Abstract:Drug designing and development is an important area of research for pharmaceutical companies and chemical scientists. However, low efficacy, off-target delivery, time consumption, and high cost impose a hurdle and challenges that impact drug design and discovery. Further, complex and big data from genomics, proteomics, microarray data, and clinical trials also impose an obstacle in the drug discovery pipeline. Artificial intelligence and machine learning technology play a crucial role in drug discovery and development. In other words, artificial neural networks and deep learning algorithms have modernized the area. Machine learning and deep learning algorithms have been implemented in several drug discovery processes such as peptide synthesis, structure-based virtual screening, ligand-based virtual screening, toxicity prediction, drug monitoring and release, pharmacophore modeling, quantitative structure–activity relationship, drug repositioning, polypharmacology, and physiochemical activity. Evidence from the past strengthens the implementation of artificial intelligence and deep learning in this field. Moreover, novel data mining, curation, and management techniques provided critical support to recently developed modeling algorithms. In summary, artificial intelligence and deep learning advancements provide an excellent opportunity for rational drug design and discovery process, which will eventually impact mankind.

The inclusion of AI in drug discovery can screen millions of compounds in a matter of hours, identifying potential drug candidates that would have taken years to find using traditional methods. AI is highly utilized in the pharmaceutical industry by optimizing processes, reducing waste, and ensuring quality control. This review covers much-needed topics, including the different types of machine-learning techniques, their applications in drug discovery, and the challenges and limitations of using machine learning in this field. The state-of-the-art of AI-assisted pharmaceutical discovery is described, covering applications in structure and ligand-based virtual screening, de novo drug creation, prediction of physicochemical and pharmacokinetic properties, drug repurposing, and related topics. This review elaborates on the opportunities and challenges of AI in healthcare and pharmaceutical research. The literature was collected from domains such as PubMed, Science Direct and Google scholar using specific keywords and phrases such as ‘Artificial intelligence’, ‘Pharmaceutical research’, ‘drug discovery’, ‘clinical trial’, ‘disease diagnosis’, etc. to select the research and review articles published within the last five years. The application of AI in disease diagnosis, digital therapy, personalized treatment, drug discovery and forecasting epidemics or pandemics was extensively reviewed in this article. Deep learning and neural networks are the most used AI technologies; Bayesian nonparametric models are the potential technologies for clinical trial design; natural language processing and wearable devices are used in patient identification and clinical trial monitoring. Deep learning and neural networks were applied in predicting the outbreak of seasonal influenza, Zika, Ebola, Tuberculosis and COVID-19. With the advancement of AI technologies, the scientific community may witness rapid and cost-effective healthcare and pharmaceutical research as well as provide improved service to the general public.

The exploitation of artificial intelligence (AI) and big data will enable in silico prediction of many parameters that are presently tested in vivo. Automation and multiplexing synthetic approaches will allow a broader range of exploration of screening of thousands of compounds with better reproducibility at lower consumption and overall lower costs thereby effectively speeding up the process of drug discovery. Other advanced techniques such as Machine Learning (ML) and Deep Learning (DL) algorithms are now being used to perform virtual screening of various lead compounds that can be developed into drugs. Integration of new technology such as AI have with drug discovery process will make the design and optimization process more robust by substantially eliminating bias and human mistakes as well as the time required for testing the candidate compounds for drug development. Abstract:Artificial Intelligence, abbreviated as AI, is described as a collection of technologies that work together to allow machines to discern, interpret, take action, and acquire knowledge with human level of intelligence. The AI domain includes technologies such as natural language processing (NLP) and machine learning (ML). Each of these is on its separate route of development and, when combined with datasets, analytics, and computerization, may assist organisations in achieving their objectives, whether it's enhancing customer service or improving supply chain. The pipeline of the development of novel drugs is quite extensive and costly apart from being complicated. On average, this entire process requires around 12 years costing approximately 2.6 billion USD. In this age of computational power, AI-based techniques are used in almost all of the steps during the process of discovering and developing drugs. AI has been able to reduce the time requirement and amount of money involved in this process thus expediting the entire procedure of new pharmaceutical inventions. In this manuscript, we have discussed some of the major applications of AI in the pharmaceutical sector. Tools that are employed to forecast the toxicity of drugs and how some of them, for example - Toxtree, ADMET, ProTox etc. have been used by researchers in the drug discovery process, have also been talked about. Further we have mentioned how AI is proving to be a powerful tool in the fight against the COVID-19 pandemic, for example, in the detection of the SARS-Co V -2 virus, development of vaccines, genomics etc. In addition to this, we have discussed about the manner in which the penetration of AI-based companies into the pharmaceutical field has resulted in some notable outcomes, for instance - prediction of the 3-dimensional structure of proteins by DeepMind's AlphaFold2, determination of new drug contender for kidney fibrosis by Insilico Medicine's Chemistry42 etc. Furthermore, we have stated the propitious future that AI is expected to bring about in the pharma world with a special focus on drug development.

Much of the initial skepticism regarding applications of AI in pharmaceutical discovery has started to vanish, consequently benefitting medicinal chemistry.Areas covered: The current status of AI in chemoinformatics is reviewed. The topics discussed herein include quantitative structure-activity/property relationship and structure-based modeling, de novo molecular design, and chemical synthesis prediction. Advantages and limitations of current deep learning applications are highlighted, together with a perspective on next-generation AI for drug discovery.Expert opinion: Deep learning-based approaches have only begun to address some fundamental problems in drug discovery. Certain methodological advances, such as message-passing models, spatial-symmetry-preserving networks, hybrid de novo design, and other innovative machine learning paradigms, will likely become commonplace and help address some of the most challenging questions. Open data sharing and model development will play a central role in the advancement of drug discovery with AI.

Abstract:Drug discovery and development affects various aspects of human health and dramatically impacts the pharmaceutical market. However, investments in a new drug often go unrewarded due to the long and complex process of drug research and development (R&D). With the advancement of experimental technology and computer hardware, artificial intelligence (AI) has recently emerged as a leading tool in analyzing abundant and high-dimensional data. Explosive growth in the size of biomedical data provides advantages in applying AI in all stages of drug R&D. Driven by big data in biomedicine, AI has led to a revolution in drug R&D, due to its ability to discover new drugs more efficiently and at lower cost. This review begins with a brief overview of common AI models in the field of drug discovery; then, it summarizes and discusses in depth their specific applications in various stages of drug R&D, such as target discovery, drug discovery and design, preclinical research, automated drug synthesis, and influences in the pharmaceutical market. Finally, the major limitations of AI in drug R&D are fully discussed and possible solutions are proposed.

Abstract:BACKGROUND The lengthy and expensive process of developing a novel medicine often takes many years and entails a significant financial burden due to its poor success rate. Furthermore, the processing and analysis of quickly expanding massive data necessitate the use of cutting-edge methodologies. As a result, Artificial Intelligence-driven methods that have been shown to improve the efficiency and accuracy of drug discovery have grown in favor. OBJECTIVE The goal of this thorough analysis is to provide an overview of the drug discovery and development timeline, various approaches of drug design, and the use of Artificial Intelligence in many aspects of drug discovery. METHODS Traditional drug development approaches and their disadvantages are explored, followed by an introduction to AI-based technology. Also, advanced methods used in Machine Learning and Deep Learning are examined in detail. A few examples of big data research that has transformed the field of medication discovery have also been presented. Also covered are the many databases, toolkits, and software available for constructing Artificial Intelligence/Machine Learning models, as well as some standard model evaluation parameters. Finally, recent advances and uses of Machine Learning and Deep Learning in drug discovery are thoroughly examined, along with their limitations and future potential. CONCLUSION Artificial Intelligence-based technologies enhance decision-making by utilizing the abundantly available high-quality data, and thereby, reduce the time and cost involved in the process. We anticipate that this review would be useful to researchers interested in Artificial Intelligence-based drug development.

Abstract:Personalized medicine, medication discovery, and development might all benefit greatly from AI’s incorporation into pharmacokinetics and pharmacodynamics. Target identification, therapeutic effectiveness prediction, drug design optimization, obstacles, and future possibilities are all explored in this survey of AI applications in these areas. An overview of pharmacokinetics and pharmacodynamics is presented first, stressing the significance of knowing how drugs are absorbed, distributed, metabolized, and excreted and the correlation between drug concentration and pharmacological effect. The article then looks into the function of AI in target identification, exploring how machine learning algorithms and data integration may be used to discover new drug targets and enhance the design of existing ones. Classification and regression methods are also investigated for their potential use in the prediction of therapeutic efficacy using AI. Patient data, molecular interaction data, and clinical response data are just a few examples of the types of data that may be used to fuel the creation of predictive models that might assist in dosage and efficacy optimization. Metrics and procedures for validating these models are addressed to evaluate their efficacy. Additionally, de novo drug design, virtual screening, and structure-based drug design are all discussed in relation to the use of AI in optimizing drug development. The paper provides examples of how AI has been applied successfully in different settings, demonstrating its potential to hasten the drug discovery process and enhance treatment outcomes. We examine data availability, interpretability, and ethical implications as challenges and limits of AI in pharmacokinetics and pharmacodynamics. To guarantee these technologies’ proper and ethical use, we also discuss the regulatory elements and rules for applying AI in drug research. Possibilities and prospects for the use of AI in pharmacokinetics and pharmacodynamics are discussed as a conclusion to the review. It stresses the significance of regulatory standards and clinical translation, as well as the incorporation of multiomics data, deep learning methods, real-time monitoring, explainable artificial intelligence, collaborative networks, and more.

Abstract:Artificial intelligence (AI) encompasses a broad spectrum of techniques that have been utilized by pharmaceutical companies for decades, including machine learning, deep learning, and other advanced computational methods. These innovations have unlocked unprecedented opportunities for the acceleration of drug discovery and delivery, the optimization of treatment regimens, and the improvement of patient outcomes. AI is swiftly transforming the pharmaceutical industry, revolutionizing everything from drug development and discovery to personalized medicine, including target identification and validation, selection of excipients, prediction of the synthetic route, supply chain optimization, monitoring during continuous manufacturing processes, or predictive maintenance, among others. While the integration of AI promises to enhance efficiency, reduce costs, and improve both medicines and patient health, it also raises important questions from a regulatory point of view. In this review article, we will present a comprehensive overview of AI's applications in the pharmaceutical industry, covering areas such as drug discovery, target optimization, personalized medicine, drug safety, and more. By analyzing current research trends and case studies, we aim to shed light on AI's transformative impact on the pharmaceutical industry and its broader implications for healthcare.
