Abstract:INTRODUCTION: The versatile multicomponent Mannich reaction occupies a salient position in organic chemistry and drug design. Sound knowledge of its scope and variations and of the biological activities of Mannich bases is crucial for the development and improvement of drugs for various diseases. Areas covered: The following article provides an overview of the latest developments in the field of drugs based on the Mannich reaction. Web-based literature searching tools such as PubMed and SciFinder were applied to obtain useful articles. In addition, pertinent literature that was recently published by the authors is discussed in this manuscript. The chemical structures of bioactive Mannich bases are also given. Expert opinion: The Mannich reaction represents a feasible and cost-effective procedure with great potential for drug development. Several newly discovered Mannich bases exhibit sound activities against various human diseases as well as favorable pharmacokinetics. Thus, scientific research about Mannich bases is prospering and appears very attractive both for chemists and for clinicians.

Abstract:The biological activity of Mannich bases, a structurally heterogeneous class of chemical compounds that are generated from various substrates through the introduction of an aminomethyl function by means of the Mannich reaction, is surveyed, with emphasis on the relationship between structure and biological activity. The review covers extensively the literature reports that have disclosed Mannich bases as anticancer and cytotoxic agents, or compounds with potential antibacterial and antifungal activity in the last decade. The most relevant studies on the activity of Mannich bases as antimycobacterial agents, antimalarials, or antiviral candidates have been included as well. The review contains also a thorough coverage of anticonvulsant, anti-inflammatory, analgesic and antioxidant activities of Mannich bases. In addition, several minor biological activities of Mannich bases, such as their ability to regulate blood pressure or inhibit platelet aggregation, their antiparasitic and anti-ulcer effects, as well as their use as agents for the treatment of mental disorders have been presented. The review gives in the end a brief overview of the potential of Mannich bases as inhibitors of various enzymes or ligands for several receptors.

Abstract:Twelve Mannich base derivatives of 2'-hydroxychalcone were synthesized through Mannich reaction, and the regioselectivity of the reaction occurred preferentially at C-3' position of B ring of chalcone backbone. All the target compounds have been identified by H-l NMR, IR spectra and elemental analyses, and further confirmed by X-ray analysis. The characteristic of this reaction was simple process, mild condition, easy purification and middle to good yields (49%similar to 72%). Some compounds showed potent CDK1/cyclin B inhibitory activities and most of compounds displayed a significant growth inhibitory action against various cancer cell lines.

Abstract:Background: Multicomponent reactions are highly useful in synthesizing natural products and bioactive molecules. Out of several MCRs, although utilized widely, some remain neglected in review articles. The Gewald and Groebke-Blackburn-Bienaymé (GBB) reactions are two such reactions. This comprehensive review assimilates applications of Gewald and Groebke-Blackburn- Bienayme reactions in synthesizing novel antimicrobial agents. It presents the antimicrobial properties of the synthesized molecules, providing an overview of their potential druggability. Objective: Developing novel antimicrobial agents is the need of the hour. Toward this objective, the scientific community is developing new methods for constructing novel architectures with potential antimicrobial properties. This review will showcase the usefulness of the Gewald, Strecker, and Groebke-Blackburn-Bienaymé (GBB) reactions in synthesizing antimicrobial molecules. Method: The articles are searched by using the Sci-finder search tool and summarize the chemistry of their synthesis and antimicrobial evaluation of the molecules Results: This review focuses on synthesizing antimicrobial molecules using the Gewald, Strecker, and Groebke-Blackburn-Bienaymé (GBB) reactions. The antimicrobial activities of the synthesized molecules are also summarized in tables. Conclusion: This review will briefly overview the application of the Gewald, Strecker, and Groebke- Blackburn-Bienaymé (GBB) reactions in synthesizing novel antimicrobial molecules. It contains several molecules with promising activity against resistant and non-resistant microbial strains. These promising molecules could be studied further to develop novel antibiotics.

The takeover of Prague’s banking cluster by multinational groups from an evolutionary perspective

Abstract:This article identifies the main drivers in the evolution of the banking cluster in Prague over the last two centuries. Conceptually, it employs the adaptive-cycle model of cluster evolution, which acknowledges the role of external factors in cluster evolution. An empirical analysis shows that the evolution of the banking cluster in Prague has been primarily driven by several episodes of major external disruptions. A particular attention is paid to the latest phase of cluster evolution, which started around the beginning of the 21st century when Prague’s banks were taken over by foreign multinational groups. We argue that, despite the numerous costs and risks associated with this “subsidiarisation” of the banking cluster in Prague, there were benefits, some of which were surprisingly manifested during the 2008–2009 global economic crisis. key words banking cluster evolution – adaptive-cycle model – financial centres – Central and Eastern Europe – global economic crisis blažek, j., bečicová, i. Abstract:Abstract Alkylamines are ubiquitous in pharmaceuticals, materials and agrochemicals. The Mannich reaction is a well-known three-component reaction for preparing alkylamines and has been widely used in academic research and industry. However, the nucleophilic components in this process rely on C(sp 2 )−H and activated C(sp 3 )−H bonds while the unactivated C(sp 3 )−H bonds involved Mannich alkylamination is a long-standing challenge. Here, we report an unprecedented multicomponent double Mannich alkylamination for both C(sp 2 )−H and unactivated benzylic C(sp 3 )−H bonds. In this process, various 3-alkylbenzofurans, formaldehyde and alkylamine hydrochlorides assemble efficiently to furnish benzofuran-fused piperidines. Mechanistic studies and density functional theory (DFT) calculations revealed a distinctive pathway that a multiple Mannich reaction and retro-Mannich reaction of benzofuran and dehydrogenation of benzylic C(sp 3 )−H bonds were key steps to constitute the alkylamination. This protocol furnishes a Mannich alkylamine synthesis from unusual C–H inputs to access benzofuran-fused piperidines with exceptional structural diversity, molecular complexity and drug-likeness. Therefore, this work opens a distinctive vision for the alkylamination of unactivated C(sp 3 )−H bonds, and provides a powerful tool in diversity-oriented synthesis (DOS) and drug discovery.

Abstract:An analysis of chemical reactions used in current medicinal chemistry (2014), three decades ago (1984), and in natural product total synthesis has been conducted. The analysis revealed that of the current most frequently used synthetic reactions, none were discovered within the past 20 years and only two in the 1980s and 1990s (Suzuki-Miyaura and Buchwald-Hartwig). This suggests an inherent high bar of impact for new synthetic reactions in drug discovery. The most frequently used reactions were amide bond formation, Suzuki-Miyaura coupling, and SNAr reactions, most likely due to commercial availability of reagents, high chemoselectivity, and a pressure on delivery. We show that these practices result in overpopulation of certain types of molecular shapes to the exclusion of others using simple PMI plots. We hope that these results will help catalyze improvements in integration of new synthetic methodologies as well as new library design.

Abstract:The synthesis and characterization of eugenol derivatives 1‐5 through Mannich condensation. The Mtb‐H37R strain is sensitive to all compounds, especially 1 and 4. Mannich bases 3, 4 and 5 show potent antioxidant activity. The high binding affinity of 4 and 5 with essential enzymes of Mtb, suggesting PanK receptor as potential molecular target. Mannich bases are interesting class of molecules thanks to their various pharmacological properties. This paper presents the synthesis and characterization of eugenol derivatives following Mannich condensation. The in vitro antituberculosis activity of the synthesized derivatives was evaluated by the Resazurin microtiter assay (REMA). The 2,2′‐diphenyl‐1‐picrylhydrazyl (DPPH) free radical scavenging and ferrous cation (FeCl2) chelating tests were used to estimate the antioxidant capacity. All eugenol Mannich bases inhibited the Mycobacterium tuberculosis (Mtb) H37Rv growing. Those of 2‐dimethylaminomethyl‐6‐methoxy‐4‐propenylphenol (1) and 2‐methoxy‐4‐propenyl‐6‐pyrrolidinylmethylphenol (4) were the most active at 10 μg/mL. Additionally, significant antioxidant profile was exhibited by the synthesized derivatives, especially for 2‐dicyclohexylaminomethyl‐6‐methoxy‐4‐propenylphenol (5) that showed an antiradical activity of EC50=20 μg/mL, twice as strong as that of eugenol precursor, and for 2‐methoxy‐6‐methylpiperazinylmethyl‐4‐propenylphenol (3) and compound 4, which were promising iron chelators with EC50 of 0.016 and 0.018 mg/mL, respectively, better than EDTA standard. The molecular docking experiments on two different antituberculosis receptors showed excellent binding affinity of compounds 4 and 5 into the active site of pantothenate kinase (PanK, type I), suggesting PanK as potential molecular target. These encouraging findings showed the importance of Mannich reaction for the development of antitubercular candidates and the improvement of natural products bioactivity.

Abstract:Objective: This article is focused on synthesis and characterisation of 1,1-(((5-(3,4,5triethoxybenzyl)pyrimidine-2,4-diyl))bis(azanediyl))bis(substituted phenyl methylene))bis(naphthalen-2-ol) moieties and evaluated for antibacterial and antioxidant activities as well as molecular docking and molecular dynamic simulation investigations. Methods: In this article we conducted one-pot three component reactions through Betti type reaction. Agar diffusion method was used to evaluate anti-bacterial properties. The DPPH method was used to determine antioxidant activity results. Results: A series of 1,1-(((5-(3,4,5triethoxybenzyl)-pyrimidine-2,4-diyl))bis(azanediyl))bis(substituted phenyl methylene))bis(naphthalen-2-ol) were synthesized via Betti type reaction and evaluated for their antibacterial and antioxidant activities. Further, all the synthesized compounds were characterized by FT-IR, 1 H, and 13 C NMR, and mass spectral analysis. Discussion: Molecular docking was carried out to study possible potential binding interaction of synthesized Betti base derivatives with 1NC6 PDB as receptor. The molecular dynamic (MD) was performed to investigate stability of binding interaction of drug-target (protein–ligand) complex. All the synthesized compounds were investigated for their antibacterial and antioxidant activity. Conclusions: Betti base derivatives were synthesized and studied on the basis of computer aided drug design including docking and molecular dynamic (MD) simulations and pharmacologically assessed for antioxidant and antibacterial activity. Molecular docking investigation revealed that compounds showed better binding energies in the range –4.1 to –7.3 kcal/mol suggests that the binding occurs efficiently in the receptor–ligand complex. These Betti base derivatives were synthesized via Betti base protocol at 70 o C through one-pot three component reaction with acceptable yields, as cost efficient, high yield, easy workup and environmental friendliness. Afterwards, obtained molecules have characterized by FT-IR, NMR, and Mass spectra which confirms the chemical structure of the synthesized compounds.

Abstract:This article emphasizes the importance of prodrugs and their diverse spectrum of effects in the field of developing novel drugs for a variety of biological applications. For the purpose of illustrating the usefulness of the prodrug approach, this review covers examples of prodrugs that have been made available or are now undergoing human trials. Additionally, it included lists of the most common functional groups, carrier linkage, and reactive chemicals that can be used to create prodrugs. The primary chemical carriers employed in the creation of prodrugs, such as esters, amides, imides, NH‐acidic carriers, amines, alcohols, carbonyl, carboxylic, and azo linkage, are also discussed. This review also discusses glycosidic and triglyceride mutually activated prodrugs, which aim to deliver the drugs after bioconversion at the intended site of action. The article also discusses the extensive chemistry and wide variety of applications of recently approved prodrugs, such as antibacterial, anti‐inflammatory, cardiovascular, antiplatelet, antihypertensive, atherosclerotic, and antiviral, etc. In order to illustrate the prodrug and mutual drugs concept's various applications and highlight its many triumphs in overcoming the formulation and delivery of problematic pharmaceuticals, this work represents a thorough guide that includes the synthetic moiety to the reader.

Abstract:The current work aims to generate novel Schiff bases by reacting substituted aldehydes with amine derivatives catalyzed by a natural acid. The developed compounds underwent diverse physicochemical analyses including liquid chromatography-mass spectrometry, Fourier transform infrared spectroscopy, scanning electron microscopy, 1H- and 13C-nuclear magnetic resonance, and X-ray diffraction. Furthermore, differential thermogravimetric, thermogravimetric, and differential thermal analysis techniques were employed in a nitrogen-free environment to determine kinetic parameters. These data were then used in model-free isoconversional methods (e.g., Friedman, Kissinger-Akahira-Sunose, and Flynn-Wall-Ozawa). The Schiff bases were evaluated for their in vitro and in silico α-amylase inhibitory activity. Schiff base-2 displayed the highest inhibition compared with the reference drug acarbose. In comprehensive MTT assay cytotoxicity investigations, both Schiff bases showed strong anticancer capabilities against the human lung cancer cell line (A549). Moreover, this study demonstrated effectiveness of synthetic compounds in screening Caenorhabditis elegans for anti-Alzheimer's and stress resistance properties. The simplicity of its biology allowed precise evaluation of the effect of compounds on neuronal function and stress response. This research enhances drug discovery efforts for Alzheimer's and stress-related disorders, potentially improving patient outcomes.

Abstract:A green and operationally simple approach for the library synthesis of novel bis-heteroarylaminomethylnaphthoquinone derivatives by a one-pot pseudo five-component domino condensation reaction of 5,5′-methylenebis(thiazol-2-amine), 2-hydroxy-1,4-naphthoquinone and various aromatic aldehydes in the presence of formic acid as the catalyst was developed. All reactions were performed in ethanol at room temperature and the products were isolated by simple filtration. The protocol proves to be an efficient and an environmentally benign in terms of good to high yields, readily available starting materials, easy workup, clean reaction profile, mild reaction conditions, green solvent and easy purification of products without chromatography. Some of the newly synthesized compounds were explored for their antimicrobial activities against two gram-positive and two gram-negative bacterial species.Graphical abstract

Abstract:β-Phenylalanine derivatives (β-PAD) represent a structural family of therapeutic interest, either as components of drugs or as starting materials for access to key compounds. As scaffolds for medicinal chemistry work, β-PAD offer the advantage of great diversity and modularity, a chiral pseudopeptidic character that opens up the capacity to be recognized by natural systems, and greater stability than natural α-amino acids. Nevertheless, their synthesis remains a challenge in drug discovery and numerous methods have been devoted to their preparation. This review is an update of the access routes to β-PAD and their various therapeutic applications.

Abstract:Aryl amines are one of the most common moieties in biologically active molecules, and approximately 37% of drug candidates contain aromatic amines. Recent advancements in medicinal chemistry, coined as “escaping from flatland”, have led to a greater focus on accessing highly functionalized C(sp3)-rich amines to improve the physiochemical and pharmacokinetic properties of compounds. This communication presents a modular and operationally straightforward three-component alkyl Petasis boron-Mannich (APBM) reaction that utilizes ubiquitous starting materials, including amines, aldehydes and alkyl boronates. Overcoming the long-standing synthetic hurdle of accomplishing the Petasis boron-Mannich reaction with alkyl boronate coupling partners was achieved via the formation of an electron donor-acceptor (EDA) complex and sequential radical-radical coupling. By adapting this transformation to high-throughput experimentation (HTE), it offers rapid access to an array of diverse C(sp3)-rich complex amines, amenable for facile drug candidate development.

Abstract:INTRODUCTION: Schiff bases are synthetically accessible and structurally diverse compounds, typically obtained by facile condensation between an aldehyde, or a ketone with primary amines. Schiff bases contain an azomethine (-C = N-) linkage that stitches together two or more biologically active aromatic/heterocyclic scaffolds to form various molecular hybrids with interesting biological properties. Schiff bases are versatile metal complexing agents and have been known to coordinate all metals to form stable metal complexes with vast therapeutic applications. Areas covered: This review aims to provide a comprehensive overview of the various patented therapeutic applications of Schiff bases and their metal complexes from 2010 to 2015. Expert opinion: Schiff bases are a popular class of compounds with interesting biological properties. Schiff bases are also versatile metal complexing ligands and have been used to coordinate almost all d-block metals as well as lanthanides. Therapeutically, Schiff bases and their metal complexes have been reported to exhibit a wide range of biological activities such as antibacterial including antimycobacterial, antifungal, antiviral, antimalarial, antiinflammatory, antioxidant, pesticidal, cytotoxic, enzyme inhibitory, and anticancer including DNA damage.

Abstract:Schiff base complexes with various metals proved out to be excellent bioactive agents in pharmaceutical and medicinal chemistry along with catalytic role in organic reactions also. Earlier reported conventional methods include refluxing at higher temperature, use of harmful solvents and more time consuming reaction procedures. Recently, several research papers have been published with novel green methodologies utilizing photo or nano catalyst, use of greener solvents like aqueous media or solvent free conditions with sustainable synthetic techniques such as microwave and ultrasound-irradiation. So keeping in mind the fast demand of Schiff base enriched motifs in the field of pharmaceutical chemistry as bioactive compound herein this review, we summarize the advancement in the synthetic methods firstly focusing on green methodologies along with recently developed Schiff bases motifs with conventional strategies also. In addition, the catalytical role and the anti-bacterial, anti-microbial, anti-cancer and anti-oxidant studies of Schiff base metal complexes appended with Cu, Co, Ni, Fe, Mn, Pd, Sn, Cd, Zn, Ru, Pt and Zr have also been discussed in the present review article.
