Abstract:The aim of this experiment was to investigate the effects of the partial substitution of fish meal with soybean products and chicken meal on the growth performance, antioxidant capacity and intestinal microbiota of Penaeus monodon. A total of 450 healthy, consistent shrimp were randomly divided into five groups, with three replicates per group and 30 shrimp per replicate. The proportion of fish meal substituted with soybean products and chicken meal in the five feed groups was 0% (FM), 40% (40SC), 60% (60SC), 80% (80SC) and 100% (100SC). The experiment lasted for 8 weeks. The results showed that, compared to the FM group, the 40SC and 60SC groups had a decrease in WG and SR, but there was no significant difference (p > 0.05). In contrast, compared to the FM group, the FCR in the 100SC group was significantly increased (p < 0.05), while there was no significant difference among the FM and 40SC, 60SC and 80SC groups (p > 0.05). Compared to the FM group, the ACP in the 80SC and 100SC groups significantly increased (p < 0.05), while the 40SC and 60SC groups had no significant difference (p > 0.05). The AKP in the 100SC group was significantly higher than that in the FM group (p < 0.05), while there were no significant differences among the other four groups (p > 0.05). There were no significant differences in T-AOC and T-SOD among all the treatment groups (p > 0.05). The next-generation sequencing of the intestinal microbiota showed that Proteobacteria was the most abundant phylum in the five groups, accounting for 37.67%, 66%, 40%, 40% and 43.33%, respectively. Compared to the FM group, the Fusobacteriota in the other four groups decreased significantly (p < 0.05). The functional prediction of FAPROTAX indicated that no functional components were observed which are harmful to the body. Considering the effects on growth performance, antioxidant capacity and intestinal microbiota, it is feasible to use soybean products and chicken meal to replace 60% of fish meal in the feed of P. monodon.

Abstract:The feeding method is of great significance for aquaculture production and cost. In recent years, research on intelligent feeding, a method based on fish appetite, has been trending. Few studies, however, have focused on fish welfare issues that can result from intelligent feeding. In this study, an adaptive feeder based on a practical intelligent feeding method was designed to evaluate whether this intelligent feeding method would impair fish welfare compared to traditional automatic feeding. The results indicated that the amount of residual feed and size inhomogeneity in the traditional group was significantly higher than that in the intelligent group. The results of the growth indicator showed that the weight gain rate (WGR) in the intelligent feeding group was significantly higher (30.17%) than in the traditional feeding group. Although no significant differences were observed in the survival rate (SR), the condition factor (CF) and the hepatosomatic index (HSI), the specific growth rate (SGR) in the intelligent feeding group was significantly increased by 8.33% while the feed conversion rate (FCR) was reduced by a remarkable 17.07% compared to the traditional feeding group. Moreover, intelligent feeding significantly improved the pepsin activity of the bass. In terms of immunity and antioxidant capacity, however, the fish in the intelligent feeding group had a significantly lower lysozyme (LZM) level than those fed with the traditional method. For superoxide dismutase (SOD), the intelligent group also displayed a lower activity value, although it was not significant. The intelligent feeding group had 0.14 nmol/mg protein higher malondialdehyde (MDA) activity than the traditional group. Regarding stress, although no statistical significance was observed, the cortisol level in the intelligent group was 1.9 ng/ml higher than in the traditional group. Together, these data suggested that the intelligent feeding method can significantly improve fish growth but may also result in stress and suppress innate immunity.
Effects of Dietary Bio-Fermented Selenium Supplementation on Growth, Immune Performance, and Intestinal Microflora of Chinese Mitten Crabs, Eriocheir sinensis

Abstract:Selenium is a vital trace mineral that is crucial for maintaining regular biological processes in aquatic animals. In this study, a four-week dietary trial was carried out to assess the impact of bio-fermented selenium (Bio−Se) on the growth and immune response of Chinese mitten crabs, Eriocheir sinensis. The crabs were randomly allocated to five dietary treatment groups, each receiving a different dose of Bio−Se. The doses included 0, 0.3, 0.6, 1.5, and 3.0 mg/kg and were accurately measured in basal diet formulations. The results showed the weight gain rate (WGR), specific growth rate (SGR), and survival rate (SR) in the 1.5 mg/kg Bio−Se group were the highest, and 3.0 mg/kg of Bio−Se has an inhibitory effect on the WGR, SGR, and SR. The activities of the immune enzymes, including glutathione peroxidase (GPX), superoxide dismutase (SOD), and acid phosphatase (ACP), of the hepatopancreas were significantly (p < 0.05) increased in the 1.5 mg/kg Bio−Se group, while they decreased (p < 0.05) in the 3.0 mg/kg feeding group compared to the 0 mg/kg feeding group. The concentration of maleic dialdehyde (MDA) exhibited the opposite pattern. Similarly, the mRNA expression levels of antimicrobial peptides (ALF-1, Crus-1, and LYS), ERK, and Relish genes were also observed to be the highest in the 1.5 mg/kg Bio−Se group compared with the other groups. Furthermore, the administration of 1.5 mg/kg of Bio−Se resulted in an increase in the thickness of the intestinal plica and mucosal layer, as well as in alterations in the intestinal microbial profile and bacterial diversity compared to the dose of 0 mg/kg of Bio−Se. Notably, the population of the beneficial bacterial phylum Fusobacteria was increased after crabs were fed the 1.5 mg/kg Bio−Se diet. In conclusion, the oral administration of 1.5 mg/kg of Bio−Se improved the growth efficiency, antioxidant capabilities, immunity, and intestinal health of E. sinensis. Through a broken-line analysis of the WGR against dietary Bio−Se levels, optimal dietary Bio−Se levels were determined to be 1.1 mg/kg. These findings contribute valuable insights to the understanding of crab cultivation and nutrition.

Abstract:The robustness of shape recovery based on deformable models depends in general, on the relative difference of position and topology of the initial model with respect to the data: a close initialization with correct topology guarantees a proper recovery of the object. Furthermore, the closeness of the initial model greatly influences the time of computation needed for the recovery. In this paper, we propose a method for initializing deformable models from range data or volumetric images. The proposed method solves two distinct problems. First, we use the topological segmentation of volumetric images in order to recover the approximate topology of the object. Second, we use an efficient mesh sampling algorithm to control the number of vertices of the initial model. The method takes into account missing data and outliers.
Fattening in Saline and Alkaline Water Improves the Color, Nutritional and Taste Quality of Adult Chinese Mitten Crab Eriocheir sinensis

Abstract:The majority of pond-reared Chinese mitten crab (Eriocheir sinensis) grow and fatten in freshwater. Previous studies illustrated that E. sinensis cultured in saline-alkaline water in outdoor environments showed a higher quality than that cultured in freshwater. However, it is still unclear whether salinity or alkalinity has an important positive effect on the quality of E. sinensis. This study aimed to investigate the gonadal development, edible yield, coloration, and nutritional and flavor quality of E. sinensis fattening in saline and alkaline water indoors. Results showed that there were no significant changes observed in gonadosomatic index (GSI) and other edible parameters among freshwater (FW), saline water (SW), and alkaline water (AW) during the 55-day fattening period (p > 0.05). Significantly higher a* and b* values of freeze-dried female carapace were observed fattening in SW and AW compared with that of FW (p < 0.05). The crude protein in gonad and male muscle, moisture in female muscle, and crude lipid in male muscle increased significantly from FW to SW and AW (p < 0.05). Better nutritional and flavor values were also detected in male hepatopancreas and muscles. In conclusion, numerous advantages of fattening in SW and AW were observed, including the improvement of carotenoid accumulation in freeze-dried carapace, DHA, EPA, total essential free amino acids (∑EFAA), total free amino acids (∑FAA), and total umami values (∑TUV) contents in male hepatopancreas and muscle.

Plant and Animal-Type Feedstuff Shape the Gut Microbiota and Metabolic Processes of the Chinese Mitten Crab Eriocheir sinensis

Abstract:In animals, growth and development are strongly correlated with the gut microbiota and metabolic profiles. In this study, gut microbiome communities, metabolic profiles, and growth performance of Eriocheir sinensis under three dietary feed types based on waterweed plants only, freshwater snails only, and waterweed plants combined with freshwater snails were studied by using 16S rRNA gene sequencing and liquid chromatography-mass spectrometry. Results indicated that different feed types dramatically affected the growth performances of E. sinensis by altering the gut microbiota and metabolic profiles. Aquatic plants, such as waterweeds, played essential roles in shaping gut microbiome communities, and the optimal Bacteroides-to-Firmicutes ratio might strongly promote growth performance. Waterweed plants also helped decrease maleficent Proteobacteria caused by excess animal-type feedstuff, such as freshwater snails, and might have positive roles in antibacterial functions in gut. A diet based on waterweeds only resulted in lipid metabolism disorders, which significantly retarded the growth of E. sinensis. In summary, E. sinensis cultured with a diet of waterweeds and freshwater snails showed superior growth performance due to their healthy gut microbiota and metabolic homeostasis. Our findings unveiled the roles of aquatic plants and animal-type food such as freshwater snail in shaping the gut microbiota and metabolic processes and provided guidance for the aquaculture of E. sinensis in future.

Abstract:The research aimed to evaluate the effects of dietary fructooligosaccharide, probiotics and their combination on the growth performance, antioxidant enzymes and stress resistance of juvenile Eriocheir sinensis. Crabs (average weight: 9.90 ± 0.07 g) were randomly assigned into 4 groups (3 tanks per group) fed experimental diets: basal diet (control group), basal diet supplemented with 5 g/kg FOS, basal diet supplemented with 2 g/kg multi-strain probiotics (each gram contains Lactobacillus acidophilus 106 CFU + Bacillus subtilis 107 CFU + Saccharomyces cerevisiae 1010 CFU) and basal diet supplemented with symbiotic (5 g/kg FOS + 2 g/kg multi-strain probiotics) (designated as control, D1, D2 and D3, respectively). After 8 weeks of feeding trial, crabs were exposed to the transportation process (3 h). Cumulative mortality was recorded at 24 h, 48 h, 72 h and 96 h after transport stress, and pre-transport and post-transport sampling of hepatopancreas occurred for assay. The results indicated that crabs fed D3 had significantly higher weight gain (WG), specific growth rate (SGR) and lower feed conversion ratio (FCR) than crabs fed control diet. Before stress, compared to the control group, crabs fed D1 and D3 diet significantly increased catalase (CAT), superoxide dismutase (SOD) activities and decreased malondialdehyde (MDA) activities, the highest activities of pepsin, trypsin and chymotrypsin were observed in crabs fed D2 diet. After stress, there was a trend that the activities of antioxidants enzymes and proteases in all groups were significantly decreased. And compared to the control group, crabs fed D1 and D3 diet significantly improved SOD and CAT activities, the lowest MDA activity was observed in crabs fed D3 diet, no significant difference was obtained in activities of total antioxidant capacity (T-AOC) and three proteases among all groups (P > 0.05). At 96 h after transportation process, the lowest cumulative mortality was observed in crabs fed D3 diet, the difference among all groups before and at 24 h, 48 h and 72 h after stress was not significant (P > 0.05). In summary, a basal diet supplemented with synbiotic could improve the growth performance and protect the hepatopancreas of Eriocheir sinensis more effectively than singular supplementation with prebiotics or probiotics.

Abstract:Chinese mitten crabs ( Eriocheir sinensis ) are traditionally fed iced trash fish, but the industry is facing problems such as low breeding safety. Black soldier fly ( Hermetia illucens ) is an alternative protein source in animal diets, including diets for aquatic animals, due to its high nutritional value. However, studies on the effects of black soldier fly on the flavor characteristics of aquatic animals are still limited. In the present study, we investigated the effects of the complete replacement of iced trash fish with black soldier fly larvae during the fattening period of Chinese mitten crab on the flavor molecule contents and evaluation indices. The levels of free amino acids and nucleotides were determined in three edible parts (muscle, hepatopancreas, and gonads) of crab. Taste activity value analysis showed that glutamic acid, glycine, alanine, and arginine were the main amino acids contributing to the umami taste and sweetness, while histidine, lysine, valine, and methionine were the main amino acids contributing to the bitterness. Equivalent umami concentration (EUC) analysis showed that female gonads had the strongest umami taste, followed by the hepatopancreas and muscle. Sweetness value (SWT) analysis showed that the sweetness of muscle was the highest. Feeding black soldier fly larvae affected the flavor characteristics of crabs with tissue and sex differences. The EUC of the female gonads and SWT of the muscle were significantly increased. Meanwhile, the EUC of the hepatopancreas and SWT of the gonads were slightly decreased in male crabs. Our results indicate that the complete replacement of iced trash fish with black soldier fly larvae during the fattening period significantly enhances the flavor characteristics of crabs based on the contents of flavoring amino acids and nucleotides. It is important for sustainable aquaculture to replace animal protein with alternative protein sources such as black soldier fly larvae.

Cottonseed Meal Protein Hydrolysate Improves the Growth Performance of Chinese Mitten Crab (Eriocheir sinensis) by Promoting the Muscle Growth and Molting Performance

Abstract:Growth retardation and prolonged marketing cycle have been noticed in the practical aquaculture of Chinese mitten crab (Eriocheir sinensis) fed with artificial feed. Plant protein hydrolysates contain a large number of small peptides and free amino acids, which can improve the growth performance of aquatic animals. However, the potential mechanisms are still not well elucidated. In this research, the influences of cottonseed meal protein hydrolysate (CPH) on the growth, feed utilization, muscle growth, and molting performance were investigated in E. sinensis. A total of 240 crabs (mean body weight 37.32 ± 0.38 g) were individually randomly distributed to six diets supplemented with 0%, 0.2%, 0.4%, 0.8%, 1.6%, and 3.2% of CPH for 12 weeks. These findings indicated that the addition of CPH at 0.4% significantly increased the survival rate, body protein gain, apparent protein utilization, trypsin and pepsin activities, and the methyl farnesoate content. When the dose reached 0.8%, the weight growth ratio, meat yield, ecdysone concentration, and the transcription of the ecdysteroid receptor all significantly increased, while the transcriptions of both myostatin and molt-inhibiting hormone significantly decreased. When CPH was added at 1.6%-3.2%, the feed conversion ratio, body crude protein content, Na+/K+-ATPase activity, and the molting ratio were all significantly improved, while the opposite was true for the transcription of the transforming growth factor-β type I receptor. The investigation results indicated that when added above 0.4%, CPH could stimulate the growth performance of E. sinensis and promote the muscle growth and molting performance.
Bacillus Subtilis B10 Improved Antioxidant Capacity, Immunity, and Lipid Metabolism of Chinese Mitten Crab (eriocheir Sinensis) During Hoarding Time

Abstract:Farmers usually prefer to sell Chinese mitten crabs (Eriocheir sinensis) until around the Spring Festival for greater economic benefits. However, the hoarding stages in winter and cold chain logistics would reduce the vitality of crabs, which would affect the economic benefits. Therefore, this experiment investigated the effects of adding Bacillus subtilis B10 on their health and vitality status after transportation, as well as the immunity and lipid metabolism in cold. The results showed that adding 1 × 109 CFU/kg B. subtilis B10 in feed could improve the vitality of crabs and reduce feet breakage caused by transportation. In male crabs, it could promote growth, and improve antioxidant capacity by increasing the expression of antioxidant gene including SOD and PRX1, increasing the activity of CuZn-SOD and T-SOD, and regulate fat metabolism by decreasing the relative expression of FAD6 and FAD9 thus adapting to environmental changes. In female crabs, it could not promote growth, but it could improve antioxidant capacity by increasing the activity of T-AOC, CuZn-SOD, and T-SOD, and regulate the expression of immune-related and apoptosis-related genes including CASP1, AKT, and AQP and regulate fat metabolism by decreasing the relative expression of FAD6, FAD9, and ELOVL6, thus adapting to environmental changes. This experiment provided a reference basis for reducing economic losses in the hoarding stage and transportation, promoting breeding methods and nutritional requirements of single-sex farming models, and application of probiotics in the field of crab farming.
Replacement of fish meal with cottonseed protein concentrate in Chinese mitten crab (Eriocheir sinensis): Nutrient digestibility, growth performance, free amino acid profile, and expression of genes related to nutrient metabolism

Abstract:This study aimed to investigate the application of cottonseed protein concentrate (CPC) in Chinese mitten crabs (Eriocheir sinensis). First, the apparent digestibility coefficient (ADC) of CPC, fish meal and soybean meal were compared in crabs (21.72 ± 0.33 g). The protein ADC of CPC was 90.42%, which was significantly higher than that of soybean meal (83.16%) (P < 0.05). The ADC of Phe, Cys and Glu of CPC were significantly higher than those of fish meal, while the ADC of Ile, Leu, Lys, Met, Thr and Ala of CPC were significantly lower (P < 0.05). Second, we investigated the effects of fish meal substitution by CPC on growth performance, free amino acid profile, and expression of genes related to nutrient metabolism in crabs. Six diets were formulated by replacing 0%, 15%, 30%, 45%, 60% and 75% fish meal with CPC, namely FM, CPC15, CPC30, CPC45, CPC60, and CPC75. A total of 630 crabs (1.68 ± 0.00 g) were randomly divided into 18 tanks (3 tanks per group) and fed 3 times daily for 9 weeks. Results showed that CPC75 group significantly reduced growth performance, feed conversion efficiency, and free Ile, Leu, Lys, Met, and Thr contents in muscle (P < 0.05). The contents of free amino acids (Arg, His, Ile, Leu, Lys, Met, Phe, Thr, Val, Ala, Cys, Glu, Gly, Ser and Tyr) in hepatopancreas decreased linearly with the increase of dietary CPC level (P < 0.05). The substitution of more than 45% fish meal with CPC significantly decreased the concentration of delicious amino acids (Ala, Glu and Gly) in hepatopancreas (P < 0.05), which might adversely affect crab flavor. The expression of genes related to antioxidant capacity, protein transport, TOR pathway and lipid metabolism was significantly downregulated by increasing dietary CPC level (P < 0.05). In conclusion, based on the quadratic regression analysis of FCR and PER, the optimal replacement levels of fish meal with CPC in crab diet containing 35% fish meal were 32.36% and 35.38%, respectively. It is recommended that Ile, Leu and Thr be supplemented in addition to Met and Lys in the application of CPC.

Abstract:Natural diets and traditional diets have been gradually replaced with formulated diets for the Chinese mitten crab Eriocheir sinensis culture, but it is unclear what effect the different feeding modes have on digestive physiology and metabolism of E. sinensis. This study investigated the effects of three feeding modes, formulated diets (FD, feeding with formulated diets), natural diets (ND, feeding with trash fish and fresh snail), and traditional diets (TD, feeding with soybean, wheat, broad bean, and corn) on the metabolism, antioxidant, and immune capacity of adult male E. sinensis. The results showed that the highest activities of lipase, trypsin, γ-glutamyltransferase, superoxide dismutase, acid phosphatase, alkaline phosphatase, and the contents of urea and malondialdehyde in the hepatopancreas, as well as the levels of triglyceride, uric acid, and malondialdehyde in the hemolymph supernatant, were detected in the ND mode, while the highest hemolymph supernatant free cholesterol was detected in the TD mode. The hemolymph supernatant glucose content of males from the FD mode was significantly lower than that of the ND and TD modes. Metabolomics analysis showed that feeding mode induced significant differences in the metabolite profiles of the hepatopancreas and hemolymph supernatant, where differential metabolites were mainly involved in amino acid and glucose metabolism. Compared with the ND and TD modes, the FD mode was more conducive to the digestion, absorption, and deposition of amino acids and glucose. Overall, this study demonstrated that feeding formulated diets that contain more balanced nutrient profiles improved nutrient utilization, antioxidant capacity, and immune indices of adult male E. sinensis.

Abstract:The present study was conducted to evaluate the effectiveness of dietary β-1,3/1,6-D-glucan,derived from Saccharomyces cerevisiae,and different feeding schedule in modulating the survival,growth,development,non-specific immunity and the resistance to Vibrio parahaemolyticus for Chinese shrimp(Fenneropenaeus chinensis) larvae.The β-glucan was administered orally at concentrations 0.25% of dietary dry matter,which were fed to the larvae in three different feeding schedules: D0(permanently control diet);D1(permanently 0.25% glucan-containing diet);3D1-3D0(three days 0.25% glucan-containing diet-three days control diet);1D1-3D0(one day 0.25% glucan-containing diet-three days control diet).So there were 4 treatments each treatment was randomly assigned to triplicate tanks,and each tank was stocked larvae at the density of 100 L-1.The results showed that survival rate,development index(DI),body length and SGR of larvae fed glucan-0.25% diet were higher than those of larvae of other three groups,although not always significant.Immune parameters, such as phenoloxidase(PO),superoxide dismutase(SOD),acid phosphatase(ACP) and alkaline phosphatase(ALP) of larvae were examined at 9 and 14 dph.The results suggested that these immune parameters of larvae fed glucan-0.25% diet were higher than those of larvae of other three groups,although not always significant.A pathogenic challenge at 14 dph showed that cumulative mortality for glucan-0.25% diet treatment was significantly higher than glucan-free diet treatment(P<0.05),but not significant compared with other two treatments fed in different feeding schedule.Results of the present study suggested that different feeding schedule is not suitable to shrimp larvae,since the highest values for different parameters examined were always observed in larvae fed permanent glucan-0.25% diet,the supplementation of β-glucan to microbound diet at concentration of approximately 0.25% and permanent feeding are recommended for the culture of shrimp larvae.

Abstract:Ammonia nitrogen is one of the most common environmental stress factors in aquaculture. The aim of this experiment was to evaluate the effects of ammonia nitrogen stress on ammonia metabolism, digestion and immune function as well as the protective effects of dietary N-carbamylglutamate (NCG) in the Chinese mitten crab (Eriocheir sinensis). Two feeds were used, namely a control feed (regular commercial feed) and an NCG feed (NCG (302.96 ± 4.07) mg/kg added). Subsequently, animals fed by these two feeds were exposed to 30 mg/L or 60 mg/L ammonia nitrogen stress (named A30 or A60) for 21 days, respectively. The rate of hemolymph ammonia excretion was significantly increased in NCG group compared to C (control) group (P < 0.05). Hemolymph ammonia levels were significantly lower in NCG A30 (NCG feed and A30) and NCG A60 (NCG feed and A60) groups compared to A30 (control feed and A30) and A60 (control feed and A60) groups (P < 0.05). Glutamate dehydrogenase and glutamine synthetase activities in the gills and hepatopancreas of NCG A60 group were significantly increased compared with the A60 group, which promoted glutamine synthesis (P < 0.05). The cumulative mortality rate of Eriocheir sinensis in NCG A30 and NCG A60 groups were effectively reduced (P < 0.05). The arginine levels of hepatopancreas were significantly increased in NCG A60 group (P < 0.05). Under ammonia nitrogen stress, NCG significantly increased the activities of trypsin, pepsin, lipase and α-Amylase in NCG feed group, and promoted feeding (P < 0.05). In hemolymph, hepatopancreas and intestine, NCG significantly increased nitric oxide synthase activity, lysozyme and phenol oxidase activities compared to the ammonia nitrogen stress group (P < 0.05). The results indicated that, NCG added in feed could decelerate the toxic effects of ammonia on Eriocheir sinensis by promoting ammonia excretion and accelerating glutamine and endogenous arginine synthesis by metabolism. Moreover, the activity of digestive enzymes and immune-related enzymes is improved.

Abstract:IMPORTANCE Childhood obesity is an important public health problem with increasing prevalence. Because treatment often has limited success, new approaches must be identified. OBJECTIVE To evaluate the effectiveness and safety of metformin for treating obesity in children aged 18 years and younger without a diagnosis of diabetes mellitus. EVIDENCE REVIEW We included randomized clinical trials identified through searches of MEDLINE, the Cochrane Library, and ClinicalTrials.gov. Our primary outcome measure was change in body mass index (BMI, calculated as weight in kilograms divided by height in meters squared). We assessed study quality, pooled data using a random-effects model, and performed subgroup and sensitivity analyses. FINDINGS Fourteen randomized clinical trials were eligible. For BMI, moderate-strength evidence indicated a reduction of -1.38 (95% CI, -1.93 to -0.82) from baseline compared with control at 6 months. A similar, if less dramatic, effect was observed in studies less than 6 months, but the pooled estimate from studies of 1 year of treatment was not statistically significant. Subgroup analyses indicated smaller, but significant, effects for those with baseline BMI below 35, those of Hispanic ethnicity, those with acanthosis nigricans, those who had tried and failed diet and exercise programs, and in studies with more girls or higher mean age (adolescents). Moderate-strength evidence indicated that with metformin, 26% reported a gastrointestinal event compared with 13% in control groups (relative risk, 2.05; 95% CI, 1.19-3.54), although there was no difference in discontinuations due to adverse events. No serious adverse events were reported. CONCLUSIONS AND RELEVANCE Metformin provides a statistically significant, but very modest reduction in BMI when combined with lifestyle interventions over the short term. A large trial is needed to determine the benefits to subgroups or impacts of confounders. In the context of other options for treating childhood obesity, metformin has not been shown to be clinically superior.

Abstract:This study aimed to evaluate the impact of substituting a portion of feed with Tenebrio molitor (TM) and Elodea nuttallii (EN) on crayfish culture. A total of 270 crayfish (5.1 ± 0.4 g) were fed three different diet combinations (A: 100% feed; B: 80% feed + 10% TM + 10% EN; C: 75% feed + 15% TM + 10% EN) for 12 weeks. The findings demonstrated that group C had an important beneficial impact on the growth performance of crayfish. This was evidenced by a rise in digestive enzyme activity (trypsin, lipase, and cellulase) in the intestinal and hepatopancreas, as well as an upregulation in the expression of growth-related genes (ghsr, igfbp7, mhc, mlc1, mef2, and pax7) in the muscle. Furthermore, the assessment of the flesh quality of crayfish muscle in group C was conducted. The findings indicated a significant increase (p < 0.05) in the energy value (moisture, crude protein, and crude lipid) within the muscle. The levels of delicious amino acids (Glu, Ala, Ser, Gly, and Tyr) and polyunsaturated fatty acids (ARA, DHA) were enhanced, resulting in an improved nutritional profile and flavor of the muscle while maintaining the Σn-3/Σn-6 ratio. The remodeling of the intestinal microbiota (abundance of Proteobacteria and ratio of Firmicutes/Bacteroidota bacteria) also revealed improved growth performance. Additional research is necessary to ascertain whether excessive use of TM or EN feed substitution can have negative effects on crayfish culture.

Abstract:The effects of selenium (Se)-biofortified corn on the total Se contents, the antioxidant enzyme levels, and the amino acids composition in Chinese mitten crab (Eriocheir sinensis) during the stage of the fifth shelling to maturity were investigated in the present study. The culture density of crabs was 600 per 667 m², and they were continuously fed 120.4 mg Se from Se-biofortified corn per 667 m² every two days for 90 days. The results showed that the total muscle Se levels in the crabs were significantly increased (p < 0.05). Activities of hemolymph supernatant enzymes including alkaline phosphatase (AKP), lysozyme (LZM), glutathione peroxidase (GPx), and superoxide dismutase (SOD) were also enhanced (p < 0.05). The protein and crude fat levels at maturity were higher than those at the fourth molt. The levels of total essential amino acids (∑EAAs) and total delicious amino acids (∑DAAs) were significantly increased (p < 0.05). We demonstrate that the feeding of Se-biofortified corn could significantly improve total muscle Se concentrations and hemolymph supernatant antioxidant enzyme activities in Chinese mitten crab, and slow down the rapid decline of ∑EAAs and ∑DAAs at maturity, thus improving the nutritional quality of Chinese mitten crab.

Abstract:Moringa oleifera is renowned for its high antioxidant activity. However, few studies have been conducted on its effects on aquatic animals. The aim of this experiment was to investigate the optimal fermentation process of M. oleifera leaves and to evaluate the effects of fermented M. oleifera leaves on crayfish (9.11 ± 0.3 g) in terms of growth performance, antioxidant capacity, and gut microbiological parameters. By optimizing the fermenting material/water ratio, fermentation time, temperature, and strain, the optimal fermentation conditions of a 10% water ratio + 48 h + 30 °C + inoculation with 2% B. amyloliquefaciens (107 CFU mL−1) were obtained. These conditions resulted in notable increases in the contents of the total protein, total phenols, flavonoids, and amino acids (p < 0.05) while also leading to a notable decrease in the content of tannins in contrast to those of unfermented M. oleifera leaves (p < 0.05). The fermented M. oleifera (FMO) leaves were incorporated at five concentrations, including 0% (control (CT)), 0.25% (0.25FMO), 0.5% (0.5FMO), 1% (1FMO), and 2% (2FMO). The results showed that the 1FMO group performed better in terms of the final body weight (FBW), weight gain rate (WGR), and specific weight gain rate (SGR) compared with the CT group (p < 0.05). In addition, amylase and lipase activities were significantly higher in the 1FMO and 2FMO groups compared with the other groups (p < 0.05). The fermented M. oleifera leaves significantly increased the catalase (CAT) activity in the crayfish (p < 0.05). The superoxide dismutase (SOD) activity was significantly increased in the 0.25FMO, 1FMO, and 2FMO groups, and the malondialdehyde (MDA) content was significantly decreased while the glutathione peroxidase (GSH-PX) content was significantly increased in the 0.5FMO, 1FMO, and 2FMO groups (p < 0.05). Furthermore, the 1FMO group was observed to significantly increase the abundance of Firmicutes while simultaneously reducing the abundance of Aeromonas (p < 0.05) and adjusting the structure of the intestinal microbiome. In conclusion, this study established the optimal fermentation conditions for M. oleifera and obtained a product with high nutrient and low tannin contents. Furthermore, the incorporation of 1% FMO was demonstrated to facilitate growth, enhance the antioxidant capacity, and optimize the gut microbiology in crayfish.

Abstract:The fattening of immature female Chinese mitten crab (Eriocheir sinensis) with high quality diets is an effective mechanism for improving ovarian development and nutritional quality. This study investigated the ovarian development and nutritional quality of female E. sinensis during an 80-day fattening period. The results showed that the gonadosomatic index (GSI) and total edible yield of female E. sinensis increased in a stepwise manner over the course of the fattening period, in synchrony with a decrease in feed consumption and feed cost. Crude protein, total lipid and total carbohydrate in ovaries increased significantly over the course of the fattening period, with the highest crude protein content in both the hepatopancreas and meat observed at Day 40. There was a stepwise increased in the content of triacylglycerol (TAG) and cholesterol (CHO) and phospholipid (PL) in ovary and meat, respectively. On the contrary, the content of free fatty acid (FFA) and PL in ovaries and hepatopancreas as well as the meat TAG and FFA decreased significantly over the course of the fattening period. In ovaries, the highest concentration of polyunsaturated fatty acid (∑PUFA) was observed at Day 60, whilst the highest total monounsaturated fatty acid (∑MUFA) in meat was detected at Day 40. The highest content of ∑MUFA in hepatopancreas, and the ∑PUFA and total long chain polyunsaturated fatty acid (∑LC-PUFA) in meat were detected at Day 80, while the highest hepatopancreas ∑PUFA was detected at Day 0. The content of total essential amino acids (∑EAA) and total amino acids (TAA) in ovaries and meat increased significantly during the fattening period of Day 0 to Day 40, while no significant increase was detected on the TAA during the period of Day 40 to Day 80. Essential amino acid scores (EAAS) of ovary and meat were >100 for all essential amino acids (EAA), and fattening culture did not significantly improve EAAS for ovary and meat. In conclusion, the fattening could improve the total edible yield and nutritional values of female E. sinensis. Considering the fattening performance and feed cost, the appropriate fattening period should be 40–60 days for pond-reared female E. sinensis post puberty molting.

Abstract:Shewanella putrefaciens is a vital bacterial pathogen implicated in serious diseases in Chinese mitten crab Eriocheir sinensis. Yet the use of probiotics to improve the defense ability of E. sinensis against S. putrefaciens infection remains poorly understood. In the present study, the protective effect of dietary R. sphaeroides against S. putrefaciens infection in E. sinensis was evaluated through antioxidant capability, immune response, and survival under bacterial challenge assays, and its protective mechanism was further explored using a combination of intestinal flora and metabolome assays. Our results indicated that dietary R. sphaeroides could significantly improve immunity and antioxidant ability of Chinese mitten crabs, thereby strengthening their disease resistance with the relative percentage survival of 81.09% against S. putrefaciens. In addition, dietary R. sphaeroides could significantly alter the intestinal microbial composition and intestinal metabolism of crabs, causing not only the reduction of potential threatening pathogen load but also the increase of differential metabolites in tryptophan metabolism, pyrimidine metabolism, and glycerophospholipid metabolism. Furthermore, the regulation of differential metabolites such as N-Acetylserotonin positively correlated with beneficial Rhodobacter could be a potential protection strategy for Shewanella infection. To the best of our knowledge, this is the first study to illustrate the protective effect and mechanism of R. sphaeroides supplementation to protect E. sinensis against S. putrefaciens infection.
