Abstract:Per- and polyfluoroalkyl substances (PFAS) are of public health concern, because of their ubiquitous and extremely persistent occurrence, and depending on their structure, their bio-accumulative, mobile and toxic properties. Human health effects associated with exposure to PFAS include adverse effects on the immune system. In 2020, EFSA (the European Food Safety Authority) defined adverse effects on the immune system as the most critical effect for human health risk assessment, based on reduced antibody responses to childhood vaccines and similar effects observed in experimental animal studies. Likewise, the U.S. EPA (Environmental Protection Agency) considers PFAS-induced immunotoxicity, especially in children, as the critical effect for risk assessment. However, the mechanisms by which antibody concentrations are impacted are not completely understood. Furthermore, other targets of the immune system functions have been reported in the literature.

Abstract:Food contact materials are all materials and articles intended to come directly or indirectly into contact with food. Before being included in the positive European "Union list" of authorized substances (monomers, other starting substances and additives) for plastic food contact materials, the European Food Safety Authority (EFSA) must assess their safety "in use". If relevant for risk, the safety of the main impurities, reaction and degradation products originating from the manufacturing process is also evaluated. Information on genotoxicity is always required irrespective of the extent of migration and the resulting human exposure, in view of the theoretical lack of threshold for genotoxic events. The 2008 EFSA approach, requiring the testing of food contact materials in three in vitro mutagenicity tests, though still acceptable, is now superseded by the 2011 EFSA Scientific Committee's recommendation for only two complementary tests including a bacterial gene mutation test and an in vitro micronucleus test, to detect two main genetic endpoints (i.e., gene mutations and chromosome aberrations). Follow-up of in vitro positive results depends on the type of genetic effect and on the substance's systemic availability. In this study, we provide an analysis of the data on genotoxicity testing gathered by EFSA on food contact materials for the period 1992-2015. We also illustrate practical examples of the approaches that EFSA took when evaluating "non standard" food contact chemicals (e.g., polymeric additives, oligomer or other reaction mixtures, and nanosubstances). Critical Reviews in Food Science and Nutrition

Abstract:Food packaging is important for today's globalized food system, but food contact materials (FCMs) can also be a source of hazardous chemicals migrating into foodstuffs. Assessing the impacts of FCMs on human health requires a comprehensive identification of the chemicals they contain, the food contact chemicals (FCCs). We systematically compiled the "database on migrating and extractable food contact chemicals" (FCCmigex) using information from 1210 studies. We found that to date 2881 FCCs have been detected, in a total of six FCM groups (Plastics, Paper & Board, Metal, Multi-materials, Glass & Ceramic, and Other FCMs). 65% of these detected FCCs were previously not known to be used in FCMs. Conversely, of the more than 12'000 FCCs known to be used, only 1013 are included in the FCCmigex database. Plastic is the most studied FCM with 1975 FCCs detected. Our findings expand the universe of known FCCs to 14,153 chemicals. This knowledge contributes to developing non-hazardous FCMs that lead to safer food and support a circular economy.

Abstract:Increasing scientific evidence suggests potential adverse effects on children’s health from synthetic chemicals used as food additives, both those deliberately added to food during processing (direct) and those used in materials that may contaminate food as part of packaging or manufacturing (indirect). Concern regarding food additives has increased in the past 2 decades in part because of studies that increasingly document endocrine disruption and other adverse health effects. In some cases, exposure to these chemicals is disproportionate among minority and low-income populations. This report focuses on those food additives with the strongest scientific evidence for concern. Further research is needed to study effects of exposure over various points in the life course, and toxicity testing must be advanced to be able to better identify health concerns prior to widespread population exposure. The accompanying policy statement describes approaches policy makers and pediatricians can take to prevent the disease and disability that are increasingly being identified in relation to chemicals used as food additives, among other uses.

Abstract:Background: Over 1800 food contact chemicals (FCCs) are known to migrate from food contact articles used to store, process, package, and serve foodstuffs. Many of these FCCs have hazard properties of concern, and still others have never been tested for toxicity. Humans are known to be exposed to FCCs via foods, but the full extent of human exposure to all FCCs is unknown. Objective: To close this important knowledge gap, we conducted a systematic overview of FCCs that have been monitored and detected in human biomonitoring studies according to a previously published protocol. Methods: We first compared the more than 14,000 known FCCs to five biomonitoring programs and three metabolome/exposome databases. In a second step, we prioritized FCCs that have been frequently detected in food contact materials and systematically mapped the available evidence for their presence in humans. Results: For 25% of the known FCCs (3601), we found evidence for their presence in humans. This includes 194 FCCs from human biomonitoring programs, with 80 of these having hazard properties of high concern. Of the 3528 FCCs included in metabolome/exposome databases, most are from the Blood Exposome Database. We found evidence for the presence in humans for 63 of the 175 prioritized FCCs included in the systematic evidence map, and 59 of the prioritized FCCs lack hazard data. Significance: Notwithstanding that there are also other sources of exposure for many FCCs, these data will help to prioritize FCCs of concern by linking information on migration and biomonitoring. Our results on FCCs monitored in humans are available as an interactive dashboard (FCChumon) to enable policymakers, public health researchers, and food industry decision-makers to make food contact materials and articles safer, reduce human exposure to hazardous FCCs and improve public health. Impact statement: We present systematically compiled evidence on human exposure to 3601 food contact chemicals (FCCs) and highlight FCCs that are of concern because of their known hazard properties. Further, we identify relevant data gaps for FCCs found in food contact materials and foods. This article improves the understanding of food contact materials' contribution to chemical exposure for the human population and highlights opportunities for improving public health.

Abstract:In this work, we explore the processes involving food in contact with food contact materials (FCMs), and the subsequent changes in these materials. We delve into the historical context of "economic poisons", the development of food safety concerns, the regulation of food additives, and the rise of harmful chemicals in food. The focus is placed on mass spectrometry-based analyses of FCMs. We have examined substances, like bisphenols, and the chemical migration from various materials into food, highlighting the difficulty in identifying and managing these substances, especially unknown ones. This work further cements the inadequacy of current regulations in effectively controlling FCMs and mitigating associated risks. We conclude that despite the information provided by mass spectrometry, challenges persist in fully comprehending and regulating the interplay between FCMs and food under actual cooking conditions, emphasising the need for ongoing research and improved regulations to protect consumer health from these evolving chemical threats.

Abstract:Progesterone and LH concentrations were measured in the plasma of blood samples taken from forty-eight pregnant ewes on Days 100, 120 and 134 of gestation. The ewes, in two groups of twenty-four were maintained from Day 100 until parturition on two planes of nutrition which supplied daily energy and protein intakes of about 4-1 or 2-3 Mcal metabolizable energy and either 192 or 111 g digestible crude protein per ewe. Within the groups, the ewes carried one, two or three fetuses and the feed intake was adjusted according to litter size to produce a uniform nutritional state within the group. On Day 100, litter size affected the concentration of plasma progesterone (P less than 0-001), but had no effect on Days 120 or 134 when the ewes were fed according to litter size. The low feed intake however caused a significant increase in plasma progesterone concentrations. The LH concentrations showed no major changes during late pregnancy and no effect of nutrition or little size on the plasma hormone concentration was observed. It was concluded that the effect of litter size on plasma progesterone concentration recorded on Day 100 or gestation was partly mediated by level of nutrition.

Abstract:Currently the only methods for non-genotoxic carcinogenic hazard assessment accepted by most regulatory authorities are lifetime carcinogenicity studies. However, these involve the use of large numbers of animals and the relevance of their predictive power and results has been scientifically challenged. With increased availability of innovative test methods and enhanced understanding of carcinogenic processes, it is believed that tumour formation can now be better predicted using mechanistic information. A workshop organised by the European Partnership on Alternative Approaches to Animal Testing brought together experts to discuss an alternative, mechanism-based approach for cancer risk assessment of agrochemicals. Data from a toolbox of test methods for detecting modes of action (MOAs) underlying non-genotoxic carcinogenicity are combined with information from subchronic toxicity studies in a weight-of-evidence approach to identify carcinogenic potential of a test substance. The workshop included interactive sessions to discuss the approach using case studies. These showed that fine-tuning is needed, to build confidence in the proposed approach, to ensure scientific correctness, and to address different regulatory needs. This novel approach was considered realistic, and its regulatory acceptance and implementation can be facilitated in the coming years through continued dialogue between all stakeholders and building confidence in alternative approaches.

Abstract:The widespread use of plastic packaging for storing, transporting, and conveniently preparing or serving foodstuffs is significantly contributing to the global plastic pollution crisis. This has led to many efforts directed toward amending plastic packaging’s end of life, such as recycling, or alternative material approaches, like increasingly using paper for food packaging. But these approaches often neglect the critical issue of chemical migration: When contacting foodstuffs, chemicals that are present in packaging transfer into food and thus unwittingly become part of the human diet. Hazardous chemicals, such as endocrine disrupters, carcinogens, or substances that bioaccumulate, are collectively referred to as “chemicals of concern.” They can transfer from plastic packaging into food, together with other unknown or toxicologically uncharacterized chemicals. This chemical transfer is scientifically undisputed and makes plastic packaging a known, and avoidable, source of human exposure to synthetic, hazardous, and untested chemicals. Here, I discuss this issue and highlight aspects in need of improvement, namely the way that chemicals present in food packaging are assessed for toxicity. Further, I provide an outlook on how chemical contamination from food packaging could be addressed in the future. Robust innovations must attempt systemic change and tackle the issue of plastic pollution and chemical migration in a way that integrates all existing knowledge.

Abstract:Plant fiber-based food contact materials (FCMs) are considered as one of the perfect substitutes for the petroleum-based traditional plastic products. Plant fibers come from natural sources and are generally considered to be nontoxic and environmentally friendly. However, it has been reported that more migration of toxic substances may be found in the plant-based products. Therefore, this article first reviews the potential risks sources of plant fiber-based FCMs throughout their entire lifecycle. It also presents a summary of the analytical methods used to identify risk substances from the perspectives of non-targeted screening and safety assessment frameworks and compiles information regarding substances detected in non-targeted screening of plant fiber-based FCMs. In addition, rapid detection methods, including biosensors and emerging detection devices, have been described. The consumption demands and growth potential of plant fiber-based FCMs are enormous. However, based on the currently reported risks, it is necessary to improve the non-targeted screening database, to develop more comprehensive risk assessment methods and to gradually establish specific regulations and standards.

The Science of The Total Environment

Abstract:This study investigated the concentration profiles and geographical variability of contaminants in house dust across Europe. A collaborative trial (CT) was organized by the NORMAN network using pooled dust and advanced chromatographic and mass spectrometric techniques combined with suspect screening and non-target screening (NTS). Over 1200 anthropogenic compounds were tentatively identified. Additionally, seventy-five individual samples were subjected to target analysis and NTS. The median concentrations of most contaminants varied <3-fold across Europe, and the contaminant profile of European dust was similar to that of North American dust, which was investigated in a previous CT. This similarity may be attributed to the use of similar consumer articles and building materials throughout the developed world. Multivariate data analysis revealed geographical trends in contaminant distribution, with north-south gradients across Europe. Geographical trends were more frequently found for compounds with rapid release (pharmaceuticals, personal care products, fragrances, pesticides, biocides) and smoke-related compounds. The concentrations of chlorinated paraffins, polycyclic aromatic hydrocarbons (PAHs), perfluorinated alkyl substances and stimulants generally increased from north to south, whereas the biocides levels decreased from north to south. Despite widespread presence of in-use contaminants in dusts, some of the highest risks come from compounds that have been restricted for decades or more. These include di(2-ethylhexyl) phthalate (DEHP), polychlorinated biphenyl (PCB) 118 and polybrominated diphenyl ethers 47, 99, and 153. DEHP remains the most abundant contaminant in European house dust, while the other compounds are classified as persistent organic pollutants (POPs). Moreover, there is a striking lack of reliable toxicity data, particularly for emerging compounds. For instance, although acceptable daily intakes (ADIs) were examined for 202 compounds, only 46 had consensus-based ADI values. The results highlight the need for proactive measures to prevent hazardous chemicals from entering the market and for careful selection of substitute chemicals, when such are needed, to avoid regrettable substitutions.

Abstract:Per- and polyfluorinated alkyl substances (PFAS) can be found in food contact materials (FCMs). Therefore, migration into food occurs and contributes to human dietary exposure. Identification and minimization of possible exposure-pathways of these omnipresent and potentially toxic chemicals are important to mitigate health risks. Nonetheless, risk assessment procedures are mostly limited to considering the final PFAS concentrations in food without distinguishing the contribution of migration. Additionally, the use of food simulants during migration tests can potentially misrepresent migration results. Critically assessing reported migration behaviors of PFAS showed that the use of food simulants (especially Tenax®) in migration tests lead to an underestimation of PFAS exposure from FCMs. Reported PFAS concentrations migrated into food as well as consumption data from databases were used to estimate the dietary exposure. PFAS exclusively migrated from FCMs can considerably contribute to consumers' dietary exposure potentially impacting human health.

Abstract:The chemical burden on the environment and human population is increasing. Consequently, regulatory risk assessment must keep pace to manage, reduce, and prevent adverse impacts on human and environmental health associated with hazardous chemicals. Surveillance of chemicals of known, emerging, or potential future concern, entering the environment-food-human continuum is needed to document the reality of risks posed by chemicals on ecosystem and human health from a one health perspective, feed into early warning systems and support public policies for exposure mitigation provisions and safe and sustainable by design strategies. The use of less-conventional sampling strategies and integration of full-scan, high-resolution mass spectrometry and effect-directed analysis in environmental and human monitoring programmes have the potential to enhance the screening and identification of a wider range of chemicals of known, emerging or potential future concern. Here, we outline the key needs and recommendations identified within the European Partnership for Assessment of Risks from Chemicals (PARC) project for leveraging these innovative methodologies to support the development of next-generation chemical risk assessment.

Abstract:A chirped pulse amplification laser with zigzag slab Nd:glass amplifiers dedicated to x-ray laser research is described. The laser provides a 1.6 ps duration pulse with approximately 7 J energy at a repetition rate of 0.1 Hz. In the power amplifier system, laser light is amplified in a two-step manner: The first step is image-relayed multipass amplification up to approximately 1 J with a 10 mm x 10 mm beam. The second step is double-pass amplification up to >10 J with a 10 mm x 90 mm beam. By using this laser system, the saturated amplification of the Ni-like Ag laser at a wavelength of 13.9 nm has been successfully demonstrated.

Abstract:<p>Reports of environmental and human health impacts of per‐ and polyfluoroalkyl substances (PFAS) have greatly increased in the peer‐reviewed literature. The goals of this review are to assess the state of the science regarding toxicological effects of PFAS, and to develop strategies for advancing knowledge on the health effects of this large family of chemicals. Currently, much of the toxicity data available for PFAS are for a handful of chemicals, primarily legacy PFAS such as perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS). Epidemiological studies have revealed associations between exposure to specific PFAS and a variety of health effects, including altered immune and thyroid function, liver disease, lipid and insulin dysregulation, kidney disease, adverse reproductive and developmental outcomes, and cancer. Concordance with experimental animal data exists for many of these effects. However, information on modes of action and adverse outcome pathways must be expanded, and profound differences in PFAS toxicokinetic properties must be considered in understanding differences in responses between sexes and among species and life stages. With many health effects noted for a relative few example compounds, and hundreds of other PFAS in commerce lacking toxicity data, more contemporary and high throughput approaches such as read across, molecular dynamics, and protein modeling are proposed to accelerate the development of toxicity information on emerging and legacy PFAS, individually and as mixtures. Additionally, an appropriate degree of precaution, given what is already known from the PFAS examples noted here, may be needed to protect human health.</p><p>This article is protected by copyright. Abstract:Rapidly evolving technological methods and mechanistic toxicological understanding have paved the way for new science-based approaches for the determination of chemical safety in support of advancing public health. Approaches including read-across, high-throughput screening, in silico models, and organ-on-a-chip technologies were addressed in a 2017 workshop focusing on how scientists can move effectively toward a vision for 21st century food safety assessments. The application of these alternative methods, the need for further development of standardized practices, and the interpretation and communication of results were addressed. Expert presentations encompassed regulatory, industry, and academic perspectives, and the workshop culminated in a panel discussion in which participants engaged experts about current issues pertaining to the application of alternative methods in toxicological testing for food safety assessments.
