Abstract:A novel quantum dots (QDs) ECL biosensor for the detection of lysozyme was developed. Lysozyme was first incubated with probes immobilized at Au electrode in order to form the aptamer-lysozyme bioaffinity complexes. And the free probes were hybridized with the 5′-biotin modified cDNA oligonucleotides to form double-stranded DNA (ds-DNA) oligonucleotides. Avidin-QDs were bound to these hybridized cDNA through the biotin-avidin-system. The ECL signal of the biosensor was responsive to the amount of QDs bonded to the cDNA oligonucleotides, which was indirectly inverse proportional to the combined target protein.
Electrochemical aptamer biosensor for DNA detection based on label-free aptamers

Abstract:Electrochemical aptasensor has been broadly advanced for nucleic acid detection. However, it is a long-term goal to design an aptasensor with high specificity, flexibility, and simplicity. In this work, we develop a strategy of triblock DNA probe, which consists of two DNA probes at both ends and ployA fragments in the middle as probe-polyA-probe. PolyA fragment has high affinity to the surface of gold electrode, so it can be assembled on the electrode surface via polyA instead of traditional Au-S bonds. When the target DNA is simultaneously hybridized with the two capture probes, the hybridization stability can be improved due to the strong base stacking effect. [Ru(NH3)6]3+, as signal probe, can be electrostatically adsorbed on the negatively charged DNA skeleton. A wide linear range (10 pM-10 μM) is obtained with a detection limit of 2.9 pM. Our electrochemical aptasensor has good repeatability, stability, and specificity. More importantly, the electrochemical sensor can successfully detect DNA in human serum samples, which proves its practical value and extensive applicability in complex environment.

The Homogeneous Fluorescence Anisotropic Sensing of Salivary Lysozyme Using the 6-Carboxyfluorescein-labeled DNA Aptamer

Abstract:A simple and sensitive fluorescence anisotropy method was developed for lysozyme, employing the coupling of fluorophore, 6-carboxyfluorescein (FAM), with lysozyme upon recognition between the target molecule and its DNA aptamer. It was found in this study that the rotational dynamic of the detecting system is crucial to obtain a high anisotropy signal that cannot always be achieved by simply increasing the molecular volume, because molecular volume increase may not be able to efficiently retard the rotational movement of the fluorophore. FAM was selected as the label of the ssDNA aptamer to effectively facilitate the change of the fluorophore from a primarily independent segmental movement to slow global rotation. The time-resolved measurements, including lifetime and dynamic fluorescence anisotropy, were conducted to study the recognition interaction and to better understand the methodology. The proposed method had a wide linear dynamic range of 12.5-300 nM and a high sensitivity with the limit of detection of 4.9 nM (3S/N). This proposed method was successfully applied to assay of human salivary lysozyme. The results based on the standard addition recovery and comparison with enzyme-linked immunosorbent assay (ELISA) demonstrated the feasibility of this method for biological samples. Using coupling between the fluorophore and the analyte can be one of the approaches working toward expanding the application of fluorescence anisotropy based on aptamer-target and antibody-antigen recognitions.

Abstract:We developed a new simple and sensitive assay for lysozyme based on gold nanoparticle plasmon resonance light scattering (PRLS) measurement and naked-eye detection using for the first time the lysozyme DNA aptamer as the recognition element. Lysozyme DNA aptamer could stabilize gold nanoparticles (AuNPs) at high ionic strength. Introducing lysozyme to the system easily triggered the aggregation of AuNPs, producing a red-to-blue color change of the solution, red-shifted plasmon absorption, and enhanced plasmon resonance light scattering. The linear range was found to be 0.2∼4 nM for 0.7 nM AuNPs, 0.3∼6 nM for 1.4 nM AuNPs and 0.6∼8 nM for 2.1 nM AuNPs. About 0.1 nM lysozyme can produce an observable enhancement of PRLS signal. For visual detection, 1 nM lysozyme can produce a very distinctive color change. Satisfactory recoveries were obtained for simulated saliva and diluted urine samples, indicating that the method has potential for analyses of clinical samples. The simplicity and high sensitivity that are consistent with the resources and needs of many laboratories makes this method a good choice for routine analysis. Abstract:This article describes an aptasensor for lysozyme that is based on a gold electrode modified with an aptamer-wrapped composite consisting of three-dimensional reduced graphene oxide, cuprous oxide, and plasma-polymerized propargylamine (Cu2O@rGO@PpPG) as the sensing layer. The nanocomposite consisting of Cu2O@rGO was synthesized by simultaneously reducing GO and Cu(II) ions with glucose and then modified by plasma-enhanced chemical vapor deposition using propargylamine as the monomer gas. The resulting amine-rich nanofilms of Cu2O@rGO@PpPG nanocomposite were deposited on a gold electrode. Differential pulse voltammetry and electrochemical impedance spectroscopy show these layers to exhibit a good amperometric response and variation of the charge transfer resistance to lysozyme after aptamer strands had been immobilized on the films via electrostatic interaction between the negatively charged phosphate groups of the aptamer and the positively charged amino groups on the electrode. The sensor, when operated at 0.22 V (vs. Ag/AgCl), can detect lysozyme in the 0.1 nM to 200 nM concentration range with a 0.06 nM limit of detection. In addition, the sensor displays excellent selectivity and repeatability. In our perception, this strategy for preparing aptasensors holds a great potential with respect to the use of plasma-modified nanocomposites in clinical analysis.
[Ru(bpy)(2)(dcbpy)nhs] Labeling/Aptamer-Based Biosensor for the Detection of Lysozyme by Increasing Sensitivity with Gold Nanoparticle Amplification

Abstract:A novel [Ru(bpy)(2) (dcbpy)NHS] labeling/aptamer-based biosensor combined with gold nanoparticle amplification for the determination of lysozyme with an electrochemiluminescence (ECL) method is presented. In this work, an aptamer, an ECL probe, gold nanoparticle amplification, and competition assay are the main protocols employed in ECL detection. With all the protocols used, an original biosensor coupled with an aptamer and [Ru(bpy)(2)(dcbpy)NHS] has been prepared. Its high selectivity and sensitivity are the main advantages over other traditional [Ru(bpy)(3)](2+) biosensors. The electrochemical impedance spectroscopy (EIS) and atomic force microscopy (AFM) characterization illustrate that this biosensor is fabricated successfully. Finally, the biosensor was applied to a displacement assay in different concentrations of lysozyme solution, and an ultrasensitive ECL signal was obtained. The ECL intensity decreased proportionally to the lysozyme concentration over the range 1.0 x 10-(13)-1.0 x 10(-8) mol L-1 with a detection limit of 1.0 x 10(-13) mol L-1. This strategy for the aptasensor opens a rapid, selective, and sensitive route for the detection of lysozyme and potentially other proteins.

Abstract:A sensitive aptasensor based on a nanocomposite of hollow titanium dioxide nanoball, three-dimensional reduced graphene oxide, and polypyrrole (TiO2/3D-rGO/PPy) was developed for lysozyme detection. A lysozyme aptamer was easily immobilized onto the TiO2/3D-rGO/PPy nanocomposite matrix by assembling the aptamer onto graphene through simple p-stacking interactions and electrostatic interactions between PPy molecular chains and aptamer strands. In the presence of lysozyme, the aptamer on the adsorbent layer catches the target on the electrode interface, which generates a barrier for electrons and inhibits electron transfer, subsequently resulting in decreased electrochemically differential pulse voltammetric signals of a gold electrode modified with TiO2/3D-rGO/PPy. Using this strategy, a low limit of detection of 0.085 ng mL(-1) (5.5 pM) for detecting lysozyme was observed within the detection range of 0.1-50 ng mL(-1) (0.007-3.5 nM). The aptasensor also presents high specificity for lysozyme, which is unaffected by the coexistence of other proteins. Such an aptasensor opens a rapid, selective, and sensitive route to lysozyme detection. This finding indicates that the TiO2/3D-rGO/PPy nanocomposite could be used as an electrochemical biosensor for detecting proteins in the biomedical field.
Feasible Electrochemical Biosensor Based on Plasma Polymerization-Assisted Composite of Polyacrylic Acid and Hollow TiO2 Spheres for Sensitively Detecting Lysozyme

Abstract:A composite made of polyacrylic acid and hollow TiO2 spheres (TiO2@PPAA) was prepared by the plasma polymerization method and subsequently used as an electrode material for detecting lysozyme. The chemical structure, surface morphology, and electrochemical performance of the TiO2@PPAA composite were mainly affected by the plasma input power used during plasma polymerization. After optimizing plasma conditions, aptamer strands exhibited high adsorption affinity toward the surface of TiO2@PPAA composite via synergistic effects between TiO2 and PPAA. Electrochemical impedance spectroscopy results showed that the developed TiO2@PPAA aptasensor presents highly sensitive detection ability toward lysozyme; the limit of detection of the proposed aptasensor is 0.015 ng mL(-1) (1.04 pM) within the range of 0.05-100 ng mL(-1) in terms of 3σ value. The film further showed excellent selectivity toward lysozyme in the presence of interfering proteins, such as thrombin, bovine serum albumin, and immunoglobulin E. Thus, this aptasensing strategy might broaden the applications of plasma polymerized nanomaterials in the field of biomedical research and early clinical diagnosis.

Abstract:This study describes a facile and general strategy for the development of aptamer-based electrochemical sensors with a high specificity toward the targets and a ready regeneration feature. Very different from the existing strategies for the development of electrochemical aptasensors with the aptamers as the probes, the strategy proposed here is essentially based on the utilization of the aptamer-complementary DNA (cDNA) oligonucleotides as the probes for electrochemical sensing. In this context, the sequences at both ends of the cDNA are tailor-made to be complementary and both the redox moiety (i.e., ferrocene in this study) and thiol group are labeled onto the cDNA. The labeled cDNA are hybridized with their respective aptamers (i.e., ATP- and thrombin-binding aptamers in this study) to form double-stranded DNA (ds-DNA) and the electrochemical aptasensors are prepared by self-assembling the labeled ds-DNA onto Au electrodes. Upon target binding, the aptamers confined onto electrode surface dissociate from their respective cDNA oligonucleotides into the solution and the single-stranded cDNA could thus tend to form a hairpin structure through the hybridization of the complementary sequences at both its ends. Such a conformational change of the cDNA resulting from the target binding-induced dissociation of the aptamers essentially leads to the change in the voltammetric signal of the redox moiety labeled onto the cDNA and thus constitutes the mechanism for the electrochemical aptasensors for specific target sensing. The aptasensors demonstrated here with the cDNA as the probe are readily regenerated and show good responses toward the targets. This study may offer a new and relatively general approach to electrochemical aptasensors with good analytical properties and potential applications.

Abstract:Over the last three decades, numerous aptamer-based biosensors have been reported. The basis of these sensors is the selective binding of target analytes by aptamers. In the last few years, a number of papers have been published questioning the binding ability of some popular aptamers such as those documented for As(III), ampicillin, chloramphenicol, isocarbophos, phorate and dopamine. In this article, these papers are reviewed, and the binding assays are described, which may provide possible reasons for obtaining false positive aptamers. Additionally, relevant aptamer selection methods and typical characterization steps are described. It is found that for small molecular targets, using an immobilized library might result in better aptamers. Furthermore, the importance of carefully designed controls to ensure the quality of binding assays is discussed, especially in the case of mutated nonbinding aptamers. Only then, with fully validated aptamers, can subsequent biosensor design bring about meaningful results.

Surfactant-Assisted Label-Free Fluorescent Aptamer Biosensors and Binding Assays

Abstract:Using DNA staining dyes such as SYBR Green I (SGI) and thioflavin T (ThT) to perform label-free detection of aptamer binding has been performed for a long time for both binding assays and biosensor development. Since these dyes are cationic, they can also adsorb to the wall of reaction vessels leading to unstable signals and even false interpretations of the results. In this work, the stability of the signal was first evaluated using ThT and the classic adenosine aptamer. In a polystyrene microplate, a drop in fluorescence was observed even when non-binding targets or water were added, whereas a more stable signal was achieved in a quartz cuvette. Equilibrating the system can also improve signal stability. In addition, a few polymers and surfactants were also screened, and 0.01% Triton X-100 was found to have the best protection effect against fluorescence signal decrease due to dye adsorption. Three aptamers for Hg2+, adenosine, and cortisol were tested for their sensitivity and signal stability in the absence and presence of Triton X-100. In each case, the sensitivity was similar, whereas the signal stability was better for the surfactant. This study indicates that careful control experiments need to be designed to ensure reliable results and that the reliability can be improved by using Triton X-100 and a long equilibration time.

Abstract:The electrostatic complexes of single-stranded deoxyribonucleic acid (ssDNA) and a cationic conjugated polyelectrolyte (CPE), poly{9,9-di[3-(1-ethyl-1,1-dimethyl ammonio)propyl]-2,7-fluorenyl-alt-1,4-phenylene dibromide} (PFN), were investigated. Fluorescence emission of PFN solution (10 mu mol/L) can be drastically quenched to about one fourth of its original intensity in the presence of a trace amount (2.6 mu mol/L) of ssDNA. The effect of oligonucleotide length on the fluorescence quenching behavior was also investigated. In contrast to single-stranded DNA with 20 bases (ssDNA-20), ssDNA with 40 bases (ssDNA-40) induces a relatively higher quenching efficiency and larger red-shift of PFN emission maximum. The binding constant of ssDNA-20 and PFN is estimated to be 1.12 x 10(21). At extremely low concentration (10 nmol/L), PFN can respond to 0.2 nmol/L (or 2 x 10(-10) mol/L) of ssDNA-20 by significant enhancement of its emission intensity. The result is contrary to the observation in the relative higher concentration, and its mechanism is postulated. Based on the high binding ability of ssDNA with cationic CPE, a label-free method for ssDNA detection is designed. It uses an electrostatic complex of cationic PFN and an anionic CPE, which exhibits fluorescence resonance energy transfer (FRET) between the two components. Abstract:Anti-lysozyme aptamers are found to preferentially bind to the edge of a tightly packed lysozyme pattern. Such edge-binding is due to the better accessibility and flexibility of the edge lysozyme molecules. Kelvin probe force microscopy (KPFM) was used to study the aptamer–lysozyme binding. Our results show that KPFM is capable of detecting the aptamer–protein binding down to the 30 nm scale. The surface potential of the aptamer–lysozyme complex is approximately 12 mV lower than that of the lysozyme. The surface potential images of the aptamer-bound lysozyme patterns have the characteristic shoulder steps around the pattern edge, which is much wider than that of a clean lysozyme pattern. These results demonstrate the potentials of KPFM as a label-free method for the detection of protein–DNA interactions. Bispecific Aptamer-Initiated 3D DNA Nanomotor Biosensor Powered by DNAzyme and Entropy-Driven Circuit for Sensitive and Specificity Detection of Lysozyme

Abstract:Dynamic DNA nanodevices have gained tremendous attention due to their extraordinary inherent functionality and advantages, however, dynamic DNA nanodevices-based biosensors are still challenging due to their high reliance on proteases and limited amplification capabilities. Herein, exploiting bispecific aptamer as initiators for the first time, we developed a three-dimensional (3D) DNA nanomotor biosensor powered by DNAzyme and entropy-driven circuit for sensitive and specific detection of lysozyme, in which walking and rolling strategies are efficiently integrated to achieve excellent signal amplification capability. Benefiting from the high selectivity of bispecific aptamer, the 3D DNA nanomotor biosensor can respond to lysozyme with high specificity and operate at high speed to release signals. The whole process is independent of protease, avoiding the influence of adverse environment on the operation stability. Under optimal conditions, it can achieve a limit of detection as low as 0.01 pg/mL with an excellent linear range of 0.05 pg/mL–500 ng/mL for lysozyme. Furthermore, the proposed strategy revealed high accuracy in the analysis of real samples, indicating a great potential for the application of nanomotor biosensors to the detection of non-nucleic acid targets.

Abstract:The aptamers are DNA or RNA oligonucleotides selected in vitro that can bind their targets with high affinity and high specificity. As a kind of new and special recognition molecules , the aptamers present notable advantages over conventional recognition molecules , such as antibody , thus they have received great attention and been widely utilized in various fields involving molecular recognition and relevant applications , especially in biosensing. Abstract:In the present work, electrogenerated chemiluminescence (ECL) of luminol was investigated in neutral condition at a gold electrode in the presence of silicon quantum dots (SiQDs). The results revealed that SiQDs can not only greatly enhance luminol ECL, but also act as energy acceptor to construct a novel ECL resonance energy transfer (ECL-RET) system with luminol. As a result, strong anodic ECL signal was obtained in neutral condition at the bare gold electrode, which is suitable for biosensing application. Lysozyme exhibited apparent inhibiting effect on the ECL-RET system, based on which an ECL aptasensor was fabricated for the sensitive detection of lysozyme. The proposed method showed high sensitivity, good selectivity, and wide linearity for the detection of lysozyme in the range of 5.0×10-14-5.0×10-9gmL-1 with a detection limit of 5.8×10-15gmL-1 (3σ). The results suggested that as-proposed luminol/SiQDs ECL biosensor will be promising in the detection enzyme.

Abstract:Owing to advantage in high sensitivity and fast response, aptamer based electrochemical biosensors have attracted much more attention. However, inappropriate interfacial engineering strategy leads to poor recognition performance, which ascribe to the following factors of immobilized oligonucleotide strand including steric hindrance, interchain entanglement, and unfavorable conformation. In this work, we proposed a DNA tetrahedron based diblock aptamer immobilized strategy for the construction of label-free electrochemical biosensor. The diblock aptamer sequence is composite of T-rich anchor domain and recognition domain, where T-rich domain enabling anchored on the edge of DNA tetrahedron via Hoogsteen hydrogen bond at neutral condition. The DNA tetrahedron scaffold offers an appropriate lateral space for target recognition of diblock aptamer. More importantly, this trivalent aptamer recognition interface can be regenerated by simply adjusting the pH environment to alkaline, resulting in the dissociation of diblock aptamer. Under the optimum condition, proposed electrochemical aptasensor manifested a satisfied sensitivity for aminoglycosides antibiotic, kanamycin with a limit of detection of 0.69 nM, which is 45-fold lower than traditional Au-S immobilization strategy. Moreover, the proposed aptasensor had also successfully been extended to ampicillin detection by changing the sequence of recognition domain in diblock aptamer. This work paves a new way for the rational design of aptamer-based electrochemical sensor.

Abstract:A 61-year-old man with upgaze paralysis was found on postmortem examination to have had a single lesion of the brain stem confined to the periaqueductal gray matter between the levels of the superior and inferior colliculi. Most previous neuropathologic cases reported in human beings have consisted of much larger lesions with common involvement of the posterior commissure and pretectum. We suggest that the lesion in this case caused upgaze paralysis either by interrupting descending fibers from the interstitial nucleus of Cajal or by destroying neurons located within the periaqueductal gray matter.

Abstract:A novel aptasensor was fabricated for the detection of lysozyme and DNA based on the fluorescence resonance energy transfer (FRET) technique between upconversion nanoparticles (UCNPs) and a dye labeled aptamer. UCNPs can act as excellent emitters due to their low autofluorescence and high penetration depth of biosamples. NaYF4:Yb, Er nanoparticles as UCNPs were synthesized and attached with a dye labeled aptamer through a cationic polymer as an electrostatic linker to quench the upconversion fluorescence intensity. The intensity can be restored after the addition of lysozyme or the complementary DNA (target DNA) because of their strong interaction with the aptamer. The sensor provided a linear concentration range from 30 to 210 nM for lysozyme and 40 to 200 nM for the target DNA, the limit of detection was 2.5 nM and 2.8 nM, respectively. The sensor was also used to monitor the lysozyme level in both human saliva and serum samples, and the results were consistent with the reported values. The method was simple and convenient without the extra procedure of bioconjugation, and could be put to use for the determination of various targets in the future.
