Abstract:Salt stress is a serious problem, because it reduces the plant growth and seed yield of wheat. To investigate the salt-tolerant mechanism of wheat caused by plant-derived smoke (PDS) solution, metabolomic and proteomic techniques were used. PDS solution, which repairs the growth inhibition of wheat under salt stress, contains metabolites related to flavonoid biosynthesis. Wheat was treated with PDS solution under salt stress and proteins were analyzed using a gel-free/label-free proteomic technique. Oppositely changed proteins were associated with protein metabolism and signal transduction in biological processes, as well as mitochondrion, endoplasmic reticulum/Golgi, and plasma membrane in cellular components with PDS solution under salt stress compared to control. Using immuno-blot analysis, proteomic results confirmed that ascorbate peroxidase increased with salt stress and decreased with additional PDS solution; however, H+-ATPase displayed opposite effects. Ubiquitin increased with salt stress and decreased with additional PDS solution; nevertheless, genomic DNA did not change. As part of mitochondrion-related events, the contents of ATP increased with salt stress and recovered with additional PDS solution. These results suggest that PDS solution enhances wheat growth suppressed by salt stress through the regulation of energy metabolism and the ubiquitin-proteasome system related to flavonoid metabolism.
Proteomic Analysis to Understand the Promotive Effect of Ethanol on Soybean Growth Under Salt Stress

Abstract:Finding solutions to mitigate the impact of salinity on crops is important for global food security because soil salinity significantly reduces plant growth and grain yield. Ethanol may play an important role in mitigating the negative salt-induced effects on crops. Soybean root growth was significantly reduced under salt stress; however, it was restored and comparable to control values by ethanol application even under stress. To study the positive mechanism of ethanol on soybean growth, a proteomic approach was carried out. The categories with the greatest changes in protein numbers were protein metabolism, transport, and cell organization in biological processes, nucleus and cytosol in cellular components, and nucleic acid binding activity in molecular functions. Proteomic data were confirmed using immunoblot analysis. Reactive oxygen species enzymes increased under salt stress; among them, mitochondrial ascorbate peroxidase was further accumulated by ethanol application. Among the cell wall and membrane-associated proteins, xyloglucan xyloglucosyl transferase and H+-ATPase increased and decreased, respectively, under salt stress; however, they were restored to control levels by ethanol application. These results suggest that soybeans were adversely affected by salt stress and recovered with ethanol application via the regulation of cell wall and membrane functions through the detoxification of reactive oxygen species.

Abstract:Salinity stress is a threat to yield in many crops, including soybean (Glycine max L.). In this study, three soybean cultivars (JD19, LH3, and LD2) with different salt resistance were used to analyze salt tolerance mechanisms using physiology, transcriptomic, metabolomic, and bioinformatic methods. Physiological studies showed that salt-tolerant cultivars JD19 and LH3 had less root growth inhibition, higher antioxidant enzyme activities, lower ROS accumulation, and lower Na+ and Cl- contents than salt-susceptible cultivar LD2 under 100 mM NaCl treatment. Comparative transcriptome analysis showed that compared with LD2, salt stress increased the expression of antioxidant metabolism, stress response metabolism, glycine, serine and threonine metabolism, auxin response protein, transcription, and translation-related genes in JD19 and LH3. The comparison of metabolite profiles indicated that amino acid metabolism and the TCA cycle were important metabolic pathways of soybean in response to salt stress. In the further validation analysis of the above two pathways, it was found that compared with LD2, JD19, and LH3 had higher nitrogen absorption and assimilation rate, more amino acid accumulation, and faster TCA cycle activity under salt stress, which helped them better adapt to salt stress. Taken together, this study provides valuable information for better understanding the molecular mechanism underlying salt tolerance of soybean and also proposes new ideas and methods for cultivating stress-tolerant soybean.

Plant-Derived Smoke Ameliorates Salt Stress in Wheat by Enhancing Expressions of Stress-Responsive Genes and Antioxidant Enzymatic Activity

Abstract:Abiotic stresses are the biggest threat to the increasing population worldwide. Salt stress is one of the most significant abiotic stresses, affecting 20% of the crop production around the world. Plant-derived smoke (PDS) has been reported as a biologically active plant product in stimulating seed germination, seedling growth and physiological characteristics of crops under abiotic stress conditions. Nevertheless, studies showing how PDS alleviates salt stress are largely unknown. Here, we report the molecular mechanism of how PDS could alleviate salt stress in wheat. Initially, PDS at 2000 ppm enhanced seed germination, root/shoot length and seedling fresh weight. However, PDS at 1000 and 500 ppm did not show any significant effect. Salt stress at 150 and 200 mM significantly reduced seed germination rate, root/shoot length and fresh weight of the wheat seedlings. Interestingly, PDS supplementation at 2000 ppm concentration was sufficient to restore seed germination under salt stress condition. Moreover, PDS improved root/shoot length and seedling biomass under 150 and 200 mM salt stress, suggesting that PDS is a potent plant product, capable of abiotic stress alleviation in crops. In comparison to the control, PDS-treated seedlings displayed increased activity of major antioxidative enzymes such as superoxide dismutase, peroxidase and ascorbate peroxidase under salt stress, resulting in reduced levels of hydrogen peroxide and lipid peroxidase, showing that PDS can possibly help in salt stress amelioration by regulating redox homeostasis. Importantly, salt stress altered the expression of germination marker genes, such as TaSAM, TaPHY, TaBGU (germination positive effectors), TaLEA and TaGARS34 (germination negative effectors), suggesting the potential role of PDS in the germination pathway under salt stress. Further, PDS modulated the transcript levels of several salt stress stress-responsive genes, including TaSOS4, TaBADH and TaHKT2. In conclusion, this study provides a molecular and physiological basis for elucidating the mechanism of how PDS functions in stress induction in wheat, as well as demonstrates the importance of PDS in agricultural practices, laying the groundwork for future research into the role of PDS in the amelioration of abiotic stresses in various plants.
Integration of Physiological and Transcriptomic Approaches in Investigating Salt-Alkali Stress Resilience in Soybean

Abstract:Salt-alkali stress is a key factor affecting plant development and production, altering essential physiological and metabolic processes within plants. Particularly, soybean ( Glycine max ) exhibits a wide range of saline-alkali tolerance in soybean germplasms. Hence, the objective of this research was to pinpoint potential genes that react to saline-alkali stress through a comparative transcriptome analysis of two distinct soybean varieties exposed to such stress conditions. The two soybean varieties, including salt-tolerant Heinong531 and salt-sensitive 20_1846, were used for RNA-seq analysis. Nine physiological parameters including malondialdehyde (MDA), superoxide dismutase (SOD), glutathione (GSH), peroxidase (POD), proline, catalase (CAT), soluble protein, scavenging rate of hydroxyl radical, and soluble sugar activity were measured under the saline-alkaline treatment series of 0,1,2,3,4 days. The results showed that the levels of MDA, proline, and catalase contents decreased significantly, while levels of GSH and POD contents significantly increased. A total of 200 saline-alkali tolerance responsive differentially expressed genes (DEGs) were identified, comprising 93 up-regulated genes and 107 down-regulated. The functional gene ontology analysis for these 200 DEGs, where 29 key DEGs were found to be involved in the regulation of 14 GO pathways responding to salt-alkaline stress which have been previously considered as vital for salt resistance mechanisms in soybean. Remarkably, the association analysis between physiological parameters and Differentially Expressed Genes (DEGs) under salt-alkali stress conditions demonstrated that distinct soybean cultivars exhibited unique patterns of gene expression, ultimately resulting in alterations in physiological indices as a response to salt stress. This study offers a compilation of potential genes worthy of additional exploration concerning their role in enhancing soybean's resistance to salt-alkali stress.

Symbiotic System Establishment between Piriformospora indica and Glycine max and Its Effects on the Antioxidant Activity and Ion-Transporter-Related Gene Expression in Soybean under Salt Stress

Abstract:The utilization of symbiosis with beneficial microorganisms has considerable potential for increasing growth and resistance under abiotic stress. The endophytic root fungus Piriformospora indica has been shown to improve plant growth under salt and drought stress in diverse plant species, while there have been few reports of the interaction of P. indica with soybean under salt stress. In this study, the symbiotic system of P. indica and soybean (Glycine max L.) was established, and the effect of P. indica on soybean growth and salt tolerance was investigated. The colonized and non-colonized soybeans were subjected to salt stress (200 mmol/L NaCl), and the impairments in chlorophyll and increasing relative conductivity that can be caused by salt stress were alleviated in the P. indica-colonized plants. The accumulation of malondialdehyde (MDA), hydrogen peroxide (H2O2), and superoxide anion (O2−) were lower than that in non-colonized plants under salt treatment, whereas the activities of antioxidant enzymes were significantly increased by P. indica colonization, including superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and glutathione reductase (GR). Importantly, without salt treatment, the Na+ concentration was lower, and the K+ concentration was higher in the roots compared with non-colonized plants. Differential expressions of ion transporter genes were found in soybean roots after P. indica colonization. The P. indica colonization positively regulated the transcription level of PM H+-ATPase, SOS1, and SOS2. The study shows that P. indica enhances the growth and salt tolerance of soybean, providing a strategy for the agricultural production of soybean plants in saline-alkali soils.

Abstract:Plant-derived smoke (PDS) is an emerging area of interest for research in plant growth during stress conditions. Two wheat varieties (NARC 2011 and NARC 2009) were imbibed in different dilutions of PDS solution and were subjected to salinity (50 mM and 100 mM NaCl). Among all three concentrations (1:100, 1:250, and 1:500), the PDS solution 1:500 has shown improvement in germination, morphological, physiological, and biochemical attributes. It has shown an increase in germination percentage by 48.9% and 27.7% at 50 mM and 100 mM salinity, respectively. Similar results were observed among plant physiological attributes (i.e., relative water content, leaf water potential, leaf osmotic potential, membrane stability index, leaf chlorophyll contents, ionic contents) and biochemical parameters (i.e., proline contents, free amino acid, soluble sugar, protein contents). Among antioxidants enzyme assay, superoxide dismutase activity has shown a reduction of 14.02% and 10.5% at 50 mM and 100 mM NaCl, respectively, by application of PDS solution (1:500 conc.). Compound 1 isolated as a result of repeated column chromatography was light brown and its name [1,4-dihydroxybenzene (hydroquinone)] was confirmed by mass spectrometry. Results have shown that lower concentrations (10 ppm and 25 ppm) exhibited more stimulatory effects on seed germination. In light of our findings, it can be suggested that PDS has shown positive and stimulatory effects on wheat variety NARC 2011 under stress conditions. Moreover, it can be used as an alternative to phytohormones in the coming future.

Dynamin GTPase was downregulated under salt stress but recovered with safranal application under salt stress. Abstract:Plant-derived smoke is known as a potent agent to promote plant growth at different developmental stages. The application of plant-derived smoke elicits germination and post-germination stages of plant species. On the other hand, abiotic stresses, such as salinity, heavy metals, and flooding, are known to weaken plant growth and development. Abiotic stresses have negative effects on growth attributes of crops, such as maize, rice, wheat, and soybean. The positive/alleviating effect of plant-derived smoke solution was observed at morphological, physiological, and molecular levels of plant under abiotic stresses. Presently, the role of plant-derived smoke in alleviation of various stresses including salt, heavy metal, temperature, and flooding stress has been comprehended and the mechanism of alleviation via regulation of cellular activities at molecular level has been discussed. The amelioration of negative effects of abiotic stresses by plant-derived smoke involves the regulation of stress-related genes that provokes the antioxidant system of plants, activating cell wall proteins and metabolic pathways that might block the uptake of salt/heavy metal ions through plant roots. This review might provide an in-depth role of plant-derived smoke to exploit new avenues enabling plants to cope with abiotic stresses.

Abstract:Soybean (Glycine max) is economically significant, but the mechanisms underlying its adaptation to simultaneous low phosphorus and salt stresses are unclear. We employed the Shennong 94–1‐8 soybean germplasm to conduct a comprehensive analysis, integrating both physiochemical and transcriptomic approaches, to unravel the response mechanisms of soybean when subjected to simultaneous low phosphorus and salt stresses. Remarkably, the combined stress exhibited the most pronounced impact on the soybean root system, which led to a substantial reduction in total soluble sugar (TSS) and total soluble protein (TSP) within the plants under this treatment. A total of 20,953 differentially expressed genes were identified through pairwise comparisons. Heatmap analysis of genes related to energy metabolism pathways demonstrated a significant down‐regulation in expression under salt and low phosphorus + salt treatments, while low phosphorus treatment did not exhibit similar expression trends. Furthermore, the weighted gene co‐expression network analysis (WGCNA) indicated that the blue module had a strong positive correlation with TSS and TSP. Notably, 2,3‐bisphosphoglycerate‐dependent phosphoglycerate mutase 1, FCS‐Like Zinc finger 8, auxin response factor 18 isoform X2, and NADP‐dependent malic enzyme emerged as hub genes associated with energy metabolism. In summary, our findings indicate that soybean roots are more adversely affected by salt and combined stress than by low phosphorus alone due to reduced activity in energy metabolism‐related pathways and hub genes. These results offer novel insights into the adaptive mechanisms of soybeans when facing the combined stress of low phosphorus and salinity.

Abstract:Drought and salinity stresses are persistent threat to field crops and are frequently mentioned as major constraints on worldwide agricultural productivity. Moreover, their severity and frequency are predicted to rise in the near future. Therefore, in the present study we investigated the mechanisms underlying plant responses to drought (5, 10 and 15% polyethylene glycol, PEG-6000), salinity (50, 100, and 150 mM NaCl), and their combination, particularly at the seed germination stage, in terms of photosynthesis and antioxidant activity, in four soybean cultivars, viz., PI408105A (PI5A), PI567731 (PI31), PI567690 (PI90), and PI416937 (PI37). Results showed that seed germination was enhanced by 10% PEG and decreased by 15% PEG treatments compared to the control, while seed germination was drastically decreased under all levels of NaCl treatment. Furthermore, combined drought and salinity treatment reduced plant height and root length, shoot and root total weights, and relative water content compared with that of control. However, the reductions were not similar among the varieties, and definite growth retardations were observed in cultivar PI5A under drought and in PI37 under salinity. In addition, all treatments resulted in substantially reduced contents of chlorophyll pigment, anthocyanin, and chlorophyll fluorescence; and increased lipid peroxidation, electrolyte leakage, and non-photochemical quenching in all varieties of soybean as compared to the control plants. However, proline, amino acids, sugars, and secondary metabolites were increased with the drought and salinity stresses alone. Moreover, the reactive oxygen species accumulation was accompanied by improved enzymatic antioxidant activity, such as that of superoxide dismutase, peroxidase, catalase, and ascorbate peroxidase. However, the enhancement was most noticeable in PI31 and PI90 under both treatments. In conclusion, the cultivar PI31 has efficient drought and salinity stress tolerance mechanisms, as illustrated by its superior photosynthesis, osmolyte accumulation, antioxidative enzyme activity, and secondary metabolite regulation, compared to the other cultivars, when stressed.

Abstract:The increasing sodium salts (NaCl, NaHCO3, NaSO4 etc.) in agricultural soil is a serious global concern for sustainable agricultural production and food security. Soybean is an important food crop, and their cultivation is severely challenged by high salt concentration in soils. Classical transgenic and innovative breeding technologies are immediately needed to engineer salt tolerant soybean plants. Additionally, unfolding the molecular switches and the key components of the soybean salt tolerance network are crucial for soybean salt tolerance improvement. Here we review our understandings of the core salt stress response mechanism in soybean. Recent findings described that salt stress sensing, signalling, ionic homeostasis (Na + /K + ) and osmotic stress adjustment might be important in regulating the soybean salinity stress response. We also evaluated the importance of antiporters and transporters such as Arabidopsis K + Transporter 1 ( AKT1 ) potassium channel and the impact of epigenetic modification on soybean salt tolerance. We also review key phytohormones, and osmo-protectants and their role in salt tolerance in soybean. In addition, we discuss the progress of omics technologies for identifying salt stress responsive molecular switches and their targeted engineering for salt tolerance in soybean. This review summarizes recent progress in soybean salt stress functional genomics and way forward for molecular breeding for developing salt-tolerant soybean plant.

Proteome analysis of soybean roots under waterlogging stress at an early vegetative stage

Abstract:To gain better insight into how soybean roots respond to waterlogging stress, we carried out proteomic profiling combined with physiological analysis at two time points for soybean seedlings in their early vegetative stage. Seedlings at the V2 stage were subjected to 3 and 7 days of waterlogging treatments. Waterlogging stress resulted in a gradual increase of lipid peroxidation and in vivo H2O2 level in roots. Total proteins were extracted from root samples and separated by two-dimensional gel electrophoresis (2-DE). A total of 24 reproducibly resolved, differentially expressed protein spots visualized by Coomassie brilliant blue (CBB) staining were identified by matrix assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometry or electrospray ionization tandem mass spectrometry (ESI-MS/MS) analysis. Of these, 14 proteins were upregulated; 5 proteins were decreased; and 5 were newly induced in waterlogged roots. The identified proteins include well-known classical anaerobically induced proteins as well as novel waterlogging-responsive proteins that were not known previously as being waterlogging responsive. The novel proteins are involved in several processes, i.e. signal transduction, programmed cell death, RNA processing, redox homeostasis and metabolisms of energy. An increase in abundance of several typical anaerobically induced proteins, such as glycolysis and fermentation pathway enzymes, suggests that plants meet energy requirement via the fermentation pathway due to lack of oxygen. Additionally, the impact of waterlogging on the several programmed cell death- and signal transduction-related proteins suggest that they have a role to play during stress. RNA gel blot analysis for three programmed cell death-related genes also revealed a differential mRNA level but did not correlate well with the protein level. These results demonstrate that the soybean plant can cope with waterlogging through the management of carbohydrate consumption and by regulating programmed cell death. The identification of novel proteins such as a translation initiation factor, apyrase, auxin-amidohydrolase and coproporphyrinogen oxidase in response to waterlogging stress may provide new insight into the molecular basis of the waterlogging-stress response of soybean.
Salicylic Acid and Pyraclostrobin Can Mitigate Salinity Stress and Improve Anti-Oxidative Enzyme Activities, Photosynthesis, and Soybean Production under Saline–Alkali Regions

Abstract:Soybean is a widespread crop in semi-arid regions of China, where soil salinity often increases and has a significant harmful impact on production, which will be a huge challenge in the coming years. Salicylic acid (SA) and pyraclostrobin are strobilurin-based bactericides (PBF). Under rainfall-harvesting conditions in covered ridges, the exogenous application of SA and PBF can improve the growth performance of soybeans, thereby reducing the adverse effects of soil salinity. The objectives of this research are to evaluate the potential effects of SA and PBF on soybean growth in two different regions, Harbin and Daqing. A two-year study was performed with the following four treatments: HCK: Harbin location with control; SA1+PBF1: salicylic acid (5 mL L−1) with pyraclostrobin (3 mL L−1); SA2+PBF2: salicylic acid (10 mL L−1) with pyraclostrobin (6 mL L−1); DCK: Daqing location with control. The results showed that in the Harbin region, SA2+PBF2 treatment reduced the evapotranspiration (ET) rate, increased soil water storage (SWS) during branching and flowering stages, and achieved a maximum photosynthesis rate. Moreover, this improvement is due to the reduction of MDA and oxidative damage in soybean at various growth stages. At different growth stages, the treatment of Harbin soybean with SA2+PBF2 significantly increased the activity of CAT, POD, SOD, and SP, while the content of MDA, H2O2, and O2− also decreased significantly. In the treatment of SA2+PBF2 in Harbin, the scavenging ability of free H2O2 and O2− was higher, and the activity of antioxidant enzymes was better. This was due to a worse level of lipid-peroxidation which successfully protected the photosynthesis mechanism and considerably increased water use efficiency (WUE) (46.3%) and grain yield (57.5%). Therefore, using plastic mulch with SA2+PBF2 treatment can be an effective water-saving management strategy, improving anti-oxidant enzyme activities, photosynthesis, and soybean production.

Abstract:Clarification of the metabolic mechanisms underlying salt stress responses in plants will allow further optimization of crop breeding and cultivation to obtain high yields in saline-alkali land. Here, we characterized 68 differential metabolites of cultivated soybean (Glycine max) and wild soybean (Glycine soja) under neutral-salt and alkali-salt stresses using gas chromatography-mass spectrometry (GC-MS)-based metabolomics, to reveal the physiological and molecular differences in salt tolerance. According to comparisons of growth parameters under the two kinds of salt stresses, the level of inhibition in wild soybean was lower than in cultivated soybean, especially under alkali-salt stress. Moreover, wild soybean contained significantly higher amounts of phenylalanine, asparagine, citraconic acid, citramalic acid, citric acid and α-ketoglutaric acid under neutral-salt stress, and higher amounts of palmitic acid, lignoceric acid, glucose, citric acid and α-ketoglutaric acid under alkali-salt stress, than cultivated soybean. Further investigations demonstrated that the ability of wild soybean to salt tolerance was mainly based on the synthesis of organic and amino acids, and the more active tricarboxylic acid cycle under neutral-salt stress. In addition, the metabolite profiling analysis suggested that the energy generation from β-oxidation, glycolysis and the citric acid cycle plays important roles under alkali-salt stress. Our results extend the understanding of mechanisms involved in wild soybean salt tolerance and provide an important reference for increasing yields and developing salt-tolerant soybean cultivars.

Abstract:Saline‐alkali stress (SS) is a common abiotic stress affecting crop cultivation worldwide, seriously inhibiting plant growth and biomass accumulation. Melatonin has been proven to relieve the inhibition of multiple abiotic stresses on plant growth. Therefore, soybean cultivars Heihe 49 (HH49, SS‐tolerant) and Henong 95 (HN95, SS‐sensitive) were pot‐cultured in SS soil and then treated with 300 μM melatonin at the V1 stage, when the first trifoliate leaves were fully unfolded, to investigate if melatonin has an effect on SS. SS increased reactive oxygen species (ROS) accumulation in soybean leaves and thereby induced DNA oxidative damage. In addition, SS retarded cell growth and decreased the mesophyll cell size, chloroplast number, photosynthetic pigment content, which further reduced the light energy capture and electron transport rate in soybean leaves, and affected carbohydrate accumulation and metabolism. However, melatonin treatment reduced SS‐induced ROS accumulation in the soybean leaves by increasing antioxidant content and oxidase activity. Effective removal of ROS reduced SS‐induced DNA oxidative damage in the soybean leaf genome, which was represented by decreased random‐amplified polymorphic DNA polymorphism, 8‐hydroxy‐20‐deoxyguanine content, and relative density of apurinic/apyrimidinic‐sites. Melatonin treatment also increased the volume of mesophyll cells, the numbers of chloroplast and starch grains, the contents of chlorophyll a and b and carotenoids in soybean seedling leaves treated with SS, thereby increasing the efficiency of effective light capture and electron transfer and improving photosynthesis. Subsequently, carbohydrate accumulation and metabolism in soybean leaves under SS were improved by melatonin treatment, which contributes to providing basic substances and energy for cell growth and metabolism, ultimately improving soybean SS tolerance.

Abstract:Soybean, an important cash crop, is often affected by soil salinity, which is one of the important types of abiotic stress that affects its growth. Poly (acrylic) acid coated Mn3O4 (PMO) has been reported to play a vital role in defending against a variety of abiotic stresses in plants. To date, the effects of PMOs on soybean have not been reported; this study explored the mechanism of PMO-enhanced soybean germination under salt stress. In this experiment, 100 mg/L PMO was used as an immersion agent with a salt treatment of 150 mM NaCl. The results showed that when compared with the PMO treatment, salt stress significantly decreased the germination rate, fresh weight, carbohydrate content, and antioxidant enzyme activity of soybean and significantly increased the contents of reactive oxygen species, malondialdehyde, and osmoregulatory substances. However, PMO treatment enhanced the antioxidant defense system and significantly reduced the malondialdehyde content of soybean. Moreover, the activities of H+-ATPase and Ca2+-ATPase were significantly higher in treated soybean than in the control, and the content of ATP was also higher in treated soybean than in the control. Generally, PMO regulates the homeostasis of reactive oxygen species and reduces ATP consumption, thereby improving the ability of soybeans to germinate under salt stress. This study provides new insights into how nanomaterials improve plant salt tolerance.

Abstract:Drought stress is a major limiting factor for soybean production. In this study, protein elicitor AMEP412 was applied on soybean seedlings to promote drought tolerance, and its correlation with alterations in the reactive oxygen species (ROS) in leaves was studied. The plants of soybean cv. Suinong 26 were sprayed with AMEP412 at the V1 stage and then treated with drought stress and rehydration. The alterations in phenotypes, physiological indices, ROS levels and antioxidant system as well as lipid peroxidation were monitored. The results showed that spraying AMEP412 significantly promoted the drought tolerance of soybean seedlings. Soybean seedlings pre-treated with AMEP412 performed better in phenotypes and physiological indexes, like leaf relative water content (RWC), leaf relative conductivity (RC) and osmolytes (free proline, soluble sugar and protein). In addition, the in situ and quantification detection of ROS revealed that AMEP412 pretreatment led to higher accumulation of ROS at the early stage and faster scavenging of ROS at the late stage. Moreover, AMEP412 pretreatment increased the activity of superoxide dismutase (SOD), guaiacol peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), glutathione reductase (GR), glutathione peroxidase (GPX), and the levels of ascorbate (AsA) and glutathione (GSH), but decreased the level of malondialdehyde (MDA) as compared with only drought treatment. Overall, the results indicated that AMEP412 was effective in alleviating the adverse effects of drought stress, which was partially attributable to the ROS accumulation and scavenging triggered by AMEP412.

Effects of salt stress on soil enzyme activities and rhizosphere microbial structure in salt-tolerant and -sensitive soybean

Abstract:Salt is recognized as one of the most major factors that limits soybean yield in acidic soils. Soil enzyme activity and bacterial community have a critical function in improving the tolerance to soybean. Our aim was to assess the activities of soil enzyme, the structure of bacteria and their potential functions for salt resistance between Salt-tolerant (Salt-T) and -sensitive (Salt-S) soybean genotypes when subject to salt stress. Plant biomass, soil physicochemical properties, soil catalase, urease, sucrase, amylase, and acid phosphatase activities, and rhizosphere microbial characteristics were investigated in Salt-T and Salt-S soybean genotypes under salt stress with a pot experiment. Salt stress significantly decreased the soil enzyme activities and changed the rhizosphere microbial structure in a genotype-dependent manner. In addition, 46 ASVs which were enriched in the Salt-T geotype under the salt stress, such as ASV19 (Alicyclobacillus), ASV132 (Tumebacillus), ASV1760 (Mycobacterium) and ASV1357 (Bacillus), which may enhance the tolerance to soybean under salt stress. Moreover, the network structure of Salt-T soybean was simplified by salt stress, which may result in soil bacterial communities being susceptible to external factors. Salt stress altered the strength of soil enzyme activities and the assembly of microbial structure in Salt-T and Salt-S soybean genotypes. Na+, NO3--N, NH4+-N and Olsen-P were the most important driving factors in the structure of bacterial community in both genotypes. Salt-T genotypes enriched several microorganisms that contributed to enhance salt tolerance in soybeans, such as Alicyclobacillus, Tumebacillus, and Bacillus. Nevertheless, the simplified network structure of salt-T genotype due to salt stress may render its bacterial community structure unstable and susceptible.
