Abstract:Antimicrobial washing (AW), antimicrobial coating (AC), and a combination of washing followed by coating (AW+AC) were evaluated for their ability to inactivate artificially inoculated foodborne pathogens and native microbiota on strawberries stored at 4°C. Strawberries were inoculated with a six-strain composite of Escherichia coli O157:H7 and Salmonella; treated by AW, AC, or AW+AC; and stored at 4°C for 3 weeks. The washing solution contained 90 ppm of peracetic acid, and the coating solution consisted of chitosan (1%, w/v), allyl isothiocyanate (1%, v/v), and corn-bio fiber gum (5%, w/v). The effectiveness of the antimicrobial treatments against E. coli O157:H7 and Salmonella pathogens and native microflora on strawberries and their impact on fruit quality (appearance, weight loss, color, and firmness) were determined. By the end of storage, pathogen populations on strawberries were 2.5 (AW+AC), 2.9 (AC), 3.8 (AW), and 4.2 log CFU for the positive (untreated) control. AW+AC treatments also inactivated the greatest population of native microflora, followed by the AC treatment alone. AW+AC treatments showed additional antimicrobial effectiveness against these two pathogens and native microflora. Both AW+AC and AC treatments preserved the color, texture, and appearance of strawberries throughout storage. The coating treatments (AW+AC and AC alone) further reduced the loss of moisture throughout storage. The AW treatment was the least effective in reducing populations of pathogens and native microflora and in maintaining the quality of strawberries throughout storage. This study demonstrates a method to improve the microbiological safety, shelf life, and quality of strawberries.
Inactivation of Natural Microflora and Inoculated Listeria Innocua on Whole Raw Shrimp by Ozonated Water, Antimicrobial Coatings, and Cryogenic Freezing

Abstract:Shrimp have been associated with outbreaks of foodborne illnesses. A survey was conducted to determine the microbiological safety and quality of frozen whole raw shrimp, purchased at markets in the Northeast United States. Shrimp were composed of 32 brands from 9 different countries of origin. The average aerobic, psychrotrophic, coliform, and Enterobacteriaceae microbial counts were 3.64, 4.06, 0.02 and 1.43 log CFU/g, respectively. Of shrimp samples that were Listeria spp. positive (21.9%), 12.5% were identified as Listeria monocytogenes. No Salmonella spp., Escherichia coli 0157:H7, Vibrio spp., or Staphylococcus aureus were detected. The survival of natural microflora and inoculated Listeria innocua on shrimp were investigated after treatments with ozonated water, antimicrobial coating and cryogenic freezing, alone or in combination. Ozonated washes, cryogenic freezing and antimicrobial coating treatments, applied singly, reduced the natural bacteria or L. innocua by <2 log CFU/g; however, in Combination, treatments provided additive or synergistic reductions of the natural bacteria and L innocua. The chitosan coating with allyl isothiocyanate, in combination with cryogenic freezing, inactivated more than 5 log of L innocua and natural microorganisms. These information may be valuable for seafood processors and distributors to adopt intervention strategies to enhance the safety and extend the microbial shelf-life of shrimp. Use of aqueous ozone rinsing to improve the disinfection efficacy and shorten the processing time of ultrasound-assisted washing of fresh produce

Abstract:In minimal processing industry, chlorine is widely used in the disinfection process and ultrasound (US) increases the disinfection efficacy of chlorine and reduces the cross-contamination incidence during washing. Tap water (TW), which has no disinfection effect, is generally used to rinse off sanitizer residues on the surface of disinfected fresh-cut vegetables. In this study, aqueous ozone (AO), a low-cost and residue-free sanitizer, was used to replace TW rinsing in combination with US (28 kHz)-chlorine (free chlorine [FC] at 10 ppm, a concentration recommended for industrial use) for the disinfection of fresh-cut lettuce as a model. US-chlorine (40 s) + 2.0 ppm AO (60 s) treatment resulted in browning spots on lettuce surface at the end of storage. In contrast, US-chlorine (40 s) + 1.0 ppm AO (60 s) did not lead to deterioration of the sensory quality (sensory crispness, color, and flavor) and a change in total color difference, and the activities of browning-related enzymes were significantly lower. Moreover, US-chlorine (40 s) + 1.0 ppm of AO (60 s) treatment led to significantly lower counts of Escherichia coli O157:H7, Salmonella Typhimurium, aerobic mesophilic (AMC), and molds and yeasts (M&Y) on days 0-7 than US-chlorine (60 s) + TW (60 s) and single 1.0 ppm AO (120 s) treatments, suggesting that AO provided an additional disinfection effect over TW, while reducing the overall processing time by 20 s. Cell membrane permeability analysis (alkaline phosphatase, protein, nucleotide, and adenosine triphosphate leakage) showed that the combination with 1.0 ppm AO caused more severe cell membrane damage in E. coli O157:H7 and S. Typhimurium, explaining the higher disinfection efficacy. 16S rRNA sequencing revealed that following US-chlorine (40 s) + 1.0 ppm of AO (60 s) treatment, Massilia and Acinetobacter had higher relative abundances (RAs) on day 7 than after US-chlorine (60 s) + TW (60 s) treatment, whereas the RAs of Escherichia-Shigella was significantly lower, indicating that the former treatment has a superior capacity in maintaining a stable microbial composition. This explains from an ecological point of view why US-chlorine (40 s) + 1.0 ppm of AO (60 s) led to the lowest AMC and M&Y counts during storage. The study results provide evidence that AO has potential as an alternative to TW rinsing to increase the disinfection efficacy of US-chlorine.

Abstract:Fresh produce-associated outbreaks of foodborne illnesses continue to occur every year in the U.S., suggesting limitations of current practices and the need for effective intervention technologies. Advanced oxidation process involves production of hydrogen radicals, which are the strongest oxidant. The objective of the present study was to evaluate the effectiveness of advanced oxidation process by combining gaseous ozone and aerosolized hydrogen peroxide. Grape tomatoes were inoculated with a 2-strain cocktail of Salmonella typhimurium on both stem scar and smooth surface. Gaseous ozone (800 and 1600 ppm) and aerosolized hydrogen peroxide (2.5, 5 and 10%) were separately or simultaneously introduced into a treatment chamber where the inoculated tomatoes were placed. During the 30 min treatments, hydrogen peroxide was aerosolized using an atomizer operated in two modes: continuously or 15 s on/50 s off. After the treatments, surviving Salmonella on the smooth surface and stem scar were enumerated. Results showed that ozone alone reduced Salmonella populations by <0.6 log CFU/fruit on both the smooth surface and the stem scar area, and aerosolized hydrogen peroxide alone reduced the populations by up to 2.1 log CFU/fruit on the smooth surface and 0.8 log CFU/fruit on stem scar area. However, the combination treatments reduced the populations by up to 5.2 log CFU/fruit on smooth surface and 4.2 log CFU/fruit on the stem scar. Overall, our results demonstrate that gaseous ozone and aerosolized hydrogen peroxide have synergistic effects on the reduction of Salmonella populations on tomatoes.

Abstract:The consumption of fresh produce is steadily increasing and chlorine washing is the most commonly used method of disinfecting fresh produce. However, chlorine washing possesses a potential risk. Hence, this study used ozone microbubble (OMB) water to disinfect Salmonella Enteritidis, S. Typhimurium, Staphylococcus aureus, and Escherichia coli on tomatoes. After injecting ozone into the microbubble generator, OMB was fulfilled in a 10 L tank for 10 or 20 min. The inoculated tomatoes were washed for 30 or 60 s. Control groups included unwashed, water-washed, microbubble-only, and ozone-only. The microbial populations were significantly lower on the OMB-treated tomatoes than controls (p 0.05). When tomatoes were treated with OMB with 10 min fulfilling and 30-s washing, the differences of tested bacteria and water washing, ozone-only, and microbubble-only were: S. Enteritidis: 4.11, 3.37, 2.54 log CFU/tomato; S. Typhimurium: 4.83, 4.50, 2.78 log CFU/tomato; E. coli: 4.31, 4.08, 2.09 log CFU/tomato; S. aureus: 4.12, 3.93, 2.82 log CFU/tomato. In addition, significant higher ozone concentrations and conductivity were detected in OMB water than other groups (p 0.05). This study demonstrated that OMB effectively inactivated bacteria on tomatoes and did not affect the physical and sensory characteristics of tomatoes.

Abstract:The purpose of this study was to evaluate the antimicrobial efficacy of an aqueous ozone (Bio-Safe) treatment and lactic acid solutions on natural microbiota and E. coli O157:H7 and Salmonella surrogates on beef carcasses and trim in a commercial beef processing plant. For every repetition, 40 carcass and 40 trim swabs (500 cm2) were collected. Samples were taken using EZ-ReachTM swabs, and plated into aerobic plate count (APC), coliform, and E. coli PetrifilmTM for enumeration. In addition, a five-strain cocktail (MP-26) of E. coli surrogates was inoculated onto trim. For every trim surrogate repetition, 30 trim pieces were sampled after attachment and after ozone intervention. Samples were diluted and counts were determined using the TEMPO® system for E. coli enumeration. Ozone and lactic acid interventions significantly reduced (p < 0.003) bacterial counts in carcasses and trim samples. Moreover, lactic acid further reduced APC and coliforms in trim samples compared to ozone intervention (p < 0.009). In the surrogate trials, ozone significantly reduced (p < 0.001) surrogate concentration. Historical data from the plant revealed a reduction (p < 0.001) of presumptive E. coli O157:H7 in trim after a full year of ozone intervention implementation. The novel technology for ozone generation and application as an antimicrobial can become an alternative option that may also act synergistically with existing interventions, minimizing the risk of pathogens such as Salmonella and E. coli O157:H7.

Abstract:Mycotoxins are health-threatening fungal metabolites that have been found in several foods around the world. Although agricultural, transportation, and storage management strategies have been employed to reduce the production of mycotoxins, they are not effective in eliminating mycotoxins. In this context, the application of ozone has emerged for the degradation of mycotoxins. Ozone has a strong oxidation rate and generates more free radicals, which can counter the functional groups of the mycotoxin by changing their molecular structures and forming products having lower molecular weight reduced number of double bonds, and reduced toxicity. Research indicates that ozone could be able to destroy mycotoxins without leaving any residues in the commodities. The ozone processing parameters, surface and nutritional properties of food, and fungal species are the primary determinants affecting the processing efficacy. Ozone can contribute to sustainable food consumption through mycotoxin degradation to achieve sustainable development goals (SDGs).
Oil-Based Sanitization in Low-Moisture Environments: Delivery of Acetic Acid with Water-in-Oil Emulsions

Abstract:Contamination with Salmonella spp. and Listeria monocytogenes is concerning across low-moisture food (LMF)-processing environments due to the pronounced survival of these organisms under dry conditions. This study treated desiccated bacteria with acetic acid delivered by oil with and without water-in-oil (W/O) emulsion. The influences of cellular desiccation, emulsion water concentration, water activity (aw), and treatment temperature were investigated. Acetic acid dissolved in oil (i.e., acidified oil) showed low levels of antimicrobial efficacy. After treatment with acidified oil (200 mM acetic acid at 22°C for 30 min), Salmonella enterica serovar Enteritidis phage type 30 cells desiccated to 75% equilibrium relative humidity (ERH) and 33% ERH were reduced by 0.69 and 0.05 log CFU/coupon, respectively. The dispersion of a low level of water (≥0.3%, vol/vol) within the acidified oil with the surfactant (i.e., acidified W/O emulsion) significantly enhanced the antimicrobial efficacy. After treatment with the acidified W/O emulsion (200 mM acetic acid at 22°C for 20 min), desiccated Salmonella (4-strain cocktail) and L. monocytogenes (3-strain cocktail) cells were reduced by >6.52 log most probable number (MPN)/coupon, regardless of the desiccation levels. Increased efficacy was observed with temperature elevation. Reduced efficacy was observed when glycerol was added to the aqueous phase of the emulsion to decrease the solution aw, indicating that the enhanced efficacy of the acidified W/O emulsion was associated with differential osmotic pressure. The antimicrobial mechanism may be due to the membrane disruption induced by acetic acid, in combination with the hypoosmotic stress provided by W/O emulsion, creating cellular lysis, as illustrated by electron micrographs. IMPORTANCE Aqueous-based cleaning and sanitation are undesirable in processing facilities that manufacture low-moisture foods such as peanut butter and chocolate. Alcohol-based sanitization is advantageous because it leaves no residue on the contact surface but requires the processing facility to close temporarily due to flammability. At >6.52 log kill of desiccated Salmonella and Listeria monocytogenes cells, the developed oil-based formulation has the potential to be an effective dry sanitation method.
An Effective Sanitizer for Fresh Produce Production: In Situ Plasma-Activated Water Treatment Inactivates Pathogenic Bacteria and Maintains the Quality of Cucurbit Fruit

Abstract:The effect of plasma-activated water (PAW) generated with a dielectric barrier discharge diffusor (DBDD) system on microbial load and organoleptic quality of cucamelons was investigated and compared to the established sanitizer, sodium hypochlorite (NaOCl). Pathogenic serotypes of Escherichia coli, Salmonella enterica, and Listeria monocytogenes were inoculated onto the surface of cucamelons (6.5 log CFU g-1) and into the wash water (6 log CFU mL-1). PAW treatment involved 2 min in situ with water activated at 1,500 Hz and 120 V and air as the feed gas; NaOCl treatment was a wash with 100 ppm total chlorine; control treatment was a wash with tap water. PAW treatment produced a 3-log CFU g-1 reduction of pathogens on the cucamelon surface without negatively impacting quality or shelf life. NaOCl treatment reduced the pathogenic bacteria on the cucamelon surface by 3 to 4 log CFU g-1; however, this treatment also reduced fruit shelf life and quality. Both systems reduced 6-log CFU mL-1 pathogens in the wash water to below detectable limits. The critical role of superoxide anion radical (·O2-) in the antimicrobial power of DBDD-PAW was demonstrated through a Tiron scavenger assay, and chemistry modeling confirmed that ·O2- generation readily occurs in DBDD-PAW generated with the employed settings. Modeling of the physical forces produced during plasma treatment showed that bacteria likely experience strong local electric fields and polarization. We hypothesize that these physical effects synergize with reactive chemical species to produce the acute antimicrobial activity seen with the in situ PAW system. IMPORTANCE Plasma-activated water (PAW) is an emerging sanitizer in the fresh food industry, where food safety must be achieved without a thermal kill step. Here, we demonstrate PAW generated in situ to be a competitive sanitizer technology, providing a significant reduction of pathogenic and spoilage microorganisms while maintaining the quality and shelf life of the produce item. Our experimental results are supported by modeling of the plasma chemistry and applied physical forces, which show that the system can generate highly reactive ·O2- and strong electric fields that combine to produce potent antimicrobial power. In situ PAW has promise in industrial applications as it requires only low power (12 W), tap water, and air. Moreover, it does not produce toxic by-products or hazardous effluent waste, making it a sustainable solution for fresh food safety.
The effectiveness of ozone and acidulant treatments in extending the refrigerated shelf life of fresh-cut potatoes

Abstract:The objective of the study was to determine the effectiveness of acidulant dip treatments (with or without aqueous ozone) to reduce enzymatic browning and to extend the shelf life of fresh-cut potato slices during refrigerated storage (4 °C) for 28 d. Potato slices subjected to aqueous ozone (2 ppm) had significantly (P≤ 0.05) higher L-values and lower a-values, but ozone did not appear to have any effect on aerobic plate counts (APCs) or polyphenol oxidase (PPO) activity. NatureSeal (NS) and sodium acid sulfate (SAS) were the most effective acidulant treatments in reducing browning (significantly [P≤ 0.05] higher L-values, lower a-values, and browning index values) regardless of ozone treatment. NS and SAS also had lower PPO activity compared to other treatments on days 0 and 28, and significantly (P≤ 0.05) lower APCs (≤2.00 log CFU/g) over refrigerated storage. Therefore, the SAS treatment was comparable to NS, a commercially available product, and showed promise as an effective antibrowning dip to reduce browning and spoilage in fresh-cut potato products. Practical Application: A 1% SAS dip treatment which included 1% citric and 1% ascorbic acid was found to be an effective antibrowning dip for fresh-cut potatoes along with NatureSeal®'s PS-10, compared to other treatments. They were both effective in maintaining low microbial counts over refrigerated storage. Additionally, aqueous ozone washes (2 ppm) showed significant benefits to reduce browning; however, ozone did not affect microbial counts or PPO enzyme activity. Therefore, the SAS treatment could have potential use in the fruit and vegetable industry to reduce browning and spoilage in fresh-cut potato products.

Abstract:The use of the OSM3 oximeter for measurement of the fraction of carboxyhaemoglobin (FCOHb) in blood allows for estimation of total circulating haemoglobin mass (Hb(tot)) by using the carbon monoxide rebreathing method. To ensure high accuracy of Hb(tot) estimation, potential sources of analytical errors should be identified and adjusted for. Based on observed differences in results of measured FCOHb between simultaneously sampled, arterialized and venous blood samples we investigated the influence of haemoglobin oxygen saturation (sO2) on results of measured FCOHb. Blood from nine healthy non-smokers was tonometered with gas mixtures containing 94% N2 or air and 6% CO2. The resulting oxygenated and deoxygenated specimens were mixed in different proportions to obtain varying sO2 values in the same blood. sO2, fractions of dyshaemoglobins, pO2, pCO2 and pH were measured at each step. FCOHb was significantly (p<0.001) higher in oxygenated (median, range: 0.6%, 0.4-0.9%) compared to deoxygenated (-0.2%, -0.5-0.0%) blood. Regression analysis identified the sO2 as the most important factor explaining 86% of the variance in observed changes in FCOHb. The observed sO2 effect has important implications on calibration procedure of OSM3, accuracy of measured FCOHb, and FCOHb dependent calculations such as estimation of Hb(tot) and related quantities. If the highest accuracy of FCOHb measurement is needed, an sO2 effect on results of measured FCOHb has to be considered and adjusted for.

Abstract:SUMMARY: The endogenous respiratory rate of non-photosynthetic Euglena gracilis var. bacillaris is remarkably constant under varied conditions of growth and incubation. After 60–90 min. of slightly more rapid respiration, the endogenous oxygen uptake averages about 6.5 μl. O2/million cells/ hr., and this rate can be maintained for at least 26 hr. The respiratory quotient of the endogenous metabolism is 1.0. Fluoroacetate inhibits endogenous oxygen consumption about 70%. A variety of evidence suggests that the polysaccharide paramylum is the major endogenous substrate. Tracer experiments indicate that the endogenous metabolism continues during the oxidation of exogenous substrates, but that the assimilated substrate mixes with, and dilutes the endogenous reserves. The time-course of this dilution process indicates that the most recently assimilated reserves are the first to be oxidized.

Abstract:The need for sustainable food production and the demand for fresh and minimally processed foods have prompted remarkable research in novel food processing technologies that ensure safe and shelf-stable food for a large population. Long-established techniques such as heating, drying, and freezing have been associated with nutrient loss and high energy consumption. This trend has drawn attention to the practice of employing ozone in several food applications owing to its significant disinfectant and antimicrobial efficiency. The aqueous form of ozone has been found to show greater efficacy than its gaseous form, with faster decomposition rates leaving no harmful residues. The current study presents an overview of the latest scientific literature on the properties, chemistry, and generation of aqueous ozone, emphasizing the factors affecting process efficiency. The review scrupulously focuses on food decontamination, starch modification, pesticide degradation, and seed germination effects of aqueous ozone, highlighting the optimum processing parameters and salient findings of some major studies. A brief insight into the limitations and future trends has also been presented. Aqueous ozone has been acclaimed to have the potential to cause significant changes in the food matrix that could result in constructive modifications with outcomes entirely dependent on the processing conditions. Indirect and direct reactions involving hydroxyl radical and molecular oxygen atoms, respectively, form the basis of the ozone reaction in aqueous media, providing a distinctive kind of advanced oxidation process that offers certain crucial benefits. With a shorter half-life in water as compared to air, the rapid decomposition of aqueous ozone to oxygen, leaving no harmful residues, adds to its advantages.

While traditional chemical-based methods are effective and widely used, rising energy costs, the development of microbial tolerances, and growing awareness of the ecological impact of chemical biocides have renewed interest in novel biocides. Ozone, in both its gaseous and aqueous forms, is recognized as a potent disinfectant against bacteria, viruses, and fungi due to its high oxidation potential. Our review highlights several studies on the applications of ozone within the food industry, including its use for surface and aerosol disinfection and its capacity to reduce viable Listeria monocytogenes, a pertinent foodborne pathogen harbouring environmental and biocide stress tolerances and biofilm former. We also explore the use of ozone in food treatment and preservation, specifically on blueberries, apples, carrots, cabbage, and cherry tomatoes. While ozone is an effective disinfectant, it is important to consider material incompatibility, and the risks associated with prolonged human exposure to high concentrations. Nevertheless, for certain applications, ozone proves to be an efficacious and valuable alternative or complementary method for microbial control. Compliance with the biocide products regulation will require ozone device manufacturers to produce proven efficacy and safety data in line with British standards based on European standards (BS EN), and researchers to propose adaptations to account for ozone's unique properties.
A Novel Technique Using Advanced Oxidation Process (UV-C/H2O2) Combined with Micro-Nano Bubbles on Decontamination, Seed Viability, and Enhancing Phytonutrients of Roselle Microgreens

Abstract:Microbial contamination commonly occurs in microgreens due to contaminated seeds. This study investigated the decontamination effects of water wash (control), 5% hydrogen peroxide (H2O2), UV-C (36 watts), advanced oxidation process (AOP; H2O2 + UV-C), and improved AOP by combination with microbubbles (MBs; H2O2 + MBs and H2O2 + UV-C + MBs) on microbial loads, seeds’ viability, and physio-biochemical properties of microgreens from corresponding roselle seeds. Results showed that H2O2 and AOP, with and without MBs, significantly reduced total aerobic bacteria, coliforms, Escherichia coli (E. coli), and molds and yeast log count in seeds as compared to the control. Improved AOP treatment of H2O2 + UV-C + MBs significantly augmented antimicrobial activity against total bacteria and E. coli (not detected,) as compared to control and other treatments due to the formation of the highest hydroxy radicals (5.25 × 10−13 M). Additionally, H2O2 and combined treatments promoted seed germination, improved microbiological quality, total phenolic, flavonoids, and 2,2-diphenyl-1-picrylhydrazyl radical (DPPH•) activity of the grown microgreens. Ascorbic acid content was induced only in microgreens developed from H2O2-treated seeds. Single UV-C treatment was ineffective to inactivate the detected microorganism population in seeds. These findings demonstrated that improved AOP treatment (H2O2 + UV-C + MBs) could potentially be used as a new disinfection technology for seed treatment in microgreens production.

Abstract:An advanced Oxidation process (AOP) based on using hydrogen peroxide, ozone and ultraviolet light (UV-C) was developed for inactivating mould spores (Penicillium expansum and Aspergillus niger) and bacterial pathogens (Salmonella Typhimurium and Listeria monocytogenes) inoculated onto lemons. Response Surface Methodology (RSM) with the central rotatable composite design was applied to study the combined effects of the percent of H2O2 (1–6% v/v), UV-C intensity (0.6–3.6 mW/cm2) and treatment time (20–60 s) as independent variables. The dependent variables were the colour change of lemons induced by AOP and log count reductions (LCR) of A. niger spores. Hydrogen peroxide concentration was the key parameter for both antimicrobial efficacy and discoloration of lemons. The optimal conditions for the AOP treatment were 1.88% v/v H2O2, 3.26 mW/cm2, UV-C intensity and 65.2 mJ/cm2 UV-C dose. From an initial loading between 7.23 and 7.62 log CFU/lemon of test microbes, the optimized treatment resulted in a LCR of 1.54 log CFU/lemon for Aspergillus spores and >5 log CFU/lemon for Salmonella and Listeria without changing the sensory characteristics of lemons.<h6>Industrial relevance</h6>Over 30% of citrus fruit is lost to post-harvest spoilage and consequently, decontamination methods are required to reduce the carriage of moulds such as Aspergillus, in addition to inactivating potential human pathogens (e.g. Salmonella and L. monocytogenes). Currently, there is a reliance on post-harvest washing to remove contamination although has low efficacy and potentially can spread contamination between batches. Advanced Oxidation Process (AOP) is an alternative decontamination method that generates antimicrobial free radicals from the UV-C degradation of hydrogen peroxide and ozone. Although antimicrobial, the free radicals also discolor lemons and hence a balance needs to be made between decontamination efficacy and fruit quality. In this study, RSM was applied to identify the optimum treatment that was subsequently verified. Although the current study was limited to lemons, the same approach could be applied to other fresh produce when designing AOP-based treatments.

Disinfection efficiency of chlorine dioxide and peracetic acid against MNV-1 and HAV in simulated soil-rich wash water

Abstract:Wash water from fresh vegetables and root vegetables is an important vehicle for foodborne virus transmission. However, there is lack of assessing rapid viral inactivation strategies in wash water characterized by a high soil content at the post-harvest stage. Considering the significance of food safety during the washing stage for fresh and root vegetable produce prior to marketing, we assessed the inactivation efficacy by using chlorine dioxide (ClO2) and peracetic acid (PAA) against a surrogate of human norovirus (murine norovirus 1, MNV-1) and hepatitis A virus (HAV), in wash water containing black soil and clay loam. The results indicated that MNV-1 and HAV were reduced to the process limit of detection (PLOD), with reductions ranging from 4.89 to 6.35 log10 PFU, and 4.63 to 4.96 log10 PFU when treated with ClO2 at 2.5 ppm for 10 mins. Comparatively, when treated with 500 ppm of PAA for 10 mins, MNV-1 and HAV were maximum reduced to 1.75 ± 0.23 log10 PFU (4.50 log10 PFU reduction) and 2.13 ± 0.12 log10 PFU (2.72 log10 PFU reduction). This demonstrated the efficacy of ClO2 in eliminating foodborne viruses in soil-rich wash water. When we validated the recovery of the virus from two types of wash water, the pH (9.24 ± 0.33 and 5.95 ± 0.05) had no impact on the recovery of MNV-1, while the recovery of HAV was less than 1 %. By adjusting the pH to a neutral level, recovery of HAV and its RNA levels was increased to 15.94 and 3.89 %. Thus, this study emphasized the critical role of pH in the recovery of HAV from the complex soil-rich aqueous environment, and the efficacy of ClO2 serving as a pivotal reference for the development of control strategies against foodborne viruses in the supply chain of fresh and root vegetables.

Abstract:Phase unwrapping is a challenging task for interferometry based techniques in the presence of noise. The majority of existing phase unwrapping techniques are path-following methods, which explicitly or implicitly define an intelligent path and integrate phase difference along the path to mitigate the effect of erroneous pixels. In this paper, a path-independent unwrapping method is proposed where the unwrapped phase gradient is determined from the wrapped phase and subsequently denoised by a TV minimization based method. Unlike the wrapped phase map where 2π phase jumps are present, the gradient of the unwrapped phase map is smooth and slowly-varying at noise-free areas. On the other hand, the noise is greatly amplified by the differentiation process, which makes it easier to separate from the smooth phase gradient. Thus an approximate unwrapped phase can be obtained by integrating the denoised phase gradient. The final unwrapped phase map is subsequently determined by adding the first few modes of the unwrapped phase. The proposed method is most suitable for unwrapping phase maps without abrupt phase changes. Its capability has been demonstrated both numerically and by experimental data from shearography and electronic speckle pattern interferometry (ESPI).
