was downregulated (p < 0.05), and the expression of anti‐inflammatory genes, including TGF-β1, IL-10, and Tlr-8, was upregulated compared to that in the control group (p < 0.05). Groups BE and E showed an increased abundance of Lactobacillaceae. Additionally, a drastic decrease in the abundance of pathogenic bacteria such as Aeromonadaceae and Enterobacteriaceae was in these groups compared to the control group. In conclusion, L. buchneri L3‐9 and its extracellular products could improve the growth performance, immune responses, and resistance to A. hydrophila in grass carp. This study provides insights for the development and application of microecological preparations.

Abstract:This study investigated the possible effects of using Pediococcus pentosaceus in common carp (Cyprinus carpio) diet on the growth performance, digestive enzyme activities, haematological indices, and innate immune responses. Four diets were prepared, including a control diet and three diets supplemented with P. pentosaceus at 107, 108, and 109 CFU g−1. After 45 days, fish fed P. pentosaceus at 108 CFU g−1 had significantly higher final body weight, weight gain, and specific growth rate when compared to the control group (P < 0.05). Furthermore, fish fed P. pentosaceus at 108 CFU g−1 had mainly enhanced the digestive enzyme activities, respect to the control fish (P < 0.05). However, the total viable heterotrophic aerobic bacteria were significantly elevated after feeding low dose of P. pentosaceus (P < 0.05). Among haematological parameters, red blood cells (RBCs), white blood cells (WBCs), hemoglobin (Hb), and hematocrit (HCT) were significantly (P < 0.05) increased in fish fed 108, and 109 CFU g−1 of P. pentosaceus. Serum total antibody level, alternative complement, protease, and lysozyme activities, as well as antibacterial activity, were significantly enhanced in fish fed with different doses of P. pentosaceus in comparison with the control group (P < 0.05). The obtained results revealed that P. pentosaceus could be used as a beneficial probiotic to improve the growth performance, digestive enzymes activity, and haemato-immunological responses in common carp culture.

Abstract:Effect of probiotics for common carp (Cyprinus carpio) based on growth performance, feed utilization (feed conversion ratio, FCR) and digestive enzyme (protease, amylase and lipase) activities was investigated. The photosynthetic bacteria and Bacillus sp. isolated from common carp ponds were added to carp basal diets as the probiotics in three forms: 1gkg−1 lyophilized photosynthetic bacteria cells (PSB), 1gkg−1 lyophilized Bacillus sp. (B) and their mix. Twelve aquaria with replicates for treatment and control were used. After a 60-day feeding experiment with probiotics supplemented and non-supplemented control diets, the diets supplemented with probiotics showed significantly better results of growth performance and FCR than those with the basal diet (control). Mean digestive enzyme activities of all probiotics treatment groups were significantly different (P<0.05) with that of the control. The protease activity was remarkably higher in the mix and Bacillus sp. compared with PSB and control. However, there was no difference between the mix and Bacillus sp. As for amylase and lipase, assays showed higher activity in the mix as compared to the rest. In conclusion, it showed that probiotics highly increased the growth performances and digestive enzyme activities, and decreased FCR. Furthermore, different probiotics forms indicated different performances and the mix produced the best results.

Abstract:This study evaluated the impacts of the probiotic, Lactobacillus sakei (L. sakei), and the extract of hawthorn, Crataegus elbursensis, on growth and immunity of the common carp exposed to acetamiprid. Fish (mean ± SE: 11.48 ± 0.1 g) feeding was done with formulated diets (T 1 (control): no supplementation, T 2: 1 × 106 CFU/g LS (Lactobacillus sakei), T3: 1 × 108 CFU/g LS, T 4: 0.5% hawthorn extract (HWE), and T 5: 1% HWE) for 60 days and then exposed to acetamiprid for 14 days. The growth performance improved in the fish fed LS at dietary level of 1 × 108 CFU/g, even after exposure to acetamiprid (P < 0.05). Intestinal Lactobacillus sakei (CFU/g) load increased (P < 0.05), following supplementation with the probiotic-enriched diet. The LS-treated fish had increases in the activity of digestive enzymes (P < 0.05). Both LS and HWE stimulated antioxidant enzymes and immune system components in serum and mucus (alkaline phosphatase (ALP), protease, total Ig, and lysozyme) (P < 0.05). However, the changes were different depending on the kind of the supplement. The malondialdehyde (MDA) levels decreased in HWE-treated fish after acetamiprid exposure (P < 0.05). Both LS and HWE reduced the liver metabolic enzymes (LDH, ALP, AST, ALT, and LDH) in serum both before and after exposure to the pesticide (P < 0.05). However, each enzyme exhibited a different change trend depending on the type of the supplement. HWE showed a stress-ameliorating effect, as glucose and cortisol levels declined in the HWE-treated fish (P < 0.05). This study indicated the immunomodulatory impacts of LS (1 × 108 CFU/g) and HWE (at dietary levels of 0.5-1%). The probiotic showed more performance compared to HWE. However, the HWE mitigated oxidative stress more efficiently than the probiotic.

Abstract:To investigate the effects of alpha-ketoglutarate (AKG) supplementation in a low protein (LP) diet on the growth performance, immune response, and intestinal health of common carp ( Cyprinus carpio ), 600 carp were randomly divided into five dietary groups: a normal protein (NP) diet containing 32% crude protein, an LP diet formulated with 28% crude protein, and LP with AKG at 0.4%, 0.8%, and 1.2% (dry matter). After an 8-week trial period, the results demonstrated that an LP diet led to a decrease in performance, immune response, and intestinal barrier function. Compared with the LP group, the final body weight and weight gain rate in the LP+0.4% AKG group were significantly higher, the feed conversion ratio was significantly decreased with the addition of 0.4% and 0.8% AKG. The supplementation with 0.4% and 0.8% AKG markedly increased the activities of T-SOD and GSH-Px, as well as the expression levels of GPX1a and GPX1b relative to the LP group, whereas the MDA content was significantly decreased in the LP+0.4% AKG group. In addition, the expression levels of tight junctions including claudin-3 , claudin-7 , ZO-1 , and MLCK were significantly up-regulated in the LP+0.4% AKG group, and the relative expression levels of the pro-inflammatory factors IL-1β and IL-6α were significantly lower with the addition of 0.4%, 0.8%, and 1.2% AKG. Moreover, the abundance of Proteobacteria in the LP+0.4% AKG group was lower than that in the LP group, and the abundance of Firmicutes and Fusobacteria was higher at the phylum level. The abundance of Citrobacter in the LP+0.4% AKG group was decreased compared to the LP group, while the abundance of Aeromonas was increased at the genus level. In short, the effects of AKG on the intestinal health of the common carp were systematically and comprehensively evaluated from the perspectives of intestinal physical barrier, chemical barrier, biological barrier, and immune barrier. We found that an LP diet supplemented with 0.4% AKG was beneficial to the growth performance and intestinal health of common carp.

Abstract:Aeromonas comprise a wide range of opportunistic bacteria that cause septicemia (blood poisoning) in fish. With expanding aquaculture, myriad methods have been used to combat these pathologies including probiotics and plant extracts which can potentially address these diseases without harming the environment. In this study, four strains of opportunistic pathogens were isolated from diseased fish, then molecular identification of 16SrDNA was performed. Results identified the four strains as Aeromonas veronii, Shewanella sp., Bacillus cereus, and Aeromonas salmonicida. Experimental trials revealed that A. veronii was the primary strain. Multiple virulencerelated factors were discovered, including aerolysin (Aer), cytotoxic enterotoxin (Act), and the outer membrane protein (OmpAII). These were found only in A. veronii. LC50 in grass carp was 3.6×10 CFU/mL over 96 h, whereas the safe concentration was 9.11×10 CFU/mL. Grass carp were challenged with 3 different concentrations of A. veronii. Following a 48 h incubation period they were treated for 24 h, 48 h, and 72 h with 8.1×10 CFU/mL concentrations. The expression of Interleukin-8 (IL-8) and Tumor Necrosis Factor-α (TNF-α) was up-regulated in the kidney, liver, spleen, and heart using qRT-PCR. Simultaneously, enzyme activity involving Pyruvate Kinase (PK), Alkaline Phosphatase (AKP), Alanine aminotransferase (ALT), Succinate Dehydrogenase (SDH), and Lysozyme (Lsz) in serum and liver tissues was significantly up-regulated. We found that probiotics enhanced growth of grass carp, improved health status, and was beneficial in reducing the prevalence of this disease. Subsequent to feeding mixed probiotics, IL-8 and TNF-α gene expression was significantly lower (p>0.05) than in the infected group, at 16 and 31 days. Expression of these genes in the liver, spleen, and intestines of healthy grass carp fed probiotics was significantly up-regulated in contrast to grass carp fed a basal diet (p<0.05). Our results suggest that use of probiotics as immune stimulator may improve the control of fish diseases in aquaculture.
