Abstract:Carbon allocation of trees to ectomycorrhizas is thought to shape forest nutrient cycling, but the sink activities of different fungal taxa for host resources are unknown. Here, we investigate fungal taxon-specific differences in naturally composed ectomycorrhizal (EM) communities for plant-derived carbon and nitrogen. After aboveground dual labeling of young beech with 15 N and 13 C, ectomycorrhizas formed with different fungal taxa exhibit strong differences in label enrichment. Secondary Ion Mass Spectrometry (SIMS) imaging of nitrogen in cross sections of ectomycorrhizas demonstrates plant-derived 15 N in both root and fungal structures. Isotope enrichment in ectomycorrhizas correlates with that in the corresponding ectomycorrhiza-attached lateral root, supporting fungal taxon-specific N and C fluxes in ectomycorrhizas. The enrichments with 13 C and 15 N in the symbiosis decrease with increasing C/N ratio of ectomycorrhizas, converging to zero at high C/N. The relative abundances of EM fungal species on roots are positively correlated with 13 C enrichment, demonstrating higher fitness of stronger than of less C-demanding symbioses. Overall, our results support that differences among the C/N ratios in ectomycorrhizas formed with different fungal species regulate the supply of the symbioses with host-derived carbon and provide insights on functional traits of ectomycorrhizas, which are important for major ecosystem processes.

Abstract:Summary Forest soils play a pivotal role as global carbon (C) sinks, where the dynamics of soil organic matter (SOM) are significantly influenced by ectomycorrhizal (ECM) fungi. While correlations between ECM fungal community composition and soil C storage have been documented, the underlying mechanisms behind this remain unclear. Here, we conducted controlled experiments using pure cultures growing on naturally complex SOM extracts to test how ECM fungi regulate soil C and nitrogen (N) dynamics in response to varying inorganic N availability, in both monoculture and mixed culture conditions. ECM species dominant in N‐poor soils exhibited superior SOM decay capabilities compared with those prevalent in N‐rich soils. Inorganic N addition alleviated N limitation for ECM species but exacerbated their C limitation, reflected by reduced N compound decomposition and increased C compound decomposition. In mixed cultures without inorganic N supplementation, ECM species with greater SOM decomposition potential facilitated the persistence of less proficient SOM decomposers. Regardless of inorganic N availability, ECM species in mixed cultures demonstrated a preference for C over N, intensifying relatively labile C compound decomposition. This study highlights the complex interactions between ECM species, their nutritional requirements, the nutritional environment of their habitat, and their role in modifying SOM.

Abstract:Studies have shown that arbuscular mycorrhizal fungi (AMF) can shape the rhizosphere microbial community of the host plant, but the underlying mechanisms are unclear. Here, we tested the hypotheses that AMF may affect the rhizosphere microbial community by mediating carbon (C) allocation of the host plant, and that this mediation may be modulated by the soil phosphorus (P) level. Using a split-root system, we conducted a microcosm experiment with three main effects (soil P level: 5 or 25 mg kg −1 ; AMF: with or without inoculation; and spatial niche (i.e., rhizosphere, hyphosphere, and mycorrhizosphere). Host plant growth benefited from AMF under low soil-P conditions. 13 CO 2 isotope labeling showed that AMF increased C allocation to the colonized root and AMF mycelia under low-P conditions, which promoted AMF growth. 13 C-DNA-SIP and 16S rRNA sequencing further indicated that the enhanced C allocation from the host altered the soil microbial community. Our results suggest that AMF enhances the C allocation of the host plant below ground, which can shape microbial community composition. These AMF effects were greater with a low than with a high level of soil P.
Differences in microbial community response to nitrogen fertilization result in unique enzyme shifts between arbuscular and ectomycorrhizal-dominated soils

Abstract:While the effect of nitrogen (N) deposition on belowground carbon (C) cycling varies, emerging evidence shows that forest soils dominated by trees that associate with ectomycorrhizal fungi (ECM) store more C than soils dominated by trees that associate with arbuscular mycorrhizae (AM) with increasing N deposition. We hypothesized that this is due to unique nutrient cycling responses to N between AM and ECM-dominated soils. ECM trees primarily obtain N through fungal mining of soil organic matter subsidized by root-C. As such, we expected the largest N-induced responses of C and N cycling to occur in ECM rhizospheres and be driven by fungi. Conversely, as AM trees rely on bacterial scavengers in bulk soils to cycle N, we predicted the largest AM responses to be driven by shifts in bacteria and occur in bulk soils. To test this hypothesis, we measured microbial community composition, metatranscriptome profiles, and extracellular enzyme activity in bulk, rhizosphere, and organic horizon (OH) soils in AM and ECM-dominated soils at Bear Brook Watershed in Maine, USA. After 27 years of N fertilization, fungal community composition shifted across ECM soils, but bacterial communities shifted across AM soils. These shifts were mirrored by enhanced C relative to N mining enzyme activities in both mycorrhizal types, but this occurred in different soil fractions. In ECM stands these shifts occurred in rhizosphere soils, but in AM stands they occurred in bulk soils. Additionally, ECM OH soils exhibited the opposite response with declines in C relative to N mining. As rhizosphere soils account for only a small portion of total soil volume relative to bulk soils, coupled with declines in C to N enzyme activity in ECM OH soils, we posit that this may partly explain why ECM soils store more C than AM soils as N inputs increase.

Abstract:Despite the general appreciation that arbuscular mycorrhizal fungi (AMF) can influence decomposition of organic materials, the underlying mechanisms remain unclear. Here we investigated whether AMF influence decomposition and change the associated soil bacterial and fungal communities through nutrient acquisition and whether this effect is altered by the level of soil phosphorus (P). We conducted a pot experiment using Medicago truncatula as host plant without or with AMF (a mixture of six species) and with two levels of soil P (4 and 24 mg kg(-1)). Hyphal-ingrowth mesh bags (which excluded roots but not hyphae or other microbes) were used to measure the effect of AMF hyphae on decomposition. Maize leaves dual-labeled with N-15:C-13 stable isotopes were used as the organic substrate. Bacterial and fungal communities were accessed via sequencing of partial 16S rRNA and ITS genes. The results showed the N-15 and C-13 content of the organic matter remaining in the mesh bags under low soil P availability were significantly lower in the treatment with AMF than without AMF. Under high soil P availability, however, no significant difference of N-15 content remaining was found between AMF and non-AMF treatments, while C-13 content remaining was higher in AMF than in non-AMF treatment. Levels of mycorrhizal colonization and (1) N-5 transport from organic matter to the host by AMF hyphae were higher at low P availability than at high P availability, suggesting that the host can acquire more nutrients through the AM from organic matter when soil P availability was low. The composition of bacterial and fungal communities were altered by AMF at both soil P levels. For bacterial community, the richness and diversity were higher with low P availability, especially for the phyla Saccharibacteria and Nitrospirae. For fungal community, AMF increased the richness but not the diversity under low P availability. Together, we show that AMF acquire nutrients from organic matter for host, influence decomposition and alter the bacterial and fungal communities, and that these effects were modulated by the soil P availability.

Abstract:Mycorrhizal fungi can affect soil organic matter cycling through several mechanisms including priming, nutrient competition, and direct enzyme production. Differences in nutrient foraging strategies between ectomycorrhizal (EcM) and arbuscular mycorrhizal (AM) fungi produce divergent belowground dynamics: where EcM can take up organic nitrogen and directly break down soil organic matter (SOM) by producing enzymes, AM fungi are limited to scavenging mineral N. EcM-associated tree species also have leaf litter with relatively higher ratios of carbon to nitrogen (C:N), and belowground saprotrophic communities more dominated by fungi. Consequently, free-living microbes in EcM-dominated soils should experience nitrogen limitation, with subsequent increases in enzyme production and decreased carbon use efficiency (CUE). However, the relative importance of the effects of substrate quality and fungal community composition on enzyme production and CUE are unclear. To assess this distinction, we sampled the organic horizon and 10 cm of the mineral horizon in northern temperate forest soils along a gradient of EcM dominance. We characterized fungal community composition by measuring EcM relative abundances from extracted fungal DNA and the fungal to bacterial (F:B) ratios from phospholipid fatty acid (PLFA) analysis. We assessed soil substrate quality as the soil C:N ratio. Soil microbial functions were measured as potential activities of five hydrolytic and two oxidative enzymes, and microbial CUE. We found that the fungal community, represented by either the F:B ratio, EcM relative abundance, or both, affected CUE and six measured enzyme activities, while the C:N ratio affected only oxidative and chitin-targeting extracellular enzyme activities. Our results highlight the use of EcM relative dominance as a predictor of soil microbial community composition and function independent of substrate quality.
Ectomycorrhizal and non-mycorrhizal rhizosphere fungi increase root-derived C input to soil and modify enzyme activities: A 14 C pulse labelling of Picea abies seedlings

Abstract:Consequences of interactions between ectomycorrhizal fungi (EcMF) and non-mycorrhizal rhizosphere fungi (NMRF) for plant carbon (C) allocation belowground and nutrient cycling in soil remain unknown. To address this topic, we performed a mesocosm study with Norway spruce seedlings [Picea abies (L.) H. Karst] inoculated with EcMF, NMRF, or a mixture of both (MIX). 14 CO2 pulse labelling of spruce was applied to trace and visualize the 14 C incorporation into roots, rhizohyphosphere and hyphosphere. Activities and localization of enzymes involved in the C, nitrogen (N) and phosphorus (P) cycling were visualized using zymography. Spruce seedlings inoculated with EcMF and NMRF allocated more C to soils (EcMF: 10.7%; NMRF: 3.5% of total recovered C) compared to uninoculated control seedlings. The 14 C activity in the hyphosphere was highest for EcMF and lowest for NMRF. In the presence of both, NMRF and EcMF (MIX), the 14 C activity was 64% lower compared with EcMF inoculation alone. This suggests a suppressed C allocation via EcMF likely due to the competition between EcMF and NMRF for N and P. Furthermore, we observed 57% and 49% higher chitinase and leucine-aminopeptidase activities in the rhizohyphosphere of EcMF compared to the uninoculated control, respectively. In contrast, β-glucosidase activity (14.3 nmol cm-2 h-1 ) was highest in NMRF likely because NMRF consumed rhizodeposits efficiently. This was further supported by that enzyme stoichiometry in soil with EcMF shifted to a higher investment of nutrient acquisition enzymes (e.g., chitinase, leucine-aminopeptidase, acid phosphatase) compared to NMRF inoculation, where investment in β-glucosidase increased. In conclusion, the alleviation of EcMF from C limitation promotes higher activities of enzymes involved in the N and P cycle to cover the nutrient demand of EcMF and host seedlings. In contrast, C limitation of NMRF probably led to a shift in investment towards higher activities of enzymes involved in the C cycle.

Abstract:In forest ecosystems, ectomycorrhizal (ECM) fungi are important for plant growth and soil biogeochemical processes. The biochemical composition of ECM mycelium is an important fungal effect trait with consequences for its decomposition rate, and consequently on soil carbon pools and plant nutrition. Although the link between ECM fungi and leaf litter–released nutrients is well known, the response of ECM fungal biochemical composition to different leaf litter species remains poorly understood. To determine how leaf litter quality influences ECM fungi’s biochemical profiles, we planted young beech trees in an oak forest and replaced the natural leaf litter with that of European beech (Fagus sylvatica), ash (Fraxinus excelsior), maple (Acer pseudoplatanus), or lime (Tilia cordata). We assessed the biochemical profiles of ECM root tips colonized by common fungal taxa in temperate forests (i.e., Cenococcum geophilum, Inocybe sp., and Lactarius subdulcis), using attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR). ECM fungal biochemical composition changed with leaf litter species. Changes were apparent in the infrared absorption bands assigned to functional groups of lipids, amides, and carbohydrates. C. geophilum and L. subdulcis exhibited large spectral differences corresponding to the initial pattern of leaf litter chemical composition between samples collected in the beech and ash leaf litter treatments. In contrast, Inocybe sp. was influenced by lime, but with no differences between samples from ash or beech leaf litter treatments. Although the spectral bands affected by leaf litter type differed among ECM fungi, they were mainly related to amides, indicating a dynamic response of the fungal proteome to soil nutritional changes. Overall, the results indicate that the biochemical response of ECM fungi to leaf litter species varies among ECM fungal species and suggests that the biochemical composition of ECM mycelium is a fungal response trait, sensitive to environmental changes such as shifts in leaf litter species.

Abstract:The presence of living roots can markedly change soil properties, microbial activities, and biogeochemical cycling in the rhizosphere compared to bulk soil. Such rhizosphere effect has been increasingly recognized to play a crucial role in carbon (C) and nitrogen (N) cycling in terrestrial ecosystems. Here, we investigated the rhizosphere effects of 12 tree species associated with two contrasting mycorrhizal types (arbuscular mycorrhizal, AM vs. ectomycorrhizal, ECM) and co-occurring in a temperate secondary forest in Northeast China. The adhering soil method was adopted to sample paired rhizosphere and bulk soils of field-grown trees in the peak growing season. Generally, we found positive rhizosphere effects on soil properties (ammonium and nitrate content, total C and N content, and C:N ratio), microbial abundances (bacteria, fungi, and actinomycetes), enzyme activities (C- and N-degradation enzymes), and C mineralization rate (C-min), but minor rhizosphere effects on soil pH, phosphorus content and phosphatase activity, net N mineralization rate (N-min), and specific C-min and N-min. (per unit soil C or N). Because of large intraspecific and particularly interspecific variations, less than half of the rhizosphere effects were statistically significant (P < 0.05), and none of the rhizosphere effects differed significantly between the two mycorrhizal types (AM vs. ECM). Moreover, most soil variables varied greatly within and across species, and thus did not differ significantly between the two mycorrhizal types (AM vs. ECM). However, soil pH, nitrate content, microbial abundance, and C-degradation enzyme activity were notably lower under ECM species than under AM species. Collectively, these results show that rhizosphere effects varied greatly among the six species within both mycorrhizal groups, and such variations within mycorrhizal group were even larger than those between mycorrhizal group. Future work should include a larger number of tree species to increase statistical power, and combine phylogenic history and functional traits of plant species with the type and traits of mycorrhizal fungi to better understand how plant-microbe interactions influence biogeochemical cycling in the rhizosphere.

Abstract:Forest soils store large amounts of carbon (C) and nitrogen (N), yet how predicted shifts in forest composition will impact long-term C and N persistence remains poorly understood. A recent hypothesis predicts that soils under trees associated with arbuscular mycorrhizas (AM) store less C than soils dominated by trees associated with ectomycorrhizas (ECM), due to slower decomposition in ECM-dominated forests. However, an incipient hypothesis predicts that systems with rapid decomposition-e.g. most AM-dominated forests-enhance soil organic matter (SOM) stabilization by accelerating the production of microbial residues. To address these contrasting predictions, we quantified soil C and N to 1 m depth across gradients of ECM-dominance in three temperate forests. By focusing on sites where AM- and ECM-plants co-occur, our analysis controls for climatic factors that covary with mycorrhizal dominance across broad scales. We found that while ECM stands contain more SOM in topsoil, AM stands contain more SOM when subsoil to 1 m depth is included. Biomarkers and soil fractionations reveal that these patterns are driven by an accumulation of microbial residues in AM-dominated soils. Collectively, our results support emerging theory on SOM formation, demonstrate the importance of subsurface soils in mediating plant effects on soil C and N, and indicate that shifts in the mycorrhizal composition of temperate forests may alter the stabilization of SOM.

Abstract:The systematic work-up for patient with clinically localised prostate cancer must allow a reconciliation of all the necessary elements to propose one or several therapeutical options. The evaluation must take into account the life's expectancy, and must precise the cancer progression and prognostic factors. The life expectancy depends out age, performance status and comorbidities. Digital rectal exam, PSA and Gleason score analysis are systematic. These tests allow the specialist to decide whether or not to complete the progression work-up by bone scan and pelvic CT. This first work-up should define if a patient will benefit from some curative local treatment. In case of palliative treatment or deferred treatment, the work-up highlights the specialist to decide the medical supervision. Some exams, systematic or not, are very specific from each therapeutic modality considered; they allow to precise individually the efficacy and toxicity prognostic factors for each therapeutic strategy. The final therapeutic decision is based at first on this objective data even if they are probabilistic, and finally on informed patients preferences.
Long-term nitrogen fertilization impacts plant-microbial interactions differently in arbuscular and ectomycorrhizal trees

Abstract:Enhanced nitrogen (N) availability in temperate forests has altered ecosystem carbon (C) and N cycling. Recent research has shown that these alterations lead to reductions in belowground C allocation by trees and that the consequences of these reductions on soil C and nutrient cycling may vary by mycorrhizal type. We hypothesized that trees that associate with ectomycorrhizal fungi (ECM) would reduce C allocation towards roots and mycorrhizal fungi to a greater extent than trees that associate with arbuscular mycorrhizal fungi (AM) in response to > 25 years of N fertilization. We further hypothesized that N induced decoupling of roots and microbes in ECM trees would be evidenced by greater declines in extracellular enzyme activities. We measured belowground C allocation to fine root biomass and mycorrhizal colonization in 6 AM and 6 ECM dominated plots in the N fertilized and reference watersheds at the Fernow Experimental Forest in West Virginia, USA. We compared these to measurements of simple-C, complex-C, nitrogen, and phosphorus acquiring enzyme activities in organic horizon, bulk mineral, and rhizosphere soil fractions. N fertilization reduced fine root biomass and mycorrhizal colonization in both AM and ECM stands. We found more consistent reductions in enzyme activities in ECM soils than AM soils under N fertilization which may have been driven by greater declines in root-C transfer to soil microbes. This mechanism helps to explain variability in soil C cycling responses across N gradient and fertilization experiments and may prove useful in predicting the fate of soil C stocks in response to N deposition.

Abstract:While it is well established that plants associating with arbuscular mycorrhizal (AM) and ectomycorrhizal (ECM) fungi cycle carbon (C) and nutrients in distinct ways, we have a limited understanding of whether varying abundance of ECM and AM plants in a stand can provide integrative proxies for key biogeochemical processes. We explored linkages between the relative abundance of AM and ECM trees and microbial functioning in three hardwood forests in southern Indiana, USA. Across each site's 'mycorrhizal gradient', we measured fungal biomass, fungal : bacterial (F : B) ratios, extracellular enzyme activities, soil carbon : nitrogen ratio, and soil pH over a growing season. We show that the percentage of AM or ECM trees in a plot promotes microbial communities that both reflect and determine the C to nutrient balance in soil. Soils dominated by ECM trees had higher F : B ratios and more standing fungal biomass than AM stands. Enzyme stoichiometry in ECM soils shifted to higher investment in extracellular enzymes needed for nitrogen and phosphorus acquisition than in C-acquisition enzymes, relative to AM soils. Our results suggest that knowledge of mycorrhizal dominance at the stand or landscape scale may provide a unifying framework for linking plant and microbial community dynamics, and predicting their effects on ecological function.

Abstract:Read the free Plain Language Summary for this article on the Journal blog. Ectomycorrhizal fungi (EMF) are common belowground tree symbionts, supplying trees with water and nutrients. In return, large amounts of C assimilated by trees can be allocated into EMF. However, the chemical forms in which the C is transferred from trees to fungi under field conditions are mostly unknown. In this study, we aimed to unravel the fate of tree‐derived C in EMF. We conducted 13CO2 pulse labelling of Pinus halepensis trees in two forest sites with adjacent EMF sporocarps, combined with a non‐targeted metabolomics profiling of root and sporocarp tissues. 13C was measured in sporocarps of Tricholoma terreum and Suillus collinitus up to 3 m from pine stems. C was assimilated in the labelled trees' needles and transferred to their roots. Starting from Day 2 after labelling, the C was transferred to adjacent sporocarps, peaking on Day 5. We identified more than 100 different labelled metabolites of different chemical groups present in roots and sporocarps. Of them, 17 were common to pine roots and both EMF species, and additional eight common to roots and one of the two EMF. The major labelled metabolites in the root tips were amino acids and tricarboxylic acid intermediates. The major labelled metabolites in sporocarps were amino acids, nucleotides, and fatty acids. We also identified labelled carbohydrates in all tissues. Labelling patterns diverged across different tissues, which can hint at how the C was transferred. Considering the young tree as a sole C source for these sporocarps, and with a diurnal assimilation of 5.4 g C, the total monthly C source is ~165 g C. On average, there were 10 sporocarps around each tree, each requiring ~1 g C. Therefore, a 10 g C investment would make 6% of total tree C allocation, and about 12% of net primary productivity. Overall, we found that this significant and ubiquitous transfer of metabolites from tree roots to EMF sporocarps is more rapid and chemically diverse than once thought. Abstract:Compared with ectomycorrhizal (ECM) forests, arbuscular mycorrhizal (AM) forests are hypothesized to have higher carbon (C) cycling rates and a more open nitrogen (N) cycle. To test this hypothesis, we synthesized 645 observations, including 22 variables related to below-ground C and N dynamics from 100 sites, where AM and ECM forests co-occurred at the same site. Leaf litter quality was lower in ECM than in AM trees, leading to greater forest floor C stocks in ECM forests. By contrast, AM forests had significantly higher mineral soil C concentrations, and this result was strongly mediated by plant traits and climate. No significant differences were found between AM and ECM forests in C fluxes and labile C concentrations. Furthermore, inorganic N concentrations, net N mineralization and nitrification rates were all higher in AM than in ECM forests, indicating 'mineral' N economy in AM but 'organic' N economy in ECM trees. AM and ECM forests show systematic differences in mineral vs organic N cycling, and thus mycorrhizal type may be useful in predicting how different tree species respond to multiple environmental change factors. By contrast, mycorrhizal type alone cannot reliably predict below-ground C dynamics without considering plant traits and climate.

Abstract:Symbiotic fungi mediate important energy and nutrient transfers in terrestrial ecosystems. Environmental change can lead to shifts in communities of symbiotic fungi, but the consequences of these shifts for nutrient dynamics among symbiotic partners are poorly understood. Here, we assessed variation in carbon (C), nitrogen (N) and phosphorus (P) in tissues of arbuscular mycorrhizal (AM) fungi and a host plant (Medicago sativa) in response to experimental warming and drought. We linked compositional shifts in AM fungal communities in roots and soil to variation in hyphal chemistry by using high-throughput DNA sequencing and joint species distribution modelling. Compared to plants, AM hyphae was 43% lower in (C) and 24% lower in (N) but more than nine times higher in (P), with significantly lower C:N, C:P and N:P ratios. Warming and drought resulted in increases in (P) and reduced C:P and N:P ratios in all tissues, indicating fungal P accumulation was exacerbated by climate-associated stress. Warming and drought modified the composition of AM fungal communities, and many of the AM fungal genera that were linked to shifts in mycelial chemistry were also negatively impacted by climate variation. Our study offers a unified framework to link climate change, fungal community composition, and community-level functional traits. Thus, our study provides insight into how environmental change can alter ecosystem functions via the promotion or reduction of fungal taxa with different stoichiometric characteristics and responses.

Abstract:Ectomycorrhizal (ECM) fungi are functionally important in biogeochemical cycles in tropical ecosystems. Extracellular enzymatic activity of ECM on a ground-area basis is the product of two attributes; exploration capacity (ECM surface-area) and specific enzymatic activity. Here, we elucidated which attribute better explained the ECM enzymatic activity in response to different levels of soil phosphorus (P) and Nitrogen (N) availability in five Bornean tropical rainforests. We determined the surface area of ECM root tips as well as the enzymatic activities per ECM surface area for carbon (C), N and P degrading enzymes in each site. We evaluated the relationship of ECM enzyme activities with the resource availabilities of C (Above-ground net primary production; ANPP), N, and P of ECM by a generalized linear mixed model. The ECM enzymatic activities on a ground-area basis were more significantly determined by specific enzymatic activity than by the exploration capacity. Specific enzymatic activities were generally negatively affected by C (ANPP) and soil P availability. ECM fungi enhance the specific enzyme activity rather than the exploration capacity to maintain the capacity of nutrient acquisition. The less dependence of ECM fungi on the exploration capacity in these forests may be related to the limitation of C supply from host trees. We highlighted the adaptive mechanisms of ECM fungi on nutrient acquisition in tropical ecosystems through the response of enzymatic activity to nutrient availability across the elements.
