Screening of Poria cocos polysaccharide with immunomodulatory activity and its activation effects on TLR4/MD2/NF-κB pathway

Abstract:PCP-W1, the Poria cocos polysaccharide with the strong immunomodulatory activity, was isolated through column chromatography and screened for in vitro immune activity in RAW 264.7 cells in this study. The structure analysis results revealed that the PCP-W1 were composed of galactose, glucose, fucose and mannose in a molar percentage of 35.87: 28.56: 21.77: 13.64. And it exhibited a random coil and branched conformational features with a molecular weight of 18.38 kDa. The main chain consisted of residues→3)-β-D-Glcp-(1 → 3,6)-β-D-Glcp-(1 → 3)-β-D-Glcp-(1 → 6)-β-D-Glcp-(1 → 6)-α-D-Galp-(1 → 6)-α-D-Galp-(1 → 2,6)-α-D-Galp-(1→6)-α-D-Galp-(1 → 6)-α-D-Galp-(1 → , while branching occurred at β-D-Glcp-(1→, α-D-Manp-(1→, and α-L-Fucp-(1 → 3)- α-L-Fucp-(1→. The pharmacodynamic studies demonstrated that PCP-W1 activated the release of NO, IL-6, IL-β, TNF-α, CD86, and ROS to induce polarization of RAW 264.7 murine macrophages towards M1-type through modulation of the TLR4/MD2/NF-κB pathway. The molecular docking results showed that PCP-W1 could primarily dock onto the hydrophobic binding site of TLR4/MD2 complex via its galactose chain. Furthermore, molecular dynamics simulation displayed stable modeling for TLR4-MD2-PCP-W1 complex. Overall, we screened the most immunoactive components of the polysaccharide, analyzed its structure, demonstrated its impact on TLR4/MD2/NF-kB pathway, and studied the interaction between TLR4/MD2 and the polysaccharide fragments. These results provide further support for the structure-activity relationship study of the immunomodulatory effects of Poria cocos polysaccharide.

Abstract:The current experiments were designed to explore the structural features and immunopotentiation activity of two homogeneous polysaccharides PCSPA and PCSPB prepared from Crocus sativus petals using DEAE-Sephadex A-50 and Sephadex G200 column chromatography. The structures of PCSPA and PCSPB were systematically characterized using extensive chemical and spectroscopic methods including colorimetry, HPGPC-RID, GC-MS, Smith degradations, methylation, solvolyticdesulfation, UV, FT-IR, NMR, SEM, and AFM. The average molecular weights of PCSPA and PCSPB were 1.98 x 10(6) and 2.53 x 10(6) Da, respectively. PCSPA consisted of Gal, Rha, Ara, and Xyl in the molar ratio of 16:5:7:3, while PCSPB were composed of Gal, Glc, Man, Rha, Ara, and Xyl with molar ratio of 16:2:7:19:15:16. Both polysaccharides contained sulfonic and acetyl groups. PCSPA and PCSPB significantly activated RAW264.7 cells by enhancing the phagocytic activity, up-regulating the expression of surface molecules, promoting the production and mRNA expression of cytokines and chemokines via MAPK and NF-kappa B pathway. Abstract:A water-soluble glucomannogalactan was isolated from P. geesteranus fruit bodies by hot water extraction and column chromatography method. The structure, chain conformation and immune effect of PGP-1c (20.9 kDa) were investigated comprehensively based on high purity and neutral sugar content (94.7 ± 0.5%). Confirmed by monosaccharide composition, methylation and NMR analysis, PGP-1c was composed of unsubstituted (1,6-α-Gal and 1,6-α-3-OMe-Gal) and monosubstituted (1,2,6-α-Gal and 1,2,6-α-3-OMe-Gal) galactose units in the backbone through α-(1 → 6) glycosidic bonds and the possible branches that a long-branched chain composed of →3)-Glc-(1 → Man and a few fucoses were connected at the O-2 of galactose with a branching degree of 48%. Through the conformational behavior of the molecular chain, it was observed that PGP-1c might have a long-branched or stacked macromolecular network caused by O-CH3 groups. Moreover, PGP-1c could promote the secretion of NO and cytokines significantly in a dose-dependent manner, which indicated a good immune-enhancing effect.

Abstract:PAP1-A, a novel heteropolysaccharide with an average molecular mass of 1.35×105Da, was isolated from Pteridium aquilinum using a combination of chromatography by DEAE Sepharose Fast Flow and Sepharose 4B. The monosaccharide component of PAP1-A was L-rhamnose, L-arabinose, L-fucose, D-xylose, D-mannose, D-glucose and D-galactose in the molar ration of 1.82: 1.53: 1.42: 1.31: 5.24: 1: 12.35. The predicted structure of PAP1-A was established to be a complex polysaccharide with a main chain consisted of α-(1→4)-linked galactose partially substituted at O-6 position, with the (1→2)-linked xylose, (1→3)-linked arabinose, (1→3)-linked rhamnose, (1→3,6)-linked mannose, and (1→6)-linked mannose, as branches. Fucose, glucose, mannose, and rhamnose were located at the termini of the branches. The immunomodulatory activity assay showed that PAP1-A could significantly promote the RAW264.7 cells proliferation and induce the production of NO from RAW264.7 cells. Therefore, PAP1-A shows as a potent immunomodulator with potential applications in the medical and food industries.

Abstract:To find the polysaccharide with hepatoprotective activity from Poria cocos and clarify its structure, a galactoglucan (PCP-1C) with a molecular weight of 17 kDa was purified from the Poria cocos sclerotium by column chromatography and activity evaluation in the present work. It was composed of galactose, glucose, mannose, and fucose in a molar percentage of 43.5: 24.4: 17.4: 14.6. Structural characterization showed that PCP-1C has a backbone consisted of 1,6-α-D-Galp, which branches composed of 1,3-β-D-Glcp, 1,4-β-D-Glcp, 1,6-β-D-Glcp, T-β-D-Glcp, T-α-D-Manp, T-α-L-Fucp and 1,3-α-L-Fucp. In vivo experiments found that PCP-1C can apparently improve the damage of liver tissue in CCl4-treated mice and relieve oxidative stress and inflammation. PCP-1C also reduced the expression of CAR and CYP2E1 in the liver. These findings indicated strong hepatoprotective effect of PCP-1C, which was attributed to the reduction of CCl4 metabolism via inhibiting the CAR/CYP2E1 signal pathway.

Abstract:Poria cocos polysaccharides (PCPs) are fungal polysaccharides derived from the traditional Chinese medicine Poria cocos . They are considered an important active ingredient for their pharmacological activity. Herein, the extraction, separation and purification, structure, and application of PCPs are reviewed. Additional research is necessary to fully understand the advanced structure of PCPs, which has implications for their structure–activity relationship. Their application mostly involves the medical industry, with less involvement in other fields. This article highlights the current research status on PCPs in the above-mentioned areas and some problems that need to be solved in future research. Additionally, it points the way for further studies on PCPs in the hopes that they will be more widely and realistically used in various industries.

Abstract:Paulownia tomentosa flower polysaccharide (PTFP) from dried cultured P. tomentosa flowers, is widely known for its immunomodulatory activities. Here, PTFP was extracted from Paulownia tomentosa flower using hot water extraction, followed by ethanol precipitation methods. Structural characterization of PTFP was revealed by scanning electron microscope, high-performance anion-exchange chromatography, gel chromatography, ultraviolet and infrared spectral. Meanwhile, adjuvant action of PTFT on the immune responses to classical swine fever vaccine in mice was evaluated to further proclaim the immune regulatory effect of PTFP. The results showed that PTFP was a type of heteropolysaccharide with a dense, rough surface and high molecular weight (667.02 kDa), mainly composed of glucose (30.93%), rhamnose (29.99%), galactose (15.66%), arabinose (6.95%), mannose (5.52%), and xylose (4.80%). The results of gel chromatography suggested that the molecular configuration of PTFP may be a spherical structure. The infrared spectrum results confirmed that the functional groups and chemical bond of PTFP contained -OH, O-H, C-H, C=O, C-O, etc. Moreover, PTFP exhibited obvious immune enhancement effect by improving concanavalin A (ConA), lipopolysaccharide (LPS), and CSFV E2-stimulated splenocyte growth and natural killer cell activity in CSFV-immunized mice. Similarly, the titers of CSFV E2-specific IgG, IgG1, IgG2a, and IgG2b antibodies and IFN-γ and IL-10 levels in CSFV-immunized mice were distinctly increased by PTFP treatment. Overall, PTFP was a macromolecular heteropolysaccharide primarily containing glucose and rhamnose, and possessed the auxiliary effect of immune enhancement on the immune responses to classical swine fever vaccine.
The differences between the water- and alkaline-soluble Poria cocos polysaccharide: A review

Abstract:Poria cocos (PC) refers to a fungal species which is also known as "Fuling" in China. For >2000 years, PC has demonstrated its therapeutic values as a kind of traditional medicine. It is believed that the various biological benefits created by PCs highly rely on the Poria cocos polysaccharide (PCP). This review recapitulates the recent progress made in PCP in four aspects: i) the methods of extraction, separation, and purification, ii) structural characterization and identification, iii) the related bioactivities and mechanism of action, and iv) structure-activity relationships. Through discussion about the objective as mentioned above, it can be found out that PCP is categorized into water-soluble polysaccharide (WPCP) and alkaline-soluble polysaccharide (APCP), which are totally different in structure and bioactivity. The structures of WPCP are multiplicity whose backbone can be (1,6)-α-galactan and (1,3)-β-mannoglucan etc. to perform various bioactivities including anti-tumor effect, anti-depressant effect, anti-Alzheimer effect, anti-atherosclerosis effect, hepatoprotection etc. The structures of APCP are much more single with backbone of (1,3)-β-D-glucan and the studies of activity concentrate on anti-tumor effect, anti-inflammatory effect and immunomodulation. Besides, the future opportunities of WPCP are primary structure identification. For APCP, scholars can focus on the conformation of polysaccharide and its relationship with activity.

Abstract:Polygonatum cyrtonema Hua. (PC) is a traditional Chinese herb with a history of use in both food and medicine. For clinical use, processed PC pieces are most commonly used, while present research has focused on crude PC polysaccharides (PCPs). In this study, a new polysaccharide, PCP-F1, with a molecular weight of 37.46 kDa, was separated from four-time processed PCPs by column chromatography and evaluated by antioxidant activity. It was composed of glucose, mannose, galactose, rhamnose, and galacturonic acid with a molar ratio of 3.5:2.5:1.3:1.8:0.8. The methylation analysis and two-dimensional NMR measurement revealed that the configuration of PCP-F1 contained nine residues in the primary structural unit by the chain of →3)-α-D-Glcp, →2)-α-D-Glcp(6→, →1)-ꞵ-D-Glcp(2→, →2)-α-D-GalAp(3,4→, →1) -ꞵ-D-Manp(3→, →2)-α-D-Glcp(3→, branched for →3)-α-D-Glcp, →2)-ꞵ-D-Galp(4→, →1)-ꞵ-D-Glcp(2→, →2,4)-α-D-Manp(6→, →3)-α-L-Rhap(4→. Radical scavenging assays indicated that PCP-F1 could scavenge radicals with a high scavenging rate, suggesting PCP-F1 possesses good antioxidant activity. The study confirms the importance of processed PC and offers the potential for exploiting it as a functional food.

Abstract:Pleurotus placentodes , a fungus, belongs to the Pleurotaceae family. The aim of the present study was to characterize the structure of a novel polysaccharide from fruiting bodies of P. placentodes (PPp-W) and evaluate its anticoagulant activity in vitro . The high-performance liquid chromatography and GC–MS analysis indicated that PPp-W with a molecular weight of 27.4 kDa was mainly composed of mannose (17.56%), glucose (6.37%), galactose (44.89%), and fucose (1.22%) with a certain amount of 3- O -methyled galactose. SEM, XRD, and AFM combined with Congo red test revealed that PPp-W was an irregular curly sheet with triple-helix conformation. The FT-IR, methylation, and nuclear magnetic resonance analysis indicated that PPp-W contained→6)-α-D-Gal p -(1→, →6)-3- O -Me-α-D-Gal p -(1→and →2, 6)-α-D-Gal p -(1→ as main chain, partially substituted at O -2 and O -6 by non-reducing ends of β - D - Man p -(1→ and β - L-Fuc p -(1→ with a small amount of α-1,3-linked-Glc p in backbone. PPp-W could significantly prolong APTT (12.9 ± 0.42 s, p < 0.001) and thrombin time (39.9 ± 0.28 s, p < 0.01) compared with the control group (11.45 ± 0.071 s and 38.05 ± 0.21 s), which showed that PPp-W had anticoagulant activity. These studies suggested that PPp-W was a 3- O -methylated heteroglycan and might be suitable for functional foods and natural drugs as an anticoagulant ingredient, which provided a basis for the application of polysaccharides from P. placentodes .

Abstract:Traditional Chinese medicine polysaccharides have numerous biological activities with broad applications in the biomedical industries. However, a clear understanding of the pharmacological activities of compound polysaccharides with multi-component structures remain challenging. This study aimed to investigate the immune boosting effect of compound polysaccharides on the influenza vaccine and assess the preliminary structure-activity relationship. The compound polysaccharide (CP) was isolated from the combined Chinese herbs lentinan, pachymaran and tremellan, and purified by gradient ethanol precipitation to obtain its subcomponents of CP-20, CP-40, CP-60, and CP-80 with decreasing molecular weights. These polysaccharides were mainly composed of glucans with different linkage patterns, including α-(1 → 3)-glucan, α-(1 → 4)-glucan and β-(1 → 6)-glucan. A significant improvement was observed in the survival of mice vaccinated with inactivated (IAV) vaccine and the isolated polysaccharides as adjuvants. A reduction in the pulmonary virus titer and weight loss were also observed. Moreover, CP-40 and CP-60, as well as the original CP, significantly enhanced the serum anti-IAV antibody titers and interleukin IL-2, IL-5, and IL-6 concentrations. These preliminary results indicate the immune boosting effect of the compound polysaccharides is highly relevant to the specific structural properties of the subcomponent, and CP-40 is worthy of further exploration as a glycan adjuvant for the IAV vaccine.

Abstract:A homopolysaccharide fraction (PEP-1A) with desired immunomodulatory activity was isolated from culture broth of Parabacteroides distasonis. Structural characterization uncovered that PEP-1A had a molecular weight of 3.40 × 106 Da and possessed mannose merely. Comprehensive analysis of FT-IR, GC-MS and 1D/2D NMR confirmed that the structure of PEP-1A was expected as follows:Finally, the activity assays on RAW 264.7 macrophages showed that PEP-1A could promote proliferation, increase the production of reactive oxygen species (ROS), enhance the phagocytosis, and promote the secretion of nitric oxide (NO) and inflammatory cytokines including IL-1β, IL-6 and TNF-α. Furthermore, PEP-1A could give play to immunoregulation effect through NF-κB, MAPK and Akt signaling pathways. Collectively, the study of PEP-1A offers a molecular underpinning for the future application of PEP-1A as a potential immunostimulant.

Abstract:A polysaccharide isolated from spores of the fungus, Ganoderma lucidum, was found to be a complex glucan. On the basis of compositional and methylation analyses, periodate oxidation, Smith degradation, 1D and 2D NMR, and ESIMS experiments of the native polysaccharide and its degraded products, the polysaccharide was shown to have a backbone of beta-(1-->3)-linked D-glucopyranosyl residues, with branches of mono-, di- and oligosaccharide side chains substituting at the C-6 of the glucosyl residues in the main chain. Conformational analysis in aqueous solution and immunological activities of the native and degraded glucans were also investigated. The results suggested that the degree of substitution on the main chain and the length of side chains may be very important factors in determining the conformation and the biological activities of beta-(1-->3)-linked glucans.

Abstract:The fine structure and chain conformation of a heteropolysaccharide (PCIPS3) from mycelium of Paecilomyces cicadae were investigated via the analysis of HPLC, IR, methylation, NMR spectroscopy and multiangle light scattering. It was determined to be a 2.23 x 10(4) g/mol heteropolysaccharide primarily composed of glucose, galactose and mannose in a molar ratio of 23.8:2.1:1.0. The PCIPS3 backbone consisted of 1,4-linked alpha-D-Glcp and 1,4-linked 6-O-Me-alpha-D-Glcp residues, which were occasionally interrupted by branched beta-Galf residues through 1,6-linkage. Moreover, the alpha (0.60) from Mark-Houwink-Sakurada (MHS) equation suggested that PCIPS3 adopted a flexible chain conformation in 0.1 mol/L NaNO3 at 25 degrees C. The worm-like chains model parameters for PCIPS3 were estimated as following: M-L = 437 nm(-1), q = 0.46 nm and 0.79 nm, which were further evidenced by AFM. Furthermore, PCIPS3 showed excellent scavenging capacities of 2,2-diphenyl-1-picrylhydrazyl radical, superoxide radical, hydroxyl radical, ORAC radical and moderate immunomodulatory activity.

Abstract:Polysaccharides obtained from mushrooms have been reported to possess immunomodulatory properties. In this study, a water-soluble polysaccharide was purified from the fruiting bodies of Pleurotus abieticola, entitled PAPS1. After its composition and structural analysis, the immunomodulatory activity was investigated in immunosuppressed mice induced by cyclophosphamide (CTX) at a dosage of 70 mg/kg by intraperitoneal injection for 7 days. After 28 days of intragastric administration, PAPS1 alleviated cyclophosphamide (CTX)-induced histopathological damage and increased the expressions of splenic CD4, CD8, CD56 and IgM in the serums of immunosuppressed mice. PAPS1 suppressed the oxidative stress indicated by preventing the increases in ROS and MDA levels. According to the intestinal microflora analysis, PAPS1 regulated 11 bacteria at the gene level, including Helicobacter and Paraprevotella, which are related to immunity and oxidative capacity. Compared with CTX-treated mice, significant increases in immune-related cytokines, such as interleukin (IL)-2, IL-6 and IL-12 in the serums of mice treated with PAPS1, were observed. Finally, PAPS1 can strongly increase the expression of nuclear factor erythroid 2-related factor 2 (Nrf2) and its downstream proteins. In conclusion, PAPS1-boosted immunity may be related to its suppression on oxidative stress via enhancing the activity of Nrf2 signaling. Thus, PAPS1 can be investigated as a candidate for immunomodulatory therapy.

Structure characterization of two functional polysaccharides from Polygonum multiflorum and its immunomodulatory

Abstract:Two purified polysaccharides named WPMP-1 and WPMP-2 were obtained from Polygonum multiflorum with an average molecular weight of 2.04kDa and 92.13kDa, respectively. Structural characterization revealed that WPMP-1was supposed to be a glucan composed of 1,4-linked α-d-Glcp, 1,4,6-linked α-d-Glcp, and terminal α-d-Glcp. WPMP-2 contained a complex branched structre formed by 1,2-linked α-d-Rhap, 1,2,4-linked α-d-Rhap, 1,3,5-linked α-l-Ara, 1,5- linked α-l-Araf, terminal α-l-Araf, 1,4-linked β-d-Galp, 1,4-linked α-d-GalpA, and 4-linked α-d-GalpA. In vitro, WPMPs were of activation effect on splenocyte and macrophages, and could also protect immunocyte against 5-Fu induced immunosupression by restoring the proliferation rate, phagocytic index and cytokines secretion level. Moreover, the acid polysaccharide WPMP-2 exhibited better immunomodulatory activity than neutral polysaccharide WPMP-1, which indicated that the immunomodulatory activity of WPMPs were positively associated with higher content of uronic acid, percentage of rhamnose, arabinose and galactose, and branching degree. These findings could promote the polysaccharides from Polygonum multiflorum to be attractive functional food supplement and immunomodulatory adjuvant.

Abstract:In 1981 the Association of American Medical Colleges created a panel on the General Professional Education of the Physician (GPEP) in response to the concern that physicians of today are not responding to the total well-being and needs of patients and their families. The panel assessed the current approaches to the education of physicians and concluded that, with new technical advances and the changing face of medicine, our present system of general professional education will be outdated and inadequate without new approaches to learning and teaching. In addition, mounting pressures have been exerted on most faculty to produce research, to generate increasing patient revenues through practice plans, and to actively participate in residency training, with less emphasis on student education. Therefore, the medical student has the responsibility to independently learn the majority of medical information, since faculty and residents are available to teach for only a limited amount of time.

Abstract:Polyporaceae is an important fungal family that has been a source of natural products with a range of pharmaceutical activities in China. In our previous study, two polysaccharides, PCWPW and PCWPS, with significant antioxidant and antidepressant activity were obtained from Poria cocos. In this study, we evaluated their potential molecular mechanisms in the immunomodulation of macrophages. PCWPW and PCWPS were characterized by GC–MS analysis to contain 1,3-linked Glcp. ELISA assays results demonstrated that the secretion of TNF-α was significantly enhanced by PCWPW/PCWPS. RNA-seq data demonstrated that PCWPS treatment modulated the expression of immune-related genes in macrophages, which was further confirmed by RT-qPCR assays. The activation of TNF-α secretion was found to be mannose receptor (MR) dependent and suppressed by MR inhibitor pretreatment. Moreover, the amount of TNF-α cytokine secretion in PCWPW/PCWPS-induced RAW264.7 cells was decreased when pretreated with NF-κB or MAPK signaling pathway inhibitors. Collectively, our results suggested that PCWPW and PCWPS possessed immunomodulatory activity that regulates TNF-α expression through the NF-κB/MAPK signaling pathway by binding to mannose receptors. Therefore, PCWPW and PCWPS isolated from Poria cocos have potential as drug candidates for immune-related disease treatment.
