Abstract:Asthma and chronic obstructive pulmonary disease (COPD) are the most common diagnoses for adults and children with respiratory tract inflammation. Recently, a novel fixed dose combination consisting of Ipratropium and Fenoterol has been released for the management and control of the symptoms of such disorders. The current research has newly developed and optimized three smart, accurate, simple, cost-effective, and eco-friendly spectrophotometric methods that enabled the simultaneous determination of the drugs under study in their combined inhaler dosage form, without the need for any previous separation steps, using water as a green solvent. The strategy employed was based on calculating one or two factors as a numerical spectrum or constant, which provided the complete removal of any component in the mixture that might overlap and the mathematical filtration of the targeted analyte. The methods developed could be classified into two types of spectrophotometric windows. Window I; involved absorption spectrum in their original zero-order forms (°D), which included recently designed methods named induced concentration subtraction (ICS) and induced dual wavelength (IDW). While window III focused on the ratio spectrum as the induced amplitude modulation (IAM) method. The extremely low absorptivity and lack of distinct absorption maximum in the zero-order absorption spectrum of Ipratropium were two intrinsic challenges that were better overcome by the proposed spectrophotometric methods than by the conventionally used ones. According to ICH guidelines, the proposed methods were validated using unified regression over range 2.0–40.0 μg/mL in the ICS method, while the linearity ranges for the IDW and IAM methods were 5.0–40.0 μg/mL of Ipratropium and 2.0–40.0 μg/mL of Fenoterol. Moreover, the three proposed methods were effectively used to assay the co-formulated marketed inhaler and further expanded to confirm the delivered dose uniformity in compliance with the USP guidelines. Finally, the established methods were evaluated for their greenness and blueness, in comparison to the official and reported analysis methods, using advanced cutting edge software metrics. Furthermore, the suggested techniques adhered well to the white analytical chemistry postulates that were recently published.

Stability-Indicating Analytical Method Development Using Quality by Design (QbD) Approach for Simultaneous Estimation of Budesonide and Levosalbutamol

Abstract:Background: Budesonide is a corticosteroid; levosalbutamol is bronchodilator. In combination, they are used in the treatment of asthma and chronic obstructive pulmonary disease. Objective: The present article portrays the development of a stability-indicating high-performance thin-layer chromatography (HPTLC) method for the simultaneous estimation of budesonide and levosalbutamol using design of experiment (DOE) principles. Method: DOE was applied in development part in which a Box-Behnken design was used to study the effect of factors on the Rf value of the drugs. Twenty-five experimental runs were performed to optimize the chromatographic conditions. Aluminum sheets precoated with silica gel 60 F254 were used as the stationary phase. The optimized mobile phase composition was found to be toluene-ethyl acetate-methanol-ammonia (4:1.8:1.8:0.2) and saturation time 20 min, quantified by densitometric analysis at 231 nm. Moreover, the drugs were subjected to acid and alkali hydrolysis, oxidation, thermal, and photodegradation. Results: The drugs undergo degradation under mainly acidic and basic conditions. Also, the degraded products were well resolved from the pure drug with significantly different Rf values. Linearity was performed in the ranges of 224-1120 ng/band for budesonide and 280-1400 ng/band for levosalbutamol. The method was validated according to the International Conference on Harmonization guidelines. Conclusions: This approach is useful to expedite method development and optimization activities in analytical laboratories. Highlights: The experimental data revealed that the volume of toluene and methanol in the mobile phase significantly affected the Rf value of both the drugs, and ammonia significantly affected the Rf value of levosalbutamol.

Abstract:Ipratropium bromide (IPR) and fenoterol hydrobromide (FEN) have recently been combined in a promising inhaler to treat two prevalent inflammatory illnesses of the airways: bronchial asthma and chronic obstructive pulmonary disease (COPD). The necessity for a single, sensitive, and trustworthy analytical approach to cover the diverse and necessary tests of in-vitro and in-vivo studies is greatly grown with the rising production of new fixed combinations. Two novel, selective and environmentally friendly LC techniques were developed in order to guarantee precise measurement of IPR and FEN in their challenging formulation. The initial technique involved high-performance thin-layer chromatography (HPTLC) in conjunction with densitometric quantification. Chromatographic separation was attained on HPTLC plates utilizing ethyl acetate - ethanol - acetic acid (5.0:5.0:0.1, by volume) as a developing system. Densitometric quantification of the separated bands was carried out at 220.0 nm over concentration ranges of 0.50-15.0 µg/band for IPR and 0.50-12.0 µg/band for FEN. High-performance liquid chromatography (HPLC) paired with diode array detection (DAD) was the core of the second technique. The optimized separation was achieved on a Zorbax SB C18 (150 × 4.6 mm, 5 μm) column with a combination of 10.0 mM potassium dihydrogen orthophosphate, pH 5.0 ± 0.1, adjusted with o-phosphoric acid and methanol (70:30, v/v) as the mobile phase and pumped at flow rate of 1.0 mL/min. The peaks were monitored at 220.0 nm using diode array detection, achieving linearity range of 5.0-200.0 µg/mL for both drugs. The ICH criteria have been verified and both methods have been confirmed to be valid, and successfully applied for assay the cited drugs in the Atrovent® comp HFA metered dose inhaler as well as delivered dose uniformity testing of the final product. Finally, whiteness appraisal and several state-of-the-art green evaluation metrics were applied to evaluate the sustainability of the proposed methods. The suggested approaches produced promising results and are the first simple and sustainable methodologies for the simultaneous quantification of both drugs in different real samples, all of which strongly suggest their application in quality control laboratories.

Abstract:Ipratropium bromide and Budesonide is combined Pharmaceutical Nasal dosage form. The label claim of this combined dosage form is 21 mcg of Ipratropium Bromide and64 mcg of Budesonide per Spray. It is for treatment of allergic rhinitis. Spray content uniformity is in-vitro investigational test which provides idea of uniformity of product throughout product life cycle. The present study aimed to validate High Performance Liquid Chromatography method for determination of spray content uniformity of these analytes. This validation study performed as per guidelines of International Conference on Harmonization and covers Precision, Linearity, Accuracy, Robustness, Ruggedness, stability of analytical solution and Specificity. The reversed phase chromatographic method uses a mobile phase prepared by mixing Buffer containing Sodium dihydrogen orthophosphate with Acetonitrile, Zorb ax CN150 mm column, 1.5 ml per min flow rate, 220 nm wavelength, 25°C column oven and 50 μL injection volume. The method found linear over the range of 50% to 150% of target concentration with correlation coefficient of 0.9997 for Ipratropium bromide and 0.9999 for Budesonide. The recoveries obtained were 103.1% - 103.8% for Ipratropium bromide and 97.0% - 100.1% for Budesonide. %Related Standard Deviation found well within the acceptance criteria for precision. No interference found at the retention time of analyte peaks in blank, placebo and degradation conditions describe specificity of method. The method showed a successful application for determination of Ipratropium bromide and Budesonide in Nasal spray pharmaceutical formulation.

Abstract:<span>Budesonide is one of the intranasal corticosteroids, referred as first-line therapy for allergic rhinitis. Its determination is a challenging task due to its extremely low plasma levels, which limits the progress in the investigation of pharmacokinetics and quality control of preparations. In this study, a sensitive and high-throughput method to determine budesonide in human plasma using budesonide-d8 as the internal standard was developed and validated. A small volume of plasma sample (0.2 mL) was diluted with 0.2 mL water, followed by a solid-phase extraction using Cleanert PEP-2 products. Extracted samples were analyzed by liquid chromatography coupled to electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS). Chromatographic separation of analytes was performed on an InertSustain AQ-C18 HP column (3 µm, 2.1 × 50 mm) under the reversed-phase condition with gradient elution. With the assay, linear calibration curves were obtained over the concentration range of 10–1200 pg/mL for budesonide, with considerable extraction recoveries (84.7–89.4%), and negligible matrix effects (<4.1). Moreover, the newly developed method was successfully applied to the evaluation of pharmacokinetics of two budesonide intranasal formulations with and without charcoal block in healthy volunteers.</span>

Abstract:A reverse phase high performance liquid chromatography method was developed and validated for simultaneous quantitative estimation of ambroxol hydrochloride, guaiphenesin and terbutaline sulphate. The chromatographic separation was achieved using a Phenomenex Luna C18 (2), 100A, 5μ (4.6 X 250mm) column using 0.012(M) sodium salt solution of 1-hexane sulphonic acid:acetonitrile 73: 27 (V/V) adjusted to pH 3 using orthophosphoric acid having isocratic elution of flow rate 1.0mL/min, detection at 220nm and injection volume 20 μL. The retention times of ambroxol hydrochloride, guaiphenesin and terbutaline sulphate were 25, 6.7 and 4.7 min, respectively. The proposed method was found to have linearity in concentration range of 63.40-95.11 μg/mL for ambroxol, 68.00-302.52 μg/mL for guaiphenesin and 4.88-7.20 μg/mL for terbutaline sulphate. Regression coefficient (R2) were 0.9997, 0.9989 and 0.9984 and percentage recovery were 101.67%, 101.68%, and 99.45% for ambroxol, guaiphenesin and terbutaline sulphate, respectively. The method validation showed excellent results for precision, accuracy, linearity and specificity. The proposed method was successfully used for the quantitative analysis of an oral liquid formulation, being developed in our laboratory, containing ambroxol hydrochloride, guaiphenesin and terbutaline sulphate.
Development of a green synchronous spectrofluorimetric technique for simultaneous determination of Montelukast sodium and Bilastine in pharmaceutical formulations

Abstract:For the treatment of rhinitis and asthma, a combination of Montelukast sodium and Bilastine has just been approved. Based on the first derivative of synchronous fluorescence, the current work developed a green, highly accurate, sensitive, and selective spectroscopic approach for estimating Montelukast sodium and Bilastine in pharmaceutical dosage form without previous separation. The selected technique focuses on measuring the synchronized fluorescence of the studied medications at a fixed wavelength range (Δλ) = 110 nm, and using the amplitude of the first derivative's peak at 381 and 324 nm, for quantitative estimation of Montelukast sodium and Bilastine, respectively. The impacts of different factors on the referred drugs' synchronized fluorescence intensity were investigated and adjusted. The calibration plots for were found to be linear over concentration ranges of 50-2000 ng mL-1 for Montelukast sodium and 50-1000 ng mL-1 for Bilastine. Montelukast sodium and Bilastine have LODs of 16.5 and 10.9 ng mL-1, respectively. In addition, LOQs were: 49.9 and 33.0 ng mL-1, for both drugs, respectively. The developed method was successfully employed to quantify the two drugs in synthetic tablets mixture and in laboratory prepared mixtures containing varied Montelukast and Bilastine ratios. To compare the results with the published analytical approach, a variance ratio F-test and a student t-test were used, which revealed no significant differences.

Abstract:The development and validation of analytical methods are crucial in guaranteeing the precision, dependability, and excellence of pharmaceutical analysis. This research investigates the field of pharmaceutical chemistry by doing a comparative examination of analytical techniques for the simultaneous determination of Bilastine and Montelukast Sodium in both bulk and tablet forms. The selected method for this analysis is planar chromatography. The simplicity, specificity, precision, and accuracy of a high-performance liquid chromatography (HPTLC) approach were investigated for their use in the simultaneous estimation of the antihistaminic combination medication Bilastine and Montelukast Sodium in bulk and its pharmaceutical dose for the treatment of allergic rhinitis. Densitometric readings were taken at 254 nm after separating substances using ethyl acetate, toluene, methanol, and ammonia (7:0.5:1.5:0.5v/v/v/v) as mobile phase and precoated aluminium silica gel plates (60F254) as stationary phase. Bilastine and montelukast Sodium have Rf values 0.2 (Bilastine) & 0.4 (Montelukast Sodium), which is considered an acceptable resolution. The International Conference on Harmonization's (ICH) requirements validated the processes for accuracy, linearity, precision, robustness, and system adaptability. Bilastine and Montelukast Sodium concentrations were determined without any disruption from the excipients. Both Bilastine and Montelukast Sodium were effectively quantified using the suggested method, which bodes well for its utility in enhancing quality assurance.
Rapid simultaneous determination of indacaterol maleate and glycopyrronium bromide in inhaler capsules using a validated stability-indicating monolithic LC method

Abstract:Background: Chronic obstructive pulmonary disease (COPD) is a major cause of morbidity and mortality worldwide. A combination of indacaterol maleate with glycopyrronium bromide has recently been approved as a once-daily maintenance therapy in patients with COPD. The very low dose (μg level/capsule) renders the analysis of such products challenges. This study reports for the first time about HPLC method for the quality control of such combination and it is a stability indicating at the same time. Results: A rapid, simple, precise and reproducible HPLC method was developed and validated for simultaneous determination of indacaterol maleate and glycopyrronium bromide using tenoxicam as an internal standard. The chromatographic separation was achieved on an onyx monolithic C18 column (100 × 4.6 mm) using a mobile phase consisting of acetonitrile and 30 mM phosphate buffer (pH 3.5) (30:70, v/v), run at a flow rate of 2 mL/min with UV detection at 210 nm. The total analysis time was less than 3 min. The HPLC method was validated for linearity, limits of detection and quantitation, precision, accuracy, system suitability and robustness. Calibration curves were obtained in the concentration ranges of 1-44 µg/mL for indacaterol maleate and 0.5-20 µg/mL for glycopyrronium bromide. Stability tests were done through exposure of the analyte solution for different stress conditions and the results indicate no interference of degradants with HPLC method. Conclusions: The method was successfully applied for the quantitative analysis of indacaterol maleate and glycopyrronium bromide both individually and in a combined pharmaceutical inhaler capsules to support the quality control and to assure the therapeutic efficacy of the two drugs. The simple procedure involved in sample preparation and the short run-time added the important property of high throughput to the method. Abstract:Two fluorescence‐based techniques to determine Ambroxol (AMX) and Guaifenesin (GFN) in their isolated forms or combined in pharmaceutical formulations without interference from each other. Ambroxol hydrochloride (AMX) and guaifenesin (GFN) are approved drugs utilized to treat coughs through their potent mucolytic and expectorant properties. Due to their massive, combined administration in many illnesses, there is a persistent need for their concurrent estimation in different pharmaceutical formulations. Two sensitive, environmentally friendly spectrofluorimetric methods were developed. AMX was determined using the first method (I) without interference from GFN. This method depends on the quenching of Erythrosine B (EB) native fluorescence at 552 nm after excitation at 527 nm due to the formation of a non‐fluorescent AMX‐EB ion‐pair complex in Britton–Robinson buffer (BRB) solution pH (3.5). The concentration plot is linear over the 0.25–5.0 μg/mL range, with a mean percent found value of 99.74%. Method (II) depends on measuring the native fluorescence of aqueous GFN solution at two analytical wavelengths, either 300 or 600 nm, after excitation at 274 nm. Relative fluorescence intensity (RFI)–concentration plots are linear over the ranges of 0.02–0.5 and 0.1–2.0 μg/ml, with mean percent found at 99.96% and 99.91% at dual wavelengths, respectively. The proposed methods were successfully applied to assay both drugs in raw materials and different single and combined pharmaceutical formulations. These methods have been thoroughly validated following International Committee on Harmonisation (ICH) guidelines. National Environmental Methods Index, Analytical Eco‐Scale, and Green Analytical Procedure Index were used to prove greenness, thereby enhancing their applicability. The proposed techniques provide straightforward, precise, and cost‐effective solutions for routine formulation analysis in quality control laboratories.

Abstract:Pharmacological treatments of asthma and chronic obstructive pulmonary disease include medications such as inhaled corticosteroids, long- or short-acting β2-adrenoreceptor agonists and anticholinergics. There is an unmet need for the monitoring of adherence and drug exposure to those therapies since poor adherence and/or inhalation technique may impact the control of the disease and the pharmacological strategy. Since plasma therapeutic drug monitoring only reflects the body exposure in the last few hours, the measurement of hair drug concentrations may be of great interest to assess the chronic exposure. A liquid chromatography - tandem mass spectrometry method was therefore developed for the quantification of corticosteroids, β2-adrenoreceptor agonists and anticholinergics in human hair. The method was validated according to the European Medicines Agency and Food and Drug Administration guidelines. Sensitivity, accuracy and precision were excellent, allowing the quantification of drugs in the pg/mg range. The method was shown suitable for the analysis of clinical hair samples, demonstrating that it could be used for hair therapeutic drug monitoring in asthma or chronic obstructive pulmonary disease patients.
Facile approaches for determination of Bromhexine Hydrochloride and its active metabolite Ambroxol Hydrochloride using Eosin Y

Abstract:New validated Spectroscopic methods were developed to assay Bromhexine Hydrochloride and its active metabolite Ambroxol Hydrochloride separately in pure form and pharmaceutical formulations. The spectrophotometric assay (method I) shows complex formation between each of the drugs and Eosin Y at 540nm at pH 3.6 and 3.4mL of 4×10-4M Eosin for Bromhexine and Ambroxol. The Spectrofluorimetric assay (method II) depends on quenching eosin native fluorescence by the studied drugs, which measured at 540nm after excitation at 302nm. The spectrophotometric absorbance-concentration plot is rectilinear over the ranges (1.0-5.0) and (1.0-10.0) μg/mL for bromhexine and ambroxol with LOD of 0.31 and 0.14μg/mL and LOQ of 0.94 and 0.42μg/mL for the two drugs respectively. The fluorometric-concentration plot is linear along the range (1.0-5.0) μg/mL and (1-10) μg/mL for the two drugs respectively with LOD of 0.13μg/mL and 0.22μg/mL and LOQ of 0.4μg/mL and 0.65μg/mL for the two drugs, respectively. Developed assays have been validated in agreement with ICH recommendations and they were used in the analysis of commercial drug formulations containing the two mucolytic drugs and the results were matching with those obtained by the comparison method.
Validated derivative and ratio derivative spectrophotometric methods for the simultaneous determination of levocetirizine dihydrochloride and ambroxol hydrochloride in pharmaceutical dosage form

Abstract:Three simple, precise, accurate and validated derivative spectrophotometric methods have been developed for the simultaneous determination of levocetirizine dihydrochloride (LCD) and ambroxol hydrochloride (ABH) in bulk powder and in pharmaceutical formulations. The first method is a first derivative spectrophotometric method ((1)D) using a zero-crossing technique of measurement at 210.4 nm for LCD and at 220.0 nm for ABH. The second method employs a second derivative spectrophotometry ((2)D) where the measurements were carried out at 242.0 and 224.4 nm for LCD and ABH, respectively. In the third method, the first derivative of the ratio spectra was calculated and the first derivative of the ratio amplitudes at 222.8 and 247.2 nm was selected for the determination of LCD and ABH, respectively. Calibration graphs were established in the ranges of 1.0-20.0 μg mL(-1) for LCD and 4.0-20.0 μg mL(-1) for ABH using derivative and ratio first derivative spectrophotometric methods with good correlation coefficients. The developed methods have been successfully applied to the simultaneous determination of both drugs in commercial tablet dosage form.
Airsupra: An Inhaled Albuterol/Budesonide Combination for Asthma

Abstract:The FDA has approved Airsupra (AstraZeneca), a metered-dose inhaler containing the short-acting beta2-agonist (SABA) albuterol and the inhaled corticosteroid (ICS) budesonide, for use as needed for treatment or prevention of bronchoconstriction and to reduce the risk of exacerbations in patients ≥18 years old with asthma. Airsupra is the first combination of a SABA and an ICS to become available in the US. It is not approved for use as maintenance therapy.

The study aimed to establish a high-performance liquid chromatography (HPLC) method for the simultaneous determination of these four drugs and to investigate their stability in 0.9% sodium chloride injection or 5% glucose injection over 48 hours. Methods: An InertSustain C18 column (150 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of acetonitrile and 50 mmol·L−1 potassium dihydrogen phosphate solution (pH 4.0) with gradient elution. The flow rate was 0.8 mL·min−1, and the column temperature was maintained at 30°C. The stability of APL, DXL, BXH, and AXH in 0.9% sodium chloride and 5% glucose injections over 48 h was determined using HPLC. Results: APL, DXL, BXH, and AXH showed good linearity within the ranges of 0.01 to 0.20, 0.003–0.06, 0.015–0.30, and 0.016–0.16 mg·mL−1, respectively (r > 0.999). The intraday and interday relative standard deviations were <2%, with recovery rates between 98.4% and 102.2%. The four drugs remained colorless and clear in infusion mixtures. The pH value fluctuated within ±0.3 over 48 hours, and the relative percentage content of the drugs ranged from 95.0% to 105.0%. Conclusion: The established HPLC method is simple, reliable, and stable, allowing for the simultaneous determination of the four antitussive and expectorant drugs. APL, DXL, BXH, and AXH were stable within 48 hours when mixed with 0.9% sodium chloride and 5% glucose injections.

Abstract:Bromhexine (BR), guaiafenesin (GUF) and salbutamol (SAL) are formulated as Ventocough syrup® (with and without sugar), labeled to contain propyl paraben and sodium benzoate as inactive ingredients. They are used to make coughing more productive and easier. A crucial element and a major issue in the pharmaceutical industry is the control of organic related impurities to obtain safe and effective treatment. Guaiacol (GUL) is reported to be GUF related impurity that was proved to be extremely toxic (toxic rating class 5), and its use should be banned. In this work, In-Silico study and ADMET estimation were conducted to predict GUL pharmacokinetic properties and its toxicity profile. Additionally, two chromatographic methods were conducted to analyze the studied components along with GUF impurity in the presence of the labeled dosage form excipients. The In-Silico study assured that GUL has oral rat acute toxicity and it is considered to be skin sensitizer. On the other hand, the developed TLC- densitometeric method depended on using a mobile phase mixture of hexane: methylene chloride: triethylamine (5.0:6.0:0.3, by volume) as a developing system. UV-Scanning was performed immediately at 275 nm for SAL, GUF and GUL, while scanning at 310 nm was used for scanning BR. Linearity was established in the ranges of 0.25–4.0, 0.25–4.0, 0.5–8.0 and 0.1–1.6 μg/band for BR, SAL, GUF and GUL, respectively. In the developed HPLC method, separation was performed on X-Bridge® C 18 column (250 × 4.6 mm, 5 μm) using a solvent mixture of 0.05M disodium hydrogen phosphate pH 3 with aqueous phosphoric acid: methanol (containing 0.3%, v/v triethylamine) (40:60, v/v). Detection was done at 225 nm and separation was achieved within 10 min. Linearity was proved in the range of 2–50 μg/mL for the proposed drugs. Validation of the developed methods was done and all the calculated parameters were within the acceptable limits recommended by ICH guidelines. After that, methods were used to examine the potency of the selected marketed dosage forms and concentrations of all drugs were within the acceptable limits. Additionally, complete separation between the studied drugs and the additives were observed. The developed methods can be used during routine quality control analysis of the proposed drugs when the required issues concern on sensitivity, selectivity and analysis time.

Abstract:Recently, a new formulation containing Mirabegron (MB) and Solifenacin succinate (SFS) has been approved for the management of over active bladder. However, only one analytical method has been reported for the simultaneous determination of both the analytes. Therefore, the current study was design to develop simple UV derivative spectroscopic and rapid RP-HPLC methods for simultaneous determination of MB and SFS. The chromatographic separation of MB and SFS was performed using Phenomenex Kinetex C18 (150mm × 4.5 mm × 5 μm) analytical column. A mixture of Water: Acetonitrile (20:80%v/v) was consider as mobile phase, at a flow rate of 1ml/min and at detector wavelength 225nm. A linear response was observe over the concentration range 2.5-12.5 μg/ml and 0.5-2.5 μg/ml respectively. The first order derivative method was develop by derivatisation of the zero absorption spectra for the first absorption spectra. The Zero crossing point of MB and SFS at 221 nm and 266 nm was obtain respectively. Beer’s law is obey in the concentration range of 7.5-20 μg/ml and 1.5-4 μg/ml for MB and SFS with correlation coefficient (R2) of 0.9984 and 0.9993 respectively. Both the methods were validated in accordance to guidelines for linearity, precision, repeatability, limit of detection (LOD), Limit of Quantification (LOQ), accuracy and robustness. Further, both the methods were validated and compared statistically using Student’s-t-test and employed for the concurrent estimation of MB and SFS in formulations. The proposed methods were simple, accurate, precise, and rapid. Therefore, they can be use for regular quality control of MB and SFS formulations and dissolution studies as well.
Stability-indicating high-performance thin-layer chromatographic method for the estimation of ambroxol hydrochloride and doxofylline in a pharmaceutical formulation using experimental design in robustness study

Abstract:A sensitive, specific, and accurate stability-indicating high-performance thin-layer chromatographic method for the analysis of ambroxol hydrochloride and doxofylline, for both bulk drug and formulation, was developed and validated according to the International Conference on Harmonization (ICH) guidelines. The densitometric analysis of both drugs was carried out in the absorbance mode at 276 nm using diethyl ether—n-butanol—ammonia (9:0.9:0.1, v/v) as the solvent system. This system was found to give compact spots for ambroxol hydrochloride at RF of 0.74 ± 0.01 and doxofylline at 0.41 ± 0.01. Moreover, both drugs were subjected to acid and alkali hydrolysis, oxidation, and photodegradation. Also, the degraded products were well resolved from the pure drug with different RF values. Linearity was found in the range of 20–100 and 100–500 ng band−1 for ambroxol hydrochloride and doxofylline. The limit of detection (LOD) and limit of quantitation (LOQ) for ambroxol hydrochloride and doxofylline were 1.17 and 3.57 ng band−1 and 30.68 and 92.97 ng band−1, respectively. “Bartlett’s test” and “Lack of fit” applied on peak area for linearity additionally proved validity of the linearity of the developed method. Good accuracy and precision were obtained as revealed from percent relative standard deviation (% RSD) less than 2. Moreover, robustness testing was performed applying fractional factorial design, 24-1. All the four factors: volume of diethyl ether, volume of n-butanol, solvent front, and chamber saturation time, evaluated in the robustness testing, were found to have an insignificant effect on the retention factor. Similarly, no interference was observed from common excipients in tablet formulation as well as degradation product, indicating specificity of the method. As the method could effectively separate both drugs from their degradation products, it can be used as a stability-indicating method.

Abstract:Inhalation therapy with a nebulizer is widely used in chronic respiratory disease. Mixing inhalation solutions/suspensions for simultaneous inhalation is more convenient and might simplify the administration procedure. However, there are no data available to address the in vitro aerosol characteristics and physico-chemical compatibility of Combivent® (containing Salbutamol and Ipratropium bromide) with other inhalation solutions/suspensions. In order to investigate the in vitro aerosol characteristics and physico-chemical compatibility of Combivent® with Budesonide, Beclomethasone, and N-acetylcysteine, the appearance, pH, osmotic pressure, chemical stability, mass median aerodynamic diameter (MMAD), fine particles fraction (FPF), particle size corresponding to X50 (particle size, which accounts for 50% of the total cumulative percentage of volume of all particles), delivery rate, and total delivery of the mixed inhalation solution/suspension were tested. There was no change in the appearance such as a change in color or precipitation formation at room temperature. The pH, osmolality, and chemicals of the mixtures were stable for 24 h after mixing. There were no significant differences between Combivent®, Budesonide, Beclomethasone, N-acetylcysteine, and the mixtures in MMAD, FPF, X50, the delivery rate, and the total delivery. This indicates that the mixtures were physically and chemically compatible. The mixing did not influence the particle size, distribution, or delivery compatibility of the mixtures.
